Summary A marked growth in the length of testes ofDrosophila hydei males occurred during pupal development. This growth continued over the first 8 days of adult life and in the young adults sperm were not produced until the testes increased approximately threefold in length to about 28 mm. The length of testes is correlated with genetic factors on the X and Y chromosomes. In males lacking a Y chromosome (X/O) or the short arm (YS) of the Y chromosome (X/YL) the testes were about half the length of testes of control males (X/Y) or double Y males (X/Y/Y). Males with deletions of the distal YL chromosome arm had testicular lengths equivalent to the controls. Males with short testes (X/O and X/YL) showed disruptions to spermatogenesis at meiosis and an absence of normal spermatid elongation. Reduction of active ribosomal RNA genes on the X chromosome in X/O caused an increased expression ofbobbed (bb) and a corresponding reduction in length of testes. Severelybobbed X/O males had very few cysts of spermatogonia and these cysts did not develop into primary spermatocytes.  相似文献

Four automatic substrate feeding strategies were developed and investigated in this study to obtain rapid, repeatable, and reliable high cell densities of Pseudomonas putida KT2440 from glucose. Growth yield data of the key nutrients, Y X/Glucose, Y X/NH4, Y X/PO4, Y X/Mg, and Y CO2/Glucose, were determined to be 0.41, 5.44, 13.70, 236, and 0.65 g g−1, respectively. Although standard exponential feeding strategy worked well when the predetermined μ was set at 0.25 h−1, an exponential glucose feeding strategy with online μ max estimation resulted in a higher average biomass productivity (3.4 vs 2.8 g l−1 h−1). A CO2 production rate based pulse glucose feeding strategy also resulted in good overall productivity (3.0 g l−1 h−1) and can be used as an alternative to pH-stat or DO-stat feeding. A cumulative CO2 production based continuous feed with real-time cumulative glucose consumption estimation resulted in much higher biomass productivity (4.3 g l−1 h−1) and appears to be an excellent and reliable approach to fully automating high-cell-density fed-batch cultivation of P. putida.  相似文献

From a study of radiation-induced X-chromosome deletions the locus of black body (b) has been localized to the proximal portion of C-band defined euchromatin. Radiation produced mostly X-chromosome deletions rather than point mutations, total X or Y chromosome loss through breakage, or increased frequency of non-disjunction. Aberrant sex ratios obtained indicate that the X chromosome carries vital loci that were deleted with b + in many cases. The X/O karyotype produces fertile adult females with a characteristic phenotype which is also produced by X deletions. Sex chromosome non-disjunction to give X/O females and X/X/Y males is normally rare but is enhanced by the presence of chromosome rearrangements even when the X and Y are not involved.  相似文献

Summary The w m Co duplication of Drosophila hydei (Dp (1; Y) 16B2-17B1) contains 13–16 bands in salivary gland chromosomes. The duplication resides preferentially in the X heterochromatin or on the Y chromosome. In some stocks frequent (up to 4×10-3) exchanges of the duplication occur between different Y chromosomes (T(X; Y) and free Y) or between the X and the Y chromosome. About 60% of the T(X; Y)-Y exchanges induce mutations in the Y chromosomal male fertility genes of the recipient Y chromosome. From the mutational spectrum generated by the T(X; Y)-Y transpositions and from the variable efficiency as acceptor of different X-Y translocations it can be concluded that the exchanges show a remarkable site specificity: distal positions in the long arm of the Y chromosome are occupied preferentially. More proximal positions in the long arm of insertions into the short arm of the Y chromosome are found only with a lower frequency. No transpositions to the autosomes have been recovered. Duplications are lost with highly differing frequencies. The losses are not linked with insertions of the w m Co element into a new position and are more frequent than transpositions. Therefore, we regard the w m Co element as a giant transposon.  相似文献

Karyotype and genetic variation of the common shrew (Sorex araneus) from Mt. Pelister in southern Macedonia has been studied. Whereas all autosomes in the chromosomal set (2 na, = 28, the only present metacentrics being af, bc, jl, and tu) were of the standard type as well as the sex chromosomes X and Y2 in males, the Y1 chromosome was a small metacentric. This chromosomal feature is unique among the common shrew populations studied cytogenetically so far. Three out of 33 loci analysed (Sdh, 6Pgd, Mdh-1) were discriminant between the Mt. Pelister population and Sorex araneus from Slovenia and two loci (Est-3, Ada) were partially discriminant. A relatively high value of Nei's genetic distance (D = 0.137) confirms unique character of the Pelister population.  相似文献

This paper deals with the relative growth of three different fruit tissues. Their morphogenetic periods and the mathematical constraints involved are described, and more precisely, the paper shows an allometric relationship (Y=nX m ) between the widths (X, Y) of the main tissues in stone fruits such as cherries, peaches and prunes. The mathematical relationships between the growth of the mesocarp and of the endocarp of somePrunus fruits are described, and it is proved that before the formation of the embryo, growth is allometric, in agreement with conclusions drawn from some experimental data. However, according to another study, the growth of the mesocarp and of the endocarp are ruled by autocatalytic and monomolecular functions, before as well as after the formation of the embryo. In this case, it is proved that if allometry exits in stone fruits, it can only be anantiometry (m=−1). To solve the dilemma, two main alternatives are proposed and discussed. We conclude that, while allometry is established on reasonable grounds before the formation of the embryo, after the formation of the embryo the mesocarp and endocarp evolve independently since a center for the coordination of growth no longer exists, and each tissue can grow according to its own independent rules.  相似文献

Using experimental data from continuous cultures of Clostridium acetobutylicum with and without biomass recycle, relationships between product formation, growth and energetic parameters were explored, developed and tested. For glucose-limited cultures the maintenance models for, the Y ATP and biomass yield on glucose, and were found valid, as well as the following relationships between the butanol (Y B/G) or butyrate (Y BE/G) yields and the ATP ratio (R ATP, an energetic parameter), Y B/G =0.82-1.35 R ATP, Y BE/G =0.54 + 1.90 R ATP. For non-glucose-limited cultures the following correlations were developed, Y B/G =0.57-1.07 , Y B/G =0.82-1.35 R ATPATP and similar equations for the ethanol yield. All these expressions are valid with and without biomass recycle, and independently of glucose feed or residual concentrations, biomass and product concentrations. Summary The frequencies of disjunction types in double-translocation heterozygous males (262Y626XY2) in Hylemya antiqua have been established in MII cells and eggs of testcrosses.Several disjunction types occurred but four predominated. A correlation was found between the frequencies of the disjunction types and the relative position of the centromeres. The frequency of numerical non-disjunction (NND) was 4%. Differences in frequency of NND between sex-linked and autosomal translocations of H. antiqua are discussed. A good correspondence between the frequencies of unbalanced karyotypes, and embryonic and larval mortality was found. The total genetic load which can be induced by the T14/T61 males is estimated to be 60–65%. Some duplication/deficiency karyotypes appeared to be viable in pupal and even adult stages. In 26262Y6262X males a regular coorientation between 2Y and X was observed, in spite of non-homologous centromeres and a complicated synapsis of 2Y. Application possibilities of the present material for genetic control of H. antiqua are discussed.  相似文献

Summary The yeast-like organism Aureobasidium pullulans efficiently converted abetd-xylose to cell mass (Y X/S=0.45 g·g–1) with negligible production of polyols (Y P/S=0.003 g·g–1) under aerobic conditions. A. pullulans grown semiaerobically exhibited different fermentation capacities in seven basal (vitaminless) medium and medium containing a mixture of seven vitamins. It was found that under semiaerobic conditions a mixture of vitamins significantly enhanced production of ethanol from abetd-xylose, resulting in a 15-fold higher yield coefficient of ethanol (Y E/S=0.22 g·g–1) as compared to that achieved in vitaminless medium. This increase in ethanol production was accomplished at the expense of cell mass. A. pullulans produced extremely low amounts of polyols throughout all aerobic and semiaerobic experiments. A. pullulans displayed strictly NADPH-linked xylose reductase and NAD+-linked xylitol dehydrogenase activities.  相似文献

Age and growth of pearly razorfish Xyrichtys novacula (Labridae) were determined in specimens caught on sandy bottoms by surrounding nets and lines (depth range 5–30 m) between January 2000 and December 2002. The study location was in the central Mediterranean Sea. For this purpose the annual growth increments (annuli) in sagittae were analysed. A total of 470 individuals of X. novacula, ranging from 49 to 200 mm TL (total length), were examined. Conversion between total length and standard length was calculated and represented by the relationship: . Edge analysis on otoliths was carried out to validate seasonality deposition. Seven age‐classes were determined from 0+ to 6+. The linear relationship between maximum otolith length (OL) and TL was summarized in the equation . The estimated parameters of the von Bertalanffy growth function were L = 175.05 mm, k = 0.80 year?1, t0 =?0.73 years and the growth performance index calculated as (Φ = 2.39). The length–weight relationship W = 0.0139 TL2.9326 described an isometric growth for the species in this area.  相似文献

Hybrid zones are geographic regions where isolating barriers between divergent populations are challenged by admixture. Identifying factors that facilitate or inhibit hybridization in sympatry can illuminate the processes that maintain those reproductive barriers. We analysed patterns of hybridization and phenotypic variation across two newly discovered hybrid zones between three subspecies of barn swallow (Hirundo rustica). These subspecies differ in ventral coloration and wing length, traits that are targets of sexual and natural selection, respectively, and are associated with genome‐wide differentiation in allopatry. We tested the hypothesis that the degree of divergence in these traits is associated with the extent of hybridization in secondary contact. We applied measures of population structure based on >23,000 SNPs to confirm that named subspecies correspond to distinct genomic clusters, and assessed coincidence between geographic clines for ancestry and phenotype. Although gene flow was ongoing across both hybrid zones and pairwise FST between subspecies was extremely low, we found striking differences in the extent of hybridization. In the more phenotypically differentiated subspecies pair, clines for ancestry, wing length and ventral coloration were steep and coincident, suggestive of strong isolation and, potentially, selection associated with phenotype. In the less phenotypically differentiated pair, gene flow and phenotypic variation occurred over a wide geographic span, indicative of weaker isolation. Traits associated with genome‐wide differentiation in allopatry may thus also contribute to isolation in sympatry. We discuss potentially important additional roles for evolutionary history and ecology in shaping variation in the extent hybridization between closely related pairs of subspecies.  相似文献

Stripe rust (caused by Puccinia striiformis) occurs annually in most wheat-growing areas of the world. Thinopyrum ponticum has provided novel rust resistance genes to protect wheat from this fungal disease. Wheat – Th. ponticum partial amphiploid line 7430 and a substitution line X005 developed from crosses between wheat and 7430 were resistant to stripe rust isolates from China. Genomic in situ hybridization (GISH) analysis using Pseudoroegneria spicata genomic DNA as a probe demonstrated that the partial amphiploid line 7430 contained ten Js and six J genome chromosomes, and line X005 had a pair of Js-chromosomes. Giemsa-C banding further revealed that both lines 7430 and X005 were absent of wheat chromosomes 6B. The EST based PCR confirmed that the introduced Js chromosomes belonging to linkage group 6, indicating that line X005 was a 6Js/6B substitution line. Both resistance observation and sequence characterized amplified region (SCAR) markers displayed that the introduced chromosomes 6Js were responsible for the stripe rust resistances. Therefore, lines 7430 and X005 can be used as a donor in wheat breeding for stripe rust resistance.  相似文献

Summary In first and second instar larvae of Drosophila melanogaster mitotic recombination has been induced by X-rays. Resulting mosaic spots were analysed in the eyes of adult females. The five examined genotypes varied in the combination of different X-chromosomes and in the presence or absence of the duplication Dp(1;3)N 264-58. X-chromosomes used have been elongated by proximally or distally linked arms of the Y-chromosome. The portion of Y-heterochromatin in the genome is negatively correlated with the frequency of twin mosaic spots (Fig. 1). The frequency of these spots is lower in flies bearing the duplication than in those without the duplication. With respect to the X-chromosome combinations, there are no marked differences in the frequencies of single mosaic spots (Fig. 2); with respect to the duplication they are absent. In the genotype Y S X/X.Y S prophases of neuroblast mitoses (Fig. 4) show normal pairing of homologous chromosome sections. Reasons for the different spot frequencies are discussed.

The morphological, yield, cytological and molecular characteristics of bread wheat X tritordeum F1 hybrids (2n =6x = 42; AABBDHch) and their parents were analysed. Morphologically, these hybrids resembled the wheat parent. They were slightly bigger than both parents, had more spikelets per spike, and tillered more profusely. The hybrids are self-fertile but a reduction of average values of yield parameters was observed. For the cytological approach we used a double-target fluorescencein situ hybridization performed with total genomic DNA fromHordeum chilense L. and the ribosomal sequence pTa71. This technique allowed us to confirm the hybrid nature and to analyse chromosome pairing in this material. Our results showed that the expected complete homologous pairing (14 bivalents plus 14 univalents) was only observed in 9.59% of the pollen mother cells (PMCs) analysed. Some PMCs presented autosyndetic pairing of Hch and A, B or D chromosomes. The average number of univalents was higher in the wheat genome (6.8) than in the Hch genome (5.4). The maximum number of univalents per PMC was 20. We only observed wheat multivalents (one per PMC) but the frequency of trivalents (0.08) was higher than that of quadrivalents (0.058). We amplified 50 RAPD bands polymorphic between the F1 hybrid and one of its parents, and 31 ISSR polymorphic bands. Both sets of markers proved to be reliable for DNA fingerprinting. The complementary use of morphological and yield analysis, molecular cytogenetic techniques and molecular markers allowed a more accurate evaluation and characterization of the hybrids analysed here.  相似文献

Hybrid sterility is a common postzygotic reproductive isolation mechanism that appears in the early stages of speciation of various organisms. Mus musculus musculus and Mus musculus domesticus represent two recently separated mouse subspecies particularly suitable for genetic studies of hybrid sterility. Here we show that the introgression of Chr X of M. m. musculus origin (PWD/Ph inbred strain, henceforth PWD) into the genetic background of the C57BL/6J (henceforth B6) inbred strain (predominantly of M. m. domesticus origin) causes male sterility. The X-linked hybrid sterility is associated with reduced testes weight, lower sperm count, and morphological abnormalities of sperm heads. The analysis of recombinant Chr Xs in sterile and fertile males as well as quantitative trait locus (QTL) analysis of several fertility parameters revealed an oligogenic nature of the X-linked hybrid sterility. The Hstx1 locus responsible for male sterility was mapped near DXMit119 in the central part of Chr X. To ensure full sterility, the PWD allele of Hstx1 has to be supported with the PWD allelic form of loci in at least one proximal and/or one distal region of Chr X. Mapping and cloning of Hstx1 and other genes responsible for sterility of B6–XPWDYB6 males could help to elucidate the special role of Chr X in hybrid sterility and consequently in speciation.  相似文献

Summary Fermentation yields of Lactobacillus plantarum were measured at controlled pH between 4.0 and 8.0 and initial lactate concentrations of 0–90 g/l. Optimal growth conditions at pH 6.0 without addition of lactate gave a growth rate of 0.57 h–1 and 20 g dry biomass/mol ATP formed (Y ATP). The pH variations resulted in a decrease in growth rate but the effect on Y ATPwas insignificant. The addition of lactate to the medium at 0 h resulted in linear decrease in the growth rate of the culture, and all the metabolic activities were completely inhibited at 110 g/l. The Y ATPand biomass/ substrate yield (Y X/S) remained fairly steady up to 33 g lactate/l, beyond which both yields decreased considerably. Offsprint requests to: M. Raimbault  相似文献

Therian X and Y sex chromosomes arose from a pair of autosomes. Y chromosomes consist of a pseudoautosomal region that crosses over with the X chromosome and a male‐specific Y‐chromosomal region that does not. The X chromosome can be structured into “evolutionary strata”. Divergence of X‐chromosomal genes from their gametologs is similar within a stratum, but differs among strata, likely caused by a different onset of suppression of crossing over between gametologs. After stratum formation, exchange of information between gametologs has long been believed absent; however, recent studies have shown limited exchange, likely through gene conversion. Herein we investigate exchange of genetic information between gametologs in old strata that formed before the split of Laurasiatheria (cattle) from Euarchontoglires (primates and rodents) with a new phylogenetic approach. A prerequisite for our test is an overall preradiative topology, that is, all X‐chromosomal gametologs are more similar among themselves than to Y‐chromosomal sequences. Screening multiple sequence alignments of the coding sequences of genes from cattle, mice, and humans identified four genes, DDX3X/Y, RBMX/Y, USP9X/Y, and UTX/Y, exhibiting a preradiation topology. Applying our test, we detected exchange of genetic information between all four X and Y gametologs after stratum formation.  相似文献
