The Spatial Distribution of Sucrose Synthase Isozymes in Barley   总被引:1,自引:0,他引:1       下载免费PDF全文
The sucrose (Suc) synthase enzyme purified from barley (Hordeum vulgare L.) roots is a homotetramer that is composed of 90-kD type 1 Suc synthase (SS1) subunits. Km values for Suc and UDP were 30 mM and 5 [mu]M, respectively. This enzyme can also utilize ADP at 25% of the UDP rate. Anti-SS1 polyclonal antibodies, which recognized both SS1 and type 2 Suc synthase (SS2) (88-kD) subunits, and antibodies raised against a synthetic peptide, LANGSTDNNFV, which were specific for SS2, were used to study the spatial distribution of these subunits by immunoblot analysis and immunolocalization. Both SS1 and SS2 were abundantly expressed in endosperm, where they polymerize to form the five possible homo- and heterotetramers. Only SS1 homotetramers were detected in young leaves, where they appeared exclusively in phloem cells, and in roots, where expression was associated with cap cells and the vascular bundle. In the seed both SS1 and SS2 were present in endosperm, but only SS1 was apparent in the chalazal region, the nucellar projection, and the vascular bundle. The physiological implications for the difference in expression patterns observed are discussed with respect to the maize (Zea mays L.) model.  相似文献

The European minkMustela lutreola Linnaeus, 1761 and the European polecatMustela putorius Linnaeus, 1758 are sympatric in southwestern France. They are related species but the former is highly threatened whereas the latter maintains good populations. Nine European mink and 14 polecats were radiotracked in the Landes de Gascogne region to compare their space use and to identify appropriate conservation measures for the European mink. Resting animals were located once a day and active animals were tracked by continuous monitoring. European mink had linear home ranges whose sizes were larger than predicted by Johnson’s model (mean±SD=2971±1888 ha in males and 257±113 ha in females). They travelled long inter-day distances (1.4±1.9 km in males and 0.4±0.6 km in females). Polecats had home ranges of various shapes (linear, circular or combined) and their sizes were consistent with Johnson’s model (707±779 ha in males and 51±58 ha in females). They also had shorter inter-day distances than mink (0.7±0.9 km in males and 0.2±0.4 km in females). However male polecats had longer activity bouts than male mink. Thus European mink exhibit large movements between small and distant activity areas while polecats compensate for their smaller range by a higher activity in restricted areas. The behaviour of the European mink appears to be an adaptation to habitats scattered over linear ranges. This extensive use of space suggests that conservation of this endangered species cannot be achieved in the confinement of Europe’s natural reserves. Conservation plans should aim at maintaining high quality habitats along entire river networks and ensuring safe movements for the animals, preventing particularly the risk of collisions with vehicles.  相似文献

Two models of the distribution of relative elemental rates ofelongation (RELEL) were tested for the extension zone (EZ) ofthe first foliage leaf of seedling wheat plants, by comparisonto patterns of separation of rings and gyres in the walls ofprotoxylem vessels. One model, containing a defined growth maximumin the basal half of the EZ, is favoured in the literature andwas derived from data published for perennial ryegrass. Theother, containing a flat, broad maximum throughout the regionof the EZ with stomates, was constructed from regressions ofinterstomatal distance against distance along the EZ in thefirst foliage leaf of wheat seedlings. The test strongly favouredthe model with the flat maximum. Although the gibberellic acid(GA) insensitivity alleles Rht1 and Rht2 reduce length of extensionzone (LEZ), leaf extension rate (LER) and final cell and leaflengths, they had no effect on maximum RELEL. Results with aninhibitor of GA synthesis indicated that control of leaf elongationby the control of LEZ may be generalizable as a mechanism bywhich GA controls LER in the grass leaf. To study the possible role of disease in the decline of endangered European mink (Mustela lutreola), we conducted a survey of antibody prevalence and renal carriage of pathogenic leptospira (Leptospira interrogans sensu lato) using serum and kidney samples collected from 1990 to 2007 from several free-ranging small carnivores and farmed American mink (Mustela vison) in southwestern France. An indirect microscopic agglutination test using a panel of 16 serovars belonging to 6 serogroups (Australis, Autumnalis, Icteroh?morrhagi?, Grippotyphosa, Panama, Sejroe) revealed antibodies in all species, with significant differences in antibody prevalences: 74% in European mink (n=99), 65.4% in European polecats (Mustela putorius, n=133), 86% in American mink (n=74), 89% in stone martens (Martes foina, n=19), 74% in pine martens (Martes martes, n=19), 35% in common genets (Genetta genetta, n=79), and 31% in farmed American mink (n=51). Serogroups Australis and Icteroh?morragi? were dominant in most free-ranging species; serogroup Grippotyphosa had high prevalences in European mink. Such high antibody prevalences have never been reported. They are probably related to the large number of known reservoirs, rats (Rattus spp.), muskrat (Ondatra zibethicus), and coypu (Myocastor coypu), in the study area. The polymerase chain reaction test specific for pathogenic leptospiral DNA detected renal carriage in 23% of 34 European mink, 22% of 18 polecats, and 15% of 33 free-ranging American mink, with no significant differences. Renal carriage shows that mustelids may shed leptospira for short periods, but their epidemiologic role is probably limited. High antibody prevalences suggest that the disease is unlikely to be highly pathogenic for these species; however, chronic forms of the disease (abortions, renal lesions) could reduce the reproductive success or life span of infected animals. Further studies on the pathogenicity of leptospirosis in these populations are needed to measure its impact on the population dynamics of these rodent predators.  相似文献

ABSTRACT. The spatial distributions of five zoochlorellae-bearing ciliate species (Euplotes daidaleos, Frontonia vernalis, Acaryophrya sp., Disematostoma bütschlii, and Stokesia vernalis) were investigated in a productive freshwater pond. The vertical profiles of all species were compared to the levels of O2, CO2, light, temperature, dissolved inorganic nitrogen, and the availability of particulate food sources. In the stratified water column, Stokesia remained close to the water surface: all other species reached peak abundance close to the oxic-anoxic boundary. The latter behavior probably accommodated the ciliates’requirements for aerobic respiration and particulate food and their dependance on the essential resources of light and dissolved nutrients, which came from opposite directions. With the collapse of stratification, Euplotes and Frontonia returned to the sediment where they reverted to a heterotrophic nutrition although they retained some zoochlorellae. Acaryophrya and Disematostoma also became heterotrophic, but they remained evenly distributed in the water column and they lost their zoochlorellae. Stokesia disappeared, presumably because it encysted. There was some evidence for vertical spatial separation of the five species in the water column.  相似文献

The spatial and seasonal distribution of Paramecium bursaria in two small Indiana ponds was studied using a sampling grid. Very small (5.0 ml) samples were taken so that the individual microhabitats could be studied. The results were evaluated in comparison to the data collected for the P. aurelia complex collected in the same manner and at the same sites. It was found that P. bursaria exist in a clumped distribution, but that the distribution was not very different from random. Paramecium bursaria also exist at the surface and at the mud-water interface. Temperature does not seem to play a statistically significant role in determining population size. The breeding system of P. bursaria is optimized for an outbreeding population of low density. In comparison, the species of the P. aurelia complex exist in a very clumped distribution, are found only at the mud-water interface, and are inbreeders. The evolutionary strategies of the two types of paramecia are discussed.  相似文献

Background

Knowledge of prevalence rates and distribution of human papillomavirus (HPV) genotypes prior high HPV vaccine coverage is necessary to assess its expected impact on HPV ecology and on cervical lesions and cancers.

Methods

Residual specimens of cervical cytology (N = 6,538) were obtained from 16 sites participating in organised cervical cancer screening pilot programs throughout France, anonymised and tested for HPV DNA using the PapilloCheck® genotyping test. Samples were stratified according to age of women and cytological grades.

Results

The age-standardised prevalence rates of HPV 16 and/or 18 (with or without other high-risk types) was 47.2% (95% Confidence Interval, CI: 42.4–52.1) in high-grade squamous intraepithelial lesions (HSILs), 20.2% in low-grade SIL (95% CI: 16.7–23.7) and 3.9% (95% CI: 2.8–5.1) in normal cytology. Overall HR HPV were detected in 13.7% (95%I CI: 11.7–15.6) of normal cytology. In women below 30 years of age, 64% of HSILs were associated with HPV16 and/or 18. In our study population, HPV16 was the most commonly detected type in all cervical grades with prevalence rates ranking from 3.0% in normal cytology to 50.9% in HSILs. HPV16 was also detected in 54% (27/50) of invasive cervical cancers including 5 adenocarcinomas.

Conclusion

HPV16 was strongly associated with cervical precancer and cancer. The high prevalence rates of HPV16/18 infection among women below 30 years of age with HSILs suggests that the impact of vaccination would be primarily observed among young women.  相似文献

Fungi are prominent drivers of ecological processes in soils, so that fungal communities across different soil ecosystems have been well investigated. However, for arable soils taxonomically resolved fine-scale studies including vertical itemization of fungal communities are still missing. Here, we combined a cloning/Sanger sequencing approach of the ITS/LSU region as marker for general fungi and of the partial SSU region for arbuscular mycorrhizal fungi (AMF) to characterize the microbiome in different maize soil habitats. Four compartments were analyzed over two annual cycles 2009 and 2010: a) ploughed soil in 0–10 cm, b) rooted soil in 40–50 cm, c) root-free soil in 60–70 cm soil depth and d) maize roots. Ascomycota was the most dominant phylum across all compartments. Fungal communities including yeasts and AMF differed strongly between compartments. Inter alia, Tetracladium, the overall largest MOTU (molecular operational taxonomic unit), occurred in all compartments, whereas Trichosporon dominated all soil compartments. Sequences belonging to unclassified Helotiales were forming the most abundant MOTUs exclusively present in roots. This study gives new insights on spatial distribution of fungi and helps to link fungal communities to specific ecological properties such as varying resources, which characterize particular niches of the heterogeneous soil environment.  相似文献

Interactions between humans and carnivores have existed for centuries due to competition for food and space. American black bears are increasing in abundance and populations are expanding geographically in many portions of its range, including areas that are also increasing in human density, often resulting in associated increases in human-bear conflict (hereafter, bear incidents). We used public reports of bear incidents in Michigan, USA, from 2003–2011 to assess the relative contributions of ecological and anthropogenic variables in explaining the spatial distribution of bear incidents and estimated the potential risk of bear incidents. We used weighted Normalized Difference Vegetation Index mean as an index of primary productivity, region (i.e., Upper Peninsula or Lower Peninsula), primary and secondary road densities, and percentage land cover type within 6.5-km2 circular buffers around bear incidents and random points. We developed 22 a priori models and used generalized linear models and Akaike’s Information Criterion (AIC) to rank models. The global model was the best compromise between model complexity and model fit (w = 0.99), with a ΔAIC 8.99 units from the second best performing model. We found that as deciduous forest cover increased, the probability of bear incident occurrence increased. Among the measured anthropogenic variables, cultivated crops and primary roads were the most important in our AIC-best model and were both positively related to the probability of bear incident occurrence. The spatial distribution of relative bear incident risk varied markedly throughout Michigan. Forest cover fragmented with agriculture and other anthropogenic activities presents an environment that likely facilitates bear incidents. Our map can help wildlife managers identify areas of bear incident occurrence, which in turn can be used to help develop strategies aimed at reducing incidents. Researchers and wildlife managers can use similar mapping techniques to assess locations of specific conflict types or to address human impacts on endangered species.  相似文献

The frequently observed positive relationship between fish population abundance and spatial distribution suggests that changes in distribution can be indicative of trends in abundance. If contractions in spatial distribution precede declines in spawning stock biomass (SSB), spatial distribution reference points could complement the SSB reference points that are commonly used in marine conservation biology and fisheries management. When relevant spatial distribution information is integrated into fisheries management and recovery plans, risks and uncertainties associated with a plan based solely on the SSB criterion would be reduced. To assess the added value of spatial distribution data, we examine the relationship between SSB and four metrics of spatial distribution intended to reflect changes in population range, concentration, and density for 10 demersal populations (9 species) inhabiting the Scotian Shelf, Northwest Atlantic. Our primary purpose is to assess their potential to serve as indices of SSB, using fisheries independent survey data. We find that metrics of density offer the best correlate of spawner biomass. A decline in the frequency of encountering high density areas is associated with, and in a few cases preceded by, rapid declines in SSB in 6 of 10 populations. Density-based indices have considerable potential to serve both as an indicator of SSB and as spatially based reference points in fisheries management.  相似文献

This study compares fossil femora attributed to extinct African bunodont lutrines with extant mustelids and ursids to reconstruct locomotor behavior. Due to the immense size differences among taxa, shape data were used to compare morphology. Based on morphological differences, the fossil femora are suggested to belong to different taxa with different locomotor abilities and habitat preferences. The Langebaanweg femur is the oldest and has a typical mustelid morphology suggesting that it was a locomotor generalist like most mustelids. The West Turkana form is more like extant nonbunodont otters, but much larger, and may have belonged to a semiaquatic taxon. The enormous Omo femur shares some features with truly aquatic taxa (e.g., Enhydra) and is the most likely to have been fully aquatic. The same may hold true for the Hadar species as it is most similar to that from the Omo. If these femora truly belong to bunodont lutrines, then they are more diverse in postcranial morphology than in dental morphology.  相似文献

Wastelands are likely to host a significant part of urban floristic diversity but have received limited attention because they are not considered interesting green zones. Here, we explore the potential role of wastelands in maintaining urban biodiversity to help define effective urban management plans. We quantified floristic diversity in 98 wasteland sites of Hauts-de-Seine, one of the most densely populated areas in France, and characterized the environmental parameters and spatial distribution of sites to identify some of the factors that influence plant species composition and to explore the impact of urban environment on the floristic interest of wastelands. Their floristic richness represented 58% of the total richness observed in the whole study area. Site richness depended on site area (the largest sites were the richest) and site age, with a maximum in sites of intermediate age (4–13 years). In the largest sites only (>2,500 m2), the floristic distance among sites was positively correlated with geographic distance, which suggests that migration of species among large sites partly controls local floristic composition. In contrast, the environmental distance among sites was not correlated with floristic distance. Finally, we showed that the presence of collective and individual dwellings within 200 m of a wasteland decreased its floristic rarity, whereas the presence of rivers or ponds increased it. We derive several recommendations to optimize the management of wastelands with respect to conservation of urban biodiversity.  相似文献

The heavy metal hyperaccumulator Thlaspi caerulescens is widespread in France on many kinds of sites and substrates, including Zn/Pb/Cd mine and smelter wastes, Ni-rich serpentine outcrops and a variety of nonmetalliferous soils. Thlaspi caerulescens is remarkable among the metallophytes of France because it accumulates Zn to high concentrations (almost always >0.1%, and often >1% in the dry matter) regardless of the total Zn concentration of the substrate. The extraordinary uptake of Zn from soils of normal Zn concentration draws attention to the need for studies of the mechanisms by which such mobilization and uptake can occur. Different populations of Thlaspi caerulescens in France show considerable variation in their ability to accumulate Cd; individuals in some populations contain as much as 0.1 to 0.4% Cd, the highest levels recorded in vascular plants. The hyperaccumulation of Ni (sometimes exceeding 1%) from serpentine soils in France is also noteworthy. Despite the generally low biomass, some very large individuals occur, giving good potential for selective breeding to improve the value of Thlaspi caerulescens for phytoremediation, especially of Cd. The high Zn uptake from all kinds of soils is a property shared by the related T. brachypetalum, and T. alpinum shows dual Zn- and Ni uptake, depending on the substrate. The extent to which other species of Thlaspi occurring in France exhibit metal accumulation is also discussed.  相似文献

Confined spatial patterns of microbial distribution are prevalent in nature, such as in microbial mats, soil communities, and water stream biofilms. The symbiotic two-species consortium of Pseudomonas putida and Acinetobacter sp. strain C6, originally isolated from a creosote-polluted aquifer, has evolved a distinct spatial organization in the laboratory that is characterized by an increased fitness and productivity. In this consortium, P. putida is reliant on microcolonies formed by Acinetobacter sp. C6, to which it attaches. Here we describe the processes that lead to the microcolony pattern by Acinetobacter sp. C6. Ecological spatial pattern analyses revealed that the microcolonies were not entirely randomly distributed and instead were arranged in a uniform pattern. Detailed time-lapse confocal microscopy at the single-cell level demonstrated that the spatial pattern was the result of an intriguing self-organization: small multicellular clusters moved along the surface to fuse with one another to form microcolonies. This active distribution capability was dependent on environmental factors (carbon source and oxygen) and historical contingency (formation of phenotypic variants). The findings of this study are discussed in the context of species distribution patterns observed in macroecology, and we summarize observations about the processes involved in coadaptation between P. putida and Acinetobacter sp. C6. Our results contribute to an understanding of spatial species distribution patterns as they are observed in nature, as well as the ecology of engineered communities that have the potential for enhanced and sustainable bioprocessing capacity.  相似文献

Gastropod assemblages from nearshore rocky habitats were studied over large spatial scales to (1) describe broad-scale patterns in assemblage composition, including patterns by feeding modes, (2) identify latitudinal pattern of biodiversity, i.e., richness and abundance of gastropods and/or regional hotspots, and (3) identify potential environmental and anthropogenic drivers of these assemblages. Gastropods were sampled from 45 sites distributed within 12 Large Marine Ecosystem regions (LME) following the NaGISA (Natural Geography in Shore Areas) standard protocol (www.nagisa.coml.org). A total of 393 gastropod taxa from 87 families were collected. Eight of these families (9.2%) appeared in four or more different LMEs. Among these, the Littorinidae was the most widely distributed (8 LMEs) followed by the Trochidae and the Columbellidae (6 LMEs). In all regions, assemblages were dominated by few species, the most diverse and abundant of which were herbivores. No latitudinal gradients were evident in relation to species richness or densities among sampling sites. Highest diversity was found in the Mediterranean and in the Gulf of Alaska, while highest densities were found at different latitudes and represented by few species within one genus (e.g. Afrolittorina in the Agulhas Current, Littorina in the Scotian Shelf, and Lacuna in the Gulf of Alaska). No significant correlation was found between species composition and environmental variables (r≤0.355, p>0.05). Contributing variables to this low correlation included invasive species, inorganic pollution, SST anomalies, and chlorophyll-a anomalies. Despite data limitations in this study which restrict conclusions in a global context, this work represents the first effort to sample gastropod biodiversity on rocky shores using a standardized protocol across a wide scale. Our results will generate more work to build global databases allowing for large-scale diversity comparisons of rocky intertidal assemblages.  相似文献
