Transplanted Morris hepatomas in Buffalo-strain rats were found to be resistant to the changes in ribonucleotide levels in rat liver caused by a high-orotate diet or an arginine-deficient diet. The increase in UTP levels and decrease in ATP levels seen in the livers of rats on a 1%-orotate diet were less marked in the livers of BUB- and DBA-strain mice on this diet. Although the changes were less than in rat liver, there was a 2-3-fold increase in UTP concentration in the livers of mice on the high-orotate diet. However, there was a similar response in nucleotide levels in the two species when the animals were maintained on an arginine-deficient diet, and there was a greater than 10-fold increase in the UTP level in the livers of both rats and mice. These diets had much less effect on the levels of deoxyribonucleotides than of ribonucleotides. In contrast to the insensitivity of hepatomas to dietary modulation of nucleotide levels, treatment of hepatoma-bearing rats with carbamoylating agents (sodium cyanate and 2-chloroethyl isocyanate) caused decreases in the levels of nucleotides in the tumors which were generally greater than in host livers. For example, 2-chloroethyl isocyanate depressed ATP levels in the Morris hepatomas 5123C and 20 under conditions in which there was no significant effect on host liver ATP. The data revealed selective modulation of nucleotide levels in normal and neoplastic liver which may be achieved by either dietary modification or drug treatment. 相似文献

Iron-sulfur centers in mitochondria prepared from Morris hepatomas with different growth rates were compared with those in host liver and nontumor-bearing rat liver mitochondria by EPR measurements (< 77° K). In the slow growing hepatoma 16, EPR signals from iron-sulfur centers located in the NADH dehydrogenase region were specifically diminished. In the rapidly growing hepatoma 7777, EPR signals of all the iron-sulfur centers showed considerably diminished intensity. In hepatoma 7800 having an intermediate growth rate, all iron-sulfur centers showed no change. Those changes in iron-sulfur centers correlated with observed respiratory activities of Morris hepatoma mitochondria. No general correlation was obtained between these parameters and the growth rate of the tumors. 相似文献

Three pot experiments, in which causes of negative yield responses to liming were investigated, are reported. The soil used,
Waimumu silt loam (Fragiochrept), differed from others that have been reported to show negative yield response to liming,
in that it is only moderately weathered and leached, only moderately acid and has previously shown positive yield responses
to liming. Deficiencies of Zn and Mg were identified, but limed (pH 6.8) soils still showed a 40% yield depression even where
all nutrients were supplied daily. Phosphorus availability was little affected by liming, and despite Mg and Zn addition,
yields were depressed at high lime (pH 6.4) and high P while plants showed leaf symptoms of Mg deficiency. Neither plant nor
soil analyses indicated low Mg levels but Ca: Mg ratios in soil were 22:1. When a pH range 5.2–6.1 was produced by liming
with CaCO3 and MgCO3 at ratios between 100:0, and 50:50 on an equivalent basis, negative yield response was eliminated at Ca:Mg of 50:50. There
was no evidence that Mg was fixed or rendered unavailable at the higher pH levels. A Ca induced Mg deficiency arising when
exchangeable Ca:Mg>20 is suggested as the cause. The role of variable surface charge in converting soils that respond positively

Steady-state populations of Escherichia coli B/r were treated with cephaloridine at minimal inhibitory concentrations. The antibiotic sensitivity of the cells and the localization of spheroplast emergence along the cell surface were examined as a function of cell length and growth rate. In fast-growing populations (greater than 1 division per h) the sites of cephaloridine interaction occurred preferentially at the cell pole in the smaller cells and at the cell center in dividing cells. At decreasing growth rates the cells became more resistant to cephaloridine, and a gradual shift from the cell pole toward the cell center was observed for the sphere position. A similar growth rate-dependent change in localization was found for sucrose-induced plasmolysis vacuoles. 相似文献

Plasticity of insect reproduction: testing models of flexible and fixed development in response to different growth rates 总被引:3,自引:0,他引:3
We tested alternative developmental hypotheses describing when during an insect oviposition cycle reproductive tactics are
determined. Newly eclosed adult females of the grasshopper Romalea guttata were raised on eight different feeding treatments consisting of a low food diet, a high food diet, and changes from high
to low food, or low to high food, at different times during the first oviposition cycle. When initial food availability was
high, a decline in food availability >7 days after adult eclosion produced no significant increase in time to oviposition
compared to constant high food. In contrast, when initial food availability was low, an increase in food availability as late
as day 14 produced a significant decrease in time to oviposition compared to constant low food. Thus, time to oviposition
is determined by feeding rate early in the oviposition cycle, but the time of this determination is later when food availability
is lower. Masses of individual eggs were unaffected by these treatments. When initial food availability was high, a decrease
in food availability on day 21 produced no significant change in numbers of eggs in a clutch compared to constant high food.
In contrast, when initial food availability was low, an increase in food availability after day 7 produced no significant
change in number of eggs in a clutch compared to constant low food. Changes in egg production resulted from oocyte resorption,
which appeared to become unresponsive to food availability between day 14 and day 21. Our results refute the hypothesis that
reproductive tactics are continuously flexible. Development toward oviposition seems to be structured so that reproductive
tactics become independent of feeding late during the first oviposition cycle. Reproductive tactics become unresponsive to
food at different times for groups initially receiving low or high food, suggesting that a particular developmental state,
rather than some absolute time, marks the shift to development that is unresponsive to␣food. Plasticity in reproductive tactics
appears to be␣controlled by hormones in a manner similar to the hormonal control of plasticity of metamorphosis in other insects.

The activities of glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase and transketolase were studied in the cytoplasmic fractions of transplanted mouse hepatomas differing in their growth rates, and in the liver, spleen and cortical layer of kidneys of tumour carriers and normal mice. It was shown that transplantation of hepatomas changes the activity of the pentose phosphate pathway enzymes in tumour carrier tissues unaffected by neoplasm. Deviations from normalcy were mainly similar to those observed in the hepatomas. The changes in the enzymatic activities were especially well-pronounced in the mice having rapidly growing hepatomas. This may be due to a generalized effect of the tumour on the organism, which is concurrent with malignancy. 相似文献

Photosynthesis and growth of winter wheat in response to waterlogging at different growth stages 总被引:3,自引:0,他引:3
A study on photosynthetic and yield effects of waterlogging of winter wheat at four stages of growth was conducted in specially designed experimental tanks during the 2007–2008 and 2008–2009 seasons. Compared with the control, waterlogging treatments at tillering and jointing-booting stages reduced photosynthetic rate (P N) and transpiration (E) significantly, it also decreased average leaf water-use efficiency (WUE, defined as the ratio of P N to E) by 3.3% and 3.4% in both years. All parameters returned quickly to the control level after soil was drained. Damage to the photosynthetic apparatus during waterlogging resulted in a lower Fv/Fm ratio, especially at the first two stages. A strong reduction in root length, root mass, root/shoot ratio, total dry mass, and leaf area index were observed. The responses from vegetative plants at tillering and jointing-booting stages were greater than in generative plants at onset of flowering and at milky stages. The number of panicles per hectare at tillering stage and the spikelet per panicle at the stages of jointing-booting and at onset of flowering were also significantly reduced by waterlogging, giving 8.2–11.3% decrease of the grain yield relative to the control in both years. No significant difference in yield components and a grain yield was observed between the control and treatments applied at milky stages. These responses, modulated by the environmental conditions prevailing during and after waterlogging, included negative effects on the growth, photosynthetic apparatus, and the grain yield in winter wheat, but the effect was strongly stage-dependent. 相似文献

The levels of ribosomes, tRNA molecules, and total protein per genome in Neurospora mycelia have been determined in eight different conditions of exponential growth. By increasing the rate of growth the number of ribosomes per genome increases dramatically while the level of total protein remains almost unchanged and the level of tRNA increases only slightly. The rates of synthesis of each of the macromolecules have been estimated. Increasing the rate of growth (mu) up to 0.5, the ratio between the rates of synthesis of tRNA and rRNA decreases reaching a constant value. The equations that best describe the dependence of the rate of synthesis of the macromolecules on the rate of growth (mu) have been determined. The rate of rRNA synthesis (rr), expressed as nucleotides polymerized, min- minus 1 per genome, is given by the equation: rr equals 6.51 times 10-7 mu-2-19. The rate of protein synthesis (rp), expressed as amino acids polymerized, min- minus 1 per genome is given by the following relationship: rp equals -1.43 times 10-7 + 3.43 times 10-8 mu. The equation describing the tRNA synthesis (rt) expressed as nucleotides, min- minus 1 per genome is rt equals 6.45 times 10-5 times exp 2.30 mu; however, more accurate determinations appear to be required for a firmer assignment of this latter equation. The significance of these equations for the studies on the regulation of rRNA and protein synthesis is discussed. For instance the rate of rRNA synthesis may set the limit for the maximal growth rate attainable by a cell, as the maximal rate of rRNA synthesis that may take place in a given cell is limited by the degree of redundancy of the rRNA genes. 相似文献

Guanylate cyclase (GTP pyrophyosphate-lyase (cyclizing), EC 4.6.1.2) activity was examined in preparations from normal rat liver and a series of Morris hepatomas. Homogenate gyanylate cyclase activites were 3.2, 1.6 and 1.2 nmol cyclic GMP formed per min/g tissue ihe non-substrate analogs of IMP were weak inhibitors of this enzyme, GMP and four of its analogs had Ki values ranging from 30 to 80 muM. Experiments were conducted in an outdoor facility to quantify growth responses of six mangrove species to rates of dissolved inorganic nitrogen and phosphorus supply mimicking the range of N and P mineralization rates in natural soils. Growth of all six species on nitrogen was nonlinear. Stem extension rates of Rhizophora apiculata and Xylocarpus granatum were enhanced to the highest rate of N supply (50 mmol m− 2 d− 1); Bruguiera gymnorrhiza, Avicennia marina, and Xylocarpus moluccensis stem growth leveled off by 10 mmol m− 2 d− 1. Stem growth of Ceriops tagal peaked at 24-26 mmol N m− 2 d− 1. Except for A. marina and C. tagal, rates of biomass increase declined at the highest supply rate, indicating NH4+ toxicity. At different rates of P supply, stem extension rates and rates of biomass increase of R. apiculata and C. tagal best-fit Gaussian curves and B. gymnorrhiza stem growth and biomass increase best-fit sigmoidal and Gaussian curves, respectively; X. moluccensis stem and biomass growth increased linearly, but stem and biomass growth rates of A. marina did not vary in relation to P supply. Stem growth of X. granatum was Gaussian but rates of biomass increase best-fit a quadratic equation. Changes in leaf and root N and P content mirrored the growth responses. As rates of N and P mineralization in natural mangrove soils overlap with the lowest rates of N and P supplied in these experiments, the growth responses imply that mangroves are intrinsically nutrient-limited at mineralization rates often encountered in nature. Such species specificity may have significant implications for recruitment success and the establishment of species gradients within mangrove forests. 相似文献

Sickle-cell anemia results from an A leads to T transversion in the second nucleotide of codon 6 of the beta-globin gene. We now report an uncommon beta-thalassemia gene that contains a deletion of this nucleotide. Thus, one mutation (GAG leads to GTG) produces sickle-cell anemia, while the other (GAG leads to GG) eliminates beta-globin production. These data establish that different alterations affecting one specific nucleotide can produce either an abnormal hemoglobin or beta-thalassemia. Moreover, the nucleotide sequence comprising codons 6-8 of the beta-globin gene appears to be particularly susceptible to mutations affecting nucleotide number. 相似文献

Characterization of three different elements in the 5'-flanking region of the fibronectin gene which mediate a transcriptional response to cAMP. 总被引:5,自引:0,他引:5
A cAMP regulatory element (CRE) at nucleotide position -170 of the fibronectin gene was characterized previously (Dean, D. C., Blakeley, M. S., Newby, R. F., Ghazal, P., Hennighausen, L., and Bourgeois, S. (1989) Mol. Cell. Biol. 9, 1498-1506). Here we identify two additional low affinity CREs at nucleotide positions -260 and -415 which differ in sequence by 1 base pair. Interestingly, these CREs did not compete for binding of nuclear proteins in gel retardation assays and partial tryptic digestion of protein-DNA complexes produced a different pattern with each CRE, indicating that they bind different proteins. CRE (-170) competed for binding of proteins to both CREs, suggesting that it may represent a composite of the two elements. CRE (-415) competed effectively for binding of nuclear proteins to the somatostatin gene CRE, suggesting that, like the somatostatin CRE, it binds the nuclear protein CREB. On the other hand, CRE (-260) appears to bind the nuclear protein PEA-2, which also binds a site in the polyoma virus enhancer. In summary, disruption of dyad symmetry in the 3' region of the CRE, as occurs with CRE (-260) and CRE (-415), results in a lower affinity site and may also change the specificity for different nuclear proteins. 相似文献
