A rust fungus found in Japan on Rhododendron kaempferi, R. kiusianum, and R. dauricum has previously been identified as Chrysomyxa rhododendri. Light and scanning electron microscopy of fresh and herbarium materials of the rust fungus, however, show that the spore surface morphology differs from the urediniospores of C. rhododendri, and the spores are slightly smaller. Furthermore, the DNA sequence of the 5′-end of the large subunit of ribosomal DNA differs from that of C. rhododendri by 3%. Telia have not been found; therefore, it is redescribed as a new anamorphic species, Caeoma tsukubaense. Through the morphological and molecular examinations of Melampsora species on willows, we clarified the taxonomic identity of the rust specimens on Salix bakko, S. hultenii and S. leucopithecia from Japan and described the following rust fungus as a new species, Melampsora salicis-bakko. This rust fungus resembled M. caprearum in morphology of teliospores, but it differed from M. caprearum mainly in the density of spines on the urediniospores. Molecular phylogenetic analyses using the rDNA ITS region (complete ITS1, 5.8S rRNA gene and ITS2) revealed that M. salicis-bakko was monophyletic, and that this rust fungus was distinct from other Melampsora species, including M. caprearum.  相似文献

Puccinia hemerocallidis and P. funkiae resemble each other morphologically; however, they are biologically and taxonomically distinct, with telia of the former being restricted to species of Hemerocallis and the latter to Hosta species. However, both fungi share a macrocyclic and heteroecious life cycle with Patrinia villosa as the spermogonial and aecial host. An additional microcyclic rust fungus, P. patriniae, is also known on P. villosa. This microcyclic fungus is similar to the two macrocyclic fungi in its telial structure and teliospore morphology. These similarities in morphology and host relationships suggest the three fungi may also share a close evolutionary relationship. To determine the phylogenetic relationships of the three species, a portion of the nuclear ribosomal DNA repeat encoding the ITS and 5.8S subunit regions was amplified by PCR, sequenced, and analyzed. The resulting phylogenetic trees showed that P. hemerocallidis and P. funkiae share a recent common ancestor and that P. patriniae is closely allied with P. hemerocallidis. The results suggest a possible evolutionary derivation of microcyclic P. patriniae from macrocyclic heteroecious P. hemerocallidis, which fits the evolutionary interpretation of correlated species known as Tranzschel's law.  相似文献

Puccinia kraussiana, a rust fungus of Africa, was found in Taiwan for the first time in eastern Asia. It was parasiting on leaves ofHeterosmilax gaudichaudiana, on which all four stages were characteristically found. The host species,H. gaudichaudiana, newly added to the host range ofP. kraussiana, constitutes also a new entry to the phanerogamic flora of Taiwan.  相似文献

The objective of this study was to find features in microcyclic rust fungi (Uredinales) on wild host plants favorable for extension of the natural range of distribution. Puccinia glechomatis, a leptosporic rust fungus and its herbal host Glechoma hederacea (Lamiaceae), both natives to Eurasia and introduced in North America, were used for this study. Although the host has been known from North America since the beginning of the nineteenth century, the rust fungus was first observed there only in recent years. Favorable features were identified by studying the life cycle of the rust, including nuclear conditions and seasonal characteristics as well as its spread in North America. The life cycle was studied macroscopically by inoculation experiments, by various light microscope techniques, and by scanning electron microscopy. The spread of the pathogen and its host were reconstructed by evaluating host plant herbarium specimens and databases, literature, and field study data. The studies on P. glechomatis show that, generally for microcyclic rust fungi, establishment and potential for spread are based on several favorable features of both the host (e.g., synanthropic occurence and dispersal, genetic stability, regeneration of vegetative plant parts) and the rust fungus (asexual reproduction/genetic stability, homothallism, propagation with host plant, formation of both leptospores and thick-walled teliospores).  相似文献

Uredinial and telial states of aMelampsora species occurring on the leaves ofSalix serissaefolia were for the first time recorded. Field observations and inoculation experiments showed that the spermogonial and aecial states of the fungus were formed onChelidonium majus var.asiaticum andCorydalis incisa. The fungus was identified asMelampsora yezoensis based on the morphological observation of all the spore states. Urediniospores were able to infectS. erocarpa andS. pierotii as well asS. serissaefolia andS. jessoensis. Salix serissaefolia, S. eriocarpa, andS. pierotii are new uredinial and telial hosts ofM. yezoensis, andC. majus var.asiaticum andC. incisa are new spermogonial and aecial hosts of the fungus. Many species of Melampsora on Populus have been reported in China, based on morphological characteristics of both uredial and telial states, and on host species, but their morphology and taxonomy are still poorly defined. In this study, 196 specimens representing Melampsora species on poplars and collected from various areas of China were used for morphological observations. The morphological characteristics of urediniospores and teliospores were examined with light and scanning electron microscopy. The specimens could be classified into five groups based on their morphology. For the sequencing of the nuclear large subunit rDNA (D1/D2), 5.8S rDNA and their internal transcribed spacers, ITS1 and ITS2 region, 54 specimens were selected from the specimens used in morphological observations. These specimens were separated into six clades by phylogenetic analyses of the D1/D2 and ITS regions. Correlations among morphological groups and phylogenetic clades based on these results suggest a revision of these species. Morphological examination and PCR-RFLP analysis of the D1/D2 region in nuclear LSU rDNA were carried out to identify a rust fungus onPinus pumila collected in the North Kurils, Russia. Morphologically, the rust from the North Kurils was similar toPeridermium kurilense andEndocronartium sahoanum var.hokkaidoense in the spore surface structure, dimension and shape. In the RFLPs of the D1/D2 region it was identical toE. sahoanum var.hokkaidoense andE. sahoanum var.sahoanum, but distinct fromCronartium ribicola. Therefore, the rust fungus onP. pumila collected in the North Kurils was identified asE. sahoanum var.hokkaidoense. A rust fungus attack was observed on Tree Spurge (Euphorbia dendroides) in two regions of southern Italy. Uredinia and telia of a Melampsora species were detected on the leaves, bracts of the cyathia and capsules of E. dendroides. On the basis of the morphological characteristics and rDNA ITS sequences of the above mentioned structures, the fungus was identified as M. gelmii. This report suggests that M. gelmii seems to be a specific taxon in the M. euphorbiae complex and to be more widespread than previously detected in Italy. Further investigations within the M. euphorbiae complex would be desirable in order to elucidate the identity of M. gelmii as a peculiar form of a still poorly explored species.  相似文献

Atkinsiella dubia, isolated from the mantle of abalone (Haliotis sieboldii), is described and illustrated as a new record from Japan. The fungus was also obtained from the gills of swimming crab (Portunus trituberculatus). Six other species of the genusAtkinsiella have hitherto been reported from various aquatic animals. The fungus is distinguished from the other six species by the morphology of its mycelia and the process of zoospore production. The most distinctive feature is that zoospores in the first motile stage ofA. dubia encyst in zoosporangia, unlike the other species. We therefore proposeHalocrusticida gen. nov. (Lagenidiales, Haliphthoraceae) for the other six species ofAtkinsiella. A key to species of the genusHalocrusticida is provided.  相似文献

Aplopsora corni sp. nov. is proposed for a rust fungus whose uredinial and basidial stage occurs onCornus controversa (Cornaceae) in Hokkaido. Nine species of rust fungi (Uredinales) were found during a 1-day field study in an Acacia koa–Metrosideros polymorpha woodland in Volcanoes National Park on Big Island, Hawaii. Two species, both found on Acacia koa, are native (endemic) rusts whereas 7 species are nonnative on nonnative hosts, highlighting the high proportion of introduced species in the Hawaiian flora even in more or less natural habitats. One species, Uromyces linearis on Panicum repens, constitutes the first record of this rust for the Hawaiian archipelago, bringing the total to 93 species on the islands, 70 (75.3%) of which are introduced. The species records are annotated with emphasis on the geographic origin of each taxon. In addition, the study has led to the molecular reevaluation of the genus Racospermyces, indicating that it is synonymous with Endoraecium, and six new combinations are proposed for the species previously placed in Racospermyces. The high number of introduced species in Hawaii coupled with the paucity of native species when compared to other global regions is discussed.  相似文献

A pea rust fungus, Uromyces viciae-fabae, has been classified into two varieties, var. viciae-fabae and var. orobi, based on differences in urediniospore wall thickness and putative host specificity in Japan. In principal component analyses, morphological features of urediniospores and teliospores of 94 rust specimens from Vicia, Lathyrus, and Pisum did not show definite host-specific morphological groups. In molecular analyses, 23 Uromyces specimens from Vicia, Lathyrus, and Pisum formed a single genetic clade based on D1/D2 and ITS regions. Four isolates of U. viciae-fabae from V. cracca and V. unijuga could infect and sporulate on P. sativum. Based on morphological characteristics and molecular phylogeny, a rust fungus on Salix hsinganica, S. sinica, S. starkeana, S. wallichiana and S. xerophila in China is described as a new species, Melampsora salicis-sinicae. This new species morphologically resembles M. capraearum and M. epiphylla in having subcuticular teliospores with a thickened apical wall, but it differs from these two species in its amphigenous telia. In addition, this new species differs from M. capraearum in its slender teliospores and thinner apical thickness of teliospores, and it also differs from M. epiphylla in the dimension of urediniospores, the position of teliospores and the apical thickness of teliospores. Phylogenetic analyses using partial sequences of the D1/D2 region of nuclear large subunit and the internal transcribed spacer (ITS1 and ITS2) regions including 5.8S of ribosomal RNA gene and the translation elongation factor 1 alpha (EF-1α) gene revealed that this rust fungus was distinct from its morphologically similar species, M. capraearum and M. epiphylla.  相似文献

We developed 15 microsatellite loci in the poplar rust fungus, Melampsora larici‐populina, using two enrichment protocols. Polymorphism of each locus was assessed on a panel of 30 isolates, comprising three subpanels (world, regional and local scales). Thirteen loci were polymorphic with three to eight alleles detected. The 15 loci were also tested on five related Melampsora species, M. allii‐populina, M. medusae f. sp. deltoidae, M. larici‐tremulae, M. rostrupii and M. pinitorqua, and partial or global cross‐amplification events were detected.  相似文献

A rust species on Calystegia soldanella in Japan has been treated as Puccinia convolvuli to date. However, morphological characteristics of specimens on C. soldanella collected from Japan are significantly different from those of specimens on other Calystegia and Convolvulus species from different areas of the world. It is proved by inoculation experiment that the rust on C. soldanella is specific to C. soldanella. Based on these results, Puccinia rust on C. soldanella from Japan is described as a new species, Puccinia calystegiae-soldanellae.  相似文献

Uromyces appendiculatus, inclusive of three varieties, is distinguished from U. vignae primarily by the position of urediniospore germ pores and putative host specificity. However, opinions concerning these morphological and physiological features as taxonomic characters have varied greatly, and distinction of these species has often been confused. To clarify the taxonomy of these two species, morphological features of urediniospores and teliospores of 225 rust fungus specimens on species of Phaseolus, Vigna, Apios, Lablab, and Dunbaria were examined by light microscopy and scanning electron microscopy. Forty-five specimens were subjected to molecular phylogenetic analyses. As a result, the position of germ pores in urediniospores and the teliospore-wall thickness were considered as good characters to separate three morphological groups. In molecular analyses, the specimens fell into two and three clades based on the nucleotide sequence at D1/D2 domain of LSU rDNA and ITS regions, respectively. One of the D1/D2 clades corresponded to one morphological group whereas another D1/D2 clade included two other morphological groups. In contrast, each of the three ITS clades corresponded to a separate morphological group. Neither morphological groups nor molecular clades were host limited. Heteroecious Phakopsora euvitis, P. vitis, and P. ampelopsidis, autoecious P. meliosmae, and an unconnected Aecidium on Meliosma are closely allied. A total of 45 collections representing the five rust fungi from Japan, Australia, and East Timor were subjected to molecular phylogenetic analyses: the D1/D2 region of nuclear large subunit rDNA and nuclear small subunit internal transcribed spacer 2 (ITS2) region including 5.8S rDNA were analyzed. Tree topologies generated from parsimony and distance methods of the D1/D2 and ITS2 sequences were similar. The 45 collections (44 for ITS2 and 33 for D1/D2, with 32 common for both analyses) are grouped into seven clades: P. ampelopsidis, P. vitis, unconnected Aecidium, P. euvitis from Japan, P. euvitis from Australia and East Timor, P. meliosmae on M. myriantha, and P. meliosmae on M. tenuis. The results confirm the phylogenetic distinctness of P. euvitis, P. ampelopsidis, and P. vitis distributed in Japan. A grapevine leaf rust fungus in Australia and East Timor has genetically diverged from P. euvitis in Japan. The unconnected Aecidium is highly likely to be an aecial anamorph of a Phakopsora fungus. Autoecious Phakopsora fungi on M. meliosmae and M. tenuis need further host-specificity and morphological studies to confirm their taxonomic status.  相似文献

The ultrastructure of the uredinial stage of the rust fungus,Puccinia polypogonis onPolypogon monspeliensis is described, using scanning and transmission electron microscopy. This study examined the urediniospores, intercellular hyphae, and haustoria of the fungus. The formation and structure of urediniospores is similar to those of otherPuccinia species. The ultrastructure of intercellular hyphae and haustoria is similar to those of other rust fungi, but with some differences. No modifications are observed in the wall of the haustorial mother cells during penetration. A collar is found only around old haustoria. In most cases, one nucleus is detected inside the haustorial body and no nucleoli are seen in the nuclei of intercellular hyphae and haustoria. The host-parasite interface, including extrahaustorial matrix and extrahaustorial membrane, is also discussed and compared with those of other rust fungi.  相似文献

The new genus and species Teracosphaeria petroica is described for a perithecial ascomycete and its anamorph occurring on decayed wood collected in New Zealand. The fungus produces immersed, non-stromatic ceratosphaeria-like perithecia in nature, with hyaline, septate ascospores produced in unitunicate, non-amyloid asci. The anamorph produced in vitro is phialophora-like with lightly pigmented phialides terminating in flaring, deep collarettes that are often noticeably brown with conspicuous periclinal thickening. Phylogenetic analysis of LSU rDNA sequence data indicates that this fungus is distinct from morphologically similar fungi classified in the Chaetosphaeriales, the Trichosphaeriales or the Magnaporthaceae. It forms a monophyletic group with recently described, chaetosphaeria-like ascomycetes, such as the pyrenomycete genus Mirannulata, and shows affinity with the anamorphic species Dictyochaeta cylindrospora. The usefulness of describing anamorph genera for morphologically reduced anamorphs, when anamorph characteristics are actually part of the holomorph diagnosis, is discussed. An apparently contradictory example of the so-called Cordana and Pseudobotrytis anamorphs of Porosphaerella spp. is also discussed.  相似文献
