Solubilized protoxins of nine Cry1 and one hybrid Cry1 delta-endotoxin from Bacillus thuringiensis were tested for their activity against larvae of the codling moth (Cydia pomonella L). Cry1Da was the most toxic, followed by Cry1Ab, Cry1Ba, and Cry1Ac, while Cry1Aa, Cry1Fa, Cry1Ia, and SN19 were still less active. Cry1Ca and Cry1Cb showed no activity. In vitro trypsin activation increased activity of all eight active delta-endotoxins, and dramatically enhanced toxicity of hybrid SN19, Cry1Aa, Cry1Ac, and Cry1Fa. The differences between toxicity of proteins before and after trypsin digestion suggests that proteolytic activation in the C. pomonella digestive tract plays a critical role for the activity of Cry proteins against this insect.  相似文献

Abstract Nuclear polyhedrosis virus (family Baculoviridae, subgroup A) in vitro systems have been well established for more than ten years, and have permitted application of modern microbiological techniques. Although widely tried, similar systems for granulosis virus (subgroup B) replication have not been obtained to our knowledge. We report here on the successful in vitro replication of Cydia pomonella granulosis virus in a Cydia pomonella primary cell line. The replication was confirmed by light and electron microscopy. This granulosis virus in vitro replication system will be an important tool for further granulosis virus research and for comparative studies on granulosis and nuclear polyhedrosis virus replication.  相似文献

Two different methods were tested to identify the sex of the early developmental stages of the codling moth Cydia pomonella (Linnaeus) (Lepidoptera: Tortricidae) with a WZ/ZZ (female/male) sex chromosome system. First, it was shown that the sex of all larval stages can be easily determined by the presence or absence of sex chromatin, which is formed by the female‐specific W chromosome in interphase nuclei. This trait can also be used to identify the sex of newly hatched larvae but it does require care and accuracy. Secondly, a new sexing technique was developed based on a molecular marker of the codling moth W chromosome. Flanking regions of an earlier described W‐specific sequence (CpW2) were isolated and sequenced and a 2.74 kb sequence (CpW2‐EcoRI), specific for the W chromosome, was obtained. Several PCR tests were conducted, which confirmed that the CpW2‐EcoRI sequence is a reliable marker for the sex identification in codling moth samples of different geographical origin. In addition, a fragment of a codling moth gene, period (Cpper) was isolated and sequenced. Results of southern hybridization of the Cpper probe with female and male genomic DNA suggested that the Cpper gene is located on the Z chromosome. Then a multiplex PCR assay was developed, which co‐amplified the CpW2‐EcoRI sequence to identify the W chromosome and the Z‐linked Cpper sequence, which served as a positive control of accurate processing of tested samples. The multiplex PCR provides an easy and rapid identification of the sex of embryos and early larval instars of the codling moth.  相似文献

Relationships between a pheromone-trap catch, adult emergence and penetration of fruit by first-instar larvae of Cydia pomonella were investigated from 1975 to 1977 in an orchard in South West England. For the first generation the times of moth emergence and catch in the pheromone trap were not significantly different; nor were male and female emergence times. The catch of moths of the first generation in the pheromone trap anticipated the appearance of their larvae in the fruit by 140–169 day-degrees > 10 °C. Eggs hatched after 94 day-degrees in the laboratory but in the orchard, wind and sunshine modified the microclimate so that the number of day-degrees required for egg development, as measured by standard meteorological instruments, was affected by wind and sunshine but development lasted on average about 90 day-degrees. This indicated a lag of 50–80 day-degrees between the curves for trap catch and oviposition: the pre-oviposition period in the orchard was shorter than expected from laboratory studies. In 1975 and 1976, some larvae developed to produce a second generation of moths which gave rise to a second generation of eggs and larvae, after an interval of 161 day-degrees in 1975, but only 41 day-degrees in 1976, indicating that in 1976 some eggs were laid before moths of the second generation were trapped. These results indicate that the first insecticidal spray against first-generation larvae should be applied about 140 day-degrees after the start of the reference week in which five moths of the spring brood are caught per trap. A second spray, if required to maintain insecticidal cover, should be applied about 100 day-degrees later. A spray should be applied against second-generation larvae immediately after the reference week for second-generation moths.  相似文献

Insecticide‐based management programmes targeting codling moth, Cydia pomonella L. (Lepidoptera: Tortricidae: Olethreutinae), in apple orchards in southern France have incurred increasing levels of fruit injury in recent years. An alternative programme incorporating the use of exclusion netting named Alt'carpo has been developed. This study aimed at studying its efficiency and gaining more insight into its mode of action. This was achieved through laboratory and field behavioural tests and observations in a network of commercial orchards in southern France. The moths were able to lay eggs through the nets and escape from net cages in the laboratory. Male moths released in the netted rows were poorly recaptured using sexual pheromone trapping, whereas over 20% of the released males were recaptured in unprotected rows. The netting reduced fruit injury by up to 91% compared to the unprotected rows in the experimental orchard. The efficacy of this netting was even higher in commercial orchards in which fruit injury did not exceed 0.1% without any application of specific insecticide. These results lead us to assume that netting alters the reproduction of the pest, mainly by preventing it from flying over the canopy to find mates.  相似文献

The codling moth, Cydia pomonella (L.) (Lepidoptera: Tortricidae), has developed resistance to various insecticides. Relative fitness of one susceptible strain (Sv) and two strains selected for resistance to diflubenzuron (Rt) and deltamethrin (Rv), respectively, was measured in the absence of insecticide selection pressure. Mating rate, fecundity, fertility, developmental time, fifth instar weight, and adult longevity were compared. Both resistant strains were less fecund and fertile, developed more slowly, weighed less, and had shorter life-spans than the susceptible strain. These results indicate that biological constraints are associated with insecticide resistance in the codling moth. We also found that fitness estimates of the Rv strain did not differ statistically from those of the Rt strain. Enhanced mixed-function oxidase and glutathione-S-transferase activities have been shown to be involved in insecticide resistance in both Rt and Rv strains. This suggests that the fitness cost described in both resistant strains was mainly associated to metabolic resistance. The impact of such deleterious pleiotropy of insecticide resistance in C. pomonella in terms of resistance management in the field is discussed.  相似文献

Regulation of autonomic physiological functions has been investigated by means of multisensor electronic methods, including electrocardiographic recording of heartbeat, strain-gauge recording of extracardiac hemocoelic pulsations (EHPs), anemometric recording of air passage through spiracles and respirographic recording of O(2) consumption and CO(2) output. Pupae of Cydia exhibit continuous respiration without remarkable bursts of CO(2). The dorsal vessel of these pupae exhibited regular heartbeat reversals characterized by shorter intervals of faster (forward oriented or anterograde) pulsations and longer intervals of slower (backward oriented or retrograde) peristaltic waves. The periodically repeated EHPs were present during the whole pupal interecdysial period. The internal physiological mechanisms regulating the cardiac (heartbeat) and extracardiac (EHP) pulsations were completely independent for most of the pupal instar. Simultaneous multisensor analysis revealed that the anterograde heartbeat of the dorsal vessel had similar but not identical frequency with EHPs. During advanced pharate adult development, frequency of cardiac and extracardiac pulsation periods profoundly increased until almost uninterrupted pulsation activity towards adult eclosion. At this time, the cardiac and extracardiac pulsations occasionally performed in concert, which enhanced considerably the efficacy of hemolymph circulation in pharate adults with high metabolic rates. The fastest hemolymph flow through the main body cavity was always associated with EHPs and with anterograde heartbeat. Simple physical diffusion of O(2) and CO(2) through spiracles (diffusion theory of insect respiration) does not play a significant role in pupal respiration. Instead, several kinds of regulated, mechanical ventilations of the tracheal system, including EHPs are responsible for effective tracheal ventilation.  相似文献

Identification of host volatile compounds attractive to codling moth Cydia pomonella, a most important insect of apple, will contribute to the development of safe control techniques. Synthetic apple volatiles in two doses were tested for antennal and behavioural activity in codling moth. Female antennae strongly responded to (Z)3‐hexenol, (Z)3‐hexenyl benzoate, (Z)3‐hexenyl hexanoate, (±)‐linalool and E,Eα‐farnesene. Two other compounds eliciting a strong antennal response were the pear ester, ethyl (E,Z)‐2,4‐decadienoate, and its corresponding aldehyde, E,E‐2,4‐decadienal, which is a component of the larval defence secretion of the European apple sawfly. Attraction of codling moth to compounds eliciting a strong antennal response was tested in a wind tunnel. Male moths were best attracted to a blend of (E,E)‐α‐farnesene, (E)‐beta‐farnesene and ethyl (E,Z)‐2,4‐decadienoate. The aldehyde E,E‐2,4‐decadienal had an antagonistic effect when added to the above mixture.  相似文献

【背景】苹果蠹蛾原产于欧亚大陆,是世界著名的严重危害苹果生产的入侵害虫,也是中国的重要对外检疫性有害生物。【方法】通过收集整理历史文献、资料,并于2005~2013年使用标准性信息素诱芯对全国各苹果、梨主产区的苹果蠹蛾发生情况进行系统监测,明确该虫在我国的分布现状及扩散历史;此外,使用特定公式对苹果蠹蛾在发生区造成的当前经济损失和扩散完成后的潜在经济损失进行了系统评估,以明确该虫在我国的现有和潜在危害。【结果】苹果蠹蛾于1953年首次在中国新疆被发现,目前在中国分布于新疆、甘肃、内蒙古、宁夏、黑龙江、辽宁和吉林7省的144个县(市、区、旗),主要集中分布在东经74.56°~106.8°以及东经120.02°~132.95°,发生面积约49410 hm~2;经济损失评估结果显示,苹果蠹蛾导致中国苹果和梨的潜在减产数量为185.01万和81.33万t。【结论与意义】该虫已在中国形成东西2个分布区,对占据中国苹果产量80%的西北黄土高原(陕西为主)和渤海湾(山东、河北、辽宁为主)两大苹果主产区构成了严重威胁,须引起高度重视。  相似文献

The resistance to deltamethrin was evaluated in diapausing larvae of 14 field populations of the codling moth (Cydia pomonella L.) From the main French orchard areas using biological assays. Resistance to deltamethrin was compared to mixed-function-oxidase (mfo) activity measured at the individual level through ethoxycoumarin-O-deethylase (ECOD) activity using a fluorescence microplate reader. The larvae were collected in corrugated paper band traps in the autumn of 1995. Analysis was previously performed on two laboratory strains, one susceptible and one resistant to deltamethrin, in order to characterize the changes in resistance during diapause development. Resistance to deltamethrin as well as the ECOD activity were stable during the diapause, and ECOD activity was always significantly lower in the susceptible strain than in the resistant one. The ECOD activity was significantly correlated to the frequency of resistant moths in the field populations. This strongly suggested the involvement of the mfo system in the resistance to deltamethrin in these populations. The intra-strain variabilities in ECOD activity of both laboratory and field resistant insects indicated that other resistance mechanisms might also be involved. Further investigations on these mechanisms are needed in order to develop complete diagnostic methods and to define suitable control strategies against each resistant population. Arch. A model was developed based on an initial ingestion phase, followed by a period of virus multiplication and host death, to describe the effects of codling moth granulosis virus (CpGV) concentration and time from initial infection on the percentage of codling moth larvae (Cydia pornonella) dying from virus infection in the laboratory. With modification, this model also described the effect of CpGV concentration on the reduction of damage to fruit by C. pomonella larvae in the field. In both cases, the effect of CpGV increased with the l/10th power of virus concentration. LT50 (time to death of 50% of larvae) was inversely related to virus concentration in the laboratory. The model predicts the threshold concentration for death of larvae, as well as LD50(C50 and LD90(C 90,) for all ages of larvae. The model also predicts that if the majority of larvae entered through the sides of apple fruits, rather than through the calyx or near the stalk (as they probably did in the field), then the efficacy of CpGV in reducing fruit damage would be lowered. This effect of site of entry is especially marked at low virus concentrations.  相似文献

Spread pachytene nuclei of codling moth Cydia pomonella (Linnaeus) (Lep., Tortricidae) females of a Syrian strain (SY) were used to investigate chromomere patterns of chromosome bivalents and determine their length. The karyotype of female codling moths consists of 28 chromosome bivalents, of which seven are clearly distinguishable using chromosome length and the number and size of the chromomeres in the pachytene stage. One autosome bivalent has two nucleolar organizing regions (NORs) that are located at the opposite ends of the chromosome and appear as distinct structural landmarks. In female codling moths, the WZ sex chromosome bivalent was easily identified in pachytene oocytes according to the heterochromatic thread of the W chromosome. This study contributed to the knowledge and identification of pachytene chromosomes of female codling moths.  相似文献

Predation of cocooned larvae of codling moth Cydia pomonella by silvereyes Zosterops lateralis was studied in an apple orchard in Nelson, New Zealand. Apple logs with known larval densities were made available to the birds for known periods of time, either in cages or exposed in the apple orchard. The numbers of silvereyes and the natural predation of codling moth were recorded in the same orchard. Predation was density dependent. On caged logs with an initial high density of 32 larvae, 1.1 larvae were consumed per bird‐hour; in contrast, one larva was consumed per 34.5 bird‐hours at three larvae per log. A curvilinear relationship was demonstrated between larval density and the bird‐hours required for predation; this relationship was consistent with the known density dependence of silvereye predation of codling moth. A regression of the total annual winter bird predation of larvae in the orchard on bird numbers was significant. However, the density dependence of predation resulted in declining rates of predation over the winter as larval density declined; the first birds to arrive in the orchard benefitted from particularly high predation rates. As a consequence, fluctuations in bird numbers during the winter had only a secondary influence on predation rates. The numbers of silvereyes in the orchard showed no relationship to the density of the codling moth population present. This study confirmed the importance of silvereyes in the predation of codling moth and a functional, not numerical, rseponse of these birds to codling moth density.  相似文献

Cydia pomonella L. is an important insect pest of pome fruits worldwide. We have isolated and characterized 17 microsatellite loci from the enriched genomic libraries constructed using a biotin/streptavidin capture protocol. Among these loci, 11 were scored polymorphic when 50 individuals from a laboratory population were screened. Their alleles numbered two to four, with the observed heterozygosity (HO) ranging from 0.114 to 0.404. Successful amplification was obtained for all these loci when the designed primers were tested, showing the promise of use in genetic mapping and population studies.  相似文献

Like in most parts of the world, the codling moth, Cydia pomonello (L.), is one of the most important pests in apple orchards in the Romanian Banat region. There, the control of this pest is often inadequate and the damage and economic losses are therefore enormous. This lack of control is caused by structural problems such as the absence of an advanced distribution network for phytopharmaceutical products, obsolete spraying equipment and the insufficient exchange of knowledge and expertise of those products and the pest itself between the local research facilities and the apple growers. The purpose of this research project is rather demonstrative, because a change in the Romanian pome culture's attitude is urgently needed. Pheromone trapping and the computerized population model RIMpro Cydia, which predicts important phenological events such as adult flight and egg hatch, were used to determine the optimal spray timing. These modern pest management techniques would not only minimize pressure on the environment, but also on the income of the Romanian apple grower, for whom phytopharmaceuticals are a high-priced investment. In July 2006 a comparative study between the supervised pest control program using Calypso, Runner and Reldan, all easily obtainable and commercially available insecticides in the region, and a conventional Romanian management programme was conducted. The control plot and the two test plots were all three located in a high codling moth pressure orchard in Timi?oara. In the control plot 9,44 % of the fruits were damaged by codling moth larvae. In the plot containing the trees that were treated according to the traditional Romanian management programme 5,56 % damage was recorded, while the supervised management plot showed only 1,1% damage. Additionally damage was measured in another orchard in Periam, about 60 km away from the other plots. There, an intensive control program using Reldan, Calypso, Actare, Karate and Insegar was applied and damage measured only 0.2%. While at first glance that approach seems most efficient, it has to be noticed that it used twice as much insecticide treatments as were used in the supervised control program. Furthermore, 1 % damage is suggested by most references as being the economical threshold for treatments. The results of this comparative study clearly show that supervised codling moth control proofs to be the most profitable. Starting from August the same year, all of the control programmes in Timi?oara were stopped. Therefore the second and possibly the third generation of codling moth larvae could develop freely. In the plot which was earlier sprayed according to the warnings of RIMpro Cydia and the pheromone trap this resulted in 27.5% damaged fruits during harvest in September. In the control plot damage reached 60.95%. These results highlight the necessity of the development of an efficient codling moth control program. Once that program is fine-tuned it should be presented to the local apple growers in order to give the Romanian pome culture a boost.  相似文献

The effects of temperature on post‐diapause development, female lifespan, and reproduction of Cydia pomonella L. (Lepidoptera: Tortricidae) were studied under controlled conditions to determine the respective parameters for a temperature‐driven phenology model. Lower thermal thresholds of 10.0 and 9.7 °C and thermal constants of 417.2 and 427.7 degree days were established for post‐diapause development of females and males, respectively. Female lifespan was found to be 202.6 degree days on average, with a lower thermal threshold of 11.2 °C. Total fecundity was highest at 27.7 °C with an average of 241.9 eggs per female. Literature data were used to quantify the lower thermal thresholds and the thermal constants for eggs, larvae, and pupae. For validation, the model was run with weather data from 13 consecutive years and the output was compared with pheromone trap catches from 39 to 63 different orchards, depending on the year. The model proved to be a reliable and useful tool to forecast codling moth phenology under Swiss conditions not only for a whole growing season but also during several years with very variable weather conditions. Hence, it can be used to support apple growers in pest management decisions on the one hand and to predict changes in codling moth phenology with respect to climate change on the other.  相似文献
