The changes in the activity of antioxidant enzymes such as superoxide dismutase (SOD: EC 1.15.1.1), catalase (CAT: EC 1.11.1.6), peroxidase (POX: EC 1.11.1.7), ascorbate peroxidase (APOX: EC 1.11.1.11) and glutathione reductase (GR: EC 1.6.4.2), free proline content, and the rate of lipid peroxidation level in terms of malondialdehyde (MDA) in roots of two rice cultivars (cvs.) differing in salt tolerance were investigated. Plants were subjected to three salt treatments, 0, 60, and 120 mol m−3 NaCl for 7 days. The results showed that activated oxygen species may play a role in cellular toxicity of NaCl and indicated differences in activation of antioxidant defense systems between the two cvs. The roots of both cultivars showed a decrease in GR activity with increase in salinity. CAT and APOX activities increased with increasing salt stress in roots of salt-tolerant cultivar Pokkali but decreased and showed no change, respectively, in roots of IR-28 cultivar. POX activity decreased with increasing NaCl concentrations in salt-tolerant Pokkali but increased in IR-28. SOD activity showed no change in roots of both cultivars under increasing salinity. MDA level in the roots increased under salt stress in sensitive IR-28 but showed no change in Pokkali. IR-28 produced higher amount of proline under salt stress than in Pokkali. Increasing NaCl concentration caused a reduction in root fresh weight of Pokkali and root dry weight of IR-28. The results indicate that improved tolerance to salt stress in root tissues of rice plants may be accomplished by increased capacity of antioxidative system.  相似文献

To investigate whether paraquat (PQ) is involved in regulation of antioxidant enzymes and lipid peroxidation under short-term salt stress, and to elucidate the physiological mechanism of salt stress mitigated by PQ, a cucumber cultivar (cv. Chunguang no. 2) was exposed to 100 mM NaCl for 48 h after pre-treatment with 10 μM PQ for 1 h. When compared to the control, salt stress increased the levels of malonaldehyde (MDA), superoxide radical (O2·−) and hydrogen peroxide (H2O2) and the activities of antioxidant enzymes, such as superoxide dismutase (EC 1.15.1.1), ascorbate peroxidase (EC 1.11.1.11) and glutathione reductase (EC 1.6.4.2) in the cucumber leaves. Under salt conditions, PQ pre-treatment prevented oxidative stress as observed by the decreases in MDA, H2O2 and O2·− that correlated with the increase in antioxidant defenses. We propose that, at low concentrations, the PQ pre-treatment can reduce the salt-induced oxidative damage by increasing the antioxidative mechanisms in cucumber plants.  相似文献

The purpose of this study was to investigate the mechanisms underlying alleviation of salt stress by mycorrhization. Solanum lycopersicum L. cultivars Behta and Piazar with different salinity tolerance were cultivated in soil without salt (EC?=?0.63 dSm?1), with low (EC?=?5 dSm?1), or high (EC?=?10 dSm?1) salinity. Plants inoculated with the arbuscular mycorrhizal fungi Glomus intraradices (+AMF) were compared to non-inoculated plants (?AMF). Under salinity, AMF-mediated growth stimulation was higher in more salt tolerant Piazar than in sensitive Behta. Mycorrhization alleviated salt-induced reduction of P, Ca, and K uptake. Ca/Na and K/Na ratios were also better in +AMF. However, growth improvement by AMF was independent from plant P nutrition under high salinity. Mycorrhization improved the net assimilation rates through both elevating stomatal conductance and protecting photochemical processes of PSII against salinity. Higher activity of ROS scavenging enzymes was concomitant with lowering of H2O2, less lipid peroxidation, and higher proline in +AMF. Cultivar differences in growth responses to salinity and mycorrhization could be well explained by differences in ion balance, photochemistry, and gas exchange of leaves. Function of antioxidant defenses seemed responsible for different AMF-responsiveness of cultivars under salinity. In conclusion, AMF may protect plants against salinity by alleviating the salt-induced oxidative stress.

Effects of exogenous nitric oxide (NO) on the germination and antioxidant enzyme during cucumber seed germination were investigated under salt stress. Seeds of cucumber (Cucumis sativus L. cv. Jinyou 1) were treated with distilled water or NaCl in the presence or absence of NO donor sodium nitroprusside (SNP) during germination. Excess 50 mM NaCl reduced significantly the seed germination rate in a short term and speed of germination. When salt concentration increased, germination of cucumber seed was reduced and the time needed to complete germination lengthened. Addition of exogenous SNP in salt solution attenuated the salt stress effects in a dose-dependent manner, as indicated by accelerating the seed germination, as well as weight increase of budding seeds, and 50 μM SNP was optimal concentration. At 150 mM NaCl, the 50 μM exogenous SNP significantly increased the activities of superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6) and protein content, while decreased the contents of malondialdehyde (MDA). There were no obvious effects of exogenous NO on peroxidase (POD, EC 1.11.1.6) and ascorbate peroxidase (APX, EC 1.11.1.6) activities under salt stress. Exogenous NO also increased the SOD and CAT isozyme expression under salt stress, which was in accordance with the improved antioxidant activities in the germinating seeds. The NO-induced salt stress resistance was associated with activated enzymes, and enhanced protein content, thus decreasing MDA content. It is concluded that exogenous NO treatment on cucumber seeds may be a good option to improve seed germination under saline conditions.  相似文献

The growth and productivity of cucumber are severely affected by salinity. To understand the complex salt response mechanism, the physiological and biochemical responses of cucumber seedlings to iso-osmotic NaCl and Ca(NO3)2 stresses were investigated. In this study, the biomass was significantly decreased under iso-osmotic NaCl and Ca(NO3)2 stresses, and the inhibitory effect of Ca(NO3)2 stress was less than that of NaCl stress. The soluble protein contents were increased under Ca(NO3)2 stress, whereas they were decreased after 6 days of NaCl stress. A sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis revealed that there were 14 differentially expressed protein bands in roots under iso-osmotic NaCl and Ca(NO3)2 stresses at 0, 3, 6, and 9 days, and seven protein bands were little expressed under NaCl stress at 6 and 9 days. Based on these results, 2-D gel electrophoresis was used to separate cucumber root proteins in response to iso-osmotic NaCl and Ca(NO3)2 stresses at 3 days. A total of 43 protein spots changed under salt stress. Of these proteins, 33 were successfully identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and liquid chromatography electro-spray ionization tandem mass spectrometry (LC-ESI-MS/MS) and categorized into classes, including those corresponding to antioxidants and defense-related proteins and energy and metabolism. The functions of the significantly differentially expressed root proteins were analyzed, which may facilitate a better understanding of different salt response mechanisms, and we suggest that cucumber seedlings showed a more powerful ability to resist Ca(NO3)2 stress.

The effects of 28-homobrassinolide (28-homoBL) on seedling growth, lipid peroxidation and antioxidative enzyme activities in the seedlings of Zea mays L. (var. Partap-1) under salt (NaCl) stress were studied. The surface-sterilized seeds were germinated in petriplates containing different concentrations of NaCl (25, 50, 75 and 100 mM) only, 28-homoBL (10−7, 10−9 and 10−11 M) only and NaCl supplemented with 28-homoBL for 7 days. The activities of superoxide dismutase (SOD, EC 1.15.1.1), guaiacol peroxidase (POD, EC 1.11.1.7), catalase (CAT, EC 1.11.1.6), ascorbate peroxidase (APOX, EC 1.11.1.11) and glutathione reductase (EC 1.6.4.2) were analysed in 7 day-old seedlings. It was observed that 28-homoBL treatments reduced the toxicity of salt on seedling growth considerably. Lipid peroxidation level was significantly increased under saline stress, but lowered with 28-homoBL applications revealing less oxidative damage. Further 28-homoBL treatments to the seedlings showed an enhancement in activities of SOD, POD, CAT and APOX. The activities of all antioxidative enzymes were further increased in seedlings treated with solution containing 28-homoBL and salt together as compared to seedlings treated with different concentration of salt solution only.  相似文献

A sand culture experiment was conducted to study ameliorative role of 28-homobrassinolide (HBL) in Brassica juncea seedlings raised from the seeds treated with water, or 10−10, 10−8 and 10−6 M 28-homobrassinolide (HBL) and grown in the presence of copper (50, 100 and 150 mg kg−1 sand) and sampled at 30 days after sowing. The plants grown in the presence of copper exhibited a significant decline in growth, chlorophyll and photosynthetic parameters. However, the activity of antioxidant enzymes: catalase (E.C. 1.11.1.6), peroxidase (E.C. 1.11.1.7) and superoxide dismutase (E.C. 1.15.1.1) and the content of proline increased in the plants grown under copper stress and/or raised from treatment with HBL. However, H2O2 content increased significantly in copper-treated plants and decreased in plants given HBL treatment. Treatment of seeds with HBL improved the growth, photosynthetic parameters and antioxidant enzymes and also improved in the plants grown under copper stress. The elevated antioxidant enzyme and proline might be responsible to overcome the toxic effects of copper in B. juncea.  相似文献

The effects of NaCl stress on the activity of antioxidant enzymes, lipid peroxidation, cell membrane stability, net photosynthetic rate, gas-exchange, and chlorophyll content were investigated in two Jerusalem artichoke cultivars, Dafeng (salt-tolerant) and Wuxi (salt-sensitive), grown under control (nutrient solution) or salt stress (nutrient solution containing 75, 150, and 225 mM NaCl) conditions for 7 days. In leaves of salt-tolerant cv. Dafeng, superoxide dismutase (EC 1.15.1.1), peroxidase (EC 1.11.1.7), and catalase (EC 1.11.1.6) activities significantly increased as compared to the controls, whereas no significant change was observed in cv. Wuxi. Lipid peroxidation and cell membrane injury were enhanced in both cultivars. Net photosynthesis and stomatal conductance decreased in response to salt stress, but cv. Dafeng showed a smaller reduction in photosynthesis than cv. Wuxi. The results indicated that stomatal aperture limited leaf photosynthetic capacity in the NaCl-treated plants of both cultivars. However, significant reduction in the leaf chlorophyll content due to NaCl stress was observed only in cv. Wuxi. These results suggested that salt-tolerant Jerusalem artichoke varieties may have a better protection against reactive oxygen species, at least in part, by increasing the activity of antioxidant enzymes under salt stress.  相似文献

Plant growth, photosynthetic parameters, chloroplast ultrastructure, and the ascorbate-glutathione cycle system in chloroplasts of self-grafted and rootstock-grafted cucumber leaves were investigated. Grafted plants were grown hydroponically and were exposed to 0, 50, and 100 mM NaCl concentrations for 10 days. Under NaCl stress, the hydrogen peroxide (H2O2) content in cucumber chloroplasts increased, the chloroplast ultrastructure was damaged, and the gas stomatal conductance, intercellular CO2 concentration, as well as shoot dry weight, plant height, stem diameter, leaf area, and leaf relative water content were inhibited, whereas these changes were less severe in rootstock-grafted plants. The activities of ascorbate peroxidase (APX; EC 1.11.1.11), glutathione reductase (GR; EC 1.6.4.2), and dehydroascorbate reductase (DHAR EC 1.8.5.1) were higher in the chloroplasts of rootstock-grafted plants compared with those of self-grafted plants under 50 and 100 mM NaCl. Similar trends were shown in leaf net CO2 assimilation rate and transpiration rate, as well as reduced glutathione content under 100 mM NaCl. Results suggest that rootstock grafting enhances the H2O2-scavenging capacity of the ascorbate–glutathione cycle in cucumber chloroplasts under NaCl stress, thereby protecting the chloroplast structure and improving the photosynthetic performance of cucumber leaves. As a result, cucumber growth is promoted.  相似文献

Five cultivars of tomato having different levels of salt stress tolerance were exposed to different treatments of NaCl (0, 3 and 6 g L−1) and ZnO-NPs (0, 15 and 30 mg L−1). Treatments with NaCl at both 3 and 6 g L−1 suppressed the mRNA levels of superoxide dismutase (SOD) and glutathione peroxidase (GPX) genes in all cultivars while plants treated with ZnO-NPs in the presence of NaCl, showed increments in the mRNA expression levels. This indicated that ZnO-NPs had a positive response on plant metabolism under salt stress. Superior expression levels of mRNA were observed in the salt tolerant cultivars, Sandpoint and Edkawy while the lowest level was detected in the salt sensitive cultivar, Anna Aasa. SDS–PAGE showed clear differences in patterns of protein expression among the cultivars. A negative protein marker for salt sensitivity and ZnO-NPs was detected in cv. Anna Aasa at a molecular weight of 19.162 kDa, while the tolerant cultivar Edkawy had two positive markers at molecular weights of 74.991 and 79.735 kDa.  相似文献

Improvement in salinity tolerance of plants is of immense significance as salt stress particularly threatens the productivity of agricultural crops. This study was designed to assess the tolerance level of six Brassica napus varieties (Super, Sandal, Faisal, CON-111, AC Excel and Punjab) under different levels of salinity (0, 50, 100, 150 & 200 mM) with three replications under CRD. Salt induced osmotic stress curtailed the plant growth attributes, photosynthetic pigments and disturbed ionic homeostasis (K+, Na+, Ca2+, Cl-) but least disturbance as compared to control was found in Super and Sandal cultivars. Punjab canola and AC Excel canola cultivars were least tolerant to salinity because these displayed greater decline in all growth and biochemical attributes. Plants subjected to NaCl induced stress exhibited considerable decline in all attributes under study with proline as exception. Antioxidants (CAT, SOD & POD) showed an obvious change in Canola plants under stress, but greatest decline was displayed at 200 mM NaCl level in all six cultivars. Over all these attributes presented a comparatively stable trend in super and sandal cultivars. This shows presence of physiological resilience and metabolic capacity in these two cultivars to tackle salinity. Similarly, all yield attributes displayed adverse behavior under 150 mM & 200 mM salinity stress. Our results demonstrated that Super and Sandal cultivars of Brassica napus exhibit good performance in salinity tolerance and can be good option for cultivation in salt affected areas.  相似文献

We investigated the effects of silicon (Si) on time-dependent changes in root tonoplast H+-ATPase and H+-PPase activities, membrane fatty acid compositions and tonoplast fluidity in two barley (Hordeum vulgare L.) cultivars differing in salt tolerance. Plants were grown in NaCl-free (control) and NaCl-supplied (60 and 120 mM, respectively) nutrient solutions with or without 1.0 mM Si. Plant roots were harvested to isolate tonoplast vesicles for assay of H+-ATPase and H+-PPase activities at days 2, 4, and 6 after treatment in the first experiment and for analysis of membrane fatty acid composition and fluidity at day 4 after treatment in the second experiment. The results showed that tonoplast H+-ATPase and H+-PPase activities in roots of salt-treated plants increased at day 2, which was more obvious at 60 mM NaCl in the salt-tolerant cultivar than in the salt-sensitive cultivar, and then decreased at day 4 and onward. These enzyme activities decreased consistently from days 2 to 6 for treatment with 120 mM NaCl. However, inclusion of 1.0 mM Si significantly enhanced both H+-ATPase and H+-PPase activities in roots of salt stressed barley, which was irrespective of NaCl level or cultivar used. The ratio of unsaturated to saturated fatty acids (U/S) increased under salt stress for both cultivars. Addition of Si to salt treatment increased the ratio of U/S in salt-tolerant cultivar but it did not in salt-sensitive cultivar compared to non-Si-amended salt treatment. Salt treatment decreased tonoplast fluidity of roots of barley significantly compared with control treatment. However, root tonoplast fluidity was significantly lower in the Si-amended salt treatment than in the non-Si-amended salt treatment. These results were in line with the previous findings that Si could help increase antioxidative defense and reduce membrane lipid oxidative damage in barley under salt stress. The possible mechanisms involved in Si-enhanced salt tolerance were discussed with respect to cell membrane integrity, stability and function in barley.  相似文献

Salt tolerance of sorghum varieties in terms of fresh weight, ion accumulations, proline content and peroxidase activity was analyzed in this study. Three sorghum varieties, Payam, Kimia, and Jambo, differing in salt tolerance, were grown in a greenhouse-hydroponic culture with a complete nutrition solution to which 0, 50, 100, 150 and 200 mM NaCl was added. Plant roots and leaves were harvested at 15 and 30 days after treatment and subjected to analysis. Clear decline in K+ and Ca2+ concentrations and increase in Na+ and proline contents were observed in the root and leaf tissues at each NaCl concentration in all varieties during the NaCl treatment. The Ca2+ concentration in leaves was higher than in roots, and had the following order in the tested cultivars: Jambo, Kimia, and Payam. Total peroxidase activity increased under salinity stress and it was proportional with the salt concentration. Payam had the largest decrease (46.95%) in fresh weight caused by NaCl, while Jambo had the lowest decrease, 28.63%. Linear regression analysis revealed significant relationships between the estimated factors and fresh weight. The profiles of isoperoxidases were modified under stress conditions. Two isoforms, A1 and A2, were detected in all three varieties with different intensities. Under NaCl stress, isoperoxidases were strongly expressed and a third isoform, A3, was specifically found in variety Jambo suggesting that A3 is implicated in salt adaptation of this variety.  相似文献

In the present study, ameliorative capabilities of wuxal amino (bio stimulant) under salt stress has been investigated through adaptive mechanisms and antioxidant potential in tomato plants. In the experiment, two different concentrations (2 cm L-1 and 3 cm L-1) of wuxal amino through foliar application and soil irrigation were applied to the salt (150 mM) treated tomato plants and then morphological traits, photosynthetic pigments, osmolytes, secondary metabolites, oxidative stress and antioxidant enzymes activity were assessed at 60 days after planting. The results revealed that salt stress decreased the growth parameters, photosynthetic pigments, soluble sugars and soluble protein whereas, content of proline, ascorbic acid, total phenols, malondialdehyde, hydrogen peroxide and the activity of antioxidant enzymes activity increased under salt stress. Moreover, Wuxal amino application through foliar or soil to salt stressed plants improved morphological traits, photosynthetic pigments, osmolytes, total phenol and antioxidant enzymes activity. Interestingly, the deleterious impact of salinity on tomato plants were significantly reduced and it can be evident from reduced MDA and H2O2 levels. These responses varied with the mode (foliar or soil) of application of Wuxal amino under different concentrations (2 cm L-1 and 3 cm L-1). It was concluded that application of Wuxal amino (2 cm L-1, foliar) and (3 cm L-1; soil) proved best and could be commercially used as eco-friendly tool for the protection of tomato plants grown under salinity stress.  相似文献

Effects of salt stress on the time course of stomatal behaviors and the activity of antioxidative enzymes such as catalase (CAT) (EC 1.11.1.6), ascorbate peroxidase (APX) (EC 1.11.1.11), and glutathione reductase (GR) (EC. 1.6.4.2) were studied in three strawberry cultivars. The responses of the cultivars ‘Camarosa’, ‘Tioga,’ and ‘Chandler’ were compared when they were irrigated with nutrient solution containing 0, 8.5, 17.0, and 34.0 mM sodium chloride (NaCl) for 30 days. A significant reduction in stomatal conductance (gs) was seen particularly on the 30th day of the salt treatments only in Camarosa, which is parallel to transpiration rate (E). CAT activities decreased in all of the salt treatments only in Tioga, while it remained almost unchanged or slightly increased depending on the period in Camarosa and Chandler. APX activity sharply increased in 17.0 and 8.5-mM NaCl treatments for 30 days in Camarosa and Tioga, respectively, whereas it linearly increased based on the NaCl treatments in Chandler. On the other hand, only Camarosa demonstrated a sharp increase in GR activity induced by salinity applied for 30 days. All the data indicated that control of the stomatal behavior, the higher salt-stress tolerance (LT50) and higher constitutive activity of antioxidant enzymes made Camarosa and Tioga relatively salt-tolerant cultivars.  相似文献

A pot study was conducted to determine the effects of arbuscular mycorrhizal (AM) fungi (Glomus mosseae and Paraglomus occultum) and salt (NaCl) stress on growth, photosynthesis, root morphology and ionic balance of citrus (Citrus tangerine Hort. ex Tanaka) seedlings. Eighty-five-day-old seedlings were exposed to 100 mM NaCl for 60 days to induce salt stress. Mycorrhizal colonization of citrus seedlings was not affected by salinity when associated with P. occultum, but significantly decreased when with G. mosseae. Compared with the non-mycorrhizal controls, mycorrhizal seedlings generally had greater plant height, stem diameter, shoot, root and total plant biomass, photosynthetic rate, transpiration rate and stomatal conductance under the 0 and 100 mM NaCl stresses. Root length, root projected area and root surface area were also higher in the mycorrhizal than in the non-mycorrhizal seedlings, but higher root volume in seedlings with G. mosseae. Leaf Na+ concentrations were significantly decreased, but leaf K+ and Mg2+ concentrations and the K+/Na+ ratio were increased when seedlings with both G. mosseae and P. occultum. Under the salt stress, Na+ concentrations were increased but K+ concentrations decreased in the mycorrhizal seedlings. Under the salt stress, Ca2+ concentrations were increased in the seedlings with P. occultum or without AM fungi (AMF), but decreased with G. mosseae. Ratios of both Ca2+/Na+ and Mg2+/Na+ were also increased in seedlings with G. mosseae under the non-salinity stress, while only the Mg2+/Na+ ratio was increased in seedlings with P. occultum under the salt stress. Our results suggested that salt tolerance of citrus seedlings could be enhanced by associated AMF with better plant growth, root morphology, photosynthesis and ionic balance.  相似文献

Mustard (Brassica juncea L.) cultivars Alankar (salt-tolerant) and PBM16 (salt-sensitive) plants were grown with 50 mM NaCl and were sprayed with 0.1, 0.5, and 1.0 mM salicylic acid (SA) to study the physiological processes determining salt tolerance and to observe the influence of SA application on the alleviation of NaCl-induced adverse effects. The content of leaf Na+, Cl, H2O2, TBARS, and electrolyte leakage and the activity of SOD were higher in PBM16 than Alankar. In contrast, nutrients content, activity of APX and GR, glutathione content, photosynthetic and growth characteristics were higher in Alankar. Treatment of 50 mM NaCl resulted in increase of Na+ and Cl, oxidative stress, activity of antioxidant enzymes and glutathione content, while nutrients content, photosynthetic, and growth characteristics decreased in both the cultivars. Application of 0.5 mM SA alleviated the negative effects of 50 mM NaCl maximally, but 1.0 mM SA proved inhibitory. The effect of SA was more conspicuous in Alankar than PBM16. It is concluded that the higher tolerance of Alankar was due to its lower leaf Na+ and Cl content, higher nutrients content, and efficient antioxidant metabolism. The application of 0.5 mM SA substantially alleviated salt-induced adverse effects in Alankar.  相似文献

This study evaluated the effects of foliar spraying melatonin (MT) on the growth of salt-stressed rice. Seedlings were treated with 50 and 100 mM of NaCl and different concentrations of MT (25, 50, 100, 200, 300, and 400 μM) for 14 days. Different concentrations of MT could promote plant growth significantly under salt stress, particularly at concentrations of 200, 300, and 400 μM. A concentration of 200 μM MT was considered as optimal and used in a subsequent experiment on biomass, water content, antioxidation, mineral nutrition, salt absorption, and distribution of salt-stressed rice seedlings. Results showed that MT’s promoting effect on plant growth under salt stress was evident with time, particularly under high salt stress. MT improved the activities of antioxidant enzymes, reduced membrane lipid peroxidation, alleviated cell injury in plant leaves, and increased N content and Si accumulation in the leaves and roots under salt stress, particularly under high salinity. This compound also inhibited Na uptake and upward transport, but it promoted or maintained the uptake and upward transport of K and Ca in salt-stressed rice. Thus, MT improved the ion homeostasis of K/Na and Ca/Na in plants, particularly in the leaves. Foliar spraying of MT alleviated salt stress on rice by promoting nutrient accumulation or translocation, improving ion homeostasis, which is evident in the leaves, and consequently enhancing its salt resistance. The antioxidative improvement caused by MT might also be related to the improved ion homeostasis.
