目的对嗜酸乳杆菌的毒性进行研究。方法采用大、小鼠急性毒性试验、Ames试验、小鼠骨髓细胞微核试验、小鼠精子畸形试验和大鼠30 d喂养等对嗜酸乳杆菌进行安全性试验研究。结果急性经口毒性试验表明,大、小鼠灌胃给予嗜酸乳杆菌,最大耐受剂量雌雄两性别均大于20.0 g/kg体重,Ames试验、微核试验和精子畸形试验结果均为阴性。大鼠30 d喂养试验结果表明各项指标均未见明显毒性反应。结论在本次实验条件下,嗜酸乳杆菌未见遗传毒性。由此可初步判定,使用嗜酸乳杆菌是安全可靠的。  相似文献

目的:对虾青素进行毒理学安全性评价,为其食用安全性提供科学依据。方法:通过急性经口毒性试验、Ames试验、骨髓细胞微核试验、精子畸形试验和大鼠30d喂养试验等毒理学评价试验,评估虾青素的食用安全性。结果:虾青素对雌、雄大鼠的急性经口最大耐受量(MTD)均大于19.0g/kg·BW。Ames试验、小鼠骨髓微核试验和精子畸形试验结果均未见该样品有致突变作用,大鼠30d喂养试验各项指标也均未见明显毒性反应。结论:虾青素急性毒性分级属无毒级,无遗传毒性,在该实验研究剂量和条件下,虾青素未见明显毒副作用。  相似文献

目的:评价牛初乳的安全性。方法:按照《保健食品检验与评价技术规范(2003版)》进行急性经口毒性试验、Ames试验、骨髓细胞微核试验、精子畸形试验和大鼠30d喂养试验等毒理学安全评价试验。结果:牛初乳大鼠经口急性毒性试验中MTD值>15.0g/kg·BW。小鼠骨髓微核试验、Ames试验、小鼠精子致畸试验对牛初乳样品没有致突变的作用。大鼠30d喂养试验各项指标也均未见明显毒性反应:对照组和各剂量组大鼠的生长发育均未见异常,血液学、血液生化、主要脏器重量、脏器系数均在正常值范围内,组织病理学检查未见与受试物有关的异常改变。结论:牛初乳属于无毒级、不会引起突变,大鼠30d喂养试验各项指标也均未见明显毒性反应。  相似文献

目的 评估益生菌粉(副干酪乳酪杆菌207-27)的毒理学安全性,为其应用提供依据。方法 通过大鼠急性经口毒性试验、细菌回复突变试验、小鼠红细胞微核试验、小鼠精母细胞染色体畸变试验及大鼠28 d经口毒性试验研究益生菌粉(副干酪乳酪杆菌207-27)的安全性。结果 大鼠急性经口毒性试验结果显示,益生菌粉(副干酪乳酪杆菌207-27)对大鼠的经口急性毒性LD50均大于15.00 g/(kg·BW),根据急性毒性分级标准属实际无毒。细菌回复突变试验、小鼠红细胞微核试验及小鼠精母细胞染色体畸变试验结果均显示阴性。大鼠28 d经口毒性试验结果表明,实验组大鼠体质量、摄食量、食物利用率、眼部状况、血液学指标、血液生化指标、脏器指数、大体及病理学检查结果与对照组相比差异均无统计学意义。结论 益生菌粉(副干酪乳酪杆菌207-27)具有良好的毒理学安全性。  相似文献

目的:对人参花食用安全性进行毒理学评价.方法:采用大鼠经口急性毒性试验、遗传毒性试验(Ames试验、小鼠骨髓细胞微核试验、小鼠精子畸形试验)、大鼠30 d喂养试验.结果:雌、雄大鼠经口最大耐受剂量(MTD)均大于17.4 g/kg·bw.3项遗传毒性试验结果均为阴性.在大鼠30 d喂养试验中,6.0 g/kg·bw,3.0 g/kg·bw及1.5g/kg· bw 3个剂量组的实验动物均生长发育良好,体重、摄食量、饮水量、血液学、血液生化学、脏器系数及病理组织学相关指标均未见异常变化.结论:人参花属于实际无毒物,未见遗传毒性,长期服用是安全的.  相似文献

本文主要研究桑黄纤孔菌子实体粉的毒理学并评价其安全性。利用小鼠急性毒性试验和遗传毒性试验(Ames试验、小鼠骨髓细胞微核试验、小鼠精子畸形试验)对桑黄纤孔菌子实体粉的毒性进行考察。试验结果显示,小鼠对桑黄纤孔菌子实体粉最大耐受量大于12g/kg;桑黄纤孔菌子实体粉在有或无S9的条件下,对组氨酸营养缺陷型鼠伤寒沙门氏菌TA97a、TA98、TA100及TA102均无潜在致突变性;桑黄纤孔菌子实体粉在小鼠骨髓细胞微核试验和小鼠精子畸形试验中均呈阴性反应,不同剂量的样品组与阴性对照组之间无显著性差异。该结果表明桑黄纤孔菌子实体粉基本无毒性,属实际无毒物质。本研究为桑黄纤孔菌子实体粉的产品开发和应用提供了科学依据。  相似文献

为确保以紫茎泽兰提取物为主要原药的植物源农药的安全性,对紫茎泽兰醇提物进行了小鼠经口急性毒性试验、大白兔急性皮肤和眼刺激试验、小鼠骨髓嗜多染红细胞微核计数及小鼠精子畸形试验等急性毒性和遗传毒性试验。结果表明,受试物对两种性别的小鼠经口急性毒性试验,LD50大于5000mg/kg,对大白兔皮肤无刺激性;小鼠骨髓嗜多染红细胞微核试验及小鼠精子畸形试验结果均为阴牲,受试物未见遗传毒性。  相似文献

以昆明小白鼠为实验动物,对类球红细菌Z1、沼泽红假单胞菌Z2及光合细菌分离株C2采用急性毒性试验、骨髓细胞微核试验和精子畸形试验进行安全性评定。结果表明,急性毒性试验:实验组小白鼠的一般状况、器官含水量、器官系数及血常规检查,与对照组差异均无显著性(P>0.05);骨髓细胞微核试验和精子畸形试验实验组与阴性对照组(生理盐水)差异均无显著性(P>0.05),与阳性对照组(环磷酰胺40 mg/kg)比较,骨髓细胞微核试验差异有极显著性(P<0.01),精子畸形率试验差异有显著性(P<0.05)。以上结果说明,三株光合细菌均不具有毒性。  相似文献

本文主要研究灵芝子实体醇提物的毒理学并评价其安全性,利用小鼠急性毒性试验和遗传毒性试验(Ames试验、小鼠骨髓细胞微核试验、小鼠精子畸形试验)对灵芝醇提物的毒性进行考察。试验结果显示,小鼠对灵芝醇提取物的最大耐受量是15 000mg/(kg?bw);灵芝醇提取物在有或无S9的条件下,对鼠伤寒沙门氏菌的突变型菌株TA97、TA98、TA100及TA102均无潜在致突变性;灵芝醇提取物在小鼠骨髓细胞微核试验和小鼠精子畸形试验中均呈阴性反应,不同剂量的样品组与阴性对照组之间无显著性差异。该结果表明灵芝醇提取物基本无毒性,属实际无毒物质。本研究为灵芝醇提取物的产品开发和应用提供了科学研究。  相似文献

3,3',4,4'-Tetrachloroazobenzene (TCAB) and 3,3',4, 4'-tetrachloroazoxybenzene (TCAOB) are dioxin-like chemicals that were investigated for toxicity in 13-week gavage studies in male and female B6C3F(1) mice and F344N rats by the National Toxicology Program. As part of the comprehensive toxicological investigation of these chemicals, peripheral blood smears from mice treated 5 days per week for 13 weeks with 0.1-30mg/kg/day TCAB or TCAOB were analyzed for the frequency of micronucleated (MN) normochromatic erythrocytes (NCE). Both chemicals produced significant increases in MN-NCE in male and female mice. In contrast to these positive results in subchronic exposure studies, no significant increases were seen in acute bone marrow MN tests in male mice administered three daily injections of 50-200mg/kg/day TCAB and TCAOB. The results with TCAB and TCAOB suggest that the routine integration of MN tests with subchronic toxicity studies may allow detection of mutagenic activity for some chemicals that fail to elicit responses in short-term, high dose tests. In addition, the integration of mutagenicity tests into general toxicity tests reduces the use of laboratory animals and the cost of the testing.  相似文献

本研究选取了一株白腐菌模式菌株进行了小鼠急性毒性试验、小鼠骨髓嗜多染红细胞微核试验以及小鼠精子畸形试验,以分析该白腐菌的食用安全性,为进一步应用白腐菌开发功能性食品提供数据支持。结果显示,小鼠经灌胃白腐菌,其LD50为6.76 g/kg(以白腐菌菌丝干重计)。微核试验和精子畸形试验结果均为阴性(P0.05)。试验结果表明,本次实验条件下,白腐菌对小鼠表现出一定的急性毒性,但未见遗传毒性,由此推断,在一定剂量范围内使用白腐菌作为食用材料是相对安全的,安全的剂量范围需要进一步扩大浓度梯度来确定。  相似文献

Five strains of male and female mice were infected with salivary gland-passaged murine cytomegalovirus (SG-MCMV) intraperitoneally. Although none of them became hyperglycemic, 13-30% of BALB/c male and female, ICR/Slc male and female and C57BL/6J male mice had abnormal values in glucose tolerance tests (GTT) 14 to 30 day post infection. Serum insulin levels in BALB/c male mice were low and infective virus was detected in the pancreas during the acute phase of infection. Histopathology showed mild edematous changes with inflammatory cell infiltrations in the pancreas. Viral antigen was located in acinar cells and occasionally in beta cells by an immunofluorescence test.  相似文献

Experiments were conducted to test the mutagenicity of wheat irradiated with 20 and 200 krad gamma-rays by feeding male and female mice for various periods starting from weaning time. The results obtained from dominant lethal tests, specific locus mutation test, studies on chromosome rearrangements in males and gonadal cell survival studies indicated no positive evidence for genetic and cytogenetic effects.  相似文献

Since male A/J mice are much more susceptible to both acute and subacute nephrotoxicity and the carcinogenic effect of ferric nitrilotriacetate than female mice, sex differences in the lipid peroxidation level after ferric nitrilotriacetate use were examined. The effects of orchiectomy and testosterone were also investigated. Male and female A/J mice were given a single intraperitoneal injection of ferric nitrilotriacetate (3 mg of iron/kg of body weight) and then thiobarbituric acid reactivity was determined in the liver and the kidney. Only male mice showed high thiobarbituric acid reactivity after 30 min, with the kidney showing higher activity than the liver. Castrated male mice showed a reduction in thiobarbituric acid reactivity, whereas testosterone-pretreated castrated male or testosterone-pretreated female mice showed increased thiobarbituric acid reactivity. In addition, daily intraperitoneal injections of ferric nitrilotriacetate resulted in the death of all normal male mice within 6 days, whereas all female and castrated male mice survived 3 months of treatment. Thus, male and female mice showed differences in ferric nitrilotriacetate-induced toxicity as reflected in the degree of lipid peroxidation and mortality.  相似文献

Nefopam (NEF) is a potent analgesic compound administered as a racemic mixture. Previous in vitro and in vivo studies with nefopam enantiomers have shown that (+)nefopam [(+)NEF] is substantially more potent than (-)nefopam [(-)NEF]. Differences between enantiomers have also been suggested in metabolic studies in vitro. The impact of these differences in vivo is not known because there is little or no information on the relative plasma concentrations of the enantiomers or on their kinetics. In this study, individual enantiomers of nefopam were synthesized and examined for acute toxicity in male and female rats and mice. Pharmacologic properties of enantiomers were examined using in vitro binding assays and antinociceptive tests in rats and mice. Additionally, a pharmacokinetic study was conducted in human volunteers. Subjects were administered 20 mg nefopam as Acupan(R) either as a 5- or 20-min intravenous infusion. In a control phase, subjects were administered only vehicle. Blood samples were collected through the following 24 h. Plasma samples were analyzed for individual enantiomers using a chiral assay developed for this purpose. The pharmacologic differences of previous studies were confirmed in receptor binding assays and in the hot plate and the formalin tests in mice. Neither enantiomer demonstrated substantial activity in the tail flick test in rats. No significant differences were revealed between LD(50) values of nefopam enantiomers after oral or intravenous administration in male and female rats or mice. There were no significant differences in AUC(0-infinity), C(max), or half-life between enantiomers following intravenous administration. Based on these findings, there is currently no compelling rationale to justify administering or monitoring individual enantiomers.  相似文献

Bacteriocin TSU4 is a novel antimicrobial peptide isolated from Catla catla gut isolate Lactobacillus animalis TSU4. It has been reported for its potential antimicrobial activity against fish pathogens and food spoilage bacteria. In vivo safety evaluation is necessary to determine its immunogenicity, toxicity, and importance in real-life applications. The present study was designed to evaluate the immunogenicity, acute and sub-chronic toxicity of bacteriocin TSU4 in BALB/c mice to ensure its safety in industrial application. Male BALB/c mice were administered intraperitoneally for immunogenicity assessment, by oral gavage with 50, 100, and 200 mg/kg/body weight for acute test and 0.5 mg/kg/day dose of bacteriocin TSU4 for sub-chronic toxicity test. Neither mortality nor any infections were observed during experimental period. There was no major increase in antibody titer during the immunogenicity test, and no mortality was observed during acute or sub-chronic toxicity tests. The LD50 value of bacteriocin TSU4 was found to be higher than 200 ± 0.45 mg/kg. No significant change in the serum biochemical markers, histopathological analysis and visual observation in spleen sizes was observed. These findings revealed that bacteriocin TSU4 is a non-immunogenic, safe, non-toxic, and could be a potential candidate for industrial applications in food preservation and aquaculture industries.  相似文献

Previous research has shown that repeated testing with a stimulus male is required for ovariectomized, hormone-primed female mice to become sexually receptive (show maximal lordosis quotients; LQs) and that drug-induced, epigenetic enhancement of estradiol receptor function accelerated the improvement in LQs otherwise shown by estrous females with repeated testing. We asked whether pre-exposure to male pheromones (‘pheromone priming’) would also accelerate the improvement in LQs with repeated tests and whether optogenetic inhibition of accessory olfactory bulb (AOB) projection neurons could inhibit lordosis in sexually experienced estrous female mice. In Experiment 1, repeated priming with soiled male bedding failed to accelerate the progressive improvement in LQs shown by estrous female mice across 5 tests, although the duration of each lordosis response and females' investigation of male body parts during the first test was augmented by such priming. In Experiment 2, acute optogenetic inhibition of AOB inputs to the forebrain during freely moving behavioral tests significantly reduced LQs, suggesting that continued AOB signaling to the forebrain during mating is required for maximal lordotic responsiveness even in sexually experienced females. Our results also suggest that pheromonal stimulation, by itself, cannot substitute for the full complement of sensory stimulation received by estrous females from mounting males that normally leads to the progressive improvement in their LQs with repeated testing.  相似文献
