Pretreatment of beet molasses to increase pullulan production   总被引:2,自引:0,他引:2  
Pretreatment of beet molasses with cation exchange resin, sulphuric acid, tricalcium phosphate, potassium ferrocyanide, and ethylenediaminetetraacetic acid and disodium salt (EDTA) to increase the production of pullulan was investigated. Among the above techniques used for the removal of heavy metals, sulphuric acid treatment gave better results regarding polysaccharide concentration, polysaccharide yield, and sugar utilization. Aureobasidium pullulans grown on beet molasses produced a mixture of pullulan and other polysaccharides. The pullulan content of the crude polysaccharide was 30–35%. The addition of nutrients improved the production of polysaccharide. A maximum polysaccharide concentration (32·0±1·0 g litre−1) was achieved in molasses solution (70 g litre 1 initial sugar concentration, pH 6·5–7·5) treated with sulphuric acid and supplemented with K2HPO4 0·5%, -glutamic acid 1%, olive oil 2·5% and Tween 80 0·5%. In this case, the highest values of biomass dry weight (33·8±1·0 g litre−1), polysaccharide yield (63·5±2·5%), and sugar utilization (97·5±1·5%) were obtained at pH 6·5, 3·5, and 4·5–7·5, respectively.  相似文献

Cell walls of glasswort (Salicornia ramosissima Woods), a halophytic Chenopodiaceae, prepared as alcohol-insoluble solids, were found to be rich in arabinose, galacturonic acid, glucose and proteins, and contained 0·7% ferulic acid and 3·8% acetic acid. Pectic and hemicellulosic polysaccharides were extracted by cyclohexanediaminotetraacetic acid, hot dilute acid, cold dilute alkali and concentrated alkali (twice), with yields of 2·9, 19·1, 4·7, 7·4 and 1·9% of the alcohol-insoluble solids, respectively. Protein-rich material precipitated upon dialysis. The dialysed fractions were fractionated by ion-exchange chromatography, and the main fractions were analysed by gel-filtration and glycosyl linkage analysis. The hot acid extract contained 46·2% arabinose and 28·9% galacturonic acid, with high degrees of methylation and acetylation (65 and 45, respectively). It could be fractionated into a low-molecular-weight arabinan rich in ferulic acid, and a pectic fraction still relatively rich in neutral sugars. The concentrated alkali extracts were rich in xylose (33·4 and 23·6%, respectively). They were separated by ion-exchange chromatography into a fucogalactoxyloglucan and a glucuronoarabinoxylan.  相似文献

Cell-wall mucilaginous polysaccharides from Enteromorpha intestinalis were extracted, purified and their physicochemical characteristics determined. The successive extracts, obtained using distilled water at 35°C, distilled water at 75°C then an aqueous solution (0·25%) of ammonium oxalate at 75°C, all contained a sulphated glucoglucuronoxylorhamnan containing about 43% rhamnose, 19·8% sulphate and 17% uronic acid (based on the dry weight of the extracts). This composition is comparable to reported values for other Ulvaceae. However, the successive extracts differ in osidic composition and physical properties. In particular, extracts using oxalate at 75°C contain galactose and have lower values of viscosity and molecular weight with respect to those obtained in the first and second extracts, probably as a result of the breakage of labile interchain bonds during extraction.  相似文献

A selection programme to increase the cellular eicosapentaenoic acid (EPA) content has been carried out with the microalga Isochrysis galbana. The selection process involved two stages of single selection. EPA content continuously increased from 2·4% dry weight (d.w.) of the ‘parent’ culture to an average value of 5·3% d.w. in the final stage. The proportion of total EPA variation attributable to the genetic variation (heritability in a broad sense) was 0·99 showing the importance of the genome in the determination of this fatty acid. The growth and fatty acid profile of an EPA-rich isolate grown as a chemostat in a cylindrical photobioreactor have been studied. A decrease in EPA content was observed (5·21% w/w to 2·8% w/w) at the lowest dilution rate D = 0·024 h−1, up close to the maximum growth rate, D = 0·038 h−1. At the same time, the biomass concentration also decreased from 1015 mg/litre to 202 mg/litre over the abovementioned range of dilution rate (D). Nonetheless, the EPA productivity increases with D, with a maximum of 15·26 mg/litre/day at D = 0·0208 h−1. Furthermore, steady-state dilution rates may be related to average internal light intensity. Reverse-phase, high-pressure liquid chromatography (HPLC) on octadecylsilyl semi-preparative columns was used to separate stearidonic acid (SA), EPA and docosohexaenoic acid (DHA) in polyunsaturated fatty acid concentrate obtained by the urea complexation method from a fatty acid solution previously obtained by direct saponification of biomass. Isolate SA, EPA and DHA fraction purity was 94·8, 96·0 and 94·9%, respectively, with yields of 100·0, 99·6 and 94·0%.  相似文献

Elemental composition and feeding rate of hydromedusae Phialidium sp. on copepods were studied in the laboratory. Regression equations for both mature and immature medusae allowed the estimation of their dry weight (DW), total C and N content as a function of their diameter. The mean C content as percentage of the DW varied from 13.13% ( ) for the immature to 19.38% (5.68) for the mature individuals. The mean N content is 4.03% (2.49) of DW of immatures and 5.85% (2.70) of the matures. Ingestion rate of Phialidium sp. fed on copepods (200–500 μm) increased with prey density but reached a maximum at high prey concentrations. A maximum ingestion rate of 8.55 (1.6) copepods · medusa −1 · h−1 was reached for prey concentrations of > 140 copepods · 1 −1 for both immature and mature medusae. Maximum daily consumption of prey weight varied from 1.41 to 978% C body weight for mature medusae and from 2.90 to 975% for the immature individuals.  相似文献

Starch from AC Hill oat grains (Avena nuda) was isolated and some of the characteristics determined. The yield of starch was 23·4% on a whole grain basis. The shape of the granule was polyhedral to irregular, with granules 6–10 μm in diameter. Lipids were extracted by acid hydrolysis and by selective solvent extraction with chloroform-methanol 2:1 v/v (CM) at ambient temperature, followed by n-propanol-water 3:1 v/v (PW) at 90–100°C. The acid hydrolyzed extracts which represented the total starch lipids (TSL) was 1·13%. The free lipids in the CM extract (1% TSL) was 6·2%, whereas the free and bound lipids in the PW extracts was 93.0%. Neutral lipids formed the major lipid class in the CM and PW extracts. The monoacyl lipid content in both CM and PW extracts was 61·0%. The total amylose content was 19·4%, of which 13·9% was complexed by native lipids. X-ray diffraction was of the ‘A’ type. Oat starch differed from wheat starch in showing a higher swelling factor, decreased amylose leaching, coleaching of a branched starch component and amylose during the pasting process, higher peak viscosity and set-back, low gel rigidity, greater susceptibility towards acid hydrolysis, greater resistance to -amylase action and a higher freeze-thaw stability. Furthermore, in comparison to wheat starch, the amylose chains of oat starch appear to be more loosely arranged in the amorphous regions, whereas in crystalline regions, oat starch chains are more compactly packed. Lipid removal from oat and wheat starches decreased their swelling factor, peak viscosity, set-back, gelatinization temperatures, freeze-thaw stability and paste clarity (at pH > 4·0), and increased their thermal stability, amylose leaching, enthalpy of gelatinization, susceptibility towards -amylase and paste clarity (at pH < 4·0). Nitrogen excretion and assimilation efficiencies in Lithognathus mormyrus (Linnaeus), a marine teleost present in high-energy surf zones in Algoa Bay, South Africa, were determined under controlled laboratory conditions at 15, 20 and 25 °C. Ammonia was the major form of nonfaecal nitrogen excreted by starved and fed L. mormyrus. Urea and amino acids were secondary excretory products. Ammonia excretion rates were temperature independent and the excretion rates of fed fish were significantly higher than starved fish at 20 and 25 °C but not at 15 °C. The mass component (b) of the mass/ammonia excretion equation was temperature independent and ranged from 0.590 to 0.669 and from 0.670 to 0.767 for starved and fed fish, respectively. The mean percentage of food energy lost via the dissolved nonfaecal excretory products (exogenous plus endogenous) was 4.12%. Assimilation efficiencies ranged from 71.89 to 98.78% for dry matter and from 96.89 to 99.88% on an energy basis. The combined nonfaecal and faecal energy loss was calculated at 10.11% of the ingested energy. The omnivorous ichthyofauna present in the surf zone ecosystem recycle 33 g N · m strip · yr−1. This constitutes < 1 % of total phytoplankton nitrogen requirements.  相似文献

Aspergillus niger catalase has been characterized by a variety of physical techniques including gel filtration, sedimentation rate and equilibrium methods and photon correlation spectroscopy. The catalase has a sedimentation coefficient (S200) of 14.2 ± 0.08 S and diffusion coefficient (D200) of 4.14 ± 0.35 × 10−7 cm2 s−1. The average molecular weight of the catalase from all available data including current sedimentation equilibrium measurements and two previous literature values is 345 000. The frictional ratio of the molecule assuming a hydration parameter similar to that of bovine liver catalase (.3 g H2O g−1) is 1.103, suggesting that Aspergillus niger catalase has an asymmetric structure with an axial ratio of approximately 3 (the Stokes radius is 5.83 ± 0.49 nm). The titration curve and amino acid analysis indicate that in the native conformation only 23% of the ionizable amino acid residues are titratable between pH 3 and 10.5. Denaturation with sodium n-dodecylsulphate increases the number of titratable groups to 46%. The ratio of anionic to cationic amino acid residues in Aspergillus niger catalase is 2.46 and the isoelectric point is 6.5. The optimum pH for catalytic activity is approximately 7.  相似文献

The infrared, visible and nuclear magnetic resonance spectra of protochlorophyll a and vinylprotochlorophyll a in dry non-polar solvents (carbon tetrachloride, chloroform, cyclohexane) are presented and interpreted in terms of dimer interaction.

The infrared spectra in the 1600–1800 cm−1 region clearly show the existence of a coordination interaction between the C-9 ketone oxygen function of one molecule and the central magnesium atom of another molecule. Infrared spectra in the OH stretching region (3200–3800 cm−1) provide a valuable test of the water content in the samples.

The analysis of the absorption and circular dichroism spectra of protochlorophyll a and vinylprotochlorophyll a in carbon tetrachloride demonstrates the existence of a monomer-dimer equilibrium in the concentration range from 10−6 to 5 · 10−4 M. The dimerization constants are (6±2) · 105 1 · M−1 for protochlorophyll a and (4.5±2) · 105 1 · M−1 for vinylprotochlorophyll a at 20 °C. The deconvolution of visible spectra in the red region has been performed in order to obtain quantitative information on the dimer structure. Two models involving a parallel or a perpendicular arrangement of the associated molecules are considered.

From 1H NMR spectra, it appears that the region of overlap occurs near ring V, in agreement with the interpretation of the infrared spectra.  相似文献

Biological slimes (biofilms) collected from the wet end of paper and board machines were examined by electron microscopy and analyzed for fatty acid composition, neutral sugar composition, and ATP. Electron microscopy revealed minuscule prokaryotic organisms (diameter, 0.2 to 0.4 μm). Larger cells morphologically resembling Sphaerotilus and Leptothrix spp. were found in slimes from machines using recycled fiber or unbleached pulp. The bacteria were embedded in a slimy matrix and often contained reserve materials microscopically resembling poly-β-hydroxybutyrate and glycogen. Fatty acid analysis of the slimes revealed bacterial signature fatty acids in concentrations equivalent to the presence of 2 × 1010 to 2.6 × 1012 (average, 7 × 1011) bacterial cells (live and dead) per g (dry weight) of slime. The slimes contained several known components of bacterial polysaccharides in addition to glucose, indicating that the slime body consisted of bacterial polysaccharides. The slimes contained uronic acids equivalent to a binding capacity of 12.5 to 50 μmol of divalent cations per g (dry weight) of slime. The uronic acid-containing polysaccharides may be responsible for the accumulation of heavy metals in the slime. Calculation of the ATP contents of the slimes resulted in an estimate of 5 × 1012 cells per g (dry weight) of slime when calibrated with pure bacterial cultures isolated from the slimes. From electron micrographs, an estimate ranging from 1 × 1010 to 1.5 × 1012 (average, 4 × 1011) cells per g (dry weight) of slime was obtained.  相似文献

龙芝2号和鹿角灵芝均为赤芝的栽培品种,且在生长发育过程中均不产生孢子。目前,尚缺乏对两者化学成分和药理活性的系统比较研究。本研究以灵芝两个无孢品种——龙芝2号和鹿角灵芝为原料,研究对比两者子实体水提物成分差异及免疫活性的强弱。采用化学及仪器分析相结合的方法,分析两者水提物中的多糖得率、含量、重均分子量分布特征及核苷、蛋白质、氨基酸含量差异,并研究了灵芝两个无孢品种水提物样品刺激RAW 264.7细胞释放NO活性。结果表明,两者多糖得率及含量差异不大,但龙芝2号中重均分子量分布范围较广,多糖分子量较大,其含有3种多糖,分子量分别为2.021×106、1.802×106和4.825×105,而鹿角灵芝中仅含有1种多糖,分子量为1.589×104;两者中含有的核苷种类相似,但各核苷的含量存在差异;鹿角灵芝和龙芝2号中蛋白质含量分别为10.70%和10.32%,两者均不含有组氨酸,蛋氨酸在两者中含量均较高,分别达到2.556%和2.591%。从鹿角灵芝和龙芝2号中得到的水提物样品均具有体外刺激巨噬细胞释放NO的活性。  相似文献

Isoamylase catalyzes the hydrolysis of -1,6-glucosidic linkages of starch and related polysaccharides. In this study, the treX gene (GenBank accession no. AE006815 REGION: 9279 … 11435) encoding the thermophilic isoamylase was PCR-cloned from the genomic DNA of Sulfolobus solfataricus ATCC 35092 to an expression vector with a T7lac promoter. Both wild-type and His-tagged isoamylases were expressed in Escherichia coli. The wild-type isoamylase was purified sequentially using heat treatment, nucleic acid precipitation, ion-exchange chromatography, and gel filtration chromatography while the His-tagged isoamylase was purified from the cell-free extract directly by metal chelating chromatography. Both enzymes were active only under their homo-trimer forms. In the absence of NaCl, both enzymes became inactive monomers. In addition, both enzymes were more stable when being stored at room temperature than at 4 °C. They had an apparent optimal pH of 5 and an optimal temperature at 75 °C. The enzyme activities remained unchanged after a 2 h incubation at 80 and 75 °C for the wild-type and His-tagged enzymes, respectively. These thermophilic isoamylases showed a potential to be used in industry to degrade the branching points of starch at a high temperature.  相似文献

Soluble inducers derived from starch were investigated for cellulase production by T. reesei. Various methods of starch treatment were compared. Acid-hydrolysed starch was found to be most effective. When 1·0 g starch was employed for cellulase production, the cellulase so produced after 6 days of fermentation, with supplementation of 1% (0·01 ml/g paper) β-glucosidase, saccharified more than 15 g shredded waste office paper in 9·34% suspension, resulting in 10 g fermentable sugars, 72% of which was glucose. The effectiveness of the starch-derived inducers was compared with that of lactose, pure cellulose, CMC, xylan and prehydrolysate of pine wood. The effects of calcium chloride and sorbose addition on cellulase production, and the kinetics of cellulase production by the starch-derived inducers are presented.  相似文献

The kinetics of malolactic fermentation in Chardonnay wine by immobilised Lactobacillus casei cells has been studied. Calcium pectate gel and chemically modified chitosan beads were used as supports for immobilisation. Repeated batch fermentations were carried out with different wine samples, some of which were treated with sulfur dioxide (free 19–25 mg/litre and total 80–88 mg/litre), in shake flask at 36, 25 and 20°C without any loss of activity. The degradation of malic acid obtained using immobilised cells was twice as high as that obtained with free cells. At an initial pH 3·2, decrease of malic acid of about 30% was observed at 25°C in one hour using L. casei cells immobilised either in pectate gel or on chitosan. Among the physico-chemical parameters studied, temperature was the main factor affecting metabolism of the organic acids as well as the rate of the malolactic fermentation. Operational stability of calcium pectate gel beads and chemically modified chitosan beads was 6 months after eight fermentations and 2 months after five fermentations, respectively, which proved the possibility of industrial application of the chosen supports in wine making.  相似文献

Immobilized preparations of the bacteria (Pseudomonas aeruginosa and Rhodopseudomonas BHU strain 1) and the cyanobacterium ( Anacystis nidulans) exhibited significant Ni adsorption in the order 91%, 72%, 75%, respectively, within 2 h contact with aqueous NiCl2 (7·05 μg Ni/0·1 mg biomass). The immobilizing agent (Ca-alginate, 1·5%, w/v) absorbed more Ni (43%) than the exopolysaccharide of cyanobacteria, Rivularia sp. (40%) or Aphanothece sp. (30%). Ni remobilization from different adsorbed systems was maximum (84%) for Ca(NO3)2 over NaCl (4·3%) at equimolar concentrations (12 m , each). Extracts from forest soil (organic C, 2-3%) were more effective in Ni remobilization (22·65%) than similar preparations from garden soil (18%) with organic C in the range of 0·98-1·1%.  相似文献

1. 1. The effect of temperature on the duration of the egg stage of 3 species of sciomyzid flies was investigated at 14, 17, 20, 23 and 26°C.

2. 2. Ilione albiseta(Scopoli): the mean duration of the egg stage (m.d.e.s) decreased from 88.53 to 34.05 days as temperature increased up to 26°C. Under changing temperature conditions (14↔20°C) there was a significant retardation in the duration of the egg stage. 50–70% of conditioned embryonated eggs (i.e. eggs maintained on moist filter paper from 10 to 20 days at 23°C) hatched within 24 in a 0.1% solution of ascorbic acid. (Embryonation was judged to have taken place when the cephalopharyngeal skeleton was visible through the chorion and there was evidence of larval movement within the egg.)

3. 3. Limnia unguicornis (Scopoli): the m.d.e.s. at the 5 constant temperatures was very variable but was significantly shorter at 26°C (22.85 days) than at 14 and 17°C (29.8 and 32.48 days respectively).

4. 4. Pherbellia cinerella (Fallén): the m.d.e.s. decreased from 14.73 days at 14°C to 4.42 days to 26°C and percentage hatch was greatest at 14°C.

ISOLATION OF AN ACID-SOLUBLE BASIC PROTEIN FROM MONKEY BRAIN   总被引:2,自引:1,他引:1  
—A basic protein, soluble in 0·1 m -perchloric acid, has been purified from brain of Macaca irus. The protein is homogeneous as indicated by ultracentrifugation, gel filtration, gel isoelectric focusing and gel electrophoresis at pH 2·9, 4·3 and 7·5. The molecular weight is estimated to be 16,000 by electrophoresis in sodium dodecyl sulphate–polyacrylamide gels. This result is in agreement with the value of 16,728 obtained from the amino acid analysis. The protein dimerizes under alkaline conditions. The predominant amino acid is glycine (15%) and the protein also contains 4% cysteine. The ratio of acidic to basic amino acids is 1·6, but a high amide content gives the protein a basic character. An isoelectric point of 9·5 is observed in gel isoelectric focusing.  相似文献

A method was developed for applying strains of Actinoplanes spp. that are hyper-parasites of oospores of Pythium ultimum to soil for reducing Pythium damping-off of plants. The method is based on the augmentation of soil with sporangia of a strain of Actinoplanes spp. borne on clay granules. In vitro sporulation of strains K30, W57, W257 and 25844 was: (1) greater for most strains on dilute Czapek-Dox agar than on four other agar media; (2) inhibited by continuous exposure to fluorescent light of intensity 4-150 μEm-2s-1, but not by exposure to 1 μEm-2s-1 or darkness; (3) greater at 20-307deg;C than at 10°C;and (4) greater at pH 6-7 than at pH 5 or 8. On solid carriers treated with dilute Czapek-Dox broth (pH 7) and incubated in the dark at 30°C for 3 weeks, strains sporulated poorly or not at all on vermiculite, perlite and rice hulls, but sporulated abundantly (107-109 colony-forming units (CFU) g-1 of granules) on montmorillonite clay granules. When strains 25844, W57 and W257 were applied as granules (4 107 - 4 × 108 CFU g-1) at 5% (w/w) to field plots infested with 750-1000 oospores of P. ultimum g-1 of soil, only strain 25844 consistently increased emergence and reduced root rot of table beets 8- 1 at 24-28 days after planting compared with controls. Strain 25844 (108 CFU g-1 of granules) at 1% (w/w) also increased the emergence of bush beans at 28 days after planting in P. ultimum-infested plots, but lower rates were ineffective. The inoculum viability of strain 25844 on clay granules declined 100-fold during 2 months of storage at 5-35°C, but thereafter remained stable for another 4 months. Strain 25844 on 6-month-old granules retained a high degree of hyper-parasitic activity toward oospores of P. ultimum. Augmentation of field soil with sporangia of Actinoplanes spp. is a valid approach to the biological control of pythium damping-off.  相似文献

Water-soluble (WSP) and insoluble non-starch polysaccharides (WIP) were isolated from wheat flour to evaluate the effects of WSP and WIP on starch gel properties. Isolated WSP and WIP were added to two types of isolated wheat starch with different amylose content at a concentration of 3% based on the dry weight of starch. 30% starch gels were prepared and stored at 5 °C for 1, 8, or 24 h. The dynamic viscoelasticity of 30% starch gels mixed with WSP and WIP was measured using parallel plate geometry, showing that WSP and WIP affected the elastic component of starch gels in opposite ways. Adding WIP increased the storage shear modulus (G′) of starch gels, while adding WSP decreased G′ and dramatically increased the loss tangent (tan δ=G″/G′).  相似文献
