The herbicide endothall (7-oxabicyclo 2.2.1 heptane-2, 3-dicarboxylic acid) was applied to cotton ( Gossypium hirsutum L. cv. Acala SJ-1) cotyledon discs. Treatment with 10−4 M endothall for 24 h resulted in injury which was expressed by increased leakage of electrolytes, development of necrotic areas, increased level of polyphenols and tissue browning. We examined whether treatments which decrease chilling injury would also decrease injury caused by the herbicide. Tissue from seedlings grown at 28°C was more sensitive to endothall than that from seedlings grown at 15°C. Pretreatment with 10−5 M abscisic acid greatly decreased the leakage of electrolytes, necrotic areas, and tissue browning caused by endothall. Similar pretreatment did not prevent the increase of polyphenols caused by the herbicide. The treatment with abscisic acid was more effective in protection against the herbicide injury when applied several hours prior to the herbicidal treatment. This time requirement indicates that the mechanism by which abscisic acid induces resistance to the herbicide depends, at least partially, on active metabolism. We suggest that the increased resistance to herbicide stress by abscisic acid is another example of a common resistance mechanism to various stresses in which abscisic acid is involved.  相似文献

Cucumber ( Cucumis sativus L. cv. Victory) seedlings were exposed to chilling at 5°C and endogenous levels of polyamines and 1-aminocyclopropane-1-carboxylic acid (ACC) were measured after chilling and after warming at 20°C. The level of spermidine was higher in the chilled seedlings than in the non-chilled seedlings. Treatment with a plant bioregulator, (2RS,3RS)-1-(4-cholorophenyl)-4,4-dimethyl-2-(1,2,4-triazol-1-yl)pentan-3-ol (paclobutrazol), reduced the chilling injury and the levels of spermidine in the chilled seedlings. The levels of ACC and production of ethylene showed sharp increases after warming following exposure to chilling. These increases were suppressed by the application of aminooxyacetic acid (AOA). However, AOA treatment did not reduce chilling injury or affect the levels of polyamines in the tissue. These data indicate that the increase in ACC and ethylene is a response of the tissue to the chilling exposure and is not a cause of the injury. The data also suggest that the syntheses of polyamines and ethylene are not competitive with each other even under chilling stress conditions.  相似文献

Maize seedlings ( Zea mays L. cv. DK 246) grown for 1–4 days in the presence of abscisic acid (ABA) or triadimefon (a fungicide) demonstrated an enhanced ability to withstand the effects of a 3-h sub-lethal (40°C) or lethal (45°C) heat shock. Both the ABA and triadimefon treatments were applied solely to the roots of seedlings; however, the ability to withstand a heat shock was induced in both the root and the shoot. The level of protection provided by these agents was dependent upon the time that plants were exposed to them; prolonged exposure reduced tolerance to subsequent stress.  相似文献

Our previous results indicated that 3-d-old dark-grown chilling-sensitive maize (Zea mays L.) seedlings did not survive 7 d of 4[deg]C chilling stress, but 69% of them survived similar stress when the seedlings were either preexposed to 14[deg]C for 3 d or pretreated with 0.1 mM H2O2 for 4 h at 27[deg]C (T.K. Prasad, M.D. Anderson, B.A. Martin, C.R. Stewart [1994] Plant Cell 6: 65-74) or 1 mM abscisic acid (ABA) for 24 h at 27[deg]C (M.D. Anderson, T.K. Prasad, B.A. Martin, C.R. Stewart [1994] Plant Physiol 105: 331-339). We discovered that chilling imposed oxidative stress on the seedlings. Since H2O2 accumulated during the periods of both acclimation and nonacclimation, we concluded that H2O2 had dual effects at low temperature: (a) During acclimation, its early transient accumulation signals the induction of antioxidant enzymes such as catalase 3 and peroxidase to scavenge H2O2; and (b) at 4[deg]C in nonacclimated seedlings, it accumulates to damaging levels in the tissues because of low levels of these and perhaps other antioxidant enzymes. Three-day-old seedlings pretreated with H2O2 (a mild oxidative stress) or ABA showed induced chilling tolerance. In the present study, we investigated whether mitochondria are a target for chilling-induced oxidative stress and, if so, what differences do acclimation, H2O2, or ABA make to protect mitochondria from irreversible chilling injury. The results indicated that chilling, in general, impairs respiratory activity, the cytochrome pathway of electron transport, and ATPase activity regardless of the treatment. In pretreated seedlings, the activities of catalase 3 and peroxidase in the mitochondria increased severalfold compared with control and nonacclimated seedlings. The increases in these antioxidant enzymes imply that mitochondria are under oxidative stress and such increases could initiate a protective mechanism in the mitochondria. Mitochondrial respiration is partially cyanide resistant during chilling stress and also after the 1st d of recovery. Upon further recovery over 3 d, in contrast to nonacclimated seedlings, the mitochondria of acclimation-, H2O2-, and ABA-treated seedlings showed the following recovery features. (a) The mitochondrial respiration changed from a cyanide-resistant to a cyanide-sensitive cytochrome pathway, (b) cytochrome oxidase activity recovered to control levels, (c) the ability of mitochondria to generate ATP was regained, and (d) the antioxidant enzyme activities remained at or above control levels. Based on these results, we conclude that chilling impairs mitochondrial function and that chilling-induced oxidative stress seems to be a factor, at least in part, for causing possible irreversible damage to the mitochondrial membrance components. Acclimation, H2O2, and ABA provide a protective mechanism by inducing antioxidant enzymes to protect mitochondria from irreversible oxidative damage that is absent in nonacclimated seedlings. Therefore, we conclude that the ability of the seedlings to recover from chilling injury is, at least in part, due to the ability of the mitochondria to resume normal function.  相似文献

The effects of paclobutrazol on the leaf membrane lipid composition of seedlings of cucumber ( Cucumis sativus L. cv. Victory) subjected to chilling temperatures were assessed. At a non-injurious temperature (12.5°C), there was no difference in the polar lipid fatty acid composition or in the glycolipid, phospholipid or free sterol content of leaves from treated vs untreated seedlings, regardless of whether paclobutrazol was administered 1 or 7 days prior to analysis. In the latter case (7 days pretreatment), there were clear effects of the bioregulator on plant growth and morphology as well as on leaf chlorophyll content. At an injurious chilling temperature (5°C), desaturation of leaf polar lipid fatty acids was markedly reduced in both treated and untreated seedlings. Chilling at 5°C resulted in losses of fresh weight and membrane lipids in leaves of both groups of plants. These losses were either reversible or irreversible, depending upon the duration of chilling and of pretreatment with paclobutrazol. Seedlings pretreated with 10 μg ml−1 paclobutrazol generally sustained less chilling injury than untreated controls, as judged by the extent of wilting, necrosis and desiccation. Seedlings of tomato (Lycopersicon esculentum, Mill.) varied diurnally in their sensitivity to chilling temperatures. If chilled near the end of the dark period when they were most sensitive, the time taken to kill half of the seedlings was approximately 3 days, whereas in samples taken 4 hours after the onset of dark, a period of 6 days of chilling was required. Sensitivity dropped rapidly after the onset of the light period. This rhythm was exogenously controlled by the diurnal changes in light, rather than in the temperature. The susceptibility of predawn seedlings could be reduced by exposure to light, by water stress, or by abscisic acid applied to the leaves. However, the subsequent changes in sensitivity to chilling did not correlate with stomatal aperture. Six other chilling-sensitive species showed similar diurnal changes in their chilling sensitivity.  相似文献

Four inbred maize lines differing in chilling tolerance were used to study changes in water status and abscisic acid (ABA) levels before, during and after a chilling period. Seedlings were raised in fertilized soil at 24/22°C (day/night), 70% relative humidity. and a 12-h photoperiod with 200 μmol m−2 s−1 from fluorescent tubes. At an age of 2 weeks the plants were conditioned at 14/12°C for 4 days and then chilled for 5 days at 5/3°C. The other conditions (relative humidity, quantum flux, photoperiod) were unchanged. After the chilling period the plants were transferred to the original conditions for recovery. The third leaves were used to study changes in leaf necrosis, ion efflux, transpiration, water status and ABA accumulation. Pronounced differences in chilling tolerance between the 4 lines as estimated by necrotic leaf areas, ion efflux and whole plant survival were observed. Conditioning significantly increased tolerance against chilling at 5/3°C in all genotypes. The genotypes with low chilling tolerance had lower water and osmotic potentials than the more tolerant genotypes during a chilling period at 5/3°C. These differences were related to higher transpiration rates and lower diffusive resistance values of the more susceptible lines. During chilling stress at 5/3°C ABA levels were quadrupled. Only a small rise was measurable during conditioning at 14/12°C. However, conditioning enhanced the rise of ABA during subsequent chilling. ABA accumulation in the two lines with a higher chilling tolerance was triggered at a higher leaf water potential and reached higher levels than in the less tolerant lines. We conclude that chilling tolerance in maize is related to the ability for fast and pronounced formation of ABA as a protective agent against chilling injury.  相似文献

Low temperature is one of the major factors limiting pepper (Capsicum annuum L.) production during winter and early spring in non-tropical regions. Application of exogenous abscisic acid (ABA) effectively alleviates the symptoms of chilling injury, such as wilting and formation of necrotic lesions on pepper leaves; however, the underlying molecular mechanism is not understood. The aim of this study was to identify genes that are differentially up- or downregulated in ABA-pretreated hot pepper seedlings incubated at 6°C for 48 h, using a suppression subtractive hybridization (SSH) method. A total of 235 high-quality ESTs were isolated, clustered and assembled into a collection of 73 unigenes including 18 contigs and 55 singletons. A total of 37 unigenes (50.68%) showed similarities to genes with known functions in the non-redundant database; the other 36 unigenes (49.32%) showed low similarities or unknown functions. Gene ontology analysis revealed that the 37 unigenes could be classified into nine functional categories. The expression profiles of 18 selected genes were analyzed using quantitative RT-PCR; the expression levels of 10 of these genes were at least two-fold higher in the ABA-pretreated seedlings under chilling stress than water-pretreated (control) plants under chilling stress. In contrast, the other eight genes were downregulated in ABA-pretreated seedlings under chilling stress, with expression levels that were one-third or less of the levels observed in control seedlings under chilling stress. These results suggest that ABA can positively and negatively regulate genes in pepper plants under chilling stress.  相似文献

Cotton (Gossypium hirsutum L.) intact seedlings and isolatedcotyledonary discs were exposed to chilling (4?C) under humidconditions which prevented dehydration. The damage resultingfrom chilling was estimated by means of electrolyte leakageand survival in whole seedlings and by the electrolyte leakageand necrotic areas in isolated cotyledonary discs. Also, theeffect of chilling on membrane phospholipids and cellular reducedglutathione was determined. Within the first two and three daysof chilling, there was a marked reduction in the reduced glutathioneand membrane phospholipid levels without electrolyte loss andnecrosis. This reduction was completely prevented by pretreatmentwith abscisic acid. Prolonging the chilling period resultedin decreased survival in whole seedlings and in progressiveincrease in electrolyte leakage and necrosis in isolated cotyledonarydiscs. Pretreatment with abscisic acid prior to chilling almostcompletely prevented this chilling injury when exposure to 4?Cwas less than 5 days. Abstract: Seedlings of Zea mays L. were grown at optimal (25 °C) and suboptimal (15 °C) temperature and then exposed to severe chilling temperature (6 °C) at their growth light intensity (450 ìmol quanta m−2 s−1) for 4 d. Photosynthetic parameters, hydrogen peroxide, antioxidant contents, and activity of scavenging enzymes were investigated before, during, and after chilling stress. This stress caused a stronger reduction in photosynthetic activity, maximum quantum efficiency of photosystem II primary photochemistry ( F v/ F m), and catalase activity in plants which had been grown at 25 °C rather than at 15 °C. Maize plants grown at suboptimal temperature de-epoxidized their xanthophyll cycle pool to a greater extent and exhibited a faster recovery from chilling stress than plants which had not been acclimated to chilling. Antioxidant content, activity of scavenging enzymes, with the exception of catalase, hydrogen peroxide formation, and the size of the xanthophyll cycle pool were hardly affected by chilling stress. However, chilling induced a temporary increase in the glutathione content and triggered the synthesis of á-tocopherol during the phase of recovery at 25 °C. The results indicate that leaves respond to chilling stress by down-regulation of photosystem II accompanied by de-epoxidation of the xanthophyll cycle pool, probably to prevent enhanced formation of superoxide radicals at photosystem I and, consequently, other reactive oxygen species.  相似文献

Inhibition of glutathione synthesis reduces chilling tolerance in maize   总被引:8,自引:0,他引:8  
 The role of glutathione (GSH) in protecting plants from chilling injury was analyzed in seedlings of a chilling-tolerant maize (Zea mays L.) genotype using buthionine sulfoximine (BSO), a specific inhibitor of γ-glutamylcysteine (γEC) synthetase, the first enzyme of GSH synthesis. At 25 °C, 1 mM BSO significantly increased cysteine and reduced GSH content and GSH reductase (GR: EC 1.6.4.2) activity, but interestingly affected neither fresh weight nor dry weight nor relative injury. Application of BSO up to 1 mM during chilling at 5 °C reduced the fresh and dry weights of shoots and roots and increased relative injury from 10 to almost 40%. Buthionine sulfoximine also induced a decrease in GR activity of 90 and 40% in roots and shoots, respectively. Addition of GSH or γEC together with BSO to the nutrient solution protected the seedlings from the BSO effect by increasing the levels of GSH and GR activity in roots and shoots. During chilling, the level of abscisic acid increased both in controls and BSO-treated seedlings and decreased after chilling in roots and shoots of the controls and in the roots of BSO-treated seedlings, but increased in their shoots. Taken together, our results show that BSO did not reduce chilling tolerance of the maize genotype analyzed by inhibiting abscisic acid accumulation but by establishing a low level of GSH, which also induced a decrease in GR activity. Many developmental and physiological changes, including alterations of enzyme activities, occur in plants under low temperature stress. In this study the total ribonuclease activity was determined in crude extracts from root tips of soybean seedlings germinated at 25 °C, subjected to chilling conditions (10°C) and recovered at optimal temperature (25°C). Measurements of RNase activity were performed every 24 hours starting from the third to the 10-th day of growth. We found that chilling caused a considerable increase in ribonuclease activity (in comparison with the control), with an activity peak on the fourth day of the cold treatment. The enzyme activity in root extracts of the plants recovered after cold stress decreased along with the time of recovery. No differences were found in approximate molecular weight (35 kDa) and pH optimum (6.0) for ribonucleases extracted from control and chilled soybean roots.  相似文献

Endogenous abscisic acid levels and induced heat shock proteins were measured in tissue exposed for 6 hours to temperatures that reduced their subsequent chilling sensitivity. One-centimeter discs excised from fully expanded cotyledons of 11-day-old seedlings of cucumber (Cucumis sativus L., cv Poinsett 76) were exposed to 12.5 or 37°C for 6 hours followed by 4 days at 2.5 or 12.5°C. Ion leakage, a qualitative indicator of chilling injury, increased after 2 to 3 day exposure to 2.5°C, but not to 12.5°C, a nonchilling temperature. Exposure to 37°C before chilling significantly reduced the rate of ion leakage by about 60% compared to tissue exposed to 12.5°C before chilling, but slightly increased leakage compared to tissue exposed to 12.5 or 37°C and held at the nonchilling temperature of 12.5°C. There was no relationship between abscisic acid content following exposure to 12.5 or 37°C and chilling tolerance. Five heat shock proteins, with apparent molecular mass of 25, 38, 50, 70, and 80 kilodaltons, were induced by exposure to 37 or 42°C for 6 hours, and their appearance coincided with increased chilling resistance. Heat shock treatments reduced the synthesis of three proteins with apparent molecular mass of 14, 17, and 43 kilodaltons. Induction of heat shock proteins could be a possible cause of reduced chilling injury in tissue exposed to 37 or 42°C.  相似文献

Low temperature during germination and early seedling growth is one of the most significant limiting factors in the productivity of plants. Tomato seedling germination is sensitive to chilling stress. Gamma-aminobutyric acid (GABA), as a non-protein amino acid, involved in various stress tolerances in plants. In this study, 5-day old tomato seedlings were exposed to chilling stress (2 ± 0.05 °C for 48 h) and then the effects of 0, 100, 250, 500 and 750 μmolL−1 concentrations of GABA on electrolyte leakage, proline and malondialdehyde (MDA) content were investigated. The resultS showed that the antioxidant enzyme activity, electrolyte leakage, MDA and proline content were significantly reduced by GABA treatments. However under chilling stress seedlings treated with GABA exhibited significantly higher sugar and proline contents as compared to un-treated seedlings. These results suggest that GABA treatment protects tomato seedlings from chilling stress by enhancing some antioxidant enzymes activity and reducing MDA content which results in maintaining membrane integrity.  相似文献

To shed light on the early protein phosphorylation events involved in plant cell signaling in response to environmental stresses, we studied changes in the phosphorylation status of the Arabidopsis cell suspension proteome after short-term low temperature and abscisic acid (ABA) treatment. We used radioactive pulse-labeling of Arabidopsis cell suspension cultures and two-dimensional (2-D) gel electrophoresis to identify proteins that are differentially phosphorylated in response to these treatments. Changes in the phosphorylation levels of several proteins were detected in response to short-term (5 min or less) cold (4°C) and chilling (12°C) stress and ABA treatment, and we observed that some of these changes were common between these treatments. In addition, we used Pro-Q Diamond phosphoprotein gel stain to study the steady-state protein phosphorylation status under the same treatments. We demonstrated that Pro-Q Diamond effectively stained phosphorylated proteins, however, the overall Pro-Q Diamond 2-D gel staining pattern of proteins extracted from low-temperature and ABA-treated cells was not consistent with the gel patterns obtained by in vivo radioactive labeling of phosphoproteins. These in vivo pulsed-labeling experiments demonstrate that the Arabidopsis phosphoproteome is dynamic in response to short-term low temperature and ABA treatment, and thus represents a strategy for the identification of signaling proteins that could be utilized in the production of chilling or freeze tolerant crop varieties.  相似文献

The effect of LAB 173 711, a synthetic analogue of abscisic acid, has been evaluated on chilling-sensitive mung bean (Vigna radiata L. cv. Local V.) seedlings. Electrical conductivity was used for assessing the degree of chilling injury. Exposure of 8-day-old mung bean seedlings to 4°C for 35 h resulted in a 50% electrolyte leakage and induced irreversible chilling injury. The seedlings gained the best protection against chilling injury by pretreatment with LAB 173 711 (5 × 10–4 M) for 3 days. The protection effect could be sustained for 4 days. The LAB 173 711 pretreatment at 28°C did not cause a significant difference in the electrolyte leakage over the ambient temperature (28°C) control. Application of LAB 173 711 at 28°C reduced visible injury and the treated seedlings had higher ethylene production and respiration rate over the untreated control. LAB 173 711 helped maintain the integrity of the cell membrane and thus reduced the leakage of soluble sugar and amino acids. These combined effects led to a higher chilling tolerance in the mung bean seedlings.  相似文献
