Summary We examined digestibility of dry matter, nutrients, and fiber, and food intake, metabolic fecal losses, weight change, and gut size of pocket gophers (Thomomys bottae) in relation to diet quality in the laboratory. Pocket gophers were maintained for 15–20 days on one of seven diets which contained from 18% to 56% neutral detergent fiber (NDF). NDF content of the diet was an excellent predictor of diet quality. Digestibility of dry matter, NDF, and nitrogen all decreased with increasing NDF content of the diet. In general, pocket gophers compensated for low diet quality by increasing dry matter intake, but those given high quality forage before the lowest quality diet reduced their intake. Thus, the response of pocket gophers to low quality diets may depend on their body condition. Because increased food intake resulted in increased total metabolic fecal losses and metabolic fecal nitrogen losses, decreasing food intake on low-quality diets may be advantageous. A further response of pocket gophers to decreased food quality was an increase in size of cecum and large intestine, suggesting that fermentation of cell walls became increasingly important as diet quality decreased.  相似文献

Inhibitors of hydroxymethylglutaryl-CoA reductase or statins have been shown to alleviate endothelial dysfunction. Their effects on constitutive nitric oxide synthase in the central nervous system may hypothetically affect the autonomic balance in sympathoexcitatory states, such as chronic heart failure (CHF). To address this issue, simvastatin (SIM) (0.3, 1.5, or 3 mg. kg-1. day-1 po) was given to rabbits with pacing-induced CHF over a 3-wk period. Normal and CHF vehicle-treated rabbits served as controls. Autonomic balance was assessed by measuring heart rate variability, including power spectral analysis (PSA). In addition, changes in resting heart rate were assessed before and after vagal and sympathetic autonomic blockade by atropine and metoprolol, respectively. The SD for all intervals was 8.9 +/- 0.7 ms in normal, 4.9 +/- 0.6 ms in CHF (P < 0.01), 3.8 +/- 0.6 ms in CHF with 0.3 mg. kg-1. day-1 SIM (P < 0.001), 5.7 +/- 0.9 in CHF with 1.5 mg. kg-1. day-1 SIM (P < 0.05), and 7.2 +/- 0.5 in CHF with 3.0 mg. kg-1. day-1 SIM. Similarly, total power was 40.5 +/- 6.3 ms2 in normal, 10.1 +/- 3.0 ms2 in CHF (P < 0.01), 6.0 +/- 1.6 ms2 in CHF with 0.3 mg. kg-1. day-1 SIM (P < 0.01), 13.2 +/- 3.9 ms2 in CHF with 1.5 mg. kg-1. day-1 SIM (P < 0.05), and 22.0 +/- 3.0 ms2 in CHF with 3.0 mg. kg-1. day-1 SIM. Both PSA data for low (0.625-0.1875 Hz) and high frequencies (0.1875-0.5625 Hz) showed recovery in CHF animals on medium and high SIM doses without changes in the low-to-high-frequency ratio. SIM beneficially affects autonomic tone in CHF as seen by the reversal of depressed HRV and total power of PSA. These data have important implications for the treatment of patients with autonomic imbalance.  相似文献

We measured nitrogen (N) transformation rates for six years to examine temporal variation across the vegetation and elevation gradient that exists within the Coweeta Hydrologic Laboratory. Net N mineralization and nitrification rates were measured using 28-day in situ closed core incubations. Incubations were conducted at various intervals, ranging from monthly during the growing season, to seasonally based on vegetation phenology. Vegetation types included oak-pine, cove hardwoods, low elevation mixed oak, high elevation mixed oak, and northern hardwoods. Elevations ranged from 782 to 1347 m. Nitrogen transformation rates varied with vegetation type. Mineralization rates were lowest in the oak-pine and mixed oak sites averaging <1.2 mg N kg soil-1 28 day-1. Rates in the cove hardwood site were greater than all other low elevation sites with an annual average of 3.8 mg N kg soil-1 28 day-1. Nitrogen mineralization was greatest in the northern hardwood site averaging 13 mg N kg soil-1 28 day-1. Nitrification rates were typically low on four sites with rates <0.5 mg N kg soil-1 28 day-1. However, the annual average nitrification rate of the northern hardwood site was 6 mg N kg soil-1 28 days-1. Strong seasonal trends in N mineralization were observed. Highest rates occurred in spring and summer with negligible activity in winter. Seasonal trends in nitrification were statistically significant only in the northern hardwood site. Nitrogen mineralization was significantly different among sites on the vegetation and elevation gradient. While N mineralization rates were greatest at the high elevation site, vegetation type appears to be the controlling factor.  相似文献

The effects of regular submaximal exercise on dietary protein requirements, whole body protein turnover, and urinary 3-methylhistidine were determined in six young (26.8 +/- 1.2 yr) and six middle-aged (52.0 +/- 1.9 yr) endurance-trained men. They consumed 0.6, 0.9, or 1.2 g.kg-1.day-1 of high-quality protein over three separate 10-day periods, while maintaining training and constant body weight. Nitrogen measurements in diet, urine, and stool and estimated sweat and miscellaneous nitrogen losses showed that they were all in negative nitrogen balance at a protein intake of 0.6 g.kg-1.day-1. The estimated protein requirement was 0.94 +/- 0.05 g.kg-1.day-1 for the 12 men, with no effect of age. Whole body protein turnover, using [15N]glycine as a tracer, and 3-methylhistidine excretion were not different in the two groups, despite lower physical activity of the middle-aged men. Protein intake affected whole body protein flux and synthesis but not 3-methylhistidine excretion. These data show that habitual endurance exercise was associated with dietary protein needs greater than the current Recommended Dietary Allowance of 0.8 g.kg-1.day-1. However, whole body protein turnover and 3-methylhistidine excretion were not different from values reported for sedentary men.  相似文献

The purpose of this paper is to use physiologically based pharmacokinetic models to demonstrate that if toxic response is a function of the time profile in physiological time of the concentration of the toxic moiety in the target tissue, then the appropriate interspecies scaling law for toxic compounds which are metabolically deactivated is mg kg-1 per unit of physiological time (mg kg-1 pt-1). At low dose rates this metric is approximately equivalent to mg kg-0.75 day-1. For reactive metabolites which are spontaneously deactivated, an approximate interspecies scaling law is mg kg-1 day-1.  相似文献

The life-history traits of pocket gophers and their chewing lice suggest that there is little opportunity for transmission of parasites among pocket gophers, with the exception of transmission from mother to offspring. Herein, we test the hypothesis that lice are transmitted maternally by using an indirect approach that compares the distribution of louse populations to the distribution of mitochondrial DNA haplotypes in the pocket gophers. Comparison of the chewing louse distributions to the distribution of mtDNA haplotypes for the gophers revealed no significant concordance, and thus falsifies the maternal transmission hypothesis.  相似文献

The digging apparatus of pocket gophers offers a unique opportunity to examine morphological constraints within a historical context because relationships among extant taxa are well resolved and the features enhancing digging performance are relatively well understood. Structural and functional considerations suggest that the muscles associated with tooth- and claw-digging in pocket gophers are subjected to contrasting levels of morphological constraints. To assess this hypothesis, we analysed the bones and muscles of the jaws and forelimbs in four genera comprising five species of pocket gophers. Morphometric analyses were performed on 12 osteological measurements selected to reflect overall skull size, variation in rostral shape and procumbency, differences in overall length of the forelimbs and processes relating to the function of lever systems used in claw-digging. In addition, dissections were made of the jaw, hyoid, neck and all of the forelimb muscles excluding the intrinsic muscles of the manus. Results of our morphometric analyses corroborate the recent suggestion that pocket gophers encompass a wide range of morphological variation extending from claw-diggers to tooth-diggers. Myologically, however, we found structural variation only in the forelimb muscles, some of which may be advantageous for digging. No changes in jaw, neck and hyoid muscles, other than differences in muscle mass or those concordant with differences in rostral shape, were noted. These results support our hypothesis that constrasting levels of morphological constraint exist between the jaw and forelimb muscles of pocket gophers. We present a discussion of the structural and functional constraints on jaws and forelimbs in gophers as well as an analysis of historical constraints acting on this group, and perhaps on mammals in general.  相似文献

Leucine kinetic and nitrogen balance (NBAL) methods were used to determine the dietary protein requirements of strength athletes (SA) compared with sedentary subjects (S). Individual subjects were randomly assigned to one of three protein intakes: low protein (LP) = 0.86 g protein.kg-1.day-1, moderate protein (MP) = 1.40 g protein.kg-1.day-1, or high protein (HP) = 2.40 g protein.kg-1.day-1 for 13 days for each dietary treatment. NBAL was measured and whole body protein synthesis (WBPS) and leucine oxidation were determined from L-[1-13C]leucine turnover. NBAL data were used to determine that the protein intake for zero NBAL for S was 0.69 g.kg-1.day-1 and for SA was 1.41 g.kg-1.day-1. A suggested recommended intake for S was 0.89 g.kg-1.day-1 and for SA was 1.76 g.kg-1.day-1. For SA, the LP diet did not provide adequate protein and resulted in an accommodated state (decreased WBPS vs. MP and HP), and the MP diet resulted in a state of adaptation [increase in WBPS (vs. LP) and no change in leucine oxidation (vs. LP)]. The HP diet did not result in increased WBPS compared with the MP diet, but leucine oxidation did increase significantly, indicating a nutrient overload. For S the LP diet provided adequate protein, and increasing protein intake did not increase WBPS. On the HP diet leucine oxidation increased for S. These results indicated that the MP and HP diets were nutrient overloads for S. There were no effects of varying protein intake on indexes of lean body mass (creatinine excretion, body density) for either group. In summary, protein requirements for athletes performing strength training are greater than for sedentary individuals and are above current Canadian and US recommended daily protein intake requirements for young healthy males.  相似文献

The adaptive responses of body protein metabolism to lactation were characterized in women at 1, 5, and 12 mo postpartum and in nulliparous controls during a controlled diet of measured protein and energy intakes by nitrogen balance, a constant infusion of [13C]bicarbonate, and a primed constant infusion of [1-13C]leucine and [alpha-15N]-lysine. Dietary energy intakes in the lactating women were 27% greater than those in the nulliparous controls. Despite these differences, lactating women had significantly lower nitrogen balances compared with the nonlactating women (-4.0 +/- 37.8 vs. +44.7 +/- 30.8 mg.kg-1.day-1). No significant differences in amino acid flux, oxidation, or incorporation into protein were detected during fasting conditions in the two groups of women. However, significantly positive associations were noted between dietary intakes and the variables of protein metabolism in the lactating women. A more complete understanding of the mechanisms that regulate the disposition of dietary nutrients into maternal body stores or milk production will enhance the determination of nutrient requirements in lactating women.  相似文献

Articulating the consequences of global climate change on terrestrial ecosystem biogeochemistry is a critical component of Arctic system studies. Leaf mineral nutrition responses of tundra plants is an important measure of changes in organismic and ecosystem attributes because leaf nitrogen and carbon contents effect photosynthesis, primary production, carbon budgets, leaf litter, and soil organic matter decomposition as well as herbivore forage quality. In this study, we used a longterm experiment where snow depth and summer temperatures were increased independently and together to articulate how a series of climate change scenarios would affect leaf N, leaf C, and leaf C:N for vegetation in dry and moist tussock tundra in northern Alaska, USA. Our findings were: 1) moist tundra vegetation is much more responsive to this suite of climate change scenarios than dry tundra with up to a 25% increase in leaf N; 2) life forms exhibit divergence in leaf C, N, and C:N with deciduous shrubs and graminoids having almost identical leaf N contents; 3) for some species, leaf mineral nutrition responses to these climate change scenarios are tundra type dependent ( Betula ), but for others ( Vaccinium vitis-idaea ), strong responses are exhibited regardless of tundra type; and 4) the seasonal patterns and magnitudes of leaf C and leaf N in deciduous and evergreen shrubs were responsive to conditions of deeper snow in winter. Leaf N is was generally higher immediately after emergence from the deep snow experimental treatments and leaf N was higher during the subsequent summer and fall, and the leaf C:N were lower, especially in deciduous shrubs. These findings indicate that coupled increases in snow depth and warmer summer temperatures will alter the magnitudes and patterns of leaf mineral nutrition and that the long term consequences of these changes may feed-forward and affect ecosystem processes.  相似文献

Foraging dynamics of muskoxen in Peary Land, northern Greenland   总被引:1,自引:0,他引:1  
Muskoxen Ovibos moschatus in northern Greenland (79-83°N) are at the northern limit of their distribution and exist under seasonal extremes dominated by nearly 10 months of winter, much of which is without sunlight. The period of summer vegetative growth is less than two months. In the Kap København area (82°30'N), diversity of plant species is low (76 species of vascular plants) and forage biomass in major vegetation types in summer varies from over 40 g m-2 in sedge-dominated fens to ≤5 g m-2 in polar barrens. Nonetheless, 90-95% of the ice-free area consists of barren ground or sparcely vegetated polar desert. During summer, muskoxen apparently foraged opportunistically to maximize intake, with sedges the major food item in fens while willows were the major dietary component when on willow-dominated slopes. Quality of summer forage was high during its early phenological stages, with 21-28% crude protein and 60-75% in vitro digestibility. Microhistological analysis of winter feces indicated dominance by graminoids. Muskoxen spent > 50% of their daily activity feeding, which fits a cline of increasing feeding time with increasing latitude in summer. Increased feeding times at high latitudes appears to be a function of both reduced forage biomass and need to maximize forage intake during the brief summer period when forage quality is high. Movement rates in summer while foraging were inversely related to available forage biomass. Seasonal activity of muskoxen peaks during the rutting period (July-September) and then declines gradually through early winter to a low in late winter (March-April).  相似文献

To examine the adaptations to low O2 and high CO2 among fossorial and nonfossorial rodents, hematological parameters were determined for laboratory rats, the valley pocket gopher (Thomomys bottae) from 250 m, and the mountain pocket gopher (T. umbrinus melanotis) from 3150 m. Hematocrit, hemoglobin concentration, and O2 capacity were higher in pocket gophers than in rats. Blood PO2 at 50% saturation and pH 7.4 was 33 mmHg for both gophers and 39 mmHg for rats. Bohr factors for all three rodents were similar (-0.55 to -0.61) but buffer value, delta log PCO2/delta pH, was -2.54 for T. umbrinus, -1.97 for T. bottae, and -0.98 for Rattus. Concentrations of total acid-soluble phosphates were 50-75% higher in gophers than in rats, while bicarbonate values were within the normal mammalian range. All three rodents had similar myoglobin concentrations in cardiac muscle. Myoglobin concentrations were significantly higher in skeletal muscles (diaphragm, gastrocnemius) of T. umbrinus when compared to T. bottae, and significantly higher in both gophers when compared to rats. These differences may constitute important adaptations to the hypoxia and hypercapnia in burrows; certain of these factors in pocket gophers respond to the additional stress of high altitude hypoxia.  相似文献

Protein loss leading to reduced lean body mass is recognized to contribute to the high levels of morbidity and mortality seen in critical illness. This prospective, randomized, controlled study compared the effects of conventional parenteral nutrition (TPN), glutamine-supplemented (0.4 g.kg-1.day-1) TPN (TPNGLN), and TPNGLN with combined growth hormone (GH, 0.2 IU.kg-1.day-1) and IGF-I (160 microg.kg-1.day-1) on protein metabolism in critical illness. Nineteen mechanically ventilated subjects [64 +/- 3 yr, body mass index (BMI) 23.8 +/- 1.3, kg/m2] were initially studied in the fasting state (study 1) and subsequently after 3 days of nutritional with/without hormonal support (study 2). All had recently been admitted to the ICU and the majority were postemergency abdominal surgery (APACHE II 17.5 +/- 1.0). Protein metabolism was assessed using a primed constant infusion of [1-13C]leucine. Conventional TPN contained mixed amino acids, Intralipid, and 50% dextrose. TPNGLN, unlike TPN alone, resulted in an increase in plasma glutamine concentration ( approximately 50%, P < 0.05). Both TPN and TPNGLN decreased the rate of protein breakdown (TPN 15%, P < 0.002; TPNGLN 16%, P < 0.05), but during these treatments the patients remained in a net negative protein balance. Combined treatment with TPNGLN + GH/IGF-I increased plasma IGF-I levels (10.3 +/- 0.8 vs. 48.1 +/- 9.1 nmol/l, study 1 vs. study 2, P < 0.05), and in contrast to therapy with nutrition alone, resulted in net protein gain (-0.75 +/- 0.14 vs. 0.33 +/- 0.12 g protein.kg-1.day-1, study 1 vs. study 2, P < 0.05). Therapy with GH/IGF-I + TPNGLN, unlike nutrition alone, resulted in net positive protein balance in a group of critically ill patients.  相似文献

1. Seasonal variations in unit perching costs, flying costs and energy budgets for Lichmera indistincta were investigated. 2. Unit perching and flying costs were greatest in winter and least in summer. Variations in perching costs occurred principally because of changes in conductance. Flying cost variations may have been due to changes in wing disc loading and/or the integrity of wing feathers. 3. Rates of net energy intake were greatest in winter and least in summer. Variations in these rates were such that they offset changes in the rates of total energy expenditure to provide diurnal energy balance. 4. Regardless of the season, net daytime energy storage proceeded at a uniform rate and resulted in the storage of sufficient energy to offset overnight expenditure.  相似文献

Seasonal changes in protein and nitrogen metabolism have not previously been reported in any Antarctic suspension-feeding species that ceases feeding for extended periods in winter. To provide comparison with data reported on Nacella concinna, a species that continues to feed in winter, we have measured feeding activity; oxygen consumption; ammonia, urea, and fluorescamine-positive substance (FPS) excretion; O : N ratios; body wall protein synthesis; RNA to protein ratios; and RNA activity at three times during the year in an Antarctic suspension-feeding holothurian. Feeding activity ceased for 4 mo during winter, and oxygen consumption rates decreased from 8.79+/-0.43 micro mol h(-1) to 4.48+/-0.34 micro mol h(-1). Ammonia excretion also decreased during winter from 2,600+/-177 nmol N h(-1) to 974+/-70 nmol N h(-1), but urea excretion rates increased from 178+/-36 nmol N h(-1) to 281+/-110 nmol N h(-1), while FPS excretion rates remained unchanged throughout the year with a seasonal mean of 88+/-13 nmol N h(-1). Oxygen to nitrogen ratios ranged between 6 and 10, suggesting that proteins were used as the primary metabolic substrate. Body wall protein synthesis rates decreased from 0.35%+/-0.03% d(-1) in summer to 0.23% d(-1) in winter, while RNA to protein ratios decreased from 33.10+/-1.0 microg RNA mg(-1) protein in summer to 27.88+/-1.3 microg RNA mg(-1) protein in winter, and RNA activity was very low, ranging between 0.11+/-0.01 mg protein mg(-1) RNA d(-1) in summer and 0.06+/-0.01 mg protein mg(-1) RNA d(-1) in winter. Heterocucumis steineni shows a larger seasonal decrease in oxygen consumption and ammonia excretion between February (summer) and July (winter) than N. concinna, while the proportional decrease in protein synthesis rates is similar in both species.  相似文献

A study of the prevalence and identity of Giardia spp. in small mammals of Grand Teton National Park was undertaken. All 90 montane voles examined were positive for Giardia, as were 4 pocket gophers, 1 water shrew, 4 water voles, and 2 meadow voles. 2003年11月-2004年10月在宁夏贺兰山国家级自然保护区苏峪口国家森林公园,在每个月的11-20日采用瞬时扫描取样法,利用SJ-1型电子事件记录器观察记录岩羊的取食、休息、运动、站立和其他行为.冬季每天的观察时间为800-1700,春、秋季为700-1800,夏季为600-2000.采用非参数估计中的2个相关样本的Wilcoxon检验、2个独立样本的Mann-Whitney U检验和K个独立样本的Kruskal-Wallis H检验对岩羊不同性别、年龄以及季节变化进行了比较.结果表明,①岩羊行为的年时间分配,取食行为最高[(63.49±7.82)%];其次是休息[(19.32±6.79)%];运动和站立较低[分别占(8.03±0.91)%和(6.66±0.57)%];其他行为所占比例最低[(2.67±0.34)%];一年中7月取食行为最低,休息最高;而在2月取食最高,运动、站立和其他行为变化不大.②春夏秋3个季节的昼间取食行为均呈现2个(清晨和黄昏)明显高峰;冬季1天中取食均达到60%以上,其他行为明显高于另外3个季节.③不同年龄年取食行为成体和亚成体>幼体;休息成体和亚成体<幼体;运动和站立幼体<成体和亚成体;亚成体的运动高于成体,而站立低于成体.④不同性别月取食行为雄性在2月最高,其余月份均低于70%;而雌性虽然也是2月最高,但从11月-次年2月均超过70%;休息行为雌雄接近;运动行为雄性高于雌性.贺兰山岩羊的昼间时间分配与许多温带有蹄类动物一样,食物数量和质量的变化以及岩羊自身不同的生长阶段和生理时期是决定其昼间时间分配的主要因素.  相似文献

Amiodarone is a potent and efficacious antiarrhythmic agent, yet associated with its use are life-threatening pulmonary fibrosis and hepatotoxicity. We have investigated the susceptibility of the male Sprague-Dawley rat to pulmonary and hepatic toxicity after repeated exposure to amiodarone and the effects of such exposure on hepatic and extrahepatic drug metabolizing enzymes. Animals received amiodarone (200 mg.kg-1.day-1 i.p., 5 days/week) for 1 week followed by 150 mg.kg-1.day-1 (5 days/week) for 3 additional weeks. No signs of pulmonary fibrosis or hepatotoxicity were observed, based on histological examination, lung hydroxyproline content, and plasma alanine aminotransferase activity. Analysis of tissues revealed extensive accumulation of amiodarone and desethylamiodarone in lung and liver, but concentrations were significantly lower in animals treated for 4 weeks than for 1 week. In a separate experiment, rats received amiodarone 150 mg.kg-1.day-1 i.p. (5 days/week) for 1 or 4 weeks. No differences in tissue concentrations of amiodarone and desethylamiodarone were detected between animals treated for 1 or 4 weeks. This regimen did not affect hepatic or extrahepatic monooxygenase activities. These results indicate that, in the male Sprague-Dawley rat, there is no observable pulmonary or hepatic toxicity following short-term amiodarone exposure, and there is enhanced elimination of amiodarone and desethylamiodarone when the daily dose of amiodarone is decreased after 1 week from 200 to 150 mg/kg.  相似文献

Systolic blood pressure responses to enalapril maleate (MK 421, a new angiotensin converting enzyme inhibitor (CEI] and hydrochlorothiazide (HTZ) were studied in conscious Dahl salt-sensitive (DS) and salt-resistant (DR) rats maintained on a high salt (8.0% NaCl) and a normal salt (0.4% NaCl) diet. The DS rats were severely hypertensive after 3 weeks on the high salt diet whereas the systolic blood pressure (SBP) of the DR rats were normotensive. Oral treatment with enalapril (15-100 mg X kg-1 X day-1) and HTZ (60-400 mg X kg-1 X day-1) caused a significant reduction of SBP in the DS rats with the high salt diet (P less than 0.001); however, this was not observed until after 4 weeks of treatment when the dosage was 30 and 150 mg X kg-1 X day-1, respectively. Furthermore, enalapril therapy alone significantly reduced the SBP of all groups of rats regardless of diet or Dahl strain (P less than 0.001), but this was not observed until the end of the 7th week of therapy in DR rats on 8.0% NaCl and the end of the 3rd week of therapy for DR and DS rats on 0.4% NaCl. These results suggest that enalapril may lower SBP by mechanisms other than those related to an action as a CEI.  相似文献
