The effect of Pseudomonas fluorescens treatment and Fusarium oxysporum f. sp. cubense inoculation on induction of phenylalanine ammonia-lyase (PAL), peroxidase (POX), chitinase, -1,3-glucanase and accumulation of phenolics in banana (Musa sp.) was studied. When banana roots were treated with P. fluorescens strain Pf10, a two-fold increase in phenolic content in leaf tissues was recorded 3 – 6 d after treatment. Challenge inoculation with F. oxysporum, the wilt pathogen, steeply increased the phenolic content in P. fluorescens-treated banana plants. Significant increase in POX activity was detected 6 – 9 d after P. fluorescens treatment. PAL, chitinase and -1,3-glucanase activities increased significantly from 3 d after P. fluorescens treatment and reached the maximum 6 d after treatment. Challenge inoculation with F. oxysporum further increased the enzyme activities. These results suggest that the enhanced activities of defense enzymes and elevated content of phenolics may contribute to bioprotection of banana plants against F. oxysporum.  相似文献

7株放线菌在辣椒根部定殖及对辣椒叶片PAL与PPO活性的影响   总被引:17,自引:0,他引:17  
采用盆栽接种试验、平皿涂抹法测数及常规酶活测定法研究了7株拮抗性放线菌在辣椒根部的定殖能力及接种24d对辣椒叶片苯丙氨酸解氨酶(PAl。)与多酚氧化酶活性(PPO)的诱导效应。结果表明:(1)供试7株放线菌单独接种均不能在辣椒根内定殖,但与辣椒疫霉P3混合接种时有5株可定殖;供试放线菌在辣椒根部的定殖能力与其体外平皿试验中产生的的拈抗圈大小基本无关;可定殖放线菌的定殖密度随时间延长而降低,至40d时均无活菌检出。(2)在放线菌单接处理中,5株菌接种后可诱导辣椒叶片PAL,活性提高,全部供试菌均能诱导PPO活性提高,其中可使两种酶同步提高的有5株菌;在放线菌+P3混接处理中,有6株接种后可诱导PAL,活性提高,5株菌能诱导PPO活性提高,其中可使两种酶同步提高的有4株菌;在接入放线菌时同时混接辣椒疫霉,能增强2株供试放线菌对辣椒叶片PAL活性及6株供试放线菌对辣椒叶片PPO活性的诱导作用;供试放线菌的定殖能力与辣椒叶片PAL及PPO活性变化无明显规律性关系。  相似文献

广藿香是著名药材,青枯病是威胁广藿香生产和质量的主要病害。针对从广藿香茎叶中分离出的1株有抑菌活性的内生真菌Alternaria sp. GHX-P17,研究其代谢产物对广藿香青枯病的防治作用及耐病机制。在室内通过人工接种Alternaria sp. GHX-P17菌株与喷施粗提物稀释液2种方式,于不同时间调查广藿香青枯病的发病率和严重度,并计算其病情指数(disease index,DI)。同时,对处理后不同时间的广藿香苯丙氨酸解氨酶(phenylalanine ammonia lyase,PAL)、过氧化物酶(peroxidase,POD)和超氧化物歧化酶(superoxide dismutase,SOD)3种保护酶活性进行测定。结果表明:(1)Alternaria sp. GHX-P17处理后青枯病DI显著降低,与对照比较,204 h后DI降低为27.16%,方差分析呈显著性差异(P<0.05)。(2)随着处理时间延长,青枯病严重度升高缓慢,严重度等级降低,到204 h时,处理组的严重度明显低于对照,防治效果达到74.65%。(3)处理后的广藿香3种保护酶PAL、POD和SOD活性增强,但3种酶活性高峰出现时间不同。PAL随时间逐渐升高; POD先升高后降低,然后又升高,出现2个峰值; SOD快速升高后逐渐降低。这表明内生真菌菌株Alternaria sp. GHX-P17可以提高广藿香3种保护酶活性,延缓了青枯菌的侵染过程,降低了青枯病的发病程度。该研究结果为植物内生真菌次级代谢产物活性成分研究及生物农药开发提供了参考。  相似文献

The activities of the enzymes responsible for cell-wall strengthening and salicylic acid (SA) content in Norway spruce seedlings were investigated after inoculation with the ectomycorrhizal fungus Pisolithus tinctorius or the pathogen Heterobasidion annosum, and after treatment with elicitors from both of these fungi. Inoculation with both fungi increased guaiacol peroxidase (POD) activity in the roots of the pathogen-inoculated seedlings during the earliest phases of colonisation, and induced the activities of several POD isoforms. Two of these were only seen in pathogen-inoculated seedlings and corresponded with increased POD activity against ferulic acid. Colonisation with H. annosum triggered an increase in phenylalanine ammonia lyase (PAL) activity in the roots of the spruce seedlings, which was followed by an accumulation of free SA. One month after inoculation levels of free SA were increased also in the shoots of H. annosum-inoculated seedlings. In contrast increase in free SA content in the roots of P. tinctorius-inoculated seedlings was only transient. Similarly to inoculation, treatment with elicitors of H. annosum increased the PAL and POD activity, as well as SA content in the roots of spruce seedlings. A positive correlation between PAL activity and SA content in the H. annosum-inoculated seedlings and accumulation of SA precursors in the phenylpropanoid pathway indicate that the plant defence mechanisms, during which SA is synthesised through the PAL pathway, are exploited by H. annosum for facilitation of colonisation.  相似文献

Sheath blight disease caused by Rhizoctonia solani Kuhn is becoming a major constraint to rice production, especially in the intensified cultivation system. To know the in rice, it is important to get the knowledge of the activity of defence-related enzymes due to the fungal infection. The pathogen induced superoxide dismutase (SOD) and chitinase activities in rice plants, while suppressing peroxidase (POD) and phenylalanine ammonia-lyase (PAL) activities at 36 and 24 h after inoculation, respectively. Induction of two POD isozymes, POD-3 and -4, up to 48 h after inoculation and disappearance of the said isomers at 72 h onwards in rice–Rhizoctonia interaction implicated the role of these isomers in susceptible host–pathogen interaction. Apart from POD and SOD, the activities of other stress-related enzymes, viz. PAL, polyphenol oxidase (PPO) and β-1,3-glucanase were also studied. From this study, it was found that these defence-related enzymes are most significantly related to host–pathogenic interaction.  相似文献

Accumulation of extracellular chitinases in Brassica napus plants infected with Turnip yellow mosaic virus (TYMV) and fungal pathogen Leptosphaeria maculans was studied in both compatible and incompatible interaction. Analysis of apoplast fluid by means of non-denaturing anodic and cathodic PAGE followed by in-gel detection of chitinase activity revealed a number of chitinase isozymes. TYMV induced 8 acidic and 4 basic isozymes in a systemic way. Except for one acidic and one basic isozyme, all other chitinases were also constitutively present in low amounts in mock inoculated control. In TYMV systemically infected plants, chitinases were detected in leaves expressing symptoms as well as in symptomless ones. Both virulent and avirulent L. maculans isolates induced production of chitinase isozymes in cotyledons in a time dependent manner. Some of them were present in plants constitutively and their content increased after inoculation. Three of five acidic and two of three basic isozymes responded to L. maculans infection. Chitinases started to accumulate before symptom appearance. First two acidic isozymes were detected 24 h after inoculation. The difference between compatible and incompatibe interaction reflected two basic isozymes.  相似文献

In this paper, we describe a study concerning the determination of phenylalanine ammonia lyase (PAL) activities leading to decline in disease progress caused by Rhizoctonia solani after application of salicylic acid (SA). Two applications of 1.4 mM SA (pH 6.5) followed by inoculation with Rhizoctonia solani resulted in a quantitative change in polyphenol oxidase (PPO), peroxidase (PRX) isoforms and increase in PAL activities from 4.38 to 19.48 unit g−1 (FM) h−1 in Bundel-1, UPC-4200 and IFC-902 cowpea genotypes. Increase in PAL activities was further observed specifically in UPC-4200 when plants were exposed with Rhizoctonia solani spores. Total soluble proteins did not change after SA treatment, however they were significantly increased in SA sprayed-inoculated UPC-4200 genotype. Of the ten detected isoforms of polyphenol oxidase, isoforms 7 and 10, and isoform 4 of peroxidase showed increased activities by SA application. The disease symptoms measured as areas under progress curve (AUDPC) indicated less A value in SA sprayed Bundel-1 and UPC-4200 genotypes over their controls.  相似文献

The induction of defence compounds and enzymes involved in the phenylpropanoid pathway were studied in the ripe and green chilli fruits inoculated with Colletotrichum capsici and Alternaria alternata. Total phenols and the activity of phenylalanine ammonia lyase (PAL), peroxidase (PO), polyphenol oxidase (PPO) and catalase (CAT) increased in the inoculated ripe and green chilli fruits compared to the corresponding healthy fruits. Total phenols and the activities of the enzymes were at the maximum 2–3 d after inoculation and thereafter declined sharply in ripe chilli fruits, whereas slowly in green chilli fruits. In comparison with ripe chilli fruits, green chilli fruits were more resistant as they showed higher accumulation of total phenols and also higher activities of enzymes.  相似文献

This study, framed in two different phases, studied the plant-growth promotion and the induction of systemic resistance in groundnut by Methylobacterium. Seed imbibition with Methylobacterium sp. increased germination by 19.5% compared with controls. Combined inoculation of Methylobacterium sp. with Rhizobium sp. also significantly increased plant growth, nodulation, and yield attributes in groundnut compared with individual inoculation of Rhizobium sp. Methylobacterium sp. challenge-inoculated with Aspergillus niger/Sclerotium rolfsii in groundnut significantly enhanced germination percentage and seedling vigour and showed increased phenylalanine ammonia lyase (PAL), β-1,3-glucanase, and peroxidase (PO) activities. Under pot-culture conditions, in Methylobacterium sp. seed—treated groundnut plants challenge-inoculated with A. niger/S. rolfsii through foliar sprays on day 30, the activities of enzymes PO, PAL, and β-1,3-glucanase increased constantly from 24 to 72 hours, after which decreased activity was noted. Five isozymes of polyphenol oxidase and PO could be detected in Methylobacterium-treated plants challenged with A. niger/S. rolfsii. Induced systemic resistance activity in groundnut against rot pathogens in response to methylotrophic bacteria suggests the possibility that pink-pigmented facultative methylotrophic bacteria might be used as a means of biologic disease control.  相似文献

Abstract

The effect of Xanthomonas oryzae pv. oryzae infection on induction of phenylalanine ammonia-lyase (PAL), peroxidase (PO), phenolics and thaumatin-like proteins (TLPs) in rice was studied. PAL activity increased significantly one day after inoculation with X. o. pv. oryzae and the maximum enzyme activity was observed two days after inoculation. The phenolic content in rice leaves increased significantly one day after inoculation and the maximum accumulation of phenols was observed two days after inoculation. Significant increase in peroxidase activity was observed in rice leaves one day after inoculation with X. o. pv. oryzae. Isozyme analysis indicated that three peroxidase isozymes (PO-1, PO-2 and PO-3) were induced after inoculation with X. o. pv. oryzae. Immunoblot analysis of protein extracts from control and pathogen inoculated rice plants revealed the induced accumulation of 16 and 24 kDa TLPs in rice leaves in response to X. o. pv. oryzae infection. TLP mRNA accumulation was induced strongly in rice leaves in response to infection by X. o. pv. oryzae.  相似文献

The defence reactions of sorghum seedlings 7 days after inoculation with Fusarium thapsinum and F. proliferatum, and interactions with wounding and exposure to light were studied to determine whether responses to these fungi differed from those to abiotic stresses. In non‐wounded plants, inoculation with both fungi increased concentrations of anthocyanins and soluble phenolics and activities of peroxidase (POX), chitinase and β‐1,3‐glucanase in the roots, and increased β‐1,3‐glucanase activity in the mesocotyls. There was no effect of inoculation on phenylalanine ammonia‐lyase (PAL) activity. Wounding by itself increased anthocyanin content of mesocotyls. Wounding also had a variety of interactions with inoculation. Exposure to light had very little effect on any defence response measured. A time course experiment showed that induction of chitinase and β‐1,3‐glucanase occurred in less than 24 h after inoculation. POX activity increased 2 days after inoculation, followed by a transient increase in PAL activity. The content of anthocyanins and soluble phenolics in roots of inoculated seedlings increased gradually compared with controls over 6 days. The responses of sorghum seedlings to inoculation with F. thapsinum and F. proliferatum were similar to those found by other workers following challenge by necrotrophic pathogens and were different from those induced by wounding and exposure to light.  相似文献

A comparative study on the extracellular ligninolytic enzymatic activity of five strains of Fusarium solani in a carbon-limited medium under shaking, revealed a differential production of these enzymes. Aryl alcohol oxidase (AAO) activity was observed only in the supernatant of strain CLPS no. 568 with levels higher than 57 mU ml−1. Free extracellular laccase activity was detected in strains CLPS nos. 493, 568 and 570, strain no. 568 being the one which showed the highest activity (over 8.6 mU ml−1). Free extracellular lignin peroxidase (LiP) activity was not detected in any isolate tested, whereas low levels of manganese-dependent peroxidase (MnP) and manganese-independent peroxidase (MIP) activities were detected in certain isolates used. The AAO activity of F. solani on primary α-alcohols such as veratryl alcohol, is reported for the first time; this enzyme activity is hydrogen-peroxide independent. Treatment with an autoclaved culture homogenate of the yeastRhodotorula rubra induces rapid accumulation of acridone epoxides, furoquinolines and furanocoumarins in cell cultures ofRuta graveolens (L). The increased accumulation is preceeded by an induction of enzymes of the biosynthetic pathways. In the case of furanocoumarins induction was shown for phenylalanine ammonia-lyase (PAL), 4-coumarate: CoA ligase (4-CL) and S-adenosyl-l-methionine: xanthotoxol O-methyltransferase (XOMT). For PAL and 4-CL time courses of induced activity showed an early maximum, 8–12 h after treatment, whereas XOMT was found to reach its maximum later, about 36–42 h after treatment. The elicitor dose-response curve showed saturation at an elicitor concentration of 1%. At any time during the whole culturing period cells responded to elicitiation but the maximum enzyme activities induced were lower at the late stages. Experiments with different suspension culture strains, a shoot teratoma culture and hydroponically grown sterile photomixotrophic plants were performed to assess the influence of differentiation on constitutive activities of these enzymes and their inducibility by elicitation. Constitutive furanocoumarin accumulation was positively correlated with the level of differentiation. The activities of chitinase, β-1,3-glucanase, peroxidase and phenylalanine ammonia lyase, constitutive and induced by Stagonospora nodorum were examined in the 10 – 14 day old seedlings of three triticale and two wheat cultivars under controlled environmental conditions and in flag leaves of two triticale cultivars in the field. Two S. nodorum isolates of different virulence were used. Both the constitutive and induced activities in triticale and wheat depended on genotype and in triticale the effect of growth conditions was also evidenced. The constitutive activities of chitinase, β-1,3-glucanase and peroxidase were several fold lower in flag triticale leaves in plants from the field than in the seedlings, growing under controlled conditions, but induction in the infected flag leaves was significantly more pronounced. In triticale genotypic differences in the response to infection were revealed only upon inoculation by S. nodorum isolate of higher virulence. The enzymatic activities increased several fold during successive days after the infection except for phenylalanine ammonia lyase. Induction of this enzyme was only transient and the activity decreased 48 or 96 h after infection when the activities of other enzymes were rising. In flag leaves in the field this activity was differentiated only after infection with more a virulent strain. A tendency appeared in triticale seedlings for association of the resistance to the pathogen with lower enzymatic constitutive activities. This relationship became more evident in triticale infected by S. nodorum and may imply that although the investigated enzymes are certainly involved in general, non-specific defense mechanism, they do not decide on the resistance to pathogen at least in the early stages of infection and cooperate with other factors in the complex pathogen-plant interaction. One can also assume that the enzymatic activities are associated with severity of infection rather than resistance to pathogen.  相似文献

Exogenous foliar application of β-aminobutyric acid (BABA) led to a significant reduction in disease severity in Brassica carinata caused by Alternaria brassicae. To get a better insight about changes in defence-related enzymes like phenylalanine ammonia lyase (PAL) and polyphenol oxidase (PPO), isoform analysis of superoxide dismutase (SOD) and peroxidase (POX) were studied. BABA-treated plants showed a significant increase in PAL, PPO enzyme activities and total phenolic content in response to pathogen inoculation. However, isoform analysis of SOD and POX revealed no change in isoform number but a quantitative change in activity was observed in response to pathogen.  相似文献

Chlorate or trimethylamine-N-oxide (TMAO) added to phototrophic cultures ofRhodobacter sphaeroides DSM 158 increased both the growth rate and the growth yield although this stimulation was not observed in the presence of tungstate. This strain, exhibited basal activities of nitrate, chlorate, and TMAO reductases independently of the presence of these substrates in the culture medium, and nitrate reductase (NR) activity was competitively inhibited by chlorate. Phototrophic growth ofRhodobacter capsulatus B10, a strain devoid of NR activity, was inhibited only by 100 mM chlorate. However, growth of the nitrate-assimilatingR. capsulatus strains E1F1 and AD2 was sensitive to 10mm chlorate, and their NR activities were not inhibited by chlorate. Both NR and chlorate reductase (CR) activities of strain E1F1 were induced in the presence of nitrate or chlorate respectively, whereas strain AD2 showed basal levels of these activities in the absence of the substrates. A basal TMAO reductase (TR) activity was also observed when these strains ofR. capsulatus were cultured in the absence of this electron acceptor. These results suggest that chlorate and TMAO can be used as ancillary oxidants byRhodobacter strains and that a single enzyme could be responsible for nitrate and chlorate reduction inR. sphaeroides DSM 158, whereas these reactions are catalyzed by two different enzymes inR. capsulatus E1F1 and AD2.  相似文献

以苹果树腐烂病菌LXS080601、感病苹果品种‘富士’和抗病砧木‘平邑甜茶’愈伤组织为材料,测定腐烂病菌侵染后,愈伤组织内过氧化物酶(POD)、多酚氧化酶(PPO)、超氧化物歧化酶(SOD)和苯丙氨酸解氨酶(PAL)活性及丙二醛(MDA)含量的动态变化。结果显示,接种LXS080601后,‘富士’愈伤组织的发病严重度和病情指数均明显大于‘平邑甜茶’;感病品种MDA含量上升速度快,于接种后3 d增幅为28.02%,且变幅较大,为–0.32%~36.39%,而抗病砧木MDA含量变化较小,仅为–2.17%~7.46%。同时,腐烂病菌侵染提高了愈伤组织内4种防御酶活性,接种后1~2 d,PPO和POD酶活性达到高峰,接种后3~4 d,PAL和SOD酶到达活性高峰;除PPO外,‘平邑甜茶’PAL、SOD和POD酶活性变化均明显高于‘富士’,且整个侵染过程酶活性维持在较高水平,而‘富士’体内3种酶活性快速下降至对照水平,表明‘平邑甜茶’通过提高抗氧化酶活性减少体内活性氧的积累,降低膜脂过氧化产物MDA的形成,增强了对腐烂病菌侵染的抗性。  相似文献

Abstract

The effect of endophytic Pseudomonas fluorescens isolates Endo2 and Endo35 on induced systemic disease protection against dry root rot of black gram (Vigna mungo L. Hepper) caused by Macrophomina phaseolina was investigated under glasshouse conditions. When the bacterized black gram plants were inoculated with dry root rot pathogen, the activities of peroxidase (PO), polyphenol oxidase (PPO), phenylalanine ammonia-lyase (PAL) were stimulated in addition to accumulation of phenolics and lignin. Activity of phenylalanine ammonia-lyase (PAL) reached the maximum 24 h after pathogen challenge inoculation, whereas the activities of PO and PPO reached the maximum at 72 h and 48 h, respectively. Isoform analysis revealed that a unique PPO3 isozyme was induced in bacterized black gram tissues inoculated with the pathogen. Phenolics were found to accumulate in bacterized black gram tissues challenged with M. phaseolina one day after pathogen challenge. The accumulation of phenolics reached maximum at the third day after pathogen inoculation. Similar observation was found in the lignin content of black gram plants. In untreated control plants, the accumulation of defence enzymes and chemicals started at the first day and drastically decreased 3 days after pathogen inoculation. These results suggest that induction of defense enzymes involved in phenylpropanoid pathway and accumulation of phenolics and PR-proteins might have contributed to restricting invasion of Macrophomina phaseolina in black gram roots.  相似文献
