Neurons program various patterns of sequential spikes as neural codes to guide animal behavior. Studies show that spike programming (capacity and timing precision) is influenced by inhibitory synaptic inputs and membrane afterhyperpolarization (AHP). Less is clear about how these inhibitory components regulate spike programming, which we investigated at the cortical neurons. Whole-cell current-clamp recording for action potentials and single channel recording for voltage-gated sodium channels (VGSC) were conducted at regular-spiking and fast-spiking neurons in the cortical slices. With quantifying the threshold potentials and refractory periods of sequential spikes, we found that fast-spiking neurons expressing AHP possess lower threshold potentials and shorter refractory periods, and the hyperpolarization pulse immediately after each of spikes lowers threshold potentials and shortens refractory periods at regular-spiking neurons. Moreover, the hyperpolarization pulses shorten the refractory periods for VGSC reactivation and threshold potentials for its sequential activation. Our data indicate that inhibitory components immediately after spikes, such as AHP and recurrent inhibition, improve spike capacity and timing precision via lowering the refractory periods and threshold potentials mediated by voltage-gated sodium channels.  相似文献

Cerebellum is involved in the motion coordination and working memory, to which the programming of sequential spikes at Purkinje cells is essential. It is not clear about the intrinsic mechanisms underlying spike capacity and timing precision as well as their postnatal maturation. We investigated the programming and intrinsic property of sequential spikes at Purkinje neurons during postnatal development by whole-cell recording in cerebellar slices. Cerebellar Purkinje neurons demonstrate the increasing of spike capacity and timing precision, as well as the lowering of refractory periods and threshold potentials during the postnatal maturation. In addition, the correlation between spike parameters and intrinsic properties converts to be more linear. This postnatal plasticity of neuronal intrinsic properties improves the timing precision and capacity of spike programming at cerebellar Purkinje neurons.  相似文献

Neural codes to guide well-organized behavior are thought to be the programmed patterns of sequential spikes at central neurons, in which the coordinative activities of voltage-gated ion channels are involved. The attention has been paid to study the role of potassium channels in spike pattern; but it is not clear how the intrinsic mechanism mediated by voltage-gated sodium channels (VGSC) influences the programming of sequential spikes, which we investigated at GABAergic cerebellar Purkinje cells and hippocampal interneurons by patch-clamp recording in brain slices. Spike capacity is higher at Purkinje cells than interneurons in response to the given intensities of inputs, and is dependent on input intensity. Compared to interneurons, Purkinje cells express the lower threshold potentials and the shorter refractory periods of sequential spikes. The increases of input intensities shorten spike refractory periods significantly. The threshold potentials for VGSC activation and the refractory periods for its reactivation are lower at Purkinje cells, and are reduced by the strong depolarization. We suggest that the VGSC-mediated threshold potentials and refractory periods are regulated by synaptic inputs, and navigate the programming of sequential spikes at the neurons.  相似文献

GABAergic neurons play a critical role in maintaining the homeostasis of brain functions for well-organized behaviors. It is not known about the dynamical change in signal encoding at these neurons during postnatal development. We investigated this issue at GFP-labeled GABAergic neurons by whole-cell recording in cortical slices of mice. Our results show that the ability of spike encoding at GABAergic neurons is improved during postnatal development. This change is associated with the reduction of refractory periods and threshold potentials of sequential spikes, as well as the improvement of linear correlations between intrinsic properties and spike capacity. Therefore, the postnatal maturation of the spike encoding capacity at GABAergic neurons will stabilize the excitatory state of cerebral cortex.  相似文献

Neurons generate spikes reliably with millisecond precision if driven by a fluctuating current—is it then possible to predict the spike timing knowing the input? We determined parameters of an adapting threshold model using data recorded in vitro from 24 layer 5 pyramidal neurons from rat somatosensory cortex, stimulated intracellularly by a fluctuating current simulating synaptic bombardment in vivo. The model generates output spikes whenever the membrane voltage (a filtered version of the input current) reaches a dynamic threshold. We find that for input currents with large fluctuation amplitude, up to 75% of the spike times can be predicted with a precision of ±2 ms. Some of the intrinsic neuronal unreliability can be accounted for by a noisy threshold mechanism. Our results suggest that, under random current injection into the soma, (i) neuronal behavior in the subthreshold regime can be well approximated by a simple linear filter; and (ii) most of the nonlinearities are captured by a simple threshold process.  相似文献

The timing of spiking activity across neurons is a fundamental aspect of the neural population code. Individual neurons in the retina, thalamus, and cortex can have very precise and repeatable responses but exhibit degraded temporal precision in response to suboptimal stimuli. To investigate the functional implications for neural populations in natural conditions, we recorded in vivo the simultaneous responses, to movies of natural scenes, of multiple thalamic neurons likely converging to a common neuronal target in primary visual cortex. We show that the response of individual neurons is less precise at lower contrast, but that spike timing precision across neurons is relatively insensitive to global changes in visual contrast. Overall, spike timing precision within and across cells is on the order of 10 ms. Since closely timed spikes are more efficient in inducing a spike in downstream cortical neurons, and since fine temporal precision is necessary to represent the more slowly varying natural environment, we argue that preserving relative spike timing at a ~10-ms resolution is a crucial property of the neural code entering cortex.  相似文献

Sensory information about the outside world is encoded by neurons in sequences of discrete, identical pulses termed action potentials or spikes. There is persistent controversy about the extent to which the precise timing of these spikes is relevant to the function of the brain. We revisit this issue, using the motion-sensitive neurons of the fly visual system as a test case. Our experimental methods allow us to deliver more nearly natural visual stimuli, comparable to those which flies encounter in free, acrobatic flight. New mathematical methods allow us to draw more reliable conclusions about the information content of neural responses even when the set of possible responses is very large. We find that significant amounts of visual information are represented by details of the spike train at millisecond and sub-millisecond precision, even though the sensory input has a correlation time of ~55 ms; different patterns of spike timing represent distinct motion trajectories, and the absolute timing of spikes points to particular features of these trajectories with high precision. Finally, the efficiency of our entropy estimator makes it possible to uncover features of neural coding relevant for natural visual stimuli: first, the system's information transmission rate varies with natural fluctuations in light intensity, resulting from varying cloud cover, such that marginal increases in information rate thus occur even when the individual photoreceptors are counting on the order of one million photons per second. Secondly, we see that the system exploits the relatively slow dynamics of the stimulus to remove coding redundancy and so generate a more efficient neural code.  相似文献

In vivo studies have shown that neurons in the neocortex can generate action potentials at high temporal precision. The mechanisms controlling timing and reliability of action potential generation in neocortical neurons, however, are still poorly understood. Here we investigated the temporal precision and reliability of spike firing in cortical layer V pyramidal cells at near-threshold membrane potentials. Timing and reliability of spike responses were a function of EPSC kinetics, temporal jitter of population excitatory inputs, and of background synaptic noise. We used somatic current injection to mimic population synaptic input events and measured spike probability and spike time precision (STP), the latter defined as the time window (Deltat) holding 80% of response spikes. EPSC rise and decay times were varied over the known physiological spectrum. At spike threshold level, EPSC decay time had a stronger influence on STP than rise time. Generally, STP was highest (6 ms) triggered spikes at lower temporal precision (>or=6.58 ms). We found an overall linear relationship between STP and spike delay. The difference in STP between fast and slow compound EPSCs could be reduced by incrementing the amplitude of slow compound EPSCs. The introduction of a temporal jitter to compound EPSCs had a comparatively small effect on STP, with a tenfold increase in jitter resulting in only a five fold decrease in STP. In the presence of simulated synaptic background activity, precisely timed spikes could still be induced by fast EPSCs, but not by slow EPSCs.  相似文献

大脑神经元胞外单细胞动作电位(即锋电位)的检测和分类是提取神经元脉冲序列、研究神经系统信息处理机制的关键.为了提高锋电位的检出率和分类的正确性,设计了一种处理多通道锋电位记录信号的算法,用于分析微电极阵列记录的大鼠海马神经元锋电位信号,电极阵列上的测量点排列紧密,4个通道可以同时记录到来自相同神经元的信号.该算法首先利用一种多通道阈值检测法检出四通道记录信号中的锋电位,然后利用一种基于复合锋电位的主成分特征参数分类法将锋电位分类.仿真数据和实验记录信号的检验结果表明:与相应的单通道算法相比,该算法的锋电位检出率和分类的正确性显著提高,并且可以增加单次实验测得的神经元数目.因此,该算法为实现神经元锋电位的自动检测提供了一种简单有效的新 方法.  相似文献

Background

Action potentials are the essential unit of neuronal encoding. Somatic sequential spikes in the central nervous system appear various in amplitudes. To be effective neuronal codes, these spikes should be propagated to axonal terminals where they activate the synapses and drive postsynaptic neurons. It remains unclear whether these effective neuronal codes are based on spike timing orders and/or amplitudes.

Methodology/Principal Findings

We investigated this fundamental issue by simultaneously recording the axon versus soma of identical neurons and presynaptic vs. postsynaptic neurons in the cortical slices. The axons enable somatic spikes in low amplitude be enlarged, which activate synaptic transmission in consistent patterns. This facilitation in the propagation of sequential spikes through the axons is mechanistically founded by the short refractory periods, large currents and high opening probability of axonal voltage-gated sodium channels.

Conclusion/Significance

An amplification of somatic incomplete spikes into axonal complete ones makes sequential spikes to activate consistent synaptic transmission. Therefore, neuronal encoding is likely based on spike timing order, instead of graded analogues.  相似文献

Spike encoding at GABAergic neurons plays an important role in maintaining the homeostasis of brain functions for well-organized behaviors. The rise of intracellular Ca2+ in GABAergic neurons causes synaptic plasticity. It is not clear how intracellular Ca2+ influences their spike encoding. We have investigated this issue at GFP-labeled GABAergic cortical neurons and cerebellar Purkinje cells by whole-cell recording in mouse brain slices. Our results show that an elevation of intracellular Ca2+ by infusing adenophostin-A lowers spike encoding at GABAergic cortical neurons and enhances encoding ability at cerebellar Purkinje cells. These differential effects of cytoplasmic Ca2+ on spike encoding are mechanistically associated with Ca2+-induced changes in the refractory periods and threshold potentials of sequential spikes, as well as with various expression ratios of CaM-KII to calcineurin in GABAergic cortical neurons and cerebellar Purkinje cells.  相似文献

It is much debated on what time scale information is encoded by neuronal spike activity. With a phenomenological model that transforms time-dependent membrane potential fluctuations into spike trains, we investigate constraints for the timing of spikes and for synchronous activity of neurons with common input. The model of spike generation has a variable threshold that depends on the time elapsed since the previous action potential and on the preceding membrane potential changes. To ensure that the model operates in a biologically meaningful range, the model was adjusted to fit the responses of a fly visual interneuron to motion stimuli. The dependence of spike timing on the membrane potential dynamics was analyzed. Fast membrane potential fluctuations are needed to trigger spikes with a high temporal precision. Slow fluctuations lead to spike activity with a rate about proportional to the membrane potential. Thus, for a given level of stochastic input, the frequency range of membrane potential fluctuations induced by a stimulus determines whether a neuron can use a rate code or a temporal code. The relationship between the steepness of membrane potential fluctuations and the timing of spikes has also implications for synchronous activity in neurons with common input. Fast membrane potential changes must be shared by the neurons to produce synchronous activity.  相似文献

Recent experimental reports have suggested that cortical networks can operate in regimes were sensory information is encoded by relatively small populations of spikes and their precise relative timing. Combined with the discovery of spike timing dependent plasticity, these findings have sparked growing interest in the capabilities of neurons to encode and decode spike timing based neural representations. To address these questions, a novel family of methodologically diverse supervised learning algorithms for spiking neuron models has been developed. These models have demonstrated the high capacity of simple neural architectures to operate also beyond the regime of the well established independent rate codes and to utilize theoretical advantages of spike timing as an additional coding dimension.  相似文献

Correlation between spike trains or neurons sometimes indicates certain neural coding rules in the visual system. In this paper, the relationship between spike timing correlation and pattern correlation is discussed, and their ability to represent stimulus features is compared to examine their coding strategies not only in individual neurons but also in population. Two kinds of stimuli, natural movies and checkerboard, are used to arouse firing activities in chicken retinal ganglion cells. The spike timing correlation and pattern correlation are calculated by cross-correlation function and Lempel–Ziv distance respectively. According to the correlation values, it is demonstrated that spike trains with similar spike patterns are not necessarily concerted in firing time. Moreover, spike pattern correlation values between individual neurons’ responses reflect the difference of natural movies and checkerboard; neurons cooperate with each other with higher pattern correlation values which represent spatiotemporal correlations during response to natural movies. Spike timing does not reflect stimulus features as obvious as spike patterns, caused by their particular coding properties or physiological foundation. As a result, separating the pattern correlation out of traditional timing correlation concept uncover additional insight in neural coding.  相似文献

Acidosis, associated with metabolic disorders, leads to the pathological changes of cognition and behavior in the clinical practices of neurology and psychology. The cellular mechanisms underlying these cerebral dysfunctions remain unclear. By using electrophysiological approach and changing extracellular pH, we have investigated the effects of acidic environment on cortical GABAergic neurons in terms of their abilities of firing spikes and responses to synaptic inputs. Artificial cerebral spinal fluid in low pH impairs the responses to excitatory synaptic inputs and the abilities of encoding sequential spikes at these GABAergic neurons. The impairments of neuronal spiking are associated with the increases of refractory periods and threshold potentials. Our studies reveal that acidosis may impair cortical GABAergic neurons and in turn deteriorate brain functions, in which their final targets are voltage-gated sodium channels and glutamate receptor-channels.  相似文献

Acupuncturing the Zusanli (ST 36) point with different types of manual acupuncture manipulations (MAs) and different frequencies can evoke a lot of neural response activities in spinal dorsal root neurons. The action potential is the basic unit of communication in the neural response process. With the rapid development of the electrode acquisition technology, we can simultaneously obtain neural electrical signals of multiple neurons in the target area. So it is crucial to extract spike trains of each neuron from raw recorded data. To solve the problem of variability of the spike waveform, this paper adopts a optimization algorithm based on model to improve the wave-cluster algorithm, which can provide higher accuracy and reliability. Further, through this optimization algorithm, we make a statistical analysis on spike events evoked by different MAs. Results suggest that numbers of response spikes under reinforcing manipulations are far more than reducing manipulations, which mainly embody in synchronous spike activities.  相似文献

In many cases, neurons process information carried by the precise timings of spikes. Here we show how neurons can learn to generate specific temporally precise output spikes in response to input patterns of spikes having precise timings, thus processing and memorizing information that is entirely temporally coded, both as input and as output. We introduce two new supervised learning rules for spiking neurons with temporal coding of information (chronotrons), one that provides high memory capacity (E-learning), and one that has a higher biological plausibility (I-learning). With I-learning, the neuron learns to fire the target spike trains through synaptic changes that are proportional to the synaptic currents at the timings of real and target output spikes. We study these learning rules in computer simulations where we train integrate-and-fire neurons. Both learning rules allow neurons to fire at the desired timings, with sub-millisecond precision. We show how chronotrons can learn to classify their inputs, by firing identical, temporally precise spike trains for different inputs belonging to the same class. When the input is noisy, the classification also leads to noise reduction. We compute lower bounds for the memory capacity of chronotrons and explore the influence of various parameters on chronotrons' performance. The chronotrons can model neurons that encode information in the time of the first spike relative to the onset of salient stimuli or neurons in oscillatory networks that encode information in the phases of spikes relative to the background oscillation. Our results show that firing one spike per cycle optimizes memory capacity in neurons encoding information in the phase of firing relative to a background rhythm.  相似文献

Cell death in cerebral ischemia is presumably initiated by neural excitotoxicity resulted from the dysfunction of inhibitory neurons in early stage. Molecular processes underlying the ischemic injury of inhibitory neurons remain to be elusive, which we investigated by biochemical manipulations with cellular imaging and patch clamp at GFP-labeled GABAergic cells in cortical slices. Ischemia induces Ca2+ elevation, acidosis and dysfunction in GABAergic cells. An elevation of cytoplasmic Ca2+ or H+ impairs the encoding of action potentials in these neurons. The effects of Ca2+ and H+ are additive in nature and occlude ischemic outcomes. Ischemia impairs spike production through prolonging spike refractory periods and raising threshold potentials. Therefore, calcium toxicity and acidosis during ischemia synergistically impair the dynamics of sodium channels and function of cortical GABAergic neurons, which lead to neural excitotoxicity. Our results also suggest that the cocktail therapeutics is needed to prevent neuronal death from ischemia.  相似文献

Sensory neurons code information about stimuli in their sequence of action potentials (spikes). Intuitively, the spikes should represent stimuli with high fidelity. However, generating and propagating spikes is a metabolically expensive process. It is therefore likely that neural codes have been selected to balance energy expenditure against encoding error. Our recently proposed optimal, energy-constrained neural coder (Jones et al. Frontiers in Computational Neuroscience, 9, 61 2015) postulates that neurons time spikes to minimize the trade-off between stimulus reconstruction error and expended energy by adjusting the spike threshold using a simple dynamic threshold. Here, we show that this proposed coding scheme is related to existing coding schemes, such as rate and temporal codes. We derive an instantaneous rate coder and show that the spike-rate depends on the signal and its derivative. In the limit of high spike rates the spike train maximizes fidelity given an energy constraint (average spike-rate), and the predicted interspike intervals are identical to those generated by our existing optimal coding neuron. The instantaneous rate coder is shown to closely match the spike-rates recorded from P-type primary afferents in weakly electric fish. In particular, the coder is a predictor of the peristimulus time histogram (PSTH). When tested against in vitro cortical pyramidal neuron recordings, the instantaneous spike-rate approximates DC step inputs, matching both the average spike-rate and the time-to-first-spike (a simple temporal code). Overall, the instantaneous rate coder relates optimal, energy-constrained encoding to the concepts of rate-coding and temporal-coding, suggesting a possible unifying principle of neural encoding of sensory signals.  相似文献
