Aims: Non‐sigmoid growth curves of Escherichia coli obtained at constant temperatures near the maximum growth temperature (Tmax) were previously explained by the coexistence of two subpopulations, i.e. a stress‐sensitive and a stress‐resistant subpopulation. Mathematical simulations with a heterogeneous model support this hypothesis for static experiments at 45°C. In this article, the behaviour of E. coli, when subjected to a linearly increasing temperature crossing Tmax, is studied. Methods and Results: Subpopulation dynamics are studied by culturing E. coli K12 MG1655 in brain heart infusion broth in a bioreactor. The slowly increasing temperature (°C h?1) starting from 42°C results in growth up to 60°C, a temperature significantly higher than the known Tmax. Given some additional presumptions, mathematical simulations with the heterogeneous model can describe the dynamic experiments rather well. Conclusions: This study further confirms the existence of a stress‐resistant subpopulation and reveals the unexpected growth of E. coli at temperatures significantly higher than Tmax. Significance and Impact of the Study: The growth of the small stress‐resistant subpopulation at unexpectedly high temperatures asks for a revision of currently applied models in food safety and food quality strategies.  相似文献

In Escherichia coli membranes, three major phospholipids are formed: phosphatidylethanolamine (PE), phosphatidylglycerol (PG) and cardiolipin (CL). We report here the survival of mutants lacking either PE or both PG and CL at an acid pHo and during long-term survival experiments. Stationary phase cultures of E. coli lacking PE are much more sensitive to acid shock (pHo 3) than the wild-type strain. Moreover, in the strain lacking PE, long-term survival in stationary phase is impaired and after 5 days no viable cells are recovered. The survival of an exponential phase culture to acid shock is known to be increased if the culture is exposed to moderately acid conditions (pHo 5) prior to a shift to pHo 3. If either PE or both PG and CL are missing, the exposure to pHo 5 does not increase the survival at pHo 3.  相似文献

Aims: The dynamics of Escherichia coli near the maximum temperature for growth in a rich medium are analysed. The effects of temperature history, medium composition and physiological state of the inoculum are evaluated. Methods and Results: Kinetics of E. coli K12 MG1655 is studied in ‘brain–heart infusion’ broth in a temperature controlled environment. Based on viable counts, ‘smooth’ growth curves are observed at 40, 41, 42 and 43°C. The exponential growth phase at 44 and 45°C is interrupted. At 46°C, a period of exponential growth is followed by inactivation. Neither the physiological state of the inoculum nor medium enrichment alters the dynamics, whilst temperature pre‐adaptation or chemical chaperones restore regular cell growth and division (‘smooth’ exponential growth). Conclusions: Atypical, nonexponential growth at 44, 45 and 46°C seems related to protein destabilization and can (partly) be restored by an appropriate medium design (i.e. addition of chemical chaperones) or temperature history (i.e. selection of a more resistant subpopulation). Significance and Impact of the Study: This study indicates that the maximum temperature for growth is dependent on the temperature history and the chemical environment. These observations and the nonexponential kinetics have important implications for the development of predictive models for food safety and quality.  相似文献

The OmpF and OmpC porins are major outer membrane proteins of Escherichia coli. Their expression is affected by medium osmolarity such that OmpF is normally produced at low osmolarity and OmpC at high osmolarity. Potassium ion accumulation is a major means by which cells maintain their internal osmolarity in high osmolarity medium in the absence of organic osmolytes such as glycine-betaine. Starvation for potassium causes cells to become turgor stressed. The effect of turgor stress and potassium ion concentration on OmpF and OmpC expression was examined. It was found that ompF gene expression was switched off by turgor stress but there was no concomitant increase in OmpC. Instead, ompC expression responded to the accumulation of potassium ions by the cell in high osmolarity medium.  相似文献

Aims: Resistance to acidic stress contributes to bacterial persistence in the host and is thought to promote their passage through the human gastric barrier. The aim of this study was to examine whether nucleosides have a role in the survival under acidic conditions in Escherichia coli. Methods and Results: We found that adenosine has a function to survive against extremely acidic stress. The deletion of add encoding adenosine deaminase that converts adenosine into inosine and NH3 attenuated the survival in the presence of adenosine. The addition of adenosine increased intracellular pH of E. coli cells in pH 2·5 medium. Addition of inosine or adenine did not increase the resistance to acidic conditions. Conclusions: Our present results imply that adenosine was used to survive under extremely acidic conditions via the production of NH3. Significance and Impact of the Study: It has been proposed that amino acid decarboxylation is the major system for the resistance of E. coli to acidic stress. In this study, the adenosine deamination was shown to induce the survival under acidic conditions, demonstrating that bacteria have alternative strategies to survive under acidic conditions besides amino acid decarboxylation.  相似文献

This study uses the enteric bacterium Escherichia coli as an experimental system to examine evolutionary responses of bacteria to an environmental acidic-alkaline range between pH 5.3 and 7.8 (15-5000 nM [H(+)]). Our goal was both to test general hypotheses about adaptation to abiotic variables and to provide insights into how coliform organisms might respond to changing conditions inside and outside of hosts. Six replicate lines of E. coli evolved for 2000 generations at one of four different constant pH conditions: pH 5.3, 6.3, 7.0, or 7.8. Direct adaptation to the evolutionary environment, as well as correlated changes in other environments, was measured as a change in fitness relative to the ancestor in direct competition experiments. The pH 5.3 group had the highest fitness gains, with a highly significant increase of 20%. The pH 7.8 group had far less significant gains and much higher variance among its lines. Analysis of individual lines within these two groups revealed complex patterns of adaptation: all of the pH 5.3 lines exhibited trade-offs (reduced fitness in another environment), but only 33% of the pH 7.8 lines showed such trade-offs and one of the pH 7.8 lines demonstrated exaptation by improving fitness in the pH 5.3 environment. Although there was also prevalent exaptation in other groups to the acidic environment, there were no such cases of exaptation to alkalinity. Comparison across the entire experimental pH range revealed that the most acidic lines, the pH 5.3 group, were all specialists, in contrast to the pH 6.3 lines, which were almost all generalists. That is, although none of the pH 5.3 lines showed any correlated fitness gains, all of the pH 6.3 lines did.  相似文献

The objective of this study was to investigate the influence of stressful growth conditions on the fitness cost of antimicrobial resistance in Escherichia coli BJ4 caused by chromosomal mutations and plasmid acquisition. The fitness cost of chromosomal streptomycin resistance increased significantly when the bacteria were grown under all stress conditions tested, while the cost in 1/3 Luria–Bertani was not significantly changed in a streptomycin+rifampicin mutant. The increase in the fitness cost depended in a nonregular manner on the strain/stress combination. The fitness cost of plasmid-encoded resistance on R751 did not differ significantly, and was generally less under stressful growth conditions than in rich media. The fitness cost associated with R751 with the multiple drug resistance cassette from Salmonella Typhimurium DT104 increased significantly only under stressful conditions at low pH and at high-salt concentrations. Strains with an impaired rpoS demonstrated a reduced fitness only during growth in a high-salt concentration. In conclusion, it was demonstrated that bacterial fitness cost in association with antimicrobial resistance generally increases under stressful growth conditions. However, the growth potential of bacteria with antimicrobial resistances did not increase in a straightforward manner in these in vitro experiments and is therefore probably even more difficult to predict in vivo .  相似文献

In contrast to enzymatic adaptation, osmotic adaption is possible with T4-infected Escherichia coli B cells. After an osmotic shift from 220 mOsM to 690 mOsM the intracellular content of potassium rises in infected cells as well as in uninfected cells. Details of the mechanism for ribosome synthesis have been incorporated in the single-cell Escherichia coli model, which enable us to predict the amount of protein synthesizing machinery under different environmental conditions. The predictions agree quite well with available experimental data. The model predicts that ribosomal protein limitations are important when the translational apparatus is in high demand. Ribosomal RNA synthesis is induced by an increase in translational activity, which, in turn, stimulates ribosomal protein synthesis. However, as the demand increases still more, the ribosomal protein mRNA must compete with the plasmid mRNA for ribosomes, and the efficiency of translation of ribosomal proteins is reduced. AIMS: To investigate the requirement of outer membrane porins for osmotic adaptation at alkaline pH in Escherichia coli. METHODS AND RESULTS: Escherichia coli mutants deficient in ompC, ompF and both genes were constructed and the growth of these mutants was observed at alkaline pH. The growth rate of the mutant deficient in both ompC and ompF was slower than that of the wild type and mutants deficient in one of these genes under hyperosmotic stress at pHs above 8.0. The decreased rate was recovered when a cloned ompC was introduced to the mutant, but the growth recovery with a cloned ompF was partial. Such growth diminution was not observed at pHs below 8.0. CONCLUSION: OmpC and OmpF were shown to participate in hyperosmotic adaptation at alkaline pH in E. coli. SIGNIFICANCE AND IMPACT OF THE STUDY: This study is the first report to demonstrate that OmpC and OmpF are required for hyperosmotic adaptation at pHs above 8.0, but not below 8.0.  相似文献

AIMS: To investigate how carbon dioxide affects the acid resistance of Escherichia coli. METHODS AND RESULTS: Escherichia coli W3110 was grown in minimal EG medium at pH 7.5, and cells were adapted at pH 5.5 at 37 degrees C with and without supply of carbon dioxide and nitrogen gases. The number of colonies grown on LB medium was measured after cells were challenged in minimal EG medium of pH 2.5 at 37 degrees C under various conditions. When carbon dioxide was supplied at both the acid adaptation and challenge stages, 94% of cells survived after the acid challenge for 1 h, while the survival rates were 50 and 67% when nitrogen gas and glutamate were supplied respectively. After the acid challenge for 3 h, the survival rate observed with the carbon dioxide gas supply was again 2.5-fold higher than those with the nitrogen gas supply. CONCLUSION: Carbon dioxide was shown to participate in the maintenance of high viability under acidic conditions. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides useful information for research into bacterial pathogenesis, fermentation and food preservation.  相似文献

Aims:  Previous work showed that the exponential phase of Escherichia coli K12 MG1655, grown in Brain Heart Infusion broth at temperatures close to its maximum growth temperature, is disturbed. Based on plate count data, microscopic images and literature, the existence of a heat-resistant subpopulation was hypothesized. Here, this hypothesis is mathematically explored via a heterogeneous model.
Methods and Results:  A heat-sensitive and a heat-resistant subpopulation are considered. A large fraction of the population is inactivated, while the remaining smaller fraction is able to resist (or adapt to) the inimical temperature and grows. A heterogeneous model that encloses a growth model (resistant population) and an inactivation model (sensitive population) is used to describe the global population dynamics. Most experimental data can be predicted when taking parameter uncertainty via Monte Carlo simulation into account.
Conclusions:  The heterogeneous model accurately describes disturbed growth curves at superoptimal temperatures, except for high initial cell densities.
Significance and Impact of the Study:  This study strengthens the hypothesis of the existence of a (small) heat-resistant subpopulation in typical inoculum cultures of E. coli K12 MG1655.  相似文献

Twenty Escherichia coli strains producing well-characterised colicins were tested for their inhibitory activity against five Shiga toxin-producing E. coli (STEC) strains using different media under aerobic and anaerobic conditions. The five STEC strains used were of serotype O26, O111, O128, O145 and O157:H7 which are frequently isolated serotypes associated with disease in humans. The main route of infection for humans is through the eating of badly cooked or handled beef. The major reservoir for STEC strains in cattle is the rumen. To mimic the situation in the rumen of cattle, overlay assays were also performed under anaerobic conditions in the presence of 30% rumen fluid. Colicins E1, E4, E8-J, K and S4 are most active against STEC strains under anaerobic conditions in the absence or presence of rumen fluid. These colicins will be used in future experiments with the aim to eradicate the presence of STEC in cattle.  相似文献

为了提高重组大肠杆菌FB讲/pSV-04发酵生产L-色氨酸的产量,减少代谢副产物乙酸的生成,考察了比生长速率和无机盐对重组大肠杆菌发酵生产L.色氨酸的影响。在确定了合适的比生长速率和无机盐浓度之后,乙酸积累很少,L-色氨酸的产量为53.4g/L,比优化前提高了141.6%。经30L发酵罐初步放大,L-色氨酸的产量达53.6g/L,发酵结果稳定,具有工业化应用前景。  相似文献

AIMS: To predict and validate survival of non-acid adapted Escherichia coli O157 in an environment mimicking the human stomach. METHODS AND RESULTS: Survival was predicted mathematically from inactivation rates at various, but constant pH values. Predictions were subsequently validated experimentally in a pH-controlled fermentor. Contrary to prediction, acid-sensitive cultures of E. coli O157 survived for a long period of time and died as rapidly as acid-resistant cultures. Experimental results showed that in an environment with changing pH, acid-sensitive cultures became acid-resistant within 17 min. Cyclo fatty acids was reported to be a factor in acid resistance. As synthesis of cyclo fatty acids does not require de novo enzyme synthesis and thus requires little time to develop, we analysed the membrane fatty acid composition of E. coli O157 during adaptation. No changes in membrane fatty acid composition were observed. CONCLUSIONS: Acid adaptation of E. coli O157 can occur during passage of the human gastric acid barrier, which can take up to 4 h. SIGNIFICANCE AND IMPACT OF THE STUDY: The ability of acid-adapted bacteria to survive the human stomach is an important virulence factor. The ability of non-acid adapted E. coli O157 to adapt within a very short period of time under extreme conditions further contributes to the virulence of E. coli O157.  相似文献

Two metabolically engineered E. coli strains HL2765k and HL27659k, while capable of producing succinate from glucose with high yields, are not able to grow and produce succinate on sucrose. Consequently, the pUR400 plasmid containing scrK, Y, A, B, and R genes was introduced into HL2765k and HL27659k, respectively. Shake flask culture studies showed that the resulting strains can utilize sucrose; the strain HL2765k pUR400 and HL27659k pUR400 can produce succinate aerobically with a molar yield of 0.78 ± 0.02 mol/mol and 1.35 ± 0.13 mol/mol, respectively. On introduction of the plasmid pHL413, which encodes the heterologous pyruvate carboxylase (PYC) from Lactococcus lactis, the molar succinate yield increased to 1.60 ± 0.01 mol of succinate per mole of sucrose by the HL2765k pUR400 pHL413 strain and to 1.84 ± 0.10 by the HL27659k pUR400 pHL413 strain. In aerobic batch bioreactor studies, the succinate production rate was faster, and succinate production reached 101.83 mM with a yield of 1.90 when dissolved oxygen (DO) was controlled at 40 ± 7%. In addition, the results showed that DO had an important effect on succinate production by influencing PYC activity. This work demonstrates the possibility of producing succinate aerobically using sucrose as the carbon source.  相似文献

目的了解杭州市第一人民医院产ESBLs大肠埃希菌的发生比例及对临床上常用的24种抗菌药物的耐药率变化.方法收集2003 2004年该院各类临床标本中分离的大肠埃希菌,采用NCCLS推荐的表型确证试验方法检测ESBLs菌株;药敏试验采用纸片扩散法.结果2003年与2004年,产ESBLs的大肠埃希菌分离率分别为46.11%(184/399)、57.44%(386/672)(P=0.0003);2年来,ESBLs阳性菌对临床常用的24种药物表现出较高的耐药性,耐药率上升非常显著(P=0.0005);非产ESBLs大肠埃希菌对大多数抗菌药物仍保持较高的敏感率;但2004年ESBLs阴性的大肠埃希菌的耐药性,比2003年显著上升(χ^2=37.785,P=0.0005).结论尽早开展产ESBLs菌的监测,合理使用抗菌药物,对于有效控制产ESBLs菌的播散与流行是一项重要措施.  相似文献

Aims:  This study aimed to determine the survival of Escherichia coli strains during steam and lactic acid decontamination interventions currently used by the beef‐processing industry, and to determine their heat resistance. Methods and Results:  Strains were grouped into cocktails of five strains each differing in their RAPD patterns for subsequent identification. Steam and lactic acid treatments on meat reduced cell counts of E. coli strain cocktails by 90–99%. The 20 slaughter plant isolates exhibited only minor variation in their resistance to steam and lactic acid treatments but were more resistant than reference strains (three strains) or isolates from live cattle (seven strains). D60 values of strains from live cattle, and reference strains ranged from 0·1 to 0·5 min, in keeping with literature data. However, D60 values of current slaughter plant isolates ranged between 15 for E. coli DM18.3 and 71 min AW 1.7. Cell counts of E. coli AW 1.7 were reduced by <5 log10 CFU g?1 in ground beef patties cooked to an internal temperature of 71°C. Conclusions:  Strains of E. coli that survive cooking of ground beef to the recommended internal temperature of 71°C can be isolated from beef‐processing facilities. Significance and Impact of the Study:  Pathogen interventions in current commercial beef slaughter may select for extremely heat‐resistant strains of E. coli.  相似文献
