FIT是调控拟南芥铁稳态的一个关键调控因子,它在转录水平上受到缺铁诱导,但其背后的调控机制还不甚清楚。该研究以拟南芥bHLH38和FIT的单、双过表达植物及bHLH Ib四突变体植物为材料,采用缺铁(-Fe)处理实验和定量RT-PCR的方法从RNA角度分析了FIT转录水平的变化。结果表明:(1)在铁充足时,bHLH38过表达植物中FIT的转录水平显著高于其在野生型中的水平。(2)在bHLH Ib四突变体植物中FIT的转录水平不受缺铁诱导。(3)FIT单过表达不能激活内源FIT的转录,而在加铁(+Fe)条件下bHLH38和FIT的双过表达则可以激活内源FIT的转录。(4)在缺铁条件下,所有植物中FIT的转录水平均与野生型中的FIT水平无明显差异。基于以上结果认为,bHLH Ib转录因子是缺铁诱导FIT转录的必要条件,而非充分条件。该研究结果为深入了解植物通过多种途径共同维持铁稳态提供了新的见解。  相似文献

Interfering RNA was used to suppress the expression of the genes At1g06680 and At2g30790 in Arabidopsis thaliana, which encode the PsbP-1 and PsbP-2 proteins, respectively, of Photosystem II. A phenotypic series of transgenic plants was recovered that expressed intermediate and low amounts of PsbP. Earlier we had documented significant alterations in a variety of Photosystem II parameters in these plant lines [Yi, X., Liu, H., Hargett, S. R., Frankel, L. K., Bricker, T. M. (2007). The PsbP protein is required for photosystem II complex assembly/stability and photoautotrophy in Arabidopsis thaliana. J. Biol. Chem. 34, 24833-24841]. In this communication, we document extensive defects in the thylakoid membrane architecture of these plants. Interestingly, strong interfering RNA suppression of the genes encoding the PsbQ protein (At4g21280 and At4g05180) was found to have no effect on the architecture of thylakoid membranes.  相似文献

Systemic acquired resistance (SAR) is a defense mechanism induced in the distal parts of plants after primary infection. It confers long-lasting protection against a broad spectrum of microbial pathogens. Lack of high-throughput assays has hampered the forward genetic analysis of SAR. Here, we report the development of an easy and efficient assay for SAR and its application in a forward genetic screen for SAR-deficient mutants in Arabidopsis (Arabidopsis thaliana). Using the new assay for SAR, we identified six flavin-dependent monooxygenase1, four AGD2-like defense response protein1, three salicylic acid induction-deficient2, one phytoalexin deficient4, and one avrPphB-susceptible3 alleles as well as a gain-of-function mutant of CALMODULIN-BINDING TRANSCRIPTION ACTIVATOR3 designated camta3-3D. Like transgenic plants overexpressing CAMTA3, camta3-3D mutant plants exhibit compromised SAR and enhanced susceptibility to virulent pathogens, suggesting that CAMTA3 is a critical regulator of both basal resistance and SAR.  相似文献

The xenobiotic monochlorobimane is conjugated to glutathione in the cytosol of Arabidopsis thaliana, transported to the vacuole, and hydrolyzed to cysteine S-bimane [Grzam, A., Tennstedt, P., Clemens, S., Hell, R. and Meyer, A.J. (2006) Vacuolar sequestration of glutathione S-conjugates outcompetes a possible degradation of the glutathione moiety by phytochelatin synthase. FEBS Lett. 580, 6384-6390]. The work here identifies gamma-glutamyl transpeptidase 4 (At4g29210, GGT4) as the first step of vacuolar degradation of glutathione conjugates. Hydrolysis of glutathione S-bimane is blocked in ggt4 null mutants of A. thaliana. Accumulation of glutathione S-bimane in mutants and in wild-type plants treated with the high affinity GGT inhibitor acivicin shows that GGT4 is required to initiate the two step hydrolysis sequence. GGT4:green fluorescent protein fusions were used to demonstrate that GGT4 is localized in the lumen of the vacuole.  相似文献
