Phycobilisomes (PBS) isolated from Nostoc muscorum contain eight spectral forms of phycobiliproteins. PBS partially dissociated by osmotic or temperature shock, were separated into 42S and 34S particles by centrifugation in sucrose density gradient. The 42S particles are enriched with phycocyanin, the 34S ones--with allophycocyanin. The 42S particles dissociate to free pigments by repeated osmotic or temperature shock and the 34S ones dissociate to subparticles I and II having the constant pigments composition. The integrity of PBS and isolated substructures is controlled by the yield of energy transfer between pigment molecules. PBS from Nostoc muscorum was found to contain the rods of three different types. The model PBS containing different spectral forms of biliproteins is proposed.  相似文献

Malondialdehyde (MDA), an in vivo metabolite of lipid peroxidation and prostaglandin biosynthesis, is mutagenic in Salmonella typhimurium. It is a reactive electrophile that can form interstrand cross-links in DNA. To explore the possibility that MDA-induced interstrand cross-links are the pre-mutagenic lesion, we have quantitated the ability of highly purified preparations of MDA to form interstrand cross-links when reacted with linear plasmid DNA. At physiological temperature and pH, MDA did not form DNA cross-links as determined by DNA denaturation followed by agarose gel electrophoresis. DNA cross-links were formed, however, when incubations with MDA were carried out at either pH 4.2 or temperatures exceeding 60 degrees. alpha-Methylmalondialdehyde (CH3MDA) was found to cross-link DNA more efficiently than MDA, but was not mutagenic in any tester strain of Salmonella. MDA polymers, formed by acid incubation of MDA, also were capable of inducing cross-links. However, an inverse relationship was observed between mutagenicity and extent of polymerization. The pattern of mutagenic response for MDA in different strains of Salmonella was compared with mitomycin C, an established mutagenic cross-linking agent. Error-prone repair and a UvrB+ phenotype, which are needed for the induction of mutations by mitomycin C, were not required for MDA mutagenesis. These findings, taken together, dissociate the mutagenicity of MDA from its ability to form interstrand cross-links with DNA.  相似文献

Spontaneous chlorate-resistant (Clr) mutants of three classes were isolated from Nostoc muscorum under three different selective conditions. A Clr-N2 class of mutants lacked nitrate reductase and showed nitrate inhibition of nitrogen fixation. A Clr-NO3 group of het+ nif- mutants formed heterocysts, but lacked nitrogen fixation and active nitrogenase enzyme. The Clr-NO2 class included those mutants deficient in both active nitrogenase and nitrate reductase, as they were unable to grow at the expense of molecular nitrogen or with nitrate nitrogen. The results suggest a common genetic determinant of active nitrogenase and nitrate reductase in the blue-green alga N. muscorum.  相似文献

Summary A model has been proposed to account for growth inhibition by L-histidine in a variant strain of Nostoc muscorum. This strain has been characterized for its response to 3-amino-1,2,4-triazole and 1,2,4-triazole-3-alanine known to act as false corepressors of the histidine biosynthesis genes. The histidine sensitive strain retained its sensitivity to triazole alanine while the inhibitory effects of aminotriazole were much reduced indicating a change in regulation of his genes. The probable interactions between nif and his genes in cyanobacteria (blue-green algae) have been discussed.  相似文献

The effects of various levels of nutrients like potassium phosphate (dibasic), calcium nitrate, and calcium chloride individually and in combinations were studied on the toxicity of the commonly used pesticides carbofuran and hexachlorocyclohexane (HCH) in growth medium to the N2-fixing blue-green alga Nostoc muscorum. It was observed that all these chemicals had some effects on toxicity. The toxicity of both carbofuran and HCH was reduced to some extent in the presence of higher concentrations of the nutrients when compared to normal levels of the chemicals in the medium. The higher doses of nutrients in combinations antagonized the effect of individual treatment and enhanced the toxicity of pesticides.  相似文献

Preincubation of the blue-green alga (cyanobacterium) Nostoc muscorum under an atmosphere of argon plus acetylene in the light led to a greater than fourfold increase of light-induced hydrogen evolution and to a 50% increase of acetylene reduction, as compared to cells that had not been preconditioned. The basic and the increased hydrogen evolution were both due to nitrogenase activity. Furthermore, after preincubation the hydrogen uptake, usually observed with unconditional cells, was abolished. Nostoc preincubated under acetylene evolved hydrogen in the light even in the presence of nitrogen for at least 2 h, with a 15-fold increase as compared to the unconditioned cells. These acetylene effects could be completely abolished by the presence of hydrogen during acetylene preincubation. These findings indicate that the hydrogen concentration in N. muscorum cells plays a role in regulation of nitrogenase activity.  相似文献

Phycobilisomes were isolated from a Nostoc sp. strain Mac in phosphate buffer (pH 7.0) by treatment with 1% Brij 56 and centrifugation on discontinuous sucrose gradients (2.0, 1.0, 0.5, and 0.25 M in the proportions 6:4:4:10 ml, respectively). Absorption spectra of isolated phycobilisomes showed the presence of phycoerythrin, phycocyanin, and allophycocyanin. The phycobilisome pigments were partially resolved by electrophoresis on acrylamide gels. Stained gels demonstrated that each main protein band corresponded to a pigmented region. The phycobilisomes appeared compact with a rounded surface and flattened base (about 40-nm diameter) at the attachment site to the photosynthetic lamellae. Fixation in glutaraldehyde caused a significant reduction in total pigment absorption, as well as shifts in the absorption maxima, particularly that of phycoerythrin.  相似文献

Heterocysts of the blue-green alga Nostoc muscorum have been isolated by prolonged treatment with lysozyme. Quantitative data are presented which show the occurrence of cytochromes c-553, f-557 and b-563 in heterocysts in amounts comparable to vegetative cells. Particularly the content of the water-soluble cytochrome c-553 can be used to evaluate the intactness of a heterocyst preparation. Cytochrome f-557 has been partially purified and found to be a c-type cytochrome corresponding to cytochrome f of higher plants and other algae. Cytochrome b-559 is present in vegetative cells but not in heterocysts. The content of plastoquinone in heterocysts is reduced to 42% of the amount present in vegetative cells. These data suggest a degradation of Photosystem II during heterocyst differentiation. Measurements of photosynthetic electron transport in heterocysts proved the inability of the photosynthetic apparatus to carry out electron transport with electrons donated by water or diphenylcarbazide. In Tris-washed thylakoids from vegetative cells, however, diphenylcarbazide can act as an electron donor to Photosystem II.  相似文献

Two plant-type ferredoxins were isolated and purified from a blue-green alga, Nostoc verrucosum. They were separable by chromatography on a DEAE-cellulose column. The slow-moving band was designated ferredoxin I (Fd I) and the fast-moving band was ferredoxin II (Fd II). The ratio of the yield of ferredoxins I and II was about 1 : 0.84. Both ferredoxins had absorption spectra similar to those of plant-type ferredoxins. Two atoms of non-heme iron and two of labile sulfur were found per mol of both ferredoxin I and ferredoxin II. Their molecular weights were identical and estimated to be about 18 000 by a gel filtration method. The biochemical activities of these Nostoc ferredoxins were studied: the NADP photoreduction activity on one hand and the NADP-cytochrome c reductase activity on the other.  相似文献

The effect of pH, light intensity and population on the toxicity of the pesticide furadan (containing 3% carbofuran as the active ingredient) was studied on the nitrogen fixing blue-green alga Nostoc muscorum. It was observed that all these factors had effects on the toxicity of the pesticide. The blue-green alga grew better in the pH range of 7.5-10, whereas the acidic pH of 5-6 retarded growth. The pesticide was more toxic to alga in the medium of pH 5-6, whereas reduction in toxicity was noticed in alkaline pH of 7.5-10. The alga grew slowly in a light intensity of 1,500 lux in comparison to a light intensity of 3,000 lux, and no growth occurred in the dark. The toxicity of the pesticide gradually decreased with increasing light intensity. The toxic effect of the pesticide was increased when the initial population level was low, and increasing the initial population reduced the toxicity.  相似文献

Nostoc flagelliforme, which is distributed in arid or semiarid steppes of the west and west-northern parts of China, has been used by the Chinese as a food delicacy and for its herbal values for hundreds of years. However, the resource is being over-exploited and is diminishing, while the market demands are increasing with the economic growth. This review deals mainly with the Chinese studies on the ecology, physiology, reproduction, morphology and culture of this species in an attempt to promote research and development of its cultivation technology. Summary Preparations of the blue-green alga, Nostoc pruniforme, treated according to the lead-sulfide staining technique of Ebel et al. (1958b) were examined by light and electron microscopy. They were found to contain spherical, electron-dense bodies, generally in close association with the nucleoplasm and polyhedral bodies, and sometimes enclosed by a membrane. In preparations extracted with cold TCA prior to the application of the staining procedure, such electron-dense structures could no longer be found; electron microscopy revealed instead spherical, electron-transparent areas with an electron-dense periphery in positions generally occupied by the electron-dense bodies in preparations not extracted with TCA.  相似文献
