社交互动的奖励特性对于社会行为的表达和适应性社会关系的发展至关重要。当个体受到应激时会导致奖赏系统异常而缺乏社会交往,出现情绪障碍并具有性二态。青春期发育中社会奖赏行为存在显著的性别差异,男性对社会奖赏敏感性大于女性;相反,女性对社会惩罚的敏感性高于男性。在青春期发育过程中,催产素/加压素(OT/AVP)、多巴胺(DA)系统在奖赏环路的性别差异及OT/AVP受体基因表达的多态性,是奖赏行为及情绪障碍性二态的原因,揭示OT-社会奖赏-情绪障碍性二态三者交互作用的神经机制,对开展精神疾病治疗有重要指导意义。  相似文献

精氨酸加压素(AVP)和催产素(OT)能够调节社会行为,并通过调节多巴胺(DA)的活动参与药物成瘾。棕色田鼠Lasiopodomys mandarinus是一种单配制田鼠,具有较复杂的社会行为。本研究以酪氨酸羟化酶(TH)标记DA神经元,对雌性棕色田鼠连续注射可卡因[20 mg·(kg·d)~(-1)]4 d,24 h后测量其运动性、焦虑水平和社会行为以及AVP、OT和TH的变化。结果发现,与对照组相比,可卡因组运动性提高,社会探究行为和攻击行为下降,但焦虑水平差异没有统计学意义。同时,可卡因组下丘脑前区AVP和下丘脑室旁核OT的免疫活性神经元数量减少;室旁核和中脑腹侧被盖区TH的免疫活性神经元数量增加。这些结果说明反复暴露于可卡因能够改变雌性棕色田鼠的行为敏感化及社会行为。AVP、OT和DA参与了这一调节过程。  相似文献

本研究通过对雌雄子午沙鼠进行新物体识别和社会认知实验,运用免疫组化方法检测其相关脑区合成催产素(OT)、加压素(AVP)和多巴胺(DA)能的神经元数量,采用酶联免疫试验(ELISA)方法检测了其血清中OT、AVP的水平,探究了雌雄子午沙鼠的两性认知差异及其神经内分泌水平的差异。结果表明,雌雄子午沙鼠对新物体的探究时间均要显著高于旧物体,雌雄子午沙鼠的辨别指数无显著差异(P>0.05);雄性子午沙鼠随着探究次数的增加对重复刺激鼠a的探究时间不断减少,对陌生刺激鼠b的探究时间显著高于刺激鼠a(P<0.05);雌性子午沙鼠没有此趋势。雄性子午沙鼠OT能神经元数量在下丘脑室旁核(PVN)和视上核(SON)均要显著少于雌性(P<0.05);雄性个体DA能神经元数量在黑质显著高于雌性(P<0.01);然而雄性个体DA能神经元数量在腹侧被盖区显著少于雌性(P<0.01);雌雄子午沙鼠血清OT、AVP水平均无显著差异。综上所述,雌雄子午沙鼠对新物体的识别能力无显著差异,然而雄性子午沙鼠的社会认知能力强于雌性。在神经内分泌水平上,雌雄子午沙鼠PVN和SON中OT能神经元数量、黑质和腹侧背盖区的DA能神经元数量均呈现出了两性差异。  相似文献

同伴间的社会互作是一种天然奖赏,能够影响成瘾药物使用的敏感性。催产素(Oxytocin,OT)能够调节社会行为,并提高社会互作的奖赏价值。然而不同背景的同伴互作以及与OT合并使用是否对可卡因的奖赏效应有不同的影响尚不清楚。棕色田鼠(Microtus mandarinus)是一种单配制田鼠,个体间具有较复杂的社会行为。利用雌性棕色田鼠,我们首先检测了可卡因(20 mg/kg)单独强化以及与不同背景关系的同伴(熟性雌性、陌生雌性和陌生雄性)同时强化诱导的条件位置偏爱(conditioned place preference, CPP)及持续时间;其次检测了实验鼠外周注射OT (1 mg /kg)并给予不同背景同伴强化对可卡因CPP的影响。结果表明,可卡因单独强化时,实验鼠能够形成可卡因CPP并能持续至3周;用熟悉的雌性同伴强化时,实验鼠对可卡因CPP的维持时间缩短;用陌生的雌性或雄性同伴强化时可抑制可卡因CPP的形成。实验鼠注射OT后,用熟悉雌性或陌生雄性同伴分别强化时会抑制或反转可卡因CPP。这些结果表明不同背景关系的同伴强化对可卡因奖赏效应的影响不同。OT可促进同伴强化的奖赏价值,降低动物对可卡因的偏爱,且该效应因强化同伴的背景而不同。  相似文献

目的:探讨白细胞介素-1β(IL-1β)对精氨酸升压素(AVP)诱导下大鼠心肌成纤维细胞(CFs)诱导型一氧化氮合酶(iNOS)-一氧化氮(NO)系统活性的影响。方法:胰酶消化法分离培养SD仔鼠CFs,硝酸还原酶法、分光光度法和逆转录-聚合酶链式反应(RT—PCR)分别测定不同浓度IL-1β与AVP协同作用下CFs的NO含量、NOS活性和iNOSmRNA表达。结果:AVP诱导下CFs iNOSmRNA表达、NOS活性和NO合成均显著增加(P〈0.05)。一定浓度范围内IL-1β与AVP协同作用,剂量依赖性地增加AVP对CFs iNOS-NO系统活性的提高作用,其中AVP+3ng/ml和AVP+5ng/ml IL-1β组的iNOS mRNA表达、NOS活性和NO合成均显著高于AVP组(P〈0。05),但IL-1β浓度增加至5ng/ml时,CFs的iNOSmRNA表达、NOS活性和NO合成不再继续升高,反而有所下降。结论:在一定浓度范围内IL-1β可与AVP协同提高CFs iNOS-NO系统活性。  相似文献

通过记录分析雌雄棕色田鼠同性间的社会互作并用免疫组织化学方法对雌激素α受体(ERα)在雌雄棕色田鼠脑内的分布进行定位,以揭示ERα在不同性别棕色田鼠社会行为中的调控作用。结果发现:雌性棕色田鼠攻击行为显著多于雄性;雌性棕色田鼠亲密行为明显少于雄性,差异极显著;此外,雌性棕色田鼠的防御行为极显著多于雄性。免疫组织化学结果显示:ERα免疫阳性细胞主要分布在弓状核(ARC)、杏仁内侧核(MeA)、杏仁中央核(Ce)、下丘脑视前区(MPOA)、下丘脑腹内侧核(VMH)和终纹床核(BST),其中ERα在BST、MPOA、MeA和Ce中的分布存在着极显著性二型性,且雌性田鼠表达的ERα较多;在ARC中的分布也存在显著性二型性,雄性田鼠表达的ERα多于雌性田鼠;ERα在VMH中的分布无明显的性二型性。结果揭示了雌雄棕色田鼠在同性间社会互作中攻击、防御以及亲密行为存在显著差异,而ERα在雌雄棕色田鼠脑内的分布模式也有显著性差异,社会互作和ERα免疫阳性细胞分布的两性差异都呈现单配制鼠类的特征,ERα在大脑分布模式的两性差别和不同种类间的差别可能是单配制啮齿类呈现相关生殖和社会行为的一个重要机制[动物学报54(6):1020-1028,2008]。  相似文献

摘要 目的:探讨长链非编码核糖核酸(LncRNA)KCNQ1OT1靶向调控miR-124-3p/高迁移率族蛋白B1(HMGB1)轴对高糖诱导肾小球系膜细胞(HMC)增殖、凋亡及纤维化的影响。方法:将人HMC分为对照组(NC组)、高糖组(30 mmol/L葡萄糖)、阴性序列(si-NC)组、KCNQ1OT1小干扰核糖核酸(RNA)(si-KCNQ1OT1)组、si-KCNQ1OT1+模拟对照序列(miR-NC)组、si-KCNQ1OT1+miR-124-3p抑制剂(miR-124-3p inhibitor)组,各组在转染后进行高糖处理。实时荧光定量聚合酶链式反应(RT-qPCR)检测LncRNA KCNQ1OT1信使核糖核酸(mRNA)、miR-124-3p mRNA、HMGB1 mRNA表达;四甲基偶氮唑盐比色法(MTT)检测细胞增殖活性;流式细胞术检测细胞凋亡率;蛋白印迹法(Western blot)检测HMGB1蛋白、增殖相关蛋白[细胞周期蛋白1(CyclinD1)]、细胞凋亡蛋白[半胱氨酸蛋白酶-3(caspase-3)、半胱氨酸蛋白酶蛋白9(caspase-9)]、细胞纤维化蛋白[纤维连接蛋白(FN)、细胞黏附分子-1(ICAM-1)]表达;双荧光素酶报告基因实验验证LncRNA KCNQ1OT1与miR-124-3p与HMGB1之间的靶向关系。结果:与NC组比较,高糖组KCNQ1OT1 mRNA、HMGB1 mRNA及HMGB1蛋白、CyclinD1蛋白、FN蛋白、ICAM-1蛋白表达、HMC活性(24 h、48 h和72 h)明显上升,miR-124-3p mRNA、caspase-3蛋白及caspase-9蛋白表达、HMC凋亡率明显下降(P<0.05);与高糖组、si-NC组比较,si-KCNQ1OT1组KCNQ1OT1 mRNA、HMGB1 mRNA及HMGB1蛋白、CyclinD1蛋白、FN蛋白、ICAM-1蛋白表达、HMC活性(24 h、48 h和72 h)明显下降,miR-124-3p mRNA、caspase-3蛋白及caspase-9蛋白表达、HMC凋亡率明显上升(P<0.05);与si-KCNQ1OT1组、si-KCNQ1OT1+miR-NC组比较,si-KCNQ1OT1+miR-124-3p inhibitor组HMGB1 mRNA及HMGB1蛋白、CyclinD1蛋白、FN蛋白、ICAM-1蛋白表达、HMC活性(24 h、48 h和72 h)明显上升,miR-124-3p mRNA、caspase-3蛋白及caspase-9蛋白表达、HMC凋亡率明显下降(P<0.05)。较miR-NC组与KCNQ1OT1-WT共转染组而言,miR-124-3p mimic组与KCNQ1OT1-WT共转染组细胞荧光素酶活性明显降低(P<0.05);较miR-NC组与HMGB1-WT共转染组而言,miR-124-3p mimic组与HMGB1-WT共转染组细胞荧光素酶活性明显降低(P<0.05)。结论:LncRNA KCNQ1OT1可以靶向下调miR-124-3p mRNA表达,上调HMGB1 mRNA及HMGB1蛋白表达,促进高糖诱导HMC增殖,抑制凋亡,促进细胞纤维化发展。  相似文献

催产素(OT)是一种9肽激素,主要由哺乳动物下丘脑产生,以神经内分泌,旁分泌或自分泌形成,在哺乳动物生殖过程中发挥重要作用,催产素受体(OTR)是与G-蛋白相耦联的膜蛋白,通过激活磷脂酶C发挥其生理作用,OT在交配,分娩,哺池时由神经垂体(垂体后叶)脉冲式释放,促进子宫平滑肌和乳腺肌上皮细胞收缩,利用精子运行,胎儿娩出和射出乳汁,OT在中枢神经系统中参与调节母性行为,在性腺中促进某些物种的黄体形成,OT与PGF2a共同作用使有蹄动物黄体退化,以上过程都依赖于OT和OTR基因的时空特异性表达,多种激素参与它们的表达调控,但OT的生理作用有时也可被其它途径所替代。  相似文献

The molecular and neural mechanisms regulating human social-emotional behaviors are fundamentally important but largely unknown; unraveling these requires a genetic systems neuroscience analysis of human models. Williams Syndrome (WS), a condition caused by deletion of ~28 genes, is associated with a gregarious personality, strong drive to approach strangers, difficult peer interactions, and attraction to music. WS provides a unique opportunity to identify endogenous human gene-behavior mechanisms. Social neuropeptides including oxytocin (OT) and arginine vasopressin (AVP) regulate reproductive and social behaviors in mammals, and we reasoned that these might mediate the features of WS. Here we established blood levels of OT and AVP in WS and controls at baseline, and at multiple timepoints following a positive emotional intervention (music), and a negative physical stressor (cold). We also related these levels to standardized indices of social behavior. Results revealed significantly higher median levels of OT in WS versus controls at baseline, with a less marked increase in AVP. Further, in WS, OT and AVP increased in response to music and to cold, with greater variability and an amplified peak release compared to controls. In WS, baseline OT but not AVP, was correlated positively with approach, but negatively with adaptive social behaviors. These results indicate that WS deleted genes perturb hypothalamic-pituitary release not only of OT but also of AVP, implicating more complex neuropeptide circuitry for WS features and providing evidence for their roles in endogenous regulation of human social behavior. The data suggest a possible biological basis for amygdalar involvement, for increased anxiety, and for the paradox of increased approach but poor social relationships in WS. They also offer insight for translating genetic and neuroendocrine knowledge into treatments for disorders of social behavior.  相似文献

The goal of our study was to explore the effect of social isolation stress of varying durations on the plasma oxytocin (OT), messenger ribonucleic acid (mRNA) for oxytocin receptor (OTR), plasma arginine vasopressin (AVP) and mRNA for V1a receptor of AVP (V1aR) expression in the hypothalamus and heart of socially monogamous female and male prairie voles (Microtus ochrogaster). Continuous isolation for 4 weeks (chronic isolation) increased plasma OT level in females, but not in males. One hour of isolation every day for 4 weeks (repeated isolation) was followed by a significant increase in plasma AVP level. Chronic isolation, but not repeated isolation, significantly decreased OTR mRNA in the hypothalamus and heart in both sexes. Chronic isolation significantly decreased cardiac V1aR mRNA, but no effect on hypothalamic V1aR mRNA expression. We did not find a gender difference within repeated social isolation groups. The results of the present study reveal that although chronic social isolation can down-regulate gene expression for the OTR in both sexes, the release of the OT peptide was increased after chronic isolation only in females, possibly somewhat protecting females from the negative consequences of isolation. In both sexes repeated, but not chronic, isolation increased plasma AVP, which could be permissive for mobilization and thus adaptive in response to a repeated stressor. The differential effects of isolation on OT and AVP systems may help in understanding mechanisms through social interactions can be protective against emotional and cardiovascular disorders.  相似文献

Oxytocin (OT) and arginine-vasopressin (AVP) are 2 peptides that are produced in the brain and released via the pituitary gland to the peripheral blood, where they have diverse physiological functions. In the last 2 decades it has become clear that these peptides also play a central role in the modulation of mammalian social behavior by their actions within the brain. Several lines of evidence suggest their involvement in autism spectrum disorder (ASD), which is known to be associated with impaired social cognition and behavior. Recent clinical trials using OT administration to autistic patients have reported promising results. Here, we aim to describe the main data that suggest a connection between these peptides and ASD. Following a short illustration of several major topics in ASD biology we will (a) briefly describe the oxytocinergic and vasopressinergic systems in the brain, (b) discuss a few compelling cases manifesting the involvement of OT and AVP in mammalian social behavior, (c) describe data supporting the role of these peptides in human social cognition and behavior, and (d) discuss the possibility of the involvement of OT and AVP in ASD etiology, as well as the prospect of using these peptides as a treatment for ASD patients.  相似文献

Oxytocin (OT) has been shown to play an important role in social bonding in animals. However, it is unclear whether OT is related to inter-species social bonding. In this study, to examine the possibility that urinary OT concentrations of owners were increased by their “dog's gaze”, perhaps representing social attachment to their owners, we measured urinary OT concentrations of owners before and after interaction with their dogs. Dog owners interacted with their dogs as usual for 30 min (interaction experiment) or were instructed not to look at their dogs directly (control experiment). We observed the behaviors of owners and their dogs during the experiments, and measured OT concentrations by radioimmunoassay in urine samples from the owners collected just before and 20 min after interaction with their dogs. Using a cluster analysis, owners could be divided into two groups: one received a longer duration of gaze from their dogs and reported a higher degree of relationship with their dogs (LG); the other received a shorter duration of gaze and reported a lower degree of relationship (SG). Urinary OT was higher in LG than SG after usual interaction with their dogs, but not in the control experiment. In the interaction experiment, a high correlation was found in LG between the frequency of behavioral exchanges initiated by the dog's gaze and the increase in urinary OT. We conclude that interactions with dogs, especially those initiated by the dog's gaze, can increase the urinary OT concentrations of their owners as a manifestation of attachment behavior.  相似文献

This study examined the effects of i.v. administration of cholecystokinin-tetrapeptide (CCK-4) on plasma release of arginine vasopressin (AVP) and oxytocin (OT) in women with premenstrual dysphoric disorder (PMDD) and control women, during both the follicular phase and the luteal phase of their menstrual cycle. Plasma AVP and OT concentrations increased following CCK-4 administration. AVP and OT response to CCK-4 was similar for PMDD and control women and unaffected by menstrual cycle phase. AVP and OT may play a role in the hypothalamo-pituitary adrenal (HPA) axis activity associated with the panic response induced by CCK-4.  相似文献

Animal studies have implicated the neuropeptides Prolactin (PRL) and Oxytocin (OT) in processes of maternal bonding and PRL has similarly been shown to play a role in the neurophysiology of fatherhood. Yet, very little is known on the involvement of PRL and OT in human fathering. Forty-three fathers and their firstborn infant were seen twice: in the second and sixth postpartum months. Paternal plasma PRL and OT were sampled at both time-points and analyzed with ELISA methods. At six months fathers were videotaped interacting with their child in social and exploratory play contexts and interactions were micro-analyzed for father-infant Affect Synchrony and father facilitation of child toy exploration. PRL and OT showed high individual stability across time and were correlated at the second observation. PRL was related to father-infant Coordinated Exploratory Play in the toy context whereas OT was associated with father-infant Affect Synchrony in the social context. Results point to the role of PRL and OT in the development of human fathering and underscore their differential relations with patterns of paternal care.  相似文献

A non-invasive assay for measurement of oxytocin (OT) and vasopressin (AVP) in primates would enable researchers to study the relationship between the endocrine system and behavior without disturbing potentially endangered animals in their natural habitats. In order to test whether or not OT specifically would be measurable in the urine of a primate, 10 microCi of tritium-labeled OT were injected into the peripheral blood supply of four adult male common marmosets (Callithrix jacchus), with continuous urinary collection over 48 h. When urine was processed by HPLC separation and beta counting for radioactive clearance, the label was present in all samples in the fraction where OT elutes. Large amounts of OT were also seen in a fraction other than that containing the OT standard, indicating that OT is measurable but that it also undergoes substantial metabolic breakdown. In a second experiment, we isolated six common marmosets for 48 h and then exposed them to social contact to evaluate the effect of changing social stimuli on endogenous urinary measurement of both OT and AVP. Both were measured after HPLC separation to isolate the intact molecule and also to control for cross-reactivity with metabolites in subsequent RIA. Cortisol was also measured to objectively evaluate the stress response. A priori assumptions were that urinary OT and AVP would be lower during a period of isolation and higher during periods of social contact. These assumptions were met, leading us to conclude that peripheral OT and AVP are measurable via urinary assay and that such an assay is a valid means of evaluating social condition in this species.  相似文献

Electrophysiological recordings in lactating rats show that oxytocin (OT) and vasopressin (AVP) neurons exhibit specific patterns of activities in relation to peripheral stimuli: periodic bursting firing for OT neurons during suckling, phasic firing for AVP neurons during hyperosmolarity (systemic injection of hypertonic saline). These activities are autocontrolled by OT and AVP released somato-dentritically within the hypothalamic magnocellular nuclei. In vivo, OT enhances the amplitude and frequency of bursts, an effect accompanied with an increase in basal firing rate. However, the characteristics of firing change as facilitation proceeds: the spike patterns become very irregular with clusters of spikes spaced by long silences; the firing rate is highly variable and clearly oscillates before facilitated bursts. This unstable behaviour dramatically decreases during intense tonic activation which temporarily interrupts bursting, and could therefore be a prerequisite for bursting. In vivo, the effects of AVP depend on the initial firing pattern of AVP neurons: AVP excites weakly active neurons (increasing duration of active periods and decreasing silences), inhibits highly active neurons, and does not affect neurons with intermediate phasic activity. AVP brings the entire population of AVP neurons to discharge with a medium phasic activity characterised by periods of firing and silence lasting 20–40 s, a pattern shown to optimise the release of AVP from the neurohypophysis. Each of the peptides (OT or AVP) induces an increase in intracellular Ca2+ concentration, specifically in the neurons containing either OT or AVP respectively. OT evokes the release of Ca2+ from IP3-sensitive intracellular stores. AVP induces an influx of Ca2+ through voltage-dependent Ca2+ channels of T-, L- and N-types. We postulate that the facilitatory autocontrol of OT and AVP neurons could be mediated by Ca2+ known to play a key role in the control of the patterns of phasic neurons.  相似文献

运用核团灌流液的放免测定和高压液相色谱法以及核团内注射拮抗剂,观察了化学刺激下丘脑视上核(SON)对中缝大核(NRM)灌流液内催产素(OT)、精氨酸加压素(AVP)和5-羟色胺(5-HT)含量的影响以及NRM内注射AVP、5-HT或OT受体拮抗剂对痛阈(PT)的影响。结果表明:SON内注射L-谷氨酸(L-Glu)10μg后动物痛阈明显升高,NRM灌流液中OT和5-HT的含量明显高于对照组水平,AVP的含量仅有一过性增加。NRM内注射oT或5-HT拮抗剂可逆转化学刺激SON引起的镇痛作用;而AVP的V_(1/2)受体拮抗剂也轻度抑制这种镇痛作用,但V_1拮抗剂对此作用无影响。以上结果提示:在刺激SON镇痛中,OT起着重要作用,L-Glu刺激SON的OT细胞释放OT,作用于NRM细胞的OT受体和V_2受体而产生镇痛作用,5-HT在此过程中也发挥重要作用。  相似文献
