Our work aims at the exploration of cortisol secretion in the Bedouin goat, native to the Algerian Sahara desert, to understand the mechanisms of adaptation to extreme hot climates. In the present study, diurnal and seasonal variations of cortisol concentrations were measured in basal conditions, as well as the response to ACTH stimulation tests across seasons in bucks. The plasma concentrations of cortisol showed no diurnal cycle but a large variation across seasons. The highest levels occurred in summer and winter when the environmental conditions are at their extreme levels. The rectal temperature showed nychthemeral and seasonal variations, and BW was also different across seasons with highest values in summer and lowest in winter. The results obtained after administration of two doses (2 or 10 μg/kg BW) of synthetic ACTH to three different age groups (kids, adults and elderly animals) showed a strong increase in plasma cortisol concentrations under all conditions with maximum levels achieved between 15 and 120 min. The analysis of the area under the cortisol curve showed no significant difference between the responses to the two doses of ACTH and between age groups, but showed seasonal variations with the lowest response in autumn than in other seasons. We conclude that season significantly affects secretion of cortisol in both basal state and under ACTH stimulation. However, the variation of adrenal reactivity to ACTH is not sufficient to explain seasonal differences, and in particular the summer peak in basal circulating cortisol concentrations. Further research should focus on the respective contribution of environmental factors (such as day length, temperature, humidity) and the mechanisms involved in cortisol regulation.  相似文献

Climate change has intensified the frequency of heat waves in the world, thereby exposing farm animals to stressful conditions. For better productive performance it is important to identify the most resilient genotypes. Thus, our objective was to evaluate the thermoregulatory responses of rams of tropical indigenous (Morada Nova and Santa Inês) and exotic breeds (Dorper and Texel), by monitoring the environmental and physiological indicators related to heat tolerance. The experiment was carried out in a tropical climate region (Cwa), in Brazil, for twelve months, which comprised spring, summer, autumn and winter. Thirty-three rams were divided into groups: Morada Nova (MN; n=8, red-coat), Santa Inês (SI; n=9, black-coat), Dorper (DO; n=8, white-coat) and Texel (TX; n=8, white-coat). The microclimatic variables were monitored, and the THI and BGHI comfort indices were calculated. Coat thickness and body surface temperatures were measured monthly, and serum triiodothyronine-T3 measurements and complete blood tests were performed. The physiological variables were evaluated every fifteen days and skin micro-biopsies were performed in the summer and winter for histological evaluation. During the warmer seasons, the THI and BGHI reached values that indicated thermal discomfort. TX showed higher coat thickness throughout the year, increased physiological variables related to thermolysis, and reduced T3 (P < 0.05). The internal temperature was permanently lower in the MN, SI and DO (P < 0.05). The body surface temperatures were affected by the coat characteristics and wool length. Hematological parameters varied in the seasons with the highest thermal conditions (P < 0.05). The MN showed larger sweat glands, while the area occupied by the sweat glands was higher in the SI. The DO showed higher hair density in the summer and winter (P < 0.05). The results indicated that the MN, SI and DO breeds overcome the thermal challenge more easily throughout the seasons due to specific adaptive morphological and physiological characteristics.  相似文献

表观重编程异常是核移植胚胎发育异常的重要原因。为了研究克隆山羊胎儿不同组织中H19基因CpG岛甲基化水平和相对表达量,本实验运用亚硫酸盐法和荧光实时定量PCR法分别检测了死亡克隆山羊胎儿和同期普通山羊胎儿(对照组)肝脏、胎盘、肾脏、肺脏和心脏组织中H19基因CpG岛甲基化水平和mRNA的相对表达量。结果表明,克隆山羊胎儿胎盘组织中H19基因第5个CpG岛的甲基化水平显著高于对照组(70%vs49.41%,P0.05),H19基因相对表达量显著低于对照组(883.3vs1264.5,P0.05);肺脏组织甲基化水平显著低于对照组(63.53%vs88.24%,P0.05),相对表达量显著高于对照组(1003.4vs515.5,P0.05);其他各组差异不显著(P0.05)。结果说明,H19基因在克隆山羊胎儿部分组织中DNA甲基化重编程异常,而且这种异常影响H19基因的正常表达,这也可能是导致克隆动物死亡的重要因素之一。  相似文献

A study was conducted to assess the impact of summer season-related heat stress and nutritional stress individually as well as simultaneously on the adaptive capability of goats. Twenty four adult Osmanabadi bucks (average body weight (BW) 16.0 kg) were divided into four groups, C (n = 6; control), HS (n = 6; heat stress), NS (n = 6; nutritional stress) and CS (n = 6; combined stress). The study was conducted for a period of 45 days. The NS and CS bucks were under restricted feed to induce nutritional stress. The HS and CS bucks were exposed to summer heat stress. The animals exhibited different physiological adaptive behavior in the morning and afternoon. Further, the higher plasma cortisol (p < 0.01) and aldosterone (p < 0.05) was recorded in CS group as compared to other groups. The highest degree of degenerative changes and hyperactivity of endocrine cells was recorded in CS group liver and adrenal gland respectively. The higher expression of adrenal and hepatic Heat Shock Protein 70 (HSP70)messenger ribonucleic acid (mRNA) was reported in CS and HS goats respectively. It can be concluded from this study that plasma cortisol and adrenal HSP70 gene expression may be considered as ideal biological markers for combined stresses in Osmanabadi bucks.  相似文献

Data on genetic differences at CSN3 in goat breeds including a DNA based typing method and the mutations responsible for variation on protein and DNA level are presented here. Isoelectric focusing (IEF) in ultrathin polyacrylamide gels with carrier ampholytes was used to demonstrate CSN3 polymorphism in milk samples of Italian (Orobica n=88; Ionica n=68) and German goat breeds (Bunte Deutsche Edelziege n=244; Weisse Deutsche Edelziege n=134; Toggenburger n=25; Thüringer Waldziege n=70). A further CSN3 band was found presenting a more cathodic migration than CSN A. After chymosin action, the genetic polymorphism was also observed in the para-kappa-casein fraction. The new allele CSN3(B) was spread mainly in Orobica (37%), Bunte Deutsche Edelziege (11%) and Ionica (10%). CSN3(B) occurred in low frequency (<3%) in Thüringer Waldziege and in Weisse Deutsche Edelziege, and could not be demonstrated in milk samples of Toggenburger. The populations were in Hardy-Weinberg equilibrium at CSN3. The codominant genetic control of CSN3(B) was confirmed by genetic studies. Discrimination of CSN3 alleles A and B was also obtained by DNA SSCP analysis. Sequencing of CSN3(B) revealed four transitions at position 247, 309, 471, and 591 compared with CSN3(A). From these transitions, the following amino acid substitutions are deduced: 44 Gln --> Arg, 65 Val --> Ile, 119 Val --> Ile, and 159 Ser --> Pro. Among the four mutations, only the 44 Gln --> Arg can be revealed by milk protein IEF analysis while at DNA level three further genetic variants should exist in addition to CSN3(A) and CSN3(B).  相似文献

Indian goat breeds are recognized as an invaluable component of the world's goat genetic resources. Microsatellite pairs were chosen from the list suggested by International Society for Animal Genetics (ISAG) and amplified in two multiplexes (Set-I: 7 microsatellites and Set-II: 11 microsatellites) for automated fluorescence genotyping to assess bottleneck and analyze genetic variability and genetic distances within and between three goat breeds viz. Zalawadi, Gohilwadi and Surti. The observed number of alleles ranged from 4 (Oar JMP-29) to 15 (ILSTS-030 and -034) with a total of 178 alleles and mean of 9.89 alleles across the three breeds. The overall heterozygosity, PIC and Shannon index values were 0.61, 0.60 and 1.50 indicating high genetic diversity. The maximum observed heterozygosity was found in Gohilwadi and minimum in Surti goat breed. The Nei's standard genetic distance was minimum between Zalawadi and Gohilwadi, and maximum between Gohilwadi and Surti. Non-significant heterozygote excess on the basis of IAM, TPM and SMM models, as revealed from Wilcoxon sign-rank tests, along with a normal ‘L’-shaped distribution of mode-shift test, indicated no bottleneck in Zalawadi and Gohilwadi goat populations, whereas mild bottleneck in the recent past for Surti breed. This research on goat genetic diversity in Gujarat state provides valuable information on Zalawadi, Gohilwadi and Surti goat genetic resources, and will assist in developing a national plan for the conservation and utilization of indigenous goat breeds.  相似文献

A study was conducted to assess the adaptive capability of three indigenous goat breeds to heat stress. Thirty six 10 months to one-year-old female goats of Osmanabadi, Malabari and Salem Black breeds were randomly divided into six groups, OC (n = 6; Osmanabadi control), OHS (n = 6; Osmanabadi heat stress), MC (n = 6; Malabari control), MHS (n = 6; Malabari heat stress), SBC (n = 6; Salem Black control) and SBHS (n = 6; Salem Black heat stress). Among the metabolic activity controlling hormones, the breed factor significantly (P < 0.05) influenced only plasma triiodothyronine (T3). However, heat stress significantly (P < 0.05) decreased thyroid stimulating hormone (TSH) in both MHS and SHS groups while significantly (P < 0.05) decreased the plasma T3 in MHS. The rumen metabolites such as acetate, propionate, butyrate and total volatile fatty acids (TVFAs) showed significant (P < 0.05) variation for both breed and treatment effect. The Salem Black breed did not show any significant variation for most of the rumen metabolites as compared to both Osmanabadi and Malabari breeds for the heat stress treatment. The study indicated the importance of the metabolic alterations in indigenous goat breeds to cope to the seasonal rhythms. The results indicated that on comparative basis, Salem Black breed adapted better to the heat stress challenges.  相似文献

Aims:  To determine the variability of the prion protein gene ( PRNP) in goats from Northern and Southern Italy.
Methods and results:  Genomic DNA isolated from goat blood was polymerase chain reaction (PCR)-amplified for the coding region of the PRNP gene and then sequenced. In total, 13 polymorphic sites were identified: G37V, T110P, G127S, M137I, I142M, I142T, H143R, R154H, P168Q, T194P, R211Q, Q222K and S240P (substitutions I142T and T194P are novel) giving rise to 14 haplotypes. Clear frequency differences between Northern and Southern breeds were found and confirmed by genetic distance analysis.
Conclusions:  Differences in allele distribution were found between Northern and Southern goats, in particular regarding the M142 and K222 alleles, possibly associated to scrapie resistance; philogeographical analysis supported the idea that Northern and Southern breeds may be considered as separate clusters.
Significance and impact of the study:  In Italy only limited studies have been carried out on caprine PRNP genotype distribution; this study is important to fill this lack of information. Moreover the finding of significant differences among allele distributions in Northern and Southern goats, especially if involved in modulating resistance/susceptibility, need to be carefully considered for the feasibility of selection plans for resistance to scrapie.  相似文献

The present study was conducted to examine differential expression pattern of HSP genes and adaptability in Indian goat breeds of semi-arid region. The study was conducted in five animals from each breed viz. Barbari, Sirohi, and Jhakrana during winter, thermo-neutral and summer seasons. The respiratory rate (RR) and rectal temperature (RT) of the goats were recorded at 09:00 h during the study period. The blood samples were collected for RNA isolation, cDNA synthesis, and quantitative analysis of HSP genes expression by quantitative RT-PCR. The RR increased significantly (p < 0.01) during summer as compared to winter and thermo-neutral season however, RT did not change (p > 0.05) during different seasons. The expression of HSP genes was significantly (p < 0.01) increased during summer (high THI) as compared to thermo-neutral season in all the goat breeds. Among HSPs, only HSP90 was upregulated (p < 0.01) in Jhakrana goats during winter as compared to thermo-neutral season. The deviation in expression of HSP genes during summer and winter with respect to thermo-neutral season was minimum in Barbari goats. Therefore, it can be concluded that Barbari goats possessed better adaptability during summer and winter as compared to Sirohi and Jhakrana goats in semi-arid climatic conditions of India.  相似文献

Summary Experimental assessment of differences between cultivars of crop species or ecotypes of wild species from different localities in their capacities for ion absorption and transport is made difficult by the problem of obtaining seed material of comparable ionic content. When young seedlings are used this problem is particularly acute if the seeds of the different cultivars have not been raised under identical soil conditions. Propagation of material from ecotypes under controlled conditions is one approach to the solution of this problem. Six maize cultivars have been selected for similarity of phosphate content and the capacity for phosphate absorption from 5 M KH2PO4 has been shown to vary by threefold whereas the proportion of the accumulated phosphate that reaches the shoot differs by much less. This level of phosphate supply approached that likely to induce deficiency and when the concentration is reduced to 1 M differences in transport capacity of up to fourfold were observed when the rate of arrival at the tip of the first leaf was continuously monitored. The rapidity with which the transport is shut off by adding 1 mM D(+) mannose to the root environment also varies significantly indicating that sizeable differences in either the accumulation of mannose or the activity of phosphomannoisomerase exist in these cultivars.Ecotypes ofArmeria maritima collected from three sites, inland serpentine, inland mine dumps and coastal salt marsh were maintained as stock plants on the same peat mixture. Samples taken from these stocks were raised on a standard culture solution to provide genetically different material grown under constant conditions. The capacities for ion uptake were shown to differ very considerably and these differences were accentuated when the plants were grown in a range of concentrations of MgSO4, NaCl and MnSO4. The absorption of phosphate and its incorporation into nucleic acids were increased temporarily in the presence of 50 mM MgSO4 but the pattern of these changes was different in the three ecotypes. The absorption of Na, Cl, and Rb was measured after treatment with a range of concentrations of NaCl and the effect of treatment with MnSO4 on subsequent absorption of Mn and SO4 was also measured. The coastal plants were significantly more efficient in their absorption of these ions when treated at the lower levels of NaCl (0.5 and 10.0 mM). The short term absorption rates were not reflected in the overall accumulation of sodium over periods of 10 weeks and the coastal plants appeared to reduce the root content of sodium by transfer to the shoot and by increased active pumping to the exterior.  相似文献

植物功能性状与环境之间的关系是功能性状研究的重点,环境因子驱使植物功能性状发生变化,进而推动群落发生演替。以平潭岛4个不同演替阶段的森林植被(灌草丛、针叶林、针阔混交林、常绿阔叶林)为研究对象,结合不同群落演替阶段的物种特征和群落结构,分析海岛不同演替阶段茎、叶功能性状的变化规律,以及功能性状与环境因子的关系。结果表明:(1)随着演替的进行,土壤养分和水分逐渐增加,土壤pH逐渐下降。比叶面积(SLA)、叶片氮含量(LNC)、叶片磷含量(LPC)、茎氮含量(SNC)、茎磷含量(SPC)下降后上升,叶厚度(LT)、叶片碳含量(LCC)、茎碳含量(SCC)与之相反,叶干物质含量(LDMC)、茎组织密度(STD)逐渐上升。(2)冗余分析表明,演替早期植物主要分布在土壤pH、容重高的贫瘠环境,拥有较高SLA、SNC、SPC、LPC的性状组合;演替后期植物主要分布在土壤养分和水分高的肥沃环境,拥有较高的STD、LDMC、LCC、LNC的性状组合。其中,土壤有机质和全氮含量是影响海岛植物演替过程中功能性状变化的关键环境因子。研究海岛植物功能性状与环境之间的关系随演替的变化规律,探讨各演替阶段功能性状和环境特征,以及功能性状如何响应环境变化。旨在为今后选择合适的树种进行海岛植被修复和重建提供依据。  相似文献

A study was conducted to assess the effect of combined stresses (thermal and nutritional) on endocrine and biochemical responses in Malpura ewes. Twenty eight adult Malpura ewes (average body weight 33.56 kg) were used in the present study. The ewes were divided into four groups viz., GI (n = 7; control), GII (n = 7; thermal stress), GIII (n = 7; nutritional stress) and GIV (n = 7; combined stress). The animals were stall fed with a diet consisting of 60% roughage and 40% concentrate. GI and GII ewes were provided with ad libitum feeding while GIII and GIV ewes were provided with restricted feed (30% intake of GI ewes) to induce nutritional stress. GII and GIV ewes were kept in climatic chamber at 40°C and 55% RH for 6 h a day between 1000 hours and 1600 hours to induce thermal stress. The study was conducted for a period of two estrus cycles. The parameters studied were Hb, PCV, glucose, total protein, total cholesterol, ACP, ALP, cortisol, T4, T3, and insulin. Combined stress significantly (P < 0.05) affected all parameters studied. Furthermore, the results revealed that, compared to thermal stress, nutritional stress had a less significant effect on the parameters studied. However, when both these stresses were coupled, they had a severe impact on all the parameters studied in these ewes. It can be concluded from this study that two stressors occurring simultaneously may impact severely on the biological functions necessary to maintain homeostasis in sheep.  相似文献

非结构性碳水化合物(Non-structural Carbohydrates, NSCs)是植物生长代谢过程中重要的能量来源。通过在华南热带次生林进行氮磷添加试验,探究不同林层植物叶片NSCs的季节变化及其对氮磷添加的响应,取样时间为2019年1月、4月、7月和10月。结果表明:1)植物叶片NSCs存在显著的种间差异,磷(P)添加对叶片淀粉和NSCs含量具有显著影响,且物种与磷添加的交互作用显著影响叶片淀粉含量。2)黑嘴蒲桃和紫玉盘叶片NSCs含量对氮(N)添加的响应较为敏感,而白车和竹节叶片NSCs含量对P添加的响应较为敏感,氮磷同时添加(+NP)对植物叶片NSCs的增效作用最好。3)植物叶片NSCs存在显著的季节性变化,且季节与林层间的交互作用对叶片可溶性糖和NSCs含量具有显著影响。4)不同林层植物对氮磷添加的响应不同,氮磷添加使林下层植物叶片可溶性糖含量增高,林冠层降低,在干季,N添加会使林下层植物叶片淀粉含量增高,林冠层降低。P添加的影响恰好与之相反。在湿季,氮磷添加使林下层和林冠层植物叶片的淀粉含量增加。5)林冠层植物叶片NSCs含量高于林下层,且林下层植物叶片NSCs含量...  相似文献

以热带地区主栽品种博优225和矮糯为材料,采用盆栽试验研究了不同水稻品种对土壤外源重金属Cd胁迫的耐性机理。结果表明:在各浓度镉胁迫下,2个品种水稻都是根累积的镉含量要高于茎叶和稻壳、糙米,即根>茎叶>稻壳>糙米;随着镉胁迫浓度的增加,细胞膜受损的伤害程度越大,矮糯质膜透性略高于博优225;随着Cd浓度的升高,水稻叶片丙二醛(MDA)、脯氨酸含量逐渐增加,且博优225的增加幅度明显大于矮糯,超氧化物歧化酶(SOD)和过氧化物酶(POD)活性均呈上升趋势,且博优225明显高于矮糯;过氧化氢酶(CAT)活性呈下降趋势;表明同样条件下博优225受Cd毒害更严重。  相似文献

There have been extensive studies in sheep and cattle considering cobalt (Co) supplementation and its effects on vitamin B12 concentrations in the body. However, there are limited studies on goats. The aim of this study was to compare two different sources of Co (sulfate v. glucoheptonate) at two different concentrations (0.25 and 0.5 mg/kg dry matter) in goat kid nutrition, and to evaluate the effects of these supplements on performance, serum vitamin B12, blood biochemistry and rumen volatile fatty acids. For this purpose, 30 weaned male goat kids were randomly allotted to five treatments. Serum vitamin B12 increased during the trial in the Co-supplemented groups. Co supplementation increased serum glucose concentrations. On day 35, Co-supplemented groups had greater glucose concentrations compared with control. Propionic+iso-butyric acid concentrations increased only in the 0.5 mg Co glucoheptonate treatment (P<0.05). Our results suggest that, despite the two sources of Co proving mostly similar, the main advantage of Co glucoheptonate compared with Co sulfate was in the ruminal synthesis of vitamin B12. However, although providing Co at National Research Council recommendation levels maintained vitamin B12 above or at normal concentrations, Co supplementation of the Co sufficient basal diet increased vitamin B12 and glucose concentrations.  相似文献

Twelve wethers (27–33 kg and 12–14 months of age) representing meat (Nubian), milk (Alpine) and mohair (Angora) producing goats were used to evaluate breed differences in protein utilization with diets containing 9 (L), 15(M) or 21% CP (H) and 2.4 Mcal ME/kg DM in a digestion trial. Fecal N, urine N and absorbed N as percent of N intake were not affected (P>0.05) by breed. Retained N as percent of absorbed N was not different (P>0.05) between breeds. Ruminal propionate (molar %) was lower (P<0.05) in Angora, but other ruminal VFA and ammonia-N were not affected (P>0.05) by diet or breed. Plasma urea-N increased (P<0.01) with dietary CP level (8.3, 22.0 and 33.3 mg/dl for L, M and H, respectively), and concentrations were lowest for Angoras and highest for Nubians (18.5 vs. 21.2 vs. 23.9 mg/dl) (P<0.01). Plasma total protein (mean 69.7 g/l), glucose (mean 83.1 mg/dl), non-esterfied fatty acids (mean 101.4 μEq/l) and cortisol (mean 10.8 ng/ml) were not affected (P>0.05) by breed or diet. Plasma glucagon concentrations increased (P<0.05) with increasing CP intake (72.4, 167.6 and 239.1 pg/ml for L, M and H, respectively). The study indicated that there was no apparent breed difference between Nubian, Alpine and Angora goats in nitrogen utilization when goats were fed pelleted diets containing 9 to 21% CP.  相似文献

The present study was conducted to assess the comparative physiological responses of stress in sheep selected for resistance or susceptibility to Haemonchus contortus in the semi-arid region during different seasons. A total of 20 adult Malpura sheep from resistant and susceptibility line were selected for study. The intensity of strongyle infection, blood biochemical and cortisol levels were estimated monthly basis while meteorological data were collected everyday throughout the year. The temperature-humidity index was the highest during dry-hot season. The faecal egg count increased significantly (p < 0.05) during June–August (rainfall season) in susceptible sheep as compared to resistant sheep. The plasma cortisol level increased significantly (p < 0.05) in both the groups during dry hot (March–May) season and it remained higher in the susceptible line till June–August. The total plasma protein reduced significantly during hot-humid with scantly rainfall (September–November) season as compared to other season in susceptible line. It is pertinent to conclude that though sheep remained in stress during dry hot (March–May) season, but effects of stress on the intensity of strongyle infection and blood biochemical were better tolerated by animals of resistant line during hot-humid season compared to susceptible line under semi-arid region.  相似文献

Sheep in hot semi-arid environments are mostly reared using extensive systems. In addition to thermal stress and feed scarcity, the animals need to walk long distances for grazing in this ecological zone. A study was conducted to assess the effect of long-distance walking on adaptive capability in terms of physiological, biochemical and endocrine responses in Malpura ewes. Fourteen adult Malpura non-pregnant ewes weighing between 33 and 35 kg were used in the study. The ewes were randomly allocated into two groups of seven animals each: GI (n = 7; Control), and GII (n = 7; walking stress). The animals were stall-fed with a diet consisting of 70% roughage and 30% concentrate. Both GI and GII ewes had uniform access to feed and water. The walking stress group (GII) ewes were made to walk 14 km in two spans between 0900 and 1500 hours with 1 h 30 min for each span (7 km) of walking. The ewes subjected to walking stress (GII) were prevented from grazing by applying a face mask made of cotton thread. The study was conducted for a period of two estrous cycles (35 days) during the autumn season (October–November). Physiological responses were recorded twice daily at 0800 and 1400 hours at weekly intervals. Blood samples were collected from the jugular vein at weekly intervals to study the effects of walking stress on blood biochemical and endocrine parameters. The results indicate that walking stress had significant (P < 0.05) influence on body weight, average daily gain, respiration rate (RR), rectal temperature (RT), haemoglobin (Hb), packed cell volume (PCV), plasma glucose, calcium, phosphorus, aspartate amino transferase (AST), alanine amino transferase (ALT), tri-iodo-thyronine (T3), thyroxin (T4), and cortisol. However, walking stress did not influence the reproductive hormone levels. The significant changes in RR, RT, plasma cortisol, T3 and T4 show that Malpura ewes have the capability to adapt to long-distance walking, and that adrenal and thyroid gland hormones play a significant role in such adaptation.  相似文献
