为了弄清血红蛋白A_2现象的发生机制,我们对“红细胞HbA_2”的化学组成进行了分析。“红细胞HbA_2”的双向电泳结果表明,它含有两种血红蛋白成分:一种相当于HbA,另一种很可能是溶血液HbA_2。其单向二次电泳结果也证明,它是由溶血液HbA_2和HbA所组成。结果初步说明,盘红细胞中HbA_2可能与HbA结合存在。两者可能有相互作用,也许这是产生血红蛋白A_2现象的原因。  相似文献

猪链球菌2型(Streptococcus suis serotype2,SS2)是一种世界范围内的引起猪链球菌病的重要的人兽共患病原菌,人感染SS2后多呈现败血症型和脑膜炎型。2005年猪链球菌在我国四川省的感染暴发和近年来不断出现的零散病例给我国的公共卫生和食品安全带来了威胁,但迄今为止,对SS2致病机制研究尚不够深入。SS2的致病过程和宿主细胞相互作用密切相关,SS2-体外宿主细胞相互作用模型的建立对于揭示SS2致病分子机理具有重要的意义。目前的研究报道主要是从猪链球菌黏附与侵袭上皮细胞或内皮细胞、猪链球菌抗吞噬细胞的吞噬和猪链球菌激活免疫细胞炎症反应方面研究猪链球菌与宿主细胞的相互作用,但与其他常致病性链球菌如化脓性链球菌和肺炎链球菌相比,有些SS2-体外宿主细胞相互作用模型还缺乏统一的标准,由于感染模型的实验参数不尽相同得到结果也有所不同。本文对近年来猪链球菌2型与宿主细胞体外相互作用及其致病机理研究进展作一综述。  相似文献

KyoT是一种LIM结构域蛋白,是以RBP-J为诱饵蛋白通过酵母双杂交系统得到的新分子,KyoT2可以抑制RBP-J介导的转录,以KyoT2为诱饵蛋白,通过酵母双杂交筛选得到了人类紧密连接蛋白2(ZO-2),的一种新的剪接体ZO-2-i3,序列分析表明,新的剪接体有19个外显子,与已发表序列比较,见ZO-2-i3分子的激酶后区发生了改变,为排除酵母双杂交实验的假阳性,实验首先在酵母中验证KyoT2与ZO-2-i3的体外直接相互作用并得到阳性结果,进而在大肠杆菌中原核表达纯化带有His标签的KyoT2蛋白,使用抗His标签的抗体,通过GST pull-down assay验证KyoT2与ZO-2-i3的体外直接相互作用,也获得阳性结果,并通过酵母实验初步确定了其作用位点,即KyoT2通过LIM2结构域与ZO-2-i3相互作用,本实验验证了KyoT2与Z-2-i3的相互作用,并初步确定其相互作用位点,对探讨KyoT的功能具有重要意义。  相似文献

当今的蛋白质组学研究,都是先裂解细胞放出蛋白质,然后对蛋白质溶液进行各种分析.对于红细胞来说,它的裂解产物也称"溶血液",其中主要成分有血红蛋白A1,A2,A3和碳酸酐酶(CA)等.本实验室用未裂解的完整的活体红细胞直接进行电泳,观察其释放出来的血红蛋白(hemoglobin,Hb),建立了淀粉-琼脂糖混合凝胶中红细胞的电泳释放实验.电泳释放可分为"初释放"(一次通电完成电泳,此时有Hb释放出来)和"再释放"(电泳过程中断电-再通电,又有Hb释放出来).本实验室在"初释放"实验中发现了"HbA2现象",并通过Hb交叉电泳发现了HbA2与HbA1的相互作用;利用初释放型双向对角线电泳发现红细胞内HbA2与HbA1结合存在;对电泳释放出来的"HbA2现象"成分做SDS-PAGE及质谱分析,发现Prx-2(Peroxiredoxin-2)可能参与"HbA2现象"的形成;在研究"再释放"实验中发现了"Hb多带再释放现象",在此基础上创建等渗再释放、低渗再释放、等低渗全程再释放及再释放型双向对角线电泳;两种红细胞(全血中的红细胞和由它分离出来的游离红细胞)再释放的比较研究;血浆成分对红细胞再释放的影响等.以上研究方法的建立为活体细胞内蛋白质存在状态的研究提供了基础,并开辟了新的研究途径和领域.  相似文献

目的:观察肝素与23种染料之间的相互作用.方法:利用淀粉琼脂糖凝胶电泳及聚丙烯酰胺凝胶电泳,观察染料加肝素后电泳行为的变化.结果:淀粉琼脂糖凝胶电泳中染料加肝素后多数结果是泳速变慢,有一些留在原点,也有前移、后退者.二氯荧光素、荧光红、荧光素、甲酚红、苯胺蓝、溴酚蓝、茜素红S、胭脂红、亮绿、氨基黑10B、丽春红G、曙红、苯胺黑、氯酚红加肝素后泳速变慢.亚甲蓝、灿烂甲酚蓝、甲基紫加肝素后电泳留在原点.刚果红、洋红加肝素后电泳变化不明显.四氯荧光素、氯化硝基四氮唑蓝加肝素后电泳荧光看不清.聚丙烯酰胺凝胶电泳中溴酚蓝加入肝素后泳速明显变慢,随着肝素量增加溴酚兰的泳速加快.与淀粉琼脂糖凝胶电泳结果一致,但它的电泳结果更明显.结论:本次所研究的23种染料,几乎都能与肝素发生相互作用,给它们的进一步深入研究打下基础.  相似文献

【目的】探讨A型流感病毒PB1-F2蛋白和人类凋亡调节因子1(MOAP-1)之间的相互作用。【方法】构建pACT2-MOAP-1重组质粒,与pGBKT7-PB1-F2质粒共转化酵母AH109,检测转化菌在四缺培养基的生长情况及β半乳糖苷酶报告基因的活性;利用GST pull-down和免疫共沉淀(Co-IP)技术进一步验证PB1-F2与宿主细胞蛋白MOAP-1的相互作用;通过过表达PB1-F2和MOAP-1,检测PB1-F2对MOAP-1蛋白表达水平的影响。【结果】酵母双杂交结果表明,PB1-F2和MOAP-1可以在酵母细胞内特异性结合。GST pull-down和Co-IP实验也进一步证实了这两种蛋白的相互作用,而且PB1-F2可上调外源MOAP-1的蛋白水平。【结论】流感病毒PB1-F2与MOAP-1存在相互作用,PB1-F2可能通过与MOAP-1的相互作用参与调控细胞生长及凋亡过程。  相似文献

广西眼镜王蛇毒用羧甲基纤维素CM-52、磷酸纤维素P-11和Sepharose CL-6B柱层析纯化,得到一个在聚丙烯酰胺凝胶电泳上为单一蛋白带,PLA_2的比活性较原蛇毒提高3.6倍,分子量的为13000,由122个氨基酸组成,_pI为8.9,具有良好的热稳定性。从碱性PLA_2对红细胞影响的电镜观察可见,对人的红细胞膜有明显的作用,而对山羊红细胞作用不明显。PLA_2无论对人还是对山羊、兔和豚鼠红细胞电泳速度都有明显的迟缓作用。  相似文献

Hemoglobin A2 (22), which is present at low concentration (1–2%) in the circulating red cells of normal individuals, has two important features that merit its study, i.e., it inhibits polymerization of sickle HbS and its elevated concentration in some thalassemias is a useful clinical diagnostic. However, reports on its functional properties regarding O2 binding are conflicting. We have attempted to resolve these discrepancies by expressing, for the first time, recombinant hemoglobin A2 and systematically studying its functional properties. The construct expressing HbA2 contains only and genes so that the extensive purification required to isolate natural HbA2 is circumvented. Although natural hemoglobin A2 is expressed at low levels in vivo, the amount of recombinant 22 expressed in yeast is similar to that found for adult hemoglobin A and for fetal hemoglobin F when the + or the + genes, respectively, are present on the construct. Recombinant HbA2 is stable, i.e., not easily oxidized, and it is a cooperative functional hemoglobin with tetramer-dimer dissociation properties like those of adult HbA. However, its intrinsic oxygen affinity and response to the allosteric regulators chloride and 2,3-diphosphoglycerate are lower than the corresponding properties for adult hemoglobin. Molecular modeling studies which attempt to understand these properties of HbA2 are described.  相似文献

目的:探究鼠双微体2同源体(MDM2)与端粒保护蛋白1(POT1)在细胞水平是否有相互作用,及其是否发挥E3泛素化连接酶的功能。方法:首先,用蛋白酶体抑制剂MG132处理稳定表达Flag-POT1的HeLa细胞,Western印迹检测Flag-POT1的表达情况;其次,在HeLa细胞中转入外源的Myc-MDM2和Flag-POT1质粒,48 h后收集细胞,通过免疫共沉淀方法验证Myc-MDM2和Flag-POT1是否具有相互作用;再次,在稳定表达Flag-POT1的HeLa细胞中转入Myc-MDM2或MDM2 siRNA,48 h后收集细胞,Western印迹检测Flag-POT1的表达水平。结果:MG132处理后,Flag-POT1的表达量明显升高且有拖尾现象,免疫共沉淀显示Myc-MDM2和Flag-POT1具有相互作用,但无论转入Myc-MDM2还是MDM2 siRNA,Flag-POT1的表达水平没有明显变化。结论:POT1通过泛素化途径降解,MDM2与POT1具有相互作用,但MDM2不是POT1主要的E3泛素化连接酶。  相似文献

2,3-Bisphosphoglycerate is a physiologically important regulator of red cell oxygen affinity during mammalian development. The rat has no fetal hemoglobin, but the newborn red cell has low 2,3-bisphosphoglycerate and high ATP concentrations, and high oxygen affinity. This report shows that red cell bisphosphoglyceromutase activity increases from near zero in the newborn rat to very high levels by four weeks of age. This increase roughly parallels the increase in red cell 2,3-bisphosphoglycerate concentration. Red cell pyruvate kinase activity declines ten-fold from birth to four weeks of age. This decrease is associated with a changeover in red cell populations from larger to smaller cells. The glycolytic rate is at least 50% higher in newborn than adult rat red cells. The data suggest that high pyruvate kinase activity and glycolytic rate contribute to the high ATP concentration in newborn rat red cells, but that their low 2,3-bisphosphoglycerate concentration is due primarily to low bisphosphoglyceromutase activity.  相似文献

The interaction and association between chitosan and hemoglobin (Hb) are studied by UV-vis and fluorescence spectroscopies, viscometry, circular dichroism, dynamic and static light scattering. Chitosan can obviously associate with Hb to form protein-chitosan complexes, which affects microstructure of Hb. The distance between the first association site of chitosan with 214-tryptophan residue in Hb is about 5.473 nm. The intrinsic UV-vis absorption and fluorescence intensities of Hb increase with an increase of chitosan concentration. The alpha-helix in Hb is drawn and changed into beta-sheet.  相似文献

To develop an enzymatic measurement of HbA(1C), two key enzymes, i.e., fructosyl peptide oxidase and Aspergillus protease were characterized. Fructosyl peptide oxidase from Eupenicillium terrenum was a flavoenzyme that could catalyze the oxidation of N-(1-deoxyfructosyl)-Val-His. The enzyme showed high specificity toward alpha-glycated molecules, therefore it seemed suitable for the HbA(1C) assay. Since high levels of FPOX expression seemed toxic to host cells, we applied a gene expression system using a bacteriophage vector and achieved high levels of expression in Escherichia coli. Next, we found that Aspergillus protease was able to digest N-(1-deoxyfructosyl)-hexapeptide, a glycated peptide that was released from the beta-chain of HbA(1C) by Glu-C endoproteinase. We showed that the N-(1-deoxyfructosyl)-Val-His released from N-(1-deoxyfructosyl)-hexapeptide by Aspergillus protease could be assayed enzymatically using fructosyl peptide oxidase, therefore these enzymes could be applied to the enzymatic measurement of HbA(1C).  相似文献

The binding force between a liposome and the C2A domain of synaptotagmin I was determined by an atomic force microscopy (AFM). Liposomes were immobilized on the surface of the L1 sensor chip and the C2A domains, which recognize phosphatidylserine, were chemically conjugated onto a gold-coated cantilever tip. The average interaction force between the C2A domain and the liposome was 306 (±57) pN while the force between untreated cantilever and the liposome was 58 (±16) pN. This work helps understand the physicochemical interactions between proteins and lipid vesicles for the design of high affinity protein probes against the apoptotic cell surface. A trimeric protein phosphatase 2A (PP2A(T55)) composed of the catalytic (PP2Ac), structural (PR65/A), and regulatory (PR55/B) subunits was isolated from rabbit skeletal muscle by thiophosphorylase affinity chromatography, and contained two additional proteins of 54 and 55 kDa, respectively. The 54 kDa protein was identified as eukaryotic translation termination factor 1 (eRF1) and as a PP2A interacting protein. The 55 kDa protein is now identified as nucleoredoxin (NRX). The formation of a complex between GST-NRX, PP2A(C) and PP2A(D) was demonstrated by pull-down experiments with purified forms of PP2A, and by immunoprecipitation of HA-tagged NRX expressed in HEK293 cells complexed endogenous PP2A subunits. Analysis of PP2A activity in the presence of GST-NRX showed that NRX competed with polycations for both stimulatory and inhibitory effects on different forms of PP2A.  相似文献

 本研究用聚丙烯酰胺凝胶垂直板电泳,双波长扫描仪对少年游泳运动员的HbA_2百分含量进行测定。发现大运动量训练对HbA_2水平是有影响的。经六天训练(星期二、五为大运动量训练),星期日休息一天,次周的星期一晨HbA_2百分含量升高,星期二大运动量训练后的次日晨HbA_2百分含量显著下降。运动性贫血的运动员,其HbA_2百分含量一周中的变化趋势与正常运动员相似,但HbA_2百分含量比正常运动员高。  相似文献
