Stereotactic body radiotherapy (SBRT) distinguishes itself by necessitating more rigid patient immobilization, accounting for respiratory motion, intricate treatment planning, on-board imaging, and reduced number of ablative radiation doses to cancer targets usually refractory to chemotherapy and conventional radiation. Steep SBRT radiation dose drop-off permits narrow ''pencil beam'' treatment fields to be used for ablative radiation treatment condensed into 1 to 3 treatments.Treating physicians must appreciate that SBRT comes at a bigger danger of normal tissue injury and chance of geographic tumor miss. Both must be tackled by immobilization of cancer targets and by high-precision treatment delivery. Cancer target immobilization has been achieved through use of indexed customized Styrofoam casts, evacuated bean bags, or body-fix molds with patient-independent abdominal compression.1-3 Intrafraction motion of cancer targets due to breathing now can be reduced by patient-responsive breath hold techniques,4 patient mouthpiece active breathing coordination,5 respiration-correlated computed tomography,6 or image-guided tracking of fiducials implanted within and around a moving tumor.7-9 The Cyberknife system (Accuray [Sunnyvale, CA]) utilizes a radiation linear accelerator mounted on a industrial robotic arm that accurately follows patient respiratory motion by a camera-tracked set of light-emitting diodes (LED) impregnated on a vest fitted to a patient.10 Substantial reductions in radiation therapy margins can be achieved by motion tracking, ultimately rendering a smaller planning target volumes that are irradiated with submillimeter accuracy.11-13Cancer targets treated by SBRT are irradiated by converging, tightly collimated beams. Resultant radiation dose to cancer target volume histograms have a more pronounced radiation "shoulder" indicating high percentage target coverage and a small high-dose radiation "tail." Thus, increased target conformality comes at the expense of decreased dose uniformity in the SBRT cancer target. This may have implications for both subsequent tumor control in the SBRT target and normal tissue tolerance of organs at-risk. Due to the sharp dose falloff in SBRT, the possibility of occult disease escaping ablative radiation dose occurs when cancer targets are not fully recognized and inadequate SBRT dose margins are applied. Clinical target volume (CTV) expansion by 0.5 cm, resulting in a larger planning target volume (PTV), is associated with increased target control without undue normal tissue injury.7,8 Further reduction in the probability of geographic miss may be achieved by incorporation of 2-[18F]fluoro-2-deoxy-D-glucose (18F-FDG) positron emission tomography (PET).8 Use of 18F-FDG PET/CT in SBRT treatment planning is only the beginning of attempts to discover new imaging target molecular signatures for gynecologic cancers.  相似文献

Rapid immobilization of inorganic nitrogen (N) in soil contributes to ecosystem N accumulation, even in old-growth and chronically-fertilized forests once thought to have poor N retention capacity. In old-growth conifer and hardwood stands in Pennsylvania, we tested the hypotheses that biotic and abiotic N immobilization are regulated by N form and forest type. We added 15NH4 +, 15NO2 ?, and 15NO3 ? to sterile (γ-irradiated) and live organic-horizon soil and define N immobilization as the mass of added 15N remaining in soil following extractions conducted 15 min, 24 h, and 21 days later. Immobilization of NO2 ? (19–25% of added N) occurred in sterile soils within 15 min and was little changed thereafter. Tracer NO3 ? immobilization was not observed, although soils had been pretreated (refrigerated) so as to quantify the lower limit of immobilization potential. Immobilization of NH4 + (27%) occurred in live conifer soils by 21 days but not in other treatments. In 21-day incubations, tracer N immobilization was greater in NO3 ?-poor and humic-rich soils. Immobilization was greater in sterile than in live soil, perhaps owing to artifacts of sterilization. Conifer stands exhibited more massive O-horizons, so NO2 ? immobilization per unit area was greater in conifer (1.46 mg N m?2) than hardwood (0.43 mg N m?2) stands, possibly accounting for lower N leaching from conifer forests. Areal immobilization rates appear to be fast enough to retain all N transformed to NO2 ?, so NO2 ? production may be a limiting step in soil N retention in old-growth ecosystems.  相似文献

Production of gluconic acid by cells of Aspergillus niger immobilized on polyurethane foam was studied in repeated-batch shake-flask and bubble-column fermentations. For passive immobilization, various amounts of polyurethane foam and spore suspension were tested in order to obtain a suitable combination for optimal concentration of immobilized biomass. Immobilized cells were sucessfully reused with higher levels of product formation being maintained for longer period (65–70h) than free cells. The highest gluconic acid concentration of about 143 g l–1 was reached on hydrol-based production medium with 0.3-cm3 foam cubes in the bubble column, where the effect of more suitable aeration and particle volume: medium volume ratio scheme was also investigated.  相似文献

Fungal biomass in the decaying cones ofPinus densiflora was investigated. Leaching, immobilization and mobilization phases were recognized in the decomposition process of cones. Fungal biomass was estimated by the agar-film technique, using a conversion factor of 0.62 mg dry wt. mm−3 of hyphal volume to biomass and a factor of 2.5 for in-efficiencency of homogenization. The fungal biomass was 4.9±2.1 (mean±S.D.) mg dry wt. g−1 dry matter in the cones on the tree, 11±6 mg g−1 in the leaching phase, 19±7 mg g−1 in the immobilization phase and 30±15 mg g−1 in the mobilization phase. It significantly increased after cones had lain on the forest floor, and also in the immobilization phase. The latter result suggests that the fungal biomass contributed to the immobilization of nitrogen in the decomposition process. The ratio of ergosterol content to fungal biomass in the cones was 2.9–8.8 μg mg−1 dry wt., lying in the range of 2–16 μg mg−1 reported for mycelia. This suggested that the estimate of fungal biomass was reasonable. Reduction in this ratio with the dry weight loss in the cones suggested that the proportion of relatively active fungal biomass decreased with the progress of decomposition.  相似文献

Evidence for abiotic immobilization of nitrogen (N) in soil is accumulating, but remains controversial. Identifying the fate of N from atmospheric deposition is important for understanding the N cycle of forest ecosystems. We studied soils of two Abies pinsapo fir forests under Mediterranean climate seasonality in southern Spain—one with low N availability and the other with symptoms of N saturation. We hypothesized that biotic and abiotic immobilization of nitrate (NO3 ) would be lower in soils under these forests compared to more mesic temperate forests, and that the N saturated stand would have the lowest rates of NO3 immobilization. Live and autoclaved soils were incubated with added 15NO3 (10 μg N g−1 dry soil; 99% enriched) for 24 h, and the label was recovered as total dissolved-N, NO3 , ammonium (NH4 +), or dissolved organic-N (DON). To evaluate concerns about possible iron interference in analysis of NO3 concentrations, both flow injection analysis (FIA) and ion chromatography (IC) were applied to water extracts, soluble iron was measured in both water and salt extracts, and standard additions of NO3 to salt extracts were analyzed. Good agreement between FIA and IC analysis, low concentrations of soluble Fe, and 100% (±3%) recovery of NO3 standard additions all pointed to absence of an interference problem for NO3 quantification. On average, 85% of the added 15NO3 label was recovered as 15NO3 , which supports our hypothesis that rates of immobilization were generally low in these soils. A small amount (mean = 0.06 μg N g−1 dry soil) was recovered as 15NH4 + in live soils and none in sterilized soils. Mean recovery as DO15N ranged from 0.6 to 1.5 μg N g−1 dry soil, with no statistically significant effect of sterilization or soil type, indicating that this was an abiotic process that occurred at similar rates in both soils. These results demonstrate a detectable, but modest rate of abiotic immobilization of NO3 to DON, supporting our first hypothesis. These mineral soils may not have adequate carbon availability to support the regeneration of reducing microsites needed for high rates of NO3 reduction. Our second hypothesis regarding lower expected abiotic immobilization in soils from the N-saturated site was not supported. The rates of N deposition in this region may not be high enough to have swamped the capacity for soil NO3 immobilization, even in the stand showing some symptoms of N saturation. A growing body of evidence suggests that soil abiotic NO3 immobilization is common, but that rates are influenced by a combination of factors, including the presence of plentiful available carbon, reduced minerals in anaerobic microsites and adequate NO3 supply.  相似文献

Screening of leachable toxic chemicals in a horseradish peroxidase-H2O2 immobilization system established that immobilization was promising for most phenolic pollutants but not for benzoic acid, 2,6-dinitrocresol, or dibutyl phthalate. The treatment did not mobilize inherently nonmobile pollutants such as anilines and benzo[a]pyrene. In a separate study, an extracellular laccase in the culture filtrate of Geotrichum candidum was selected from five fungal enzymes evaluated as a cost-effective substitute for horseradish peroxidase. This enzyme was used in demonstrating the immobilization and subsequent fate of 14C-labeled 4-methylphenol and 2,4-dichlorophenol in soil columns. When applied to Lakewood sand, 98.1% of 4-methylphenol was leached through with distilled water. Two days after immobilization treatment with the G. candidum culture filtrate, only 9.1% of the added 4-methylphenol was leached with the same volume of water. Of the more refractory test pollutant 2,4-dichlorophenol, 91.6% had leached at time zero and 48.5% had leached 1 day after the immobilization treatment. However, 2 weeks after immobilization, only 12.0% of the 2,4-dichlorophenol was leached compared with 61.7% from the control column that received no immobilization treatment. No remobilization of the bound pollutants was detected during 3- and 4-week incubation periods. Enzymatic immobilization of phenolic contaminants in soil appears to be a promising technique for the reduction of groundwater pollution by such substances.  相似文献

The objective of this paper was the investigation of a suitable Sepabeads? support and method for immobilization of lipase from Candida rugosa. Three different supports were used, two with amino groups, (Sepabeads? EC-EA and Sepabeads? EC-HA), differing in spacer length (two and six carbons, respectively) and one with epoxy group (Sepabeads? EC-EP). Lipase immobilization was carried out by two conventional methods (via epoxy groups and via glutaraldehyde), and with periodate method for modification of lipase. The results of activity assays showed that lipase retained 94.8% or 87.6% of activity after immobilization via epoxy groups or with periodate method, respectively, while glutaraldehyde method was inferior with only 12.7% of retention. The immobilization of lipase, previously modified by periodate oxidation, via amino groups has proven to be more efficient than direct immobilization of lipase via epoxy groups. In such a way immobilized enzyme exhibited higher activity at high reaction temperatures and higher thermal stability.  相似文献

Objective: To determine the ability of air displacement plethysmography (ADP) to predict visceral adipose tissue (VAT) volume in children. Research Methods and Procedures: Fifty‐five (33 boys/22 girls) white children 13 to 14 years old were studied. Anthropometric measures were collected for body mass, stature, BMI, and waist‐to‐hip ratio (WHR), and body fat percentage was estimated from triceps and subscapular skinfolds, bioelectrical impedance analysis, and ADP. VAT volume was determined using magnetic resonance imaging, using a multiple slice protocol at levels L1 to L5. Results: Boys had significantly (p ≤ 0.05) less VAT volume than girls [645.1 (360.5) cm3 vs. 1035.8 (717.3) cm3]. ADP explained the greatest proportion of the variance in VAT volume compared with the other anthropometric measures. Multiple regression analysis indicated that VAT volume was best predicted by ADP body fat percentage in boys [r2 = 0.81, SE of the estimate (SEE) = 160.1, SEE coefficient of variation = 25%] and by WHR and BMI in girls (r2 = 0.80, SEE = 337.71, SEE coefficient of variation = 33%). Discussion: Compared with the other anthropometric measures, ADP explains the greatest proportion of the variance in VAT volume in children 13 to 14 years old. For boys, ADP is the tool of choice to predict VAT volume, yet using the more simply collected measures of BMI and WHR is recommended for girls. However, large SE of the estimates remained, suggesting that if precision is needed, there is no surrogate for direct imaging of VAT.  相似文献

Hystrix javanica is endemic species in Indonesia. Study about fetal development of Hystrix javanica are very rare because of sample limitation. This study was carried out to describe the morphometrics and x-ray analysis of three fetuses in different stage to give basic information about fetal development of Hystrix javanica. Three fetus samples fixed in Bouin’s solution was used in this study. Observation was carried out to identify the characteristic of three fetus samples. This included the pattern of hair, body measurements, body volume, and body weight. X-ray analysis was carried out to know the ossification process in the fetal development. Statistical analysis was carried out using Microsoft 365® Excel program software. Three fetus samples had different specific hair pattern, that was hairless, smooth hairs, and smooth hairs with dense-non dense pattern. Body volume of 1st, 2nd, and 3rd fetus were 23mL, 90mL, and 170mL, respectively. Body weight of 1st, 2nd, and 3rd fetus were 19.5g, 79.22g, and 153.18g, respectively. Pearson’s correlation analysis shown strong relationship between total body length, front body length, back body length, horizontal body diameter, vertical body diameter, head length, and head diameter against body volume and body weight of three fetuses. Significant positive correlation was shown between horizontal body diameter, vertical body diameter, and head diameter against body volume and body length with P value < 0.05. Faint radiopaque images showed in the 2nd fetus sample and strong radiopaque images showed in the 3rd fetus sample. Radiopaque images were identified in the teeth, cranium, vertebrae, and extremities bones. In this study we concluded that there was a specific hair pattern in different fetal stage. All body measurements have positive correlation with body volume and body weight and x-ray analysis shown that the ossification of the bone was started to happen while the smooth hair was growth.  相似文献

Objective: This study presents total body volume (TBV) and regional body volume, and their relationships with widely used body composition indices [BMI, waist circumference (WC), and percentage body fat (% fat)] in severely obese adults (BMI ≥35 kg/m2). Research Methods and Procedures: We measured TBV, trunk volume (TV), arm volume (AV), leg volume (LV), and WC and estimated % fat in 32 severely obese persons with BMI 36 to 62 kg/m2 (23 women; age, 19 to 65 years; weight, 91 to 182 kg) and in 58 persons with BMI <35 kg/m2 (28 women; age, 18 to 83 years; weight, 48 to 102 kg) using a newly validated 3‐day photonic image scanner (3DPS, Model C9036–02, Hamamatsu Co., Japan) and calculated TV/TBV, AV/TBV, and LV/TBV. Results: Men had significantly larger TBV and higher TV/TBV and AV/TBV, but significantly lower LV/TBV than women, independently of BMI. TV/TBV increased while AV/TBV and LV/TBV decreased with increasing BMI, WC, and % fat, and the rate of increase in TV/TBV per % fat was significantly greater in severely obese individuals than in individuals with BMI <35 kg/m2. The relationships for TBV with % fat were much lower than with BMI or WC. Conclusion: Body volume gains were mainly in the trunk region in adults, irrespective of sex or BMI. For a given BMI, WC, or % fat, men had a significantly larger TV than women. The implication is that men could have higher health risks due to having higher trunk body weight as a proportion of total body weight compared with severely obese or less severely obese women.  相似文献

Optimal operation condition was investigated for immobilized rice callus culture using a turbine blade reactor (TBR2) with polyurethane foam supports. By using polyurethane foam block as immobilization support, the inhibition of cell growth at a high agitation speed was avoided because the hydrodynamic stress against immobilized cell was probably reduced. Experimental results in each operational condition were assessed by means of rice callus growth, immobilization ratio in TBR and those regeneration frequencies in regeneration culture using solid medium. Concerning with pore size of polyurethane foam and support size, three-millimeter cube support of polyurethane foam with an average pore size of 1.3 mm was the most suitable support. The maximum immobilization ratio was 50% under 5% support volume by volume of growth medium. For improving the immobilization ratio of rice callus in the TBR, the optimum TBR operation and modification were investigated further. By repeating a periodic operation 3 times (agitating at 300 rpm for 5 min and then 50 rpm for 2 min, and then 200 rpm of constant agitation speed during the remaining time), almost all supports could entrap rice callus and homogeneous immobilization was attained. The immobilization ratio was improved as compared with that using a constant operation at 200 rpm. Next, the TBR was modified by setting an air sparger inside the stainless mesh cylinder. In the modified TBR, the floating support by air bubbles was reduced, and the immobilization ratio increased further and reached 86.3% when we increased the support volume to 15% under periodic operation on a daily basis. The regeneration frequency of immobilized callus was also slightly increased by periodic operation and modification of the TBR.  相似文献

Naturally senesced needles from ponderosa pine (Pinus ponderosa Dougl.), grown from seed in open-top chambers under three levels of CO2 (350, 525 and 700 μl l-1) and three levels of N fertilization (0, 10 and 20 g N m-2 yr-1), were used in a field litterbag decomposition study and in a laboratory study on potential microbial and nonmicrobial N immobilization. The litterbag studies revealed no statistically significant effects of either CO2 or N treatment on mass loss, N concentration, or N content over a 26-month period. The laboratory study of potential 15N immobilization revealed no statistically significant effects of CO2 or N treatment on either total or microbial immobilization. Elevated (CO2) did have a significant negative effect on nonmicrobial immobilization, however. Natural abundance of 15N was significantly greater with elevated (CO2) in both live and naturally senesced needles under all N treatments. This pattern combined with 15N natural abundance in soils suggests that saplings grown under elevated (CO2) were either taking up more N from surface horizons or from a more recalcitrant soil N pool in either horizon. Polyamide matrices, such as membranes, gels and non-wovens, have been applied as supports for enzyme immobilization, although in literature the enzyme immobilization on woven nylon matrices is rarely reported. In this work, a protocol for a Trametes hirsuta laccase immobilization using woven polyamide 6,6 (nylon) was developed. A 24 full factorial design was used to study the influence of pH, spacer (1,6-hexanediamine), enzyme and crosslinker concentration on the efficiency of immobilization. The factors enzyme dosage and spacer seem to have played a critical role in the immobilization of laccase onto nylon support. Under optimized working conditions (29 U mL−1 of laccase, 10% of glutaraldehyde, pH = 5.5, with the presence of the spacer), the half-life time attained was about 78 h (18% higher than that of free enzyme), the protein retention was 30% and the immobilization yield was 2%. The immobilized laccase has potential for application in the continuous decolourization of textile effluents, where it can be applied into a membrane reactor.  相似文献

Paramecia are an excellent model system for studying the mechanisms involved in sensory transductions and intracellular Ca2+ regulation. These cells have two functionally distinct plasma membrane domains, body and cilia. The body plasma membrane is responsible for transduction of sensory stimuli into receptor potentials and the ciliary membrane is required for Ca2+ action potentials. Although ciliary membrane vesicles (cmv) have been purified and well characterized, body plasma membranes have not. We have generated body plasma membrane vesicles (bmv) by homogenization of deciliated cells and purified them from the microsome fraction by a two-phase aqueous polymer separation. The major criteria for purity of the bmv fraction are: (i) It is enriched 15-fold for a known plasma membrane marker (immobilization antigen) while the marker activities for other membranes were all decreased. The protein banding pattern of bmv is generally similar to cmv on SDS-PAGE. (ii) It contains a vanadate-sensitive Ca2+-ATPase activity that has been suggested to be a plasma membrane Ca2+ pump. The specific activity of this bmv Ca2+-ATPase is increased 4-fold over that of the homogenate. (iii) The phospholipid, fatty acid, and sterol composition of the bmv fraction are indicative of plasma membranes because they are qualitatively similar to cmv. The bmv also contains a membrane-bound NADPH-dependent cytochrome c reductase activity, suggesting that it may play a role in body plasma membrane function. This purified bmv preparation is useful for studying the role of the body plasma membrane in Ca2+ regulation, sensory transduction, protein and lipid trafficking, and plasma membrane fusion events.  相似文献

The aim of this study was to investigate the efficacy of two protocols for field immobilization of white-eared opossums (Didelphis albiventris) and compare their effects on immobilization, cardiopulmonary variables, and recovery times. Twenty one opossums were randomly divided into two groups; G1 received ketamine (15 mg kg?1)-dexmedetomidine (0.15 mg kg?1) intramuscularly (IM) and G2 received the ketamine-dexmedetomidine combination and isoflurane once induction was achieved. Oxygen was delivered by face mask (1.5 L minute?1). Thirty minutes after induction, isoflurane was discontinued (G2) and both groups were administered atipamezole (1.5 mg kg?1) IM. Respiratory (?R) and heart rate (HR), oxyhemoglobin saturation (SpO2), and rectal temperature (T) were recorded every 5 min. Induction time, time to first movement (RT1), and time to achieve standing (RT2) were recorded. ANOVA and non-parametric tests were used. Level of immobilization was assessed by observation of movements and evaluation of muscle relaxation. The mean induction time was 4.71 min. RT1 and RT2 were significantly longer in G2. No significant differences were found in SpO2 or ?R. HR did not vary significantly along time, but was higher in G2. Rectal temperature did not show differences between treatments, but decreased significantly with time in G2. Four of nine animals in G1 showed movements, while no animals in G2 did and muscle relaxation was determined to be better in this latter group. Both protocols were adequate for short-term field immobilization, with minimal alterations of HR and T and relatively short recovery times. Isoflurane provided better immobilization with statistically significant prolongation of recovery times.  相似文献

In many forests of Europe and north-eastern North America elevated N deposition has opened the forest N cycle, resulting in NO3 ? leaching. On the other hand, despite this elevated N deposition, the dominant fate of NO3 ? and NH4 + in some of these forests is biotic or abiotic immobilization in the soil organic matter pool, preventing N losses. The environmental properties controlling mineral N immobilization and the variation and extent of mineral N immobilization in forest soils are not yet fully understood. In this study we investigated a temperate mixed deciduous forest, which is subjected to an average N deposition of 36.5 kg N ha?1 yr?1, but at the same time shows low NO3 ? concentrations in the groundwater. The aim of this study was to investigate whether the turnover rate of the mineral N pool could explain these low N leaching losses. A laboratory 15N pool dilution experiment was conducted to study gross and net N mineralization and nitrification and mineral N immobilization in the organic and uppermost (0–10 cm) mineral layer of the forest soil. Two locations, one at the forest edge (GE) and another one 145 m inside the forest (GF1), were selected. In the organic layers of GE and GF1, the gross N mineralization averaged 10.9 and 11.1 mg N kg?1 d?1, the net N mineralization averaged 6.1 and 6.8 mg N kg?1 d?1 and NH4 + immobilization rates averaged 3.8 and 3.6 mg N kg?1 d?1. In the organic layer of GE and GF1, the average gross nitrification was 3.8 and 4.6 mg N kg?1 d?1, the average net nitrification was ?25.2 and ?31.3 mg N kg?1 d?1 and the NO3 ? immobilization rates averaged 29.0 and 35.9 mg N kg?1 d?1. For the mineral (0–10 cm) layer the same trend could be observed, but the N transformation rates were much lower for the NH4 + pool and not significantly different from zero for the NO3 ? pool. Except for the turnover of the NH4 + pool in the mineral layer, no significant differences were observed between location GE and GF1. The ratio of NH4 + immobilization to gross N mineralization, gross N mineralization to gross nitrification, and NO3 ? immobilisation to gross nitrification led to the following observations. The NH4 + pool of the forest soil was controlled by N mineralization and NO3 ? immobilization was importantly controlling the forest NO3 ? pool. Therefore it was concluded that this process is most probably responsible for the limited NO3 ? leaching from the forest ecosystem, despite the chronically high N deposition rates.  相似文献

Hyperthermia is described as the major cause of morbidity and mortality associated with capture, immobilization and restraint of wild animals. Therefore, accurately determining the core body temperature of wild animals during capture is crucial for monitoring hyperthermia and the efficacy of cooling procedures. We investigated if microchip thermometry can accurately reflect core body temperature changes during capture and cooling interventions in the springbok (Antidorcas marsupialis), a medium-sized antelope. Subcutaneous temperature measured with a temperature-sensitive microchip was a weak predictor of core body temperature measured by temperature-sensitive data loggers in the abdominal cavity (R2=0.32, bias >2 °C). Temperature-sensitive microchips in the gluteus muscle, however, provided an accurate estimate of core body temperature (R2=0.76, bias=0.012 °C). Microchips inserted into muscle therefore provide a convenient and accurate method to measure body temperature continuously in captured antelope, allowing detection of hyperthermia and the efficacy of cooling procedures.  相似文献

The C mineralization and N transformations during the decomposition of sunflower stalks (Helianthus annuus L.) and wheat straw (Triticum aestivum L.) with and without addition of (NH4)2SO4 (27.53 atom% 15N) were studied in a Vertisol. Soil samples were incubated under aerobic conditions for 224 days at 22 °C. The plant residues were added at a rate of 5.2 g kg-1 soil. Nitrogen was applied at a rate of 50.7 mg N kg-1 soil. Carbon dioxide emission and inorganic N content in soil were periodically determined. Gross N immobilization and remineralization were calculated on the basis of the isotopic dilution technique. At the end of the incubation period a 15N balance was established. Respectively, 68 and 45% of the applied residue-C mineralized from the sunflower stalks and wheat straw after 224 days. Both crop residues caused losses of up to 25% of added 15N after 224 days of incubation. These 15N losses were about three times larger than in the control soil, and were probably due to denitrification. The net immobilization of soil derived N following residue incorporation was largest in the case of wheat straw, depleting all soil inorganic N. In the wheat straw treatment with added (NH4)2SO4 soil inorganic N remained available, resulting in an enhanced initial C mineralization and N immobilization compared to the treatment without added N. In the case of the sunflower stalks, the high inorganic N content of the stalks suppressed the effects of N addition on C mineralization and N immobilization/mineralization. Gross N immobilization amounted to 31.9 and 28.2 mg N g-1 added C after 14 days for wheat straw and sunflower stalks, respectively. At the end of the incubation, about 35% of the newly immobilized N was remineralized in both plant residue treatments. Gross N immobilization plotted against decomposed C suggests that fairly uniform C-N relationships exist during the decomposition of divers C substrates. The results demonstrate that low fertilizer N use efficiencies may be expected in a wheat-sunflower cropping system with incorporation of crop residues, as the fertilizer N applied becomes largely immobilized in the soil organic fraction. Cardiac output and stroke volume were estimated for a 200 g largemouth blackbass (Micropterus salmoides) by a modified whole-body thermodilution method using the relation between thermal equilibration rates and heartbeat frequencies. The reciprocal of the thermal time constant, k (min–1), was related to the heartbeat frequency, F (beats min–1), by the equation k=0.00146 F + 0.309; the slope is the weight-specific stroke volume (ml g–1) and the intercept is the weight-specific heat transfer constant (cal °C–1 min–1 g–1). Stroke volume was 0.292 ml (0.00146 ml/g body weight), yielding cardiac output values ranging from 44 ml kg–1 min–1 (at 30 beats min) to 158 ml kg–1 min–1 (at 108 beats min–1), or 4.4 to 15.8% of body weight. Active (convective) heat transfer due to blood flow constituted an estimated 11 to 34% (mean 22.5%) of total heat transfer, depending on heartbeat frequency; this variability constitutes physiological thermoregulation.  相似文献

We report the effect of random and oriented immobilization of Rhizomucor miehei lipase (RML) on its functional properties. For this purpose, silica nanoparticles (MCM-41 and SBA-15) were prepared, characterized and functionalized by glycidyloxypropyl trimethoxysilane. Direct immobilization of RML on these supports was performed via the variety of amino acid residues on the surface of RML which promotes random immobilization. To perform oriented immobilization, partial modification of epoxy functionalized supports was carried out by introducing iminodiacetic acid groups followed by addition of Cu2+. In this way, immobilization is mainly directed via the most accessible histidine group, followed by intramolecular reaction of the other nucleophilic residues of the enzyme and the remaining epoxy groups on the support. The results showed higher thermal stability for immobilized derivatives compared to the soluble enzyme. Co-solvent stability of the derivatives was also studied in presence of six polar organic solvents (DMSO, THF, acetonitrile, 1-propanol, 2-propanol and dioxane). Influence of the immobilization procedure on activity and selectivity of the immobilized preparations was studied in selective hydrolysis of fish oil. All the derivatives discriminate between cis-5,8,11,14,17-eicosapentaenoic acid (EPA) and cis-4,7,10,13,16,19-docosahexaenoic acid (DHA) in favor of EPA. Remarkable improvement in selectivity was obtained using oriented immobilization of RML.  相似文献
