Signal transduction during cold, salt, and drought stresses in plants
Abiotic stresses, especially cold, salinity and drought, are the primary causes of crop loss worldwide. Plant adaptation to environmental stresses is dependent upon the activation of cascades of molecular networks involved in stress perception, signal transduction, and the expression of specific stress-related genes and metabolites. Plants have stress-specific adaptive responses as well as responses which protect the plants from more than one environmental stress. There are multiple stress perception and signaling pathways, some of which are specific, but others may cross-talk at various steps. In this review article, we first expound the general stress signal transduction pathways, and then highlight various aspects of biotic stresses signal transduction networks. On the genetic analysis, many cold induced pathways are activated to protect plants from deleterious effects of cold stress, but till date, most studied pathway is ICE-CBF-COR signaling pathway. The Salt-Overly-Sensitive (SOS) pathway, identified through isolation and study of the sos1, sos2, and sos3 mutants, is essential for maintaining favorable ion ratios in the cytoplasm and for tolerance of salt stress. Both ABA-dependent and -independent signaling pathways appear to be involved in osmotic stress tolerance. ROS play a dual role in the response of plants to abiotic stresses functioning as toxic by-products of stress metabolism, as well as important signal transduction molecules and the ROS signaling networks can control growth, development, and stress response. Finally, we talk about the common regulatory system and cross-talk among biotic stresses, with particular emphasis on the MAPK cascades and the cross-talk between ABA signaling and biotic signaling.

Plant growth and crop production are limited by environmental stress. We used a large population of transgenic Arabidopsis expressing rice full-length cDNAs to isolate the rice genes that improve the tolerance of plants to environmental stress. By sowing T2 seeds of the transgenic lines under conditions of salinity stress, the salt-tolerant line R07047 was isolated. It expressed a rice gene, OsSMCP1, which encodes a small protein with a single C2 domain, a Ca2+-dependent membrane-targeting domain. Retransformation of wild-type Arabidopsis revealed that OsSMCP1 is responsible for conferring the salt tolerance. It is particularly interesting that R07047 and newly constructed OsSMCP1-overexpressing Arabidopsis showed enhanced tolerance not only to high salinity but also to osmotic, dehydrative, and oxidative stresses. Furthermore, R07047 showed improved resistance to Pseudomonas syringae. The OsSMCP1 expression in rice is constitutive. Particle-bombardment-mediated transient expression analysis revealed that OsSMCP1 is targeted to plastids in rice epidermal cells. It induced overexpression of several nuclear encoded genes, including the stress-associated genes, in transgenic Arabidopsis. No marked morphological change or growth retardation was observed in R07047 or retransformants. For molecular breeding to improve the tolerance of crops against environmental stress, OsSMCP1 is a promising candidate.

Being sessile, plants are subjected to a diverse array of environmental stresses during their life span. Exposure of plants to environmental stresses results in the generation of reactive oxygen species (ROS). These activated oxygen species tend to oxidize various cellular biomolecules like proteins, nucleic acids, and lipids, a process that challenges the core existence of the cell. To prevent the accumulation of these ROS and to sustain their own survival, plants have developed an intricate antioxidative defence system. The antioxidative defence system comprises various enzymatic and nonenzymatic molecules, produced to counter the adverse effect of environmental stresses. A sizable number of these molecules belong to the category of compounds called secondary metabolites. Secondary metabolites are organic compounds that are not directly involved in the growth and development of plants but perform specialized functions under a given set of conditions. Absence of secondary metabolites results in long-term impairment of the plant’s survivability. Such compounds generally include pigments, phenolics, and so on. Plant phenolic compounds such as flavonoids and lignin precursors have been reported to accumulate in response to various biotic and abiotic stresses and are regarded as crucial defence compounds that can scavenge harmful ROS. Another important category of plant metabolites, called brassinosteroids, exhibit stress regulatory and growth-promoting activity and are classified as phytohormones. Elucidation of the physiological and molecular effects of secondary metabolites and brassinosteroids have catapulted them as highly promising and environment-friendly natural substances, suitable for wider application in plant protection and crop yield promotion. The present review focuses on our current understanding of how plants respond to the generation of excessive ROS and the role of secondary metabolites and brassinosteroids in countering the adverse effects of environmental stresses.

Salinity stress is one of the major abiotic stresses affecting plant growth and productivity globally. In order to improve the yields of plants growing under salt stress bear remarkable importance to supply sustainable agriculture. Acclimation of plants to salinized condition depends upon activation of cascade of molecular network involved in stress sensing/perception, signal transduction, and the expression of specific stress-related genes and metabolites. Isolation of salt overly sensitive (SOS) genes by sos mutants shed us light on the relationship between ion homeostasis and salinity tolerance. Regulation of antioxidative system to maintain a balance between the overproduction of reactive oxygen species and their scavenging to keep them at signaling level for reinstating metabolic activity has been elucidated. However, osmotic adaptation and metabolic homeostasis under abiotic stress environment is required. Recently, role of phytohormones like Abscisic acid, Jasmonic acid, and Salicylic acid in the regulation of metabolic network under osmotic stress condition has emerged through crosstalk between chemical signaling pathways. Thus, abiotic stress signaling and metabolic balance is an important area with respect to increase crop yield under suboptimal conditions. This review focuses on recent developments on improvement in salinity tolerance aiming to contribute sustainable plant yield under saline conditions in the face of climate change.

Agriculture productivity is severely hampered by soil salinity, drought and other environmental stresses. Studies on stress-resistant plants (halophytes, xerophytes, accumulating plants for specific toxic ions) have illuminated some mechanisms of stress tolerance in plants at metabolic or molecular levels, which gave some clues on how to genetically engineer stress-tolerant crops. With the isolation of more stress-responsive genes, genetic engineering with modified expression of stress responsive genes may be an effective way to produce stress-tolerant crops. In the present report, two genes (PEAMT and BADH) encoding the corresponding key enzymes for choline and glycine betaine (an important osmoprotectant) biosynthesis in plants were isolated in oilseed rape, an important oil crop in the world. Effects of salt stress on their expression were studied with quantitative PCR and their potential use in the genetic engineering of oilseed rape was discussed.

In natural habitats, especially in arid areas, plants are often simultaneously exposed to multiple abiotic stresses, such as salt, osmotic and heat stresses. However, most analyses of gene expression in stress responses examine individual stresses. In this report, we compare gene expression in individual and combined stresses. We show that combined stress treatments with salt, mannitol and heat induce a unique pattern of gene expression that is not a simple merge of the individual stress responses. Under multiple stress conditions, expression of most heat and salt stress‐responsive genes increased to levels similar to or higher than those measured in single stress conditions, but osmotic stress‐responsive genes increased to lower levels. Genes up‐regulated to higher levels under multiple stress condition than single stress conditions include genes for heat shock proteins, heat shock regulators and late embryogenesis abundant proteins (LEAs), which protect other proteins from damage caused by stresses, suggesting their importance in multiple stress condition. Based on this analysis, we identify candidate genes for engineering crop plants tolerant to multiple stresses.

Endophytes are micro‐organisms including bacteria and fungi that survive within healthy plant tissues and promote plant growth under stress. This review focuses on the potential of endophytic microbes that induce abiotic stress tolerance in plants. How endophytes promote plant growth under stressful conditions, like drought and heat, high salinity and poor nutrient availability will be discussed. The molecular mechanisms for increasing stress tolerance in plants by endophytes include induction of plant stress genes as well as biomolecules like reactive oxygen species scavengers. This review may help in the development of biotechnological applications of endophytic microbes in plant growth promotion and crop improvement under abiotic stress conditions.

Significance and Impact of the Study

Increasing human populations demand more crop yield for food security while crop production is adversely affected by abiotic stresses like drought, salinity and high temperature. Development of stress tolerance in plants is a strategy to cope with the negative effects of adverse environmental conditions. Endophytes are well recognized for plant growth promotion and production of natural compounds. The property of endophytes to induce stress tolerance in plants can be applied to increase crop yields. With this review, we intend to promote application of endophytes in biotechnology and genetic engineering for the development of stress‐tolerant plants.
