When an electric field is applied along the fiber axis, the intensities of all observable optical diffraction lines of skeletal muscle fibers increase. This electro-optical effect was extensively studied and it was confirmed that the effect is due to the interaction between electric dipole moments of thin filaments and the applied field. From the present study on the intensity modulation due to applied field in sinusoidal and square forms, we confirmed that (1) the thin filament is a semiflexible rod, (2) the second order mode of the bending motion of thin filaments contributes to the electro-optical effect of muscle fibers at higher frequencies of a sinusodidal field or shorter durations of a square field, (3) the induced moment has no appreciable effect, and (4) the estimated value of the flexural rigidity of thin filaments strongly depends on the concentrations of free calcium ions in the myofibrillar space.  相似文献

The effect of MgPPi on the rigor force of glycerinated fibres in the range of ionic strength 75-250 mM at two temperatures 18 degrees and 5 degrees C was studied. At 18 degrees C the maximum of this effect was above the range of average ionic strength. At 5 degrees C the greatest effect of MgPPi was observed at low ionic strength.  相似文献

The occurrence and distribution of an array of neuropeptides and dopamine-beta-hydroxylase in the circumvallate papillae of monkey, pig, cow, ferret, cat, rat and mouse was studied by immunocytochemistry. The animals were chosen to represent species with different diets. Substance P/neurokinin A- and calcitonin gene-related peptide-containing fibers were numerous in the circumvallate papillae of all animals examined, with the highest frequency in monkey, pig, cow, rat and mouse; in ferret and cat moderate numbers were detected. Vasoactive intestinal peptide/peptide histidine isoleucine amide-containing fibers were numerous in the circumvallate papillae of pig, while they were moderate in number in monkey, ferret and mouse. Neuropeptide Y-containing fibers were few to moderate in number in the circumvallate papillae of all species. Galanin-containing fibers were numerous in the pig circumvallate papillae, while only a few fibers could be detected in monkey, cow, cat, rat and mouse. Somatostatin-containing fibers were seen only in the cat circumvallate papillae, gastrin-releasing peptide-containing fibers in the cow and cat, cholecystokinin/gastrin-containing fibers in the pig and cow. Dopamine-beta-hydroxylase-containing fibers were detected in all animals studied. They were few to moderate in number in the circumvallate papillae. There was no obvious link between the peptidergic innervation pattern and the food habits.  相似文献

Silks are protein fibers with remarkable mechanical properties. The discovery of the structural features that govern these properties is a challenge for biochemistry and structural biology. This review summarizes the results of the biochemistry of silk proteins as well as the knowledge of the molecular biology of the respective genes. The in-depth oxypropylation of different types of cellulose fibers, namely Avicel, Rayon, Kraft, and Filter Paper, was investigated. New biphasic mono-component materials were obtained, which could be hot-pressed to form films of cellulose fibers dispersed into a thermoplastic matrix. The success of this chemical modification was assessed by FTIR spectroscopy, X-ray diffraction, scanning electron microscopy, differential scanning calorimetry, thermogravimetric analysis and contact angle measurements. The optimization of this process led to the establishment of the optimal molar ratio between the cellulose OH groups and propylene oxide, which varied as a function of the specific morphology of the fibers.  相似文献

Electron micrographs of deoxyhemoglobin S fiber cross sections provide an end-on view of the fiber whose appearance is sensitive to small changes in orientation. We have developed a procedure to exploit this sensitivity in order to determine the hand of these particles. In a sickle hemoglobin fiber the hemoglobin molecules form long pitch helical strands which twist about the particle axis with a pitch of about 3000 A. Tilting a 400-A-thick cross section by a few degrees aligns one of the long pitch helices so that it is nearly parallel to the direction of view. When a strand of hemoglobin molecules in a fiber is aligned in this manner it appears as a strongly contrasted bright spot. It is this spot, rather than the fiber axis, which appears to be the apparent center of rotation of the cross section. The direction of the displacement of the spot from the particle axis depends upon the particle hand and tilt direction. We have used this property to determine that sickle hemoglobin fibers are right-handed particles. This method may be applicable to other particles with long pitch helices as well.  相似文献

The transient kinetics of ATP hydrolysis in chemically skinned psoas muscle fibers of the rabbit have been measured. Muscles fibers in the rigor state (absence of nucleotide) were relaxed rapidly by the photochemical release of [2-3H]ATP from caged-ATP (P3-1-(2-nitro)phenylethyl[2-3H]adenosine 5'-triphosphate) in the absence of calcium ions. Rapid freezing of the fiber to stop hydrolysis, followed by analysis of the tritiated nucleotide content allowed the course of the hydrolysis to be determined. The timecourse of ATP hydrolysis was biphasic, with an initial rapid phase occurring at a rate of approximately 60 s-1 at 12 degrees C for fibers exposed to greater than 0.7 mM ATP. The amplitude of the rapid phase was as previously reported (Ferenczi, M. A., E. Homsher, and D. R. Trentham, 1984, J. Physiol. (Lond.)., 352:575-599).  相似文献

Using a novel purification procedure, the protein composition of the tail fibers of bacteriophage T4 has been determined. Fibers contain four proteins whose molecular weights, as estimated by sodium dodecyl sulfate/acrylamide gel electrophoresis, are 150,000, 125,000, 40,000 and 24,000. The two largest proteins have been previously identified as the products of genes 34 (P34) and 37 (P37), respectively (King and Laemmli, 1971; Ward and Dickson, 1971). The two smaller proteins have now been identified as the products of genes 35 (P35) and 36 (P36), respectively. The products of the two other known phage genes required for fiber assembly, 38 and 57, have been identified as non-structural phage proteins with molecular weights of 26,000 and 10,000, respectively.  相似文献

Regional differences in histochemical properties along the length of rat intrafusal muscle fibers were examined. Outside the muscle spindle capsule the nuclear bagfibers lose their hitherto characteristic ATPase activity and stain in a manner similar to that of extrafusal type I muscle fibers, perhaps in relationship to spindle skeletofusimotor innervation.  相似文献

Dermal elastic fibers are believed to have a primary role in providing elastic stretch and recoil to the skin. Here we compare the structural arrangement of dermal elastic fibers of chick skin and different animal species. Most elastic fibers in chick skin are derived from cells that line the feather follicle and/or smooth muscle that connects the pterial and apterial muscle bundles to feather follicles. Elastic fibers in the dermis of animals with single, primary hair follicles are derived from cells lining the hair follicle or from the ends of the pili muscle, which anchors the muscle to the matrix or to the hair follicle. Each follicle is interconnected with elastic fibers. Follicles of animals with primary and secondary (wool) hair follicles are also interconnected by elastic fibers, yet only the elastic fibers derived from the primary follicle are connected to each primary follicle. Only the primary hair follicles are connected to the pili muscle. Human skin, but not the skin of other primates, is significantly different from other animals with respect to elastic fiber organization and probably cell of origin. The data suggest that the primary role for elastic fibers in animals, with the possible exception of humans, is movement and/or placement of feathers or hair.  相似文献

Scar tissue, obtained from humans, was stained for elastin fibers by a new staining method. Elastin fibers were noted at sites where they must have been formed de novo. The morphology and the distribution of elastin in various types of scars are described. Practically no elastin was found in keloids.  相似文献

Water in barnacle muscle has been studied using NMR techniques. Fresh fibers are compared with membrane-damaged fibers treated with solutes that greatly alter fixed charge and total water content. Both water (97%) and solute (3%) protons are visible in continuous wave spectra of oriented fresh fibers. No local field inhomogeneities were detected, nor are cell solutes significantly bound. In pulse experiments, all cell water is visible and exhibits a single exponential decay. In fresh fibers, T2 approximately or equal to 40 ms; faster decaying signals are assigned to immobile and mobile protons on macromolecules. T1 and T1p are frequency dependent. Using equations derived for a two-compartment model with fast exchange, we calculate the following: tau b, the correlation time for anisotropic rotational motion of bound water; Sb, its order parameter; tau ex, the correlation time for exchange between bound and free fractions; f, the fraction of water bound; and Hr, the grams of water bound per gram of macromolecule. Whereas f varies inversely with total water content, the other parameters are virtually constant, with values: tau b approximately or equal to 1.3 X 10(-8) S; tau ex approximately or equal to 8 X 10(-6) s; Sb approximately or equal to 0.06; and Hr approximately or equal to 0.1g H2O/g macromolecule. Thus, the NMR relaxation detectable properties of water bound to macromolecules are unaffected by solutes that greatly alter the macromolecular surface charge.  相似文献

Catalase has been localized immunocytochemically with anti-bovine catalase in long thin filament structures in aerobic type I fibers in the skeletal muscles of normal and genetically dystrophic hamsters. The filaments range in length from 1 to 60 micron, are orientated regularly along the long axis of the fibers, and also seem to surround and project from muscle nuclei. The enzyme thus appears to be more prominent in the sarcoplasmic reticulum than in peroxisomes, and in this situation is suitably placed for destroying toxic hydrogen peroxide which may be continously generated in aerobic fibers.  相似文献

The supramolocular structure of hemoglobin S has been studied by electron microscopy and computer-based image reconstruction. Negatively stained fibers prepared by the lysis of sickled cells or the stirring of hemoglobin S hemolysates have been observed to be almost exclusively of the 20-nm diameter form. These fibers have a periodic variation in diameter between the extremes of 18 nm and 23 nm. Computed Fourier transforms of the fibers show a, highly complex pattern of reciprocal space maxima, with 30 maxima on 20 layer-lines clearly resolved. The Bessel orders of the maxima were assigned with the aid of a newly developed technique, a combined real-space Fourier-space reconstruction method (REFORM). This method utilizes the filtered image produced by the inverse Fourier transform of the low-resolution maxima to calculate in real space the crosssection of a helical fiber. The REFORM analysis indicated that the fibers have an elliptical cross-section and are composed of 14 hexagonally packed filaments with 10 outer filaments surrounding four inner filaments. On the basis of this cross-section, the Bessel orders of all the maxima were assigned, permitting the calculation of three-dimensional reconstructions by Fourier Bessel synthesis. From these reconstructions details of the location of hemoglobin S molecules of each filament were obtained. Hemoglobin S molecules are staggered in adjacent filaments to produce a closely packed helical structure. Reconstructions of fibers at various stages of disassembly revealed a stable intermediate containing 10 filaments which could be characterized in terms of the loss of two pairs of specific outer filaments. A partially disassembled fiber with only six filaments at positions corresponding to three inner and three outer filaments of the parent structure was also identified. The six-filament structure appears to be produced from the 10-filament structure by the loss of two specific pairs of filaments. Thus pairs of filaments are evidently significant structural units in the stabilization of the complete fibers and the orientation of the molecules in these pairs may be related to the filament pairs known to occur in crystals of hemoglobin S.  相似文献
