Genetic diversity and population differentiation of the giant honey bee (Apis dorsata) in Thailand were examined. Six PCR-RFLP mitotypes were generated from digestion of the COI-COII, Cytb-tRNAser, ATPase6-8, and lrRNA genes with Dra I and Hin fI. Low genetic diversity (h=0.074, π=0.032%) and a lack of genetic population differentiation between A. dorsata originating from geographically different regions were observed from mtDNA polymorphisms (P > 0.05). In contrast, microsatellite (A14, A24, and A88) polymorphisms revealed a relatively high level of genetic diversity in A. dorsata (H o=0.68–0.74, average number of alleles per locus=6.0–9.0). Both A24 and A88 indicated significant population differentiation between bees from the north-to-central region (north, northeast, and central regions), peninsular Thailand, and Samui Island.  相似文献

Concordance between the mitochondrial haplotypes of the Eastern honey bee, Apis cerana, and its ectoparasitic Varroa mites across the Isthmus of Kra in Thailand has suggested that local host–pathogen co-evolution may be responsible for the geographic distribution of particular genotypes. To investigate nuclear microsatellites population structure in A. cerana, single workers of A. cerana colonies from Thailand were genotyped at 18 microsatellite loci. The loci showed intermediate to high levels of heterozygosity and a range of allele numbers. The analyses confirmed a fundamental subdivision of the Thai A. cerana population into the “Asia Mainland” and “Sundaland” regions at the Isthmus of Kra. However, the nuclear microsatellite differentiation was less distinct than mtDNA haplotype differences, suggesting male-biased dispersal and population admixture. Overall, samples showed a weak isolation-by-distance effect. The isolated population on Samui island was most differentiated from the other samples. The results do not support our initial hypothesis of local host–pathogen co-evolution, which predicts a strict correspondence between the nuclear genome and the lineage of parasitic Varroa mite of the A. cerana samples, because the gene flow indicated by our nuclear microsatellite markers should also mix potential Varroa resistance alleles among subpopulations. Instead, our study suggests that the coincidental distribution of Varroa lineages and A. cerana population structure in Thailand are the result of biogeographic history and current migration patterns.  相似文献

The biogeography and intraspecific variability of the eastern cavity‐nesting honeybee, Apis cerana, are not very well known. We studied the variability of this species in China using morphometrical methods together with restriction and sequence analysis of two different regions of mitochondrial DNA. Samples of A. cerana were collected from feral or traditionally managed colonies in 19 locations of the Chinese mainland, covering the main ecological regions. Worker bees from each sample were dissected and morphometric characters were measured. The data were analysed with multivariate statistical procedures. Data were supplemented by previously published Chinese samples from the Oberursel data bank and reference samples of A. cerana from adjacent countries. A mitochondrial DNA fragment containing a non‐coding region was amplified and analysed with the restriction enzyme DraI. This fragment was sequenced for two samples. For a subset of samples, the subunit 2 of the mitochondrial NADH gene was amplified and sequenced. Morphometric analysis revealed a high degree of variation, strongly associated with ecological zones and correlated with geographical and climatic parameters. Two main clusters were apparent, one comprised the bees from the southern tropical seasonal rain forest region, showing strong associations to the bees of Vietnam, Thailand and Myanmar. The second main cluster included the bees from the temperate deciduous broad‐leaved forest region, the subtropical evergreen broad‐leaved forest zone, the high, cold meadow and steppe region and the North, and showed increasing similarity to the bees of Korea and Japan. In particular, the bees from Qingzang plateau, on the fringe of the Ganshu province, were set apart by their exceptional body size, darkness and pilosity. There was no variation in DraI restriction patterns within China. Sequence variation of the mitochondrial ND2 region was consistent with geographic patterns of morphological variation. Bees of the South Yunnan region were set apart by characteristically broad abdominal sterna and wax mirrors, with this locally restricted trait transcending the main transition line at the northern limit of the tropical seasonal rain forest region. Varroa jacobsoni was first described as a natural ectoparasitic mite of the Eastern honeybee (Apis cerana) throughout Asia. It later switched host to the Western honeybee (A. mellifera) and has now become a serious pest of that bee worldwide. The studies reported here on genotypic, phenotypic and reproductive variation among V. jacobsoni infesting A. cerana throughout Asia demonstrate that V. jacobsoni is a complex of at least two different species. In a new classification V. jacobsoni is here redefined as encompassing nine haplotypes (mites with distinct mtDNA CO-I gene sequences) that infest A. cerana in the Malaysia–Indonesia region. Included is a Java haplotype, specimens of which were used to first describe V. jacobsoni at the beginning of this century. A new name, V. destructor n. sp., is given to six haplotypes that infest A. cerana on mainland Asia. Adult females of V. destructor are significantly larger and less spherical in shape than females of V. jacobsoni and they are also reproductively isolated from females of V. jacobsoni. The taxonomic positions of a further three unique haplotypes that infest A. cerana in the Philippines is uncertain and requires further study.Other studies reported here also show that only two of the 18 different haplotypes concealed within the complex of mites infesting A. cerana have become pests of A. mellifera worldwide. Both belong to V. destructor, and they are not V. jacobsoni. The most common is a Korea haplotype, so-called because it was also found parasitizing A. cerana in South Korea. It was identified on A. mellifera in Europe, the Middle East, Africa, Asia, and the Americas. Less common is a Japan/Thailand haplotype, so-called because it was also found parasitizing A. cerana in Japan and Thailand. It was identified on A. mellifera in Japan, Thailand and the Americas.Our results imply that the findings of past research on V. jacobsoni are applicable mostly to V. destructor. Our results will also influence quarantine protocols for bee mites, and may present new strategies for mite control.  相似文献

In south India there are two distinct colour morphs of cavity nesting honey bees: the yellow ‘Plain’ morph and the black ‘Hill’ morph which are collectively known as Apis cerana. We show that the Hill morph is associated with a widely distributed mitochondrial haplotype that is present throughout mainland populations of south east Asian A. cerana. In contrast, the Plain morph, which is apparently confined to low to moderate elevations in India and Sri Lanka, is associated with a unique mitochondrial haplotype that is not present in other cavity nesting honey bees. We further show that in a region of sympatry (Bangalore, Karnataka State) the drone mating flight times of the two colour morphs barely overlap. Combined, drone flight data and the complete separation of mitochondrial haplotypes suggest that the two morphs are reproductively isolated. The Plain morph is distinguished from the Hill morph by the first three abdominal tergites of the worker, which are completely yellow in the Plain morph, whereas in the Hill morph they are black or black with yellow patches. Although the two morphs are generally distinguishable in the field by overall colouration, microscopic examination of the first 3 abdominal tergites is preferred. Numerous shells of bivalves, which study attributed to 37 species, were collected from samples of ground taken with a Van Veen dredge sampler at the northeast coast of Sakhalin Island at depths of about 10–60 m in June–October 2003–2009. Tellina lutea, Mactromeris polynyma, Serripes groenlandicus, Macoma spp., Liocyma fluctuosum, and Siliqua alta occurred most often in the samples. The assessment of growth in 18 bivalve species revealed common patterns of age variations that were irrespective of their way of life, maximum body size, and maximum age. The greatest annual increment in shell length was observed in the first 4 years of life, and the asymptotic size of the mollusks was reached with a 10-fold decrease in the annual growth rate. The life span of the mollusks varied from 2 up to 41 years and sometimes significantly differed from that of the studied species in other areas of their range, clearly displaying the ecological conditionality of this parameter. Species with maximum ages of less than 10 years were the most numerous in the bivalve community of the region. The results of the research are discussed in relation to the nature of the environment at the northeast coast of Sakhalin Island.  相似文献

The small dwarf honey bee, Apis andreniformis, is a rare and patchily distributed Apis spp. and is one of the native Thai honey bees, yet little is known about its biodiversity. Thirty (27 Thai and 3 Malaysian) and 37 (32 Thai and 5 Malaysian) colonies of A. andreniformis were sampled for morphometric and genetic analysis, respectively. For morphometric analysis, 20 informative characters were used to determine the variation. After plotting the factor scores, A. andreniformis from across Thailand were found to belong to one group, a notion further supported by a cluster analysis generated dendrogram. However, clinal pattems in groups of bee morphometric characters were revealed by linear regression analysis. The body size of bees increases from South to North but decreases from West to East, although this may reflect altitude rather than longitude. Genetic variation was determined by sequence analysis of a 520 bp fragment of the mitochondrial cytochrome oxidase subunit b (cytb). DNA polymorphism among bees from the mainland of Thailand is lower than that from Phuket Island and Chiang Mal. Although two main different groups of bees were obtained from phylogenetic trees constructed by neighbor-joining and unweighted pair-group method using arithmetic averages programs, no clear geographic signal was present. Thus, while the minor group (B) contained all of the samples from the only island sampled (Phuket in the south), but not the southem mainland colonies, it also contained samples from the far northem inland region of Chiang Mai, other samples of which were firmly rooted in the major group (A).  相似文献

Reproduction ofVarroa jacobsoni Oudemans (Acari: Varroidae) and the number ofVarroa mites that were found dead on the bottom board of the hive, were studied in relation to the period the mites spent on adult honey bees,Apis mellifera L. (Hymenoptera: Apidae), prior to invasion into brood cells. The maximum period on adult bees was 23 days. To introduce mites, combs with emerging worker brood, heavily infested with mites, were placed into a colony and removed the next day. At the beginning of the first day following emergence from brood cells, 18% of the mites introduced into the colony was found on the bottom of the hive. Part of these mites may already have died inside the capped brood cells, and then fallen down after cleaning of cells by the bees. At the second and third day following emergence, respectively 4% and 2% of the mites on adult bees at the previous day was recovered on the bottom, whereas from the fourth day on only 0.6% of the mites on adult bees was recovered on the bottom per day. After invasion into brood cells, 8–12% of the mites did not produce any offspring. Of the mites that did reproduce, the total number of offspring was 4.0–4.4 per mite during one reproductive cycle, part of which may reach maturity resulting in 1.2–1.3 viable daughters, and 8–10% of the mites produced only male offspring. Reproduction was independent of the period the mites had spent on adult bees prior to invasion into brood cells.  相似文献

Yolk-sac larvae of Solenostomus paradoxus are described from 19 specimens (2.8–5.1 mm in body length) discharged from the brood pouch of a single female collected by hand at Libong Island, Trang, southern Thailand, on 30 January 2002. Although the larvae of S. paradoxus were closely similar to those of S. cyanopterus in general appearance and pigmentation patterns, the former differed in lacking clusters of melanophores on the edge of the dorsal finfold and dark oblique streaks on the dorsal and ventral finfolds near the tail tip.  相似文献

为了解澜沧江流域北部中华蜜蜂(Apis cerana cerana)的分布,探究中华蜜蜂的食源和营养生态位沿海拔梯度的变化特征,调查了澜沧江流域北部各海拔区域中华蜜蜂的种群分布,运用蜂蜜孢粉学(melissopalynology)分析了各海拔区域中华蜜蜂蜂蜜中花粉的组成特征和变化规律,并综合分析了海拔、中华蜜蜂营养生态位和蜂蜜中花粉的相关性,探讨了自然环境与中华蜜蜂的分布,海拔梯度与蜜粉源植物、中华蜜蜂的食源和营养生态位的关系。结果表明:澜沧江流域北部中华蜜蜂的食源种类丰富,中华蜜蜂种群主要分布于2200—2800 m海拔区域。不同海拔区域中华蜜蜂采食花粉的种类和数量不同,中华蜜蜂种群分布多的海拔区域,蜂蜜中花粉的种类较多,但花粉数量相对少。随着海拔梯度升高,中华蜜蜂蜂蜜中花粉的数量表现为先降后增,而花粉的种类则表现为先增后降。不同海拔区域的中华蜜蜂营养生态位存在差异,推测各海拔区域蜜粉源植物分布、中华蜜蜂种内和种间授粉昆虫及食草动物等竞争因素不同,但各海拔梯度间的变化差异不显著。海拔与中华蜜蜂营养生态位呈正相关(r=0.051),相关性不显著;海拔与蜂蜜中花粉数量呈正相关(r=0.047),与蜂蜜中花粉种类呈正相关(r=0.144),相关性都不显著;中华蜜蜂营养生态位与蜂蜜中花粉的种类呈负相关(r=-0.305),相关性显著(P!0.05);与花粉的数量呈负相关(r=-0.064),相关性不显著。蜂蜜中花粉的数量与种类呈正相关(r=-0.303),且相关性显著(P!0.05)。  相似文献

The internal transcribed spacer (ITS) regions of 18–26S nuclear ribosomal DNA was sequenced to address phylogenetic relationships and to measure the extent of differentiation among six species of the Far EasternIndigofera. ITS 1 had 230–240 base pairs (bp) long while ITS 2 had 211–213 bp long. The 5.8S rRNA coding region was 161 bp long. Sequence divergence calculated by Kimura's two parameter method between species ranged from 0.00 to 13.49%. A single most parsimonious tree was produced from 77 variable nucleotide sites, which had a consistency index of 0.97 and a retention index of 0.83. ITS sequence data suggested that the continental species ofI. kirilowii (2n=16) is diverged from the common ancestor of other species at first, and then the island species ofI. decora (2n=48) andI. venulosa and the Korean Peninsular species ofI. grandiflora (2n=16) andI. koreana (2n=32) are diverged from the ancestor. The molecular data supports that the speciation in the Far EasternIndigofera occurred with polyploidization from continental ancestor to peripheral peninsular and island species.  相似文献

Mitochondrial DNA D-loop (control) region (426-bp) was used to infer the genetic structure of Spanish mackerel (Scomberomorus commerson) from populations in Southeast Asia (Brunei, East and West Malaysia, Philippines, Thailand, Singapore, and China) and northern Australia (including western Timor). An east–west division along Wallace’s Line was strongly supported by a significant AMOVA, with 43% of the total sequence variation partitioned among groups of populations. Phylogenetic and network analyses supported two clades: clade A and clade B. Members of clade A were found in Southeast Asia and northern Australia, but not in locations to the west (Gulf of Thailand) or north (China). Clade B was found exclusively in Southeast Asia. Genetic division along Wallace’s Line suggests that co-management of S. commerson populations for future sustainability may not be necessary between Southeast Asian nations and Australia, however all countries should share the task of management of the species in Southeast Asia equally. More detailed genetic studies of S. commerson populations in the region are warranted.  相似文献

中华蜜蜂为多种植物传粉,具有重要的生态服务价值。为进一步了解中华蜜蜂选择蜜粉源植物的多样性与特征,采用聚合酶链式反应(PCR)、DNA条形码、高通量测序等技术,研究了源自陕西宝鸡、贵州望谟和广西凤山蜂巢蜜中的花粉来源。通过对样品花粉rbcL基因片段扩增及测序、分析,发现各样本间可操作分类单元(OTU)水平上蜜粉源植物Alpha多样性指数无显著差异,同一地点样品不同重复间存在较高的相关性,而不同地区样本相关性较低。三地共鉴定出27目46科71属82种蜜粉源植物,其中宝鸡市、望谟县、凤山县样本中分别鉴定出57种、30种和40种,各地蜜粉源植物的物种组成存在较大差异。这些蜜粉源植物中,菊目植物种类最多,其次为毛茛目,分别有12种和7种。其中草本植物和木本植物(含乔木、灌木和木质藤本等)分别有44种和38种。研究对蜜粉源植物花序特点进行了分析,但并未发现相关性状对中华蜜蜂选择具有的影响。从属的区系成分分析,总体上温带分布区类型的植物属多于热带分布区类型的属,所占比例分别为46.5%和36.6%,但在凤山县样本中出现热带分布区类型多于温带分布区类型的情况。在不同样品中,红足蒿(Artemisia rubripes)、向日葵(Helianthus annuus)、芜青(Brassica rapa)、粗糠柴(Mallotus philippensis)、枇杷(Eriobotrya japonica)等分别占有一定优势,其与中华蜜蜂的关系值得进一步研究。根据蜜粉源植物的系统进化关系,中华蜜蜂访问的蜜粉源植物处于不同进化地位的分支,其中在菊类分支和蔷薇类分支比较集中。研究揭示了中华蜜蜂传粉植物组成特点,为进一步认知中华蜜蜂传粉服务功能的重要性提供依据。  相似文献

The microsporidium Nosema ceranae is detected in honeybees in Thailand for the first time. This endoparasite has recently been reported to infect most Apis mellifera honeybee colonies in Europe, the US, and parts of Asia, and is suspected to have displaced the endemic endoparasite species, Nosema apis, from the western A. mellifera. We collected and identified species of microsporidia from the European honeybee (A. mellifera), the cavity nesting Asian honeybee (Apis cerana), the dwarf Asian honeybee (Apis florea) and the giant Asian honeybee (Apis dorsata) from colonies in Northern Thailand. We used multiplex PCR technique with two pairs of primers to differentiate N. ceranae from N. apis. From 80 A. mellifera samples, 62 (77.5%) were positively identified for the presence of the N. ceranae. Amongst 46 feral colonies of Asian honeybees (A. cerana, A. florea and A. dorsata) examined for Nosema infections, only N. ceranae could be detected. No N. apis was found in our samples. N. ceranae is found to be the only microsporidium infesting honeybees in Thailand. Moreover, we found the frequencies of N. ceranae infection in native bees to be less than that of A. mellifera.  相似文献

Monterey pine (Pinus radiata D. Don) has only five extant native populations: three disjunct populations along the coast of California, USA and two on Mexican islands. All populations have been influenced by human activity, but the island populations in particular have been affected by introduced biota. On Guadalupe Island, the pine population has suffered drastically from overgrazing by introduced goats. We visited both island populations and described their status, took measurements, and made seed collections. We counted approximately 200 mature pine trees and virtually no seedlings on Guadalupe Island: a reduction of approximately half the population in the last 50 years. The trees are all large (mean diameter of 144 cm) –considerably larger than trees from the other four populations – and arguably near the end of their natural lifespan. The population on Cedros Island is much more robust, with thousands of trees. None sampled were as large as those on Guadalupe Island (mean diameter of 20 cm) and many groves were young and even-aged – presumably the consequence of natural regeneration after a recent fire. Tissue samples from trees on both islands did not show evidence of infection from the pitch canker pathogen, Fusarium circinatum, that has caused significant mortality in the three mainland populations. Caution is recommended in any restoration activity for the Guadalupe Island pines. Inbreeding levels could indicate the need for some planting or seeding intervention but there are also risks associated with this. Natural regeneration – after goat removal – is preferred.  相似文献

The trnTtrnF region is located in the large single-copy region of the chloroplast genome. It consists of the trnL intron, a group I intron, and the trnTtrnL and trnLtrnF intergenic spacers. We analyzed the evolution of the region in the three genera of the gymnosperm lineage Gnetales (Gnetum, Welwitschia, and Ephedra), with especially dense sampling in Gnetum for which we sequenced 41 accessions, representing most of the 25–35 species. The trnL intron has a conserved secondary structure and contains elements that are homologous across land plants, while the spacers are so variable in length and composition that homology cannot be found even among the three genera. Palindromic sequences that form hairpin structures were detected in the trnLtrnF spacer, but neither spacer contained promoter elements for the tRNA genes. The absence of promoters, presence of hairpin structures in the trnLtrnF spacer, and high sequence variation in both spacers together suggest that trnT and trnF are independently transcribed. Our model for the expression and processing of the genes tRNAThr(UGU), tRNALeu(UAA), and tRNAPhe (GAA) therefore attributes the seemingly neutral evolution of the two spacers to their escape from functional constraints. In this work, Apis mellifera carnica and A. m. macedonica honey bees from Serbia, Bosnia and Herzegovina and Republic of Macedonia were analysed using molecular techniques in order to improve our knowledge about biogeography of A. mellifera on the Balkan peninsula. This is the first time that the indigenous honey bees from Bosnia and Herzegovina and Republic of Macedonia have been analyzed using a molecular approach. Sampling was carried out from 560 stationary apiaries where bees were kept in traditional hives (woven skeps). The COI–COII regions of 1680 samples were PCR-amplified and sequenced. To reveal the haplotype of studied bees, the obtained sequences were aligned with published sequence data of haplotypes that belong to A. mellifera C phylogenetic lineage. The C2D mtDNA haplotype was found in all honey bees sampled from Serbia, Bosnia and Herzegovina and Republic of Macedonia, These results show that A. m. carnica and A. m. macedonica share the same C2D mtDNA haplotype. COI gene segments of 1680 samples were PCR-amplified and digested with restriction enzymes NcoI and StyI in order to discriminate A. m. macedonica from A. m. carnica. Amplified fragment patterns produced by both restriction enzymes matched with diagnostic pattern characteristic for A. m. macedonica in case of samples from east, south and south-west parts of Serbia, and Republic of Macedonia, fragments of samples from northern part of Serbia and Bosnia and Herzegovina did not include NcoI, and StyI restriction sites. These results indicate that honey bees from east, south and south-west parts of Serbia, and Republic of Macedonia belong to the A. m. macedonica, and honey bees from northern part of Serbia and Bosnia and Herzegovina belong to another subspecies, probably to the A. m. carnica. Therefore A. m. macedonica has much wider area of distribution than it was previously considered.  相似文献

A new species, Polydactylus siamensis, is described on the basis of eight specimens from Thailand. The species is similar to P. plebeius (Broussonet, 1782) in having five pectoral filaments and several dark stripes along the scale rows above and below the lateral line, but differs from the latter in having lower counts of pectoral fin rays (15 vs. 16–18 in P. plebeius), pored lateral line scales (54–58 vs. 60–68), scale rows above and below the lateral line (7 and 10 or 11, respectively vs. 8 or 9 and 12 or 13, respectively) and gill rakers (9 or 10 upper series, 13 or 14 lower and 22–24 total vs. 9–14, 13–18 and 24–32, respectively), and a longer upper jaw (mean 17% [range 16–17%] of standard length vs. 15% [13–16%]). Polydactylus siamensis is currently known only from Bangkok and Songkhla, Gulf of Thailand, and Phuket Island, Andaman Sea, whereas P. plebeius is widely distributed in the Indo-Pacific. In its native Europe, the bumblebee, Bombus terrestris (L.) has co-evolved with a large array of parasites whose numbers are negatively linked to the genetic diversity of the colony. In Tasmania B. terrestris was first detected in 1992 and has since spread over much of the state. In order to understand the bee’s invasive success and as part of a wider study into the genetic diversity of bumblebees across Tasmania, we screened bees for co-invasions of ectoparasitic and endoparasitic mites, nematodes and micro-organisms, and searched their nests for brood parasites. The only bee parasite detected was the relatively benign acarid mite Kuzinia laevis (Dujardin) whose numbers per bee did not vary according to region. Nests supported no brood parasites, but did contain the pollen-feeding life stages of K. laevis. Upon summer-autumn collected drones and queens, mites were present on over 80% of bees, averaged ca. 350–400 per bee and were more abundant on younger bees. Nest searching spring queens had similar mite numbers to those collected in summer-autumn but mite numbers dropped significantly once spring queens began foraging for pollen. The average number of mites per queen bee was over 30 fold greater than that reported in Europe. Mite incidence and mite numbers were significantly lower on worker bees than drones or queens, being present on just 51% of bees and averaging 38 mites per bee. Our reported incidence of worker bee parasitism by this mite is 5–50 times higher than reported in Europe. That only one parasite species co-invaded Tasmania supports the notion that a small number of queens founded the Tasmanian population. However, it is clearly evident that both the bee in the absence of parasites, and the mite have been extraordinarily successful invaders.
