Antarctic yeast strains were investigated for exopolysaccharide biosynthesis and the Sporobolomyces salmonicolor AL1 strain was selected. It was studied for exopolysaccharide biosynthesis on different carbon and nitrogen sources. The investigations showed that sucrose and ammonium sulphate were suitable culture medium components for polymer biosynthesis. Exopolysaccharide formation by the yeast strain was accompanied by a decrease in the culture medium pH value from the initial pH 5.3 to pH 1.7–2.0. During the biosynthetic process, the dynamic viscosity of the culture broth increased to the maximum value of 15.37 mPas and the polysaccharide yield reached 5.63 g/l on a culture medium containing 5.00% sucrose and 0.25% ammonium sulphate at a temperature of 22 °C for 120 h. The crude polysaccharide obtained from Sp. salmonicolor AL1 featured high purity (90.16% of carbon content) and consisted of glucose (54.1%), mannose (42.6%) and fucose (3.3%). Pure mannan containing 98.6% of mannose was isolated from it.  相似文献

牛筋草离孺孢生物学特性及代谢产物活性测定   总被引:1,自引:0,他引:1  
对牛筋草离孺孢属(Bipolaris sorokiniana)NJ-08菌株的生物学特性及代谢产物除草活性测定结果表明:此菌株的菌丝体生长速度慢, 但产孢量大, 致病性强。菌丝体生长最适温度25°C, 致死温度是55°C/10 min, 最适pH值为8, 碳源以葡萄糖最佳, 供试氮源则不利于菌丝体生长。产生分生孢子最适温度25°C, 最适pH值为8, 碳源以葡萄糖最好, 氮源以硝酸钠产孢量最高。分生孢子萌发温度在20°C~35°C之间没有差异, 适宜pH值为5~7, 碳源以D-木糖的孢子萌发率最高, 氮源以蛋白胨的孢子萌发率最高, 不同处理间存在明显差异。NJ-08菌株产生的代谢产物作用于牛筋草, 可使叶片黄化或枯死, 代谢产物对指示植物及杂草如柱花草、地桃花、猪屎豆和辣椒的胚根胚芽生长都有明显的抑制作用, 抑制率均超过了90%, 显示出很好的除草活性。  相似文献

An alkalophilic bacterial isolate identified as Bacillus pantotheneticus, isolated from saline-alkali soils of Avadh region of UP, India, was studied for the production of alkaline protease. The mutant of the isolated species showed 44% improved production over the parent strain. Organic nitrogen sources supported better protease production than the inorganic sources. The production of alkaline protease was (242 U/ml) in the medium containing molasses, which was comparable with molasses and wheat bran (285 U/ml) as carbon and nitrogen sources, respectively. Protease production was best at pH 10 and temperature 30 degrees C. The Km (for casein) was 11 mg/ml and Vmax was 380-microg tyrosine/ml/min. The enzyme was stable between pH 7 and 10.7 and temperature between 30 and 60 degrees C with a pH and temperature optimum at 8.4 and 40 degrees C, respectively. The results indicated that molasses was an optimal substrate for alkaline protease production.  相似文献

Sporulation of Streptomyces venezuelae in submerged cultures   总被引:2,自引:0,他引:2  
Shaken cultures of Streptomyces venezuelae ISP5230 in minimal medium with galactose and ammonium sulphate as carbon and nitrogen sources, respectively, showed extensive sporulation after 72 h incubation at 37 degrees C. The spores formed in these cultures resembled aerial spores in their characteristics. The ability of the spores to withstand lysozyme treatment was used to monitor the progress of sporulation in cultures and to determine the physiological requirements for sporulation. In media containing ammonium sulphate as the nitrogen source, galactose was the best of six carbon sources tested. With galactose S. venezuelae ISP5230 sporulated when supplied with any of several nitrogen sources; however, an excess of nitrogen source was inhibitory. In cultures containing galactose and ammonium sulphate, sporulation was suppressed by a peptone supplement. The onset of sporulation was accompanied by a drop in intracellular GTP content. When decoyinine, an inhibitor of GMP synthase, was added to a medium containing starch and ammonium sulphate, a slight increase in sporulation was seen after 2 d. The suppression of sporulation by peptone in liquid or agar cultures was not reversed by addition of decoyinine. A hypersporulating mutant of S. venezuelae ISP5230 was altered in its ability to assimilate sugars. In cultures containing glucose the mutant sporulated more profusely than did the wild-type and did not acidify the medium to the same extent. However, the suppressive effect of glucose on sporulation was not merely a secondary result of acid accumulation.  相似文献

The effects of the carbon and nitrogen sources, initial pH and incubation temperature on laccase production by the endophytic fungus Monotospora sp. were evaluated. The optimal temperature and initial pH for laccase production by Monotospora sp. in submerged culture were found to be 30 degrees C and 8.5, respectively. Maltose (2 g l(-1)) and ammonium tartrate (10 g l(-1)) were the most suitable carbon and nitrogen source for laccase production. Under optimal culture medium, the maximum laccase activity was determined to be 13.55 U ml(-1), which was approximately four times higher than that in basal medium. This is the first report on laccase production by an endophytic fungus.  相似文献

The effect of temperatures and exogenous supply of different carbon and nitrogen sources on the conidial germination and appresoria formation inC. capsici has been studied. 25 °C was observed to be the best temperature for conidial germination. At 18 °C, conidia germinated only in hanging drops and did not germinate on 2 % agar. Amongst carbon sources, 1-rhamnose supported maximum conidial germination and appresoria formation. Potassium nitrate supported very good conidial germination and appresoria formation. Ammonium nitrate, ammonium sulphate and ammonium chloride were found to be inhibitory. A few aminoacids stimulated conidial germination but dl-alanine, dl-methionine, tyrosine and l-glutamine were found to be inhibitory.  相似文献

Germination of spores of Chrysosporium crassitunicatum, Nannizzia fulva (+), Nannizzia fulva (–) and Trichophyton equinum was studied in the presence of various carbon and nitrogen sources. Effect of different temperatures on spore germination was also determined. Maximum spore germination within 24 hours was recorded when glucose was used as a carbon source for all the test fungi. Except sodium nitrate all the inorganic nitrogen sources enhanced the spore germination at 0.05% concentration. Most of the organic nitrogen sources used were found to be stimulatory for the spore germination of test fungi. Optimum temperature i.e. 28 °C supported maximum spore germination of all the test fungi within 24 hours. C. Crassitunicatum, N. fulva(+), N. fulva(–) could germinate upto 35 °C but beyond that no spore germination was noticed in these fungi. T. equinum could germinate at a higher temperature of 40 °C but the percentage of germination was very low.  相似文献

对前期筛选得到的具有生防潜力的杀蜱真菌Beauveria bassiana AT17菌株进行相关生物学性状的研究,建立一套规范的实验室培养的技术和方法,同时对其液固双相发酵技术进行优化,在于提供优质高效的生防材料,为其真菌制剂的规模化生产提供实践和理论基础。通过采用单因素筛选方案对其最适基础培养基、温度、pH值、碳源、氮源、微量元素及液固双相发酵配方进行研究,发现该菌株在PDA、PPDA、PDAY、SDAY、SMAY 5种培养基上均能较好生长,在PDA上生长最快,在PDAY上产孢量最大。温度对菌丝的生长和产孢影响显著,25°C菌丝生长最快,且产孢量最大。B.bassiana AT17菌株在pH 4-10范围内均可生长,在偏碱性环境内生长最快,在pH 5-6的偏酸性环境内产孢量最大。综合评价真菌各项指标后,葡萄糖和酵母粉为菌丝生长和产孢的最佳碳源和氮源,固体物料麦麸+玉米粉+米粉与基础培养液3/4 SDAY按2:1均匀混合后,添加0.05%K+可作为菌株固体发酵的最佳物料配方组合。  相似文献

从碳源、氮源、酸碱度及生长温度等方面对引起马铃薯干腐病的硫色镰孢Fusarium sulphureum的生物学特性进行了研究。结果表明,该病原菌在不同发育阶段对营养和环境条件的要求存在差异。在固体培养基上,菌落生长最佳碳源为葡萄糖、麦芽糖,最佳氮源为蛋白胨,pH8?为最佳;在液体培养基中,菌丝体生长以麦芽糖为最佳碳源,以硝酸钠为最佳氮源,pH6?为最佳;在分生孢子萌发阶段,在以羧甲基纤维素钠、蛋白胨和谷氨酸为碳、氮源的营养液中,分生孢子萌发率最高,最适pH 6–8。该病菌最适生长温度为25℃,分生孢子致死  相似文献

To enhance the production of micrococcin GO5, a bacteriocin produced by Micrococcus sp. GO5, cultivation conditions and medium composition were optimized. The optimal initial pH and temperature for bacteriocin production were 7.0-9.0 and 37 degrees C, respectively. Micrococcus sp. GO5 displayed the highest micrococcin GO5 activity when grown in modified MRS medium that contained lactose or sucrose, rather than glucose, as a carbon source. The maximum bacteriocin activity was obtained in modified MRS medium containing 0.5% tryptone and 1.0% yeast extract as nitrogen sources instead of the other nitrogen sources present in MRS medium. Bacteriocin production was greatly affected by the concentration of K(2)HPO(4); strain GO5 produced eight-fold more bacteriocin in medium containing 2.0-2.5% K(2)HPO(4) than in medium containing 0.2% K(2)HPO(4). The optimal concentration of MgSO(4).7H(2)O for bacteriocin production was 0.5%. The production of micrococcin GO5 was increased 32-fold in shake flask culture and 16-fold in a bioreactor using the optimized medium (TY medium), compared with culturing in MRS medium.  相似文献

The aim of this work was to make a survey describing factors that influence the production of extracellular enzymes by white-rot fungus Ceriporiopsis subvermispora responsible for the degradation of lignocellulolytic materials. These factors were: carbon sources (glucose, cellulose, hemicellulose, lignin, maltose and starch), nitrogen sources (ammonium sulphate, potassium nitrate, urea, albumin and peptone), pH, temperature and addition of three different concentrations of Cu2+ and Mn2+. The cellulase and xylanase activities were similar in medium with different carbon sources and the highest cellulase and xylanase activities were measured in medium with urea and potassium nitrate as nitrogen sources, respectively. The highest laccase activity was observed in medium with lignin and peptone as carbon and nitrogen sources. In other experiments, time course of production of lignocellulolytic enzymes by white-rot fungus C. subvermispora in medium with lignin or glucose as carbon sources was observed.  相似文献

以沈阳地区芹菜斑枯病菌的纯培养菌株为试材,通过对菌丝生长、孢子萌发的营养及环境条件的多因子试验,明确了芹菜斑枯病菌菌丝生长以在CDAMS—VI培养基上最好,其次是CSA、CCSA和PDA,菌丝生长及产孢对碳源、氮源种类有明显的选择性。菌丝生长的最适pH值为4~5,最适温度为21℃,致死温度为39℃(处理40min),光照明显抑制菌丝生长。病菌分生孢子在水和芹菜汁中不能萌发,在水琼脂上萌发最好;孢子萌发的适宜pH值为4.6~6.6,适温为17—25℃,致死温度为42℃(处理20min),RH100%最适宜孢子萌发。光照和变温处理可促进分生孢子器的形成,病菌在PDA上从孢子萌发到分生孢子器形成并释放出分生孢子需要16~18d。  相似文献

Biosurfactant production by Pichia anomala PY1, a thermotorelant strain isolated from fermented food, was examined as grown in media containing various carbon and nitrogen sources. The optimal conditions for biosurfactant production included 4% soybean oil as carbon source at pH 5.5 at 30 degrees C for 7 d. Under these conditions, the surface tension of the medium decreased to 28 mN/m with oil displacement measured at 69.43 cm(2). Comparative studies of biosurfactant production in media containing glucose or soybean oil were performed. The biosurfactants obtained were isolated and purified by chromatographic methods. The molecular weights of samples were further investigated by mass spectrometry. In medium containing glucose, biosurfactants of molecular weights of 675, 691, and 707 were obtained, while those isolated from medium containing soybean oil were of molecular weights of 658, 675, and 691. These results reveal that sophorolipid compounds containing fatty acids of C20 and C18:1 were produced from both media.  相似文献

A species of bacterium with high chitosanase activity was isolated from soil samples in Haiyan City, China, and identified as an Acinetobacter species. This strain, named Acinetobacter sp. strain C-17, produced a chitosanase that was inducible and secreted into the medium. The optimal conditions for enzyme production were cells used to inoculate a medium containing 1% chitosan (pH 7.0) followed by culture at 30 degrees C. The chitosanase activity reached 1.7 U/ml when strain C-17 was incubated in a 250-ml flask under the optimal conditions for 24 h, and reached 2.8 U/ml when cells were incubated in a 3-l fermentor. The optimal pH and temperature for hydrolysis of chitosanase were 7.0 and 36 degrees C, respectively. The chitosanase activity was stable in the pH range of 5-8 and temperature range of 30-40 degrees C. The chitosanase of the strain was extracted by zinc acetate and ammonium sulfate precipitation. The molecular mass was estimated to be 35.4 kDa by SDS-PAGE.  相似文献

A new actinomycete strain designated as BT-408 producing polyketide antibiotic SBR-22 and showing antibacterial activity against methicillin resistant Staphylococcus aureus has been characterized and found to be a novel strain of Streptomyces psammoticus. Nutritional and cultural conditions for the production of antibiotic by this organism under shake-flask conditions have been optimized. Glucose and ammonium nitrate were found to be best carbon and nitrogen sources respectively for growth and antibiotic production. Similarly initial medium pH of 7.2, incubation temperature of 30 degrees C and incubation time of 96 h were found to be optimal. Optimization of medium and cultural conditions resulted in 1.82-fold increase in antibiotic yield.  相似文献

Biosurfactant-producing bacteria were isolated from two petroleum contaminated sites in western Canada. Seven potential biosurfactant/bioemulsifier-producing isolates were screened and characterized. All of the seven isolates were able to form emulsions. Emulsion-stabilizing capacity was also measured up to 48 hrs. Strain C-111-2 and C-203-2 would lead to highly reduced surface tension. For strain C-203-2, the optimum conditions that supported bacteria growth and production were investigated. The influences of carbon sources, medium pH values, and temperature were taken into account. The experimental results indicated that the crude oil and glucose were promising carbon sources for biosurfactants production; the isolated strains produced a maximum concentration of biosurfactant in a neutral pH environment and showed a higher surface activity under the temperature level of 35°C than that under 10°C. To further optimize the carbon and nitrogen source for biosurfactant production, response surface methodology (RSM) was applied to explore the favorable concentration of two carbon sources: glucose, crude oil, and one nitrogen source, NaNO3. The optimal concentration of 8.1g/L, 4% and 3.9 g/L for glucose, crude oil, and NaNO3, respectively, which can be obtained through RSM analysis.  相似文献

The effects of the carbon and nitrogen sources, initial pH and incubation temperature on laccase production by Trametes modesta were evaluated using the one-factor-at-a-time method. The final optimisation was done using a central composite design resulting in a four-fold increase of the laccase activity to 178 nkat ml(-1). Response-surface analysis showed that 7.34 g l(-1) wheat bran, 0.87 g l(-1) glucose, 2.9 g l(-1) yeast extract, 0.25 g l(-1) ammonium chloride, an initial pH of 6.95 and an incubation temperature of 30.26 degrees C were the optimal conditions for laccase production. Laccase produced by T. modesta was fully active at pH 4 and at 50 degrees C. The laccase was very stable at pH 4.5 and at 40 degrees C but half-lives decreased to 120 and 125 min at higher temperature (60 degrees C) and lower pH (pH 3).  相似文献

Decolorization of the textile dyes by newly isolated bacterial strains   总被引:17,自引:0,他引:17  
Six bacterial strains with the capability of degrading textile dyes were isolated from sludge samples and mud lakes. Aeromonas hydrophila was selected and identified because it exhibited the greatest color removal from various dyes. Although A. hydrophila displayed good growth in aerobic or agitation culture (AGI culture), color removal was the best in anoxic or anaerobic culture (ANA culture). For color removal, the most suitable pH and temperature were pH 5.5-10.0 and 20-35 degrees C under anoxic culture (ANO culture). More than 90% of RED RBN was reduced in color within 8 days at a dye concentration of 3,000 mg l(-1). This strain could also decolorize the media containing a mixture of dyes within 2 days of incubation. Nitrogen sources such as yeast extract or peptone could enhance strongly the decolorization efficiency. In contrast to a nitrogen source, glucose inhibited decolorization activity because the consumed glucose was converted to organic acids that might decrease the pH of the culture medium, thus inhibiting the cell growth and decolorization activity. Decolorization appeared to proceed primarily by biological degradation.  相似文献

从驯化后的活性污泥中筛分、诱变出一株性能较好的异养硝化菌JZ1-1.经形态及生理生化特性分析,鉴定菌株JZ1-1为胶样菌属(Colloides sp.).分别考察了碳源、C/N、pH、溶解氧、温度和铵态氮初始浓度对JZ1-1硝化性能的影响.结果表明:菌株对柠檬酸钠的利用较好;C/N为10 ~14、30℃、pH 6-9和转速150 r·min-1以上有利于铵态氮的降解;菌株对中高浓度铵态氮废水(100 mg·L-1≤铵态氮浓度≤500 mg·L-1)的降解效果显著.经5次继代培养,菌株的稳定性较好.  相似文献
