马鞍菌属新种和新记录(一)

有关马鞍菌属 Helvella 的历史,属种概念以及中国已知种的检索表已作过报道(刘波等,1985)。本文描述了近年来采到的两个新种:粘马鞍菌 Helvella glutinosa sp.nov.和伞形马鞍菌 H.galeriformis sp.nov.;四个新记录:白柄马鞍菌 H.albellu Quél.,小马鞍菌H.helvellula(Dur.et Mont.)Dissing,乳白马鞍菌 H.lactea Boud.和脉马鞍菌 H.phlebo-phora Pat.et Doass.。此外,斑点马鞍菌 H.maculata Weber 虽已作过报道和描述(李静丽等,1986),然而由于它是一个非常具特征性的种,而且有一限制的地理分布,因此本文也对它进行了更充分的描述和讨论。     

云南剌皮属的研究

本文报道云南剌皮属Heterochaete Pat.10种,其中1个新种,3个国内新纪录种。新种是思茅剌皮H.simaoensis Hu et J.Z.Lisp.nov。本文所研究的标本藏于湖南师范大学真菌标本室(MHHNU)和中国科学院微生物研究所真菌标本室(HMAS)。     

关于 Archineottia与Holopogon 两属合并的问题

本文根据蕊柱结构将Archineottia 归并于Holopogon,并发表了5个新组合：无喙兰Holopogon gaudissartii (Hand.-Mzt.) S. C. Chen、 印度无喙兰H. microglottis (Duthie) S. C. Chen、锡金无喙兰H. pantlingii (W. W. Smith) S. C. Chen、叉唇无喙兰 H. smithianus (Schltr.) S. C. Chen、 日本无喙兰H. japonicus (M. Furuse) S. C. Chen。     

New Taxa of Heracleum L. (Umbelliferae) from China

Three new species of the genus Heracleum L. (Umbelliferae) are described from Sichuan Province, China. They are H. wenchuanense; H.wolongense and H. xiaojinense.     

啮齿动物螺杆菌多重PCR检测方法的建立

目的建立一种可同时检测肝、胆汁、啮齿类三种螺杆菌的多重PCR方法。方法根据已公布的肝、胆汁、啮齿类三种螺杆菌16SrRNA基因序列设计三对特异性引物进行多重PCR并对反应条件进行优化。结果三对引物能分别扩增出特异性的417 bp、364 bp、324 bp目的条带。最佳退火温度为52℃,镁离子浓度为2.0mmol/L,dNTP浓度为200μmol/L,引物浓度为0.625μmol/L。在此条件下多重PCR同时检测的肝、胆汁、啮齿类三种螺杆菌敏感度均为10 fg。结论本实验建立的多重PCR是一种敏感、特异、高效的方法,为同时检测啮齿动物中肝、胆汁、啮齿类三种螺杆菌奠定了良好的基础。     

滇藏蝠蛾属三新种记述（鳞翅目：蝙蝠蛾科）

本文记述了分布于云南、西藏高寒草甸区的蝠蛾属3新种:云南蝠蛾Hepialus yunnanensis sp.nov.、芒康蝠蛾Hepialus markamensis sp.nov.和草地蝠蛾Hepialus pratensis sp.nov.     

植物细胞中蓝光诱导ROS生成的识别和亚细胞定位检测

以2’,7’-二氯二氢荧光素二乙酯(dichlorofluorescein diacetate,H2DCF-DA)为荧光探针孵育拟南芥叶表皮条,利用荧光光谱和激光共聚焦扫描显微技术,对高辐照蓝光诱导下叶肉细胞活性氧(reactive oxygen spe-cies,ROS)的生成,进行了分子识别和亚细胞定位检测。结果表明:植物细胞在蓝光诱导下,可以产生大量的ROS。过氧化氢酶清除实验表明:高辐照蓝光诱导产生的ROS,主要成分是H2O2,并且主要定位在叶绿体和细胞膜上。     

2004年下半年广东野生鸟类禽流感研究初报

2004年下半年我们对广东省的野生鸟类进行禽流感取样调查,共取得血样263份、拭子样337份,血清检测采用琼脂扩散（AGP）和血凝抑制试验（HI）,病毒检测用鸡胚法鉴定。结果：1）病毒检测中分别在放生的白眉鸭（Arias querquedula）和石鸡（Alectoris chukar）体内检测到H5和H9病毒各1例;2）血清检测中H5的总阳性率为97.7％,而H9达98．5％,其中,不论是候鸟与留鸟间,还是放生个体、市场个体及野外个体间在H5差异不大且都高于94％,H9也是如此。由此,我们认为：1）鸟类放生存在传播禽流感的可能性;2）该时期野生鸟类中普遍出现H5和H9的高阳性率现象,可能存在H5和H9在野外的大范围传播情况。     

变性高效液相色谱检测沙门氏菌、空肠弯曲菌和肠出血性大肠杆菌

应用多重PCR 反应(multiplex PCR,mPCR)结合变性高效液相色谱(denaturing high-performance liquid chromatography,DHPLC)技术建立食品中沙门氏菌、空肠弯曲菌和肠出血性大肠杆菌O157:H7的快速检测方法.以编码沙门氏菌的fimY基因、编码空肠弯曲菌的gyrA基因和编码肠出血性大肠杆菌O157:H7的rfbE基因为靶基因,选择3对引物,建立并优化了快速鉴别沙门氏菌、空肠弯曲菌和肠出血性大肠杆菌O157:H7的多重PCR体系,扩增产物分别为284、159和499 bp,并验证了该多重PCR具有特异性.沙门氏菌、空肠弯曲菌和肠出血性大肠杆菌O157:H7标准菌株稀释成不同梯度,做灵敏度检测.试验结果表明该方法有很好的特异性,且灵敏度高,检测限可达到:沙门氏菌1.5 CFU/ml、空肠弯曲菌15 CFU/ml、肠出血性大肠杆菌O157:H7 15 CFU/ml.在随机采集的226份冷冻鸡肉类样品中,检出了7份样品为沙门氏菌阳性、10份为空肠弯曲菌阳性、1份为肠出血性大肠杆菌O157:H7阳性.研究建立的多重PCR-DHPLC方法可特异、灵敏地实现对沙门氏菌、空肠弯曲菌和肠出血性大肠杆菌O157:H7的快速检测.     

磷饥饿下番茄幼苗根系液泡膜H+-ATPase活性的适应性变化

以‘世纪星’番茄为材料。研究了磷饥饿下番茄幼苗的生长状况及其根部液泡膜H^＋-ATPase活性的适应性变化。结果表明：磷饥饿下番茄幼苗的平均高度均低于对照苗,而主根长度均显著长于对照。磷饥饿提高了番茄幼苗根部液泡膜H^＋-ATPase的水解活性,随着磷胁迫时间的延长,该酶的活性逐渐增大,在磷饥饿7d时达到最大,后又略有降低;而对照番茄幼苗根部该酶的活性变化很小。动力学分析表明：磷饥饿使番茄幼苗根部液泡膜H^＋-ATPase的Km值明显降低,但对该酶的Kmax影响不大。这说明磷饥饿提高了该酶对其底物的亲和力。此外,磷饥饿并不改变液泡膜H^＋-ATPase酶的最适pH值（仍为7．5）。     

保加利亚乳杆菌H+-ATPase缺陷型菌株的筛选

【目的】从传统乳制品中筛选具有新霉素抗性的H+-ATPase缺陷的德氏乳杆菌保加利亚亚种自发突变株,为最终开发弱后酸化的酸奶发酵剂奠定基础。【方法】利用API 50 CH细菌鉴定系统和16s rRNA基因序列分析对菌株进行鉴定。新霉素作为筛选压力,筛选具有新霉素抗性自发突变菌株,比较亲本和突变菌株的H+-ATPase活力及其代谢情况。【结果】从内蒙古地区的传统发酵酸奶中分离鉴定出一株德氏乳杆菌保加利亚亚种（Lactobacillus delbrueckii subsp. bulgaricus）,并命名为KLDS 1.9201。以此为出发菌株,筛选出两株H+-ATPase缺陷的自发突变株,分别命名为KLDS 1.9201-1、KLDS 1.9201-4,它们的H+-ATPase活力分别比亲本KLDS 1.9201降低了46%和60%。在MRS培养基中生长24 h后,KLDS 1.9201、KLDS 1.9201-1和KLDS 1.9201-4对初始葡萄糖的代谢率分别为65%、41%和31%,终产物中乳酸的浓度分别为26g/L、18g/L和15g/L,突变菌株的生物量均低于亲本。【结论】H+-ATPase活力降低的德氏乳杆菌保加利亚亚种的自发突变株具有较低的生长速率和弱产酸能力,它们可被用于制作弱后酸化的酸奶发酵剂。     

NaCl胁迫对盐芥质膜和液泡膜ATPase活性的影响

以盐生植物盐芥和中生植物拟南芥幼苗为材料,研究了盐胁迫对它们叶片和根质膜、液泡膜H+-ATPase、Ca2+-ATPases和K+-ATPase活性以及H+-ATPase、Na+/H+ 逆向转运蛋白表达的影响.结果显示:在NaCl胁迫下,盐芥叶片和根质膜的H+-ATPase活性分别比对照显著升高41%～212%和35%～53%,液泡膜的H+-ATPase分别显著升高281%～373%和4%～38%,而拟南芥却比相应对照都显著降低;相同盐浓度胁迫下,盐芥叶片的H+-ATPase活性比根部高4～8倍,盐芥根也远高于拟南芥.在NaCl胁迫下,盐芥叶片和根的液泡膜H+-ATPase蛋白质β亚基含量变化与其酶活性变化趋势一致,质膜Na+/H+ 逆向转运蛋白的表达量与Na+含量变化趋势一致.盐胁迫下盐芥根中Ca2+-ATPases和K+-ATPase活性的增加与根中Ca2+和K+含量呈显著正相关.研究发现,在盐胁迫条件下,盐芥能有效增强H+-ATPase蛋白和Na+/H+逆向转运蛋白表达,显著提高其根系与叶片质膜和液泡膜的H+-ATPase、Ca2+-ATPase和K+-ATPase活性,维持细胞质中较高的Ca2+和K+水平,从而缓解盐胁迫的伤害,增强耐盐性.     

中国山龙眼属植物二新种

山龙眼属植物在我国南部的常绿林中为较常见的树种。作者在编写《ＦｌｏｒａｏｆＣｈｉｎａ》的山龙眼科植物时,发现华南地区的山龙眼属植物中有２个新种;它们是生长于广东阳春县西南部的阳春山龙眼和生长于广西十万大山的东兴山龙眼;现予报道．

Soils contain two different activities for oxidation of hydrogen

Abstract Hydrogen oxidation rates were measured in a neutral compost soil and an acidic sandy loam at H₂ mixing ratios of 0.01 to 5000 ppmv. The kinetics were biphasic showing two different K _m values for H₂, one at about 10–40 nM dissolved H₂, the other at about 1.2–1.4 μM H₂. The low- K _m activity was less sensitive to chloroform fumigation than the high- K _m activity. If sterile soil was amended with Paracoccus denitrificans or a H₂-oxidizing strain isolated from compost soil, it exhibited only a high- K _m (0.7–0.9 μM) activity. It also failed to utilize H₂ mixing ratios below a threshold of 1.6–3.0 ppmv H₂ (160–300 mPa). A similar result was obtained when fresh soil samples were suspended in water, and H₂ oxidation was determined from the decrease of dissolved H₂. However, H₂ was again utilized to mixing ratios lower than 0.05 ppmv, if the supernatant of the soil suspension or the settled soil particles were dried onto sterile soil or purified quarz sand. Obviously, soils contain two different activities for oxidation of H₂: (1) a high- K _m, high-threshold activity which apparently is due to aerobic H₂-oxidizing bacteria, and (2) a low- K _m, low-threshold activity whose origin is unknown but presumably is due to soil enzymes.     

Emerging influenza virus: A global threat

Since 1918, influenza virus has been one of the major causes of morbidity and mortality, especially among young children. Though the commonly circulating strain of the virus is not virulent enough to cause mortality, the ability of the virus genome to mutate at a very high rate may lead to the emergence of a highly virulent strain that may become the cause of the next pandemic. Apart from the influenza virus strain circulating in humans (H1N1 and H3N2), the avian influenza H5N1 H7 and H9 virus strains have also been reported to have caused human infections, H5N1 H7 and H9 have shown their ability to cross the species barrier from birds to humans and further replicate in humans. This review addresses the biological and epidemiological aspects of influenza virus and efforts to have a control on the virus globally.     

马鞍菌属新种和新组合

本文报道了产于我国的马鞍菌属Helvella的三个新种及一新组合,它们是亚梭孢马鞍菌H.subfusispora Liu et Cao,太原马鞍菌H·taiyuanensis Liu,Du et Cao中国马鞍菌H.sinensis Liu et Cao以及紫棱柄马鞍菌H.purpurea(Zang)Liu et Cao。并附有我国马鞍菌种的检索表。
