以尿囊素作为鉴别试剂,对肉苁蓉属中的管花肉苁蓉与同属的其它品种植物进行鉴别.采用HPLC法,在以下色谱条件下(Hypersil ODS-2色谱柱4.6 mm×250 mm,5 μm,流动相为乙腈-7.5%二氧六环水溶液1:99,流速0.5 mL/min,柱温25℃,检测波长224 nm)绘制肉苁蓉属植物不同品种的色谱图.在盐生肉苁蓉或荒漠肉苁蓉的HPLC色谱图中,与对照品尿囊素色谱峰相应的位置处,显示保留时间相同的色谱峰,而在管花肉苁蓉供试品的HPLC色谱图中,无尿囊素色谱峰,提示不含尿囊素的肉苁蓉为管花肉苁蓉.该鉴别方法简便、灵敏,能快速、准确地将管花肉苁蓉与同属其它品种植物鉴别开来.  相似文献

从发育的角度研究了中国特有单种属DichotomanthesKurz及与其系统学研究有关的外类群Prinsepiautilis的花粉形态 ,扫描电镜观察显示Dichotomanthes花粉粒自脱离四分体胼胝质膜开始至成熟二核花粉粒不同发育时期 ,花粉形态和外壁纹饰未见变化 ,仅花粉体积随成熟度增加而有所增大。而Prinsepiautilis ,其花粉粒刚脱离四分体时形状和成熟花粉明显不同 ,成熟花粉极面观为三裂圆形 ,赤道面观为圆形 ,外壁具清晰的平行条纹 ,但幼嫩花粉粒的形状很特别 ,极面观为深三裂圆形 ,赤道面观亦见花粉在两条沟之间下陷而沟部外突 ,明显为角萌发孔花粉 ,且花粉体积较成熟者小 ,而外壁纹饰同成熟者相比无根本性差异。前述两种植物花粉在不同成熟期体积有明显差异 ,而外壁纹饰在不同成熟期不存在质的变化并相对稳定 ,说明花粉外壁纹饰这一性状在蔷薇科中具有较为重要的分类学意义。DichotomanthesKurz具典型Rosaceae花粉的三孔沟结构 ,外壁具条纹 -穴状纹饰。将其孢粉学特征同Rosaceae 4个亚科有关类群的同类资料相比较 ,并结合其它形态解剖与细胞学等研究结果 ,支持将Dichotoman thes置入Maloideae下而不赞同将其另立亚科或置于Prunoideae之下。此外 ,由于Prinsepiautilis的花粉在其发育初期具角萌发孔花粉 ,与Cunoniacea  相似文献

The genus Oreocharis as circumscribed here consists of 27 species including 5 varieties, of which 5 species and 4 varieties are described as new in the present paper. In the work analysed were the external morphology and geographic distribution and examined under SEM were pollen exine of 22 species and seed coat of 16 species. As a result, three types of the corolla, two types of the anther, three types of the pollen exine and three types of the seed coat are distinguished here in the paper. It is discovered that the corolla in the genus is relatively stable, though diverse, and highly correlated with the characters of pollen grains and seeds. The corolla clearly bilabiate but constricted at the throat, occurring in O. auricula, O. cordatula, O. aurantiaca, etc., for an example, is correlated with smooth, reticulate pollen exine and partial tectum and the reticulate and smooth seed coat. For this reason the subdivision of the genus in the paper is mainly based on the characters of the corolla, but combined with those of the anther, pollen and seed coat. The genus is divided into four sections in the present classification. Dasydesmus Craib, based on a single species. O. bodinieri, is reduced here, and the reasons are given. The genus is distributed mainly in the subtropics, and less frequently in the tropics, of China south of 32.5°N and east of 98.5°E, with only two species beyond the border, O. hirsuta in Thailand (only a single locality in Chiengmai) and O. aurea also found in north Vietnam (see Fig. 1, Table 3). Sect. 1. Stomactin (Clarke) Fritsch. Corolla urceolate-tubular, constricted at the throat, with limb distinctly bilabiate; anthers broad-oblong; seed coat reticulate, smooth, rarely minutely tuberculate; pollen exine fine-reticulate, tectum partial and smooth, luminae slightly unequal in size. Sect. 2. Orthanthera K. Y. Pan Corolla campanulate or campanulate-tubular; anthers broad-oblong; seed coat reticulate, muri smooth, rarely spiny-processed; pollen exine fine-reticulate, with partial and smooth tectum and luminae slightly unequal in size, rarely exine insular and fine-tuberculate, tectum perforate. Setc. 3. Oreocharis Corolla thin-tubular; anthers broad-oblong; seed coat densely spinyprocessed, rarely fine-tuberculate; pollen exine insular, densely spiny-processed, rarely finereticulate and smooth, luminae unequal in size. Sect. 4. Platyanthera K. Y. Pan Corolla campanulate; anthers hippocrepiform; seed coat densely spiny-processed; pollen exine fine-reticulate, tectum perforate, luminae small, nearly equal in size. In the section Stomactin, although the constriction of corolla at its throat is a specialized character, the characters of seed coat, pollen grains and anthers are apparently primitive. Therefore it may be said at least that more primitive characters are preserved in the section. In the section Oreocharis, on the contrary, the characters of corolla, seed coat and pollen exine are all advanced. And in the section Platyanthera, the seed coat, pollen (with perforate tectum) and anthers have developed rather specialized characters.  相似文献

本文对中国东北牛儿苗科Geraniaceae老鹳草属Geranium和牛儿苗属Erndium植物计2属,14种,2变种及1变型的花粉形态进行了较系统的研究。并对本科花粉形态进行了分类处理,编制了花粉形态科种检索表。结果表明,本科花粉如按萌发孔类型、外壁雕纹式样可以分为二大类:即三孔沟,外壁雕纹瘤状(Geranium)三沟,外壁雕纹网状(Erodium)。在老鹳草属种及种下等级的分类处理中,花粉粒大小、萌发沟长度、外壁覆盖层网眼均匀性、直径大小、网脊上纹饰及外壁雕纹瘤纹突起均匀性、大小及瘤上纹饰都具有较大的分类学意义。花粉形态分类与经典分类对本科分类群的处理意见相吻合。大大丰富了本科分类微形态的证据。  相似文献

The pollen morphology of some native Greek Aegilops species is investigated in LM (quantitative pollen characters) and SEM (exine sculpture) using acetolysed material. Furthermore, quantitative data are subjected to a multivariate analysis. The tetraploid Ae. cylindrica and its diploid parent Ae. caudata show a distinct morphological affinity as regards their quantitative pollen profile and the features of the exine sculpture. The tetraploid Ae. triuncialis is morphologically divergent from both its parents Ae. caudata and Ae. umbellulata due to the very large values of its quantitative pollen characters. However, the SEM survey of the exine sculpture indicates a rather high degree of similarity between Ae. triuncialis and Ae. umbellulata. No significant differences have been found between the two varieties of Ae. caudata (caudata and polyathera) concerning the quantitative pollen characters as a total or the morphology of the exine sculpture. The resultant clustering of the taxa on the basis of the quantitative pollen characters as well as the recorded similarities of their exine pattern are related to their sectional classification based on cytogenetical and morphological data.  相似文献

The present paper reports the pollen morphology of 36 species and varieties of Brassica L. in China mostly for the first time. They were examined both under LM and SEM. Pollen grains of Brassica L. in China are long-spheroidal, subspheroidal, spheriodal, oblate, rarely reniformis or subsquare. According to the aperture characters pollen grains may be divided into four types: 2,3,4-colpate, 3-colpate, 3,4-colpate, 3,4-colporate. The exine sculpture may also be divided into three types: 1, reticulate, 2, cerebroid-reticulate, 3, foveolatereticulate. Detailed characters of pollen grains for each taxon are presented in Table 1. The 2,3,4-colpate type of pollen grains may be considered as primitive one and the 3,4-colpate or 3,4-colporate type of pollen grains is the most advanced in Brassica. The tricolpate aperture and reticulate exine have been reported by the previous authors, whereas, 3,4-colporate and cerebroid-reticulate or foveolate-reticulate pollen grains are newly reported.  相似文献

LESTER, R. N. & EZCURRA, C, 1991. Enzyme etching treatment as an aid in the study of seed surface sculpture in Justicia and Ruellia (Acanthaceae). Enzyme etching treatment of seeds of Justicia was tested before scanning electron microscope examination. On all species tested the treatment removed the outer periclinal walls of the epidermal cells of the seeds, revealing the shape and morphology of the anticlinal walls. This treatment provides additional characters of the structure of the testa that may be useful in assessing relationships between species in this large and complex genus, but it proved useless in Ruellia. Here the seeds produce a coat of mucilage when moistened that completely protects the testa from the action of even extensive enzyme treatment.  相似文献

The pollen morphology of 26 species and 5 hybrids of the genus Malus was investigated with aid of SEM. It is found that the pollen morphology of Malus is rather similar in shape, size, position and number of aperture and exine sculpture. The characters of polle morphology of sections and series are as follows: Sect. Malus Ser. Baccatae: Striae regular, parallel to colpi, and conjunct at pole; Ser. Pumilae: the same as in Ser. Baccatae, but more or less curved near pole. Sect. Docyniopes: Striae regular and parallel to colpi, but bent near pole. Sect. Chloromeles: Striae irregular, dense and interlock. Sect. Sorbormalus: Striae irregular, sparse and not interlock. Ser. Sieboldiance: pollen grains prolate, with perforation among striae, colpi narrow; Ser. Kansuenses: Pollen grains spheroidal, colpi wide in the middle but narrow at both ends, striae relatively dense and regular, mostly dichotomous, perforation present; Ser. Yunnanenses; Pollen grain spheroidae, colpi wide in the middle but narrow at both ends, striae obviously irregular, less dichotomous, perforation absent. The major evolutionary trend of exine sculpture of pollen may be from densely thin-striate to sparsely striate with perforation. Characters of the exine sculpture of hybrids can be used to recognize the rela-tionship between the parental species.  相似文献

Pollen morphology of the genus Skimmia was studied. Of six species of the genus, five species were investigated by light microscopy and scanning electron microscopy. Pollen grains of Skimmia had striate to striate-reticulate sculpture and variable aperture number (3-7), and these characters differed from even the phylogenetically closest relatives such as Dictamnus and Casimiroa which have 3-colporate pollen grains with reticulate exine. In Skimmia laureola, pollen dimorphism is suggested in the aperture number in accordance with the floral system. Pollen grains of Skimmia japonica, a morphologically very variable species, were extensively surveyed covering its whole distribution area. The grains were also striate to striate-reticulate, and the exine sculpture was not different between varieties. However, the aperture number of S. japonica showed a geographical variation ranging from 4 to 6; in Sakhalin, Hokkaido, most part of Honshu and Taiwan, grains with 4-5 apertures were dominant, while in the southern part of Japan, including the Ryukyu Islands and the Izu Islands, those with 5-6 apertures were common.  相似文献

In this work examined were pollen grains of 6 species of Nomocharis and 4 species of Lilium, all under LM and SEM, but Nomocharis saluenensis Balf. f. under TEM. The pollen grains are monocolpate (distal) in Nomocharis, monocolpate or 2-3-porate in the species of Lilium examined. Two types of pollen grains in Nomocharis are recognized based upon the exine stratification and sculpture. The pollen characters and gross morphology, and their evolutionary trends in both Nomocharis and Lilium are discussed. Our conclusions are as follows: 1. Lilium henrici Franch. 、L. souliei (Franch.) Sealy 、L. lophophorum (Bur. et Franch.) Franch. and L. nanum Klotz. et Garcke differ from Nomocharis in the aperture and sculpturing elements. Their pollen morphology supports their being placed in Lilium. 2. The pollen grains of both Lilium lophophorum (Bur. et Franch.) Franch. and L. nanum aggregate are monocolpate or 2-3-porate, which shows an evolutionary trend of aperture from monocolpate to porate, as suggested by Walker and others [Walker et al., (1975), and Chanda (1979)]. 3. In Balfour’s (1918) classification system of Nomocharis, N. pardanthina Franch. and N. meleagrina Franch. belong to Sect. Nomocharis, while N. forrestii and N. biluoensis are placed in Sect. Ecristata, but their pollen grains are all reticulate. They refer to type I (see Table 2). Thus, it does not seem reasonable placing in a single section, Sect. Ecristata, N. forrestii and N. biluoensis with the reticulate exine and N. saluenensis and N. aperta with the two-row-ritipilate exine. 4. Nomocharis saluenensis Balf. f. differs from Lilium in the exine sculpture, which seems to support placing it in Nomocharis. 5. Muri of N. forrestii Balf f. and N. biluoensis s.y. Liang show a tendency of connecting each other by heads of pili, and these two species are intergraded with the other species of the type I with respect to gross morphorlogical characters. It is thus considered that the reticulate pollen grains in Nomocharis is derived from ritipilate ones. This opinion is also held by Nair (1965). 6. Nomocharis differs from Lilium in the exine ornamentation, but some species of Nomocharis are very similar to Lilium in the gross morphology. Pollen morphology of 16 Chinese species representing 7 genera in the tribe Cynoglosseae of Boraginoideae(Boraginaceae) was examined under LM and SEM, and 5 species under TEM. Pollen grains are cocoon-shaped, rarely subprolate, prolate or ovoid, very small, 7~ 15.7μm× 3.5 ~ 13.9 μm in size, P/E = 1.6~2.02; 3-colporate apertures alternate with 3-pseudocolpi, with equatorial endocingulus except those in Bothriospermum; exine surface is usually smooth, with or without perforations in two poles, rarely with tuberculate ornamentation; exine is rather thin and includes ectexine and endexine, while ectexine consists of imperforate tectum, columellae and foot-layer. The tribe Cynoglosseae has many common characters of pollen grains, but there are some differences among genera. A key to the genera is given based on pollen morphology. Bothriospermum, without endocingulus, may be a primitive genus in this tribe, and its pollen morphology is more similar to that of Eritrichieae than to Cynoglosseae, thus it seems more reasonable to put it into Eritrichieae. Solenanthus may be the most advanced genus in the tribe Cynoglosseae for its pollen grains of sub-isopolar and ovoid shape.  相似文献

利用扫描电子显微镜对紫金牛属圆齿组20种1变种植物的花粉形态进行了研究,并比较分析了腋序组2种及锯齿组1种的花粉。经观察花粉为近球形或近扁球形,具3孔沟,除锯齿组的月月红外,其余种均形成合沟。外壁纹饰可分为四类:穴网状、细网状、皱波状及细颗粒状(带刺突)。结果显示,花粉形态特征可作为属下分组及组下分类处理的依据。  相似文献

The aim of this study is verification of the taxonomic usefulness of the pollen grain features studied, based on pollen morphology of 32 wild species from all 4 subgenera and all 10 sections of the genus Rosa, mainly for delimitation of subgenera, sections, and species. The measurements and observations were carried out with both light microscopy and scanning electron microscopy. Only correctly formed pollen grains (30 per specimen) were measured, and 960 pollen grains were examined in total. They were analyzed for 14 quantitative features of pollen grains and exine sculpturing and the following qualitative ones: outline, shape, and operculum structure. Our study revealed that the diagnostic features of pollen grains studied were: exine sculpture, length of polar axis, and pollen shape (P/E ratio). On the basis of the above characters, five species were isolated and the remaining ones were included in several groups isolated on the basis of exine sculpture types. The following three exine sculpture types occurred in the species studied: granular-verrucate (in R.?stellata), striate-psilate (in R.?multibracteata and R.?multiflora), and striate (the remaining species). R.?banksiae is characterized by small pollen grains, while R.?setigera has strongly elongated pollen with P/E ratio >1.5. Exine sculpture features considered to be diagnostic should be treated as auxiliary because they fail to differentiate individual species, although they can be helpful in distinguishing groups of species of similar exine sculpture. The arrangement of the species examined on a dendrogram only slightly corroborates division of the Rosa genus into subgenera and sections currently adopted in taxonomy (Rehder 1940). An interesting result was reported for the species studied from the Caninae (R.?agrestis, R.?canina, R.?dumalis, R.?jundzillii, and R.?rubiginosa) section which, despite hybrid nature, with the exception of R.?villosa, grouped in the same, most separated group of species.  相似文献

Pollen morphology in the subtribe Aspidistrinae is reported and the main viewpoints are summarized as follows: (1) Two major types of pollen grains, i.e., monosulcate,ellipsoidal pollen and inaperturate, spheroidal pollen, are identified in the genus Tupistra. The species with monosulcate, ellipsoidal pollen usually have monomodal karyotypes, brightcolored flower, smaller stigma, and stamens inserted at upper or middle part of perianth tube, while the inaperturate and spheroidal pollen is always found in the species with bimodal karyotypes, dingy-colored flower, swollen stigma and stamens inserted at the base of perianth tube. (2) Pollen grains in the genus Rhodea are monosulcate and ellipsoidal, while those in the genus Aspidistra are inaperturate and spheroidal, correspondingly similar to the second type of pollen grains in the genus Tupistra. (3) The most primitive pollen in the subtribe Aspidistinae is regarded as monosulcate and ellipsoidal, having perforate or reticulate exine sculpture. The inaperturate, spheroidal pollen with verrucate, gemmate or rugulate exine sculpture is considered derived; (4) Unlike those reported in other groups of the family Liliaceae, the infraspecific pollen shape, aperture type and exine sculpture in Aspidistrinae are basically stable and may serve as a taxonomic character.  相似文献

Seed size and shape, seed coat surface pattern, seed coat thickness, and endosperm structure were investigated in Androsace septentrionalis, Cortusa matthioli, Hottonia palustris, Primula elatior, Soldanella carpatica (Primulaceae), Anagallis arvensis, A. minima, Cyclamen purpurascens, Glaux maritima, Lysimachia nemorum, L. vulgaris, Trientalis europaea (Myrsinaceae), and Samolus valerandi (Theophrastaceae). Three seed size categories were distinguished on the basis of biometric measurements. Almost all seeds examined were found to be small with an angular shape classified as sectoroid or polyhedral. A new type of seed shape, suboval, was identified for H. palustris. Cyclamen purpurascens seeds differed from seeds of all other species examined because of their large size, subglobose shape, and concave hilar area. Despite the different shape types, the length/width ratio of the seeds examined was constant, while their hilum length/width ratio was a highly variable feature. Three types of seed surface patterns were observed: (1) reticulate, (2) tuberculate with secondary striations, and (3) poroid-alveolate with the presence of a spongy outer layer. For seeds of Anagallis arvensis, A. minima, Cortusa matthioli, Lysimachia nemorum, and Soldanella carpatica, secondary seed sculpture was described. The seed coats of all species examined were two-layered, and great differences in testa thickness were found (9.9?C128.6???m). In general, seeds of the Myrsinaceae species were more often characterized by thick testa. Different proportions in thickness of the inner and outer testa layers were observed. In seeds with reticulate seed patterns, the inner testa layer was twice to several times thicker than the outer layer, while the opposite proportions were observed in seeds with the tuberculate or poroid-alveolate seed sculpture pattern. In seeds of all species examined, oxalate crystals were present on the surface of the inner testa layer. Thus, the presence or absence of oxalate crystals in testa is not a feature distinguishing species with angular or subglobose seeds. Four types of endosperm structure were identified according to the thickness of the endosperm cell walls and the relief of their inner surface: (1) with evenly thickened and smooth cell walls, (2) with evenly thickened cell walls and circular or helical thickenings on their inside surfaces, (3) with very thick, but not evenly thickened cell walls with constrictions (??pitted??), and (4) with very thin papery and undulate cell walls. There is no rule concerning the seed shape, type of the seed sculpture, testa thickness, or endosperm structure that corresponds to the family affiliation of the species examined.  相似文献

Pollen morphology of Chinese 19 species representing 6 genera in the tribe Zizipheae Brongn., Rhamnaceae, was examined under light microscope and scanning electron microscope. The pollen grains oblate or suboblate, rarely subspheroidal. Polar axis 13.1-26.1μm long, equatorial axis 16.5-29.6μm long, 3-colporate, colpus generally narrow, ora lalongate, with two ends connected with the thinned part of exine, forming a H-shape. There are 4 thickenings where colpi and ora pass across, or sometimes the thickenings indistinct, forming a ring. Stratification couspicuous or inconspicuous. Ornamentation of exine obscure, rarely obscurely reticulate under light microscope, but conspicuously fine-reticulate, finely reticulate-foveolate, foveolate, striate-reticulate and shortly striate under scanning electron microscope. A key to the genera based on pollen grains is provided and the general morphology of 6 genera: Paliurus, Ziziphus, Chaydaia, Berchemia, Berchemiella and Rhamnella are described. Based on 4 thickenings and H-shape, pollen grains of the tribe Zizipheae may be divided into two groups: 1. the four thickenings rather larger and conspicuous, or stretched along the colpus in some species, H-shape distinct, e.g. in Berchemiella, Chaydaia and Rhamnella, and 2. the four thickenings smaller, sometimes forming a ring, H-shape more or less conspicuous, e.g. in Berchemia, Paliurus and Ziziphus. With respect to the concept of the genera Berchemiella and Berchemia, there were two different ideas among a number of taxonomists. H. Koidzumi, J. Ohwi and S. Kitamura place the former in Berchemia as a section, but on the contrary, T. Nakai, T. Yamazaki and K. Suessenguth consider Berchemiella and Berchemia as two separate genera. The data from pollen morphology support the latter treatment. Berchemia is closely related to Berchemiella, Chaydaia and Rhamnella in the habit and the floral morphology, but differs from the others by its disc well-developed, eularged and becoming cup-shaped in fruit. They are also different from each other in the characters of pollen grains. T. Yamazaki places the genus Chaydaia in Rhamnella as a section, but according to the pollen morphology to treat Chaydaia and Rhamnella as two independent genera woild be reasonable.  相似文献

本文研究并报道了百合科蜘蛛抱蛋亚族的花粉形态,主要结论如下:1.开口箭属含两种花粉类型即:(1)单槽、椭圆形;(2)无萌发孔、球形。具单槽、椭圆形花粉的种类含单型性核型,花形较小,柱头小而花柱短,雄蕊高于柱头;无萌发孔、球形花粉的种类则为单型性核型,花形较大,柱头常高度膨大,雄蕊常着生在花被筒基部。2.万年青属花粉为椭圆形,具单槽;蜘蛛抱蛋属花粉则为球形,无萌发孔;开口箭属花粉一部分与万年青属相似,另一部分则与蜘蛛抱蛋属雷同。3.蜘蛛抱蛋亚族中最原始的花粉类型为单槽椭圆形,具孔状或网状外壁纹饰。球形,无萌发孔,外壁具皱波状、瘤状或芽孢状纹饰的花粉为派生类型。4.蜘蛛抱蛋亚族种下花粉形状、萌发孔类型和外壁纹饰基本稳定,可作为分种特征。  相似文献

The pollen morphology of 28 species of Parnassia L. was investigated with light microscopy and scanning electron microscopy (SEM). The shape of pollen grains in this genus varies from subspheroidal to prolate in equatorial view and is three-lobed circular in the polar view. Pollen grains are usually radially symmetrical, isopolar, tricolporate or syntricolporate, with reticulate sculpture. The pollen characteristics among species are fairly similar to each other. Morphological information regarding the pollen grains shows that Parnassia is a natural genus. Based on exine ornamentation observed under SEM, three types of pollen grains were recognized: (i) type I, with foveolate-reticulate sculpture; (ii) type II, with a finely reticulate sculpture; and (iii) type III, with a coarsely reticulate sculpture. Most sections of this genus have one type of sculpture of pollen morphology, but Sect. Nectarotrilobos has three types of sculpture and Sect.Saxifragastrum has two types of sculpture. The present paper describes the pollen morphology of 31 species of the genus Quercus from China. The pollen grains were all examined with light microscope and scanning electron microscope, and those of some species under transmission electron microscope. Pollen grains of the genus are spheroidal or subspheroidal, 16.8-50.4μm in diameter, 3colporoidate or 3-colpate. The exine is 2-layered, 0.4-1.8μm thick, sexine thicker than nexine, granulated tuberculate or verrucate.  相似文献
