Adenosine-5′-carboxaldehyde (1a) was treated with nitromethane under alkaline conditions, to give the two stereoisomeric 5′-C-(nitromethyl) derivatives (2 and 3) of adenosine. Catalytic hydrogenation of 2 gave 9-(6-amino-6-deoxy-β-D-allofuranosyl)adenine (4), which, on treatment with nitrous acid, yielded 9-(β-D-allofuranosyl)hypoxanthine (6). Similar treatment of 3 gave the α-L-talo nucleosides 5 and 7. Reaction of 2′,3′-O-p-anisylidene adenosine-5′-carboxaldehyde (1b) with ethoxycarbonylmethylene-triphenylphosphorane afforded 9-(ethyl 5,6-dideoxy-β-D- ribo-hept-5-enofuranosyluronate)adenine (8), which was hydrolyzed to the corresponding uronic acid (9). Catalytic hydrogenation of 8 gave 9-(ethyl 5,6-dideoxy-β-D-ribo-heptofuranosyluronate)adenine (10). Reduction of 8 with lithium aluminum hydride yielded two new analogs of adenosine: 9-(5,6-dideoxy-β-D-ribo-heptofuranosyl)adenine (12) and 9-(5,6-dideoxy-β-D-ribo-hept-5-enofuranosyl)adenine (13).  相似文献

The use of the microorganism Sporotrichum sulfurescens (ATCC 7159) to oxygenate organic molecules has been extended to several dialkylbenzenes. Oxygenation of 1,4-di-t-butylbenzene (1) gave 4-t-butyl(1-hydroxy-2-methyl)isopropylbenzene (2) and 1,4-di-(1-hydroxy-2-methyl)isopropylbenzene (3); of 1,4-diisopropylbenzene (4) gave (R,R)-1,4-di-(1-hydroxy)isopropylbenzene (5); of 1,3-diisopropylbenzene (6) gave 1,3-di-(2-hydroxy)isopropylbenzene (7), 3-(1-hydroxy)isopropyl-(2-hydroxy)isopropylbenzene (8), and 1,3-di-(1-hydroxy)isopropylbenzene (9); and of p-isobutylisopropylbenzene (20) gave 1-(p-2-hydroxyisopropylphenyl)-2-methylpropan-2-ol (15) and 1-(p-1-hydroxyisopropylphenyl)-2-methylpropan-2-ol (16). Monohydroxydialkylbenzenes also served as useful substrates in this reaction as suggested by the fact that 2 is an intermediate in the formation of 3 from 1. Oxygenation of 1-(p-isopropylphenyl)-2-methylpropan-2-ol (14), conveniently prepared from 2-(p-isopropylphenyl)propene (12) via oxygenative isomerization with thallium trinitrate to 13 followed by addition of methyl magnesium bromide, gave 15 and 16. Oxygenation of 2-(p-isobutylphenyl)propan-2-ol (18) gave 15, 2-(p-isobutylphenyl)-propan-1,2-diol (21), and 1-(p-2-hydroxyisopropylphenyl)-2-methylpropan-3-ol (22). Compound 16, obtained from substrate 14, was converted to (2R)-2-[4-(2-hydroxy-2-methylpropyl)phenyl]propionic acid (11), the enantiomer of a metabolite of the antiinflammatory agent, 2-(4-i-butyl)phenylpropionic acid (10).  相似文献

Two new prenylated flavonoids, lanneaflavonol (1) and dihydrolanneaflavonol (2) together with the known compounds myricetin-3-O-α-rhamnopyranoside (myricitrin) (3) and myricetin-3-O-α-arabinofuranoside (betmidin) (4), lupeol (5) and sitosterol (6) were isolated from the roots of Lannea alata. Compounds 14 exhibited good antibacterial and radical scavenging activity with the glycosides 3 and 4 showing better antioxidant activity than the aglycones 1 and 2 and myricetin-3-O-α-arabinofuranoside (4) showing the best antimicrobial activity followed by the aglycones 1 and 2. Betmidin (4) with an arabinose moiety at the 3-O-position showed the best antibacterial activity against Gram-positive bacteria, followed by the prenylated dihydroflavonol (2), whilst the prenylated linear flavonol (1) showed limited activity against Gram-negative bacteria. The arabinofuranoside (4) followed by the rhamnopyranoside (3) showed the best antioxidant activity comparable to that of ascorbic acid. The synthesis and anticonvulsant properties of new N-Mannich bases of 3-phenyl- (9ad), 3-(2-chlorophenyl)- (10ad), 3-(3-chlorophenyl)- (11ad) and 3-(4-chlorophenyl)-pyrrolidine-2,5-diones (12ad) were described. The key synthetic strategies involve the formation of 3-substituted pyrrolidine-2,5-diones (58), and then aminoalkylation reaction (Mannich-type) with formaldehyde and corresponding secondary amines, which let to obtain the final compounds 9ad, 10ad, 11ad and 12ad in good yields. Initial anticonvulsant screening was performed in mice (ip) using the maximal electroshock (MES) and subcutaneous pentylenetetrazole (scPTZ) seizures tests. The most effective compounds in mice were tested after oral administration in rats. The acute neurological toxicity was determined applying the minimal motor impairment rotarod test. The in vivo results revealed that numerous compounds were effective especially in the MES test (model of human tonic-clonic seizures). The most active in the MES seizures in rats was 1-[(4-benzyl-1-piperidyl)methyl]-3-(2-chlorophenyl)pyrrolidine-2,5-dione (10c) which showed ED50 value of 37.64 mg/kg. It should be stressed that this molecule along with 9a, 9d and 10d showed protection in the psychomotor seizure test (6-Hz), which is known as an animal model of therapy-resistant epilepsy. Furthermore compounds 9a, 9d and 10d were also tested in the pilocarpine-induced status prevention (PISP) test to assess their potential effectiveness in status epilepticus. For the most promising molecule 9d an influence on human CYP3A4 isoform of P-450 cytochrome was studied in vitro.  相似文献

Ibicella lutea and Proboscidea louisiana, both of the Martyniaceae family, are known for rich glandular trichomes on their leaves and stems. Chemical investigations of the glandular trichome exudates on leaves of the two plants furnished three types of secondary metabolites, glycosylated fatty acids, glycerides (2-O-(3,6-diacetyloxyfattyacyl)glycerols and 2-O-(3-acetyloxyfattyacyl)glycerols) and dammarane triterpenes. The glycosylated fatty acids from I. lutea were determined to be 6(S)-(6-O-acetyl-β-d-glucopyranosyloxy)-octadecanoic acid (1A), -eicosanoic acid (1B) and -docosanoic acid (1C), as well as their respective deacetyl congeners (2A, 2B and 2C), whereas P. louisiana furnished 8(S)-(6-O-acetyl-β-d-glucopyranosyloxy)-eicosanoic acid (3A) and -docosanoic acid (3B) and their respective deacetyl congeners (4A and 4B), together with 2B. Both plants contained 12 identical 2-O-[(3R,6S)-3,6-diacetyloxyfattyacyl]glycerols (5A-L), in which the fatty acyl moieties contained between 17 and 21 carbon atoms. The corresponding mono-acetyloxy compounds, 2-O-[(3R)-3-acetyloxyfattyacyl]glycerols (6AL) were detected in both plants. Among these glycerides, ten compounds (5A, 5C, 5F, 5H, 5K, 6A, 6C, 6F, 6H and 6K) had iso-fattyacyl structures and four (5E, 5J, 6E and 6J) had anteiso-fattyacyl structures. A previously unknown dammarane triterpene, betulatriterpene C 3-acetate (7), was isolated together with three known dammarane triterpenes, 24-epi-polacandrin 1,3-diacetate (8), betulatriterpene C (9) and 24-epi-polacandrin 3-acetate (10) from I. lutea, whereas 12 dammarane triterpenes, named probosciderols A–L (1223), and the known compound betulafolienetriol (11) were isolated from P. louisiana. The structures of these compounds were elucidated by spectroscopic analysis including 2D-NMR techniques and chemical transformations. The 6-O-acetylglucosyloxy-fatty acids 1AC (42%) and the dammarane triterpenes 710 (31%) were the two most abundant constituents in the glandular trichome exudate of I. lutea, whereas the dammarane triterpenes 1123 (47%) and the glucosyloxy-fatty acids (4A, 4B and 2B) (38%) were the most abundant constituents in the glandular trichome exudate of P. louisiana.  相似文献

Spiroleptosphol B (2), spiroleptosphol C (3), norleptosphol C (4) and hydroleptosphol (5) were isolated from ascomycete Leptosphaeria doliolum. Detailed 1H and 13C NMR spectral analyses revealed these were structural analogues of spiroleptosphol (1) which we have recently isolated from the same fungi. Spiroleptosphol B (2) carried an unprecedent 5,3-dioxatricyclo[4.4.0.11.4]undecane framework in place of the spirobicyclo ring system of 1. Spiroleptosphol C (3) was a 17-(R)-hydroxy derivative of 1. Norleptosphol C (4) was deduced to be the monocyclic structure biosynthetically resulted by decarboxylation from 3. Although 5 gave broaden 1H NMR spectrum, it was gradually transformed to 2 which suggested being a hydrolysate of 1.  相似文献

The 1-methyl derivatives (3 and 4) of 3-(1-phenyl- (1) and 3-(1-p-bromophenylhydrazono-L-threo-2,3,4-trihydroxybutyl)-2-quinoxalinone (2) were prepared by methylation. Periodate oxidation of 3 gave 1-methyl-3-[1-(phenylhydrazono)glyoxal-1-yl]-2-quinoxalinone (5), which, on reduction with sodium borohydride, gave the corresponding 3-[2-hydroxy-1-(phenylhydrazono)ethyl] derivative (8). Reaction of 5 with hydroxylamine or benzoylhydrazine gave the corresponding 2-oxime (6) and 2-(benzoylhydrazone) (7), respectively. Acetic anhydride causes one molecule of 3 or 4 to undergo elimination of two molecules of water, with simultaneous acetylation and ring closure to afford pyrazoles 9 and 10, respectively. Pyrolysis of the triacetate of 3 led to the elimination of acetic acid from the sugar and the hydrazone residue, to give the 3-[5-(acetoxymethyl)-1-phenylpyrazol-3-yl] derivatives (9). Acetic acid was found to effect the same rearrangement, but without acetylation, of 1, 2, and 3 to give the 3-[5-(hydroxymethyl)] derivatives 11, 12, and 13, respectively. The structure of these pyrazoles was confirmed by a series of reactions, including methylation and acetylation. The n.m.r. and i.r. spectra of the compounds were investigated.  相似文献

2,3,5-Tri-O-benzyl-D-ribofuranosyl bromide (17) has been converted into methyl 3-(2,3,5-tri-O-benzyl-β-d-ribofuranosyl)propiolate (8) and its α anomer 10 in 21 and 42% yields, respectively, by reaction with the silver salt of methyl propiolate. Attemps to prepare 8 from (β-d-ribofuranosyl)ethyne (1) by standard methods were unsuccesful. The reactions of the esters 8 and 10 and the ethyne 1 with several 1,3-dipoles have been examined. With diazomethene, 8 and 10 gave the pyrazole esters 20 and 28, respectively, whereas the ethyne 1 reacted more slowly to give a mixture of 23 (37%) and 26(31%). The ester 10 was converted into the triazoles 32 (51%) and 36 (34%) by reaction with benzyl azide. Treatment of the ester 10 with phenylhydrazine gave the pyrazolone 38 in 71% yield. A number of the products of dipolar addition have been converted into new d-ribofuranosyl-pyrazoles and -triazoles by hydrogenolysis.  相似文献
