Aim: The aim of this study was to analyze the expression levels of plasma soluble Fas (sFas) and soluble Fas ligand (sFasL) in patients with orthotopic liver transplantation (OLT) procedures routinely performed without venovenous bypass. Methods: The sFas and sFasL were analyzed in the blood of 20 consecutive patients who underwent transplantation. Blood samples were drawn from the radial artery at serial time points before, during, and after surgery. Plasma levels of sFas and sFasL were detected by Enzyme Linked-Immuno-Sorbent Assay. Plasma aspartate transaminase (AST) and alanine transaminase (ALT) were assayed by routine clinical chemistry testing. Results: Marked elevation of plasma AST and ALT were detected at the reperfusion and postoperation time points (P < 0.001), with a peak on the first postoperative day. The mean plasma concentration of sFas and sFasL remained unchanged from preoperative to anhepatic phase (T1 to T3) (P  0.268). The sFas and sFasL concentrations were significantly higher at 15 and 60 min after reperfusion compared to the preoperative value (P  0.048). Postoperatively, sFas and sFasL concentration were decreased to preoperative levels on the first postoperative day (P  0.127). Conclusion: The sFas and sFasL seem to be involved in reperfusion injury during OLT. The understanding of Fas may provide new insights into the mechanisms of ischemia/reperfusion injury during OLT.  相似文献

A group of 20 healthy volunteers [10 women, 10 men; median age 25 (20–33) years] were examined by means of pulsed wave Doppler echocardiography, blood sample analysis and psychological testing before and after listening to three different examples of music: a waltz by J. Strauss, a modern classic by H. W. Henze, and meditative music by R. Shankar. To assess small haemodynamic changes, mitral flow, which reflects left ventricular diastolic behaviour, was measured by Doppler ultrasound. Heart rate, arterial blood pressure and plasma concentrations of adrenocorticotropic hormone, cortisol, prolactin, adrenaline, noradrenaline, atrial natriuretic peptide (ANP) and tissue plasminogen activator (t-PA) were determined simultaneously. Transmitral flow profile is characterized by early E-wave and late atrial induced A-wave. Velocity-time integrals were measured and the atrial filling fraction was calculated. The mental state was measured by using a psychological score (Zerssen) with low values (minimum 0) for enthusiastic and high values (maximum 56) for depressive patterns. Music by J. Strauss resulted in an increase of atrial filling fraction (AFF; 29% vs 26%;P<0.05) and ANP (63 pg·ml–1 vs 60 pg·ml–1;P<0.05). The mental state was improved (Zerssen: 6.5 vs 11 points;P<0.05). After the music of H. W. Henze prolactin values were lowered (7.7 ng·ml–1 vs 9.1 ng·ml–1;P<0.01). The music of R. Shankar led to a decrease of cortisol concentrations (57 ng·ml–1 vs 65 ng·ml–1;P<0.001), noradrenaline concentrations (209 g·l–1 vs 256 g·l–1;P<0.01) andt-PAantigen concentrations (1.1 ng·ml–1 vs 1.4 ng·ml–1;P<0.05). Heart rate and blood pressure remained unchanged during the whole experiment. We concluded that different types of music induced changes of left ventricular diastolic function and plasma hormone concentrations. After rhythmic music (Strauss) AFF and ANP increased significantly, the mental state being improved. Meditative music (Shankar) lowered plasma cortisol, noradrenaline and t-PA concentrations; the observed increase of early left ventricular filling was not statistically significant. Prolactin concentrations decreased after modern music (Henze). Thus, it would seem to be possible to detect cardiovascular changes following different types of music by Doppler ultrasound and hormone analysis, meditative music having promising therapeutic implications in the treatment of conditions of stress.This paper contains data from J. Alicyclobacillus and viable aerobic counts were monitored at nine different production stages of pear concentrate, with the functioning of either a recirculating or a one-pass flume water system. Significantly (P < 0.05) higher levels of Alicyclobacillus were detected in the final pasteurized product (102–104°C for 90 s) when the recirculating flume water system was operational. An average of 1.19 Log10 c.f.u. ml−1 vegetative cells and 1.35 Log10 c.f.u. ml−1 endospores were recovered, whereas 0 c.f.u. ml−1 vegetative cells and endospores were detected when the one-pass flume water system was operational. Alicyclobacillus levels did not differ significantly (P > 0.05) in condensate water during the functioning of the two flume water systems, with 1.81 Log10 c.f.u. ml−1 vegetative cells and 1.01 Log10 c.f.u. ml−1 endospores (recirculating system) and 0.78 Log10 c.f.u. ml−1 vegetative cells and 0.42 Log10 c.f.u. ml−1 endospores (one-pass system) recovered, respectively. As a result, water treatment protocols should be established if untreated recirculating flume or condensate water is to be used in order to prevent Alicyclobacillus contamination and accumulation in the processing environment.  相似文献

Gout patients have a high incidence of atherosclerotic coronary heart disease. Low serum paraoxonase (PON) activity is considered a risk factor for atherosclerosis. The relationships among paraoxonase-1 (PON1) activity, oxidative stress parameters, and atherosclerosis in gout is not known. Therefore, we determined the plasma levels of malondialdehyde (MDA), oxidized low-density lipoprotein (Ox-LDL), and activities of PON1/superoxide dismutase (SOD) activities in 49 gout patients (mean age 44.2 ± 7.0 years) and 42 healthy, age-matched controls (mean age 45.0 ± 9.3 years). PON1 was measured spectrophotometrically, MDA by thiobarbituric acid method, SOD by Griess reaction, and Ox-LDL by sandwich ELISA. Lipid and other biochemical parameters were determined by routine laboratory methods. In gout patients, PON1/SOD activities and MDA/Ox-LDL levels were 131.3 ± 25.3/75.3 ± 28.9 kU l−1 and 6.12 ± 1.67 nmol ml−1/690.1 ± 180.2 μg l−1, respectively. In controls, these were 172.5 ± 27.8/94.0 ± 26.3 kU l−1 and 4.10 ± 1.25 nmol ml−1/452.3 ± 152.1 μg l−1, respectively. Thus, in gout patients, there was a significant decrease in PON1 (P < 0.01) and SOD (P < 0.05) activities, and an increase in MDA (P < 0.01) and Ox-LDL (P < 0.01) levels compared with controls. PON1 activity correlated positively with SOD (P < 0.05), and negatively with MDA (P < 0.01) and Ox-LDL (P < 0.01). These results suggest that gout patients were in a state of oxidative stress and the protective effects of HDL against atherosclerosis maybe dependent on PON1 activity. These findings may explain in part the reported increase in cardiovascular mortality in gout patients.  相似文献

High mobility group box 1 protein (HMGB1) plays an important role in myocardial ischemia and reperfusion (I/R) injury. Ethyl pyruvate (EP), a potent reactive oxygen species scavenger, has been reported to inhibit myocardial apoptosis and reduce myocardial I/R injury. The aim of this study was to investigate the mechanism by which EP reduces myocardial I/R injury in rats. Anesthetized male rats were once treated with EP (50 mg/kg, i.p.) before ischemia, and then subjected to ischemia for 30 min followed by reperfusion for 4 h. Lactate dehydrogenase (LDH), creatine kinase (CK), malondialdehyde (MDA), superoxide dismutase (SOD) activity and infarct size were measured. HMGB1 expression was assessed by immunoblotting. The results showed that pretreatment of EP (50 mg/kg) could significantly reduce the infarct size and the levels of LDH and CK after 4 h reperfusion (all P < 0.05). EP could also significantly inhibit the increase of the MDA level, the decrease of the SOD level (both P < 0.05). Meanwhile, EP could significantly inhibit the expression of HMGB1 induced by I/R. The present study suggested that ethyl pyruvate could attenuate myocardial I/R injury by inhibiting HMGB1 expression.  相似文献

Pekin ducks (Anas platyrhynchos) were bilaterally adrenalectomized (biADX), injected with 1 mg of triamcinolone (TRIAM) kg bw−1 im and given 0.9% saline drinking water during a 24 h recovery period followed by chemical sympathectomy with 6OH DOPA 3 h before the start of experimental observations. Baseline plasma dopamine (DA) concentrations decreased from 283 ± 88.5 pmol ml−1 to 42.4 ± 11.1 pmol ml−1; epinephrine (E) from 142 ± 46 pmol ml−1 to 18.4 ± 9.2 pmol ml−1 and norepinephrine (NE) from 742 ± 84 pmol ml−1 to 406 ± 38 pmol ml−1 1 day after biADX + TRIAM but before chemical sympathectomy. Baseline MABP increased from 132 ± 3.2 mmHg to 209 ± 14.3 mmHg (P < 0.05) in response to TRIAM. After chemical sympathectomy with 6OH DOPA there was an additional 90% decrease in plasma NE to 42 ± 9.4 pmol ml−1 and a concurrent 60% decrease in MABP to 83.4 ± 6.9 mmHg (P < 0.05). Nasal fluid secretion was maintained by the continuous infusion of hypertonic saline (1,000 mosmol kg H2O−1 at a rate of 0.3 ml kg−1 min−1). Rates of nasal fluid secretion and fluid electrolyte concentrations were unchanged following biADX + TRIAM + 6OH DOPA. Angiotensin II (ANG II; dose 1 μg kg bw−1 i.v.), attenuated nasal fluid secretion showing that the response to ANG II was not NE- dependent. Plasma NE concentrations decreased following Tyramine i.v. (33 ± 8.5 pmol ml−1) there being no vasopressor response. This is the first report of the ANG II induced attenuation of duck salt gland secretion in the absence of measurable E and NE.  相似文献

The smooth skate, Malacoraja senta, and thorny skate, Amblyraja radiata, are two commercially exploited batoids found within the Gulf of Maine. During the past five years, we conducted a large study to accurately describe important biological life history parameters previously lacking for these species. As part of that project, the current study reports our findings on the hormonal profiles associated with the reproductive cycles of M. senta and A. radiata. Blood samples were obtained from mature M. senta and A. radiata of both sexes from all months of the year, and plasma testosterone (T), estradiol (E2) and progesterone (P4) concentrations were determined using radioimmunoassay (RIA). In female M. senta and A. radiata, monthly T concentrations ranged from 4,522 pg ml−1 to 1,373 pg ml−1 and 31,940 pg ml−1 to 14,428 pg ml−1, E2 concentrations from 831 pg ml−1 to 60 pg ml−1 and 8,515 pg ml−1 to 2,902 pg ml−1, and P4 concentrations from 3,027 pg ml−1 to 20 pg ml−1 and 3,264 pg ml−1 to 331 pg ml−1, respectively. No statistical differences were detected between any months for any hormone. Estradiol concentrations were not correlated with ovary weight, shell gland weight, or diameter of the largest follicles in either species. Monthly T concentrations in male M. senta and A. radiata ranged from 23,146 to 12,660 pg ml−1 and from 57,500pg ml−1 to 24,737 pg ml−1, while E2 concentrations ranged from 7.5 pg ml−1 to undetectable and 103 to 30 pg ml−1, respectively. No statistical differences were observed between months for either steroid. Testosterone concentrations were weakly correlated with testes weight and percent of stage VI spermatocysts in A. radiata, however, no correlation was detected between T and stage VI spermatocysts in M. senta. Collectively, these data support the previous conclusion that M. senta and A. radiata of both sexes are capable of reproducing year round in the western Gulf of Maine.  相似文献

Objectives: Our previous study has shown that slow or “controlled” reperfusion for the ischemic heart reduces cardiomyocyte injury and myocardial infarction, while the mechanisms involved are largely unclear. In this study, we tested the hypothesis that enhancement of survival and prevention of apoptosis in hypoxic/reoxygenated cardiomyocytes by hypoxic postconditioning (HPC) are associated with the reduction in peroxynitrite (ONOO) formation induced by hypoxia/reoxygenation (H/R). Methods: Isolated adult rat cardiomyocytes were exposed to 2 h of hypoxia followed by 3 h of reoxygenation. After 2 h of hypoxia the cardiomyocytes were either abruptly reperfused with pre-oxygenized culture medium or postconditioned by two cycles of 5 min of brief reoxygenation and 5 min of re-hypoxia followed by 160 min of abrupt reoxygenation. Results: H/R resulted in severe injury in cardiomyocytes as evidenced by decreased cell viability, increased LDH leakage in the culture medium, increased apoptotic index (P values all less than 0.01 vs. normoxia control group) and DNA ladder formation, which could be significantly attenuated by HPC treatment applied before the abrupt reoxygenation (P < 0.05 vs. H/R group). In addition, H/R induced a significant increase in ONOO formation as determined by nitrotyrosine content in cardiomyocytes (P < 0.01 vs. normoxia control). Treatment with the potent ONOO scavenger uric acid (UA) at reoxygenation significantly decreased ONOO production and protected myocytes against H/R injury, whereas the same treatment with UA could not further enhance myocyte survival in HPC group (P > 0.05 vs. HPC alone). Statistical analysis showed that cell viability closely correlated inversely with myocyte ONOO formation (P < 0.01). Conclusion: These data demonstrate that hypoxic postconditioning protects myocytes against apoptosis following reoxygenation and enhances myocytes survival, which is partly attributable to the reduced ONOO formation following reoxygenation. We used culture- and molecular-biology-based methods to investigate microbial diversity in the traditional Mongolian fermented milks “Airag” (fermented mare’s milk) and “Tarag” (fermented milk of cows, yaks, goats, or camels). By rRNA or functional gene sequencing, we identified 367 lactic acid bacteria (LAB) strains and 152 yeast strains isolated from 22 Airag and 31 Tarag samples. The total concentration of LAB in Airag (107.78 ± 0.50 c.f.u. ml–1; mean ± SD) was significantly lower (P < 0.01) than in Tarag (108.35 ± 0.62 c.f.u. ml−1), whereas the total concentration of yeasts in Airag (107.41 ± 0.61 c.f.u. ml-1) was significantly higher (P < 0.01) than in Tarag (105.86 ± 1.29 c.f.u. ml-1). Lactobacillus helveticus and Lactobacillus kefiranofaciens were isolated from Airag as the predominant LAB strains at levels of about 107 c.f.u. ml−1, whereas Lactobacillus delbrueckii subsp. bulgaricus, L. helveticus, and Streptococcus thermophilus were the predominant isolates from Tarag at about 107 c.f.u. ml−1. The lactose-fermenting Kluyveromyces marxianus was isolated predominantly from Airag as its major alcoholic fermentation component. Non-lactose-fermenting yeasts such as Saccharomyces cerevisiae, Issatchenkia orientalis, and Kazachstania unispora were the predominant isolates from Tarag, at about 105 c.f.u. ml−1. The apparent geographic differences in the L. kefiranofaciens and S. thermophilus contents of Tarag strongly suggested that differences among the animal species from which the milk was sourced, rather than geographic distances, were the most important factors influencing the diversity of the microbial composition of traditional fermented milks in Mongolia.  相似文献

Transient episodes of ischemic preconditioning (PC) render myocardium protected against subsequent lethal injury after ischemia and reperfusion. Recent studies indicate that application of short, repetitive ischemia only during the onset of reperfusion after the lethal ischemic event may obtain equivalent protection. We assessed whether such ischemic postconditioning (Postcon) is cardioprotective in pigs by limiting lethal injury. Pentobarbital sodium-anesthetized, open-chest pigs underwent 30 min of complete occlusion of the left anterior descending coronary artery and 3-h reflow. PC was elicited by two cycles of 5-min occlusion plus 10-min reperfusion before the 30-min occlusion period. Postcon was elicited by three cycles of 30-s reperfusion, followed by 30-s reocclusion, after the 30-min occlusion period and before the 3-h reflow. Infarct size (%area-at-risk using triphenyltetrazolium chloride macrochemistry; means +/- SE) after 30 min of ischemia was 26.5 +/- 5.2% (n = 7 hearts/treatment group). PC markedly limited myocardial infarct size (2.8 +/- 1.2%, n = 7 hearts/treatment group, P < 0.05 vs. controls). However, Postcon had no effect on infarct size (37.8 +/- 5.1%, n = 7 hearts/treatment group). Within the subendocardium, Postcon increased phosphorylation of Akt (74 +/- 12%) and ERK1/2 (56 +/- 10%) compared with control hearts subjected only to 30-min occlusion and 15-min reperfusion (P < or = 0.05), and these changes were not different from the response triggered by PC (n = 5 hearts/treatment group). Phosphorylation of downstream p70S6K was also equivalent in PC and Postcon groups. These data do not support the hypothesis that application of 30-s cycles of repetitive ischemia during reperfusion exerts a protective effect on pig hearts subjected to lethal ischemia, but this is not due to a failure to phosphorylate ERK and Akt during early reperfusion.  相似文献

The aims of the present study were to evaluate if seasonality in semen characteristics and plasma testosterone concentrations exist in Markhoz male goats. Ten Markhoz (Angora) bucks were housed and fed according to standard recognized practices. During the observation period, semen was collected monthly with the aid of an electro-ejaculator and examined microscopically immediately after collection. Physical parameters of semen and the semen index were recorded. Blood samples were also taken monthly throughout the observation period and the plasma testosterone concentration monitored. Bucks demonstrated a higher semen quality (P < 0.05) in autumn and summer (semen index of 965 × 106 and 752 × 106 ml−1, respectively), compared to spring and winter (semen index of 606 × 106 and 512 × 106, respectively). This coincided with a higher (P < 0.05) plasma testosterone concentration in autumn and summer (8.1 and 10.1 ng ml−1, respectively), compared to that obtained in spring (3.0 ng ml−1) and winter (2.5 ng ml−1). During autumn and summer, the ejaculate volume (average of 1.2 and 1.0 ml), sperm output (1159 × 106 and 1005 × 106 sperm ml−1), sperm mass motility (4.2 and 4.3), sperm progressive motility (83.9 and 82.0%) and percentage live sperm (90.7 and 88.2%, respectively) of the bucks were higher (P < 0.05) than in the spring (0.6 ml, 880 × 106 sperm ml−1, 3.3, 71.5% and 80.2%) and winter (0.7 ml, 863 × 106 sperm ml−1, 4.0, 71.5% and 84.9%, respectively). During autumn and summer, the percentage of sperm abnormalities (5.0 and 9.2%) was significantly lower than that in spring (12.9%) and winter (11.2%). The semen pH was slightly alkaline being significantly (P < 0.05) lower in the autumn (7.1) than in spring (7.3). Data showed season of the year to influence all semen parameters evaluated—indicating that optimal buck performance may be obtained in late summer and autumn. It can thus be said that Markhoz bucks have distinct seasonal spermatogenic activity, with poorer semen characteristics being recorded during winter and spring. This may be a critical obstacle when implementing an intensive breeding system of three kidding seasons in 2 years, with natural mating being implemented.  相似文献

The gene encoding enterotoxigenic Escherichia coli B-subunit heat-labile toxin (LTB) antigen was co-transformed into hairy root cultures of Nicotiana tabacum (tobacco), Solanum lycopersicum (tomato) and Petunia parodii (petunia) under the CaMV35S promoter. Tobacco and petunia roots contained ~65–70 μg LTB g−1 tissue whilst hairy roots of tomato contained ~10 μg LTB g−1. Antigen at ~600 ng ml−1 was detected in growth medium of tobacco and petunia. Tobacco roots with higher LTB levels showed growth retardation of ~80% whereas petunia hairy roots with similar levels of LTB showed only ~35% growth retardation, relative to vector controls. Regeneration of plants from LTB-containing tobacco hairy roots was readily achieved and re-initiated hairy roots from greenhouse-grown plants showed similar growth and LTB production characteristics as the original hairy root cultures.  相似文献

In 2005 and 2006, adult sockeye salmon (Oncorhynchus nerka) were captured en route to spawning grounds and placed in either a slow (∼ 0.1 m·s−1) or fast (∼0.4 m·s−1) water velocity treatment for 18 days in order to assess how migrational energy depletion during the final stages of maturation affected physiological condition and survival. Fish in the fast treatment utilized more energy than the slow treatment in 2005 (0.91 MJ kg−1 vs. 0.43 MJ kg−1; P = 0.010), and 2006 (0.72 MJ kg−1 vs. 0.37 MJ kg−1; P = 0.021). Non-treatment fish captured upon arrival at spawning grounds showed energy levels intermediate to the two treatments in 2005 and lower than both in 2006, suggesting that energy use during the treatments were within levels normally experienced by this population. No differences in survival were found between treatments (P > 0.05), although females had lower survival than males in both years (both P < 0.01). After 18 days, surviving fish from the fast treatment showed signs of elevated physiological stress relative to fish from the slow treatment. Specifically, plasma osmolality was lower in fast fish in 2005 (P < 0.001), as was plasma chloride in both years (both P < 0.02). In 2006, plasma lactate was higher (P = 0.014) in fast fish. Within the ranges of energetic depletion that were examined here, a more energy-intensive migration can have a substantial influence on the physiological condition and stress of adult sockeye salmon, but not on survival.  相似文献

The purpose of the present study was to determine the inhibitory activities of two bacteriocins, produced by lactobacilli, against genital mycoplasmas. In this study, infections produced by genital mycoplasmas were studied; of these, 1.3% were caused by Mycoplasma hominis, 10.7% by Ureaplasma urealyticum and 5.6% by U. urealyticum + M. hominis. U. urealyticum was isolated from 75 out of 123 patients with genital mycoplasmas, while M. hominis was isolated from 9 patients (7.3%) and both U. urealyticum and M. hominis from 39 patients (31.7%). Bacteriocins, L23 and L60, produced by Lactobacillus fermentum and L. rhamnosus, respectively, appear to be two novel inhibitors of bacterial infection with potential antibacterial activity. Both bacteriocins proved to be active against 100% of strains tested; MICs of bacteriocin L23 ranged between 320 and 160 UA ml−1 for 78% of the M. hominis strains and between 320 and 80 UA ml−1 for 95% of the U. urealyticum strains. In addition, bacteriocin L60 was still active at 160 UA ml−1 for a high percentage (56%) of M. hominis strains, and at 80 UA ml−1 for 53% of the U. urealyticum strains. Interestingly, these antimicrobial substances produced by lactobacilli showed an inhibitory activity against genital mycoplasmas even when diluted. Altogether, our study indicates that the bacteriocins, L23 and L60, are good candidates for the treatment or prevention of genital infections in women.  相似文献

A new cell culture system (MRH) was developed for the first time from 2 months old freshwater prawn, Macrobrachium rosenbergii. Primary cultures were developed from heart tissues by explant culture technique. Cell outgrowth was obtained from the heart explant after 14 days of explant culture. The culture medium used was Leibovitz-15 supplemented with 20% Fetal Bovine Serum along with 1% prawn hemolymph serum, 0.1% glucose, 0.5% NaCl and antibiotics (Penicillin 10,000 Units ml−1, Streptomycin 10,000 μg ml−1, Amphotericin B 500 mg ml−1) with a final osmomolality of 470–550 mmol kg−1. The pH of the growth medium found suitable for the growth of the cells was 7.20. The viability of cells was found to be 60% when revived after a month of storage in liquid nitrogen.  相似文献

Staphylococcus aureus is an important pathogen for both humans and animals, and it has been an ubiquitous etiological agent of bovine mastitis in dairy farms worldwide. Elimination of S. aureus with classic antibiotics is difficult, and the current study aimed to evaluate the efficacy of ethanolic extracts of propolis (EEP) against S. aureus cultivated in complex media or milk. EEP (0–0.5 mg ml−1) decreased growth of S. aureus in BHI media and 1 mg ml−1 was bactericidal against washed cell suspensions (107 CFU ml−1). Propolis extracts also killed S. aureus cells resuspended in milk, but the bactericidal dose was at least 20-fold greater. Cultures that were transferred for at least 60 generations with sub-lethal doses of propolis did not change much their sensibility to EEP. Atomic force microscopy images revealed changes in morphology and cell size of S. aureus cells exposed to EEP (0.5 mg ml−1). Our results indicate that propolis extracts might be effective against mastitis-causing S. aureus strains in vivo, but milk constituents affect the inhibitory activity of propolis. Considering that propolis-resistance appears to be a phenotype not easily selected, the use of EEP combined or not with other antimicrobial agents might be useful for mastitis control in vivo.  相似文献

The South China Sea (SCS) is the world’s largest marginal sea being notable for vertical mixing at various scales resulting in a sequence of chemical and biological dynamics in surface waters. We investigated the ingestion, gut content, evacuation and clearance rates of copepods collected from six stations (including a South East Asia Time Series station) along a transect line in the tropical of a SCS cruise during September 27, 1999 to October 2, 1999. The goal of the present study was to understand the feeding ecology of copepods in the upper water layers (0–5 m) of the northern SCS during autumn. We measured the gut pigment contents of 33 copepod species by the gut fluorescence method. The gut chlorophyll a values of most small size copepods (<1 mm) were lower than 1.00 ng Chl a individual−1. The highest gut pigment content was recorded in Scolecithrix danae (7.07 ng Chl a individual−1). The gut pigment contents of 33 copepod species (including 70 samples and 1,290 individuals) estimated is negatively correlated with seawater temperature (Pearson correlation r = −0.292, P = 0.014) and is positively correlated with the chlorophyll a concentration of ambient waters (Pearson correlation r = 0.243, P = 0.043). Mean gut pigment content, ingestion and clearance rates (from 80 samples and 1,468 individuals) show that larger copepods (>2 mm) had significantly higher values than medium sized copepods (1–2 mm) and smaller sized copepods. The present study shows that the performance of feeding on phytoplankton was variable in different sized copepod groups, suggesting that copepods obtained in the tropical area of the southeastern Taiwan Strait might be opportunistic feeders.  相似文献

Groups of juvenile tench (7.02 ± 0.28 g) were reared under four different light regimes; blue light, red light (80 Wm−2 12L:12D photoperiod) white light (912 ± 210 lux, 80 Wm−2, 12L:12D photoperiod) and no light (0 lux) (0L:24D). Visibility of fish out of shelters was used as an indicator of activity and was monitored by video recording. Blood plasma cortisol concentrations were also measured. Fish under blue or white light were significantly less active during the photophase than those under red or no light (P < 0.01). Red light produced similar activity patterns to fish receiving 24 h darkness. Plasma cortisol concentrations were also significantly influenced (P < 0.05) with the fish under white light having the highest plasma cortisol concentration (317 ± 62 ng cm−3) compared to fish in the dark treatment (106 ± 36 ng cm−3). Thus, the provision of coloured light filters increases activity in juvenile tench and may reduce their intrinsic stress level.  相似文献

A newly isolated Geobacillus sp. IIPTN (MTCC 5319) from the hot spring of Uttarakhand's Himalayan region produced a hyperthermostable α-amylase. The microorganism was characterized by biochemical tests and 16S rRNA gene sequencing. The optimal temperature and pH were 60°C and 6.5, respectively, for growth and enzyme production. Although it was able to grow in temperature ranges from 50 to 80°C and pH 5.5–8.5. Maximum enzyme production was in exponential phase with activity 135 U ml−1 at 60°C. Assayed with cassava as substrate, the enzyme displayed optimal activity 192 U ml−1 at pH 5.0 and 80°C. The enzyme was purified to homogeneity with purification fold 82 and specific activity 1,200 U mg−1 protein. The molecular mass of the purified enzyme was 97 KDa. The values of K m and V max were 36 mg ml−1 and 222 μmol mg−1 protein min−1, respectively. The amylase was stable over a broad range of temperature from 40°C to 120°C and pH ranges from 5 to 10. The enzyme was stimulated with Mn2+, whereas it was inhibited by Hg2+, Cu2+, Zn2+, Mg2+, and EDTA, suggesting that it is a metalloenzyme. Besides hyperthermostability, the novelty of this enzyme is resistance against protease.  相似文献
