The bone marrow mesenchymal stem cells (MSCs) are multipotent stem cells which can differentiate into mesenchymal cells in vitro. In this study, MSCs in duck were isolated from bone marrow by density gradient centrifuge separation, purified and expanded in the me- dium. The primary MSCs were expanded for 11 passages. The different-passage MSCs were induced to differentiate into osteoblasts and neuron-like cells. Karyotype analysis indicated that MSCs kept diploid condition and the hereditary feature was stable. The different- passage MSCs expressed CD44, ICAM-1 and SSEA-4, but not CD34, CD45 and SSEA-1 when detected by immunofluorescence staining There was no significant difference among the positive rates of passages 2, 6 and 8 (P 〉 0.05), but a significant difference existed among those of passages 2, 6, 8 and 11 (P 〈 0.05). After the osteogenic inducement was added, the induced different-passage MSCs expressed high-level alkaline phosphatase (ALP), and are positive for tetracycline staining, Alizarin Red staining and Von Kossa staining. After the neural inducement was added, about 70% cells exhibited typical neuron-like phenotype, the induced different-passage MSCs expressed Nestin, neuron-specific enolase (NSE) and glial fibrillary acidic protein (GFAP) when detected by immunofluorescence staining. There was no significant difference among the positive rates of passages 3, 4 and 6 (P〉0.05), but a significant difference existed among those of passages 3, 4, 6 and 8 (P〈0.05). These results suggest that MSCs in duck were capable of differentiating into osteoblasts and neuron-like cells in vitro.  相似文献

Duck plague (Duck virus enteritis) was disgnosed in a resident population of Muscovy ducks (Cairina moschata) on a small game farm in Alberta. This disease has not been reported previously in Canada. Clinical signs consisted of cyanosis, depression and acute death. Necropsy of two Muscovy ducks revealed lesions typical of the disease. There were ulcerations with pseudomembranes in the small intestine, ulcerations with caseous plaques in the esophagus and eosinophilic intranuclear inclusion bodies in the spleen. Clinical disease with mortality was reproduced in young ducklings injected with tissue homogenates from field cases. All surviving inoculated ducklings seroconverted to highly significant titres of neutralizing antibodies to duck virus enteritis (DVE) virus. All attempts to isolate the agent in embryonating duck eggs or primary tissues cultures of duck and chicken kidney were negative. Identification of the DVE virus was accomplished by serum neutralization with ducklings as the host system.  相似文献

The possibility of using RAPD markers for the detection of differences between lines of Peking duck constituting a local population maintained at the Blagovarskii State Farm for Pedigree Poultry was demonstrated. Genetic distances based on the RAPD markers precisely and accurately reflect even small changes that occurred in the genetic structure of Peking duck lines during breeding of parental forms. The pattern of inheritance of RAPD markers obtained using primer HM13 was studied in the F1 progeny of two families. The results can be used for improvement of available high-productivity lines of ducks breeding of new lines, and promotion of the combining abilities of lines.  相似文献

As an approach to identifying hepatocyte receptors for the avian hepadnavirus duck hepatitis B virus (DHBV), hybridomas were prepared from mice immunized with permissive duck hepatocytes. Monoclonal antibodies (MAbs) were screened for the ability to inhibit binding of DHBV particles to primary duck hepatocytes and to block infection. We identified two MAbs which partially blocked binding and caused marked inhibition of infection of primary duck hepatocytes with DHBV. Lack of cross-reactivity with DHBV envelope proteins suggested that inhibition of infection was due to specific interaction between the antibodies and a host cell surface molecule. Both MAbs immunoprecipitated a 55-kDa protein (p55) expressed in duck liver and several other duck tissues. p55 homologs were also identified in other birds and mammals. We predict from our data that only a small proportion of total cellular p55 molecules are expressed at the surfaces of hepatocytes and that p55 is involved in some early step in the infectious pathway.  相似文献

Only one metallothionein appears in domesticated duck upon zinc induction. The complete amino acid sequence has been elucidated. This metallothionein has the same sequence as the chicken metallothionein, as determined by chemical sequencing of overlapping peptides produced by selective proteinase digestion and confirmed by mass spectrometry. Clinical and gross anatomy features of myopathic ducks are reported. Light microscopic examination of the leg muscles showed necrosis, degeneration and inflammation, and some degree of regeneration was also observed. There was infiltration and replacement of muscle fibres with fibrous tissue. Electron microscopy showed the early stages of myofilament dissolution and accumulation of glycogen in the presynaptic terminal of the neuromuscular junction.  相似文献

The persistence of 3 low-pathogenicity avian influenza viruses (LPAIV) (H4N6, H5N1, and H6N8) and one human influenza virus (H1N1) as well as Newcastle disease virus (NDV) and enteric cytopathogenic bovine orphan (ECBO) virus was investigated in lake sediment, duck feces, and duck meat at 30, 20, 10, and 0°C using a germ carrier technique. Virus-loaded germ carriers were incubated in each substrate, and residual infectivity of the eluted virus was quantified on cell culture after regular intervals for a maximum of 24 weeks. Data were analyzed by a linear regression model to calculate T(90) values (time required for 90% loss of virus infectivity) and estimated persistence of the viruses. In general, the persistence of all of the viruses was highest in lake sediment, followed by feces, and was the lowest in duck meat at all temperatures. For the avian influenza virus subtypes, T(90) values in sediment ranged from 5 to 11, 13 to 18, 43 to 54, and 66 to 394 days at 30, 20, 10, and 0°C, respectively, which were 2 to 5 times higher than the T(90) values of the viruses in the feces and meat. Although the individual viruses vary in tenacity, the survival time of influenza viruses was shorter than that of NDV and ECBO virus in all substrates. The results of this study suggest that lake sediment may act as a long-term source of influenza viruses in the aquatic habitat, while the viruses may remain infectious for extended periods of time in duck feces and meat at low temperatures, allowing persistence of the viruses in the environment over winter.  相似文献

Duck hepatitis B virus (DHBV) obtained from the serum of congenitally infected ducks was used to infect primary duck hepatocyte cultures 1 to 4 days after plating. Virus replication was demonstrated by the appearance, beginning at 2 days after infection, of intracellular covalently closed-circular and single-stranded DHBV DNA replicative intermediates which were not present in the inoculating virus preparation. With increasing time after infection there was further amplification of intracellular relaxed circular, covalently closed-circular, and single-stranded DHBV DNA. Cultures of primary duck hepatocytes are competent for infection with DHBV only during the first 4 days of culture. Synthesis of DHBV core antigen and DHBV surface antigen was detected by immunofluorescence in 10% of the hepatocytes in culture. De novo synthesis and release of infectious virus was also demonstrated. Therefore, all stages of viral replication were carried out by these experimentally infected primary hepatocyte cultures. This system makes it possible to study DHBV replication in vitro.  相似文献

选用1600羽出壳1 d的樱桃谷鸭随机分为8组,分别饲喂不同配比和组合的抗生素、沸石、微生态制剂(EM)、中草药和甘氨酸微量元素螯合物等添加剂饲料,探讨了不同饲料添加剂对肉鸭生长性能的影响,并对鸭舍的有害气体含量进行了研究。结果表明:饲料中添加沸石、EM、中草药对提高肉鸭的日增重和降低料肉比有显著效果(P<0.05)。在1~14 d阶段,以混合添加沸石和EM对肉鸭体重、采食量和日增重的效果为最好(P<0.05);添加抗生素、混合添加抗生素与沸石均能明显(P<0.05)降低1~14 d樱桃谷鸭料肉比。在15~35 d阶段,添加沸石、EM、中草药均能提高肉鸭的饲料利用率,以混合添加沸石与中草药及甘氨酸微量元素螯合物效果最好,可显著(P<0.05)提高肉鸭体质量、采食量和日增重,降低料肉比。添加沸石、EM、中草药均能降低鸭舍内NH3、H2S和CO2的浓度,尤以添加沸石、甘氨酸微量元素螯合物、沸石与中草药混合物分别对降低NH3、H2S、CO2浓度效果最好。说明沸石、中草药和EM可有效提高肉鸭生长性能,降低鸭舍内有害气体浓度。  相似文献

The major mode of natural infection of duck hepatitis B virus (DHBV) in Pekin ducks is vertical transmission, with 95 to 100% of the embryos from DHBV-infected dams eventually becoming infected. Maternally transmitted virus is present in large quantities in the yolk of unincubated eggs and is taken up by the embryo during early development. Synthesis of DHBV DNA in the embryo begins at about 6 days of incubation and coincides with the formation of the liver. DHBV DNA synthesis is incomplete, however, until 8 to 10 days of incubation, as shown by comparing the electrophoretic patterns of DHBV-specific nucleic acid species from embryonic livers at successive stages of development. From 8 days of incubation and continuing throughout embryonic development, subviral particles, which resemble viral replication intermediates isolated from infected livers of post-hatch ducklings, appear in the circulation. These particles contain a polymerase activity that utilizes an RNA template to synthesize viral DNA. Our results suggest that certain host functions, which appear during embryonic development, may be required for DHBV replication and assembly. It is possible that in mammals a similar developmental process occurs. The failure to find human hepatitis B virus in the circulation of most babies, born to hepatitis B virus carrier women, in the first few weeks after birth may reflect such a process.  相似文献

The role of harvest in the dynamics of waterfowl populations continues to be debated among scientists and managers. Our perception is that interested members of the public and some managers believe that harvest influences North American duck populations based on calls for more conservative harvest regulations. A recent review of harvest and population dynamics of North American mallard (Anas platyrhynchos) populations (Pöysä et al. 2004) reached similar conclusions. Because of the importance of this issue, we reviewed the evidence for an impact of harvest on duck populations. Our understanding of the effects of harvest is limited because harvest effects are typically confounded with those of population density; regulations are typically most liberal when populations are greatest. This problem also exists in the current Adaptive Harvest Management Program (Conn and Kendall 2004). Consequently, even where harvest appears additive to other mortality, this may be an artifact of ignoring effects of population density. Overall, we found no compelling evidence for strong additive effects of harvest on survival in duck populations that could not be explained by other factors. © 2012 The Wildlife Society.  相似文献
