This laboratory study examined the effects of the specific serotonin reuptake inhibitor fluoxetine on growth following molting and on a range of behaviors in the crayfish Orconectes rusticus. For growth experiments, male Form I and Form II crayfish were weighed and measured and placed individually in water containing 0–500 μg/L of fluoxetine. They were held in fluoxetine or control water until they molted and were reweighed two weeks post-molt. In behavior experiments, juvenile and adult animals were held individually in 0, 2, 200, or 500 μg/L of fluoxetine for 10 days and tested in an open field arena to assess locomotion, thigmotaxis, sheltering, and habituation to a novel environment. Under our laboratory conditions, crayfish exposed to fluoxetine at 500 μg/L showed significantly enhanced growth: post-molt Form I animals had greater body weight and post-molt Form II animals had greater carapace length, relative to controls. In open field tests, juvenile crayfish exposed to 2 and 500 μg/L fluoxetine displayed significantly reduced locomotion compared to controls. The results indicate that crayfish growth and locomotion can be manipulated by short-term exposure to ambient fluoxetine, suggesting that this means of exposure may offer a useful and noninvasive way to examine drug effects in freely moving animals. However, effects were only observed at concentrations well above fluoxetine levels currently reported in the environment. This suggests that O. rusticus may be relatively resistant to this form of pharmaceutical pollution but whether effects would occur following long-term exposure to lower concentrations is unknown.  相似文献

D. M. Skinner was the first biologist to systematically investigatethe effect of multiple limb autotomy on ecdysis in decapod crustaceans.She proposed the existence of (1) limb autotomy factor anecdysis(LAFan) which initiates precocious molting, and (2) limb autotomyfactor proecdysis (LAFpro) which postpones proecdysis. Meantime to ecdysis did not differ significantly among groups ofsmall juvenile Callinectes sapidus with zero, two, four, andsix limbs removed. Variance was significantly less for the groupmissing six limbs; i.e., autotomy of six limbs synchronizedthe molt-cycle. These patterns were consistent with the hypothesesthat LAFan exerts an additive effect, i.e., more anecdysialcrabs enter proecdysis as more limbs are autotomized and thata delay of proecdysis occurs above a threshold (>4) of autotomizedlimbs. Multiple limb autotomy provides investigators with easyaccess to crabs in metecdysial molt-stages which is useful instudying interactions between rhizocephalan barnacles and theirhosts. While 71% of metecdysial C. sapidus exposed to infectivelarvae of the sacculinid rhizocephalan, Loxothylacus texanus,developed the external stage of the parasite, no similarlysizedanecdysial crab was parasitized, suggesting that these crabsare not susceptible to infection during anecdysis. Size of C.sapidus at infection was inversely proportional to the numberof ecdyses between infection and emergence of the parasite,but not correlated to final host size. These data suggest thatthere is a minimum threshold for host size; smaller hosts undergomore ecdyses before attaining the threshold. Rhizocephalansare being considered as biological control agents and thereis a need to understand how they find and infect hosts. Becauselimb autotomy is such a useful research tool, D. M. Skinner'scontributions to our understanding of how limb autotomy influencesthe crustacean molt cycle will continue to pay significant dividendsin crustacean biology.  相似文献

Chitin synthase (CHS) is the key regulatory enzyme in chitin synthesis and excretion in insects, and a specific target of insecticides. We cloned a CHS B gene of Bombyx mori (BmChsB) and showed it to be midgut specific, highly expressed during the feeding process in the larva. Knockdown of BmChsB expression in the third‐instar larvae increased the number of nonmolting and abnormally molting larvae. Exposure to nikkomycin Z, a CHS inhibitor, reduced the amount of chitin in the peritrophic membrane of molted larvae, whereas abnormally elevated BmChsB mRNA levels were readily detected from the end of molting and in the newly molted larvae. Exogenous 20‐hydroxyecdysone (20E) and methoprene, a juvenile hormone analogue, significantly upregulated the expression of BmChsB when the levels of endogenous molting hormone (MH) were low and the levels of endogenous juvenile hormone (JH) were high immediately after molting. When levels of endogenous MH were high and those of endogenous JH were low during the molting stage, exogenous 20E did not upregulate BmChsB expression and exogenous methoprene upregulated it negligibly. When the endogenous hormone levels were low during the mulberry‐leaf intake process, BmChsB expression was upregulated by exogenous methoprene. We conclude that the expression of BmChsB is regulated by insect hormones, and directly affects the chitin‐synthesis‐dependent form of the peritrophic membrane and protects the food intake and molting process of silkworm larvae.  相似文献

Many environmental pollutants have toxic effects that can alternormal limb regeneration and molting in Crustacea. The mostcommon effect of heavy metals is that of retardation of regenerationof limbs accompanied by a delay in ecdysis; in some cases regenerationis affected without altering the molt cycle. Chlorophenols anddithiocarbamates caused inhibition of regeneration without affectingmolting in shrimp. Organic toxicants such as aromatic hydrocarbonsanddioxins also result in a decrease in the growth increment permolt, while DDT was found to accelerate regeneration and molting.A number of toxicants also produce morphological alterationsin the regenerated limbs of crabs. These may be relatively minor,such as reduced number of pigment cells, setae, or tuberclesin the regenerated limbs (mercury and cadmium), or may be moremajor deformities, such as abnormal bending in the limb or claw(tributyltin), or defects inchitin formation in the exoskeleton(diflubenzuron).  相似文献

We investigated the effects of environmentally relevant concentrations of nonylphenol (NP) on survival, growth, sexual development and molting of the mysid Americamysis bahia (Crustacea: Mysidacea), including assay of 20-hydroxyecdysone (20E) levels. Seven-day-old mysids were exposed to NP concentrations of 0.3, 1, 3, 10 and 30 microg/L for 14 days. Among growth-related traits we assayed, an effect on body length was the most prominent, with the effects observed at concentrations as low as 1 microg/L NP. In addition, the total number of molting exposed to NP for 14 days was significantly decreased in the treatment groups at 10 or 30 microg/L relative to the controls. These results clearly indicate that subchronic exposure to environmentally relevant concentrations of NP affects growth of mysids. Furthermore, we investigated the effect of NP on production of 20E in female mysids during the molting period. Production of 20E in pre-molting mysids exposed to 30 microg/L of NP was lower than those in control groups, and exposure of female mysids to NP disrupted molting cycles. Taken together, the results suggest that environmentally relevant concentrations of NP can disrupt growth of A. bahia via inhibition of 20E levels and an associated delay in molting.  相似文献

Size increase at molt is reduced following multiple limb regeneration in the shore crabs, Hemigrapsus oregonensis and Pachygrapsus crassipes. Limb loss per se does not influence postmolt size. Effect of increasing number of regenerating limbs is additive. Postmolt size is programmed early in the premolt period of the preceding instar and is probably not readily influenced by water uptake mechanics at ecdysis. A simple model for growth, molting, and regeneration in heavily calcified Crustacea is developed from the viewpoint of adaptive strategies and energetic considerations.  相似文献

Aquatic organisms chronically exposed to cadmium can increase their resistance to a subsequent elevated exposure. In order to investigate mechanisms involved in acclimation process in the Chinese crab Eriocheir sinensis, we compared Cd level as well as metallothionein-like protein (MTLP) content in different tissues after direct acute exposure (i.e. 500 microg Cd L(-1) for 3 days), and after acute following chronic (i.e. 10 or 50 microg Cd L(-1) for 30 days) exposure. Cadmium accumulation occurred in the following order: anterior gill>hepatopancreas>posterior gill>carapace>hemolymph>muscle. As high concentrations as 188 microg Cd g(-1) w.w. were reported in anterior gills and seem to reach a saturation level. In these gills, the highest MTLP induction was observed after a direct acute exposure, for which a correlation with Cd content occurred. However, the Cd-binding potential by MTLPs was exceeded for any exposure condition. In hepatopancreas, the highest Cd level was reported for crabs acclimated during 30 days to 50 microg Cd L(-1) before challenging with an acute exposure. Moreover, we showed that MTLPs were induced during the acclimation process. In this organ, MTLPs are theoretically sufficient to bind all Cd. These results suggest that during a chronic exposure to 50 microg Cd L(-1), Chinese crabs acquire the capacity to hold more cadmium in hepatopancreas where it can be sequestrated by MTLPs. On the contrary, MTLP induction seems to be a rapid response to acute exposure in anterior gill, but is not sufficient to sequester all Cd. Other sequestration and/or detoxification mechanisms must take place in anterior gill to cope with high Cd levels.  相似文献

Molting and limb regeneration are tightly coupled processes, both of which are regulated by ecdysteroid hormone synthesized and secreted by the Y-organs. Regeneration of lost appendages can affect the timing and duration of the proecdysial, or premolt, stage of the molt cycle. Autotomy of all eight walking legs induces precocious molts in various decapod crustacean species. In the land crab Gecarcinus lateralis, autotomy of a partially regenerated limb bud before a critical period during proecdysis (regeneration index <17) delays molting so that a secondary limb bud (2 degrees LB) forms and the animal molts with a complete set of walking legs. It is hypothesized that 2 degrees LBs secrete a factor, termed limb autotomy factor-proecdysis (LAF(pro)), that inhibits molting by suppressing the Y-organs from secreting ecdysone. Molting was induced by autotomy of eight walking legs; autotomy of primary (1 degrees ) LBs reduced the level of ecdysteroid hormone in the hemolymph 73% by one week after limb bud autotomy (LBA). Injection of extracts from 2 degrees LBs, but not 1 degrees LBs, inhibited 1 degrees LB growth in proecdysial animals, thus having the same effect on molting as LBA. The inhibitory activity in 2 degrees LB extracts was stable after boiling in water for 15 min, but was destroyed by boiling 15 min in 0.1 N acetic acid or incubation with proteinase K. These results support the hypothesis that LAF(pro) is a peptide that resembles a molt-inhibiting hormone.  相似文献

The surface area of pericardial sacs of the semiterrestrialcrab, Ocypode cordimana, is greater than that of pericardialsacs of the intertidal crabs, Ocypode macrocera and Ocypodeplatytarsis. Responses of the pericardial sacs with relationto conservation of water and molting vary with the species.In the semiterrestrial crab, Ocypode cordimana, the pericardialsacs store water during proecdysis which is used to stretchthe new cuticle after ecdysis. In contrast, the pericardialsacs of the intertidal crabs, Ocypode macrocera and Ocypodeplatytarsis, do not store water during proecdysis. But the sacsof these crabs swell for a brief period during ecdysis, apparentlyto accommodate excess fluid entering the body while the legsare being withdrawn from the old exoskeleton. Although the significanceremains unknown, deposits of uric acid are found in the pericardialsacs of Ocypode cordimana but not in the pericardial sacs ofOcypode macrocera and Ocypode platytarsis. Exposure of specimensof Ocypode cordimana to dry sand reduces the swelling of pericardialsacs, while proecdysial growth of the limbs remains unaffected.The pericardial sacs of Ocypode cordimana swell at a rapid rateif the crabs are maintained in darkness rather than in light.Implications of the present findings are discussed with relationto the habitat of the crabs.  相似文献

Ecdysteroids, the molting hormones in crustaceans and other arthropods, play a crucial role in the control of growth, reproduction and embryogenesis of these organisms. Insecticides are often designed to target specific endocrine-regulated functions such as molting and larval development such as methoprene, a juvenile hormone analogue.The aim of this study was to examine the effects of methoprene on molting in a non-target species, the estuarine mysid Neomysis integer (Crustacea: Mysidacea). Mysids have been proposed as standard test organisms for evaluating the endocrine disruptive effect of chemicals. Juveniles (< 24 h) were exposed for 3 weeks to the nominal concentrations 0.01, 1 and 100 μg methoprene/l. Daily, present molts were checked and stored in 4% formaldehyde for subsequent growth measurements. Methoprene significantly delayed molting at 100 μg/l by decreasing the growth rate and increasing the intermolt period. This resulted in a decreased wet weight of the organism. The anti-ecdysteroidal properties of methoprene on mysid molting were also evaluated by determining the ability of exogenously administered 20-hydroxyecdysone, the active ecdysteroid in crustaceans, to protect against the observed methoprene effects. Co-exposure to 20-hydroxyecdysone did not mitigate methoprene effects on mysid molting. This study demonstrates the need for incorporating invertebrate-specific hormone-regulated endpoints in regulatory screening and testing programs for the detection of endocrine disruption caused by man-made chemicals.  相似文献

对暴露于不同浓度的甲胺磷(0.01、0.1、1.0、10.0、100.0、1 000.0和10 000.0 μg·L-1)和17β-雌二醇(0.001、0.01、0.1、1.0、10.0、100.0、和1 000.0 μg·L-1)溶液中的萼花臂尾轮虫的种群动态进行了研究.结果表明:甲胺磷和17β-雌二醇对轮虫种群动态的影响存在差异.甲胺磷和17β-雌二醇对实验期间轮虫的平均种群增长率均有显著影响,但对轮虫种群中的混交雌体受精率的平均值无显著影响;甲胺磷对实验期间轮虫种群中的携卵雌体数/非携卵雌体数和混交雌体百分率的平均值均有显著影响,而17β-雌二醇则无显著影响;甲胺磷对实验期间轮虫的休眠卵总产量无显著影响,而17β-雌二醇的影响较显著.与对照相比,10.0~10 000.0 μg·L-1的甲胺磷和100.0 μg·L-1的17β-雌二醇均使轮虫的平均种群增长率显著提高;0.1~10 000.0和10.0 μg·L-1的甲胺磷分别显著地降低了实验期间轮虫种群中的携卵雌体数/非携卵雌体数和混交雌体百分率的平均值;1 000.0 μg·L-1的17β-雌二醇显著降低了轮虫的休眠卵总产量.在实验浓度范围内,轮虫平均种群增长率(Y,d-1)与甲胺磷浓度(X,μg·L-1)之间的关系为Y=-2×10-8X2 0.0002X 0.3474.种群增长率可用于监测和评价甲胺磷对轮虫种群动态的影响.  相似文献

The aim of this study was to evaluate the effects of waterborne cadmium on hyper-osmoregulatory capacity of the Chinese mitten crab Eriocheir sinensis acclimated to freshwater. For this purpose, crabs were submitted to acute (0.5 mg Cd L(-1) for 1, 2 or 3 days), chronic (10 or 50 microg Cd L(-1) for 30 days) or chronic, immediately followed by acute, exposure. While no effect was observed after 1 or 2 days, hemolymph osmolality, Na(+) and Cl(-) concentrations were significantly reduced after 3 days of acute exposure. Under this latter condition, the respiratory anterior gill ultrastructure, Na(+)/K(+)-ATPase and cytochrome c oxidase activities were significantly impaired. In contrast, the osmoregulatory posterior gill was unaffected for all treatments. As a consequence, we suggest that the observed hyper-osmoregulatory capacity impairment is the result of increased dissipative flow of ions and/or water through anterior gills. In contrast to acute exposure, chronic exposure did not induce any observable effect. However, crabs submitted to a known deleterious acute condition (0.5 mg Cd L(-1) for 3 days) directly after chronic exposure to 50 microg Cd L(-1) for 30 days showed normal hyper-osmoregulatory capacity with no change in gill Na(+)/K(+)-ATPase activity, and only little disturbance of anterior gill ultrastructure. These results demonstrate that a chronic cadmium exposure can induce acclimation mechanisms related to osmoregulation in this euryhaline decapod crustacean.  相似文献

Bering Sea snow crabs (Chionoecetes opilio) are a commerciallyimportant crab harvested in the Bering Sea. Optimal managementof this species requires an understanding of the biology ofthis crab that is currently incomplete. Fisheries managers applya continuous growth model in their management of snow crab,which assumes that male crabs increase in size throughout theirlifespan. Male snow crabs undergo a morphometric molt that leadsto a disproportionate increase in chelae size and it is stilldebated whether this molt is associated with a terminal molt.This study was conducted to determine whether adult male C.opilio are anecdysic. Using current knowledge of the hormonalregulation of crustacean growth, snow crab physiology was manipulatedto induce an increase in molting hormones (ecdysteroids). Sincefemale snow crabs are known to undergo a terminal molt afterattaining reproductive maturity, we compared ecdysteroid levelsin eyestalk-ablated terminally molted females, small-clawedmales and large-clawed males. Snow crabs were collected fromthe Bering Sea and maintained in circulating seawater at approximately6°C. Animals were either eyestalk-ablated or left intact.Ecdysteroid levels in hemolymph were quantified using an enzyme-linkedimmunosorbant assay (ELISA). Circulating ecdysteroids were significantlyhigher in small-clawed male crabs when compared to large-clawedmales or terminally molted females. Eyestalk-ablation increasedcirculating ecdysteroids in small-clawed males, but had no significanteffect on circulating ecdysteroids in large-clawed males orin terminally molted females.  相似文献

This study examines the effects of Fenoxycarb on larval growth, and lipid class and fatty acid composition in first crabs of the mud crab Rhithropanopeus harrisii reared through total larval development in nominal water concentrations from 1 to 100 microg/l. In first crabs of R. harrisii, dry weight (microg) decreased significantly (P < 0.05) from 228.8+/-38.2 microg (n = 9) in the controls to 131.8+/-10.1 microg (n = 4) in animals exposed throughout larval development to 100 microg/l. A significant (P < 0.05) reduction was found between total lipid content in the controls and first crabs reared at concentrations greater than 50 microg/l. In relative terms (% dry weight), different lipid classes predominated in the controls and the various fenoxycarb exposure concentrations. There were no significant (P > 0.05) differences among the treatment groups in phospholipid level, while the triglyceride content was significantly lower in crabs exposed to 10 and 100 microg/l. No significant differences in the percent of free fatty acids were found in crabs exposed to 1-10 microg/l and the controls. Free sterols in crabs exposed to concentrations higher than 10 microg/l were below the detection limit. Control animal fatty acid profiles were dominated by palmitic, stearic, and oleic acid, accounting for 48% of total fatty acids (TFA). The fatty acid composition of crabs exposed to 100 microg/l significantly (P < 0.05) differed from the controls. The results suggest that fenoxycarb has substantial effects on growth, lipid class and fatty acid composition in developing larvae of R. harrisii at water concentrations greater than 10 microg/l.  相似文献

Methoprene (an analogue of juvenile hormone) application and feeding on a protein diet is known to enhance male melon fly, Bactrocera cucurbitae Coquillett (Diptera: Tephritidae), mating success. In this study, we investigated the effect of these treatments on male B. cucurbitae's ability to inhibit female remating. While 14‐d‐old females were fed on protein diet, 6‐d‐old males were exposed to one of the following treatments: (i) topical application of methoprene and fed on a protein diet; (ii) no methoprene but fed on a protein diet; (iii) methoprene and sugar‐fed only; and (iv) sugar‐fed, 14‐d‐old males acted as controls. Treatments had no effect on a male's ability to depress the female remating receptivity in comparison to the control. Females mated with protein‐deprived males showed higher remating receptivity than females first mated with protein‐fed males. Methoprene and protein diet interaction had a positive effect on male mating success during the first and second mating of females. Significantly more females first mated with sugar‐fed males remated with protein‐fed males and females first mated with methoprene treated and protein‐fed males were more likely to remate with similarly treated males. Females mating latency (time to start mating) was significantly shorter with protein‐fed males, and mating duration was significantly longer with protein‐fed males compared with protein‐deprived males. These results are discussed in the context of methoprene and/or dietary protein as prerelease treatment of sterile males in area‐wide control of melon fly integrating the sterile insect technique (SIT).  相似文献

Recent declines in red king crab (Paralithodes camtschaticus) stocks in its native and introduced habitat have sparked interest in the development of aquaculture methods for this commercially important species. Little is known about the basic biology of this species and the factors controlling its growth rate. In this paper we present concentrations of circulating ecdysteroids (the hormones that control molting) in hemolymph of intermolt red king crabs in three coastal areas of the Barents Sea. Two molting hormones (20-hydroxyecdysone and ecdysone) were assayed. Mean levels of these ecdysteroids varied from 0.0 to 190.0 μg ml−1 and from 0.0 to 13.4 μg ml−1, respectively. These levels in general were higher in comparison with other decapod species. Concentrations of ecdysteroids were similar in male and female crabs and in injured (animals with at least one autotomized limb) and intact red king crabs. In contrast, the levels of circulating ecdysteroids were much higher in small (predominantly immature crabs) than in large adult animals because the latter have a lower molting probability (once per year) than smaller crabs (2–3 times per year). Our data can be used in further investigations of red king crab growth rates and their application to the development of aquaculture methods for this species.  相似文献

Chitin synthase is the key regulatory enzyme for chitin synthesis and excretion in insects, as well as a specific target of insecticides. The chitin synthase A gene (BmChsA) cloned from Bombyx mori, the model species of lepidopteran, is an epidermis-specific expressed gene during the molting stage. Knockdown BmChsA gene in 3rd instar larvae increased the number of non-molting and abnormal molting larvae. Exposure to nikkomycin Z, a chitin synthase inhibitor downregulated the expression of BmChsA and decreased the amount of epidermis chitin during the molting process. The thickness of the new epidermis and its dense structure varied greatly. The exogenous hormones significantly upregulated the expression of BmChsA with low levels of endogenous MH and high levels of endogenous JH immediately after molting. With low levels of endogenous hormones during the mulberry intake process, BmChsA was rarely upregulated by exogenous hormones. With high levels of endogenous MH and low levels of endogenous JH during the molting stage, we did not detect the upregulation of BmChsA by exogenous hormones. The expression of BmChsA was regulated by endocrine hormones, which directly affected the chitin synthesis-dependent epidermal regeneration and molting process.  相似文献

Adult male shore crabs (Carcinus maenas) were maintained on a fish-based diet supplemented with 0, 5 or 10% chitin for 11 weeks. Significantly greater mortality was found during this period in the control group (0% chitin) than those fed 10% chitin. Crabs fed 5 or 10% chitin had lower numbers of cultivatable bacteria in the hepatopancreas than those on the basal diet alone. The addition of chitin had no significant effect on the serum concentrations of protein and glucose, and the levels of glycogen in the hepatopancreas. The total number of circulating hemocytes in the blood was unaffected by the addition of chitin to the diet, however, at week 6 there were significantly more hyaline hemocytes in those crabs fed 10% chitin than the control group. The in vitro phagocytic activity of hemocytes was unaffected by chitin supplementation and crabs challenged with Vibrio alginolyticus showed a similar pattern of susceptibility in the three dietary groups (0, 5 or 10% chitin). Overall although crabs on a chitin-supplemented diet showed greater survival, this was not explained in terms of elevation in the cellular defences of these animals. The enhanced survival of crabs-fed chitin is probably as a result of the removal of potentially pathogenic bacteria from the hepatopancreas. Because chitin appears to 'purge' bacteria from the gut, this may prove to be a useful addition to diets on animals undergoing oral probiotic treatment.  相似文献
