The effects of malting conditions on proteolytic activities in three improved sorghum varieties: ICSV400, SK5912 and KSV8 were studied. Grains were steeped for 45 h using 6 h wet and 3 h dry cycles and germinated for 8 days. Moisture contents and their effects on proteolytic activity were monitored at various intervals using standard methods. Significant positive correlations existed between moisture content and carboxypeptidase and proteinase activities during steeping. Optimum carboxypeptidase and proteinase enzyme activities were consistently recorded in both ICSV400 and SK5912 at the 40th h of steeping while those of KSV8 occurred on the 45th h. This suggests that protein hydrolysis of KSV8 is different from that of ICSV400 and SK5912. Similarly, optimum proteolytic activities occurred in all the sorghum varieties on the 5th day of germination suggesting optimum germination time for optimum protein modification. Variety and steeping time affected both carboxypeptidase and proteinase activities significantly (P<0.001) during germination. Significant positive correlations existed in the sorghum varieties ICSV400 and SK5912 between free amino nitrogen (FAN) contents and carboxypeptidase activities during both steeping and germination. In contrast, variety KSV8 showed no significant correlation between FAN contents and carboxypeptidase activities during either steeping or germination. On the other hand, while there was a highly significant positive correlation between CWS-P development and proteinase activities in all the sorghum varieties during steeping, no such relationship existed during germination.  相似文献

Summary Three sorghum genotypes, L.187, SK5912 and RCFA×L.187 (the latter being a hybrid) were field grown under four nitrogen application rates and replicated four times. The experiments were of complete randomized plot design and conducted in 1976, 1977 and 1978.Nitrate Reductase Activity (NRA) was measured at 5, 7, 9, 11, 13, and 15 weeks and at 18, 19, 20 and 21 weeks after planting in 4th vegetative leaf and flag leaf respectively. Flag leaf, 4th leaf and grain protein contents were also measured.Nitrogen application generally increased NRA in both 4th leaf and flag leaf in the 3 genotypes at all the sampling dates.NRA of the hybrid, RCFA×L.187 was consistently higher than those of SK5912 and L.187 (both straight varieties).NRA of 4th leaf was negatively correlated with leaf protein but flag leaf protein tended to increase with applied nitrogen.Flag leaf NRA was positively and significantly correlated with grain protein indicating an important part played by the flag leaf relative to protein accumulation of the developing grain.  相似文献

Sorghum midge, Contarinia sorghicola Coq. (Diptera: Cecidomyiidae) is an important pest of grain sorghum, and host-plant resistance is one of the most effective means of controlling this pest. We studied the antibiosis mechanism of resistance in sorghum to C. sorghicola in a diverse array of midge-resistant and midge-susceptible genotypes. Data were recorded on adult emergence, postembryonic developmental period, number of mature eggs in the ovary, fecundity, larval survival from artificially implanted eggs; and the tannins, soluble sugars, and protein content of 10-day old and mature grains during the 1982-91 rainy and post-rainy seasons. Adult emergence was significantly lower in the midge-resistant genotypes compared with the susceptible controls. Initiation of adult emergence was delayed by 4–8 days on DJ 6514, IS 8571, IS 9807, IS 10712, IS 19474, IS 19512, ICSV 830 and ICSV 197. Postembryonic developmental period was prolonged on DJ 6514, IS 15107, IS 3461, IS 7005, IS 19474, ICSV 831 and ICSV 197. However, the delay in adult emergence or the extended developmental period was not observed during the post-rainy season in some genotypes. These differences in the expression of antibiosis to midge in resistant genotypes over seasons may be attributed to the effect of environmental conditions on the insect development and chemical composition of sorghum grain. Amounts of tannins and proteins were generally greater in the midge-resistant lines compared with the susceptible ones (except tannins in DJ 6514) while the soluble sugars were low in the midge-resistant lines (except TAM 2566). These differences in chemical composition of the grain between genotypes and variations over seasons have been discussed in relation to the expression of antibiosis mechanism of resistance to the sorghum midge. Antibiosis to sorghum midge was also evident in terms of smaller size of larvae, lower number of eggs in the ovary, reduced fecundity, and larval survival. Midge-resistant lines have diverse effects on the biology of this insect. Antibiosis along with other components of resistance can be used to develop cultivars with stable resistance to C. sorghicola.  相似文献

Forty varieties of vegetable, cereal, fodder and grass seeds at 10, 13 and 16% moisture content were fumigated with methyl bromide at 10 and 18oC, to determine the maximum safe moisture content required to avoid unacceptable phytotoxicity. Dosages (concentration x time products) of 200 and 400 mg h/1 of technical and commercial methyl bromide were used and treated seeds were compared with controls in germination tests. Results were not consistent, but in c. 30 % of the comparisons lower moisture contents, lower temperature and lower dosage each resulted in better germination than the corresponding higher values. The reductions in germination of treated samples were usually less than 5 % and the overall average reduction was only 2.3%. This work showed that under controlled conditions the treatments are safe for most of the tested varieties of seed, but for some seeds, particularly white clovers, Agrostis and runner beans, the highest moisture content (16%) should be avoided.  相似文献

The changes in crude protein, free amino acids, amino acid composition, protein solubility, protein fractionation and protein digestibility after germination of sorghum were investigated. Sorghum varieties (Dorado, Shandaweel-6, Giza-15) were soaked for 20 h followed by germination for 72 h; the results revealed that crude protein and free amino acids in raw sorghum varieties ranged from 10.62 to 12.46% and 0.66 to 1.03 mg/g, respectively. Shandaweel-6 was the highest variety in crude protein and free amino acids content. After germination, crude protein was decreased and free amino acids were increased. There was an increase in content of valine and phenylalanine amino acids after germination. On the other hand, there was a decrease in most of amino acids after germination. After germination protein solubility was significantly increased. Regarding protein fractions, there was an increase in albumin, globulin and kafirin proteins and a decrease in cross linked kafirin and cross linked glutelin after germination.  相似文献

Superoxide dismutase (SOD) activity in the white sorghum farafara and ICRISAT sorghum variety – ICSV 400 was at a high activity in the embryo at about 8 SOD units/tissue. This activity was almost completely destroyed at 80 °C. Totox index of the brewing grains were 366 for ICSV 400, 312 for farafara, 112 for maize grits and 90 for barley malt. Worts from sorghum/maize and sorghum/barley malt brews all had hydroperoxy linoleic acid (15–19 μM) which remained undetected after wort boiling. Sorghum/maize brews formed very little trub (wort proteinous sediments) in the whirlpool and trub increased in sorghum/barley brews with increased usage of barley malt. Sorghum/maize brews had free fatty acids (FFA) at 22 mg/l in pitching wort but in sorghum/barley brew (50/50) only 9 mg/l. Objective

To enhance the production of phenolic compounds during barley germination using chitooligosaccharide as an elicitor to improve the antioxidant capacity of malt.

Results

When used as an elicitor for barley germination, chitooligosaccharide with a molecular weight of 3 kDa, added at 10 mg/kg barley kernels during the first steeping cycle, led to the maximum production of phenolic compounds. Compared with the control with no chitooligosaccharide added to the steeping water, the total phenolic content was increased by 54.8%. Increases in the total phenolic content of the barley malt occurred when chitooligosaccharide was applied during the first or both the first and the second steeping cycles. Thus the antioxidant capacity of barley malt was increased significantly by adding chitooligosaccharide during the steeping process.

Conclusion

Applying chitooligosaccharides during the steeping process increased the content of phenolic compounds thus improving the antioxidant capacity of the barley malt.

Sorghum grain (two varieties) was modified to different water contents (12% to 16% wet weight basis) and heated to 60°C, 70°C and 80°C for periods of 4, 8 and 12 min. Germination, seedling vigour, seedling dry matter, free fatty acid (FFA) content, fungal contamination and infestation with the insect pest Rhyzopertha dominica (F.) were all markedly affected by heat treatment. The effectiveness of the heat treatment was also influenced by the size of the sample used. Heat treatment at 60°C and 70°C resulted in germination being unaffected or stimulated while at 80°C and the higher water contents significantly reduced, when compared to untreated controls. The dry matter of seedlings, and seedling vigour was positively correlated with germination and heat treatment. Heat treatment had no effect on FFA. All stages of the insect pest, R. dominica, were destroyed at 70°C and an 8 min exposure time. However, the water content of the sorghum was critical in determining the efficacy against this pest. The percentage fungal contamination of grain was reduced from 90% to about 25% by heat treatment. However, some grain fungi, particularly Eurotium spp., Aspergillus niger and Penicillium spp. could still be isolated from sorghum grain treated at 80°C for up to 12 minutes.  相似文献

Objectives

To study enhanced barley germination by chitooligosaccharide as an elicitor for improving the quality of malt.

Results

Barley germination for both radical and leaf sprouts was enhanced when chitooligosaccharide was added to the steeping water in the first steeping cycle. The activities of hydrolases (α-/β-amylase, proteinase and β-glucanase) and antioxidases (superoxide dismutase and catalase) in the resultant malt were increased in a dose-dependent manner when chitooligosaccharide was supplemented in the steeping water. Maximal promotion was at 1 mg chitooligosaccharide/l for α-/β-amylase and proteinase, and at 10 mg/l for β-glucanase, superoxide dismutase and catalase. Malt quality, including free α-amino nitrogen content, Kolbach index, malt extract content, diastatic power, wort viscosity and the ratio of glucose, maltose and maltotriose, was significantly improved by chitooligosaccharide in seed priming at 1 mg/l.

Conclusion

Application of chitooligosaccharide in the steeping water promotes barley germination and improves the quality of malt.

Large differences in the leachate conductivities of single seeds after 24 h soaking were observed in five Brussels sprout cultivars. Within each cultivar non-germinable seeds tended to have higher conductivities than germinable seeds, but the conductivity above which seeds failed to germinate varied between cultivars. No conductivity reading or partition value could therefore be considered as an absolute boundary between germinable and non-germinable seeds to allow the prediction of seed germination for all seed lots of the same species. However, the mean conductivity for each sample was significantly correlated with germination (r = 0.83, P < 0.05) and was clearly higher for samples with lower germinations. Differences in the partition values for the five cultivars may have been attributable to the natural age of the seeds. Seeds of Asmer Aries artificially aged rapidly at 20% moisture content for up to 48 h at 45° C showed increasing leachate conductivity with increased time of ageing (r = 0.97, P < 0.01) and decreased germination (r = - 0.88, P < 0.05). Again however the values for germinable and non-germinable seeds of different artificial ages varied. Leakage from seeds was limited by the testa in the first 6 h of imbibition. Solute leakage from embryos of the five cultivars was therefore greater than those for intact seeds but a clear separation of germinable and non-germinable seeds was still not possible from the conductivity readings. The implications of the observed increases in conductivity with increased seed age and decreased germination for the development of a rapid vigour test for small seeded vegetables in conjunction with controlled deterioration are discussed.  相似文献

During steeping and germination in micromalting equipment, the number of colonies of filamentous fungi increased from two to five times, colonies of yeasts from five to ten times, and bacteria from 50 to more than 100 times the numbers present in the grain before malting. Kiln drying according to a commercial schedule reduced the number of all types of microflora below the number present before kilning, but all were present in larger numbers in the kilned malt than in the original grain. In barley stored at room temperature and at a moisture content of 15 to 18%, members of the Aspergillus glaucus group increased with increasing time and increasing moisture content, and germination percentage of the seeds decreased. Stored free of storage fungi at room temperature, barley with a moisture content just over 15% retained a high germination percentage for 5 months, but at a moisture content of 16% the germination decreased to zero.  相似文献

Amount of certain amino acids required by herbivores, and their availability in host plants are of crucial importance for insect growth, development, and life processes. Therefore, we carried out profiling and quantification of 17 amino acids in diverse sorghum genotypes, and on the Chilo partellus larvae reared on them, to understand the association and contribution of different amino acids in plant resistance to insects. Sorghum germplasm lines IS 2205 and IS 2123 had severe detrimental effects on the development and survival of C. partellus followed by varieties ICSV 700 and ICSV 708 in comparison to susceptible check, Swarna. Profiling of sorghum seedlings, seeds, and the C. partellus larvae fed on these genotypes for 17 amino acids revealed that Arginine, Glycine, Isoleucine, Leucine, Proline, and Valine in sorghum seedlings and the C. partellus larvae had significant and positive association, suggesting their role in the development and survival, while negative association of Cystine indicated its contribution in plant defense. Furthermore, C. partellus acquired less of cyclic and aliphatic amino acids per unit amount from the test resistant genotypes, while more from the susceptible check, than their presence in the seedlings. Present studies suggest that Alanine, Cystine, Glycine, and Proline contents in C. partellus larvae; Cystine and Proline contents in sorghum seedlings; and Methionine content in sorghum seeds, have significant and negative association, and contribute to explain?>93% and >96%, respectively, of the variability in antibiosis mechanism and overall resistance to C. partellus. These studies have implications for antibiosis and nutritional mechanism of host plant–insect interactions in sorghum against C. partellus.  相似文献

Twelve varieties of improved and local (landrace) sorghum were tested under artificial inoculation conditions. Out of 12 sorghum varieties evaluated with covered kernel smut, notably local cultivar ‘Tetron’ was found to be highly resistant. Disease incidence and severity on the rest of the cultivars varied from 21 to 47% and 40 to 53% respectively. Differences among cultivars in yield loss were also observed. The highest yield loss (40%) was recorded on 97 MW 6129 (NVT-11 4) and for Tetron yield loss was nil. The effect of disease on germination was noticed but cannot be considered as conclusive due to moisture stress at the time of germination.  相似文献

The changes in phytate, phytase activity and in vitro bioavailability of iron and zinc during soaking and germination of three white sorghum varieties (Sorghum bicolor L. Moench), named Dorado, Shandweel-6, and Giza-15 were investigated. Sorghum varieties were soaked for 20 h and germinated for 72 h after soaking for 20 h to reduce phytate content and increase iron and zinc in vitro bioavailability. The results revealed that iron and zinc content was significantly reduced from 28.16 to 32.16% and 13.78 to 26.69% for soaking treatment and 38.43 to 39.18% and 21.80 to 31.27% for germination treatments, respectively. Phytate content was significantly reduced from 23.59 to 32.40% for soaking treatment and 24.92 to 35.27% for germination treatments, respectively. Phytase enzymes will be activated during drying in equal form in all varieties. The results proved that the main distinct point is the change of phytase activity as well as specific activity during different treatment which showed no significant differences between the varieties used. The in vitro bioavailability of iron and zinc were significantly improved as a result of soaking and germination treatments.  相似文献

[背景] 在白酒发酵过程中,原料中的谷物蛋白可为微生物的生长提供氮源等营养物质,进而形成多种代谢产物。谷物蛋白可分为清蛋白、球蛋白、醇溶蛋白和谷蛋白。然而,谷物蛋白对微生物多样性及其代谢产物多样性的调控尚不明确。[目的] 揭示白酒发酵过程中与微生物多样性及其代谢产物多样性显著相关的关键谷物蛋白种类及其调控作用。[方法] 通过Osborne法测定不同品种高粱中谷物蛋白的组成;采用多组学联用技术解析4种高粱在发酵过程中的微生物菌群多样性及代谢产物多样性;通过模拟发酵揭示原料中影响微生物群落及其代谢多样性的关键蛋白。[结果] 4种高粱中的谷物蛋白组成存在显著差异(ANOSIM:R=0.85,P=0.001);4种高粱在发酵第5天时,S4高粱的细菌多样性显著(P<0.05)高于其他3种高粱,S3高粱中微生物的代谢产物多样性显著(P<0.05)高于其他3种高粱;清蛋白和球蛋白含量与发酵第5天的优势细菌多样性(R2=0.34,P<0.05;R2=0.58,P<0.05)和代谢产物多样性呈显著正相关(R2=0.58,P<0.05;R2=0.36,P<0.05),被定义为关键蛋白;模拟发酵实验验证了优势细菌多样性和代谢产物多样性可随着2种关键蛋白即清蛋白和球蛋白含量的升高而升高。当清蛋白含量在3.0 g/L时,优势细菌多样性及代谢产物多样性可分别达到0.72和0.65;当球蛋白含量在3.0 g/L时,优势细菌多样性及代谢产物多样性可分别达到0.66和0.81。[结论] 研究揭示了酿造原料中的清蛋白和球蛋白对发酵过程中细菌多样性及代谢产物多样性的调控作用,为提高白酒发酵的可控性及质量提供了依据。  相似文献

Canopy CO2-exchange rates (CER), air temperatures, and dew points were measured throughout ten days during the 1987 growing season for cotton (Gossypium hirsutum L.), grain sorghum [Sorghum bicolor (L) Moench], and five soybean [Glycine max (L) Merr.] cultivars, and throughout seven days in 1988, on maize (Zea maize L.). The objective was to determine if the decline in CER per unit light during the afternoon is associated with a vapor pressure deficit (VPD) increase. Some of the soybean and maize plots were kept as dry as possible. A VPD term significantly contributed (P0.05) to a canopy CER regression model in 54 of 80 data sets in 1987. Grain sorghum was less sensitive than the well-watered soybean genotypes to an increasing VPD (P0.05) on three of the ten measurement days and less sensitive than cotton (P0.05) on only one day. Cotton demonstrated less VPD sensitivity than soybean (P0.05) on one day. The moisture stressed soybean plots showed a greater CER sensitivity to VPD (P0.05) than the well-watered soybean plots. In 1988, the frequently irrigated maize plots were less sensitive to VPD (P0.05) than the rain-fed plots early in the season, before the rain-fed plots were excessively damaged by moisture stress. These results indicate that the afternoon declines in canopy CER found in a number of different species are associated with increases in the VPD; recent work of others suggests that this may be due to partial stomatal closure.Abbreviations CER carbon dioxide exchange rate - VPD vapor pressure deficit - PPFD photosynthetic photon flux density - DAP days after planning  相似文献

Eleven-kilogram parcels of 2-row and 6-row barley initially at 18% moisture content were implanted in dry bulk oats in a farm granary in Manitoba for 60 weeks between August 1983 and October 1984. Temperature, moisture content, O2 and CO2 levels, fat acidity values, seed germination, microfloral incidence and abundance and the presence of major mycotoxins (aflatoxins, sterigmatocystin, ochratoxin A, citrinin, penicillinic acid, patulin) were monitored. Ochratoxin A reached maximum levels of 0.97 ppm by week 24 in the 6-row barley, and 0.05 ppm by week 28 in the 2-row; no other mycotoxins were detected. The effect of cultivar type was significant (P<0.01) with greater effects in the 6-row barley for the following parameters: fat acidity value, germination, incidence of Alternaria alternata, Penicillium spp. and Helminthosporium sativum, total fungal propagule count and ochratoxin A levels. The effect of time was significant (P<0.05) for all variables except oxygen, carbon dioxide, Aspergillus versicolor, and total fungal propagule count. Sorghum shoot fly, Atherigona soccata is one of the serious constraints to sorghum production, and host plant resistance is an important component for controlling this pest. We studied the expression of resistance to A. soccata in a diverse array of sorghum genotypes in relation to composition of leaf surface chemicals during the seedling stage. The sorghum genotypes IS 1054, IS 1057, IS 2146, IS 4664, IS 2312, IS 2205, SFCR 125, SFCR 151, ICSV 700, and IS 18551 exhibited antixenosis for oviposition, and suffered less deadhearts due to sorghum shoot fly, A. soccata. Compounds undecane 5- methyl, decane 4- methyl, hexane 2, 4- methyl, pentadecane 8- hexyl, and dodecane 2, 6, 11- trimethyl, present on the leaf surface of sorghum seedlings, were associated with susceptibility to shoot fly; while 4, 4- dimethyl cyclooctene was associated with resistance to shoot fly. The compounds associated with resistance/susceptibility to shoot fly, can be used as marker traits to select for resistance as well as for diversifying and increasing the levels of resistance to this pest. The role of biochemical compounds for developing sorghum varieties with resistance to shoot fly, A. soccata has been discussed.  相似文献

The increase in salinity of the medium resulted in the decrease α-amylase and protease activities in all cotton varieties tested, however it was more pronounced in NIAB-86. Decrease in concentration of reducing and non-reducing sugars, slower mobilization of reserve protein and reduced amino acidslevels were observed with increase in salinity levels. However, varieties K-115 showed better performance than others. The variety K-115 also had a capacity to mobilization and had higher levels of sugars, total free amino acids and reserve protein during germination and early seedling growth stages. However, varieties K-115 showed better performance than others. Variety K-115 showed highest germination followed by NIAB-Karishma and NIAB-86. The variety K-115 also had a capacity to mobilization and had higher levels of total free amino acids and less reserve protein during germination and early seedling growth stages.  相似文献
