Many species of Melampsora on Populus have been reported in China, based on morphological characteristics of both uredial and telial states, and on host species, but their morphology and taxonomy are still poorly defined. In this study, 196 specimens representing Melampsora species on poplars and collected from various areas of China were used for morphological observations. The morphological characteristics of urediniospores and teliospores were examined with light and scanning electron microscopy. The specimens could be classified into five groups based on their morphology. For the sequencing of the nuclear large subunit rDNA (D1/D2), 5.8S rDNA and their internal transcribed spacers, ITS1 and ITS2 region, 54 specimens were selected from the specimens used in morphological observations. These specimens were separated into six clades by phylogenetic analyses of the D1/D2 and ITS regions. Correlations among morphological groups and phylogenetic clades based on these results suggest a revision of these species. A rust fungus found in Japan on Rhododendron kaempferi, R. kiusianum, and R. dauricum has previously been identified as Chrysomyxa rhododendri. Light and scanning electron microscopy of fresh and herbarium materials of the rust fungus, however, show that the spore surface morphology differs from the urediniospores of C. rhododendri, and the spores are slightly smaller. Furthermore, the DNA sequence of the 5′-end of the large subunit of ribosomal DNA differs from that of C. rhododendri by 3%. Telia have not been found; therefore, it is redescribed as a new anamorphic species, Caeoma tsukubaense. We analyzed the phylogenetic relationships of 49 specimens comprising 14 morphologically similar species of Pucciniastrum distributed in Japan based on the sequence data of the large subunit rDNA (D1/D2), 5.8S rDNA, and internal transcribed spacer
(ITS) regions. Neighbor-joining and parsimony analyses generated six major groups for both the D1/D2 and ITS regions. Pucciniastrum circaeae and P. epilobii formed a single group. P. hydrangeae-petiolaris, P. coryli, P. fagi, P. hikosanense, P. tiliae, and P. boehmeriae were each a distinct clade, and P. fagi formed a close relationship with P. hikosanense. However, these analyses did not support the monophyly of the following species: P. kusanoi, P. actinidiae, P. corni, P. styracinum, P. yoshinagai, and P. miyabeanum.

Phylogeny of the genus Pythium is analyzed based on sequences of the large subunit ribosomal DNA D1/D2 region and cytochrome oxidase II gene region of Pythium isolates and comprehensive species of related taxa belonging to the Oomycetes. The phylogenetic trees show that the genus Pythium is a highly divergent group and divided into five well- or moderately supported monophyletic clades. Each clade is characterized by sporangial morphology such as globose, ovoid, elongated, or filamentous shapes. Based on phylogeny and morphology, the genus Pythium (s. str.) is emended, and four new genera, Ovatisporangium, Globisporangium, Elongisporangium, and Pilasporangium, are described and segregated from Pythium s. lato. 相似文献

A new polypore, Perenniporia tibetica, collected in Xizang (Tibet), southwestern China, is described and illustrated on the basis of morphological and molecular characters. Perenniporia tibetica is characterized by an annual growth habit, resupinate basidiocarps with a creambuff to pinkish-buff pore surface and white to cream-colored rhizomorphs, a dimitic hyphal system with slightly dextrinoid and distinctly cyanophilous skeletal hyphae, and basidiospores that are ellipsoid, truncate or not, strongly dextrinoid, and cyanophilous, 6.7–8.7 × 5.3–6.8 μm. Its preliminary phylogenetic relationships are inferred based on sequence data from the ribosomal ITS and LSU regions, both suggesting that P. tibetica forms a distinct lineage in the genus Perenniporia. 相似文献

During a survey of bryophilous fungi from boreal and montane habitats in central Alberta, a hitherto undescribed species of Cladophialophora was recovered from Polytrichum juniperinum, Aulacomnium palustre, and Sphagnum fuscum. On potato dextrose agar (PDA) colonies grew slowly, attaining a diameter of 25 mm after 30 d, were dark grey, velvety, radially sulcate, and convolute and cracked at the centre. Micronematous conidiophores gave rise to branched chains of small (1–2 × 8–22 μm), cylindrical to fusiform conidia with truncate, swollen scars at each end. Phylogenies built on the ITS and ribosomal SSU regions indicate the isolates form a monophyletic clade within the family Herpotrichiellaceae (Chaetothyriales) that is composed of two geographically based groups, each with 99 % within-group sequence similarity and 97–98 % between-group sequence similarity. A teleomorph has not been found but would likely be similar to species of Capronia. In vitro inoculation of the isolates onto axenically grown P. juniperinum produced no discernible host symptoms, and host penetration could not be detected using light microscopy. The production of polyphenol oxidases by the fungus and the role of other Cladophialophora species as latent endophytes and saprobes suggest that a potential role for the fungus is the degradation of the polyphenol-rich cell walls of mosses. A new species ofNeosartorya, N. multiplicata (anam.Aspergillus multiplicatus), isolated from soil collected at Houli, Taichung, in Taiwan, is described and illustrated. The species is characterized by its restricted growth on Czapek agar, white ascomata, nearly globose ascospores with ribbed surface ornamentation of several linear ridges, and a limited development of conidia on common media. Pinus amamiana is an endangered Pinus species found only on Yakushima Island and Tanegashima Island, Japan. We surveyed remaining P. amamiana forests and found some sporocarps of Rhizopogon (Boletales), many species of which exhibit strict host specificity to a narrow range of Pinaceae trees and play critical roles in host establishment. Based on morphological characteristics and molecular phylogeny, here we describe Rhizopogon yakushimensis sp. nov. This new species belongs to a new clade, phylogenetically related to the subgenera Versicolores and Rhizopogon. We also confirmed its ectomycorrhizal association with P. amamiana by comparing rDNA ITS sequences between the sporocarps and ectomycorrhizal root tips. 相似文献

Nannizziopsis mirabilis, isolated from a soil sample collected from New Jersey, USA, is described as a new species. The ascomata are white to pale yellow, with a peridium of a network of loosely interwoven hyphae and simple, more or less straight, clavate appendages. The ascospores are hyaline, globose to subglobose, and ornamented with spiral bands or sometimes polygonal pits. The associated anamorph is assignable to the form genusChrysosporium. 相似文献

A pea rust fungus, Uromyces viciae-fabae, has been classified into two varieties, var. viciae-fabae and var. orobi, based on differences in urediniospore wall thickness and putative host specificity in Japan. In principal component analyses, morphological features of urediniospores and teliospores of 94 rust specimens from Vicia, Lathyrus, and Pisum did not show definite host-specific morphological groups. In molecular analyses, 23 Uromyces specimens from Vicia, Lathyrus, and Pisum formed a single genetic clade based on D1/D2 and ITS regions. Four isolates of U. viciae-fabae from V. cracca and V. unijuga could infect and sporulate on P. sativum. Powdery mildew fungi found on leaves, shoots, and stems of Phyllanthus acidus, P. amarus, and P. reticulatus proved to be a fungus having morphology unique in the Erysiphaceae. Light micrographs of a new germination pattern are added to discuss differences to other four germination patterns of the powdery mildews. The rDNA sequences (28S and 18S regions) of the fungi found on Phyllanthus spp. form a distinct monophyletic clade strongly supported by bootstrap (100%) in 18S + 28S trees, which indicates that the fungus is an isolated fungal group among the Erysiphaceae in tribal level. Because we cannot find the teleomorphic state of this fungus, a new subgenus Microidium of anamorphic genus Oidium is proposed to accommodate this organism. 相似文献

Uredinial and telial states of aMelampsora species occurring on the leaves ofSalix serissaefolia were for the first time recorded. Field observations and inoculation experiments showed that the spermogonial and aecial
states of the fungus were formed onChelidonium majus var.asiaticum andCorydalis incisa. The fungus was identified asMelampsora yezoensis based on the morphological observation of all the spore states. Urediniospores were able to infectS. erocarpa andS. pierotii as well asS. serissaefolia andS. jessoensis. Salix serissaefolia, S. eriocarpa, andS. pierotii are new uredinial and telial hosts ofM. yezoensis, andC. majus var.asiaticum andC. incisa are new spermogonial and aecial hosts of the fungus.

A new species ofRoumegueriella (Ascomycetes; Hypocreales),R. pulchella, is described and illustrated. This fungus is characterized by its rapid growth on Czapek-yeast extract and YpSs agars at 37°C, bright yellow non-ostiolate ascomata, translucent membranaceous peridium, broadly clavate asci, and hyaline one-celled subglobose-ovoid ascospores ornamented with prominent spines. A rust species on Calystegia soldanella in Japan has been treated as Puccinia convolvuli to date. However, morphological characteristics of specimens on C. soldanella collected from Japan are significantly different from those of specimens on other Calystegia and Convolvulus species from different areas of the world. It is proved by inoculation experiment that the rust on C. soldanella is specific to C. soldanella. Based on these results, Puccinia rust on C. soldanella from Japan is described as a new species, Puccinia calystegiae-soldanellae. 相似文献

A new species of Ulocladium was discovered from diseased leaves of Lycopersicon esculentum and Duchesnea indica from Hunan Province of China. Morphologically, this species is very close to U. consortiale, U. cucurbitae, and U. subcucurbitae in producing narrow ellipsoid conidia at 1–3 days, but the conidial size range of this species at this stage could distinguish it from three well-known species. It also
exhibits the multiplex conidium morphology at the different growth-stages (1–3 days and 4–7 days). The results of maximum
parsimony (MP) and neighbor-joining (NJ) phylogenetic analyses of combined glyceraldehyde-3-phosphate dehydrogenase (gpd) gene and Alternaria alternata major allergen (Alt a 1) genes show that U. solani and U. subcucurbitae cluster in a unique and separate subclade with no clear affinities to a specific sistergroup, and demonstrate that the Ulocladium species group is monphyletic, but two clades of this section are recognized.
