 选用千粒重大小不同的小麦品种,研究了去除顶端两个小穗对两类品种(大粒品种和小粒品种)穗部性状、籽粒平均灌浆速率、单穗平均增重速率、植株光合速率及14C同化物运输分配的影响。试验结果表明,去除顶端两个小穗后,两类品种的籽粒平均灌浆速率和单穗平均增重速率(分别表征籽粒库容活性和穗粒库容活性)相应提高,穗粒重表现为补偿性增长。两类品种比较,小粒品种的增长幅度大于大粒品种。穗粒库容活性增强使得小粒品种灌浆中后期的植株光合速率提高,使两类品种分配到籽粒中的14C同化物比例增加。从而表明,无论是植株光合速率还是同化物的运转分配皆受库容活性的调控,调控方式和幅度因品种类型而不同。  相似文献

2-Carboxyarabinitol 1-phosphate (CAIP) is involved in the regulationof ribulose 1,5-bisphosphate carboxylase (rubisco) activityin many plants, but the biochemical pathway for its synthesisis unknown. In an attempt to induce synthesis of 14C-CAIP invivo, intact leaflets of Phaseolus vulgaris were pulse-labeledwith 14CO2 and chased with 12CO2 under conditions which resultin leaves accumulating unlabeled CAIP. Sugar-phosphates wereisolated from leaf extracts by anion exchange chromatographyand constituent metabolites were separated by 2D-thin-layerchromatography. No l4C-labeled CAIP was recovered from extractsprepared from leaves experiencing a range of exposure/chaseconditions, a range of leaf/plant ages, or from other speciesdiffering in their ability to accumulate unlabeled CAIP. Appropriatecontrol experiments indicated no loss of 14C-standard whichhad been added at the time of killing the leaf. The data suggestthat carboxyarabinitol 1-phosphate is not synthesized in vivoas some "misfire" catalysis by rubisco, and that the precursorto its synthesis is far "downstream" of CO2 fixation. Lehnherr, B. M?chler, F. and N?sberger, J. 1985. Effect of CO2concentration during growth on a CO2 concentrating mechanismin white clover as predicted from differential 14CO2/12CO2 uptake.-J. exp. Bot. 36: 1835-1841. White clover was grown at 20 and100 Pa p(CO2). The CO2 response of net photosynthesis and differentialuptake of 14CO2 and 12CO2 by leaves were measured at varioustemperatures and at various O2 and CO2 partial pressures andcompared with predictions from ribulose bisphosphate carboxylase/oxygenasekinetics. Discrepancies between the observed gas exchange characteristicsfor the leaves and those predicted from the enzyme kineticswere interpreted as being due to a CO2 concentrating mechanism.Plants grown at 20 Pa p(CO2) showed a higher affinity for CO2than plants grown at 100 Pa p(CO2) when measured at 10 ?C. Nodifference in affinity was found at 30 ?C. The postulated CO2concentrating effect was greater in plants grown at low CO2than in plants grown at high CO2 concentration and occurredonly at low temperature and low CO2 partial pressure. It issuggested that plants grown at the lower CO2 partial pressurehave a higher affinity for CO2 due to a more efficient CO2 concentratingsystem than plants grown at the higher CO2 partial pressure. 目的观察在进行14C-尿素呼气试验(UBT)时常规进行胃内酸化是否可提高14C-UBT试验诊断的准确性。方法250例因消化不良需要进行胃镜检查的门诊患者分为传统14C-UBT组和酸化14C-UBT组。传统14C-UBT组按常规方法进行试验,酸化14C-UBT组的检测过程与传统14C-UBT基本相同,唯一的差别是以0.1 mol/L的柠檬酸水200 m l(加少量甜味剂)送服14C尿素胶囊。比较2次UBT的14CO2呼出量、诊断的敏感性、特异性及准确性。结果(1)2组幽门螺杆菌(H.pylori)阳性的患者平均14CO2呼出量酸化组高于传统组,而2组H.pylori阴性的患者平均14CO2呼出量酸化组低于传统组,且统计学上差异均有显著性(P<0.05)。(2)传统14C-UBT组诊断的准确性、特异性、敏感性分别为92%、91%、92%;而酸化14C-UBT组分别为98.5%、100%、98%。2组间的诊断准确性及特异性差异有显著性(P<0.05);敏感性差异无显著性(P>0.05)。结论在14C-尿素呼气试验时常规进行胃内酸化可提高诊断的准确性。  相似文献

Labelled carbon dioxide was supplied for 22 hrs to a leaf of the leader or to the lateral shoot in two-year-old apple seedlings. The distribution of radioactive assimilates within the plant following this treatment was investigated by using radioautography. The transport of labelled assimilates from the young leaf of the leader was very meagre and affected only parts of the stem and the leaves situated in the close vicinity of the treated leaf. The14C-labelled assimilates from the mature leaf of the leader were transported in a considerable amount to the apex and to the other leaves of the leader. They were also found in an appreciable amount in the stem and the roots, as well as in some lateral shoots. After supplying14CO2 to the lateral shoot remarkable transport of labelled assimilates was observed. Radioactivity was detected in the tip and in the youngest leaves of the leader, as well as in the roots. Their path in the stem was studied by dissecting the plant and examining the cross section from each internode. This method revealed that the assimilates from the treated leaf or shoot were transported downward only on one side of the stem in a helical pattern. The lateral shoots situated on the radioactive side of the stem were also labelled, whereas those situated on the opposite (non-radioactive) side were not labelled.  相似文献

A simple closed system for the serial determination of 14CO2 in small volumes of fluid samples, is described. The device consists of commercially available scintillation vials and silicone tube seals. 14CO2 is selectively liberated by citric acid and absorbed in a scintillation vial by Hyamine. Experiments on the effect of dichloroacetate on pyruvate dehydrogenase activity in rat hindlimbs perfused with [1-14C]pyruvate demonstrate the applicability of the method.  相似文献

Decimal serial dilutions of eight common bacterial species were prepared, and the detection times were determined by measuring the (14)CO(2) metabolized from the (14)C-labeled glucose substrate. The detection time was proportional to the logarithm of the initial inoculum, and high sensitivity, down to 1 colonyforming unit, was demonstrated.  相似文献

High activity of phosphoenolpyruvate (PEP)-carboxykinase, orADP: oxalacetate (OAA) carboxy-lyase activity (a kind of EC4. 1. 1. 32) was discovered in enzyme extracts or partiallypurified preparations obtained from the brown algae, Eiseniabicyclis, Dictyota dichotoma, Spatoglossum pacificum; and Hizikiafusiformis. Enzyme activities were determined by measuring theradioactivity incorporated in the products of dark 14CO2-fixationand by spectrophotometric determinations. Except for the lowactivity of "malic enzyme" (EC 1. 1. 1.40), no activities ofother carboxylases, i.e. PEP-carboxylase, PEP-carboxytransphosphorylase,and pyruvate carboxylase could be detected in algal extractsprepared under various conditions. Malate dehydrogenase (EC1. 1. 1. 37), fumarase (EC 4. 2. 1. 2), and glutamic: oxalacetictransaminase (EC 2. 6. 1. 1) were also detected. The algal PEP-carboxykinase required ADP and Mn2+ for maximumactivity in the carboxylation reaction; and ATP and Mn2+, butnot GTP, for maximum activity in both the decarboxylation andOAA-14CO2-exchange reactions. The optimum pH of purified PEP-carboxykinase was in the regionof 7.0 to 7.3 in both the carboxylation and decarboxylationreactions, and its Km values for HCO3, PEP, and ADP were10 mM, 0.3 mM, and 0.07 mM, respectively, in the carboxylationreaction, and values for OAA and ATP were 0.05 mM and 0.4 mM,respectively, in the decarboxylation reaction. Furthermore,the decarboxylation reaction was markedly inhibited by 20 mMHCO3. Agricultural soils are important sources of atmospheric N2O and CO2. However, in boreal agro-ecosystems the contribution of the winter season to annual emissions of these gases has rarely been determined. In this study, soil N2O and CO2 fluxes were measured for 6 years in a corn-soybean-wheat rotation in northeast China to quantify the contribution of wintertime N2O and CO2 fluxes to annual emissions. The treatments were chemical fertilizer (NPK), chemical fertilizer plus composted pig manure (NPKOM), and control (Cont.). Mean soil N2O fluxes among all three treatments in the winter (November–March), when soil temperatures are below −7°C for extended periods, were 0.89–3.01 µg N m−2 h−1, and in between the growing season and winter (October and April), when freeze-thaw events occur, 1.73–5.48 µg N m−2 h−1. The cumulative N2O emissions were on average 0.27–1.39, 0.03–0.08 and 0.03–0.11 kg N2ON ha−1 during the growing season, October and April, and winter, respectively. The average contributions of winter N2O efflux to annual emissions were 6.3–12.1%. In all three seasons, the highest N2O emissions occurred in NPKOM, while NPK and Cont. emissions were similar. Cumulative CO2 emissions were 2.73–4.94, 0.13–0.20 and 0.07–0.11 Mg CO2-C ha−1 during growing season, October and April, and winter, respectively. The contribution of winter CO2 to total annual emissions was 2.0–2.4%. Our results indicate that in boreal agricultural systems in northeast China, CO2 and N2O emissions continue throughout the winter.  相似文献

The pattern of oxidative metabolism of pyruvate may be assessed by comparing the steady-state 14CO2 production from four isotopes in identical samples. The assay requires measuring the ratios of steady-state 14CO2 production from two isotope pairs, [2-14C]pyruvate:[3-14C]pyruvate and [1-14C]acetate:[2-14C]acetate. These ratios are defined as the "pyruvate 14CO2 ratio" and the "acetate 14CO2 ratio," respectively. If pyruvate is metabolized exclusively via pyruvate dehydrogenase (PDH), the two ratios will be identical. Alternatively, if any pyruvate enters the tricarboxylic acid (TCA) cycle via pyruvate carboxylation (PC), the pyruvate 14CO2 ratio will be less than the acetate 14CO2 ratio. If pyruvate enters the TCA cycle only through PC (with oxaloacetate and fumarate in equilibrium) the pyruvate 14CO2 ratio will approach a value of 1.0. An equation is presented for the quantitative evaluation of pyruvate oxidation by these two pathways. We have used this method to detect relative changes in the pattern of pyruvate metabolism in rat liver mitochondria produced by exposure to 1 mM octanoyl carnitine, a compound known to alter the PC:PDH activity ratio. The major advantages of the method are (i) that it provides a sensitive method for detecting pyruvate carboxylation at physiological pyruvate concentrations and (ii) that it provides a method for distinguishing between effects on pyruvate transport and effects on pyruvate oxidation.  相似文献

In a field study, we examined the relationship between vegetation, abiotic factors and the CO2 exchange dynamics of a cut-away peatland 20 years after production had ended. The main objective was to determine the effect of rewetting on the CO2 exchange dynamics, measured separately in Eriophorum vaginatum tussocks and intertussocks (almost non-vegetated surfaces) using closed-chamber techniques, one growing season before and three growing seasons after the rewetting treatment. Rewetting lowered total respiration (R TOT) and increased gross photosynthesis (P G), which resulted in a higher incorporation of CO2 into the system. The seasonal CO2 balance for the almost continuously submerged section of the rewetted site became positive 2 years after rewetting (9.1 g CO2-C m−2), and it was still higher in the 3rd year (64.5 g CO2-C m−2), i.e. the system accumulated carbon. In the driest section of the rewetted site the seasonal balance increased strongly, but the balance was still negative during the 3 years following rewetting with losses from the system of 44.1, 26.1, 38.3 g CO2-C m−2 in 1995, 1996 and 1997 respectively. At the control site seasonal balance during 1995–1997 varied between ecosystem C losses of 41.8 and 95.3 in an area with high Eriophorum cover and between 52.1 and 109.9 g CO2-C m−2 with lower cover. Simulation of a cut-away peatland with dense Eriophorum vegetation (Eriophorum cover 70%) showed that if the water level (WT) is low, the seasonal CO2 balance of the ecosystem can reach the compensation point of no net C change (P G = R TOT) only if weather conditions are favourable, but with a high WT the seasonal CO2 balance would be positive even under varying weather conditions. It can be concluded that with dense Eriophorum vegetation a restored cut-away peatland acts as a functional mire and becomes a sink for atmospheric CO2. A method for estimation of 14CO2 present in blood and tissular samples is described. It is basically based on the introduction of large amounts of a gas mixture (95% O2, 5% CO2) in the samples which serves to remove the CO2 label by gas dilution. The gas phase is later captured in scintillation vials containing an organic-soluble base that retains the carbon label. The results obtained by means of this methodology show much better recoveries for blood samples than those obtained when the classic acid-diffusion method is used. In addition, it is a very fast procedure which does not alter the pH or protein integrity of the biological sample.  相似文献

The incorporation of 14CO2 by an exponentially growing culture of the autotrophic bacterium Methanobacterium thermoautotrophicum has been studied. The distribution of radioactivity during 2s–120s incubation periods has been analyzed by chromatography and radioautography. After a 2 s incubation most of the radioactivity of the ethanolsoluble fraction was present in the amino acids alanine, glutamate, glutamine and aspartate, whereas phosphorylated compounds were only weakly labelled. The percentage of the total radioactivity fixed, which was contained in the principal early labelled amino acid alanine, increased in the first 20 s and only then decreased, indicating that alanine is derived from primary products of CO2 fixation.The labelling patterns of alanine produced during various incubation times have been determined by degradation. After a 2 s 14CO2 pulse, 61% of the radioactivity was located in C-1, 23% in C-2, and 16% in C-3. The results are consistent with the operation of a previously proposed autotrophic CO2 assimilation pathway which involves the formation of acetyl CoA from 2 CO2 via one-carbon unit intermediates, followed by the reductive carboxylation of acetyl CoA to pyruvate.  相似文献

The contribution of individual vascular bundles of the stemto the flow of assimilates from a selected source leaf to thesink regions was investigated inUrtica dioica L., a plant witha decussate leaf arrangement. Two homologous sets of eight vascularstrands were recognized, arranged in mirror symmetry in thestem internodes. In each set, three of the bundles were identifiedas traces of one leaf merging into the vascular system of thestem one node below the origin of the leaf. The main bundleof a stem-half bifurcates at each end of the internode intotwo subdominant bundles, which combine in the next but one nodeto form the dominant bundle again. Each set of vascular strandsalso contains two minor bundles which pass more or less withoutinterruption through the whole stem. The uppermost mature source leaf (leaf number 5 as counted fromthe tip) was exposed to14CO2in a closed gas circuit. The concentrationof the carbon-labelled CO2was maintained at the ambient CO2levelto maintain the natural source strength of the leaf. By theend of the usual nocturnal dark phase, carbon from the sourceleaf had been imported predominantly by sink leaves of the sameorthostichy. Lesser, but significant amounts of radiocarbonwere also incorporated into the sink leaves of the adjacenttwo orthostichies via the marginal leaf traces. In spite ofthe junction of the vascular strands in the nodes and an interfascicularconnection of the stem bundles, randomization of the photosynthatesfrom individual leaves was minimal in the vascular system ofthe stem in the upward direction, and also low in the flux tothe roots. Substantial amounts of radioactivity were also foundin the lately-formed xylem elements of the vascular strandsand their interfascicular connections, indicating active secondarygrowth.
