Summary The possibility of using polyurethane foam as a support for the immobilization ofZymomonas mobilis cells to carry out sucrose conversion to ethanol was investigated. Sucrose hydrolysis efficiencies of 90% and higher, volumetric reactor productivity of 20 gL–1h–1 and final ethanol concentration of 6.3% (v/v) at a dilution rate of 0.4 h–1 show the good performance of polyurethane foams for whole cell immobilization.  相似文献

Summary Columnar reactors containing immobilized cells of Zymomonas mobilis were utilized for the continuous production of ethanol from glucose. Two different immobilization strategies were investigated. In one case, cells were entrapped in borosilicate glass fiber pads, while in the other, cells were immobilized via flocculation. The reactors were operated in both the fixed-bed and expanded-bed manner. Ethanol productivities as high as 132 g/l·h were achieved. Data obtained from studies employing 5.0 and 10.0% glucose concentrations are presented. Summary Immobilization of Saccharomyces cerevisiae as a monocellular layer adhering to a glass support has been achieved by adsorbing Al or Fe(III) ions on the cells or by coating the support with Al(OH)3 or Fe2O3 particles. The initial rate of glucose to ethanol conversion was measured in pure glucose solutions for free cells in suspension, either untreated or submitted to the chemical treatments used for immobilization, and for immobilized cells, using a small reactor (50–70 ml) in closed circuit. The conversion rate of immobilized cells is in the range of 3.5 to 5×10-14 mol/cell·h while that of untreated suspended cells is in the range of 5 to 7×10-14 mol/cell·h.  相似文献

Summary A broad pH range of 4.5–7.5 for maximum ethanol productivity and ethanol yield was observed with a passively immobilizedZ. mobilis system. Total retained biomass (as suspended flocs and entrapped cells) was >50 g/l for medium pH values between 4.0–8.0. The entrapped cells to suspended flocs ratio was highest at pH 4.0, whereas at pH above 5.2 it was close to 1.0. The observed enhancement of cell immobilization on the packing support at low pH seemed to be related to formation of bacterial filaments.  相似文献

Summary A laboratory-scale, two-stage continuous process for the production of curdlan-type exopolysaccharide has been operated in steady-state for 500hr. Two continuous flow, constant volume fermenters are connected in series. A stable, curdlan-producing strain of Alcaligenes faecalis var myxogenes ATCC 31749 is grown aerobically in a nitrogen-limited chemostat operating near Dmax at 0.24 hr–1. The effluent is introduced directly into a second larger constant volume fermenter which is being simultaneously fed a glucose solution at a fixed rate. Under sub-optimal conditions associated with curdlan production, the observed Qp was 0.05 g curdlan/g cell/hr. At a biomass level of 4 g/L in the second stage, curdlan was present at 10 g DW/L and the volumetric productivity was 0.2 g/g cell/hr. The substrate (glucose) conversion efficiency was 42%. The process is described in patents applied for on behalf of George Weston Ltd. (Toronto, Canada).  相似文献

Summary Thermophilic degradation of sugar beet pulp was studied in batch cultures at 55°C by different associations of bacteria, includingClostridium thermocellum,Methanobacterium sp. andMethanosarcina MP.C. thermocellum produced acetate, succinate, methanol, ethanol, H2 and CO2. The coculture ofC. thermocellum andMethanobacterium sp. produced trace amounts of ethanol and succinate; acetate concentration was about three times higher than in theC. thermocellum monoculture. Summary Mead was produced by immobilized cells of Hansenula anomala in calcium alginate gels. The immobilized cell beads of 3 mm diameter packed in column reactors of dimensions 2.2x60, 4x40 and 8x80 cm, produced mead containing maximum concentrations of ethanol and ethyl acetate of 70 g/l and 730 mg/l, respectively at a dilution rate of 0.1 h–1. The maximum alcohol productivity achieved was 23.1 g/l·h at a dilution rate of 0.33 h–1. With intermittent regenerations of the cells the reactor operated continuously for 110 days. This process enables the quick production of matured mead by a single culture and the elimination of the traditionally used long aging periods.  相似文献

Summary An immobilization technique has been developed for the conversion of both cellobiose and xylose to ethanol, which may be considered as one stage of a process for the conversion of cellulosic biomass to ethanol. Relatively inexpensive charcoal was used as a support material, with 23 mg dry weight of Clostridium saccharolyticum cells per g dry weight of support. Tests were run for 170 h at 0.15 1/h dilution rate. From a 3% (w/v) sugar mixture, 0.7% (w/v) ethanol was obtained with over 97% cellobiose and 62% xylose utilization.  相似文献

Summary The specific ethanol productivity withSaccharomyces cerevisiae grown aerobically in a chemostat at a growth rate of 0.17 hr–1 was found to increase from zero to 13 mmol/g cell dry matter·h when the potassium content in the substrate used was decreased to 0.05 mol/kg glucose. 78% of the glucose metabolized were converted to ethanol under these aerobic growth conditions.  相似文献

Candida shehatae NCL-3501 utilized glucose and xylose efficiently in batch cultures. The specific rate of ethanol production was higher with mixtures of glucose and xylose (0.64–0.83 g g–1 cells d–1) compared to that with individual sugars (0.38–0.58 g g–1 cells d–1). Although the optimum temperature for growth was 30°C, this strain grew and produced appreciable levels of ethanol at 45°C. A stable ethanol yield (0.40–0.43 g g–1 substrate utilized) was obtained between 10 g L–1 and 80 g L–1 of initial xylose concentration. Conversion efficiency was further improved by immobilization of the cells in calcium alginate beads. Free or immobilized cells ofC. shehatae NCL-3501 efficiently utilized sugars present in rice straw hemicellulose hydrolysate, prepared by two different methods, within 48 h. Ethanol yields of 0.45 g g–1 and 0.5 g g–1 from autohydrolysate, and 0.37 g g–1 from acid hydrolysate were produced by free and immobilized cells, respectively.  相似文献

Arachis hypogaea L. is a tropical crop that is slow-growing at temperatures below 25°C. Unadapted CO2-assimilation rate (A) showed insufficient variation between 15 and 30°C in the short term (hours) to explain this marked reduction in growth. However, at longer periods (12 d), A was depressed as were growth rate and leafproduction rate. To examine the possible relationship between growth, A and sink demand plants were transferred from 30°C, which is near the optimum for growth, to a suboptimal temperature (19°C). In the first 2 d of cooling, A decreased by 50–70%, the stomata stayed open, and the intercellular CO2 concentration (ci) rose, i.e. the decrease in A of the cooled plants was the result of non-stomatal factors. Changes in dark respiration did not account for the decline in A.Clear evidence was obtained of sink control of A by independently manipulating the temperature of different leaves on the plant. Cooling (to 19°C) most of the plant (the sink) led to a 70% decline in A of the remaining leaves at 30°C after 3 d, whereas the converse treatments (30°C sink, 19°C source) resulted in small changes (17%). In plants at 19°C which were exposed to low CO2 concentration to prevent photosynthesis, A was not reduced when measured at normal CO2 concentrations, indicating that carbohydrate accumulation was responsible for the decline in A. Summary The productivity of continuous ethanol fermentation has been increased using fixed bed reactors where a high density of yeast cells was maintained on a packing of wood chips. Two different systems have been used: 1. A tubular reactor which produced alcohol solutions containing up to 13.5% (V/V) ethanol. High CO2 retention and a poor mass transfer between bulk medium and immobilized biomass prevented production rates higher than 2.2 g/l·h. 2. A multistage reactor where a better utilisation of the reactor volume led to improved performances. Solutions containing 132 g/l of ethanol (16.5% V/V) were produced with a productivity increased up to 4.8 g/l·h. A better distribution of the active biomass and a lower gradient of alcohol concentration between support and bulk medium are possible reasons for this improvement.  相似文献

Summary Candida shehatae ATCC 22984 and Pichia stipitis CBS 5776 were tested for ethanol production from xylose, glucose-xylose mixtures, and aspen wood total hydrolysates. Adaptation of these yeasts to wood hydrolysate solutions by recycling resulted in improved substrate utilization and ethanol production. Compared to the non-adapted cultures, recycled C. shehatae and P. stipitis in aspen hydrolysate increased g ethanol/g sugar consumed from 0.39 and 0.41 to 0.45 and 0.47; while ethanol production from a 70:30 glucose-xylose solution (total sugars 140 g/L) was 45 g/L in 24 h and 60 g/L in 72 h with the adapted yeasts compared to 15 g/L and 28 g/L in the same times with the parent strains.  相似文献

Calli were obtained from Ginkgo biloba embryos grown on Murashige and Skoog (MS) medium. The G. biloba cells could grow on either MS or Gamborg B5 mineral salt medium supplemented with sucrose (3% and 2%, respectively) and naphthaleneacetic acid (NAA) and kinetin (K) in concentrations ranging from 0.1 to 2.0 mg·L–1. Best growth and maintenance of callus cultures were achieved using MS medium supplemented with 2 mg·L–1 NAA and 1 mg·L–1 K (N2K1MS). Light was required to maintain healthy growth of the callus tissue.In both MS and B5 based media, sucrose was hydrolyzed extracellularly before being taken up by Ginkgo cell suspension cultures. Specific growth rates of 0.13 d–1 and 0.08 d–1 were obtained in MS medium supplemented with 1 mg·L–1 NAA, 0.1 mg·L–1 K and 30 g·L–1 sucrose (N1K0.1MS) and B5 medium supplemented with the same growth regulator regime and 20 g·L–1 sucrose (N1K0.1B5) respectively. Complete phosphate and ammonium uptake was observed in 11 days when cultured in MS medium and 10 days and 4 days respectively when cultured in B5 medium. During the culture, G. biloba cells consumed only 64% and 29% of the nitrate content of N1K0.1MS and N1K0.1B5 media respectively. Maximum dry biomass concentrations were 13.4 g·L–1 and 7.9 g·L–1, and yields on carbohydrate were 0.39 and 0.45 in N1K0.1MS and N1K0.1B5 media respectively. The better performance of MS cultures came from the higher sucrose and nitrogen salts concentrations of this medium.Abbreviations B5 Gamborg mineral salt medium - d.w. Summary A computer-assisted on-line glucose analyzer was developed for feed-back control of cell growth. Using this system the glucose consumption rate for Escherichia coli was determined to be linear during batch culture at 0.37 g/hr. On-line feed-back control of glucose concentration at 1.5±0.5 g/L was used with fed-batch cultures to produce 31.2 g dry weight of E. coli cells/L in 12 h.  相似文献

Summary Nitrosoguanidine-induced, stable theromotolerant mutant (ZMI2) ofZymomonas mobilis ZM4 was found to possess almost normal cell morphology, and a better ethanol tolerance at 42°C than the parent strain (ZM4). Its kinetic parameters, in converting different concentrations of glucose to ethanol, were comparable to ZM4 at 30°C, and significantly superior at 42°C. In a 200 g/L glucose medium in a pH-stat (5.0) at 42°C, the mutant yielded more ethanol (71.0 g/L) (improved to 73.7 g/L at pH 5.5) and alcohol dehydrogenase (ADH) than the parent strain. A model has been developed to calculate the ethanol production in a well-mixed fluidized bed reactor. This model takes into account diffusion and the reaction inside porous glass beads and the activity of suspended cells in the fluidized bed reactor. The associated model parameters have been determined from the literature and by kinetic studies with Zymomonas mobilis in a continuous stirred tank reactor. The model permits good predictions of steady-state data in a fluidized bed reactor at residence times longer than 1–1.5 h. The immobilization of Z. mobilis in a fluidized bed reactor results in high ethanol space-time yields of more than 50 g·–1·h–1 at a glucose conversion of 80% (glucose in substrate: 120 gl–1). At 99% conversion a space-time yield of 30 g·–1·–1 can be achieved when two fluidized bed reactors operate as cascade.  相似文献

Summary Wastewater from cotton yarn and fabric finishing was successfully treated in an upflow anaerobic filter at 35°C up to a COD loading of 1 Kgr COD/m3 · day; the COD removal varied from 50 to 90% and production of biogas was 0,2–0.4 L/g influent COD, having 70–80% CH4.
