Four heavy metal staining methods have been applied to frog lung surfactant. Among them, the iodoplatinate method is the only one that almost exclusively visualizes the phospholipid moiety being produced in the lamellated bodies of the pulmonary epithelial cells and forming the backbone of organized structures within the extracellular lining layer. The other three techniques-ruthenium red-osmium tetroxide, osmium tetroxide-ferrocyanide, acidic phosphotungstic acid in chromatic (Rambourg technique)--more or less give electron contrast to glycoproteins and to a lesser extent to the hydrophilic parts of phospholipids. They all show the extracellular lining layer to be a two component system: the content of the lamellar bodies form--when released--membranous configurations, similar to those observed in mammalian lungs; they unfold in an amorphous hypophase, which is apparently secreted by goblet cells of the pulmonary epithelium.  相似文献

Summary Location of acid phosphatase outside and inside the osteoclast was studied by electron microscopic histochemistry. Osteoclasts with a ruffled border apposed to the bone showed enzyme activity in a) membrane-limited cytoplasmic bodies of different dimensions, b) some Golgi vesicles and inner Golgi cisternae, c) vacuoles and vacuole-like profiles, d) extracellular channels and channel expansions in the ruffled border, e) cell-bone interspace. The possibility of bone degradation by lysosomal enzymes both in the cytoplasmic vacuoles and in the cell-bone interspace is discussed.This research was supported by the Danish Medical Research Council. Summary Small tissue blocks of various organs of the rat were incubated for gradually increasing times in a neutral buffer-sucrose medium modelling the main parameters of the histochemical incubations. Following incubation the blocks were fixed in paraformaldehydeosmium and embedded in Durcupan. The electron microscopic study revealed that even after 40 minutes of incubation prior to fixation, fine structure is preserved satisfactorily from the histochemical point of view. With consideration to further advantages of such a proceeding, the incubation of unfixed tissue blocks for electron-histochemistry is recommended.  相似文献

Summary Scanning and transmission electron microscopy were used to study the inner architecture of the frog lung. In some specimens the alveolar surface mucus layer was removed to permit the examination of underlying features. The inner surface of the frog's lung is covered by a layer of microvilli belonging to only one type of epithelial cells. The boundaries of these epithelial cells are demarcated by small ridges. Different degrees of lung expansion cause variations of the surface topography. The morphology of certain surface features is examined in detail. Several methods of drying the specimens are compared.The author wishes to thank Dr. I. E. The influence of long-term (6 hours) immobilization stress on morphofunctional state of lung air-blood barrier was studied in experiments of the rats. It was shown that stress provoked the marked ultrastructural changes in the lungs, which were as follows: lung tissue oedema, pronounced thickening of lung air-blood barrier and its separate layers, edema-hemorrhagic syndrome, alveolar epithelial injury, disturbance of lung surfactant systems. Such a pathological complex may be designated as a "stress lung".  相似文献

Summary A technique is described, by means of which the undesirable effects by freezing and thawing necessary in cryostat histochemistry might be avoided using embedding in polyetylene glycol, which does not affect enzyme activity nor ultrastructural appearance of cellular detail. The 52 alveolar brush cells (ABC) were revealed in the semi-thin sections of the rat lung tissue, metachromatically stained with toluidine blue. Characteristic features of the ABC on the light microscope level were the following: pyramidal body shape, basal position of the nucleus, darker stain tinge of the cytoplasm than that of other alveolar cells, the presence of microvilli on the small free cell surface. There is one ABC per 21 alveocytes, type 2, and 15 alveocytes, type 1. 41.1% of the ABC are localized in places of adjacent alveolus walls junction, 32.7%--on the alveolus wall facing the alveolus cavity, 16.8%--near the alveolus entrance; 9.4% of the cells are directed into the cavities of two neughbo ring alveoli or settle down near the pores of Kohn. In parallel electron microscopy there was revealed in ABC a form of granular cytoplasmic reticulum (unusual for other alveolocytes) in the form of blocks made up of 5-8 cysternae as if adherent one to another, bundles of filaments and microtubules, vacuoles in the apical cytoplasm. The ultrastructure of ABC, their topography, and incidence in the alveoli of rats evidenced their chemoreceptor nature.  相似文献

Synopsis Sympathetic ganglia of 13 to 19-week-old human foetuses were cultured in small pieces with and without nerve growth factor for up to 5 weeksin vitro. The cultures were studied using phase-contrast, fluorescence and electron microscopy. Monoamines were demonstrated with the formaldehyde-induced fluorescence method, with and without pretreatment of the cultures with catecholamines or monoamine oxidase inhibitor.In the long-term cultures, primitive sympathetic cells, sympathicoblasts of types I and II, and young sympathetic neurons showed a fine structure identical to that described earlierin vivo. There were virtually no satellite or Schwann cells in the cultures. The neurons showed a considerable capacity to grow new nerve fibres in culture, even without nerve growth factor. Nerve terminals with accumulations of other nervous structures. Large granular vesicles were regularly found in the sympathicoblasts after glutaraldehyde-osmium tetroxide fixation. After permanganate fixation, dense-cored vesicles typical of adrenergic neurons were not seen, either in the perikarya, or in the processes, although it was possible to demonstrate specific fluorescence. No small intensely fluorescent (SIF) cells were observed.Variable formaldehyde-induced fluorescence was observed in the nerve cell perikarya and nerve fibres. The intensity of the fluorescence increased after treatment of the cultures with monoamine oxidase inhibitor and after incubation with catecholamines.  相似文献

The presence of toxic heavy metals in natural environments entails a potential health hazard for humans. Metal contaminants in these environments are usually tightly bound to colloidal particles and organic matter. On the other hand, the potential of these metals towards chelation by different chelating agents presents a good characteristic for their removal from the environment. On this basis, two chitosan/anionic surfactant complexes were prepared and evaluated for their ability to remove heavy metals from aqueous solutions. The experimental results of the uptake of metal ions including Cu2+, Sn2+, Co2+ and Ni2+ are reported in this study. The results show that modified chitosan with short‐spacer group cross‐linkers has a higher potential for heavy metal uptake than long‐chain cross‐linker‐modified chitosan. Also, increasing the electronegativity of the heavy metals increases their uptake from the medium. Increasing the time of exposure of the heavy metals to the modified polymer increases the efficiency of the metal uptake process.  相似文献

Varying dose levels of methylmercuric chloride (MMC), 1000 ppm (5 mg through 15 mg/kg of body weight), were administered via an intragastric tube to pregnant ICR Swiss/Webster mice on day 9 of gestation. The animals were killed on gestational day 18 and the fetuses removed. Fetal lung sections were processed for light and electron microscopy. A group of animals treated with physiological saline in a similar manner served as the controls. The fetal lungs from treated animals were hypoplastic and retarded in development. The severity of pulmonary changes increased with the dose-levels of MMC. Vacuolation and lysis of mitochondria were seen in fetal lungs. Mitochondrial damage increased in severity with dose-level of methylmercuric chloride.  相似文献

Summary Mitosis in the imperfect yeast-like basidiomyceteBullera alba was studied by comparative light and electron microscopy. During mitosis the chromatin containing part of the nucleus moved into the progeny cell, and the nucleolus containing part of the nucleus remained in the parent cell. The two portions of the nucleus then separated and the nucleolar part degenerated. Metaphase and anaphase took place in the progeny cell. Subsequently one mass of chromatin returned to the parent cell, and two new nuclei were formed. The study concentrated on the nuclear envelope, nucleolus, spindle pole body, chromatin, spindle, and cytoplasmic microtubules. Mitosis inB. alba was compared with reports of mitosis in other basidiomycetes, theUredinales, and theAscomycotina and was deemed closest to the heterobasidiomycete yeasts.Histochemical evidence for the presence of lipid, glycogen, and polyphosphate in the cytoplasm was presented.  相似文献
