Summary Interspecific hybrids of the mungbean, Vigna radiata (L.) Wilczek (2n=22) and V. glabrescens (2n=44) were generated with the aid of embryo culture. V. glabrescens x V. radiata hybrids were recovered via germination of the immature embryos. Reciprocal hybrids were obtained via shoot formation from embryonic callus. The authenticity of the hybrids was determined by morphological characteristics, chromosome number, and isozyme patterns. The hybrids were highly sterile upon selfing, but backcrossing to the diploid parent yielded viable seeds. Some of the plants resembled the diploid parent morphologically while others resembled neither parent. The backcross plants were sufficiently fertile to give a large number of mature, selfed seeds. Plants obtained differed morphologically and in their isozyme patterns from either parent, indicating introgression. These progeny populations will be used as bridging materials to transfer pest resistance from the wild tetraploid to the cultivated mungbean.  相似文献

Pelargonium incarnatum is a tetraploid species (2n = 40) andP. patulum is diploid (2n = 22). A natural hybrid, collected at the same locality as the parent species, has 2n = 42 chromosomes. The meiosis of the hybrid is practically normal and its pollen fertility corresponds to that ofP. incarnatum. The flavonoid patterns of the parent species and the hybrid are very similar. The origin and phylogenetic implications of this intersectional hybrid are discussed.  相似文献

A putative natural hybrid betweenOrchis laxiflora andO. morio (Orchidaceae) from southern Italy, formerly known asO. alata, was characterized both on morphological and molecular grounds in order to confirm its hybrid status and to trace its maternal lineage. The morphological characters of the putative hybrid showed intermediacy between those of the parent species, and restriction fragment length polymorphism (RFLP) analysis of a region of the nuclear ribosomal DNA confirmed its hybrid origin. Chloroplast DNA RFLP analysis indicated thatO. morio provided the maternal genome.  相似文献

Towner , Joseph W. (U. California, Los Angeles.) Cytogenetic studies on the origin of Tagetes patula. I. Meiosis and morphology of diploid and allotetraploid T. erecta × T. tenuifolia. Amer. Jour. Bot. 48 (9): 743–751. Illus. 1961.—Cytological and morphological evidence is presented to show that Tagetes patula L. is an allotetraploid species (2n = 48) which probably originated by hybridization between the diploids T. erecta L. and T. tenuifolia Cav., or species closely related to them. The putative parent species are separated by strong reproductive isolation barriers, and controlled crosses between them yielded very few good seed. Most of the hybrid seedlings died before reproductive age, and the few survivors were sterile and extremely weak. At meiosis in the diploid hybrid, bivalent frequency per cell was 0–11 and averaged 4.4. Although 43.6% of the quartet stage consisted of restitution dyads, pollen was essentially all non-viable and seed set was less than 1%. A true-breeding, fertile, allotetraploid T. erecta-tenuifolia was synthesized by colchicine-doubling of the chromosome number of the F1 hybrid. In the synthetic allotetraploid, average chromosomal association was 23.97 bivalents plus 0.05 univalents, and 97.4% of MI cells had 24 bivalents; there were no multivalents, bridges, or fragments. Tagetes erecta-tenuifolia showed a striking morphological similarity to T. patula. Both the natural and synthetic allotetraploids were intermediate between T. erecta and T. tenuifolia for nearly every character. Of 10 species of Tagetes whose chromosome numbers are known, 5 are tetraploid (2n = 48).  相似文献

To examine interspecific relationships and test the hypothesis of hybrid origin within Lycoris species, this study used data from parsimony analyses with nuclear ITS sequences for 19 taxa representing 14 species of Lycoris and two outgroup taxa. The ITS sequences resolved three infrageneric clades. One clade included L. chinensis, L. longituba, L. longituba var. flava, L. anhuiensis, and L. aurea; the second one consisted of L. sprengeri, L. radiata, L. radiata var. radiata, L. radiata var. pumila, L. haywardii, L. rosea, L. sanguinea var. sanguinea, and L. sanguinea var. koreana; and the third included L. caldwellii, L. straminea, L. albiflora, L. flavescens, and two hybrids. The results strongly support the hypothesis that L. straminea originated from hybridization between L. chinensis and L. radiata var. pumila, and the allotriploid L. caldwellii and L. albiflora derived from hybridization between L. chinensis and L. sprengeri. As nucleotide additivity was observed in the artificial hybrids and several presumed hybrids, the likelihood of hybrid origin of Lycoris species is supported.These authors contributed equally to this work.  相似文献

Poly(ADP-ribose) polymerase (PARP) is a nuclear enzyme that recognizes and binds to the nicks and ends of DNA, and catalyses successive ADP-ribosylation reactions. To clarify the function of PARP at the molecular level, we searched proteins which interact with PARP. In the auto-modification domain of PARP in Drosophila, there is a putative leucine-zipper motif which can interact with other protein molecules. To find interacting proteins we examined the auto-modification domain of Drosophila PARP, using the Far-Western screening method. From six independent cDNA clones isolated, we characterized two clones, PBP-3 and PBP-12. The predicted amino acid sequences from 109 to 269 of PBP-3 and from 184 to 312 of PBP-12 had more than 62% identities to mammalian L23a (rpl23a) and L22 (rpl22), the ribosomal proteins of the large subunit. This indicated that PBP-3 and PBP-12 are Drosophila homologues of L23a and L22, respectively. These Drosophila ribosomal protein L22 and L23a have additional Ala-, Lys- and Pro-rich sequences at the amino terminus, which have a resemblance to the carboxy-terminal portion of histone H1. Thus, Drosophila L22 and L23a might have two functions, namely the role of DNA-binding similar to histone H1 and the role of organizing the ribosome.  相似文献

Homoploid hybrid plant species are rare, and the mechanisms of their speciation are largely unknown, especially for homoploid hybrid tree species. Two contrasting hypotheses have been proposed to explain the origin of Hippophae goniocarpa: (1) it is a diploid hybrid originating from H. rhamnoides ssp. sinensis × H. neurocarpa ssp. neurocarpa, and (2) it originated via marginal differentiation from H. rhamnoides ssp. sinensis. Regardless of which of these hypotheses is true (if either), previous studies have suggested that H. rhamnoides ssp. sinensis is the only maternal donor for this hybrid species. In this study, we aim to elucidate the maternal composition of H. goniocarpa and to test the two hypotheses. For this purpose, we sequenced the maternal chloroplast DNA trnL‐F region of 75 individuals representing H. goniocarpa, H. rhamnoides ssp. sinensis, and H. neurocarpa ssp. neurocarpa in two co‐occurring sites of the taxa. Seven haplotypes were identified from three taxonomic units, and their phylogenetic relationships were further constructed by means of maximum parsimony, maximum likelihood, and network analyses. These seven haplotypes clustered into two distinct, highly divergent lineages. Two haplotypes from one lineage were found in H. rhamnoides ssp. sinensis, and five (representing the other lineage) in H. neurocarpa ssp. neurocarpa. Hippophae goniocarpa shared four common haplotypes from both lineages, but the haplotypes detected from the two populations differed to some extent, and in each case were identical to local haplotypes of the putative parental species. Thus, both H. rhamnoides ssp. sinensis and H. neurocarpa ssp. neurocarpa appear to have together contributed to the maternal establishment of H. goniocarpa. These results clearly demonstrate that the marginal origin hypothesis should be rejected, and support the hybrid origin hypothesis. Hippophae goniocarpa exhibits a sympatric distribution with its two parent species, without occupying new niches or displaying complete ecological isolation. However, this species has effectively developed reproductive isolation from its sympatric parent species. The development of early ovules, megasporogenesis, and megagametogenesis in Lotus corniculatus L. (2n = 4x = 24), L. conimbricensis L. (2n = 2x = 12), and their protoplast fusion hybrid were studied. The protoplast fusion hybrid plant was known to be completely male sterile and appeared female sterile. The objectives of this study were to examine the development of the ovules and megagametophytes of the protoplast fusion hybrid of L. corniculatus × L. conimbricensis and to compare the development of the ovules and megagametophytes of the fusion hybrid with that of the parent types. The normal development of the ovules and megagametophytes of L. corniculatus, L. conimbricensis, and their protoplast fusion hybrid were similar to those of several other species of Fabaceae. Approximately 24% of the protoplast fusion hybrid ovules developed into apparently normal megagametophytes beyond the critical reduction divisions. The abortion of ovules in the protoplast fusion hybrid occurred at any stage of megasporogenesis or megagametogenesis, but most abortions appeared to be due to postmeiotic failures. Evidence that normal-appearing megagametophytes of the fusion hybrid are capable of being fertilized and forming zygotes is needed.  相似文献

Amelanchier bartramiana, which has been shown to be sexual and self-incompatible, and A. laevis, in which apospory and self-compatibility occur, grow in a mixed population in western Maine. Also present there is the putative interspecific hybrid, A. × neglecta. Principal components and canonical discriminant analyses of seven morphological characters from 96 parental and putative hybrid individuals substantiate morphological intermediacy of the hybrids. A Nomarski differential interference microscopy study of megasporogenesis and megagametogenesis in five hybrid plants shows a predominantly aposporous mode of reproduction, and controlled pollinations indicate that seed formation between A. bartramiana and A. laevis is possible and that the hybrids are self-compatible. The hybrids, which are tetraploids (n = ca. 34) like both parents in Maine, produce viable pollen and seed, but there is little evidence for backcrossing with either parent.  相似文献

Summary One of the aims of the interspecific crossing programme between Hordeum vulgare L. and H. bulbosum L. has been to introgress desirable genes into barley from the wild species. However, despite their close taxonomic relationship there have been few reports of achieving this objective using amphidiploid hybrids. In order to broaden the range of available hybrids, partially fertile triploids between H. vulgare (2n = 2x = 14) and H. bulbosum (2n = 4x = 28) were developed and crossed with H. vulgare as female parent. From 580 progeny which were screened, eight putative single monosomic chromosome substitution lines and one double monosomic substitution were identified by cytological analysis. These involved the substitution of H. vulgare chromosome 1 (two lines), 6 (four lines), 6L (one line), 7 (one line) and 1 + 4 (one line) by their respective H. bulbosum homoeologues. The H. bulbosum chromosome was frequently eliminated during plant development, but it was observed regularly in pollen mother cells of two lines. However, pairing between the H. bulbosum chromosome and its H. vulgare homoeologue was low. Several of the lines were more resistant than their H. vulgare parents to powdery mildew (Erysiphe graminis DC. f.sp. hordei Em. Marchai), net blotch (Drechslera teres Sacc.) and scald (Rhynchosporium secalis (Oudem.) Davis). Apart from their use in studying genome relationships, their value to plant breeders will depend on the ease of inducing translocations between the parental chromosomes.  相似文献

Summary Successful crossing is reported between L. atlanticus Gladst. (2n = 38) and L. cosentinii Guss. (2n = 32), using lines of both species selected for crossability followed by selection of relatively fertile progenies. In one cross, 82E75, from a single F2 segregating plant, 22 F3 seeds were obtained. Some other less crossable combinations were completely sterile in the f1 or F2. Backcrossing to both parent species was successful, but some crosses gave relatively more seed by using F2 plants for backcrossing rather than F2's. It is concluded that potential exists for introgression of useful genes in both directions.  相似文献

The crossability between Brassica tournefortii (TT, 2n = 20) and Brassica rapa (AA, 2n = 20) and the cytomorphology of their F1 hybrids were studied. Hybrids between these two species were obtained only when B. tournefortii was involved as a female parent. The hybrid plants were intermediate for most of the morphological attributes and were found to be free from white rust under field conditions. The F1 plants showed poor pollen fertility, although occasional seed set was achieved from open pollination. Self-pollination or backcrosses did not yield any seeds in these plants. The occurrence of chromosome association ranging from bivalents (0–7), trivalents (0–2) to a rare quadrivalent (0–1) in the dihaploid hybrids indicates pairing between the T and A genomes. The homoeologous pairing coupled with seed set in the F1 plants offer an opportunity for interspecific gene transfers from B. tournefortii to B. rapa and vice-versa through interspecific hybridization. 垫状植物是高山冰缘带植物区系的重要组成成分,是植物适应高寒环境趋同进化的特殊生活型。该研究以采自西藏和云南高山冰缘带的5种石竹科垫状植物种子为材料,采用滤纸萌发法、常规压片法镜检观察分析其核型,为青藏高原垫状植物积累核型资料,并为探讨极端环境下特殊生活型植物的核型进化机制提供依据。结果表明:5种植物均为二倍体,染色体核型分别为:密生福禄草(Arenaria densissima),2n=2x=20m+2sm,2A;山生福禄草(A.oreophila),2n=2x=22m,1A;团状福禄草(A.polytrichoides),2n=2x=20m+2sm,2A;大花福禄草(A.smithiana),2n=2x=20m+2sm,1A;囊种草(Thylacospermum caespitosum),2n=2x=18m+4sm,2A。囊种草与福禄草亚属植物的染色体数目一致,为二者在分子系统发育框架下形成的独立分支提供核型证据。并讨论了5种垫状植物古多倍体物种形成的核型进化与中新世青藏高原及周边地区地质抬升的关联。  相似文献

Axenic cultures of Trentepohlia species are necessary for the study of growth and hysiological characters of the algae. We describe the use of a Sherman micromanipulator to isolate filaments from samples of T. aurea and T. odorata collected from their natural habitats. These filaments were then used as inocula for the establishment of axenic cultures. In the case of T. aurea, further treatment with lactic acid was necessary.  相似文献

The taxonomic delimitation of Hesperis bicuspidata (Willd.) Poir. and H. stellata F. Dvo?ák is problematic, and we therefore studied morphological, palynological, seed‐coat surface and karyological features of both species to determine their degree of similarity. The morphology was described using 13 characters, each based on ten measurements on different specimens. Hesperis stellata and H. bicuspidata both show large variation in vegetative and reproductive organs; petal length varies between 18 and 26 mm and petal width is 3–7 mm; the corolla is lilac, whitish lilac, violet, dark violet or white in color, and fruits of both species are generally glabrous or have very short bifurcate hairs. Pollen and seed morphology were examined by LM and SEM. Pollen of both species is prolate‐spheroid or subprolate, tricolpate and reticulate. Their exine structures are the same. Hesperis bicuspidata is diploid (2n=14) but different chromosome types were present in the two studied specimens. Hesperis stellata is tetraploid (2n=28) showing the same chromosome types as the samples of H. bicuspidata. Eighteen morphometrical characters of H. bicuspidata, H. stellata, H. pisidica and H. armena were analysed by ANOVA. Notes on ecology and distribution are also presented for H. bicuspidata. Based on the results, H. stellata is reduced to a synonym of H. bicuspidata.  相似文献

棱果沙棘为同域分布的中国沙棘和肋果沙棘同倍化自然杂交形成的。本文利用母系遗传的cpDNA trnS-G序列检测青海祁连棱果沙棘及其亲本中国沙棘和肋果沙棘同域分布的两个地区(拱北湾、八宝河滩)共93个个体的遗传关系。结果表明棱果沙棘及其亲本在拱北湾和八宝河滩分别有12个和7个单倍型,两地区的棱果沙棘都与其亲本共享单倍型,其中拱北湾棱果沙棘共36个个体中有28个与中国沙棘共享3个单倍型(H2,H4,H5),有2个个体与肋果沙棘共享单倍型(H11),八宝河滩的棱果沙棘共10个个体中有7个与中国沙棘共享一个单倍型(H4),3个与肋果沙棘共享单倍型(H7)。应用最大简约法(MP)分别对两地区的棱果沙棘及其亲本trnS-G序列构建的系统发育树中棱果沙棘的大部分个体都与中国沙棘聚在一起,另外,棱果沙棘4种特有单倍型(H3、H7、H8、H9)的6个个体在系统树上也与中国沙棘聚为一支。以上结果进一步证明了二倍体自然杂交种棱果沙棘为双向杂交起源,但其主要母本来源应为中国沙棘。  相似文献

Results of a karyological analysis of eight species collected from northeast Anatolia, Turkey are presented. The following chromosome numbers were determined:H. medianiforme (Litv. etZahn)Juxip 2n=36,H. karagollense (Zahn)P.D. Sell etC. West 2n=36,H. argillaceoides (Litv. etZahn)Juxip 2n=36,H. amblylepis Boiss. 2n=27,H. lanceolatum Vill. 2n=27,H. asterodermum (Woronow etZahn)Juxip 2n=27,H. umbellatum L. 2n=18,H. laevigatum Willd. 2n=27. The chromosome numbers of five of these seven species are presented for the first time. The results show a high proportion of triploids and tetraploids within the taxa studied.  相似文献

The two closely related species,Phaseolus aureus andP. mungo were crossed successfully using the former as the female parent. A detailed cytological study of the hybrid, which showed morphological characters of both the parents, was carried out.At the pachytene stage, the pairing behaviour of 9 out of the 11 chromosomes belonging to the haploid complement of the two parents was investigated. The remaining two chromosomes are represented by morphologically distinct bivalents. The two species differ by a translocation, a deletion and a duplication. A consideration of the data from comparative morphology, geographical distribution, protein characters and genetical studies in the F2 and back cross generations, indicates a very close relationship between the two diploid species. Chromosome differentiation seems to constitute the major difference. The karyological evidences seem to indicate thatmungo is derived fromaureus.  相似文献

以紫薇(Lagerstroemia indica)、尾叶紫薇(L.caudata)、屋久岛紫薇(L.fauriei)和福建紫薇(L.limii)4种紫薇属植物为材料,利用染色体荧光原位杂交技术(FISH)获得了4种紫薇属植物的有丝分裂中期染色体FISH图及核型参数,分析了45SrDNA在紫薇属植物染色体上的数量和分布特点。结果表明,4种紫薇属植物染色体上均具有1对45SrDNA杂交位点,位于较长染色体短臂的近端部,紫薇、尾叶紫薇、屋久岛紫薇和福建紫薇的核型公式分别为2n=48=2M+24m+22sm、2n=48=30m+18sm、2n=48=2M+20m+26sm和2n=48=2M+32m+14sm,均为2A型。该研究首次获得了紫薇属植物45SrDNA荧光原位杂交核型,为紫薇属植物亲缘关系研究和细胞生物学研究提供了分子细胞学依据。  相似文献
