Formaldehyde (HCHO) and choline, trigonelline and N-trimethyl-L-lysine (TML) were detected and measured in tissue cultures of Datura innoxia Mill. by TLC and OPLC. The higher level of HCHO in young tissues appears to originate from the methylation reactions. This is in correlation with the high level of fully N-methylated substances. In spite of this the level of the fully N-methylated substances decreased with the aging of tissue. HCHO level also decreased with aging. However, in old tissues the amount of HCHO again increased. This HCHO originates mainly from demethylation reactions. Antioxidative response to cadmium in roots and leaves of tomato plants   总被引:1,自引:0,他引:1  
Treatment of tomato seedlings (Lycopersicon esculentum Mill. cv. 63/5 F1) with increasing CdCl2 concentrations in the culture medium resulted in Cd accumulation more important in roots than in leaves. Biomass production was severely inhibited, even at low Cd concentration. Cd reduced chlorophyll content in leaves and enhanced lipid peroxidation. An increase in antioxidative enzyme (superoxide dismutase, ascorbate peroxidase, guaiacol peroxidase, glutathione reductase) activities was more pronounced in leaves than in roots, while catalase activity increased only in roots. In addition, changes in isoenzyme composition were observed using the non-denaturing polyacrylamid gel electrophoresis.  相似文献

The aim of this study was to investigate the effect of tissue nitrogen concentration, as a consequence of nitrogen supply rate, on the susceptibility of tomato plants to three pathogens. We varied tissue N concentration by supplying N at different rates by adding nitrate in different, exponentially increasing amounts to the nutrient solution on which the tomato plants were grown. Separate experiments were carried out to test susceptibility of tomato plants to the bacterial speck-causing Pseudomonas syringae pv tomato, to the wilt agent Fusarium oxysporum f.sp. lycopersici and to tomato powdery mildew caused by Oidium lycopersicum. The effect of tissue N concentration appeared to be highly pathogen-dependent: there was no effect on susceptibility to F. oxysporum, but susceptibility to P. syringae and O. lycopersicum increased significantly with increasing N concentration. We have previously demonstrated the opposite for susceptibility to Botrytis cinerea: decreasing susceptibility with increasing N concentration. The apparent contradictory effects are discussed in relation to the effect of N supply on both the nutritional value of the plant tissue to the pathogen and on the concentration of resistance-related compounds. We conclude that the effect of changing both characteristics on disease susceptibility is highly pathogen-specific and is probably dependent on differences in resource requirements of the pathogen or the sensitivity of the pathogen to plant resistance reactions or on both these factors. Summary This paper details the solid-phase synthesis by N -9-fluorenylmethyloxycarbonyl (Fmoc) chemistry of a series of bivalent consolidated ligands, branched peptides with lengths of 22 to 25 residues. The target peptides were designed to, and in fact do, interact with greater specificity and higher affinity with the SH2 and SH3 domains of Abelson kinase in an SH(32) dual domain construct. Fmoc-O-phospho-l-tyrosine[Fmoc-Tyr(PO3H2)-OH] was used to introduce the required phosphotyrosine residues, and Fmoc-N -1-(4,4-dimethyl-2,6-dioxocyclohexylidene)ethyl-l-lysine [Fmoc-Lys(Dde)-OH] was used to introduce a branch point that allowed proper orientation of individual ligands. The resultant product peptides were characterized by amino acid analyses and electrospray mass spectra.This paper is based on a presentation given at the Symposium on Peptide Structure and Design as part of the 31st Annual ACS Western Regional Meeting held in San Diego, CA, USA, October 18–21, 1995.  相似文献

Using 13C-labelled internal standards and gas chromatography-mass spectrometry/multiple-ion monitoring the levels of xanthoxin (Xan) and 2-trans-xanthoxin (t-Xan) have been determined in stressed and non-stressed leaves of wildtype tomato (Lycopersicon esculentum Mill cv. Ailsa Craig), and the wilty mutants, notabilis (not), flacca (flc) and sitiens (sit). Levels of Xan were very low in all tissues. Ratios of t-Xan: Xan ranged from 10:1 to <500:1. In the wild-type and flc, t-Xan levels increased following stress. The results from feeding experiments using [13C]Xan and t-Xan demonstrated that whilst wild-type and not plants readily converted Xan into abscisic acid (ABA), flc and sit plants converted only a small amount of applied Xan into ABA. In all plants t-Xan was not converted into ABA. These results indicate that the flc and sit mutants are impaired in ABA biosynthesis because they are unable to convert Xan into ABA, whereas the not mutant is blocked at a metabolic step prior to Xan. Another possible ABA precursor, ABA-1,4-trans-diol (ABA-t-diol) was found to occur in wild-type and mutant tissue. All four tissues could convert [2H]ABA-t-diol to ABA. Two lines of tomato (Lycopersicon esculentum Mill.) representing extremes in utilization effciency of absorbed Ca were studied to detect internal differences in Ca transport and distribution and factors responsible for strain differences in susceptibility to low Ca-stress. Differences in efficiency of Ca use were expressed as CaER (mg of dry weight produced for each mg of Ca absorbed by the plant).Ca-efficiency in line 113(E) appeared to be associated with a slow continuous movement of absorbed Ca, allowing for continued growth of the shoot apex and upper lamina under Ca-deficiency conditions. In the inefficient line 67(I), in contrast, Ca was rapidly deposited in the lower leaves with little upward movement in the plant after absorption.Fractionation of tissue Ca into various chemical forms suggested that Ca inefficiency also was associated with higher concentrations of insoluble Ca in the shoot tissue. The efficient line, although sustaining growth at lower levels of Ca, was capable of maintaining a higher ratio of soluble to insoluble Ca in all shoot tissues.Calcium was concentrated in the lower plant tissues of the inefficient strain, limiting its availability for continued shoot growth.Autoradiographs of lines fed45Ca during the final 8 days of a 24-day experiment suggested that upward movement was sustained in line 113(E), in spite of vastly reduced transpiration rates and a root system characterized by leakage of K ions from the roots back into the solution.  相似文献

Plant cell walls play a major role in the outcome of host-parasite interactions. Wall fragments released from the plant, and/or the fungal pathogen, can act respectively as endogenous and exogenous elicitors of the defence response, and other wall components, such as callose, lignin, or hydroxyproline-rich glycoproteins, can inhibit pathogen penetration and/or spreading. We have previously demonstrated that calli from tomato cultivars resistant in vivo to Fusarium oxysporum f.sp. lycopersici show a high amount of polysaccharides in vitro. The aim of the present work was to assess the possible role of polysaccharide content and/or synthetic capacity in determining the competence of plant cells for active defence. For this purpose, tomato cell clones with increased and decreased polysaccharide (FL+, FL-) and callose (A+, A-) content have been selected by means of specific stains as visual markers and tested for the effect of these changes on the extent of response to Fusarium. The analysis of several parameters known to be indicative of active defence (cell browning after elicitor treatment, peroxidase and -glucanase induction and inhibition of fungal growth in dual culture) clearly shows that FL+ and A+ clones have acquired an increased competence for the activation of defence response. The results are thoroughly discussed in terms of an evaluation of the relative importance of constitutive and/or inducible polysaccharide synthetic capacity for plant response to pathogens, and their possible regulation by plant physiological backgrounds.  相似文献

Applying the method of pyrolysis coupled with gas chromatography (PYR-GC) the content of endogenous acetylcholine (ACh) was investigated in the extracts obtained from tomato (Lycopersicon esculentum Mill.). Seven-day-old seedlings of wild type (WT) and phytochrome mutants au (aurea), hp (high pigment), fri (far-red light insensitive) and tri (temporarily red light insensitive) were studied. In the analyzed material the presence of choline and acetylcholine was discovered. The highest content of ACh (381 mmole/g of fresh weight) was found in tomato cotyledons, whereas the lowest amount (162 nmole/g of fresh weight) in roots. The level of ACh in the plants grown under the continuous light was higher than in etiolated ones. However, no considerable differences in the concentrations of ACh in au and tri seedlings grown under the continuous light and in darkness were observed. The irradiation of etiolated seedlings of wild type with red light was accompanied by the increase of endogenous level of ACh. The pulse of far-red light applied directly after red light reversed this stimulating effect. A similar effect of both light wavelengths on the content of ACh was also found in the case of the tri mutant. On the other hand, in the case of fri mutant, pulse of red light caused the drop in the content of ACh, whereas far-red applied after red light caused visible increase in the level of the investigated substance. In tissues of au mutant no effect of red and far-red lights on the concentration of ACh was established.  相似文献

This research aims to examine the effect of cadmium uptake on lipid composition and fatty acid biosynthesis, in young leaves of tomato treated seedlings (Lycopersicon esculentum cv. Ibiza F1). Results in membrane lipids investigations revealed that high cadmium concentrations affect the main lipid classes, leading to strong changes in their composition and fatty acid content. Thus, the exposure of tomato plants to cadmium caused a concentration-related decrease in the unsaturated fatty acid content, resulting in a lower degree of fatty acid unsaturation. The level of lipid peroxides was significantly enhanced at high Cd concentrations. Studies of the lipid metabolism using radioactive labelling with [1-14C]acetate as a major precursor of lipid biosynthesis, showed that levels of radioactivity incorporation in total lipids as well as in all lipid classes were lowered by Cd doses. In total lipid fatty acids, [1-14C]acetate incorporation was reduced in tri-unsaturated fatty acids (C16:3 and C18:3); While it was enhanced in the palmitic (C16:0), palmitoleic (C16:1), stearic (C18:0) and linoleic (C18:2) acids. [1-14C]acetate incorporation into C16:3 and C18:3 of galactolipids [monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG)] and some phospholipids [phosphatidylcholine (PC) and phosphatidylglycerol (PG)] was inhibited by Cd stress. Our results showed that in tomato plants, cadmium stress provoked an inhibition of polar lipid biosynthesis and reduced fatty acid desaturation process.  相似文献

We analysed the stable isotope composition of emitted N2O in a one-year field experiment (June 1998 to April 1999) in unfertilized controls, and after adding nitrogen by applying slurry or mineral N (calcium ammonium nitrate). Emitted N2O was analysed every 2–4 weeks, with additional daily sampling for 10 days after each fertilizer application. In supplementary soil incubations, the isotopic composition of N2O was measured under defined conditions, favouring either denitrification or nitrification. Soil incubated for 48 h under conditions favouring nitrification emitted very little N2O (0.024 mol gdw –1) and still produced N2O from denitrification. Under denitrifying incubation conditions, much more N2O was formed (0.91 mol gdw –1 after 48 h). The isotope ratios of N2O emitted from denitrification stabilized at 15N = –40.8 ± 5.7 and 18O = 2.7 ± 6.3. In the field experiment, the N2O isotope data showed no clear seasonal trends or treatment effects. Annual means weighted by time and emission rate were 15N = –8.6 and 18O = 34.7 after slurry application, 15N = –4.6 and 18O = 24.0 after mineral fertilizer application and 15N = –6.4 and 18O = 35.6 in the control plots, respectively. So, in all treatments the emitted N2O was 15N-depleted compared to ambient air N2O (15N = 11.4 ± 11.6, 18O = 36.9 ± 10.7). Isotope analyses of the emitted N2O under field conditions per se allowed no unequivocal identification of the main N2O producing process. However, additional data on soil conditions and from laboratory experiments point to denitrification as the predominant N2O source. We concluded (1) that the isotope ratios of N2O emitted from the field soil were not only influenced by the source processes, but also by microbial reduction of N2O to N2 and (2) that N2O emission rates had to exceed 3.4 mol N2O m–2 h–1 to obtain reliable N2O isotope data.  相似文献

The apparent involvement of ornithine decarboxylase (ODC) and putrescine in the early stages of fruit growth in tomato (Lycopersicon esculentum Mill.) has been previously described. Further evidence presented here supports the direct involvement of ODC and putrescine in the cell division process in tomato fruits. In tomato fruits grown in vitro, in which basic growth processes are inhibited, the activity of ODC and arginine decarboxylase (ADC) and the level of free polyamines were reduced. While ODC and ADC activity was correlated with the period of cell division in the tomato fruit, the free polyamine content was correlated with the DNA content, cell size, and fruit fresh weight. The addition of exogenous putrescine, however, did not restore the basic growth processes in the fruits grown in vitro.  相似文献

Temporal and spatial changes in 13C and 15N of seston (mainly phytoplankton) and isotopic relationship between seston and the lake anchovy (Coilia ectenes) were studied in the large eutrophic freshwater Lake Chaohu in China. Much of the spatial and temporal variation in 13C of lake anchovies was explained by variation in seston, indicating a strong link between pelagic primary production and higher order consumers. Because the lake is shallow, there were no significant differences in 13C and 15N of seston between surface and overlying waters. Spatially, the relatively high 13C and 15N of seston in the western part of the lake might be due to high levels of anthropogenically derived N and C introduced from the surrounding cities through sewage drainage systems. The trophic position of the lake anchovy in the food web of Lake Chaohu was estimated to be 2.9–4.1 (3.5 ± 0.4), which agrees well with the previous stomach content analysis suggesting that the lake anchovy fed both on zooplankton and small planktivorous fishes.  相似文献

Lactobacillus plantarum ATCC 8014 grew poorly on raffinose agar plates, but large mutant colonies appeared in high frequency from a thin film of background growth. The -galactosidase and -galactosidase activities ofL. plantarum ATCC 8014 and a mutant strain were studied in static cultures and pH-controlled fermenter cultures. Both -galactosidase and -galactosidase production were inducible in the parental strain; the induction was not needed in the mutant. The -galactosidase activity of both strains was repressed by glucose but not by -methyl-D-glucoside. The mutant phenomenon might be an obstacle in connection to traditionalLactobacillus identification by means of carbohydrate fermentation.  相似文献

The tomato nuclear genome was determined to have a G+C content of 37% which is among the lowest reported for any plant species. Non-coding regions have a G+C content even lower (32% average) whereas coding regions are considerably richer in G+C (46%).5-methyl cytosine was the only modified base detected and on average 23% of the cytosine residues are methylated. Immature tissues and protoplasts have significantly lower levels of cytosine methylation (average 20%) than mature tissues (average 25%). Mature pollen has an intermediate level of methylation (22%). Seeds gave the highest value (27%), suggesting de novo methylation after pollination and during seed development.Based on isoschizomer studies we estimate 55% of the CpG target sites (detected by Msp I/Hpa II) and 85% of the CpNpG target sites (detected by Bst NI/Eco RI)are methylated. Unmethylated target sites (both CpG and CpNpG) are not randomly distributed throughout the genome, but frequently occur in clusters. These clusters resemble CpG islands recently reported in maize and tobacco.The low G+C content and high levels of cytosine methylation in tomato may be due to previous transitions of 5mCT. This is supported by the fact that G+C levels are lowest in non-coding portions of the genome in which selection is relaxed and thus transitions are more likely to be tolerated. This hypothesis is also supported by the general deficiency of methylation target sites in the tomato genome, especially in non-coding regions.Using methylation isoschizomers and RFLP analysis we have also determined that polymorphism between plants, for cytosine methylation at allelic sites, is common in tomato. Comparing DNA from two tomato species, 20% of the polymorphisms detected by Bst NI/Eco RII could be attributed to differential methylation at the CpNpG target sites. With Msp I/Hpa II, 50% of the polymorphisms were attributable to methylation (CpG and CpNpG sites). Moreover, these polymorphisms were demonstrated to be inherited in a mendelian fashion and to co-segregate with the methylation target site and thus do not represent variation for transacting factors that might be involved in methylation of DNA. The potential role of heritable methylation polymorphism in evolution of gene regulation and in RFLP studies is discussed.  相似文献

Summary The hypothesis that relative water motion and boundary layer diffusion processes affect carbon isotope ratios of aquatic plants was tested in tidal pool and surge zone comparisons of the surfgrass Phyllospadix spp. No evidence was found that submerged plants growing in still upper tidal pools were isotopically different from those growing submerged in lower tidal surge zones. Significant decreases in 13C/12C ratios for plants growing emersed in the intertidal may have been caused by uptake of atmospheric carbon dioxide. Marine algae (Egregia menziesii and Halosaccion americanum) growing at the same location and tidal elevations as the seagrasses showed somewhat different isotopic fractionation patterns, suggesting that causes of isotopic variability in the seagrasses were not necessarily the same as those in the two marine algae.  相似文献

The concentrations of arginine, protein and total nitrogen (N) and the abundance of15N were measured in 3-and 4-year-old needles of Scots pine trees fertilized with either 0 (C), 36 (N1) or 73 (N2) kg N ha-1 year-1 annually for 22 years (average doses of N). Remaining green needles and needles that were shed were compared and removal of N from total, protein and arginine pools was calculated. Earlier investigations had shown that high arginine concentrations are found in needles of trees that have an excessive N supply (Näsholm and Ericsson 1990). This study aimed to elucidate the fate of the accumulated arginine during needle senescence. It was speculated that a low removal of arginine during senescence would implicate that the primary function of arginine is in N detoxification and not in N storage. Moreover, litter quality would be altered if needles are shed with high concentrations of arginine and this might affect the turnover of N in forest ecosystems. In remaining green needles, the concentration of total N increased with increasing N supply. Protein N concentrations were higher in fertilized trees, but did not differ between the two N treatments. Arginine N was low in C and N1 trees but high in N2 trees. Senescent needles from C and N1 trees had about equal total N concentrations while in N2 trees this concentration was significantly higher. Protein N in senescent needles did not differ between treatments. Arginine N, however, was less than 0.1 mg g–1 dw in C and N1 trees but was higher than 1.5 mg g–1 dw in N2 trees. Removal of N was highest in N1 trees followed by C trees while N2 trees removed least N from senescing needles. The high concentration of total N in senescent needles from N2 trees was to a great extent explained by a high arginine concentration.The 15N value of remaining, green needles was higher (less negative) in N2 trees than in C and N1 trees. The same pattern was found for senescent needles. Comparisons of 15N values between remaining, green and senescent needles within each treatment showed a significant increase in 15N for all treatments during senescence possibly indicating losses of N as NH3 (g) from needles during senescence. It is concluded that arginine, accumulated in response to high N supply, is retranslocated only to a small extent during needle senescence. The ecological and physiological implications of this finding are discussed.  相似文献

Summary Natural carbon and nitrogen isotope ratios were measured in different compartments (needles and twigs of different ages and crown positions, litter, understorey vegetation, roots and soils of different horizons) on 5 plots of a healthy and on 8 plots of a declining Norway spruce (Picea abies (L.) Karst.) forest in the Fichtelgebirge (NE Bavaria, Germany), which has recently been described in detail (Oren et al. 1988a; Schulze et al. 1989). The 13C values of needles did not differ between sites or change consistently with needle age, but did decrease from the sun-to the shade-crown. This result confirms earlier conclusions from gas exchange measurements that gaseous air pollutants did no long-lasting damage in an area where such damage was expected. Twigs (13C between-25.3 and-27.8) were significantly less depleted in 13C than needles (13C between-27.3 and-29.1), and 13C in twigs increased consistently with age. The 15N values of needles ranged between-2.5 and-4.1 and varied according to stand and age. In young needles 15N decreased with needle age, but remained constant or increased in needles that were 2 or 3 years old. Needles from the healthy site were more depleted in 15N than those from the declining site. The difference between sites was greater in old needles than in young ones. This differentiation presumably reflects an earlier onset of nitrogen reallocation in needles of the declining stand. 15N values in twigs were more negative than in needles (-3.5 to-5.2) and showed age- and stand-dependent trends that were similar to the needles. 15N values of roots and soil samples increased at both stands with soil depth from-3.5 in the organic layer to +4 in the mineral soil. The 15N values of roots from the mineral soil were different from those of twigs and needles. Roots from the shallower organic layer had values similar to twigs and needles. Thus, the bulk of the assimilated nitrogen was presumably taken up by the roots from the organic layer. The problem of separation of ammonium or nitrate use by roots from different soil horizons is discussed.  相似文献

Infection by tobacco mosaic virus improved the suitability of tomato, Lycopersicon esculentum Mill. for survival of Colorado potato beetle, Leptinotarsa decemlineata (Say), larvae. This improvement was due, at least in part, to the increase in total nitrogen content of virus-infected plants. The simultaneous increase in tomatine content had no discernable effect on L. decemlineata survival. Our results are consistent with the suggestions that virus infection may improve the suitability of partially resistant or non-preferred hosts, and that virus infection may facilitate the adaption of phytophagous insects to such marginal host plant species.
Résumé Pour vérifier l'hypothèse selon laquelle une contamination par un virs végétal peut accroître la consommation de plantes délaissées ou partiellement résistantes, nous avons examiné les interactions entre Leptinotarsa decemlineata Say, Lycopersicon esculentum Mill., et le virus de la mosaïque du tabac (TMV). La survie des larves de L. decemlineata a augmenté avec la contamination par TMV; les teneurs en azote et en tomatine étaient toutes deux plus élevées dans les plantes contaminées par le virus. La survie a augmenté linéairement avec la teneur en azote, mais sans dépendre de la teneur en tomatine. L'influence positive pour L. decemlineata de la contamination par le virus peut être attribuée au moins en partie à la teneur en azote plus élevée. Nos résultats correspondent à l'hypothèse selon laquelle la consommation de plantes délaissées ou partiellement résistantes peut être augmentée par la contamination virale, et que cette contamination peut faciliter l'adaptation d'insectes phytophages sur des plantes marginalement consommables.
