Benzodiazepines (BDZs) are the most widely prescribed class of psychoactive drugs in current therapeutic use, despite the important unwanted side-effects that they produce such as sedation, myorelaxation, ataxia, amnesia, ethanol and barbiturate potentiation and tolerance. Searching for safer BDZ-receptor (BDZ-R) ligands we have recently demonstrated the existence of a new family of ligands which have a flavonoid structure. First isolated from plants used as tranquilizers in folkloric medicine, some natural flavonoids have shown to possess a selective and relatively mild affinity for BDZ-Rs and a pharmacological profile compatible with a partial agonistic action. In a logical extension of this discovery various synthetic derivatives of those compounds, such as 6,3-dinitroflavone were found to have a very potent anxiolytic effect not associated with myorelaxant, amnestic or sedative actions. This dinitro compound, in particular, exhibits a high affinity for the BDZ-Rs (Ki = 12–30 nM). Due to their selective pharmacological profile and low intrinsic efficacy at the BDZ-Rs, flavonoid derivatives, such as those described, could represent an improved therapeutic tool in the treatment of anxiety. In addition, several flavone derivatives may provide important leads for the development of potent and selective BDZ-Rs ligands.  相似文献

The H3L histone variant gene in Paracentrotus lividus (sea urchin) shows almost all typical features of the replication-dependent histone genes, but it codes for the H3.3 histone protein with the S.//. A.IG amino acid motif, which is typical of the variants synthesized in a replication-independent manner. H3L-like histone genes have been found in several unrelated organisms. These genes are intronless and encode for the typical H3.3 histone proteins. The newly described family of H3L-like variants, nearly ubiquitous within the animal kingdom, could represent the common ancestor of H3 and H3.3 histone genes.  相似文献

In his important contribution to the literature on gymnosperm phylogeny, “Basic Features of Gymnosperm Systematics and Phylogeny as Evidenced by the Fossil Record,” Meyen (1984) uses the classical comparative method of the morphologist and the “congregational” method, i.e., grouping by common characters, of the taxonomist. The latter may have led him to categorize some taxa on the basis of superficially similar, non-homologous characters because he used no apparently objective method to distinguish homoplasy. For this and other reasons, Meyen’s hypothesis of evolutionary relationship among gymnosperms cannot, at present, be accepted as any nearer the truth than several competing proposals. The major innovation of Meyen’s proposed phylogeny is the recognition of the clade, Ginkgoopsida, coordinate with Cycadopsida and Pinopsida. Ginkgoopsida encompasses Calamopityaceae, Callistophytales, Glossopteridales, Peltaspermales, Caytoniales, and Ginkgoales, among others. These taxa are considered to be related because, in Meyen’s view, they share the common character of primary platyspermy — i.e., the presumed platyspermic seed-type of the basal group, Calamopityaceae, is considered to have evolved directly from the pteridophytic condition of an ancestral form. This basis for Ginkgoopsida is weakened by the facts that no seeds have been discovered in organic connection with any calamopityacean and, except for the poorly-preserved seed-like structure,Spermolithus, all seeds that occur earlier in the geologic record than fossils of the Calamopityaceae are radiospermic. It is possible, therefore, that even if the platyspermic seed,Lyrasperma, found in association with the calamopityacean,Stenomyelon, were borne by that plant, it is secondarily platyspermic, having evolved from a radiospermic ancestor. The foundation upon which Ginkgoopsida was erected seems, therefore, to be rather tenuous. Other characters used by Meyen, both reproductive and vegetative are discussed. Some which he considers significant, are interpreted to be unimportant in denoting phylogeny, while others are interpreted to support alternative hypotheses. Meyen proposes that the Ginkgoopsida evolved from archaeopterid progymnosperms. This viewpoint seems to be based largely on his erroneous belief thatArchaeopteris was probably a seed plant that bore compound leaves. There is no definitive evidence that supports the view thatArchaeopteris bore seeds. On the other hand, there is strong evidence to support the contention thatArchaeopteris produced simple leaves (Carluccio et al., 1966; Beck, 1971), not compound leaves. Meyen’s phylogenetic proposal is based on data that can be and, in part, have been differently interpreted by others. Consequently, it deserves the careful and critical evaluation of all students of gymnosperm phylogeny.  相似文献

A new anticonvulsant, nonbarbiturate compound, Doriden (ethyl-a-phenyl-glutarimide) was studied for clinical activity as a hypnotic drug in mildly disturbed patients on the acute Psychiatric Service of Los Angeles County General Hospital. It was found to have approximately the same hypnotic activity as pentobarbital in the dosages recommended (1 gm. Doriden=200 mg. pentobarbital).  相似文献

Cl channels play important roles in many physiological processes, including transepithelial ion absorption and secretion, smooth and skeletal muscle contraction, neuronal excitability, sensory perception, and cell volume regulation. The molecular identity of many types of Cl channels is still unknown. Recently, three research groups have arrived independently at the identification of TMEM16A (also known as anoctamin-1) as a membrane protein strongly related to the activity of Ca2+-activated Cl channels (CaCCs). Site-specific mutagenesis of TMEM16A alters the properties of the channels, thus suggesting that TMEM16A forms, at least in part, the CaCC. TMEM16A is a member of a family that includes nine other membrane proteins. All TMEM16 proteins have a similar structure, with eight putative transmembrane domains and cytosolic amino- and carboxy-termini. TMEM16B expression also evokes the appearance of CaCCs, but with biophysical characteristics (voltage dependence, unitary conductance) different from those associated to TMEM16A. The roles of the other TMEM16 proteins are still unknown. The study of TMEM16 proteins may lead to identification of novel molecular mechanisms underlying ion transport and channel gating by voltage and Ca2+.  相似文献

Two thousand five hundred forty-five cases of upper gastrointestinal tract hemorrhage were studied especially with a view to determining the indications for urgent surgical treatment.Decisions as to whether and when to operate were as follows:Immediate operation for patients over 50 years with a good history of ulcer and a severe initial bleed.Operation after the first repetition of bleeding in patients (1) over 50 with a good history and a mild initial bleed, (2) over 50 with inconclusive history but severe initial bleed, (3) under 50 with a good history and a severe initial bleed.In all other cases, operation was used only if conservative treatment failed.Absolute indications for operation were (a) association with perforation, (b) association with stenosis, (c) persistence of severe ulcer pain after hemorrhage, (d) continuous bleeding.Since operation is to be avoided if possible in cases of esophagitis, erosive gastritis and small acute or subacute ulcers, emergency gastroscopy has valuable uses.Where operation is deemed necessary and no obvious lesion found at laparotomy, blind gastrectomy* appears to be the most satisfactory procedure.The mortality rate associated with upper gastrointestinal tract bleeding in patients less than 60 years of age was low (2.5 per cent). Even in cases in which operation was required, it was 6.2 per cent. Over 60 years the mortality rises steeply with increasing age, and in cases of operation the rise is even steeper.By using the methods of selection the overall mortality rate was appreciably reduced.  相似文献
