以樟树果实为材料,运用800~400 nm光谱扫描分析了pH值、金属离子、加热时间及温度、光照等稳定性因子对樟树果实红色素吸收光谱的影响。结果表明:最大吸收波长是517 nm,酸性条件对色素吸收光谱无明显影响,当pH为8.6的强碱性环境,红色素结构变化,最大吸收峰漂移至402 nm;100℃内,色素性质稳定,当加热时间延长至120 min,最大吸收峰在512.5 nm;红色素耐光性较好,但避光更利于保存;金属离子对红色素吸收光谱的影响强弱为:Fe3+>Al3+>Mg2+>Ca2+>Na+>K+,Fe3+短时内引起色素变色、发生沉淀,最大吸收峰漂移,浓度越高影响越大,Na+、K+对红色素吸收光谱无明显影响。  相似文献

Poly (A) took stable double-stranded form in 0.02 m phosphate buffer pH 5.8 when its S-value was higher than 3.5. Poly (C) also took stable double-stranded form in 0.01 m acetate buffer pH 3.9 containing 0.1 m NaCl if its S-value was higher than 4.8.

Poly (I) with S-values higher than 6.5 stably formed triple-stranded form. Poly (A) with S-values over 3S and poly (I) with S-values over 7S were observed to aggregate in the presence of high concentration of salt, but poly (C) and poly (U) did not aggregate under the same condition. Effect of salt concentration differed with different polynucleotides and with different salts.  相似文献

Dark-grown pea seedlings (Pisum sativum L.) were irradiated for a short period each day with low intensity red light (662 nm), red light immediately followed by far red light (730 nm), or far red light alone. Other plants were transferred to a white light regime (14 hours light/10 hours dark). There was no change in the amount of RNA in the tissue on a fresh weight basis after the various treatments. However, compared with dark-grown seedlings, those plants irradiated with red light showed an increase in the net RNA content per stem apex. In addition there was a two- to three-fold increase in ribosomal RNA of the etioplasts relative to the total ribosomal RNA. These increases were comparable to those found in plants grown in the white light regime. The changes were much smaller if the dark-grown plants were irradiated either with red light followed by far red light, or with far red light alone. Thus continuous light is not essential for the production of ribosomal RNA in plastids, and the levels of ribosomal RNA found in chloroplasts can also be attained in etioplasts of pea leaves in the dark provided the leaf phytochrome is maintained in its active form.  相似文献

Wheat seedlings (Triticum aestivum L. cv Gamenya) were grown for 4 days in culture solutions of differing pH prior to studying their subsequent short-term absorption of 32Pi from solutions of the same or different pH.

Increasing pH of the absorption solution from 5.5 to 7.0 or 8.0 depressed 32Pi absorption from 1 and 10 micromolar Pi but had little effect at 100 and 1000 micromolar Pi. Increasing the pH of the culture solution from 4.5 to 6.5 doubled or trebled subsequent 32Pi absorption from nearly all absorption solutions over a wide range of Pi concentrations, pH, and nutrient compositions.

When seedlings were transferred between culture pH treatments 4.5 and 6.5, their capacity for 32Pi absorption remained unchanged for at least 5 hours and adjusted by 60 to 80% after 24 hours and completely after 48 hours. This suggests that the root's capacity to absorb Pi responds to pH through slow structural changes in its mechanism of Pi absorption. P content and concentration of wheat seedlings reflected the response of 32Pi absorption to culture pH.

It is suggested that absorption pH affects an activity component of the process for Pi absorption and culture pH affects a capacity component. Failure to recognize the capacity component of the pH response explains why previously published results for short-term 32Pi absorption conflict with those for long-term P accumulation in plants.

Using 31P nuclear magnetic resonance spectroscopy, we followed cytoplasmic and vacuolar pH in pea (Pisum sativum cv Alaska) internode segments during treatment with indoleacetic acid (IAA) or fusicoccin (FC) in continuously perfused, oxygenated buffer. Although IAA and FC induced normal H+ extrusion, elongation, and glucan synthase activity responses during the measurements, neither the cytoplasmic nor the vacuolar pH showed significant change at any time between 5 minutes and 1 to 3 hours of treatment. Changes in cytoplasmic pH as small as about 0.04 pH unit were detected after treatment with 1-naphthyl acetate. Therefore, cytoplasmic pH changes do not appear to mediate IAA or FC stimulation of H+ extrusion or other metabolic responses to these effectors.  相似文献

The administration of pteroylmonoglutamic acid and either sodium bicarbonate or phenytoin sodium in normal subjects and epileptics results in lower serum levels of folic acid than when administered alone. Bicarbonate administration was shown to produce a more alkaline pH in the jejunum, while an alkaline pH was found in some epileptics on phenytoin therapy. The physicochemical mechanisms whereby folic acid absorption is reduced in an alkaline medium are considered, and the implications on folate absorption in other disease states are discussed. The mechanism by which phenytoin sodium alters the intrajejunal milieu is obscure.  相似文献

By measuring the derivative absorption spectra of chloroplasts of different populations of Pinus armandi Franch., it was found that southern populations maintained higher absorption at 680 nm than at 670 nm, but some of the northern populations deviated the maximum absorption from 680 nm to 670 nm, which indicated that the activity of PS Ⅱ in some of the northern populations declined. Clear geographical differences also have been found in the positions of emission peaks of PS Ⅰ and PS Ⅱ in the fluorescence emission spectra at 77 K. Analysis of the fluorescence excitation spectra at 77 K revealed geographical changes in the absorption.status of Chl a. Besides, the experimental results indicated that the intact needles of Pinus armandi are not ideal materials to be used in detecting the geographical variation in photochemical reaction process because the presence of thicker coat, resin etc. can conceal the spectral differences in different populations.  相似文献

Experiments in which [1-14C]glycolate uptake is carried out in conjunction with measurements of stromal pH indicate that only glycolic acid and not the glycolate anion is crossing the pea (Pisum sativum var. Six healthy male subjects performed three exercise tests in which the power output was increased by 100 kpm/min each minute until exhaustion. The studies were carried out after oral administration of CaCO3 (control), NH4Cl (metabolic acidosis), and NaHCO3 (metabolic alkalosis). Ventilation (VE), O2 intake (VO2), and CO2 output (VCO2) were monitored continuously. Arterialized-venous blood samples were drawn at specific times and analyzed for pH, PCO2, and lactate concentration. Resting pH (mean +/- SE) was lowest in acidosis (7.29 +/- 0.01) and highest in alkalosis (7.46 +/- 0.02). A lower peak power output (kpm/min) was achieved in acidosis (1,717 +/- 95) compared with control (1,867 +/- 120) alkalosis (1,867 +/- 125). Submaximal VO2 and VCO2 were similar, but peak VO2 and VCO2 were lower in acidosis. Plasma lactate concentration was lower at rest and during exercise in acidosis. Although lactate accumulation was reduced in acidosis, increases in hydrogen ion concentration were similar in the three conditions. We conclude that acid-base changes influence the maximum power output that may be sustained in incremental dynamic exercise and modify plasma lactate appearance, but have little effect on hydrogen ion appearance in plasma.  相似文献

Inhibitors of angiotensin I-converting enzyme were isolated from an enzymatic hydrolysate of bovine casein. The amino acid sequences of these inhibitors were Phe-Phe-Val-Ala-Pro-Phe-Pro- Glu-Val-Phe-Gly-Lys (CEI12), Phe-Phe-Val-Ala-Pro (CEI5), and Ala-Val-Pro-Tyr-Pro-Gln-Arg (CEIβ7). CEI5 is a penta-peptide derived from the hydrolysate of CEI12 with proline-specific endopeptidase, and CEIβ7 is a hepta-peptide derived from β-casein. These inhibitors potentiated bradykinin in the contraction of the uterus and the ileum of rats. The ileum was more sensitive to these inhibitors than the uterus. The bradykinin-potentiating activity of these inhibitors on the ileum lasted for more than 90 min even after washing the organ.  相似文献
