Partial α-amylase gene sequences were determined and α-amylase gene expression was quantified in four species of carnivorous, omnivorous, and herbivorous prickleback fishes (family Stichaeidae) to assess the effects of ontogeny, diet, and species on expression of this gene. Pairwise comparison of α-amylase nucleotide sequences revealed 96–98 % identity, and comparison of amino acid portions revealed 93–95 % similarity among the four prickleback species. Expression was determined using in situ hybridization and intensity of expression quantified using image analysis. Alpha-amylase expression level was compared in three feeding categories of the four species: (1) small, wild-caught carnivorous juveniles; (2) larger, wild-caught juveniles of the carnivorous species and the three that had shifted to herbivory or omnivory; and (3) larger, juveniles produced by feeding a low-starch artificial diet to small juveniles until they reached the size of the larger wild-caught juveniles. The results showed no dietary effect in any species but significant ontogenetic and species-level effects in Cebidichthys violaceus, as well as in the sister species Xiphister mucosus and X. atropurpureus. Based on a phylogeny for the Stichaeidae produced for this study using two mtDNA genes and one nuclear gene, the ontogenetic dietary shifts to herbivory/omnivory evolved independently in C. violaceus and in the clade containing the two species of Xiphister. All three of these species increased α-amylase gene expression with increase in size and had higher expression than Anoplarchus purpurescens, which is a member of a third, stichaeid clade comprising carnivores. These results show the importance of α-amylase in the herbivores and omnivores.  相似文献

Three digestive enzymes in four species of closely related prickleback fishes (family Stichaeidae: Cebidichthys violaceus, Xiphister mucosus , Xiphister atropurpureus and Anoplarchus purpurescens ) were analysed to assess whether diet or phylogeny played a larger role in influencing digestive enzyme activity. Cebidichthys violaceus and X. mucosus are primarily herbivorous, whereas X. atropurpureus and A. purpurescens are mainly carnivorous. The two Xiphister species are sister taxa, and A. purpurescens is in a clade adjacent to that of the three other species. Pepsin and trypsin specific activities did not differ significantly among the four species, but α‐amylase activity was significantly higher in the two Xiphister species, followed by C. violaceus , and then A. purpurescens . The wide disparity between the two carnivores, the striking similarity between the two sister taxa, and the significant difference between the two herbivores indicate that activity of α‐amylase follows a pattern influenced more by phylogeny than by diet in these fishes.  相似文献

Symbioses with gut microorganisms provides a means by which terrestrial herbivores are able to obtain energy. These microorganisms ferment cell wall materials of plants to short-chain fatty acids (SCFA), which are then absorbed and used by the host animal. Many marine herbivorous fishes contain SCFA (predominantly acetate) in their hindgut, indicative of gut microbial activity, but rates of SCFA production have not been measured. Such information is an important prerequisite to understanding the contribution that gut microorganisms make in satisfying the energy needs of the fish. We have estimated the rates of acetate production in the gut of three species of temperate marine herbivorous fish from northeastern New Zealand: Kyphosus sydneyanus (family Kyphosidae), Odax pullus (family Odacidae), and Aplodactylus arctidens (family Aplodactylidae). Ex vivo preparations of freshly caught fish were maintained with their respiratory and circulatory systems intact, radiolabeled acetate was injected into ligated hindgut sections, and gut fluid was sampled at 20-min intervals for 2 h. Ranges for acetate turnover in the hindguts of the studied species were determined from the slope of plots as the log of the specific radioactivity of acetate versus time and pool size, expressed on a nanomole per milliliter per minute basis. Values were 450 to 570 (K. sydneyanus), 373 to 551 (O. pullus), and 130 to 312 (A. arctidens). These rates are comparable to those found in the guts of herbivorous reptiles and mammals. To determine the contribution of metabolic pathways to the fate of acetate, rates of sulfate reduction and methanogenesis were measured in the fore-, mid-, and hindgut sections of the three fish species. Both rates increased from the distal to proximal end of the hindgut, where sulfate reduction accounted for only a small proportion (<5%) of acetate methyl group transformed to CO2, and exceeded methanogenesis from acetate by >50-fold. When gut size was taken into account, acetate uptake from the hindgut of the fish species, determined on a millimole per day per kilogram of body weight basis, was 70 (K. sydneyanus), 18 (O. pullus), and 10 (A. arctidens).  相似文献

Few investigations have empirically analyzed fish gut function in the context of chemical reactor models. In this study, digestive enzyme activities, levels of gastrointestinal fermentation products [short chain fatty acids (SCFA)], luminal nutrient concentrations, and the mass of gut contents were measured along the digestive tract in herbivorous and carnivorous minnows to ascertain whether their guts function as “plug-flow reactors” (PFRs). Four of the species, Campostoma anomalum, C. ornatum, C. oligolepis, and C. pauciradii, are members of a monophyletic herbivorous clade, whereas the fifth species, Nocomis micropogon, is a carnivore from an adjacent carnivorous clade. In the context of a PFR model, the activities of amylase, trypsin and lipase, and the concentrations of glucose, protein, and lipid were predicted to decrease moving from the proximal to the distal intestine. I found support for this as these enzyme activities and nutrient concentrations generally decreased moving distally along the intestine of the four Campostoma species. Furthermore, gut content mass and the low SCFA concentrations did not change (increase or decrease) along the gut of any species. Combined with a previous investigation suggesting that species of Campostoma have rapid gut throughput rates, the data presented here generally support Campostoma as having guts that function as PFRs. The carnivorous N. micropogon showed some differences in the measured parameters, which were interpreted in the contexts of intake and retention time to suggest that PFR function breaks down in this carnivorous species.  相似文献

Most young fishes lack the ability to function as herbivores, which has been attributed to two aspects of the digestive system: elevated nitrogen demand and a critical gut capacity. We compared the digestive morphology and biochemistry of two size classes of the marine herbivore Hyporhamphus regularis ardelio, pre-ontogenetic trophic shift (pre-OTS, <100mm) and post-ontogenetic trophic shift (post-OTS, >100mm), to determine what limits the onset of herbivory and how their digestive processes fit with current models of digestion. Two gut-somatic indices comparing gut length to body length (relative gut length) and body mass (Zihler's Index) demonstrated a significant decrease (RGL 0.59→0.49, P<0.01; ZI 3.24→2.44, P<0.01) in gut length relative to body size. There was little difference in enzyme activity between the two classes, with juveniles showing similar levels of carbohydrase and lipase and less protease compared with adults, indicating that juveniles did not preferentially target nitrogen and were as capable of digesting an herbivorous diet. These findings suggest that herbivory in this fish is not limited by the function of the post-oesophageal digestive tract, but rather the ability of the pharyngeal mill to mechanically process plants. Our findings offer partial support for the current model of stomachless digestion, indicating that further refinement may be necessary.  相似文献

Synopsis The distribution patterns, diets, and substratum (refuge) requirements of early juveniles of two sympatric stichaeid fishesCebidichthys violaceus andXiphister mucosus, were investigated in a rocky intertidal habitat at Diablo Canyon, California. Monthly investigations were conducted at low tide for four consecutive months, to assess ontogenetic differences in distribution, diet, and refuge requirements within and between the two species. Distinct differences in vertical zonation were exhibited by both stichaeids throughout the study. Interspecific zonation patterns were similar to those recorded for adults of both species. Diet analyses showed that early juveniles of both stichaeids were zooplanktivorous, differing markedly from the primarily herbivorous diets of adults. Changes in diet were largely due to the selection of larger prey taxa as both fishes, and their mouth size, grew over the study period. Predation by both fishes on water-column planktors (calanoid copepods, zoea and polychaete larvae) was greatest following initial intertidal settlement and habitat establishment by early juvenile fishes. Greater dependence on substrate-oriented and/or benthic prey (harpacticoid copepods, gammarid amphipods and mysid shrimp) was exhibited by both fishes as they grew in size. Affinities for sand, gravel, and pebbles during monthly field surveys were similar for both species throughout the study. As the fishes grew, their substratum preferences changed in relation to the substrata which provided the best refuge. Results from laboratory experiments indicated that young stichaeids select very specific substrata based on fish age (size) and substratum suitability (i.e. adequate refuge). Similarities in diet and substratum preferences, and changes in those preferences over time, appear to be the result of morphological similarities (body size and shape and mouth gape) for both species at a given age.  相似文献

A range of digestive tract specializations were compared among dietary categories in the family Liolaemidae to test the hypothesis that herbivores require greater gut complexity to process plant matter. Additionally, the hypothesis that herbivory favors the evolution of larger body size was tested. Lastly, the association between diet and hindgut nematodes was explored. Herbivorous liolaemids were larger relative to omnivorous and insectivorous congeners and consequently had larger guts. In addition, small intestine length of herbivorous liolaemids was disproportionately longer than that of congeners. Significant interaction effects between diet and body size among organ dimensions indicate that increases in organ size occur to a greater extent in herbivores than other diet categories. For species with plant matter in their guts, there was a significant positive correlation between the percentage of plant matter consumed and small intestine length. Herbivorous liolaemids examined in this study lacked the gross morphological specializations (cecum and colonic valves) found in herbivores in the families Iguanidae and Agamidae. A significantly greater percentage of herbivorous species had nematodes in their gut. Of the species with nematodes, over 95% of herbivores had nematodes only in the hindgut. Prevalence of nematodes in the hindgut of herbivores was 2 x that of omnivores and 4 x that of insectivores.  相似文献

Differences in the allometric scaling between gut capacity (with body mass, BM1.00) and food intake (with BM0.75) should theoretically result in a scaling of digesta retention time with BM0.25 and therefore a higher digestive efficiency in larger herbivores. This concept is an important part of the so-called ‘Jarman–Bell principle’ (JBP) that explains niche differentiation along a body size gradient in terms of digestive physiology. Empirical data in herbivorous mammals, however, do not confirm the scaling of retention time, or of digestive efficiency, with body mass. Here, we test these concepts in herbivorous reptiles, adding data of an experiment that measured food intake, digesta retention, digestibility and gut capacity in 23 tortoises (Testudo graeca, T. hermanni , Geochelone nigra, G. sulcata, Dipsochelys dussumieri) across a large BM range (0.5–180 kg) to a literature data collection. While dry matter gut fill scaled to BM1.07 and dry matter intake to BM0.76, digesta mean retention time (MRT) scaled to BM0.17; the scaling exponent was not significantly different from zero for species > 1 kg. Food intake level was a major determinant of MRT across reptiles and mammals. In contrast to dietary fibre level, BM was not a significant contributor to dry matter digestibility in a General Linear Model. Digestibility coefficients in reptiles depended on diet nutrient composition in a similar way as described in mammals. Although food intake is generally lower and digesta retention longer in reptiles than in mammals, digestive functions scale in a similar way in both clades, indicating universal principles in herbivore digestive physiology. The reasons why the theoretically derived JBP has little empirical support remain to be investigated. Until then, the JBP should not be evoked to explain niche differentiation along a body size axis in terms of digestive physiology.  相似文献

The foraging ecology of mammalian herbivores is strongly shaped by plant secondary compounds (PSCs) that defend plants against herbivory. Conventional wisdom holds that gut microbes facilitate the ingestion of toxic plants; however, this notion lacks empirical evidence. We investigated the gut microbiota of desert woodrats (Neotoma lepida), some populations of which specialise on highly toxic creosote bush (Larrea tridentata). Here, we demonstrate that gut microbes are crucial in allowing herbivores to consume toxic plants. Creosote toxins altered the population structure of the gut microbiome to facilitate an increase in abundance of genes that metabolise toxic compounds. In addition, woodrats were unable to consume creosote toxins after the microbiota was disrupted with antibiotics. Last, ingestion of toxins by naïve hosts was increased through microbial transplants from experienced donors. These results demonstrate that microbes can enhance the ability of hosts to consume PSCs and therefore expand the dietary niche breadth of mammalian herbivores.  相似文献

Monocultures typical of intensive agriculture offer ideal conditions to specialized herbivores while depriving their natural enemies of habitat and nutritional resources. The resulting release of herbivores from both bottom-up and top-down control causes pest outbreaks in economically important crops. Boosting locally occurring natural enemy populations through species-specific habitat management to restore natural herbivore control has been much advocated but remains rarely tested in the field. Here, we investigated whether adding specifically selected flowering plants to monocultures increases parasitation rates of herbivores and crop yield. We performed replicated field experiments in 2 years and found that adding cornflowers (Centaurea cyanus) into cabbage (Brassica oleracea) fields significantly increased larval and egg parasitation and egg predation of the herbivore, reduced herbivory rates, and increased crop biomass in at least 1 year. These findings show that addition of a single, well-chosen flowering plant species can significantly increase natural top-down pest control in monocultures but success is variable. This is relevant on two applied levels. First, well chosen companion plants may partially substitute pesticides in agriculture if the approach is optimized, reducing negative effects such as unspecific killing of non-target organisms, residues in food, contamination of soils and water-bodies and increasing pesticide resistances. Our results suggest that, from an agro-economical point of view, egg parasitoids or predators may be the best targets for habitat management because strong natural selection acts on larval parasitoids to keep their hosts alive for their own development. Second, the addition of non-crop vegetation to monocultures benefits biodiversity conservation directly through resource diversification and indirectly through the reduction of pesticide application that increased natural control makes possible.  相似文献

The structure and ontogeny of lateral‐line canals in the Rock Prickleback, Xiphister mucosus, were studied using cleared‐and‐stained specimens, and the distribution and morphology of neuromasts within lateral‐line canals were examined using histology. X. mucosus has seven cephalic canals in a pattern that, aside from four branches of the infraorbital canals, is similar to that of most teleostean fishes. Unlike most other teleosts, however, X. mucosus features multiple trunk lateral‐line canals. These include a short median posterior extension of the supratemporal canal and three paired, branching canals located on the dorsolateral, mediolateral, and ventrolateral surfaces. The ventrolateral canal (VLC) includes a loop across the ventral surface of the abdomen. All trunk canals, as well as the branches of the infraorbitals, are supported by small, dermal, ring‐like ossifications that develop independently from scales. Trunk canals develop asynchronously with the mediodorsal and dorsolateral canals (DLC) developing earliest, followed by the VLC, and, finally, by the mediolateral canal (MLC). Only the mediodorsal and DLC connect to the cephalic sensory canals. Fractal analysis shows that the complexity of the trunk lateral‐line canals stabilizes when all trunk canals develop and begin to branch. Histological sections show that neuromasts are present in all cephalic canals and in the DLC and MLC of the trunk. However, no neuromasts were identified in the VLC or its abdominal loop. The VLC cannot, therefore, directly function as a part of the mechanosensory system in X. mucosus. The evolution and functional role of multiple lateral‐line canals are discussed. J. To determine what capabilities wood-eating and detritivorous catfishes have for the digestion of refractory polysaccharides with the aid of an endosymbiotic microbial community, the pH, redox potentials, concentrations of short-chain fatty acids (SCFAs), and the activity levels of 14 digestive enzymes were measured along the gastrointestinal (GI) tracts of three wood-eating taxa (Panaque cf. nigrolineatus “Marañon”, Panaque nocturnus, and Hypostomus pyrineusi) and one detritivorous species (Pterygoplichthys disjunctivus) from the family Loricariidae. Negative redox potentials (?600 mV) were observed in the intestinal fluids of the fish, suggesting that fermentative digestion was possible. However, SCFA concentrations were low (<3 mM in any intestinal region), indicating that little GI fermentation occurs in the fishes’ GI tracts. Cellulase and xylanase activities were low (<0.03 U g?1), and generally decreased distally in the intestine, whereas amylolytic and laminarinase activities were five and two orders of magnitude greater, respectively, than cellulase and xylanase activities, suggesting that the fish more readily digest soluble polysaccharides. Furthermore, the Michaelis–Menten constants (K m) of the fishes’ β-glucosidase and N-acetyl-β-d-glucosaminidase enzymes were significantly lower than the K m values of microbial enzymes ingested with their food, further suggesting that the fish efficiently digest soluble components of their detrital diet rather than refractory polysaccharides. Coupled with rapid gut transit and poor cellulose digestibility, the wood-eating catfishes appear to be detritivores reliant on endogenous digestive mechanisms, as are other loricariid catfishes. This stands in contrast to truly “xylivorous” taxa (e.g., beavers, termites), which are reliant on an endosymbiotic community of microorganisms to digest refractory polysaccharides.  相似文献

理解动物的营养生态位是充分理解其整个生态学的基础,对于害兽控制和物种保护也很重要,食肉动物的小肠很发达,这可能与对食物的高消化能力有关;杂食性动物有更复杂的胃肠器官,其后端有可进行发酵的盲肠,消化物的平均滞留时间(mean retention times,MRTs)更长;最长的平均滞留时间见于食草动物,其消化道内高密度的微生物种群对不同滞留区内的消化物进行发酵,但是,并不是所有的食草动物都能够最大程度地消化植物纤维,只有反刍动物、骆驼和个体较大的后肠发酵动物(hindgut fermenter)能够具有这种能力,对比而言,许多其它的食草动物,如前肠发酵的有袋类和小型的后肠发酵动物如兔子、田鼠和负鼠等,它们具备可以使植物纤维消化效率最大的消化系统,可以在食物中的纤维素含量非常高的情况下仍能处理大量的食物。这些不同的消化策略使哺乳动物具有广幅的营养生态位。  相似文献

Biofouling ascidians are ubiquitous in coastal ecosystems and are among the main colonizers of aquaculture gear. Our study tested the hypothesis that the transport, removal, and transfer of fouling ascidian species by aquaculturists provide a mechanism for concentration and distribution of harmful-algal cells to new areas. Wild-caught specimens of common, biofouling ascidian species (Styela clava, Ciona intestinalis, Molgula manhattensis, Botrylloides violaceus, Didemnum vexillum, and Botryllus schlosseri) were exposed individually to cultured strains of co-occurring harmful algae (Prorocentrum minimum, Alexandrium fundyense, Alexandrium monilatum, Karenia brevis, Aureococcus anophagefferens, or Heterosigma akashiwo) at simulated bloom cell densities of each HAB species. After feeding, ascidians were transferred to ultrafiltered seawater. Immediately after exposure, and after 24 and 48 h in ultrafiltered seawater, biodeposits were collected and observed microscopically for the presence of intact, potentially viable cells. Subsamples of biodeposits were transferred into culture tubes with ultrafiltered seawater and monitored for algal growth during 8 weeks. Cells of all HAB species were found to pass intact through the ascidian digestive system, remained viable, and in many cases were capable of re-establishing populations at least 48 h post-ingestion. The results of our study will inform industry and managers of the potential threat and ecological impact of spreading biofouling ascidians, and practices to mitigate adverse impacts. Additionally, these management practices have been formally incorporated into a new cost-share program developed to help shellfish producers prevent the further spread of ascidians and associated HAB species.  相似文献

Coral reefs are characterized by intense herbivory. Spatial patterns in herbivory—particularly along the depth gradient—influence the distribution and abundance of algae. Depth gradients in herbivorous reef fishes are generally assumed to be temporally stable, but this assumption has rarely been questioned. Here, we use underwater visual census and herbivore exclusion experiments to study the community composition and temporal patterns in habitat use by roving herbivorous fishes in an environment characterized by profound seasonal changes in algal biomass and distribution and extreme summer temperatures. Among the 18 species of roving herbivores recorded, parrotfishes were dominant in species richness and biomass, while regional endemic species represented 77 % of the total biomass. During most of the year, roving herbivores aggregate in the shallow reef zones and their biomass declines with depth. The herbivore community on the reef flat is distinct from that in deeper zones. The former is characterized by Siganus rivulatus, Acanthurus gahhm and Hipposcarus harid, while the deeper reef zones are characterized by S. ferrugineus, Chlorurus sordidus and Ctenochaetus striatus. In summer, the distinct community structures among reef zones are lost as reef flat herbivores tend to exploit deeper reef zones and some reef crest species venture on to the reef flat. This summer change in herbivore distribution is also reflected in reduced turf biomass and increased yield to herbivores in the deeper reef zones. Habitat use is related to the feeding mode such that browsers dominate the reef flat and scrapers the reef crest, while the seasonal changes correspond to changes in availability of targeted algal resources. These seasonal changes appear to be driven by the extreme temperatures in summer, reaching 36 °C on the shallow reef flat.  相似文献

1. Hindgut volume varies considerably between carnivores, omnivores and herbivores. But a common feature in all mammals is an extensive microbial fermentation of polysaccharides in the hindgut. Large amounts of short chain fatty acids (SCFA) are produced. Total concentrations of SCFA are generally ca 100 mmol/l. SCFA metabolism contributes considerably to the energy metabolism of the animal. 2. In hindgut fermenting herbivores ileal outflow provides fluid and the buffering capacity essential for microbial metabolism. 3. SCFA are rapidly absorbed. Absorption is passive and, unexpectedly, nearly independent from luminal pH. This is attributed to the presence of a constant pH-microclimate at the epithelial surface. 4. The permeability of the proximal compared to the distal colon of guinea pig is higher for acetate, equal for propionate and lower for butyrate. This difference is due to partial absorption of SCFA in the dissociated form in the proximal segment. 5. Protons required for SCFA transport in the undissociated form may be partially explained by HCO3 accumulation or by Na-H exchange. Findings are controversial.  相似文献

应用光学显微镜观察龟足(Capitulum mitella)消化系统的形态和组织结构。龟足的消化系统包括消化腺和消化道。消化腺一对,呈长囊状,含有分泌细胞(B细胞)、吸收细胞(R细胞)、储存细胞(F细胞)和胚细胞(E细胞)4种类型细胞。消化道呈U型,由口、食道、胃、肠、直肠和肛门组成,各部分的结构由内到外可分为黏膜层、黏膜下层、肌层和外膜4层。口器为咀嚼型,包括一片上唇、一对触须、一对大颚以及两对小颚。食道细短,具几丁质层但无基膜,管壁向腔内突起形成明显的纵褶突;食道前段的环肌特别发达,同时独有放射肌。胃略呈球袋状,肠较长;胃和肠的组织结构相似,没有几丁质层,上皮细胞都有发达的微绒毛。直肠细长,外膜分布有16组纵肌;直肠前段的组织结构与胃、肠相似,而直肠后段有几丁质层覆盖,黏膜层、黏膜下层、肌层和外膜渐退化,16组纵肌渐发达。肛门16组更加发达的纵肌挤入上皮细胞下方,在外膜外另出现一层明显的环肌。龟足消化道各部分的组织结构差异明显,反映了它们功能的差异。  相似文献
