Thiosulfate oxidation and mixotrophic growth with succinate or methanol plus thiosulfate was examined in nutrient-limited mixotrophic condition for Methylobacterium oryzae CBMB20, which was recently characterized and reported as a novel species isolated from rice. Methylobacterium oryzae was able to utilize thiosulfate in the presence of sulfate. Thiosulfate oxidation increased the protein yield by 25% in mixotrophic medium containing 18.5 mmol.L-1 of sodium succinate and 20 mmol.L-1 of sodium thiosulfate on day 5. The respirometric study revealed that thiosulfate was the most preferable reduced inorganic sulfur source, followed by sulfur and sulfite. Thiosulfate was predominantly oxidized to sulfate and intermediate products of thiosulfate oxidation, such as tetrathionate, trithionate, polythionate, and sulfur, were not detected in spent medium. It indicated that bacterium use the non-S4 intermediate sulfur oxidation pathway for thiosulfate oxidation. Thiosulfate oxidation enzymes, such as rhodanese and sulfite oxidase activities appeared to be constitutively expressed, but activity increased during growth on thiosulfate. No thiosulfate oxidase (tetrathionate synthase) activity was detected.  相似文献

Various cultures (previously described), which oxidize thiosulfate in mineral media have been studied in an attempt to determine the products of oxidation. The transformation of sodium thiosulfate by Cultures B, T, and K yields sodium tetrathionate and sodium hydroxide; secondary chemical reactions result in the accumulation of some tri- and pentathionates, sulfate, and elemental sulfur. As a result of the initial reaction, the pH increases; the secondary reactions cause a drop in pH after this initial rise. The primary reaction yields much less energy than the reactions effected by autotrophic bacteria. No significant amounts of assimilated organic carbon were detected in media supporting representatives of these cultures. It is concluded that they are heterotrophic bacteria. Th. novellus oxidizes sodium thiosulfate to sodium sulfate and sulfuric acid; the pH drops progressively with growth and oxidation. Carbon assimilation typical of autotrophic bacteria was detected; the ratio of sulfate-sulfur formed to carbon assimilated was 56:1. It is calculated that 5.1 per cent of the energy yielded by the oxidation of thiosulfate is accounted for in the organic cell substance synthesized from inorganic materials. This organism is a facultative autotroph. The products of oxidation of sodium thiosulfate by Th. thioparus are sodium sulfate, sulfuric acid, and elemental sulfur; the ratio of sulfate sulfur to elemental sulfur is 3 to 2. The pH decreases during growth and oxidation. The elemental sulfur is produced by the primary reaction and is not a product of secondary chemical changes. The bacterium synthesizes organic compounds from mineral substances during growth. The ratio of thiosulfate-sulfur oxidized to carbon assimilated was 125:1, with 4.7 per cent of the energy of oxidation recovered as organic cell substance. 对含有不同浓度铅的水稻土添加硫代硫酸钠进行土培试验,研究硫作用下铅污染水稻土微生物活性及群落结构的变化.结果表明:添加硫底物后,土壤的氧化还原电位(Eh)升高,呼吸强度增强,硫氧化菌的数量显著增加.硫促进了土壤中硫氧化菌等微生物的生长并引起群落结构的变化,克隆测序表明,加硫处理下土壤微生物的特异性条带与拟杆菌、硫杆菌、β-变形菌和嗜酸菌具有很高的相似性.硫处理下土壤碳酸盐结合态及铁锰结合态铅含量发生了显著变化.  相似文献

Sodium 2-propenyl thiosulfate, a water-soluble organo-sulfane sulfur compound isolated from garlic, induces apoptosis in a number of cancer cells. The molecular mechanism of action of sodium 2-propenyl thiosulfate has not been completely clarified. In this work we investigated, by in vivo and in vitro experiments, the effects of this compound on the expression and activity of rhodanese. Rhodanese is a protein belonging to a family of enzymes widely present in all phyla and reputed to play a number of distinct biological roles, such as cyanide detoxification, regeneration of iron-sulfur clusters and metabolism of sulfur sulfane compounds. The cytotoxic effects of sodium 2-propenyl thiosulfate on HuT 78 cells were evaluated by flow cytometry and DNA fragmentation and by monitoring the progressive formation of mobile lipids by NMR spectroscopy. Sodium 2-propenyl thiosulfate was also found to induce inhibition of the sulfurtransferase activity in tumor cells. Interestingly, in vitro experiments using fluorescence spectroscopy, kinetic studies and MS analysis showed that sodium 2-propenyl thiosulfate was able to bind the sulfur-free form of the rhodanese, inhibiting its thiosulfate:cyanide-sulfurtransferase activity by thiolation of the catalytic cysteine. The activity of the enzyme was restored by thioredoxin in a concentration-dependent and time-dependent manner. Our results suggest an important involvement of the essential thioredoxin-thioredoxin reductase system in cancer cell cytotoxicity by organo-sulfane sulfur compounds and highlight the correlation between apoptosis induced by these compounds and the damage to the mitochondrial enzymes involved in the repair of the Fe-S cluster and in the detoxification system.  相似文献

Fluorescence spectroscopy at 77 K showed that the application of glucose lead to the depletion of phycobilisomes (PBS) and photosystems (PS) 2 and 1, and that PS2 was more sensitive to glucose than PS1. The application of sodium thiosulfate, an effective scavenger of reactive oxygen intermediates, counteracted the effects of glucose. Sodium thiosulfate effectively protected photosynthetic apparatus, PS2, PS1, and PBS against glucose-induced depletion. Sodium thiosulfate showed strong capability to inhibit the disappearance of chlorophyll induced by glucose. At a relatively low concentration of glucose, the application of sodium thiosulfate can even be helpful for the assembly of photosynthetic apparatus. Hence the reactive oxygen species might be involved in the depletion of the photosynthetic apparatus in the cyanobacterium Synechocystis sp. PCC 6803 cells grown in the presence of glucose.  相似文献

Sodium 2-propenyl thiosulfate was identified in boiled garlic (Allium sativum). When canine erythrocytes were incubated with sodium 2-propenyl thiosulfate, the methemoglobin concentration and Heinz body percentage in erythrocytes were both increased, indicating that the compound induced oxidative damage in canine erythrocytes. Sulfamate is an analogue of thiosulfate, and the sodium and potassium salts of sulfamic acid inhibited the chemolithoautotrophic growth on thiosulfate of Acidithiobacillus ferrooxidans and Halothiobacillus neapolitanus. The chemo-organotrophic growth of Paracoccus versutus on sucrose was similarly inhibited by sulfamate. Thiosulfate oxidation by suspensions of H. neapolitanus was, however, unaffected by sulfamate, showing that sulfamate did not directly affect thiosulfate uptake, activation or oxidation. Inhibition of P. versutus was not relieved by cysteine and methionine, indicating that sulfate uptake and sulfur amino acid biosynthesis were not directly affected by sulfamate. Sulfamate was not degraded by any of the bacteria, and so could not serve as an alternative to thiosulfate as an energy-yielding substrate. Sulfamate is also an analogue of ammonia and might act like hydrazine by inhibiting ammonium uptake or an essential enzyme activity.  相似文献

Two enzymes containing thiosulfate sulfur transferase activity were purified fromChlorobium vibrioforme f.thiosulfatophilum by ion exchange chromatography, gel filtration and isoelectrofocusing. Enzyme I is a basic protein with an isoelectric point at pH 9.2 and has a molecular weight of 39,000. TheK m-values for thiosulfate and cyanide of the purified basic protein were 0.25 mM (thiosulfate) and 5 mM (cyanide). Enzyme II is an acidic protein. The enzyme has an isoelectric point at pH 4.6–4.7 and a molecular weight of 34,000. TheK m-values of the acidic protein were found to be 5 mM for thiosulfate and 125 mM for cyanide.In addition to thiosulfate sulfur transferase activity, cellfree extracts ofChlorobium vibrioforme f.thiosulfatophilum also contained low thiosulfate oxidase activity and negligible thiosulfate reductase activity. In the present work it was shown that biosynthesis of silver sulfide nanoparticles from silver nitrate and sodium thiosulfate solutions of millimolar concentration occurs efficiently by living Shewanella oneidensis MR-1 cells, as well as by ultrasonically-disrupted cells and by the membrane fraction of the cells. The size of nanoparticles synthesized in the presence of living cells was 7.8 ± 1.5 nm, while in the presence of ultrasonically-disrupted cells — it was 6.5 ± 2 nm. The shape of nanoparticles in both cases was close to spherical. It was also shown, that synthesis of nanoparticles occurs in a cell-free solution of sodium thiosulfate that has been incubated with cells previously and to which then a silver nitrate solution was added. In this case the nanoparticles were of elongated shape and their size was (11 ± 4) × (24 ± 6) nm. In the control experiment, when only silver nitrate and sodium thiosulfate solutions not incubated with cells were used, the nanoparticles were not detected. It was shown that biosynthesis of nanoparticles occurs both in aerobic and anaerobic conditions. Nanoparticles are not formed by using thermally inactivated cells as it was shown by us previously. The results show the important role of the native structures of cells for the nanoparticles formation.  相似文献

Histological and histochemical study of the pancreas of albino rats and experimental pancreatitis showed the use of sodium thiosulfate to considerably inhibit the progress of necrotic changes and circulatory disturbance. The preparation prevented recidivation of pancreatitis and inhibited sclerotic changes in the gland. Sodium thiosulfate stimulated the regenerative process including regenerative hypertrophy and expressed epimorphosis.  相似文献

【背景】沼气和天然气等清洁能源中往往会含有一定量的硫化氢,硫化氢的存在不仅污染环境,而且对人类危害很大。【目的】以硫代硫酸钠为唯一硫源从巴丹吉林沙漠盐碱湖岸边沉积物中分离筛选得到一株硫氧化菌BDL05,并研究其硫氧化特性。【方法】通过形态观察、生理生化特征及16S rRNA基因序列分析对硫氧化菌BDL05进行鉴定。【结果】菌株BDL05为革兰氏阴性菌,弧状,其16S rRNA基因序列与Thiomicrospira microaerophila ASL 8-2的相似性达99.8%,将其命名为Thiomicrospira microaerophila BDL05。该菌氧化硫代硫酸盐的最适pH为9.3,最适总钠盐浓度为0.8mol/L,在以硫化钠为硫源的气升式反应器中单质硫的生成率为94.7%,生成速率为3.0 mmol/(L·h)。【结论】菌株Thiomicrospira microaerophila BDL05为嗜盐嗜碱硫氧化菌,其耐盐耐碱性较强,比生长速率快,硫化钠氧化能力较强,是一株在气体生物脱硫方面具有应用价值的菌株。  相似文献

【目的】从典型自然环境中筛选耐盐高效硫氧化菌,研究其生长特性,并进行初步脱硫实验。【方法】以硫代硫酸钠为唯一能源底物的培养基富集脱硫菌,经过3次平板划线培养、纯种分离后得到纯种培养。经过革兰氏染色、平板菌落形态观察及形态学特征研究,并结合16S rRNA基因序列分析及分子系统发育树的构建结果,确定菌株的种类。【结果】从上海外高桥某发电厂冷却水池中筛选分离出一株硫代硫酸盐去除率高、耐盐性较强的细菌,命名为CYJN-1。该菌为革兰氏阴性菌,短杆状,鉴定为那不勒斯菌(Halothiobacillus neapolitanus)。H. neapolitanus CYJN-1具有较强适应盐度变化的能力,菌株生长的盐度范围为0?5% (NaCI,质量体积比)。菌株最适生长条件为:温度30 °C、pH 7.0、底物浓度为20 g/L。在此条件下,该菌对硫代硫酸钠的去除率可达98%。【结论】H. neapolitanus CYJN-1耐盐性较强,硫代硫酸盐去除率高,在生物脱硫、生物冶金等领域都具有潜在的应用前景。  相似文献

Intraperitoneal injection of acrylonitrile at 1.51-2.26 mmole/kg (80-120 mg/kg) or propionitrile at 0.54-1.51 mmole/kg (30-83 mg/kg) on the morning of Day 8 of gestation in the hamster induced exencephaly, encephalocoeles, and rib fusions and bifurcations in the offspring. These doses of the aliphatic nitriles also resulted in obvious toxicity to the dams. Multiple intraperitoneal injections of sodium thiosulfate at 4.03 mmole/kg (1 gm/kg) protected both dams and embryos against toxicity. When the larger doses of either acrylonitrile or propionitrile were given in the presence of sodium thiosulfate, teratogenic effects were observed in the absence of overt signs of maternal poisoning. A survey of the literature describes many studies which demonstrate that acrylonitrile and propionitrile are converted in vivo to toxicologically significant concentrations of cyanide and that sodium thiosulfate, an established cyanide antagonist, can provide protective actions against poisoning by either acrylonitrile or propionitrile. The observations suggest that the teratogenic effects of both acrylonitrile and propionitrile are related to the metabolic release of cyanide.  相似文献

The binding of sulfate, selenate and thiosulfate by the sulfur-transferase rhodanese (EC 2.8.1.1) in the crystalline state has been studied by X-ray analysis at resolutions between 0.23 nm and 0.4 nm. The three ions appear to occupy a common site between the N eta atoms of Arg-29 and the main-chain NH group of Glu-148 at the surface of the enzyme molecule. A second binding site for the three ions is situated at the entrance to the active centre, between the side chains of Arg-186 and Lys-249. Selenate and thiosulfate are bound equally well at both anion-binding sites. Sulfate, however, binds better at the first position, near Arg-29, than at the second site near Arg-186. In the complex of sulfur-rhodanese with thiosulfate, the outer sulfur atom of the anion near the active centre points towards the extra sulfur atom which is bound as a persulfide to the S gamma of the essential Cys-247. The distance between the outer sulfur atom of the thiosulfate ion and the persulfide sulfur atom appears to be about 0.3 nm. The thiosulfate difference Fourier also shows a distinct, localized conformational change involving residues 71, 72 and 249. This is the result of the replacement of an ammonium ion in the sulfate and selenate media by a sodium ion in the sodium thiosulfate solution. Rhodanese is apparently able to accomodate ions with different radii at this cation-binding site by minor structural alterations.  相似文献

Summary Amongst the family of the Enterobacteriaceae the ability to reduce tetrathionate to thiosulfate and thiosulfate to sulfite and sulfide occurs in the genera Proteus, Citrobacter and Salmonella. These reductions are coupled to a respiratory chain which functions under anaerobic conditions. Only during transport of electrons to tetrathionate oxidative phosphorylation has been demonstrated. Isolation and purification of the cytoplasmic membrane bound tetrathionate and thiosulfate reductase fromProteus mirabilis makes clear that this bacterium forms only one enzyme for both reductions. This enzyme has a molecular weight of 133,000 daltons and can be divided into two subunits with molecular weights of 43,000 and 90,000 daltons by treatment with sodium dodecyl sulfate and 2-mercaptoethanol. The reduction of tetrathionate is activated by its primary product thiosulfate. Nitrate or oxygen represses and inactivates the tetrathionate and thiosulfate reductase. Nevertheless the smaller subunit of this enzyme appears to be formed and assembled into the cytoplasmic membranes after anaerobic growth in the presence of nitrate. The capability of Phascolosoma arcuatum to detoxify sulfide in anaerobic conditions was examined. Sulfane sulfur, which underwent cold cyanolysis, was the major excretory end product of sulfide detoxification during anoxia. Thiosulfate was not excreted into the external medium. Instead, it was absorbed by P. arcuatum and its absorption was stimulated by the presence of sodium sulfide (Na2S) in the incubation medium. The effective formation and excretion of sulfane sulfur by P.␣arcuatum required the presence of both Na2S and sodium thiosulfate (Na2S2O3). Results obtained indicate that rhodanese might be involved in sulfide detoxification in this sipunculid. Rhodanese could act as a catalyst in the transfer of sulfur atoms from thiosulfate to HS. The body wall and the introvert were the main sites of sulfide detoxification. However, it is unlikely that epibiotic bacteria associated with the outside surface of the worm were involved in the detoxification process. A time-course study on the contents of thiosulfate and sulfane sulfur in the body wall of P. arcuatum incubated anaerobically in the presence of Na2S + Na2S2O3 verified that thiosulfate absorbed was utilized to detoxify sulfide to sulfane sulfur. A method is offered for he differential diagnosis of cancer cells. It depends on the use of methylene blue decolorized with sodium thiosulfate (denoted here HLM, i.e. “hyposulfite methylene blue”); this is prepared by dissolving 800 mg. sodium thiosulfate in 10 ml. of 0.1% aqueous methylene blue and adding 3-5 drops of dilute (1:3) HCl. Frozen sections are treated with this reagent for 2-3 minutes, rinsed with a large amount of distilled water, then stained 2-3 minutes with 0.05% aqueous acid fuchsin. Staining should be performed in a darkened room. If all due precautions are observed, normal tissue appears blue, malignant tissue red.  相似文献

In the longitudinal muscle strip of guinea pig ileum phenoxybenzamine (POB) produces a maximum parallel shift of 0.7 log units in the dose-response curve to histamine. In the presence of sodium thiosulfate in the wash fluid the parallel shift whith retention of maximum response increases to about 2 log units, and a similar value is obtained for Nethyl-N-(2-bromoethyl)-1-naphthylamine. The The agent N-ethyl-N- (2-chloroethyl)benzylamine produces a significantly smaller shift of dose-response curve of 1.53 log units before the maximum response becomes depressed. The receptor-specific depression of maximum response produced by higher doses of POB is reversed by sodium thiosulfate and by bovine serum albumin, while the parallel shift in dose-response curve is unaffected by both treatments. These findings may be explained by a hypothesis involving interaction of 2-haloalkylamines at two sites.  相似文献

A medium was devised for the rapid presumptive identification of Aeromonas hydrophila. It also offered good differentiation of Klebsiella, Proteus, and other enteric species. Mannitol fermentation, inositol fermentation, ornithine decarboxylation and deamination, indole production, motility, and H2S production from sodium thiosulfate and cysteine could be recorded in a single tube of the medium.  相似文献
