Effect of soil pH and sewage sludge on VA mycorrhizal infection of soybeans   总被引:1,自引:0,他引:1  
Summary Small plots were amended in 1976 or 1978 with four kinds of sewage sludge. The sludges represented samples considered to be relatively free of heavy metals as well as sludges highly contaminated with heavy metals. Sludges were added to a silt loam soil at rates of 224 or 448 Mgha−1. The soils were maintained at a high or low pH regime. In 1984, soybeans (Glycine max L. Merril. var. ‘Clark’) were planted and grown to the R4 stage. After harvest, roots were removed from the soil, washed, and examined for VA mycorrhizal infection. It was found that the heavy metal content of the sludge alone was generally not related to determining the extent of mycorrhizal infection. A heat treated sludge, high in heavy metals, exhibited the highest degree of mycorrhizal infection when the soil was maintained at a pH of 6.2. With this treatment, 52% of the root segments examined were infected by mycorrhiza. When the same sludge was added to a soil with a slightly lower pH (5.7) none of the roots examined were infected by mycorrhiza. When soybean roots were examined from soils that received no sludge and were maintained at either a low (5.6) or high (6.2) pH, there was no significant difference in mycorrhizal infection between the pH regimes. These results therefore indicate that sewage sludge may inhibit mycorrhizal infection if the sludge contains a high concentration of heavy metals and the sludge is applied to the soil with a low pH. A moderately thermophilic and acidophilic sulfur-oxidizing bacterium named S2, was isolated from coal heap drainage. The bacterium was motile, Gram-negative, rod-shaped, measured 0.4 to 0.6 by 1 to 2 μm, and grew optimally at 42–45°C and an initial pH of 2.5. The strain S2 grew autotrophically by using elemental sulfur, sodium thiosulfate and potassium tetrathionate as energy sources. The strain did not use organic matter and inorganic minerals including ferrous sulfate, pyrite and chalcopyrite as energy sources. The morphological, biochemical, physiological characterization and analysis based on 16S rRNA gene sequence indicated that the strain S2 is most closely related to Acidithiobacillus caldus (>99% similarity in gene sequence). The combination of the strain S2 with Leptospirillum ferriphilum or Acidithiobacillus ferrooxidans in chalcopyrite bioleaching improved the copper-leaching efficiency. Scanning electron microscope (SEM) analysis revealed that the chalcopyrite surface in a mixed culture of Leptospirillum ferriphilum and Acidithiobacillus caldus was heavily etched. The energy dispersive X-ray (EDX) analysis indicated that Acidithiobacillus caldus has the potential role to enhance the recovery of copper from chalcopyrite by oxidizing the sulfur formed during the bioleaching progress.  相似文献

【目的】土壤重金属污染问题日益受到关注,其中钒污染逐渐成为研究热点。淋洗是土壤修复的重要手段,但存在污染大、成本高的缺点。生物淋洗技术因其经济高效且环保的特点能够应用于土壤的修复,但其对钒污染土壤的修复,认识仍非常有限。【方法】本研究采用嗜酸性氧化亚铁硫杆菌对钒污染土壤进行了生物淋洗试验,通过影响因素试验探究了钒的最佳浸出条件,并应用扫描电子显微镜-能量色散X射线谱分析了钒在淋洗过程中的变化,最后对代谢产物进行了解析。【结果】微生物次生代谢产物能促进土壤中钒的溶出。氧化亚铁硫杆菌对土壤钒的浸出效率较高,生物淋洗20 d后土壤中钒的浸出率达到27.4%,进一步的影响因素试验表明,在固体浓度为3%、接种体积为10%、初始pH值为1.8、初始Fe2+的浓度为3.0 g/L的条件下,土壤中钒的浸出效果最佳。SEM-EDS分析证实生物淋洗后土壤中钒含量减少,其中以非残渣态形式存在的钒更容易被浸出。代谢组学分析显示氧化亚铁硫杆菌在浸出过程中产生了大量代谢产物来应对重金属胁迫。【结论】生物淋洗技术能够有效地实现土壤钒污染的修复,本研究为钒污染土壤提供了一种环境友好的修复方式。  相似文献

Symptoms of fairy rings caused by Lepista sordida have been reported on Zoysiagrass (Zoysia spp.) turf maintained at fairway height (2 cm), but not on bentgrass (Agrostis spp.) maintained at putting green height (0.5 cm). The mycelia of this fungus inhabit primarily the upper 0–2 cm layer of the soil extending into the thatch. To compare conditions for the mycelial growth in Z. matrella turf to those in A. palustris turf, we examined the effects of nutrients, temperature, water potential, and pH in the field as well as in the laboratory. Greater growth of the mycelia was observed in medium that included hot water extracts from soil of the 0–1 cm zone in Z. matrella turf compared to that from A. palustris. The upper soil layer in Z. matrella turf contained more organic matter from clippings than that in A. palustris. The temperature and water potential of the 0–2 cm soil zone in Z. matrella turf were also more favorable for the mycelial growth. The soil pH values of this zone in Z. matrella turf were less favorable compared to A. palustris but within the range for accelerating mycelial growth. Biological autotrophic sulfur oxidation processes have been proposed to remove heavy metals from wastewater treatment sludge by bioleaching. We made a characterization of the microbial population in batch and continuous sludge bioleaching reactors using fluorescent in situ hybridization of fluorescently-labeled oligonucleotidic probes targeting rRNA in a ‚top to bottom approach’. Batch incubations of sludge with 0.2% (w/v) elemental sulfur resulted in a pH value of 5. Alpha-Proteobacteria hybridizing with probe ALF1b were dominant in this incubation. Members of the Acidophilium-group (hybridizing with probe Acdp821) of Nitrospira/Leptospirillum phylum (Ntspa712 probe) and from the archaeal domain (ARCH915) were also detected. When sludge was incubated with 1% elemental sulfur in batch or continuous reactor experiments, final pH values were always below 2. Active microbial communities consisted almost exclusively of gamma-Proteobacteria (hybridizing with probe GAM42a). However, further hybridization experiments with probe Thio820 targeting Acidithiobacillus ferroxidans and Acidithiobacillus thioxidans gave negative results. A new probe, named THIO181, encompassing all known members of the genus was designed. Hybridization perfomed with THIO181 and GAM42a showed a perfect co-localization of the hybridization signals. Further hybridization experiments with probe THIO181 and THC642, specific for the species Acidithiobacillus caldus, confirmed that this bacteria was largely responsible for the sulfur oxidation reaction in our acidophilic sludge bioleaching reactors.  相似文献

An anionic trypsin (TRY-EP) was purified from North Pacific krill (Euphausia pacifica) by ammonium sulfate precipitation, ion-exchange and gel-filtration chromatography. The purified enzyme was identified as a trypsin by LC-ESI-MS/MS analysis. The relative molecular mass of TRY-EP was 33 kDa, with isoelectric point of 4.5. The histidine, tryptophan, arginine, lysine, aspartic acid and glutamic acid residues were functional groups to TRY-EP. TRY-EP was activated by Ca2+ and Mg2+ and inhibited by some heavy metal ions (Zn2+, Cu2+, Pb2+ and Hg2+), organic solvents (ethanol, glycerin, DMSO and acetone) and specific trypsin inhibitors (benzamidine, CEOM, SBTI and TLCK). TRY-EP was active over a wide pH (6.0–11.0) and temperature (10–70°C) range, with optimum of pH 9.0 and 40–50°C. TRY-EP was stable between pH 6.0 and 11.0 and below 30°C. Compared with some trypsins from the Temperate and Tropical Zone organisms, TRY-EP and other trypsins from the Frigid Zone organisms have higher affinity to substrate and 2–42-fold physiological efficiency.  相似文献

Cupriavidus metallidurans CH34 and related strains are adapted to metal contaminated environments. A strong resistance to environmental stressors and adaptation make it ideal strains for survival in decreasing biodiversity conditions and for bioaugmentation purposes in environmental applications. The soil bacterium C. metallidurans is able to grow chemolithoautotrophically on hydrogen and carbon dioxide allowing a strong resilience under conditions lacking organic matter. The biofilm growth on soil particles allows coping with starvation or bad conditions of pH, temperature and pollutants. Its genomic capacity of two megaplasmids encoding several heavy metal resistance operons allowed growth in heavy metal contaminated habitats. In addition its specific siderophores seem to play a role in heavy metal sequestration besides their role in the management of bioavailable iron. Efflux ATPases and RND systems pump the metal cations to the membrane surface where polysaccharides serve as heavy metal binding and nucleation sites for crystallisation of metal carbonates. These polysaccharides contribute also to flotation under specific conditions in a soil-heavy metals–bacteria suspension mixture. An inoculated moving bed sand filter was constructed to treat heavy metal contaminated water and to remove the metals in the form of biomass mixed with metal carbonates. A membrane based contactor allowed to use the bacteria as well in a versatile wastewater treatment system and to grow homogeneously formed heavy metal carbonates. Its behaviour toward heavy metal binding and flotation was combined in a biometal sludge reactor to extract and separate heavy metals from metal contaminated soils. Finally its metal-induced heavy metal resistance allowed constructing whole cell heavy metal biosensors which, after contact with contaminated soil, waste, solids, minerals and ashes, were induced in function of the bioavailable concentration (Cd, Zn, Cu, Cr, Co, Ni, Tl, Pb and Hg) in the solids and allowed to investigate the speciation of immobilization of those metals. Molecular characterization based on 16s rDNA gene sequence analysis of bacterial colonies isolated from endosulfan contaminated soil showed the presence of Ochrobacterum sp, Burkholderia sp, Pseudomonas alcaligenes, Pseudomonas sp and Arthrobacter sp which degraded 57–90% of α-endosulfan and 74–94% of β-endosulfan after 7days. Whole cells of Pseudomonas sp and Pseudomonas alcaligenes showed 94 and 89% uptake of α-isomer and 86 and 89% of β-endosulfan respectively in 120 min. In Pseudomonas sp, endosulfan sulfate was the major metabolite detected during the degradation of α-isomer, with minor amount of endosulfan diol while in Pseudomonas alcaligenes endosulfan diol was the only product during α-endosulfan degradation. Whole cells of Pseudomonas sp also utilized 83% of endosulfan sulfate in 120 min. In situ applications of the defined consortium consisting of Pseudomonas alcaligenes and Pseudomonas sp (1:1) in plots contaminated with endosulfan showed that 80% of α-endosulfan and 65% of β-endosulfan was degraded after 12 weeks of incubation. Endosulfan sulfate formed during endosulfan degradation was subsequently degraded to unknown metabolites. ERIC-PCR analysis indicated 80% survival of introduced population of Pseudomonas alcaligenes and Pseudomonas sp in treated plots.  相似文献

The Effects of Bacterial Leaching on Metal Partitioning in Sewage Sludge   总被引:2,自引:0,他引:2  
The partitioning of Mn, Al, Zn, Cu and Ti ions in municipal sewage sludge was investigated before and after bioleaching processes effectuated by Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans. Oxidation–reduction potential increase and pH decrease were obtained as a result of bacterial activity. A less pronounced and constant decrease was obtained with A. ferrooxidans, whereas A. thiooxidans presented a lag phase before a steep pH decrease. Metal solubilization was accomplished in experimental systems supplemented with energy source, Fe2+ for A. ferrooxidans and S0 for A. thiooxidans. Solubilization efficiency differed for each metal except for Al, and was relatively similar for either organism. Metal partitioning was conducted using a five-step sequential extraction procedure before and after the bioleaching. The results indicated that Zn and Mn ions were mostly associated with the organic fraction, whereas Cu, Al and Ti ions with the sulphide/residue fraction. The bioleaching process caused prompt solubilization of metals mostly associated with the more labile fractions (exchangeable, adsorbed and organically bound metals), whereas those associated to the less labile ones (EDTA and sulphide/residue fractions) were exchanged towards more labile fractions.  相似文献

The lelvels of seven heavy metals and their toxicity towardGanoderma lucidum under various cultivation conditions were assessed. The contents of Mn, Cu, Zn, Cd, Hg, Pb and U in the fruitbodies of cultivatedG. lucidum, and sawdust substrates were determined to be at trace levels for U, 0.01–0.1 μg/g for Cd and Hg, and 1–5 μg/g for Pb, 10–120 μg/g for Mn, Cu and Zn. The effects of heavy metals, on the growth of mycelia ofG. lucidium in pure cultures were examined over a wide range of concentrations (10–3,000 μg/ml), and their toxicities were found to decrease in the order: Hg>Cd>Cu>U>Pb>Mn=Zn. The translocation and accumulation of Zn from contaminated substrates (at 10 μg/g) in fruitbodies were investigated by using65Zn tracer, andG. lucidum was found to take up Zn with an efficiency of >60%, leading to accumulation of >100 μ/g, in fruitbodies and >80 μ/g Zn in basidiospores.  相似文献

A defined mixed bacterial culture was established which catalyzed dissimilatory sulfate reduction, using glycerol as electron donor, at pH 3.8–4.2. The bacterial consortium comprised a endospore-forming sulfate reducing bacterium (isolate M1) that had been isolated from acidic sediment in a geothermal area of Montserrat (West Indies) and which had 94% sequence identity (of its 16S rRNA gene) to the Gram-positive neutrophile Desulfosporosinus orientis, and a Gram-negative (non sulfate-reducing) acidophile (isolate PFBC) that shared 99% gene identity with Acidocella aromatica. Whilst M1 was an obligate anaerobe, isolate PFBC, as other Acidocella spp., only grew in pure culture in aerobic media. Analysis of microbial communities, using a combination of total bacterial counts and fluorescent in situ hybridization, confirmed that concurrent growth of both bacteria occurred during sulfidogenesis under strictly anoxic conditions in a pH-controlled fermenter. In pure culture, M1 oxidized glycerol incompletely, producing stoichiometric amounts of acetic acid. In mixed culture with PFBC, however, acetic acid was present only in small concentrations and its occurrence was transient. Since M1 did not oxidize acetic acid, it was inferred that this metabolite was catabolized by Acidocella PFBC which, unlike glycerol, was shown to support the growth of this acidophile under aerobic conditions. In fermenter cultures maintained at pH 3.8–4.2, sulfidogenesis resulted in the removal of soluble zinc (as solid phase ZnS) whilst ferrous iron remained in solution. Potential syntrophic interactions, involving hydrogen transfer between M1 and PFBC, are discussed, as is the potential of sulfidogenesis in acidic liquors for the selective recovery of heavy metals from wastewaters.  相似文献

The present study was undertaken to investigate the potential of enriched indigenous sulfur-and-iron-oxidizing microorganisms in the bioleaching of Cu, Ni, Zn and Fe from textile sludges by using elemental sulfur and ferrous sulfate (FS), respectively, as an energy source under batch conditions. The experiments were performed with three different textile sludges (S1, S2 and S3) at initial neutral pH of the sludges procured from different parts of the country i.e., UP, Haryana and Punjab. The three sludges used were not only procured from different parts of the country but also differ in physiochemical characteristics. The extent of heavy metals solubilization in each sludge was found to be different using sulfur- and iron-oxidizing microorganisms. The results of the study indicate that sulfur-oxidizing microorganisms were found more efficient in the bioleaching process, irrespective of any sludge. The use of sulfur-oxidizing microorganisms led to higher solubilization of heavy metals and after 7 days of bioleaching about 84–96% Cu, 64–78% Ni, 81–92% Zn and 74–88% Fe were removed compared to 62–73% Cu, 62–66% Ni, 74–78% Zn and 70–78% Fe using iron-oxidizing microorganisms. This study had shown the feasibility of applying the bioleaching process to textile sludge contaminated with heavy metals. The results of the present study indicate that the bioleached sludge would be safer for land application.  相似文献

Iron- and sulfur-oxidizing bacteria identified as Thiobacillus ferrooxidans and T. thiooxidans were successfully enriched from various soil samples contaminated with heavy metals and organic compounds. Depending on the growth medium, the soil sample, and the type of contaminant, the indigenous isolates solubilized > 50% of most of the heavy metals present in the solid sample (As, Cd, Co, Cr, Cu, Ni, V, Zn, B, Be). Leaching with T. ferrooxidans strains resulted in total extraction of Cd, Co, Cu, and Ni. With sulfur-oxidizing bacteria > 80% of Cd, Co, Cu, and Zn was mobilized from rainwater sludge. Pb and Ba were not detected in the leachate, given the insolubility of their sulfate compounds. An increase in pulp density up to 20%, indicating 6.6% total organic carbon in the soil and rubble leach experiment (sample 557), did not inhibit the growth of the indigenous T. ferrooxidans strain. In view of these results, bioleaching appears to have some potential for remediation of heavy metal contaminated soils.  相似文献

This greenhouse study aimed to determine the effect of colonization by the arbuscular mycorrhizal (AM) fungus (Glomus intraradices Schenck & Smith) on the “wild” tobacco (Nicotiana rustica L. var. Azteca), under soil–zinc (Zn) conditions. Plants of N. rustica were grown in AM or non-AM inoculated substrate and subjected to four soil–[Zn] concentrations (0, 50, 100, and 250 mg Zn kg−1 dry soil). The AM root colonization increased markedly from 14 to 81% with the increasing soil–[Zn] and the mycorrhizal structures were significantly more abundant at the highest soil–[Zn], suggesting that Zn may be involved directly or indirectly in AM root colonization. In addition, total Zn content or Zn concentrations in shoots and roots were shown to increase as soil–[Zn] increased in both AM and non-AM plants. As for the growth parameters studied, there were no significant differences between treatments despite the increase in Zn content or concentration. The AM roots subjected to the highest soil–[Zn] had a significant reduction by about 50% of total Zn content and Zn concentration compared to non-AM roots. Still, the relative extracted Zn percentage decreased dramatically as soil–[Zn] increased. Soil pH was significantly lower in non-AM than AM treatments at the highest soil–[Zn]. In summary, AM plants (particularly roots) showed lower Zn content and concentration than non-AM plants. In this regard, the AM fungi have a protective role for the host plant, thus playing an important role in soil-contaminant immobilization processes; and, therefore, are of value in phytoremediation, especially when heavy metals approach toxic levels in the soil.  相似文献

An “ex situ” microbial method for the removal of heavy metals from soil is described. Elemental sulfur was added to generate the lixiviant in shaker flask experiments in which soil sampled from a polluted agricultural field was treated. The biotic oxidation of sulfur to sulfuric acid resulted in significant drop in pH of the bioleaching liquor from 6.94 to 1.8 after 50 days. In batches operated at very low (10 g/kg) sulfur concentrations, pH changed from 6.94 to 5.45. The 50 g/kg soil sulfur concentration was found to be most beneficial to the solubilization process because more than 95% of metals such as zinc (Zn), cadmium (Cd), and nickel (Ni) were recovered while approximately 67% of manganese (Mn) got solubilized. The least concentration of dissolved metals was lead (Pb) – (25%) and chromium (Cr) – (10%). Sulfate accumulation rose to 47% in samples spiked with 50 g/kg soil of sulfur. At lower sulfur concentrations, the sulfates generated were higher than the amount of sulfur added. The microbial process compared well to the abiotic process involving extraneous addition of sulfuric acid except that very high concentrations of acid had to be used. The treatment of the bioleaching wastewater promoted precipitation of the dissolved metals into insoluble hydroxides making discharge of the effluent into the environment safe. The leached soil recovered sufficiently for agricultural use after quick lime and animal manure was used to improve, stabilize, and restore its physical, chemical, and biological conditions.  相似文献

Grass fallow shifting cultivation is an important land use practice in the highlands of Bhutan. Part of the nutrient pool contained in soil organic matter is made available for the traditional buckwheat (Fagopyrum tataricum) crop through a highly labor intensive system exposing 250–500 MT soil ha−1 to temperatures of 500°C and above. Dry topsoil is collected in mounds and burned using plant biomass or manure and soil organic matter as fuel. Labor input ranged from 150–401 days ha−1 with land preparation accounting for 65–85% of the total requirement. The burning increased soil pH from 6.0 to 6.9 and available K from 34 to 69 mg kg−1. Soil organic C and total N were reduced from 3.3 and 0.17% to 0.8 and 0.08%, respectively. Release of P from soil organic matter and plant material and reduction in C/N ratio resulting in increased N availability are considered the most essential effects required for good crop yields. Through the burning about 16 MT of C and 470 kg N ha−1 are released into the atmosphere. Fallow periods of 15–20 years are required for the system to be sustainable. The toxic and growth inhibiting effects of methyl tert-butyl ether (MTBE) on the hydrocarbon-degrading Pseudomonas veronii T1/1 strain (isolated from gasoline contaminated soil) were studied. In our experiments, the MIC of MTBE was found to be 60 mM and the EC50 was 51.7 mM. In the concentration range 0–30 mM, MTBE did not significantly influence the growth parameters of this bacterium, but at concentrations over 30 mM MTBE exerted a significant growth inhibiting effect. In the presence of 70 mM MTBE, the specific growth rate dropped from 0.4731 to 0.1201 h−1, while the length of the lag period increased from 5.41 to 17.01 h and the yield coefficient declined from 0.2652 to 0.0718 g g−1. MTBE at 100 mM inhibited the growth of this strain completely. These findings may have important environmental implications, as high concentrations of MTBE could influence the efficiency of soil and groundwater bioremediation processes significantly.  相似文献

Decolorization of textile reactive azo dyes by a strain of bacteria (SL186) isolated from a contaminated site was investigated. SL186 was identified as Clostridium bifermentans by phenotypic characterization and 16S rDNA sequence comparison. Under anaerobic conditions, SL186 had decolorized the dyes Reactive Red 3B-A, Reactive Black 5, and Reactive Yellow 3G-P by over 90% after 36 h post-inoculation. The bacterium retained decolorizing activity over a wide range of pH values (6–12), with peak activity at pH 10. Additionally, SL186 decolorized a relatively high concentration of Reactive Red 3B-A dye (1,000 ppm) by over 80% and raw industrial effluent effectively. The addition of glucose increased the decolorization rate a little. Spectrophotometric analyses of the reactive dyes showed no distinct peak indicating aromatic amines. However, a new peak was detected between 300 and 450 nm from the decolorized raw industrial effluent. These results suggest that C. bifermentans SL186 is a suitable bacterium for the biological processing of dye-containing wastewater.  相似文献

Three strains of gram-positive, motile, rod-shaped and boron (B)-tolerant bacterium were isolated from naturally B containing soil of Hisarcik area in the Kutahya Province, Turkey. The strains, designated as T-14A, T-15ZT and T-17s, produced spherical or ellipsoidal endospores in a terminal bulging sporangium. The strains required B for the growth and can tolerate more than 450 mM B. These also tolerated up to 7.0% (w/v) NaCl in the presence of 50 mM B in agar medium but grew optimally without NaCl. The temperature range for growth was 16–37°C (optimal of 30°C), whereas the pH range was 6.5–9.0 (optimal of 7.5–8.5). The DNA G + C content was 41.1–42.2 mol% and the predominant cellular fatty acid was iso-C15:0. The major respiratory quinone system was detected as MK-7 and the diamino acid of the peptidoglycan was meso-diaminopimelic acid. Based on phenotypic and chemotaxonomic characteristics, phylogenetic analysis of 16S rRNA gene sequences data and DNA–DNA re-association values, we concluded that the three strains belong to a novel species of the genus Bacillus, the type strain of which is T-15ZT and for which we proposed the name, B. boroniphilus sp. nov. Ferns are good ecological indicators of arsenic accumulation in contaminated abandoned mines. The rhizomes of ferns were analyzed for arsenic and other heavy metals using inductively coupled plasma-mass spectrometry. In addition, we examined the ribulose-bisphosphate carboxylase (rbcL) gene isolated from the indigenous species of ferns at the pithead, tailings and soil sites in the vicinity of the Myoungbong abandoned mine, Republic of Korea. A search of the GenBank database revealed that the predicted amino acid sequence of this gene was homologous with 99% of Pteridium aquilinum As-2 (EU476184), 99% of Deparia petersenii As-6 (EU476188), 97% of Deparia lobatocrenata As-8 (EU476190), and 98% of Deparia conilii As-7 (EU476189). High concentrations of heavy metals (arsenic, 17.6 mg/kg in rhizomes and 2690 mg/kg in soil; copper, 17.1 mg/kg in rhizomes and 40.7 mg/kg in soil; cadmium, 2.9 mg/kg in rhizomes and 12.2 mg/kg in soil; lead, 41.9 mg/kg in rhizomes and 135 mg/kg in soil; zinc, 27.0 mg/kg in rhizomes and 172 mg/kg in soil) were found in P. aquilinum As-2 (EU476184) from the Myoungbong mine area, which may play an important role as ecological indicators of the contaminated areas.  相似文献
