血管生成素样蛋白3(angiopoietin—like protein 3,angptl 3)是血管生成素和血管生成素样蛋白家族成员,因其与该家族成员有共同的结构特征而得名,即氨基端与分泌相关的信号肽,介导同源寡聚体形成的螺旋样结构域,一段短的连接肽和羧基端介导配体活性的纤维蛋白原样结构域。Angptl 3是一种分泌蛋白,其mRNA主要在人、鼠的肝脏表达。目前研究表明angptl 3的生物学效应表现在两方面:调节血管生成和脂质代谢。它对血管生成的诱导作用较弱,而对脂质代谢的调节作用较显著,因而在心血管病领域受到极大关注。本文将就angptl 3的研究进展做一综述。  相似文献

目的探讨RNA干扰血管生成素样蛋白7 (Angptl7)基因对血管紧张素Ⅱ(AngⅡ)诱导的血管平滑肌细胞(VSMC)炎症因子的影响及其作用机制。 方法体外培养人VSMC,分为常规F12K培养基培养(对照)和1 μg/mL AngII培养24 h。VSMC用AngⅡ(1 μg/mL)处理24 h后,采用siRNA-Angptl7和阴性对照siRNA-NC在Lipofectamine 2000介导下转染VSMC。RT-qPCR检测mRNA表达水平;Griess反应测定一氧化氮(NO)含量;蛋白免疫印记法检测相关蛋白的改变;酶联免疫吸附法检测VSMC中炎症因子肿瘤坏死因子α(TNF-α)、白细胞介素-1β (IL-1β)和IL-6水平。多组间比较采用单因素方差分析,组间两两比较采用LSD-t检验,两组间比较采用独立样本t检验。 结果与对照比较,1 μg/mL AngⅡ处理可促进VSMC中Angptl7 mRNA (0.97±0.06比3.05±0.21)和蛋白表达(1.01±0.12比1.61±0.14),亦可促进VSMC中IL-1β[(45.21±8.10)比(126.17±11.77) pg/mL]、IL-6[(50.50±7.51)比(108.50±9.51)pg/mL]和TNF-α的表达[(60.77±9.58)比(185.67±17.35)pg/mL],差异有统计学意义(P均< 0.01)。与对照和转染siRNA-NC相比,转染siRNA-Angptl7下调Angptl7蛋白表达(0.99±0.12,0.98±0.12比0.44±0.14,P < 0.01)。与AngⅡ干预组相比,siRNA-Angptl7降低AngⅡ介导的VSMC炎症反应相关蛋白TNF-α、IL-6和IL-1β的表达,核因子κB (NF-κB)/诱导型一氧化氮合酶(iNOS)/环氧化酶2 (COX-2)信号通路相关蛋白NF-κB、iNOS和COX-2表达及NO含量亦降低,差异有统计学意义(P均< 0.01)。与siRNA-NC相比,siRNA-Angptl7组AngⅡ诱导的VSMC炎症反应相关蛋白TNF-α (0.99±0.13比0.51±0.12)、IL-6 (1.00±0.12比0.38±0.05)和IL-1β的表达(0.99±0.14比0.48±0.11),NF-κB (1.00±0.10比0.42±0.08)、iNOS (1.02±0.12比0.42±0.10)和COX-2表达(1.00±0.11比0.52±0.12)均降低,NO含量[(54.78±2.76)比(18.08±3.61)μmol/L]亦降低,差异有统计学意义(P均< 0.01)。 结论AngⅡ可通过Angptl7促进VSMC炎症反应,下调Angptl7蛋白表达可以抑制VSMC的炎症反应,其作用机制可能与抑制NF-κB/iNOS-COX-2信号通路有关。  相似文献

利用半定量RT-PCR和免疫组化的方法同时从mRNA水平和蛋白质水平对血管生成素样蛋白2在不同病理阶段的2型糖尿病肾病模型小鼠--db/db小鼠肾脏中的表达情况进行了较为系统的分析.结果发现:a.在糖尿病前的db/db小鼠(4周龄的db/db小鼠),血管生成素样蛋白2与作为正常对照的db/m小鼠相比,差异不是很大,随着肥胖的加剧,高血糖、蛋白尿的出现,血管生成素样蛋白2在db/db小鼠肾脏中的表达无论从mRNA水平还是从蛋白质水平均显著升高.b.从免疫组化的分析结果来看,血管生成素样蛋白2主要分布于小鼠肾脏的肾小球部分,主要是沿毛细血管袢呈线性分布,其位置与足细胞的位置重叠,足细胞是小鼠肾脏中血管生成素样蛋白2的主要分泌细胞.c.小鼠肾脏血管生成素样蛋白2的表达水平似乎还与鼠龄相关:虽然变化幅度不是很大,但在周龄较大的小鼠(如20周龄以上),其表达水平相对较高.上述工作不仅印证了先前对2型糖尿病肾病患者肾小球基因表达谱的分析结果,更加明确了血管生成素样蛋白2与糖尿病肾病的相关性,同时揭示了血管生成素样蛋白2在正常小鼠和糖尿病肾病小鼠肾脏中的表达、分布和变化规律,有利于进一步揭示血管生成素样蛋白2的功能及其在糖尿病肾病发生、发展过程中的可能作用,探讨糖尿病肾病的分子机制.  相似文献

通过RT-PCR从人外周血白细胞中钓取血管生成素(angiogenin,Ang)cDNA,构建诱饵蛋白载体pAS-2-1-Ang,对其自身转录激活活性进行鉴定后,通过酵母双杂交系统筛选人肝细胞cDNA文库,获得两个双阳性克隆.序列分析和同源检索表明所获候选蛋白分别为人上皮细胞素和λ晶体蛋白.构建Ang及候选蛋白标签融合表达载体并共转染COS-7细胞,利用免疫共沉淀和蛋白质印迹方法在哺乳动物细胞中验证了Ang与候选蛋白间的相互作用.为阐明Ang促血管新生的分子机制创造了条件.  相似文献

通过RT PCR的方法从人外周血白细胞扩增血管生成素 (Ang)cDNA .在计算机分子结构模建的基础上 ,通过柔性连接臂构建了Ang与Gfp融合基因 ,并在大肠杆菌DH5α中实现了高效表达 .重组蛋白占菌体总蛋白的 32 %.融合蛋白经初步纯化后 ,在紫外线激发下可见明显的绿色荧光 ,同时能够显著地促进鸡胚尿囊膜毛细血管的新生 ,而且所获融合蛋白在体外具有促进人脐静脉血管内皮细胞增殖的作用 .这种双功能融合蛋白的表达为阐明Ang的核转位过程奠定了基础 ,同时为阐明血管新生的分子机制创造了条件  相似文献

目的探讨血管内皮生长因子(VEGF)和血管生成素(angiopoietin,Ang)在胃癌的表达及其与肿瘤血管生成和临床病理因素的关系。方法采用免疫组化SP法检测84例胃癌和30例癌旁正常组织中VEGF、Ang-1、Ang-2的表达,应用CD34抗体标记微血管内皮细胞,计数微血管密度(MVD),结合临床病理资料进行分析。结果胃癌组织VEGF、Ang-2阳性表达率、MVD值明显高于癌旁正常组织(P(0.05)。VEGF表达与肿瘤大小、侵袭深度、临床分期、淋巴结转移有关(P(0.05),而与患者年龄、性别、组织学类型和分化程度无关,其阳性组的Ang-2阳性表达率、MVD值明显高于阴性组,VEGF的表达与Ang-2、MVD呈正相关。胃癌组织Ang-2表达与肿瘤大小、侵袭深度、淋巴结转移有关(P(0.05),与MVD呈正相关。胃癌Ang-1表达略低于对照组,但无统计学差异(P(0.05),Ang-1的表达与肿瘤侵袭深度和MVD值呈负相关。结论胃癌中VEGF、Ang-2蛋白的过度表达以及Ang-1蛋白的低表达可能在肿瘤血管生成和肿瘤浸润、转移中起重要作用。  相似文献

钙粘着蛋白(Cadherins)作为细胞粘着分子中的一个大家族,其成员多达几十种,它们在形态发生(包括细胞的聚集、迁移和分化等)过程中具有极其重要的作用。本文就钙粘着蛋白结构、分布及其功能的研究进展作了较为全面的概述,同时简要讨论了钙粘着蛋白的信号调节机制等。目前,有关钙粘着蛋白的研究趋势已从单纯强调新的分子的发现转变为更为深入、细致地研究它们的结构,它们与配体、受体和反受体的相互作用,信号传导及其调节机制,以及它们在复杂的生命活动中的生物学功能。对于它们的研究正方兴未艾。  相似文献

Recent experiments show that membrane ATPases are capable of absorbing free energy from an applied oscillating electric field and converting it to chemical bond energy of ATP or chemical potential energy of concentration gradients. Presumably these enzymes would also respond to endogenous transmembrane electric fields of similar intensity and waveform. A mechanism is proposed in which energy coupling is achieved via Coulombic interaction of an electric field and the conformational equilibria of an ATPase. Analysis indicates that only an oscillating or fluctuating electric field can be used by an enzyme to drive a chemical reaction away from equilibrium.In vivo, the stationary transmembrane potential of a cell must be modulated to become locally oscillatory if it is to derive energy and signal transduction processes.  相似文献

基因组选择(genomic selection, GS)是畜禽经济性状遗传改良的重要方法。随着高密度SNP芯片和二代测序价格的下降,GS技术越来越多被应用于奶牛、猪、鸡等农业动物育种中。然而,降低全基因组SNP分型成本、提高基因组育种值(genomic estimated breeding value,GEBV)估计准确性仍然是GS研究的主要难题。本文从全基因组SNP分型策略和GEBV估计模型两个方面进行了综述,并对目前GS技术在主要畜禽品种中的应用现状进行了介绍,以期为GS在农业动物育种中的深入开展提供借鉴和参考。  相似文献

14-3-3蛋白是高度保守的、所有真核生物细胞中都普遍存在的、在大多数生物物种中由一个基因家族编码的一类蛋白调控家族。它几乎参与生命体所有的生理反应过程,人们在各种组织细胞中发现了各种不同的14-3-3蛋白。作为与磷酸丝氨酸/苏氨酸结合的第一信号分子,14-3-3蛋白在细胞的信号转导中起着至关重要的作用,尤其是它直接参与调节蛋白激酶和蛋白磷酸化酶的活性,被称为蛋白质与蛋白质相互作用的”桥梁蛋白”;它可以与转录因子结合形成复合体,调节相关基因的表达。一些研究表明,14-3-3蛋白调控机制的紊乱可以直接导致疾病的发生,在临床上14-3-3蛋白常常可以作为诊断的标志物。  相似文献

We report the initial characterization of an Arabidopsis thaliana cDNA (atdrg1), a member of a new class of GTP-binding proteins (G-proteins) in plants. The predicted ATDRG1 protein contains all five structural motifs characteristic of the G-protein superfamily. Apart from these motifs, the amino acid sequence differs substantially from all known G-proteins except for a recently discovered new family named developmentally regulated G-proteins (DRGs). Sequences closely related to atdrg1 are found in species as distant as human (80% amino acid conservation), Drosophila (74%), yeast (77%) and Caenorhabditis elegans (77%). The remarkable evolutionary conservation of these proteins suggests an important, but as yet unclear role. Phylogenetic analysis of the available homologous sequences strongly suggests a diphyletic origin of the eukaryotic DRG proteins. Northern analysis shows high levels of atdrg1 mRNA in all Arabidopsis tissues studied, and homologues of atdrg1 are present throughout the plant kingdom. In situ hybridization reveals that atdrg1 is highly expressed in actively growing tissues and reproductive organs. Southern analysis indicates the presence of either one or two copies of atdrg1 in the Arabidopsis genome. Immunolocalization studies show that the protein is present in cytoplasmic vesicles found mainly in actively growing tissues suggesting a putative role for ATDRG1 in either the regulation of vesicle transport or the regulation of enzymes involved in storage protein processing.  相似文献

Molecules can enter the nucleus by passive diffusion or active transport mechanisms, depending on their size. Small molecules up to size of 50-60 kDa or less than 10 nm in diameter can diffuse passively through the nuclear pore complex (NPC), while most proteins are transported by energy driven transport mechanisms. Active transport of viral proteins is mediated by nuclear localization signals (NLS), which were first identified in Simian Virus 40 large T antigen and had subsequently been identified in a large number of viral proteins. Usually they contain short stretches of lysine or arginine residues. These signals are recognized by the importin super-family (importin α and β) proteins that mediate the transport across the nuclear envelope through Ran-GTP. In contrast, only one class of the leucine-rich nuclear export signal (NES) on viral proteins is known at present. Chromosome region maintenance 1 (CRM1) protein mediates nuclear export of hundreds of viral proteins through the recognition of the leucine-rich NES.  相似文献

Dopamine receptors are a subclass of the super family of G protein-coupled receptors, that transduce their effects by coupling to specific G proteins. Within the dopamine receptor family, the adenylyl cyclase stimulatory receptors include the D1 and D5 subtypes. The D1 and D5 dopamine receptors are genetically distinct, sharing >80% sequence homology within the highly conserved seven transmembrane spanning domains, but displaying only 50% overall homology at the amino acid level. When expressed in transfected GH4C1 rat pituitary cells, both D1 and D5 receptors stimulate adenylyl cyclase and have identical affinities toward dopaminergic agonists and antagonists. In order to analyze specific signaling pathways mediated by activation of either D1 or D5 receptors, we have identified the G proteins that are coupled to these receptors. Through functional analyses and competition binding studies, and from immunoprecipitation techniques, using antisera against the various α subunits of G proteins, we have established that both D1 and D5 receptors couple to Gsα. In addition, D1 receptors are also coupled to Goα. Since Goα has been implicated in the regulation of Ca2+, K+, and Na+ channels, this finding would suggest that D1 receptors can mediate the functional activity of these ion channels. There is also evidence to indicate that D5 receptors couple to Gzα, a novel G protein abundantly expressed in neurons. Thus, despite similar pharmacological properties, such differential coupling of D1 and D5 receptors to G proteins other than Gsα, indicates that dopamine can transduce varied signaling responses upon the simultaneous stimulation of both these receptors.  相似文献

Calmodulin labeled with125I or34S has been used to screen expression libraries to isolate cDNAs encoding calmodulin-binding proteins (CBPs) from several eukaryotic systems. The use of radiolabeled calmodulin has, however, several disadvantages. We have developed a nonradiactive method to isolate cDNAs for CBPs using biotinylated calmodulin. Screening of a cDNA library in an expression vector with biotinylated calmodulin resulted in the isolation of cDNAs encoding CBPs. Avidin and biotin blocking steps, prior to incubation of the filters with biotinylated calmodulin, are found to be essential to eliminate the cDNAs that code for biotin-containing polypeptides. The cDNA clones isolated using this nonradioactive method bound calmodulin in a calcium-dependent manner. The binding of biotinylated calmodulin to these clones was completely abolished by ethylene glycolbis(\-aminoethylether)-N,N′-tetraacetic acid (EGTA), a calcium chelator. Furthermore, the isolated cDNAs were confirmed by probing the clones with35S-labeled calmodulin. All the isolated clones bound to radiolabeled calmodulin in the presence of calcium but not in the presence of EGTA. The method described here is simple, fast, and does not involve preparation and handing of radiolabeled calmodulin. All the materials used in this method are commercially available; hence, this procedure should be widely applicable to isolate cDNAs encoding CBPs from any eukaryotic organism.  相似文献
