菜粉蝶Pieris rapae是十字花科蔬菜重要的农业害虫。为了深入了解中国菜粉蝶的群体遗传特征,通过高通量测序获取了中国5个群体48头菜粉蝶的线粒体全基因组,并分析了不同群体间的遗传结构与多样性。结果表明:菜粉蝶群体间有较高的线粒体单倍型多样性;相对封闭的山区和盆地地区的核苷酸多样性较低;群体间的平均遗传分化指数(FST)仅0.042,表明不同群体间的遗传分化水平较低,这也暗示了群体间基因交流频繁,推测这与甘蓝等蔬菜频繁运输有关;线粒体进化树和网络图均未形成明显的地理谱系,南北方群体之间没有明显的遗传结构差异;总群体的Tajima''s D和Fu''s Fs值均为负值,且达到了显著水平,这意味着中国地区的菜粉蝶可能经历过群体扩张事件。  相似文献

Abstract We developed microsatellite markers and combined them with mitochondrial markers to analyse the population genetic structure of the queenless ant Diacamma indicum. This species, lacking winged queens, is likely to have a restricted female dispersal but exhibits various life history traits suggesting higher dispersal abilities than the other Diacamma species. Only 4 of 11 microsatellites were polymorphic and only 1 had more than 4 alleles over 166 individuals originating from 7 populations from the south of India. Only one mitochondrial DNA (mtDNA) haplotype was detected throughout India (including one population in the north) and Sri Lanka. Such a level of polymorphism is particularly low compared with other Diacamma species having much smaller ranges in the south of India. A strong genetic differentiation was observed between populations separated by more than a few kilometres. We also analysed the genetic differentiation between the Indian populations and two populations from the Japanese island of Okinawa, which are morphologically similar and might belong to the same species. The genetic differentiation was high for both markers, suggesting an absence of ongoing gene flow between these populations.  相似文献

【目的】Wolbachia 是一种广泛存在于节肢动物中的胞内共生细菌,影响寄主的生物学特性。花蓟马 Frankliniella intonsa (Trybom)是重要的害虫,对农作物及园林植物造成危害。本研究旨在明确 Wolbachia 在花蓟马中的感染情况,并分析其与寄主线粒体DNA多样性的关系。【方法】采集中国境内26个花蓟马自然种群,运用多位点序列分型技术(multilocus sequence typing, MLST)对其体内 Wolbachia 感染率及株系进行分析;利用线粒体 COI 分子标记研究花蓟马的遗传分化及遗传多样性;通过比较感染和未感染 Wolbachia 个体 COI 数据,探究 Wolbachia 多样性与寄主线粒体DNA多样性之间的关系。【结果】花蓟马中 Wolbachia 的感染率为0%~60%,共检测到5种 Wolbachia 株系(wFint1,wFint2,wFint3,wFint4及wFint5),均属于B大组且形成一个单系群。Wolbachia感染情况与这些花蓟马种群(除CC, GZ, TA和TY, N<5)的线粒体DNA多样性相关,表现为不感染 Wolbachia 的种群中线粒体DNA单倍型多样性(Hd)与核苷酸多样性(Pi)均高于感染 Wolbachia 的种群,且 Wolbachia 感染率与 Hd 呈显著负相关( P <0.05)。AMOVA分析表明花蓟马线粒体DNA遗传分化与Wolbachia 感染情况有关。【结论】 Wolbachia 可能在侵染花蓟马种群后出现遗传分化;Wolbachia 感染与寄主线粒体DNA多样性有关。  相似文献

【目的】木领针蓟马Helionothrips mube近年来成为芋头Colocasia esculenta上的一种常见害虫。本研究旨在探讨其中国西南地区地理种群间的遗传变异。【方法】通过Sanger法测定了21个地理种群132头木领针蓟马的线粒体COI基因序列,利用MEGA, DnaSP, Arlequin和Network等软件对木领针蓟马种群间的遗传多样性、遗传分化、分子变异等进行分析。【结果】获得的木领针蓟马132条线粒体COI序列(643 bp)中共发现34个变异位点、16种单倍型,其中单倍型H1出现频率最高、分布最广。木领针蓟马总群体的遗传多样性较高(Hd=0.712, Pi=0.00413, K=2.655),遗传分化程度极大(Fst=0.3443),基因交流水平不高(Nm=0.96)。分子方差分析(AMOVA)表明,木领针蓟马的遗传分化主要来自种群内部,Mantel检测出种群地理距离与遗传距离呈正相关。总群体中性检验Tajima’s D值显著负值,Fu’s Fs值不显著,错配分布曲线呈多峰。综合种群间遗传距离、单倍型系统发育树及中介网络图结果,表明四川成都(CHD)、云南昌宁(CHN)和贵州遵义(ZY)3个种群的遗传分化程度均高于其他种群。【结论】中国西南地区木领针蓟马总群体的遗传多样性较高,遗传分化明显,种群间的基因交流受到地理距离的影响,总群体在较近的历史时期内没有出现扩张现象。  相似文献

甘肃鼢鼠(Myospalax cansus)是一种终年营地下独居生活的小型掘土类动物。本文通过测定mt DNA的控制区部分序列(530 bp)变异,分析青海东部地区8个甘肃鼢鼠地理种群遗传多样性与遗传结构。158个样本共发现26个变异位点,定义了39种单倍型,整体的平均单倍型多样性高(h=0.953 2)、核苷酸多样性低(π=0.006 36)。歧点分布和中性检验均说明青海东部甘肃鼢鼠种群在历史上存在着快速扩张的事件。基于邻接法构建的网络关系图中,单倍型呈星状分布,没有按地理位置形成对应类群。基因流(Nm)数据显示多数地理种群间基因交流贫乏,AMOVA结果显示种群内与种群间遗传变异分别为48.82%和51.18%,遗传分化明显。IBD分析表明,甘肃鼢鼠的遗传分化与地理距离呈正相关,说明距离隔离对甘肃鼢鼠种群分化具有重要作用。甘肃鼢鼠的这种遗传多样性与种群遗传结构特点,可能是地下生活方式靠挖掘迁移带来的较小扩散能力的结果。  相似文献

【目的】揭示中国草地螟Loxostege sticticalis不同地理种群的遗传分化程度。【方法】采用PCR技术扩增中国西北和华北地区草地螟11个地理种群的线粒体 COI, Cytb和COII基因序列,基于其序列变异及单倍型贝叶斯系统发育树和单倍型网络图分析,探讨不同地理种群间的遗传距离、分子系统发生关系及遗传分化程度。【结果】草地螟11个地理种群的线粒体 COI, Cytb和COII基因序列分别有24, 12和69个变异位点(分别占总序列的3.6%, 2.7%和8.8%),检测到的单倍型分别为22, 14和16个,单倍型多样度(Hd)分别为0.7600, 0.5842和0.7341,核苷酸平均差异度(K)分别为1.704, 0.752和3.997,不同单倍型间的遗传距离平均值分别为0.004, 0.005和0.013。总种群的Tajima’s D和Fu’s Fs值皆不显著,表明草地螟不同地理种群间的遗传分化不明显,群体大小稳定。根据各地理种群的单倍型建立的系统发育树和单倍型网络图表明,各单倍型散布在不同的地理种群中,无明显的地理分布格局。【结论】草地螟各地理种群的遗传距离与地理距离间不具有显著的相关性,其遗传分化不明显。  相似文献

中缅树鼩广泛分布于东南亚地区,在我国主要分布于西南地区及海南岛。本研究以中国10个地理种群共112只中缅树鼩为研究对象, PCR扩增得到1398 bp的细胞色素氧化酶亚基Ⅰ(COⅠ)基因序列,并对其进行分析。 结果显示:112个中缅树鼩COⅠ基因共定义了64个单倍型,其单倍型多样性(Hd)平均值为0.9730,核苷酸多样性(Pi )平均值为0.04494;AMOVA方差分析显示种群间的变异占总变异的93.09%,说明中缅树鼩地理种群变异主要发生在种群间;整体遗传分化固定指数(FST)为0.93091,说明各地理种群中缅树鼩已出现明显的遗传分化;结合中性检验与碱基错配分布图结果表明中缅树鼩在历史进程中未经历过种群扩张现象; 基于单倍型构建的系统进化树与NETWORK网络图显示10个地理种群的中缅树鼩聚为4支:海南种群一支,大新种群一支,片马种群一支,其他种群一支。结果表明:中国不同地理种群的中缅树鼩具有较高的遗传多样性,各地理种群间已经出现了较为明显的遗传分化,地理阻隔作用可能是其分化的主要原因。  相似文献

The paradox of successful invading species is that they are likely to be genetically depauperate compared to their source population. This study on Colorado potato beetles is one of the few studies of the genetic consequences of continent-scale invasion in an insect pest. Understanding gene flow, population structure and the potential for rapid evolution in native and invasive populations offers insights both into the dynamics of small populations that become successful invaders and for their management as pests. We used this approach to investigate the invasion of the Colorado potato beetle (Leptinotarsa decemlineata) from North America to Europe. The beetles invaded Europe at the beginning of the 20th century and expanded almost throughout the continent in about 30 years. From the analysis of mitochondrial DNA (mtDNA) and amplified fragment length polymorphism (AFLP) markers, we found the highest genetic diversity in beetle populations from the central United States. The European populations clearly contained only a fraction of the genetic variability observed in North American populations. European populations show a significant reduction at nuclear markers (AFLPs) and are fixed for one mitochondrial haplotype, suggesting a single successful founder event. Despite the high vagility of the species and the reduction of genetic diversity in Europe, we found a similar, high level of population structure and low gene flow among populations on both continents. Founder events during range expansion, agricultural management with crop rotation, and selection due to insecticide applications are most likely the causes partitioning genetic diversity in this species.  相似文献

为探究经过2个选育世代后选育群体遗传多样性和遗传结构的变化, 研究对4个野生群体(邗江、九江、石首和吴江)和2个选育世代(F1和F2)进行了线粒体DNA控制区(D-loop)序列的遗传变异分析。实验结果表明, 4个野生群体在单倍型数目(H)、单倍型多样性(Hd)、核苷酸多样性(π)、平均核苷酸差异数(K)水平上均高于2个选育世代, 在2个选育世代内表现为F1代群体的核苷酸多样性(π)和平均核苷酸差异数(K)大于F2代群体, 但单倍型多样性(Hd)小于F2代群体; 单倍型分析结果表明, 6个群体间无共享单倍型, 4个野生群体间共发现2种共享单倍型(Hap1和Hap3), 石首群体和2个选育世代共享1种单倍型(Hap15); 遗传分化指数(Fst)分析结果表明, 邗江、九江、吴江3个野生群体和2个选育世代间存在较大的遗传分化(Fst范围为0.41475—0.55128), 石首群体与F1代群体之间存在较小的遗传分化, 与F2代群体之间存在中等水平的遗传分化, 同时F1代群体与F2代群体之间存在较小的遗传分化; 基于6个群体276个个体构建的邻接(Neighbor-Joining, NJ)进化树和基于27种单倍型构建的单倍型网络图也得到了相似的结论, 即邗江、九江、吴江3个野生群体和2个选育世代间的亲缘关系较远, 石首群体和2个选育世代两两之间的亲缘关系较近。以上结果表明, 经过2个世代的选择育种, 选育群体的遗传结构已发生了变化, 并且随着选育的进行, 选育世代的遗传多样性下降的较为明显, 这警示着我们在今后的育种工作中应适当改变现有的选育方案, 并实时监测选育群体的遗传多样性, 以便为今后进一步的选育工作打下坚实的基础。  相似文献

Southern India, one of the last strongholds of the endangered Asian elephant (Elephas maximus), harbours about one-fifth of the global population. We present here the first population genetic study of free-ranging Asian elephants, examining within- and among-population differentiation by analysing mitochondrial DNA (mtDNA) and nuclear microsatellite DNA differentiation across the Nilgiris-Eastern Ghats, Anamalai, and Periyar elephant reserves of southern India. Low mtDNA diversity and 'normal' microsatellite diversity were observed. Surprisingly, the Nilgiri population, which is the world's single largest Asian elephant population, had only one mtDNA haplotype and lower microsatellite diversity than the two other smaller populations examined. There was almost no mtDNA or microsatellite differentiation among localities within the Nilgiris, an area of about 15,000 km2. This suggests extensive gene flow in the past, which is compatible with the home ranges of several hundred square kilometres of elephants in southern India. Conversely, the Nilgiri population is genetically distinct at both mitochondrial and microsatellite markers from the two more southerly populations, Anamalai and Periyar, which in turn are not genetically differentiated from each other. The more southerly populations are separated from the Nilgiris by only a 40-km-wide stretch across a gap in the Western Ghats mountain range. These results variably indicate the importance of population bottlenecks, social organization, and biogeographic barriers in shaping the distribution of genetic variation among Asian elephant populations in southern India.  相似文献

【目的】大豆食心虫Leguminivora glycinivorella (Matsumura)是一种危害大豆的主要害虫,在中国北方地区危害较重。本研究旨在探讨大豆食心虫在中国东北不同地理种群间的遗传变异。【方法】测定了10个不同地理种群153个个体的线粒体细胞色素氧化酶亚基Ⅰ (mtCOI)基因的657 bp序列,利用DnaSP 5. 0和Arlequin 3. 5. 1. 2等软件对大豆食心虫种群间的遗传多样性、基因流水平和分子变异进行分析。【结果】结果表明:10个地理种群间的COI基因共有36个变异位点和17个单倍型,其中1个单倍型为10个种群所共享。总种群的单倍型多样性指数Hd为0.456,各地理种群单倍型多样度范围在0~0.634之间。总群体的固定系数Fst为0.12545,遗传分化系数Gst为0.06326,总基因流Nm为3.49,且各种群间的基因流均大于1,种群间基因交流的水平较高。【结论】大豆食心虫种群内遗传多样性水平处于中低等水平。总群体和各种群的Tajima’s D检验结果皆不显著,说明中国东北地区大豆食心虫在较近的历史时期内没有出现种群扩张现象。AMOVA分子变异分析结果表明,大豆食心虫的遗传分化主要来自种群内部,而种群间未发生明显的遗传分化。各地理种群的单倍型在系统发育树上和中介网络图上散布在不同的分布群中,缺乏明显的地理分布格局。各种群的遗传距离与地理距离之间没有显著线性相关性,种群间的基因交流并未受到地理距离的影响。  相似文献

Chinese sea bass (Lateolabrax maculatus), an important commercial and recreational fishery species in China, whose natural resources have decreased dramatically for decades due to overfishing and environmental changes. To provide guidelines for the source conservation and management, genetic diversity and population structure of L. maculatus were analyzed based on the mitochondrial cytochrome oxidase subunit I (COI) gene. Totally 192 individuals were sampled from five locations: Qingdao (QD), Chongming (CM), Dongtou (DT), Lieyu (LY) and Fangcheng (FC). After sequencing of a 586 bp fragment of COI gene, 20 haplotypes were defined. H4 (haplotype 4) and H8 were the dominant haplotypes and existed in all populations. Haplotype diversity (Hd) and nucleotide diversity (π) of each population ranged from 0.572 to 0.721 and from 0.00129 to 0.00271, respectively. The highest Hd and π were found in DT and LY populations, while the lowest value of Hd and π were in the QD population. Genetic distance ranged from 0.0031 to 0.0027 within populations and from 0.0018 to 0.0035 between populations. Analysis of molecular variance (AMOVA) revealed that significant genetic divergence was found in QD and FC populations. In addition, neutrality tests and mismatch distribution analysis indicated that this species experienced potential population expansion events.  相似文献

研究基于线粒体D-loop区和Cyt b基因部分序列, 分析了广西全州、融水、环江三地禾花鲤(Cyprinus carpio)的遗传结构。在3个群体中共鉴定到19种D-loop与Cyt b序列的组合单倍型, 总的单倍型多样性(Hd)和核苷酸多样性(π)分别为0.916±0.010和0.008±0.004, 禾花鲤线粒体DNA具有单倍型多样性高和核苷酸多样性低的特点。环江群体单倍型多样性最低, 但是核苷酸多样性却最高, 反映了环江群体存在最明显的谱系混杂。分子方差分析(AMOVA)显示遗传变异主要来源于群体内部(71.54%), 禾花鲤三个群体间有显著的遗传分化(Fst=0.285; P<0.01)。系统发育分析表明, 多数样本的线粒体单倍型属于华南鲤(C. carpio rubrefusus)类型, 同时三地禾花鲤中均检测到一定频率的西鲤(C. carpio carpio)或远东鲤(C. carpio haematopterus)类型的线粒体单倍型, 提示禾花鲤可能受到鲤养殖品种的杂交渐渗。研究初步揭示了广西禾花鲤的种质资源现状, 为禾花鲤这一地方特色“稻鱼共作”品种的选育和苗种管理提供了必要的依据。  相似文献

Surveys of allozyme polymorphisms in the carpet shell clam Ruditapes decussatus have revealed sharp genetic differentiation of populations. Analysis of population structure in this species has now been extended to include nuclear and mitochondrial genes. A partial sequence of a mitochondrial COI gene and of the internal transcribed spacer region (ITS-1) were used to study haplotype distribution, the pattern of gene flow, and population genetic structure of R. decussatus. The samples were collected from twelve populations from the eastern and western Mediterranean coasts of Tunisia, one from Concarneau and one from Thau. A total of twenty and twenty-one haplotypes were detected in the examined COI and ITS1 regions respectively. The study revealed higher levels of genetic diversity for ITS1 compared to COI. The analysis of haplotype frequency distribution and molecular variation indicated that the majority of the genetic variation was distributed within populations (93% and 86% for COI and ITS1 respectively). No significant differentiation was found among eastern and western groups on either side of the Siculo-Tunisian strait. However, distinct and significant clinal changes in haplotypes frequencies between eastern and western samples were found at the most frequent COI haplotype and at three out of five major ITS1 haplotypes. These results suggest the relative importance of historical processes and contemporary hydrodynamic features on the observed patterns of genetic structure.  相似文献

本研究基于7个叶绿体DNA片段(cpDNA)和2个核DNA片段(ITS和PZ8)的测序数据,对龙血树柴胡(Bupleurum dracaenoides Huan C.Wang,Z.R.He&H.Sun)8个居群的153个样本进行了遗传多样性和分布式样研究。cpDNA片段分析结果显示:龙血树柴胡在物种水平具有较高的遗传多样性(Hd=0.862;Pi=0.00567),但居群内遗传多样性低,遗传变异主要存在于居群间,遗传分化显著(Fst=0.959);而核DNA片段ITS和PZ8的数据分析结果显示,其遗传多样性较低(Hd=0.532,Pi =0.00121和Hd=0.349,Pi=0.00060),遗传变异主要存在于居群内,居群间仅存在一定程度的遗传分化。中性检验和失配分布分析结果发现龙血树柴胡没有经历过近期种群扩张事件,8个居群的153个样本从遗传成份上可被分为两组。研究结果将为龙血树柴胡的资源保护和发掘提供参考。  相似文献

The genetic diversity of 289 spotted scat (Scatophagus argus) from seven populations along the northern coast of the South China Sea was studied by analyzing the full-length sequences of the mitochondrial control region (D-LOOP). The S. argus D-LOOP sequence was 1,004–1,010 bp long and contained 156 variant sites. The seven studied S. argus populations had a high degree of genetic diversity (haplotype diversity [Hd] = 0.99135; nucleotide diversity (π) = 0.01313). There was no obvious genetic differentiation among the seven geographical populations and gene exchange was frequent (Fst = −0.01867–0.01117, p > .05). Four distinct mitochondrial lineages were identified in the phylogenetic tree and the haplotype network. The between-lineage Fst was 0.71690–0.84940 (p < .001), but these lineages showed no obvious phylogeographic pattern. Based on D-LOOP mutation rates, we estimated that the four lineages diverged approximately 513,800–93,600 years ago, during the Eocene ice age, at which time falling sea levels may have led to population segregation. We estimated that S. argus population expansion occurred approximately 2.29–0.68 million years ago, during the late Pleistocene. During this period, sea levels rose again, allowing previously separated lineages to come into sympatry, which eventually gave rise to a highly genetically diverse population without pyhlogeographic structure. Here, we characterized the genetic structure and differentiation of seven S. argus populations from the northern coast of the South China Sea. Our results suggested that the seven S. argus populations from the northern coast of the South China Sea have a relatively low level of genetic variation and can be considered a single unit for the purposes of fishery development, utilization, and management.  相似文献

【目的】为了明确我国甜菜夜蛾Spodoptera exigua地理种群间的遗传分化及基因流,阐明该种害虫在我国的种群历史动态。【方法】本研究对采自我国20个地理种群的529头甜菜夜蛾样品进行线粒体COI基因序列测定与分析,利用DnaSP 5.0和Arlequin 3.11软件分析种群间遗传多样性、遗传分化、基因流水平及分子变异,构建了单倍型系统发育树与单倍型网络图。【结果】在所分析的所有529个序列样本中,共检测出10个单倍型,其中Hap_1为所有种群所共享。总群体遗传多样性较低(Hd=0.257±0.025,Pi=0.0007±0.0001,Kxy=0.323),群体间遗传分化较小(FST=0.211),基因流水平较高(Nm=1.870)。AMOVA分析表明,甜菜夜蛾遗传变异主要来自种群内,种群间变异水平较低。各种群间遗传分化程度与地理距离无显著相关性(R2=0.005,P>0.05)。各单倍型相互散布在不同种群中,未形成明显系统地理结构。中性检验(Tajima’s D=-2.177, P<0.05; Fu’s FS=-8.629, P<0.05)与错配分布分析表明,我国甜菜夜蛾种群曾经历种群近期扩张。【结论】研究结果揭示,甜菜夜蛾各种群间的基因交流并未受到地理距离的影响,验证了甜菜夜蛾具有高度的迁飞能力。  相似文献

为阐明锁阳(Cynomorium songaricum)的遗传结构与遗传多样性, 以甘肃省河西走廊地区及青海省共18个居群的188个锁阳个体为研究对象, 利用现代分子生物学技术, 采用序列分析方法从核-质基因方面对遗传结构进行了分析。结果显示, 锁阳ITS序列总长度为687 bp, 含有7个变异位点, 定义9个单倍型, 整体单倍型多态性Hd=0.294 20, 核苷酸多样性π=0.000 49。在整个单倍型网络中介图中, 单倍型H1位于中心位置, 并在所有的居群中均有分布, 为核心的古老单倍型。分子方差分析结果显示, 锁阳种群变异主要来源于种群内。根据ITS序列得到的群体间遗传分化系数以及Mantel检验结果, 锁阳种群间的遗传距离与地理距离之间不存在相关性, 表明现存的锁阳居群是相对近期发生生境片段化的产物。中性检验结果表明, 锁阳拒绝中性进化, 群体历经扩张或者基因座位受到负选择作用, 其中性零假说不能被排除。该研究为锁阳的系统分类、资源鉴定以及保护措施的制定提供了分子证据。  相似文献
