A possible alternative to liver transplantation is the internal bioartificial liver (IBAL) consisting in the transplantation of isolated encapsulated hepatocytes. The goal of IBAL is to allow an auxiliary liver until native liver regeneration. The hepatocytes could be allogeneic or xenogeneic (animal origin). Large animal source of liver cells should allow immediate isolation of fresh hepatocytes when IBAL is required. The peritoneal cavity is probably the site of implantation of IBAL. In the future, IBAL could be envisioned as the treatment of metabolic deficiencies or acute and chronic liver failure. It could ensure a therapeutic bridge until transplantation.  相似文献

Stinging mechanisms generally deliver venomous compounds to external targets. However, nematocysts, the microscopic stinging organelles that are common to all members of the phylum Cnidaria, occur and act in both external and internal tissue structures. This is the first report of such an internal piercing mechanism. This mechanism identifies prey items within the body cavity of the sea anemone and actively injects them with cytolytic venom compounds. Internal tissues isolated from sea anemones caused the degradation of live Artemia salina nauplii in vitro. When examined, the nauplii were found to be pierced by discharged nematocysts. This phenomenon is suggested to aid digestive phagocytic processes in a predator otherwise lacking the means to masticate its prey.  相似文献

Uptake of phosphate by Streptococcus lactis ML3 proceeds in the absence of a proton motive force, but requires the synthesis of ATP by either arginine or lactose metabolism. The appearance of free Pi internally in arginine-metabolizing cells corresponded quantitatively with the disappearance of extracellular phosphate. Phosphate transport was essentially unidirectional, and phosphate concentration gradients of up to 10(5) could be established. Substrate specificity studies of the transport system indicated no preference for either mono- or divalent phosphate anion. The activity of the phosphate transport system was affected by the intracellular Pi concentration by a feedback inhibition mechanism. Uncouplers and ionophores which dissipate the pH gradient across the cytoplasmic membrane inhibited phosphate transport at acidic but not at alkaline pH values, indicating that transport activity is regulated by the internal proton concentration. Phosphate uptake driven by arginine metabolism increased with the intracellular pH with a pKa of 7.3. Differences in transport activity with arginine and lactose as energy sources are discussed.  相似文献

Rhopalosiphum padi virus (RhPV) is an insect virus of the Dicistroviridae family. Recently, the 579-nucleotide-long 5' untranslated region (UTR) of RhPV has been shown to contain an internal ribosome entry site (IRES) that functions efficiently in mammalian, plant, and insect in vitro translation systems. Here, the mechanism of action of the RhPV IRES has been characterized by reconstitution of mammalian 48S initiation complexes on the IRES from purified components combined with the toeprint assay. There is an absolute requirement for the initiation factors eIF2 and eIF3 and the scanning factor eIF1 to form 48S complexes on the IRES. In addition, eIF1A, eIF4F (or the C-terminal fragment of eIF4G), and eIF4A strongly stimulated the assembly of this complex, whereas eIF4B had no effect. Although the eIF4-dependent pathway is dominant in the RhPV IRES-directed cell-free translation, omission of either eIF4G or eIF4A or both still allowed the assembly of 48S complexes from purified components with approximately 23% of maximum efficiency. Deletions of up to 100 nucleotides throughout the 5'-UTR sequence produced at most a marginal effect on the IRES activity, suggesting the absence of specific binding sites for initiation factors. Only deletion of the U-rich unstructured 380-nucleotide region proximal to the initiation codon resulted in a complete loss of the IRES activity. We suggest that the single-stranded nature of the RhPV IRES accounts for its strong but less selective potential to bind key mRNA recruiting components of the translation initiation apparatus from diverse origins.  相似文献

The structure and motility of the Mollicutes (Spiroplasma, Mycoplasma, and Acholeplasma) are briefly reviewed. The data are presented from the perspective of prokaryotic and eukaryotic motors, cytoskeletons, and cell motility. The Mollicutes are eubacteria derived from Clostridia by regressive evolution and genome reduction to produce the smallest and simplest free-living and self-replicating cells. Structurally, the Mollicutes are characterized by a complete lack of a cell wall and the presence of an internal cytoskeleton. Spiroplasma, which are helical cells with a flat, ribbon-like cytoskeleton, are amenable to structural and geometrical analysis. Motility and shape changes can be explained and modeled by the cytoskeleton acting as a linear motor.  相似文献
