The nuclear ribosomal DNA internal transcribed spacer (ITS-1 and ITS-2) sequences were determined for 10 of 12 Japanese non-digitate Laminaria species, Kjell-maniella gyrata (Kjellman) Miyabe, Costaria costata (Turner) Saunders, Alaria praelonga Kjellman and Chorda filum (L.) Stackhouse collected at Hokkaido. Phyloge-netic analyses (maximum parsimony and distance matrix) of these sequences, including published data for L. sac-charina (L.) Lamouroux from Canada, showed strong nucleotide conservation among these species of Laminaria, but two phylogenetically distinct species groups were recognized. A L. japonica group encompassing L. yapon/ca Areschoug, L. religiosa Miyabe, L. ochotensis Miyabe, L. diabolica Miyabe, L. longipedalis Okamura, L. angustata Kjellman and L. longissima Miyabe; and a L. saccharina group including L. coriacea Miyabe, L. sac-charina, L. cichorioides Miyabe and L. yendoana Miyabe. As to other laminarialean genera, Kjellmaniella gyrata was most closely related to the genus Laminaria, being related to the second Laminaria species group based on both parsimony and distant tree values.  相似文献

Laminaria population variables and understory community composition were monitored just prior to, and for two summers following, a September 1980 experimental total harvest ofL. longicruris De la Pylaie andL. digitata (L.) Lamouroux within two plots in Lobster Bay, Nova Scotia. Both plots, distinguished mainly by depth, were characterized by highLaminaria standing crop and no recent history of extensive sea urchin grazing. Within the shallower plot (2–3 m below MSL), recovery could not be assessed thoroughly due to ice damage, but within the deeper plot (3–4 m below MSL),L. longicruris regrew cropped biomass and attained maximum observed abundance within one year. BothLaminaria species required two years to mature to pre-harvest population characteristics. Survivorship of 0–1 year old and mature populations of both species was generally low (0–67 % per year); however, the higher maximum life expectancy ofL. digitata (> 4 years vs 2 years) can result in that species persisting to the disadvantage ofL. longicruris. Analysis of understory community composition for both harvested plots and their adjacent controls weakly distinguished the harvested plots one summer after harvesting from all others. It is doubtful the distinction is attributable to harvesting and in neither site was there evidence of a critical change in the understory community. Management implications for the commercial harvest of the brown algaLaminaria are discussed.  相似文献

The salinity tolerances of Baltic and Atlantic populations ofFucus vesiculosus andChorda filum have been measured using net photosynthesis as an index of tissue damage. AtlanticFucus proved to have a broader salinity tolerance than AtlanticChorda, a result which is consistent with others published on the tolerances of intertidal and sublittoral marine species. The optimum salinity for all Atlantic plants was 11 or 34‰, but that of all Baltic plants was 6‰. BalticFucus andChorda were different in breadth of tolerance, in spite of the fact that they inhabit the same sublittoral habitat. This difference is interpreted in relation to their respective life-forms,Fucus being perennial andChorda annual.Fucus is therefore present as a macrothallus at all times of year, including the critical low-salinity period of the spring ice-melt.Chorda evades damage by existing as a resistent microthallus at this time. It is concluded that the distinctive character of Baltic marine algae deserves nomenclatural recognition at some level below that of the species. The rank of subspecies would appear the most appropriate of those listed in the Code, but none of those available is able adequately to express the patterns of variation now being reported. Eight strains isolated from birds, reptiles, and water constitute a new DNA hybridization group that is 37–58% related toEdwardsiella tarda and less than 10% related to other species of Enterobacteriaceae (SI nuclease method). This homogeneous group (78–100% relatedness within the group) constitutes a new species that is namedEdwardsiella hoshinae sp. nov. (type strain, CIP 78.56 ATCC 33379). Strains of this species produce acid fromd-mannitol, sucrose,d-trehalose, and salicin, and give a positive malonate test. Seven other strains that produced acid fromd-mannitol and sucrose (but not fromd-trehalose and salicin) and were malonate negative were found to belong to theEdwardsiella tarda DNA hybridization group. The base composition of the DNAs ofE. tarda andE. hoshinae is 55–58 mol% G+C.  相似文献

Sporophytes of Laminaria digitata (L.) Lamour. were assayed for their content of accumulated iodine, which ranged from 0.4% of dry weight in adult plants up to 4.7% for young plantlets. Sporophyte tissue from Laminaria saccharina (L.) Lamour. and L. digitata took up iodide according to Michaelis-Menten kinetics. Hydrogen peroxide and various substances known to interfere with oxidative metabolism were shown to either inhibit or enhance the uptake of iodide, confirming that apoplastic oxidations play a key role in iodide uptake in Laminaria. Consistently, iodide uptake was triggered in L. saccharina protoplasts by incubation in the presence of hydrogen peroxide. Similarly, the uptake of iodide was enhanced in L. digitata gametophytes by addition of haloperoxidase, suggesting that this enzyme catalyses the oxidation of iodide by hydrogen peroxide and plays a key role in iodine uptake. Oxidative stress resulted in a marked efflux of the intracellular iodine. In both influx and efflux experiments, a marked proportion (10–30%) of the tracer was not accounted for, indicating volatilisation of iodine. Kelps are important providers and constituents of marine ecological niches, the coastal kelp forests. Kelp species have differing distribution ranges, but mainly thrive in temperate and arctic regions. Although the principal factors determining biogeographic distribution ranges are known, genomics could provide additional answers to this question. We sequenced DNA from two Laminaria species with contrasting distribution ranges, Laminaria digitata and Laminaria solidungula. Laminaria digitata is found in the Northern Atlantic with a southern boundary in Brittany (France) or Massachusetts (USA) and a northern boundary in the Arctic, whereas L. solidungula is endemic to the Arctic only. From the raw reads of DNA, we reconstructed both chloroplast genomes and annotated them. A concatenated data set of all available brown algae chloroplast sequences was used for the calculation of a robust phylogeny, and sequence variations were analyzed. The two Laminaria chloroplast genomes are collinear to previously analyzed kelp chloroplast genomes with important exceptions. Rearrangements at the inverted repeat regions led to the pseudogenization of ycf37 in L. solidungula, a gene possibly required under high light conditions. This defunct gene might be one of the reasons why the habitat range of L. solidungula is restricted to lowlight sublittoral sites in the Arctic. The inheritance pattern of single nucleotide polymorphisms suggests incomplete lineage sorting of chloroplast genomes in kelp species. Our analysis of kelp chloroplast genomes shows that not only evolutionary information could be gleaned from sequence data. Concomitantly, those sequences can also tell us something about the ecological conditions which are required for species well‐being.  相似文献

This paper describes the macroflora and macrofauna associated with two bull kelp species,Laminaria hyperborea andL. digitata, at the island of Helgoland, North Sea. During a study period of seven months (March–September 1987), 29 macroflora species and 125 macrofauna species were found. The dominant taxonomic groups were Polychaeta (25 species), Bryozoa (17), Amphipoda (14), Hydrozoa (10) and Ascidiae (8). The species maximum was in July. In general,L. hyperborea was preferred as a substrate for settlement toL. digitata. Composition of the communities associated with kelp changed during the season according to exposure to wave action, and according to location on the kelp thallus. The rhizoid community of both kelps bore more species at exposed locations. Wave-exposedL. digitata lacked obvious faunal settlement on both phylloid and cauloid. Phylloid and cauloid ofL. hyperborea were chosen as an attractive substrate at both sheltered and wave-exposed locations, showing an association of encrusting bryozoan and hydrozoan colonies.  相似文献

The taxonomy and evolutionary relationships of species in the genus Laminaria are poorly understood. Previous studies have demonstrated significant plasticity of morphological characters used to describe taxa, and interfertility has been reported among putative species. We analyzed nuclear ribosomal DNA (rDNA) sequence variation in eight species of Laminaria (L. agardhii Kjell., L. digitata (Huds.) Lamour., L. groenlandica Rosenv. [sensu Druehl 1968], L. longicruris De la Pyl., L. longipes Bory, L. saccharina (L.) Lamour., L. setchellii Silva, and L. yezoensis Miyabe) to elucidate evolutionary relationships in this genus. Restriction maps were constructed using a small subunit rDNA probe from Costaria costata (Turn.) Saunders, an rDNA repeat from the nematode Caenorhabditis elegans, and 11 hexameric restriction endonucleases in an annealing analysis of genomic DNA. Laminaria rDNA restriction maps were compared to each other and to that of the outgroup taxon, C. costata. rDNA restriction maps of Laminaria species and C. costata were similar. Restriction fragment length polymorphisms mapped to both the coding regions and the nontranscribed spacer of rDNA. Laminaria species were distinguished with this method. The restriction maps of L. agardhii, L. saccharina, and L. longicruris were identical, supporting a previous hypothesis that these species are conspecific. Comparison of restriction maps of Laminaria species suggested that the generic subdivision of Sections Simplices and Digitatae may be invalid.  相似文献

A cytological investigation of British members of the orderLaminariales has revealed the presence of a very large X-chromosomein dividing nuclei of female gametophytes and young sporophytesof Saccorhiza polyschides. This chromosome pairs with a smallY-chromosome at meiosis. A chromosome which is conspicuouslylarger than the others has also been detected in the femalegametophytes (and young sporophytes) of Laminaria spp., Chordafilum, and Alaria esculenta. There is some evidence that thisalso pairs with a smaller chromosome. Growth of young sporophytes of the brown algae Laminaria digitata, L. saccharina and L. solidungula from Spitsbergen were measured in the laboratory after being exposed for 21 days to either photosynthetically active radiation (PAR=P) or to full light spectrum (PAR + UV-A + UV-B=PAB) using of cutoff glass filters. The plants were grown at 8±2°C and 16 h light : 8 h dark cycles with 6 h additional ultraviolet radiation (UVR) exposure in the middle of the light period. Growth was measured every 10 min using growth chambers with online video measuring technique. Tissue morphology and absorption spectra were measured in untreated young sporophytes while chlorophyll (Chl) a content and DNA damage were measured in treated thalli at the end of the experiment. In all species, growth rates were significantly higher in sporophytes exposed to P alone compared to sporophytes exposed to PAB. Tissue DNA damage is dependent on thallus thickness and absorption spectra characteristics of pigments and UV-absorbing compounds. In sporophytes exposed to UVR, energy demands for repair of DNA damage and synthesis of UV-absorbing compounds for protection effectively diverts photosynthate at the expense of growth. Photosynthetic pigment was not significantly different between treatments suggesting a capacity for acclimation to moderate UVR fluence. The general growth pattern in sporophytes exposed to P alone showed an increasing growth rate from the onset of light (0500–0900 hours) to a peak at the middle of the light phase (0900–1500 hours), a decline towards the end of the light phase (1500–2100 hours) and a minimum “low” growth in the dark (2100–0500 hours) relative to growth during the entire light phase. Under PAB, different growth patterns were observed such as growth compensation at night in L. digitata, delayed growth recovery in L. saccharina and minimal but continuous growth in L. solidungula. Growth as an integrative parameter of all physiological processes showed that the effect of UVR is correlated to the depth distribution of these species.  相似文献

The growth of threeFrankia strains (HFPCcl3, ORS021001, ORS020607), isolated from nodules ofCasuarina, was unaffected by 5–15 μmol/L quercetin. Re-isolation ofFrankia strain HFPCc13 fromCasuarina root nodules was carried out with the addition of 15 μmol/L quercetin to three different media. Using constant heat sap flow sensors, xylem water fluxes in ten tree species and two liana species were monitored for 5–10 days during the beginning of the wet season in May, 1993. For a subset of the trees, a branch was also monitored at the top of the crown for 5 days. Xylem flux (J S) was related diurnally in all plants to vapor pressure deficit (D) measured within the upper-third of the canopy, and to incoming shortwave radiation R S above the canopy. Cross-correlation analysis was used to estimate time lags between diurnal patterns of J S and D or R S, and between J S in stems and branches. The maximum correlation coefficient from cross-correlation of J S with R S (range=0.57–0.92) was often higher than the maximum of J S with D (range=0.43–0.89), indicating that diurnal J S was more dependent on R S than D. Time lags (lag corresponding to maximum correlation) of J S at stem-base with D was shorter (0–45 min) than with radiation (5–115 min), highly variable within a species, and uncorrelated to the height or exposure of tree crowns or liana in the canopy. On a stand level, not accounting for the diel lag between stem sap flux and canopy flux resulted in errors in estimated canopy transpiration of up to 30%. We investigated patterns of genetic structure in two sister kelp species to explore how distribution width along the shore, zonation, latitudinal distribution and historical factors contribute to contrasting patterns of genetic diversity. We implemented a hierarchical sampling scheme to compare patterns of genetic diversity and structure in these two kelp species co‐distributed along the coasts of Brittany (France) using a total of 12 microsatellites, nine for Laminaria hyperborea and 11 for Laminaria digitata, of which eight amplified in both species. The genetic diversity and connectivity of L. hyperborea populations were greater than those of L. digitata populations in accordance with the larger cross‐shore distribution width along the coast and the greater depth occupied by L. hyperborea populations in contrast to L. digitata populations. In addition, marginal populations showed reduced genetic diversity and connectivity, which erased isolation‐by‐distance patterns in both species. As L. digitata encounters its southern range limit in southern Brittany (SBr) while L. hyperborea extends down to mid‐Portugal, it was possible to distinguish the effect of habitat continuity from range edge effects. We found that L. digitata did not harbour high regional diversity at its southern edge, as expected in a typical rear edge, suggesting that refuges from the last glacial maximum for L. digitata were probably not located in SBr, but most likely further north. For both species, the highest levels of genetic diversity were found in the Iroise Sea and Morlaix Bay, the two regions in which they are being currently harvested. Preserving genetic diversity of these two foundation species in these areas should, thus, be a priority for the management of this resource in Brittany.  相似文献

Azospirillum isolates were obtained from rhizosphere soil and roots of three cactaceae species growing under arid conditions. All Azospirillum isolates from rhizosphere and roots ofStenocereus pruinosus andStenocereus stellatus were identified asA. brasilense; isolates of surface-sterilized roots fromOpuntia ficus-indica were bothA. brasilense andA. lipoferum. Azospirilla per g of fresh root in the three species ranged from 70×103 to 11×103. The most active strains in terms of C2H2 reduction (25–49.6 nmol/h·ml) and indoleacetic acid (IAA) production (36.5–77 μg/ml) were those identified asA. brasilense and isolated from Stenocereus roots.A. lipoferum isolated from Opuntia roots produced low amounts of IAA (6.5–17.5 μg/ml) and low C2H2-reduction activity (17.8–21.2 nmol/h·ml).  相似文献

Laminaria longissima and Zaohoucheng No.1 (a commercial variety selected from Laminaria japonica) differ to a certain extent in their morphological characteristics and biological habits. It was assumed that varieties bred through their hybridization should exhibit high yield potential and tolerate relatively high seawater temperatures. Female gametophyte clones isolated from L. longissima were crossed with male clones isolated from Zaohoucheng No.1. Laminaria variety 90-1 was obtained after gametophyte crossing, continuous self-crossing and selection. This variety was genetically homozygous; the indices of variation of blade length, width and thickness of the final two selection cycles were 7–8%; i.e., not different significantly. Variety 90-1 grew faster, lost less tissue and had higher yield potential than two widely used commercial varieties of L. japonica (all commercial varieties currently used in China originate from this latter species). The blade of variety 90-1 increased 3.71 cm day−1 on average during the whole period of cultivation, almost two-fold that of two controls, and growth was maintained even when seawater temperature was higher than 18°C–3°C higher than the temperature tolerated by other Laminaria varieties. Variety 90-1 increased yield by more than 70% over two controls and also synthesized desirable amounts of iodine, mannitol and algin. In blade length, variety 90-1 was more similar to L. longissima than to L. japonica, but more similar to L. japonica in blade width and thickness. Since the adoption of variety 90-1 in 1999, its culturing area has increased each year to reach its current area of 7,000 ha, i.e., almost one-third of the total cultivation acreage of Laminaria in China. Breeding of variety 90-1 has demonstrated that it is feasible to develop elite Laminaria varieties by crossing gametophytes from different Laminaria species in combination with successive self-crossing and selection.  相似文献

Pigment composition, fluorescence parameters, and oxygen evolution of the deep water Laminaria abyssalis Oliveira and of the shallow water L. digitata Lamoroux were determined in response to high irradiances. This was performed in the presence and absence of an inhibitor of violaxanthin de‐epoxidase (dithiothreitol) or an inhibitor of the chloroplast‐encoded protein synthesis (chloramphenicol). Photochemical quenching in L. digitata was almost 3‐fold that seen in L. abyssalis, whereas both nonphotochemical quenching and PSII photochemical yield were doubled. Laminaria digitata possessed a xanthophyll‐cycle pool nearly double that of L. abyssalis. After photoinhibitory treatment, L. digitata displayed substantial violaxanthin de‐epoxidation, whereas in L. abyssalis de‐epoxidation only took place in limited amounts. Both species were able to fully recover their epoxidation status after transfer back to dim light. Overnight incubation with dithiothreitol fully blocked de‐epoxidation in both species, and both displayed similar fluorescence properties. Chloramphenicol caused no change in their fluorescence parameters. With high light treatment, L. abyssalis was completely and irreversibly inhibited both in the presence and absence of inhibitors, whereas L. digitata showed 60% inhibition of its photosynthetic activity and full recovery in the absence of inhibitors. In the presence of dithiothreitol, L. digitata did not recover to the preillumination conditions and chloramphenicol delayed the recovery of the oxygen evolution activity. We suggest that the xanthophyll cycle is the main mechanism of photoprotection of these Laminaria species and that the higher susceptibility of L. abyssalis to photoinhibition may be due to its limited de‐epoxidation capacity and reduced xanthophyll‐cycle pool size.  相似文献

The nameLeclercia adecarboxylata is proposed for a group of the family Enterobacteriacae previously known asEscherichia adecarboxylata. Leclercia adecarboxylata can be phenotypically differentiated from all other species of Enterobacteriaceae. The members of this species are positive for motility, indole production, methyl red, growth in the presence of KCN, malonate, beta-galactosidase, beta-xylosidase, esculin hydrolysis, gas production fromd-glucose, and acid production fromd-cellobiose,d-lactose, melibiose,l-rhamnose, adonitol,d-arabitol, dulcitol, and salicin; the strains were negative for Voges-Proskauer, citrate (Simmons), H2S (Kligler), lysine and ornithine decarboxylases, arginine dihydrolase, phenylalanine deaminase, gelatinase, DNase, Tween-80 hydrolysis, and acid production from myoinositol and alpha-methyl-d-glucoside. Fermentation ofd-raffinose,d-sucrose, andd-sorbitol is variable with strains. DNA relatedness of 11 strains ofL. adecarboxylata to three strains including the type strain of this species averaged 80% in reactions at 65°C. DNA relatedness to other species in Enterobacteriaceae was 2%–32%, indicating that this species was placed in a new genusLeclercia gen. nov. The type strain ofL. adecarboxylata is ATCC 23216.  相似文献

Propolis ethanolic extract (PEE) at 3 and 4 g/L and ultragriseofulvin (UG) at 0.75 and 1 g/L reduced the percentage of conidia germination in twoAspergillus flavus isolates. PEE at 1–4 g/L decreased the mycelial dry mass ofA. flavus isolates by 11–80%, and aflatoxin B1 production by 34–100%. UG concentrations of 0.25–1 g/L reduced the growth and aflatoxin B1 production of the isolates by 16–88 and 48–98%, respectively. Any increase in PEE and UG concentration was accompanied by a clear decrease in the per cent conidia germination, growth and aflatoxin B1 production. A method to extract high-quality DNA from potato tubers was developed and tested on 3 wild potato species (Solanum raphanifolium, S. megistracrolobum, S. bukasovii) and on the tetraploid B3 bred population, (population number 393228, derived fromS. tuberosum subsp.tuberosum). The average yield of extracted DNA varied from 10–30 μg of DNA per gram of processed tissue. The DNA was pure and suitable for ligation-mediated polymerase chain reaction (LM-PCR) amplification, producing clear, distinctive, and reproducible banding patterns in polyacrylamide gels.  相似文献
