Five genotypes of swede (Brassica napus var. napobrassica), two genotypes of kale (B. oleracea var. acephala), and two genotypes of rape (B. napus var. napus) were each inoculated at the 8–10 true leaf stage with five cabbage root fly (Delia radicum) eggs. The percentage pupation after larval feeding on individual plant genotypes ranged from 45 to 78%, and the mean pupal weight from 6.5 to 13.0 mg. After 5 weeks, larval feeding damage had reduced root weight by up to 47%, compared with uninoculated plants. The dry matter content of undamaged roots was higher in the kales and rapes than in the swedes. Whilst the dry matter content of the rapes and swedes were not changed by D. radicum damage, that of the kales was elevated. The ethanol-soluble sugar content of the root was reduced in all cases by D. radicum larval damage. However, the effect of D. radicum damage on the concentrations of individual sugars (glucose, fructose and sucrose) was crop- and genotype-dependent. In the roots of kales and rapes, the glucose and fructose concentrations were either very low or unaffected by D. radicum damage, whilst both glucose and fructose were generally reduced in swede roots by D. radicum damage. The root sucrose concentration was either reduced or not significantly affected by D. radicum damage in all of the crop types tested. The percentage pupation and the mean pupal weight of D. radicum were inversely correlated to root freeze-dried matter content. D. radicum pupal weight was positively correlated with root fructose, glucose and ethanol-soluble sugar contents.  相似文献

In this study, interactive effects of plant competition and herbivory on plant quality and herbivore development were examined in a greenhouse experiment where cabbage plants [Brassica oleracea L. var. capitata (Brassicaceae)] were intercropped with red clover [Trifolium pratense L. (Fabaceae)]. Cabbages were grown with two red clover densities and attack rates by the root feeding herbivore the turnip root fly, Delia floralis Fall. (Diptera: Anthomyiidae). Above ground and below ground cabbage biomass was reduced through intercropping and larval damage. Intercropping also resulted in lower nitrogen and higher carbon root levels compared with levels in the roots of monocultured cabbage. Furthermore, both root nitrogen and carbon levels increased with herbivory. Root neutral detergent fibre (NDF) and lignin content increased in response to both increased plant competition and higher egg densities. For lignin, an interaction effect was observed in the form of elevated levels in intercropped plants subjected to larval damage, while levels in roots of monocultured cabbage remained unchanged. The quality changes brought about by clover competition affected D. floralis development negatively, which resulted in reduced pupal weight. In addition, increased egg density also decreased larval growth. The effects on the development of D. floralis in relation to host plant quality are discussed. Handling editor: Gimme Walter  相似文献

Field trials in two years indicated that egg-laying antixenosis was the major component of resistance in swede cvs Angus and Melfort against root flies (Delia floralis and D. radicum). Between four and eight times as many D. floralis eggs were laid on the susceptible cv. Doon Major as on the resistant cv. Angus. The degree of antixenosis effective against D. radicum was more variable in 1985, due to low numbers of adult flies, but in 1986, when adult fly numbers were higher, D. radicum laid 34 times as many eggs on susceptible cv. Sator Øtofte as on Angus. Root antibiosis against larval feeding is a second but less important component of resistance against D. floralis. In egg inoculation tests, 1.3-2.0 times more D. floralis pupae developed on Doon Major than on Angus or Melfort. Larval feeding on resistant cultivars was restricted to surface root tissue only, compared with deeper tunnelling on susceptible cultivars. Tolerance to root damage may also occur under certain conditions. Although increased dry matter content was significantly correlated with root antibiosis, its effect on larval feeding was apparently not due to increased tissue hardness. It seems likely that chemical differences in the outer root tissues influence larval feeding and development.  相似文献

Red beet hairy root cultures, obtained after genetic transformation with Agrobacterium rhizogenes, are completely heterotrophic and synthesize betalaines (BNs). Upon subjecting the hairy roots to treatments containing different sugars (3% w/v) it was found that sucrose was rapidly utilized, followed by maltose, and a very limited use of glucose, but the other hexoses – fructose, lactose, xylose and galactose or glycerol totally suppressed both growth and BN synthesis. No habituation or adaptability to maltose or glucose occurred, evidenced by the lack of growth upon re-culture in respective medium. Glycerol, was not taken up alone, but was utilized to a considerable extent in the presence of low levels of sucrose for growth only but not BN synthesis. Red beet hairy root culture did not exogenously hydrolyse sucrose to hexoses, as there were only traces of reducing sugar present in the medium soon after inoculation, without an increase later, confirmed by HPLC. There was an increase in medium osmolarity in the presence of fructose indicating the exudation of certain compounds from the roots. Red beet hairy roots appear useful as a model system to study sugar metabolism/signalling due to their sensitivity to different sugars that may directly link to morphological changes and BN synthesis.  相似文献

A field cage method was evaluated as an alternative to field trials for identifying sources of resistance to turnip root fly in brassicas. This involved the controlled release of adult flies and the use of egg traps to quantify egg-laying on potted plants. Overall levels of resistance to Delia floralis, and its components, were assessed for 15 swede genotypes. Significant levels of antixenosis and antibiosis type resistance were identified in several cultivars and breeding lines. There was a close agreement between the field cage results and those from previous field trials, using comparable susceptible and resistance control cultivars. Compared with field selection, the field cage method reduced by c. 40% the time required to quantify resistance and each of its components.  相似文献

In Brassica crops differences in susceptibility to root fly attack can be largely attributed to antixenotic resistance. Plants of four genotypes (two swedes and two kales) with widely differing resistance in field trials, were compared in laboratory choice assays for their susceptibility to oviposition by the root flies Delia radicum (L.) and D. floralis (Fallen) (Diptera, Anthomyiidae). For both species the preference among the genotypes corresponded to the susceptibility of the genotypes in the field. The preference ranking in response to surrogate leaves treated with methanolic surface extracts of the four genotypes was identical to the preference among potted plants, demonstrating that chemical factors on the leaf surface mediate host preference for oviposition in these species.For both species of fly, glucosinolates are major oviposition stimulants and for D. radicum an additional, nonglucosinolate oviposition stimulant, presently called CIF, is known. We describe a procedure for chromatographic separation of glucosinolates from CIF in leaf surface extracts. In oviposition-choice assays with D. radicum, the CIF-fractions of the two swede genotypes applied to surrogate leaves received a 1.8 and 4.6 times higher proportion of eggs than the respective glucosinolate-fractions, confirming the major importance of CIF as an oviposition stimulant. The genotype of swede that was preferred by both fly species in tests with plants and methanolic leaf surface extracts, also stimulated oviposition more in tests with the glucosinolate-fractions or the CIF-fractions derived from the surface extracts, respectively. Thus, glucosinolates and CIF together account for the observed preference among the genotypes and may also be responsible for their susceptibility under field conditions. In the two kale genotypes the preference for plants or surface extracts differed from the preference among the corresponding glucosinolate- and CIF-fractions, indicating that additional, as yet unknown chemical factors may also be involved.For both groups of stimulants tarsal chemoreceptors allow electrophysiological monitoring of glucosinolate- and CIF-activity in fractionated surface extracts. For D. radicum the chemosensory activity of both glucosinolate- and CIF-fractions corresponded to the respective behavioural activity in the oviposition preference tests, suggesting that preference for oviposition among genotypes can be predicted from the electrophysiological activity of their fractions. The chemosensory response of D. floralis, in particular to the CIF-fractions, was less pronounced than the response of D. radicum, indicating interspecific differences in the perception of the major oviposition stimulants. We discuss the potential application of electrophysiological techniques in support of other screening methods used in breeding for root fly resistance in Brassica crops.  相似文献

Petroleum ether, acetone, water and aqueous extract of Pedalium murex L. (Pedalaceae) leaves, root and fruit were tested against fourth instar larvae of the tobacco cutworm, Spodoptera litura (Fabr.) under laboratory conditions using leaf dip method. Larval mortality; larval, pupal and adult periods; pupal weight, pupation and adult emergence; larval and pupal deformities of S. litura were recorded. All the tested solvent extracts of P. murex were effective against S. litura life stages by causing mortality in a dose dependent manner. However, the efficacy was more significant with respect to acetone (leaf and root) and petroleum ether (root) at higher concentrations (0.8%) which leads to 100% larval mortality. The water and aqueous extracts of root caused 86.6 and 88.0% larval mortality respectively at 4% concentration. Among the plant parts tested, root had more importance followed by leaf and fruit. Plant extracts extended the larval, pupal and adult periods; reduced the pupal weight, pupal and adult emergence and caused larval–pupal deformities. Total life time was highly prolonged in acetone extracts of P. murex root. Though all the extracts were found to have insecticide activity, acetone and petroleum ether extracts of P. murex root can be used as an effective alternative to modern synthetic insecticides. Bioactive principles from these extracts can be isolated, identified and integrated in S. litura management.  相似文献

The effects of nitrogen fertilizer on sweet potato, Ipomoea batatas (L.) Poir., resistance to the sweetpotato weevil, Cylas formicarius elegantulus (Summers), was studied. Adult weevil feeding and oviposition preference, larval survival, and pupal weight were used as measures of sweet potato resistance. Sweet potato resin glycosides and caffeic acid concentrations in the periderm tissue of storage roots also were measured. Sweet potato genotypes (Beauregard, Excel, W-244, W-250, and Sumor) with varying levels of resistance to sweetpotato weevil were grown in the field under three nitrogen regimes (0, 45, and 135 kg N/ha). Harvested storage roots were evaluated in the laboratory for feeding and oviposition activity of sweetpotato weevil female adults under no-choice and choice test conditions. Larval survival rate and pupal weight were determined by rearing the insects individually on storage root sections. Nitrogen level had a significant effect on the number of eggs deposited, but not on the number of feeding punctures. Sweetpotato weevils laid fewer eggs on plants with the highest level of nitrogen. Nitrogen levels did not significantly affect larval survival and pupal weight. Genotype had a significant effect on feeding, oviposition, and larval survival. Beauregard had higher levels of feeding, oviposition, and larval survival compared with the other genotypes. No interaction effects between nitrogen and genotype were significant. Resin glycosides and caffeic acid concentrations were significantly different among genotypes and between years. Nitrogen levels significantly affected the concentrations of caffeic acid in 1997.  相似文献

  • 1 The relative importance of the resource concentration hypothesis and the enemies hypothesis was investigated for the turnip root fly Delia floralis in a cabbage–red clover intercropping system compared with a cabbage monoculture.
  • 2 Delia floralis egg densities were measured as well as the activity‐densities of generalist predators in a field experiment during two growing seasons. In the second year, a study of egg predation with artificially placed eggs was conducted, in addition to a predator exclusion experiment, to estimate total predation during the season. Parasitization rates were estimated from samples of pupae.
  • 3 Delia floralis oviposition was greater in the monoculture during both years. The predator activity‐densities differed between treatments and study years. The known natural enemies of Delia spp., Bembidion spp. and Aleochara bipustulata showed a strong response to a cultivation system with higher activity‐densities in the monoculture. The response, however, appeared to be caused primarily by habitat preferences and not by D. floralis egg densities.
  • 4 The reduction in the number of D. floralis pupae in the intercropping may be explained by a disruption in oviposition behaviour caused by the presence of clover because neither predation, nor parasitization rates differed between cultivation systems.

In general, plant material grown in vitro has low photosynthetic ability to achieve positive carbon balances. Therefore, a continuous supply of carbohydrates from the culture medium is required, and sucrose has been the most commonly used carbon source. In this paper, we investigate the effects of different sucrose concentrations and the presence and absence of light on the endogenous levels of soluble carbohydrates and starch as well as on the proliferation and growth of Dendrobium Second Love (Orchidaceae) in vitro. The possibility of using etiolated stem segments as a means for micropropagating this hybrid was also verified. The results obtained indicated that the presence and absence of light and the sucrose concentrations used influenced the amounts of soluble carbohydrates and starch and the proliferation of D. Second Love shoots and roots. An increase in sucrose concentration caused a progressive increase in the amounts of total carbohydrates and starch. Under both light conditions, sucrose was the main sugar found in the shoots followed by glucose and fructose. The addition of sucrose to the culture medium up to 2% and 4% was advantageous to the number of shoots produced per explant and the root longitudinal growth in the presence and absence of light, respectively. Shoot and root dry matter and the number of roots formed per explant increased as sucrose concentration was raised up to 6% in both light treatments. The use of dark-grown shoot segments proved to be a useful and reliable alternative for the micropropagation of this hybrid.  相似文献

Life history traits and stress tolerance were studied in four domestic species of DrosophilaD. melanogaster, D. simulans, D. auraria and D. immigrans– to understand how they adapt to their environments. In all species, larval weight approximately doubled in 1 day. The relative egg weight (egg weight : pupal weight) was smaller and the larval period was longer in D. immigrans than in the other three species. The pupal period was the longest in D. auraria. However, the adaptive significance of these differences in larval and pupal periods was not clear. The pupal case was generally thicker in the larger species, probably to support the larger pupal body. The start of oviposition was earliest and reproductive effort was greatest in female D. simulans, followed by female D. melanogaster. In contrast, starvation tolerance and the increase in bodyweight after eclosion was greater in D. immigrans and D. auraria than in the other two species. Pupal desiccation tolerance was greatest in D. melanogaster and lowest in D. auraria, and the less tolerant species seemed to select more humid sites for pupation. Adult tolerance to desiccation was greatest in D. melanogaster and lowest in D. simulans. In contrast, adult cold tolerance was greater in D. auraria and adult heat tolerance was lower in D. immigrans than in the other species. These differences in life history traits and stress tolerance represent the Drosophila species differential adaptations, and are assumed to allow coexistence of the species.  相似文献

Role of sugars in nitrate utilization by roots of dwarf bean   总被引:4,自引:0,他引:4  
Nitrate uptake and in vivo, nitrate reductase activity (NRA) in roots of Phaseolus vulgaris, L. cv. Witte Krombek were measured in nitrogen-depleted plants of varying sugar status, Variation in sugar status was achieved at the start of nitrate nutrition by excision, ringing, darkness or administration of sugars to the root medium. The shape of the apparent induction pattern of nitrate uptake was not influenced by the sugar status of the absorbing tissue. When measured after 6 h of nitrate nutrition (0.1 mol m?3), steady state nitrate uptake and root NRA were in the order intact>dark>ringed>excised. Exogenous sucrose restored NRA in excised roots to the level of intact plants. The nitrate uptake rate of excised roots, however, was not fully restored by sucrose (0.03–300 mol m?3). When plants were decapitated after an 18 h NO3? pretreatment, the net uptake rate declined gradually to become negative after three hours. This decline was slowed down by exogenous fructose, whilst glucose rapidly (sometimes within 5 min) stimulated NG?3 uptake. Presumably due to a difference in NO3? due to a difference in NO3? uptake, the NRA of excised roots was also higher in the presence of glucose than in the presence of fructose after 6 h of nitrate nutrition. The sugar-stimulation of, oxygen consumption as well as the release of 14CO2 from freshly absorbed (U-14C) sugar was the same for glucose and fructose. Therefore, we propose a glucose-specific effect on NO3? uptake that is due to the presence of glucose rather than to its utilization in root respiration. A differential glucose-fructose effect on nitrate reductase activity independent of the effect on NO3? uptake was not indicated. A constant level of NRA occurred in roots of NO3? induced plants. Removal of nutrient nitrate from these plants caused an exponential NRA decay with an approximate half-life of 12 h in intact plants and 5.5 h in excised roots. The latter value was also found in roots that were excised in the presence of nitrate, indicating that the sugar status primarily determines the apparent rate of nitrate reductase decay in excised roots.  相似文献

The role of flowering in root‐fungal symbiosis is not well understood. Because flowering and fungal symbionts are supported by carbohydrates, we hypothesized that flowering modulates root‐beneficial fungal associations through alterations in carbohydrate metabolism and transport. We monitored fungal colonization and soluble sugars in the roots of Arabidopsis thaliana following inoculation with a mutualistic fungus Phomopsis liquidambari across different plant developmental stages. Jasmonate signalling of wild‐type plants, sugar transport, and root invertase of wild‐type and jasmonate‐insensitive plants were exploited to assess whether and how jasmonate‐dependent sugar dynamics are involved in flowering‐mediated fungal colonization alterations. We found that flowering restricts root‐fungal colonization and activates root jasmonate signalling upon fungal inoculation. Jasmonates reduce the constitutive and fungus‐induced accumulation of root glucose and fructose at the flowering stage. Further experiments with sugar transport and metabolism mutant lines revealed that root glucose and fructose positively influence fungal colonization. Diurnal, jasmonate‐dependent inhibitions of sugar transport and soluble invertase activity were identified as likely mechanisms for flowering‐mediated root sugar depletion upon fungal inoculation. Collectively, our results reveal that flowering drives root‐fungus cooperation loss, which is related to jasmonate‐dependent root soluble sugar depletion. Limiting the spread of root‐fungal colonization may direct more resources to flower development.  相似文献

Roots of eighteen potato genotypes, differing in tolerance of G. pallida, were grown from tuberpieces on agar in Petri dishes. Juveniles of G. pallida were inoculated directly onto root tips. Root length was measured at various times after inoculation. Inoculation reduced root growth within one day. At later stages, genotypes differed strongly in growth of inoculated roots. Between four and seven days after inoculation, growth of inoculated roots was not significantly correlated with growth of untreated roots, and was only poorly correlated with tolerance assessed in the greenhouse or in the field. However, multiple regression analysis revealed that the tolerance of the tested genotypes was associated with both the rate at which they induced hatching and the growth of roots after inoculation. The combination of these two variables accounted for high percentages explained variance.  相似文献

Information concerning the sugar status of plant cells is of greatimportance during all stages of the plant life cycle. The aim of this work wasto study primary carbohydrate metabolism in hairy roots of red beet. Growth ofhairy roots of red beet in vitro and changes in concentration of major nutrientsand sugar in the media were measured over a growth cycle of 16 days. We havealso determined the levels of key enzymes in the pathways of sucrose metabolism.Sucrose concentration decreased as hairy root growth proceeded while no changein glucose and fructose levels in the medium was found during the first 3 daysindicating that external sucrose is preferably taken to the cell before it ishydrolyzed by extracellular invertase. The increase in glucose and fructoselevels in the media after 5 days of culture indicates extracellular hydrolysisof sucrose which was further supported by the activity of acid invertaseobserved during that time in the culture medium. The uptake of mineral nutrientsby hairy root of red beet was monitored continuously during the culture cycle.The preferential use of NH4 + overNO3 at the beginning of the culture andacidification of culture media were the two most notable results concerningnitrogen nutrition during hairy root growth of red beet.  相似文献

Extracellular invertase of Rhizobium japonicum and its role in free sugar metabolism in the developing root nodules of Sesbania grandiflora L. was studied. The enzyme hydrolysed sucrose extracellularly, and its release was substrate inducible. 0.1 Mβ-mercaptoethanol released the cell-bound form of this enzyme. The production of invertase was low when glucose, galactose, mannose, fructose and raffinose were used as carbon sources in the growth medium. In the developing nodules sucrose was the major sugar. The content of fructose was low in comparison with that of glucose – suggesting that in the nodules, fructose is converted to glucose prior to its entry into the bacterial cell. The content of glucose synchronised with the pattern of change in the activity of invertase in the nodules.  相似文献

细根对氮沉降的生理生态响应将显著影响森林生态系统的生产力和碳吸存。为了揭示氮沉降对杉木细根的生理生态影响,对一年生杉木(Cunninghamia lanceolata)幼苗进行了模拟氮沉降试验,并测定施氮1年后杉木幼苗细根生物量、细根形态学特征(比根长、比表面积)、元素化学计量学指标(C、N、P、C/N、C/P、N/P)、细根代谢特征(细根比呼吸速率、非结构性碳水化合物)。结果表明:(1)杉木细根生物量随氮添加水平的升高而显著降低,尤其是0—1 mm细根生物量;细根比根长和比表面积随氮添加水平升高而显著增大。(2)氮添加后杉木细根C含量、C/N、C/P显著降低,高氮添加导致1—2 mm细根N含量和N/P显著升高,而低氮添加导致1—2 mm细根P含量显著升高、N/P显著降低,而0—1 mm细根的N、P含量则保持相对稳定。(3)氮添加后杉木细根比呼吸速率无显著变化,细根可溶性糖含量随氮添加增加而显著增加,而淀粉含量和NSC显著降低。综合以上结果表明:氮添加后用于细根形态构建的碳分配减少,这可能会减少土壤中有机碳的保留,0—1 mm细根的形态更易发生变化,但是其内部N、P养分含量相对更稳定以维持生理活动,细根NSC对氮添加的响应表明施氮可能导致细根受光合产物的限制。  相似文献
