Effects of salinity and phosphate on ion distribution in lupin leaflets   总被引:1,自引:0,他引:1  
Lupin ( Lupinus luteus L. cv. Weiko III) were grown in nutrient solution over a range of inorganic phosphate (Pi) concentrations, with or without 50 m M NaCl. Plants with high Pi (2 m M ) and salt showed progressive leaf necrosis and had higher concentrations of total phosphate than plants grown with high Pi alone. Most of the extra total phosphate in salt treated plants was in the Pi form. Pi supply did not influence Na+, K+ or Cl concentrations in epidermal vacuoles or mesophyll cells. However, epidermal vacuoles accumulated more monovalent cations (Na+ and K+) than Cl, and in vacuoles of plants grown with 0.1 m M Pi additional Pi was accumulated, possibly to maintain charge balance. Plants grown with 2 m M Pi did not accumulate additional Pi in epidermal vacuoles, but showed higher phosphorus levels in cell walls. It is suggested that at moderate phosphorus concentrations Pi plays a role in epidermal osmotic adjustment, possibly explaining the beneficial role of additional phosphorus on salt stressed plants. At high Pi supply with salt, Pi does not contribute to osmotic adjustment and instead accumulates in cell walls. However, the cause of leaf damage under conditions of high phosphorus supply and salinity is still not entirely clear.  相似文献

Applying silicon in the form of metasilicic acid (H4SiO3) or silicic acid (H4SiO3) to Bradyrhizobium -infected, hydroponically grown cowpea seedlings resulted in a significant ( P 0.05) increase in the number of nodules, nodule dry matter, and nitrogen fixed on a per plant basis. Total dry matter of plants increased with silicon supply, and the differences were significant ( P 0.05) at the higher silicon concentrations. Cowpea plants cultured in sand were also assessed for their response to silicic acid. Nodule number and nodule mass increased with silicon supply to sand cultured plants, though nitrogen fixation was unaltered. Although silicon is not essential for growth of cowpea, it is important for nodule formation and nodule functioning in hydroponically grown plants. Consequently, data collected and conclusions drawn from earlier glasshouse experiments, which have excluded silicon from nutrient solutions, may be flawed. Future studies on nodulation and nitrogen fixation using legumes in liquid culture must therefore include silicon as a nutrient element.  相似文献

Ozone-tolerant Bel B and ozone-sensitive Bel W3 tobacco cultivars were subjected to acute ozone fumigation (200 p.p.b. for 3 h) and the subcellular localization of H2O2 was then studied. H2O2 accumulated on the cell walls and plasma membrane of both cultivars but the accumulation pattern differed greatly. H2O2 production was high in both cultivars immediately after fumigation, but, in the tolerant Bel B cultivar, after 7 h was only detected in some spongy cells adjacent to epidermal cells. Instead, in the sensitive Bel W3 cultivar, accumulation was still abundant in the cell walls of palisade, spongy and epidermal cells at this time. Significant changes in apoplastic ascorbate pool were noted in both cultivars in the first hours after fumigation. As the reduced ascorbate content remained unchanged, the marked increase in total ascorbate must have originated from the striking increase in dehydroascorbate, particularly in the ozone-sensitive Bel W3. Exposure of plants to ozone resulted in a marked transient increase in both free and conjugated salicylic acid (SA) as well as an increase in the activity of benzoic acid 2-hydroxylase which catalyses SA biosynthesis. SA induction differed greatly in the two cultivars, in that: (1) SA accumulation was far greater in the ozone-sensitive Bel W3 cv. and (2) the maximum SA peak was delayed in Bel W3 and observed only 7 h after fumigation ended. These results suggest that a high SA content, as documented in the ozone-sensitive Bel W3 cultivar, could trigger the production of ROS with subsequent SA-mediated cell-death.  相似文献

Chloride transport in sheath and blade tissue and the cellular distribution of Cl- were investigated in an attempt to determine the physiological basis of the preferential accumulation of Cl- in sheaths of salt-stressed sorghum ( Sorghum bicolor L.). Import and export of 36Cl- in leaf sheaths and blades of intact sorghum were followed over a 2 week period. X-ray microanalysis of frozen-hydrated bulk tissue samples was used to determine the accumulation of Cl- and other elements in the vacuoles of sheath and blade cells.
Sheath tissue accumulated Cl- despite a relatively high Cl- turnover rate. Chloride was shown to accumulate in most cell types of the sheath, particularly in adaxial epidermal cells. After an initial increase in the concentration of Cl-, blade tissue regulated Cl- levels within certain limits. Chloride levels in blades were greater in the abaxial and adaxial epidermal cells than in other cell types. The epidermal cells of blades accumulated Cl- to approximately the same concentration as sheath epidermal cells. The Cl- concentration in the photosynthetically active mesophyll and bundle sheath cells, however, remained low.
Thus, the partitioning of Cl- previously observed in the leaves of salinized sorghum apparently results from the ability of bundle sheath and mesophyll cells to maintain concentrations of Cl- at lower levels than do epidermal cells. In addition, the relatively large sheath parenchyma cells tend to serve as reservoirs for the storage of Cl-.  相似文献

Phytoliths are amorphous silicon dioxide (SiO2.nH2O) inclusions abundant in leaves, in-ternodes and glumes in members of Poaceae. They may occur as inclusions filling the entire lumen of the silica cells, bulliform cells and trichomes or may be part of the outer epidermal cell walls. Since phytoliths are resistant to fungal or animal digestive juices, a large quantity of phytoliths accumulate in the soil where grasses grow. Compared with the pollen grains of grasses which tend to be uniform, phytoliths vary in sue and morphology and can be of value in identification at different taxonomic levels and in the dating of past vegetation. The size and shape of phytoliths of about 100 species of grasses from Tamil Nadu, India, have been determined. Silica bodies were observed either after isolation or in cleared leaf blades. Size and shape of phytoliths were determined under a microscope or from micrographs of the specimens. Size and shape can be used to assign the phytoliths to their respective subfamilies and to distinguish some of the grasses at the generic level. Drawings of silica cells and an identification key are provided for 80 species.  相似文献

SUMMARY. Methods for determination of low levels of biogenic silica (0.2–0.4 mg SiO2) in aqueous samples after digestion with three wetalkaline extraction procedures compared favourably in both precision of replicates and recovery of silica utilized by diatoms in budgeted cultures. Leaching samples with 0.2 M NaOH for 10–15 min at 100°C was the least time consuming procedure. Also interference from silicate minerals was lower for this method than leaching with either 0.5 or 5% Na2CO3 for 2 h at 85°C. The use of filters to concentrate samples enables detection of low levels of biogenic silica with colorimetric procedures. Polycarbonate filters are recommended in preference to cellulose acetate or polyvinyl chloride filters for sample collection. Time-course experiments are recommended for establishing digestion times and determining the presence of mineral silicate interference. Wet-alkaline digestion methods are recommended for routine analysis of biogenic silica in suspended matter in preference to infra-red analysis, alkaline fusion and hydrofluoric acid/nitric acid methods.  相似文献

Structural characteristics of xyloglucan are constant in the pericarp cell walls of kiwifruit ( Actinidia deliciosa ) throughout fruit enlargement and maturation. Most of the xyloglucan (XG) persists in the cell walls of ripe kiwifruit. XG from the pericarp tissues of 36-h ethylene-treated kiwifruit was extracted as hemicellulose II (HC-II) with 4.28 M KOH containing 0.02% NaBH4, and purified using iodine precipitation and subsequent anion-exchange chromatography. This purifying protocol increased XG purity from 50 mol% in HC-II fraction to 62 mol% in the purified XG powder. The molar ratio of glucose: xylose: galactose: fucose in the purified XG was 10: 6.9: 2.1: 0.3. Gel permeation chromatography indicated that purified XG had an average molecular-mass of 161 KDa, a value that exceeds the 95 KDa Mr determined for total polymeric sugars. Sugar linkage analysis confirmed the lack of fucose in the kiwifruit XG, but a small amount of arabinoxylan and low Mr glucomannan remained associated with this fraction.  相似文献

通过盆栽试验,研究了UV-B增强下不同施硅量和硅肥种类对水稻生长及甲烷(CH4)排放的影响.UV-B辐射设2个水平,即对照(自然光,A)和增强20%(E);硅肥设4个水平,即Si0(不施硅,0 kg SiO2·hm-2)、Si1(硅酸钠,100 kg SiO2·hm-2)、Si2(硅酸钠,200 kg SiO2·hm-2)和Si3(钢渣硅肥,200 kg SiO2·hm-2).结果表明: 施硅能缓解UV-B增强对水稻生长的抑制作用,使分蘖数、叶绿素含量、地上部和地下部生物量增加.施硅对水稻生长的促进作用随施硅量(硅酸钠)的增加而增加,钢渣硅肥的促进作用大于硅酸钠.UV-B增强可提高稻田CH4的排放通量和累积排放量,施硅显著降低CH4的排放通量和累积排放量,且CH4排放随施硅量的增加而减少.在施硅量相同的情况下,钢渣硅肥的减排效果优于硅酸钠.表明在水稻生产中,施用钢渣硅肥不仅能实现废弃物利用,而且可有效降低UV-B增强下CH4的排放量.  相似文献

ABSTRACT. Using hydrofluoric acid, scanning electron microscope-assisted X-ray microanalysis, and energy-filtered transmission electron microscopy, we present the first definite proof of biomineralized silicon [(SiO2)] n in a ciliophoran protist, Maryna umbrellata , a common inhabitant of ephemeral pools. In the trophic specimen, the amorphic silicon (glass) granules are accumulated in the anterior half of the body. When entering the dormant stage, most glass granules are excreted to form the surface cover of the globular resting cyst. Most likely, the silicon granules are synthesized in vesicles of the Golgi apparatus. First, nanospheres with a size of 20–40 nm are formed in a fibrous matrix; they grow to be spongious complexes, eventually becoming amorphous glass granules with an average size of 819 nm × 630 nm. In the transmission electron microscope, the silicon granules show the characteristic fracture pattern of glass known from many other silicon-bearing organisms. A literature survey suggests that silicon is very rare in ciliates. The fine structure and genesis of silicon granules in M. umbrellata are very similar to those of other organisms, including vascular plants and animals, indicating a common mechanism. Light perception and protection against mechanical stress and predators might be functions of the silicon granules in M. umbrellata . The palaeontological significance of glass cysts in ciliates is also discussed.  相似文献

GA1, GA8, GA17, GA19, GA20 and GA29 were identified by combined gas chromatography-mass spectrometgry (GC-MS) in immature seeds and pericarp of Lycopersicon esculentum Mill. (tomato). Higher levels of these GAs were present in the seeds than in the pericarp; seeds in addition contained GA15, GA24, GA25, and GA44. Fruits of the Lycopersicon pimpinellifolium Mill. mutant I were smaller and contained lower GA1 concentrations, but higher GA20 concentrations, than those of mutants III and IV. In contrast, differences in fruit size in L. esculentum due to position on the truss did not correlate with GA1 concentration in either the pericarp or seeds.  相似文献

Cell walls extracted from cv. Rutgers, 7711 (ripening inhibited), and nor (non-ripening) tomato ( Lycopersicon eseulentum Mill.) pericarp tissue at various stages of post-maturation development have been separated into four distinct fractions and their carbohydrate composition characterized. The amount of ionically-associated, chelator-soluble (CDTA, cyclohexanediaminetetraacetic acid) uronic acid in 'Rutgers' fruit cell walls remained constant during ripening, whereas the amount of residual pectin, which was extracted with cold alkali (Na2CO3) and was apparently covalently bound, decreased. These changes did not occur in rin and nor mutant fruit at a similar chronological age. The galactose content in pectic polysaccharide preparations extracted from tomato cell walls with CDTA and Na2,CO3, decreased by 65% during ripening. A similar but diminished decrease also occurred in rin and nor fruit. A non-cellulosic polysaccharide(s) was present in walls which resisted extraction with Na-acetate/CDTA, Na2CO3, and 4 M KOH. In 'Rutgers' fruit, the content of galactose in this polysaccharide(s) decreased 44% during ripening, whereas little or no significant change was observed in rin or nor mutant fruit.  相似文献

Abstract: Isolates of Paxillus involutus (Batsch) Fr. collected from different hosts and environmental conditions were screened for their ability to form ectomycorrhizal symbiosis with hybrid poplar P. × canescens ( = Populus tremula L. × P. alba) in vitro. The ability to form ectomycorrhiza varied between the fungal isolates and was not correlated with the growth rate of the fungi on agar-based medium. The isolate MAJ, which was capable of mycorrhiza synthesis under axenic conditions, and the incompetent isolate NAU were characterized morphologically and anatomically. MAJ formed a typical hyphal mantle and a Hartig net, whereas NAU was not able to penetrate the host cell walls and caused thickenings of the outer cell walls of the host. MAJ, but not NAU, displayed strong H2O2 accumulation in the outer hyphal mantle. Increases in H2O2 in the outer epidermal walls and adjacent hyphae of the incompetent isolate were moderate. No increases of H2O2 in response to the mycobionts were found inside roots. Suggested functions of H2O2 production in the outer hyphal mantle of the compatible interaction are: growth regulation of the host's roots, defence against other invading microbes, or increasing plant-innate immunity. The system established here for P. × canescens compatible and incompetent fungal associations will be useful to take advantage of genomic information now available for poplar to study tree-fungal interactions at the molecular and physiological level.  相似文献

Arthrobacter nicotianae KCC B35 isolated from blue-green mats densely covering oil sediments along the Arabian Gulf coast grew well on C10 to C40 n -alkanes as sole sources of carbon and energy. Growth on C20 to C40 alkanes was even better than on C10 to C18 alkanes. Biomass samples incubated for 6 h with n -octacosane (C28) or n -nonacosane (C29) accumulated these compounds as the predominant constituent alkanes of the cell hydrocarbon fractions. The even chain hexadecane C16 and the odd chain pentadecane C15 were the second dominant constituent alkanes in C28 and C29 incubated cells, respectively. n -Hexadecane-incubated cells accumulated in their lipids higher proportions of C16-fatty acids than control cells not incubated with hydrocarbons. On the other hand, C28 and C29-incubated cells did not contain any fatty acids with the equivalent chain lengths, but the fatty acid patterns of the cell lipids suggest that there should have been mid-chain oxidation of these very long chain alkanes. This activity qualifies A. nicotianae KCC B35 to be used in cocktails for bioremediating environments polluted with heavy oil sediments.  相似文献

Pectins from persimmon ( Diospyros kaki L.) fruit pericarp were sequentially extracted with 0. 05 M trans -1,2-diaminocyclohexane-N,N, N', N'-tetraacetic acid (CDTA), 0. 05 M Na2CO3 (1°C) and Na2CO3 (20°C) and the carbohydrate composition and metabolism during development determined. Young persimmon fruits contained a large proportion of pectins, 46% by dry weight, that decreased to 20% with ripening. This decrease occurred in the CDTA and Na2CO3 (1°C) fractions, mainly composed of uronic acids, and represents a net loss of uronic acids, arabinose and galactose. The amount of non-cellulosic neutral sugars was especially high in the Na2CO3 (20°C) fraction. The loss of pectins was also accompanied by a depolymerisation of the polysaccharides extracted in the three pectic fractions. However, none of these changes can be attributed to the action of polygalacturonase activity. Proteins were extracted from the pericarp tissue, but endopolygalacturonase (EC 3. 2. 1. 15) activity, determined as a decrease in viscosity of polygalacturonic acid, was not observed in the extract. Determination of exopolygalacturonase (EC 3. 2. 1. 67) activity by measuring the release of reducing groups from polygalacturonic acid was also negative. The results presented indicate that polygalacturonase is not responsible for the metabolism of pectins during persimmon fruit ripening.  相似文献

The extreme dwarf d x tomato ( Lycopersicon esculentum Mill.) mutant has very short internodes which were found to contain shorter and fewer epidermal cells. The leaves are highly abnormal. The mutant showed a substantial stem growth response to GA3, without approaching normal stature or morphology. The active gibberellin GA1 and its precursors GA19 and GA20 were identified by coupled gas chromatography-mass spectrometry (GC/MS) in d x shoots. Quantitative GC/MS revealed that GA20 accumulated to far higher levels than normal in stems and leaves of the mutant.  相似文献

White fluorescent light (5 W m−2) inhibited subhook growth in derooted Alaska pea cuttings. In the inner tissue of the subhook, it inhibited the increase in osmotic potential during 18 h incubation. In the epidermis, on the other hand, light did not affect the osmotic potential. Light increased the minimum-stress relaxation time (T0) of the inner tissue cell walls, but did not change T0 of the epidermal cell wall. Light decreased tissue stress determined by the split test and the ability of the inner tissue to extend by water absorption. The short-term light effect on subhook growth. T0, and the tissue stress almost disappeared when pea cuttings were transferred to darkness. These facts suggest that light changes the mechanical properties of the cell wall in the inner tissue of shoots, and decreases tissue stress, which is considered to be the driving force of shoot growth.  相似文献

Rice (Oryza sativa) takes up arsenite mainly through the silicic acid transport pathway. Understanding the uptake and sequestration of arsenic (As) into the rice plant is important for developing strategies to reduce As concentration in rice grain. In this study, the cellular and subcellular distributions of As and silicon (Si) in rice roots were investigated using high-pressure freezing, high-resolution secondary ion mass spectrometry, and transmission electron microscopy. Rice plants, both the lsi2 mutant lacking the Si/arsenite efflux transporter Lsi2 and its wild-type cultivar, with or without an iron plaque, were treated with arsenate or arsenite. The formation of iron plaque on the root surface resulted in strong accumulation of As and phosphorous on the epidermis. The lsi2 mutant showed stronger As accumulation in the endodermal vacuoles, where the Lsi2 transporter is located in the plasma membranes, than the wild-type line. As also accumulated in the vacuoles of some xylem parenchyma cells and in some pericycle cells, particularly in the wild-type mature root zone. Vacuolar accumulation of As is associated with sulfur, suggesting that As may be stored as arsenite-phytochelatin complexes. Si was localized in the cell walls of the endodermal cells with little apparent effect of the Lsi2 mutation on its distribution. This study reveals the vacuolar sequestration of As in rice roots and contrasting patterns of As and Si subcellular localization, despite both being transported across the plasma membranes by the same transporters.  相似文献
