The survival rates in 200 patients with terminal renal failure treated by maintenance haemodialysis and renal transplantation were analysed. The overall survival rate (+/- SE) was 81-4 +/- 3% at two years, 68-4 +/- 4% at five years, and 60-8 +/- 4-8% at eight years. Survival was inversely related to age: in those aged 15-34 years overall survival was 89-7 +/- 3-7% at two years, 87-8 +/- 4-1% at five years, and 83-9 +/- 5-5% at eight years. Eighty per cent of the surviving patients were able to work full time. The survival rate in patients returned to dialysis after graft rejection was 87-5% +/- 8-3 at three years.  相似文献

基质金属蛋白酶(MMPs)家族的作用是降解所有细胞外基质,其活性受其特异性组织抑制因子(TIMPs)的抑制。细胞外基质成分的降解与重组在动物生殖生长过程中起重要作用,其变化可以通过MMPs和TIMPs两者表达水平的变化进行监测。大鼠虽然没有月经形成,但是在其子宫内膜也出现类似灵长类的生殖生物学变化。本文从MMPs和TIMPs两者的表达水平,对大鼠子宫内膜的这些变化进行了研究。于大鼠动情周期的不同时期,将其处死、取子宫制备酶粗提液和组织切片,采用酶谱法(zymoyranhn)和原位杂交方法研究动情周期大鼠子宫中MMP-2和-9的活性变化以及MMP-2、-9和TIMP-1、-2、-3mRNA的表达。并通过光密度扫描方法对酶谱结果进行半定量分析。所用杂交探针见Table1。酶谱结果显示:在动情周期大鼠子宫中只检测到67kDa的MMP-2活性,而没有检测到MMP-9的活性(Fig.1)。MMP-2的活性在动情前期最高,动情期和动情后期次之,间情期最低(Fig.2)。原位杂交结果显示:MMP-2、-9、TIMP-1、-2、-3mRNA主要在子宫内膜基底部的基质细胞中表达。MMP-2和-9mRNA在动情前期、动情期和动  相似文献

Haplotype frequencies of the beta-globin gene cluster of Han Chinese at Beijing, Xi'an, and Kunming were estimated, and their mutual genetic relationships were examined and compared to those of Buryats, Khalkhs, Evenkis, Oroqens, Koreans, and Colombian Amerindians. A major 5' subhaplotype (5' to the delta-globin gene), a major 3' subhaplotype (in and 3' to the beta-globin gene), and a major haplotype (combination of 5' and 3' subhaplotypes) are represented as + - - - -, - +, and + - - - - - +, respectively, and found in all three Han Chinese. A rare 5' subhaplotype, - - - - -, which is one of the possible ancestral types, was found only in Han Chinese at Kunming at low frequency (0.013), and a rare 3' subhaplotype, - -, was also observed in all three Han Chinese at low frequencies (0.009-0.014). The present haplotype frequency study suggested that the highest genetic affinity was found between Han Chinese at Beijing and those at Xi'an; the next highest was between Han Chinese at Beijing and Koreans, followed by that between Han Chinese at Beijing and Khalkhs, then that between Han Chinese at Xi'an and those at Kunming or Khalkhs, and finally that between Han Chinese at Beijing and those at Kunming. A genetic boundary between northern and southern Han Chinese was not evident in the present study.  相似文献

Experiments were conducted on laboratory populations of Eriworm, Philosamia ricini Boisduval to study the effect of temperature and humidity on development, growth, survival and oviposition. Experiments were performed at four different temperatures (22 +/- 2 degrees C, 26 +/- 2 degrees C, 28 +/- 2 degrees C and 34 +/- 1 degrees C) each with three different humidities (50%, 70% and 90% relative humidity). Shortest developmental period (42 days) was observed in 28 +/- 2 degrees C and 70% RH. Longest developmental period was (63.6 days) observed in 22 +/- 2 degrees C at 50% RH. Highest larval weight (9.1 g) was found in 28 +/- 2 degrees C at 70% RH. Best pupal weight was observed in 26 +/- 2 degrees C at 70% RH. Weight of silk material was found to be maximum in 26 +/- 2 degrees C at 70% RH. Survival was best in 28 +/- 2 degrees C and 26 +/- 2 degrees C at 70% RH. Oviposition was found to be highest in 28 +/- 2 degrees C at 70% RH.  相似文献

Three different modes of Na+ channel action, the F mode (fast inactivating), the S mode (slowly inactivating), and the P mode (persistent), were studied at different potentials in exceptionally small cell-attached patches containing one and only one channel. Switching between the modes was independent of voltage. In the F mode, the mean open time (tau o) at -30 and -40 mV was 0.14 and 0.16 ms, respectively, which was significantly larger than at -60 and 0 mV, where the values were 0.07 and 0.08 ms, respectively. The time before which half of the first channel openings occurred (t 0.5), decreased from 0.58 ms at -60 mV to 0.14 ms at 0 mV. The fit of steady-state activation with a Boltzmann function yielded a half-maximum value (V 0.5) at -48.1 mV and a slope (k) of 5.6 mV. The mean open time in the S mode increased steadily from 0.12 ms at -80 mV to 1.09 ms at -30 mV, but was not prolonged further at -20 mV (1.07 ms). Concomitantly, t 0.5 decreased from 1.61 ms at -80 mV to 0.22 ms at -20mV. Here the midpoint of steady-state activation was found at -61.2 mV, and the slope was 8.7 mV. The mean open time in the P mode increased from 0.07 ms at -60 mV to 0.45 ms at 0 mV and t 0.5 declined from 2.14 ms at -60 mV to 0.19 ms at +20 mV. Steady-state activation had its midpoint at -14.7 mV, and the slope was 10.9 mV. It is concluded that a single Na+ channel may switch among the F, S, and P mode and that the three modes differ by a characteristic pattern of voltage dependence of tau 0, t 0.5, and steady-state activation.  相似文献

采用露点水势仪对长白山阔叶红松林不同深度(0~10 cm的壤土、20~30 cm的白浆土、50~60 cm的黄土)土壤水分特征曲线进行分析.结果表明:不同土层土壤水分特征曲线整体趋势均表现为快速下降-缓慢下降-基本平稳;用Gardner等提出的幂函数方程可较好地反映该区土壤含水量与土壤水势之间的数量关系,拟合方程的相关系数在0.9239~0.9400;不同土层土壤的持水能力大小依次为黄土>壤土>白浆土,土壤含水量随土壤水势降低而减小的速度依次为壤土>白浆土>黄土.不同土层脱湿过程的土壤水分特征曲线位于吸湿过程之下,说明土壤水分存在滞后现象,且滞后时效依次为壤土>黄土>白浆土.  相似文献

The effects of naloxone and beta-endorphin on LH secretion by pig pituitary cells were studied in primary cultures. On Day 4 of culture, cells (10(5) seeded/well) were challenged with 10(-9), 10(-8) or 10(-7) M gonadotrophin-releasing hormone (GnRH), 10(-10), 10(-9), 10(-8) or 10(-7) M-beta-endorphin or 10(-6) M-naloxone individually or in combinations. Secreted LH was measured at 4 h and 24 h after treatment and cellular content of LH was measured after 24 h. Basal LH secretion (control) was 23.5 +/- 7.6 and 36.9 +/- 10.3 ng/well at 4 h and 24 h, respectively. Relative to control at 4 h, 10(-9), 10(-8) or 10(-7) M-GnRH stimulated (P less than 0.05) LH secretion 140%, 210% and 250%, respectively. At 24 h, LH secretion was increased (P less than 0.05) by GnRH compared to control, but the dose-response to GnRH was absent. Naloxone increased (P less than 0.01) LH secretion 166 +/- 13% at 4 h and 141 +/- 13% (P less than 0.06) at 24 h. Secretion of LH after simultaneous addition of 10(-8) M-GnRH plus naloxone was greater (P less than 0.01) than after GnRH alone at 4 h but not at 24 h. beta-Endorphin at 10(-10), 10(-9), 10(-8) or 10(-7) M failed to alter basal LH secretion at 4 h but decreased secretion at 24 h, while cellular LH content was similar to control at 24 h.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The early, rapid phase of tension recovery (phase 2) after a step change in sarcomere length is thought to reflect the force-generating transition of myosin bound to actin. We have measured the relation between the rate of tension redevelopment during phase 2 (r), estimated from the half-time of tension recovery during phase 2 (r = t0.5(-1)), and steady-state force at varying [Ca2+] in single fibers from rabbit psoas. Sarcomere length was monitored continuously by laser diffraction of fiber segments (length approximately 1.6 mm), and sarcomere homogeneity was maintained using periodic length release/restretch cycles at 13-15 degrees C. At lower [Ca2+] and forces, r was elevated relative to that at pCa 4.0 for both releases and stretches (between +/- 8 nm). For releases of -3.4 +/- 0.7 nm.hs-1 at pCa 6.6 (where force was 10-20% of maximum force at pCa 4.0), r was 3.3 +/- 1.0 ms-1 (mean +/- SD; N = 5), whereas the corresponding value of r at pCa 4.0 was 1.0 +/- 0.2 ms-1 for releases of -3.5 +/- 0.5 nm.hs-1 (mean +/- SD; N = 5). For stretches of 1.9 +/- 0.7 nm.hs-1, r was 1.0 +/- 0.3 ms-1 (mean +/- SD; N = 9) at pCa 6.6, whereas r was 0.4 +/- 0.1 ms-1 at pCa 4.0 for stretches of 1.9 +/- 0.5 (mean +/- SD; N = 14). To test the hypothesis that the hypoxic ventilatory response (HVR) of an individual is a constant unaffected by acclimatization, isocapnic 5-min step HVR, as delta VI/delta SaO2 (l.min-1.%-1, where VI is inspired ventilation and SaO2 is arterial O2 saturation), was tested in six normal males at sea level (SL), after 1-5 days at 3,810-m altitude (AL1-3), and three times over 1 wk after altitude exposure (PAL1-3). Equal medullary central ventilatory drive was sought at both altitudes by testing HVR after greater than 15 min of hyperoxia to eliminate possible ambient hypoxic ventilatory depression (HVD), choosing for isocapnia a P'CO2 (end tidal) elevated sufficiently to drive hyperoxic VI to 140 ml.kg-1.min-1. Mean P'CO2 was 45.4 +/- 1.7 Torr at SL and 33.3 +/- 1.8 Torr on AL3, compared with the respective resting control end-tidal PCO2 of 42.3 +/- 2.0 and 30.8 +/- 2.6 Torr. SL HVR of 0.91 +/- 0.38 was unchanged on AL1 (30 +/- 18 h) at 1.04 +/- 0.37 but rose (P less than 0.05) to 1.27 +/- 0.57 on AL2 (3.2 +/- 0.8 days) and 1.46 +/- 0.59 on AL3 (4.8 +/- 0.4 days) and remained high on PAL1 at 1.44 +/- 0.54 and PAL2 at 1.37 +/- 0.78 but not on PAL3 (days 4-7). HVR was independent of test SaO2 (range 60-90%). Hyperoxic HCVR (CO2 response) was increased on AL3 and PAL1. The costs of walking (Cw) and running (Cr) were measured on 10 runners on a treadmill inclined between -0.45 to +0.45 at different speeds. The minimum Cw was 1.64 +/- 0.50 J. kg(-1). m(-1) at a 1.0 +/- 0.3 m/s speed on the level. It increased on positive slopes, attained 17.33 +/- 1.11 J. kg(-1). m(-1) at +0.45, and was reduced to 0.81 +/- 0.37 J. kg(-1). m(-1) at -0.10. At steeper slopes, it increased to reach 3.46 +/- 0.95 J. kg(-1). m(-1) at -0.45. Cr was 3.40 +/- 0.24 J. kg(-1). m(-1) on the level, independent of speed. It increased on positive slopes, attained 18.93 +/- 1.74 J. kg(-1). m(-1) at +0.45, and was reduced to 1.73 +/- 0.36 J. kg(-1). m(-1) at -0.20. At steeper slopes, it increased to reach 3.92 +/- 0.81 J. kg(-1). m(-1) at -0.45. The mechanical efficiencies of walking and running above +0.15 and below -0.15 attained those of concentric and eccentric muscular contraction, respectively. The optimum gradients for mountain paths approximated 0.20-0.30 for both gaits. Downhill, Cr was some 40% lower than reported in the literature for sedentary subjects. The estimated maximum running speeds on positive gradients corresponded to those adopted in uphill races; on negative gradients they were well above those attained in downhill competitions.  相似文献

在温度(150.5)C,盐度15.50.5的条件下,研究了Cu2+和Zn2+对西藏拟溞(DaphniopsistibetanaSars)存活、生长和生殖的影响。结果表明,西藏拟溞在各Cu2+活度组中的存活率差异不显著,而在10-8.60mol/L组中的体长增长率显著高于其他各组。当水环境中Cu2+活度为10-8.30和10-8.13mol/L时,西藏拟溞的内禀增长率(rm)为0.2211和0.2171/d,显著高于对照组,而西藏拟溞在不同Cu2+活度组中的产卵率均高于对照组,为0.9705-1.1742。西藏拟溞在各Cu2+活度组中的存活率和生长率差异均不显著。当Zn2+活度为10-7.04-10-6.82mol/L时,西藏拟溞的rm为0.2249-0.2296/d,显著高于对照组。西藏拟溞在Zn2+活度为10-7.04mol/L组中的产卵率最大,为1.0178,10-6.82mol/L组次之,为0.867。当Zn2+活度为10-7.04-10-6.82mol/L时,西藏拟溞一生生殖次数显著高于对照组(1.58),为1.92-2.17次。因此,综合来看,当水环境中的Cu2+活度为10-8.60-10-8.13mol/L、Zn2+活度为10-7.04-10-6.82mol/L时能明显的促进西藏拟溞的种群增长、发育和生殖。论文讨论了Cu2+和Zn2+对西藏拟溞的促长机理。    相似文献

Three experiments were designed to analyze the effects of cooling rate on survival of stallion spermatozoa in a milk-based extender, at 0 to 96 hours after reaching the desired temperature. The samples were warmed to 37 degrees C and were evaluated by computer-assisted analysis of sperm motility. In Experiment 1, rate of cooling between 37 and 20 degrees C was evaluated. Sperm motion was not affected by cooling at plunge, -0.42 or -0.28 degrees C/minute. However, storage of spermatozoa at 5 degrees C after slow cooling below 20 degrees C was superior to storage at 20 degrees C. In Experiment 2, 3 cooling rates from 37 degrees to 5 degrees C were evaluated. Cooling at either -0.05 or -0.7 degrees C/minute was superior (P<0.05) to plunging spermatozoa to 5 degrees C. Cooling at -0.05 degrees C/minute rather than -0.7 degrees C/minute maximized the percentage of motile spermatozoa and their curvilinear velocity. In Experiment 3, cooling rates from 20 to 5 degrees C were evaluated, with all samples cooled at -0.7 degrees C/minute from 37 to 20 degrees C. Sperm motion was similar (P>0.05) after cooling below 20 degrees C at -0.012, -0.05 or -0.10 degrees C/minute, and the 2 slower rates were superior (P<0.05) to cooling at -0.3 degrees C/minute. The radioresistance of coxsackievirus B-2 was studied when the virus was suspended in Eagle minimal essential medium, distilled water, cooked ground beef, and raw ground beef and irradiated at various temperatures in a cobalt-60 gamma radiation source. The number of surviving viruses at given doses of radiation was determined by a plaque assay system. All destruction curves indicated a first-order reaction. When the virus was irradiated in minimal essential medium at temperatures of -30, -60, and -90 C, D values (in Mrad) were 0.69, 0.59, and 0.64, respectively. When the virus was suspended in water and irradiated at -90 C, the D value was 0.53. Cooked ground beef containing the virus was irradiated at temperatures ranging from 16 to -90 C. The D values were 0.70 (16 C), 0.76 (0.5 C), 0.68 (-30 C), 0.78 (-60 C), and 0.81 (-90 C). Raw ground beef containing the virus was irradiated at -30, -60, and -90 C, and the D values were respectively 0.75, 0.71, and 0.68. The D values indicate that the rate of viral inactivation was dependent on the suspending menstrum.  相似文献

Development times and survivorship of immature shore flies and longevity and reproduction of adult shore flies, Scatella tenuicosta Collin, reared on algae-infested filter paper, were studied at three temperatures (constant 20, 26, and 28.5 degrees C) through life table analysis. The development time for each individual life stage and the total time from egg to adult decreased with increasing temperature. Duration of the third (ultimate) larval instar ranged from 3.3 +/- 0.09 d at 20 degrees C to 1.4 +/- 0.04 d at 28.5 degrees C and was 1.7-1.9 times longer than the approximately equal first and second instars. Development of male and female shore flies from egg to adult needed an average of 14.5 +/- 0.13, 8.2 +/- 0.05, and 7.0 +/- 0.04 d at 20, 26, and 28.5 degrees C, respectively, and needed an estimated 154.4 +/- 1.2 thermal units (degree days). At these respective temperatures, adult females lived 21.8 +/- 2.2, 19.9 +/- 2.4, and 15.0 +/- 1.4 d and produced 379 +/- 62, 710 +/- 119, and 477 +/- 83 eggs during oviposition periods of 14.3 +/- 2.1, 15.0 +/- 2.2, and 10.8 +/- 1.4 d; daily lifetime egg production averaged 16.3 +/- 2.3, 33.5 +/- 3.8, and 29.7 +/- 3.5. Developmental stage-specific mortality was relatively low for all life stages at all temperatures, with maximum percent mortalities of 5.7% occurring in both the egg stage and in the third instar. The highest net reproductive rate (R(o)) was obtained for insects reared at 26 degrees C and was 329.6. The intrinsic rate of natural increase (r(m)) was highest at 28.5 degrees C and was 0.430. Generation time and doubling time of the population were shortest at 28.5 degrees C and were 12.4 and 1.6 d, respectively. Systolic blood pressure in the arm was measured in infants at the ages of 4 to 6 days and 5 to 7 weeks by the Doppler ultrasound technique. At the age of 4 to 6 days the mean blood pressure (+/- SE of mean) in 469 sleeping infants was 70-7 +/- 0-3 mm Hg, rising at 5 to 7 weeks to 89-7 +/- 0-9 mm Hg (in 144 infants). In 252 infants awake at 5 to 7 weeks blood pressure was 96-8 +/- 0-6 mm Hg. In 391 infants in whom measurements were made on both occasions blood pressure at 4 to 6 days was significantly related to blood pressure at 5 to 7 weeks. Thus those infants with relatively high blood pressures at 4 to 6 days showed a weak tendency to have relatively high blood pressures at 5 to 7 weeks. In this trend continues with age it would suggest that the tendency to develop hypertension may already be demonstrable at the age of 4 to 6 days.  相似文献

Sulfate transport was examined using membrane vesicles (pH 7.0 inside) prepared from rat liver lysosomes. Sulfate uptake was dependent upon external pH with increased uptake at lower buffer pH. The Km for uptake was 160 microM at pH 5.0 while at pH 7.0, a lower affinity system with a Km of 1.4 mM was present. The protonophore carbonyl cyanide m-chlorophenylhydrazone increased uptake at pH 5.0 while valinomycin/KCl had no effect. In contrast, at pH 7.0, valinomycin-induced changes in membrane potential stimulated uptake. Countertransport of sulfate at pH 7.0 was inhibited by 4,4'-diisothiocyano-2,2'-disulfonic acid stilbene, N-(4-azido-2-nitrophenyl)-2-aminoethanesulfonic acid, and a variety of anions: SO4(2-) greater than MoO4(2-) greater than Cl- greater than HPO4- greater than HCO3-. Trans-stimulation of sulfate uptake at pH 7.0 was observed with MoO4(2-) and, to a lesser extent, with S2O3(2-) while Cl-, HPO4-, and HCO3- had little effect. However, chloride loading of vesicles resulted in marked stimulation of sulfate uptake at pH 5.0. It appears that sulfate and protons exit lysosomes in exchange for chloride by a specific, pH-regulated anion transport system.  相似文献

The Wa strain of human rotavirus, grown in MA-104 cells, was suspended either in tryptose phosphate broth or feces from a case of rotaviral diarrhea. It was then aerosolized into a rotating drum using a Collison nebulizer. The drum air was sampled using an all-glass impinger containing tryptose phosphate broth as collecting fluid. At 20 +/- 1 degree C, the virus aerosolized from tryptose phosphate broth was found to survive best at 50 +/- 5% relative humidity, where its half-life was 44.2 +/- 6.3 h. At 30 +/- 5% and 80 +/- 5% relative humidity, the half-life of the virus was 24.5 +/- 3.5 and 3.8 +/- 1.0 h, respectively. At 6 +/- 1 degree C, the airborne survival of the virus at the mid and low relative humidity levels was further enhanced, but at the high relative humidity it remained very similar to that seen at 20 +/- 1 degree C. When aerosols of fecally suspended human rotavirus were held at 20 +/- 1 degree C with 50 +/- 5% relative humidity, nearly 80% of the airborne virus particles remained infectious even at the aerosol age of 24 h. These findings may help in our understanding of the epidemiology of rotaviral infections.  相似文献

Antioxidant activity of d-α-, dl-β-, d-γ- and d-δ-tocopherol was investigated with fatty acid methylester of milk fat from which unsaponifiable matter had been removed. Autoxidation was carried out at 50°C and its degree was indicated by peroxide value, α- or β-Tocopherol was more effective at lower concentrations (0.003 and 0.01%) than at higher concentrations (0.05, 0.1 and 0.5%). The antioxidant activity of γ- and δ-tocopherol was increased with the increase of tocopherol concentration within the range of 0.001 to 0.5%. The order of antioxidant activity of these tocopherols, which was compared in terms of the time to reach 30 meq of peroxide value, varied with the concentration; γ > β > δ > α at 0.001%, α > γ > β > δ at 0.003%, γ > δ > β > α at 0.01%, and δ > γ > β > α at the concentrations more than 0.05%. α-Tocopherol at the concentration of 0.003%, which corresponded to the concentration in original milk fat, was more effective than other tocopherols at the same concentration and α-tocopherol at other concentrations. Synergism due to the combination of β-, γ-, or δ-tocopherol with 0.003% of α-tocopherol was not observed.  相似文献

Lipolytic activity measured at pH 8.6 in bovine corpora lutea exhibited classical properties of lipoprotein lipase (LPL) in terms of serum and heparin stimulation and NaCl inhibition. LPL activity was measured in 23 corpora lutea collected at different stages of the estrous cycle and early pregnancy. The LPL activity in cyclic corpora lutea (mumole FA released/hr/100 mg acetone powder) was low at Days 4-8 of the estrous cycle (3.1 +/- 1.5: mean +/- SE) and at Days 19-20 (1.6 +/- 0.6). However, high activity of the enzyme was found at Days 12-15 of the cycle (11.8 +/- 1.8); these concentrations were significantly (P less than 0.01) elevated over those found at Days 4-8 and 19-20. The enzyme activity began to decline at Days 16-18 of the estrous cycle (5.1 +/- 1.7). Low enzyme activity was found in the corpora lutea removed from two cows at Day 22 of pregnancy. Progesterone concentrations were measured in 16 of the 23 corpora lutea and a good correlation (r = 0.75, P less than 0.01) was found between lipoprotein lipase and progesterone concentrations of the tissue. While the stimulatory effect of vanadate, an anion of pentavalent vanadium, on adenylate cyclase (AC) has been repeatedly demonstrated in various tissues only a few studies have been hitherto devoted to the effect of vanadyl, a cation of tetravalent vanadium, but these have provided contradictory results. In the present experiments synaptic plasma membranes from normal rat cerebral cortex were used for estimation of the vanadyl effect (in the concentration range from 10(-5) mol.1(-1) to 10(-3) mol.1(-1) on the basal adenylate cyclase activity. Four types of incubation media were used. In the presence of Tris-maleate and creatine phosphate + creatine phosphokinase (CP + CK) maximal stimulation (33%) was reached at 10(-4) mol.1(-1). In the same buffer but in absence or (CP + CK) maximum was already obtained at 10(-5) mol.1(-1) (49%); at 10(-3) mol.1(-1) no effect was observed. In Tric.HCl buffer with (CP + CK) maximal stimulation appeared at 10(-5) mol.1(-1), whereas at 10(-3) mol.1(-1) inhibition (-25%) was observed. In a medium containing Tris.HCl without (CP + CK) the biphasic nature of vanadyl effect was less markedly expressed: maximal stimulation (+55%) occurred at 10(-4) mol.1(-1). Thus vanadyl stimulates AC, but at relatively low concentrations (10(-5)-10(-4); at higher concentration it tends to exert an inhibitory action. Vanadate had a qualitatively similar effect, but the stimulation was more pronounced and the tendency to inhibition was shifted to higher concentrations.  相似文献
