在真菌(Fusarium oxysporum f.vasinfectum (Atkinson) Snyder et Hansen)寡聚糖诱导悬浮培养南方红豆杉(Taxus chinensis (Pilger) Rehd.var.mairei (Lemee et Lévl.) Cheng et L.K.Fu)细胞生产紫杉醇的体系中发现细胞出现凋亡,次生代谢增强.电镜观察到细胞核质和原生质出现凝集现象,液泡内出现大量的高电子致密体.核DNA经琼脂糖凝胶电泳,呈200 bp的整数倍的梯状条带(ladders);而对照组细胞核DNA完整,呈大片段,细胞完整,细胞器发达,但紫杉醇合成速率很低.加入寡聚糖后,细胞防御系统开启,细胞生长停止,次生代谢物酚类物质大量积累且次生壁加厚,多酚氧化酶活性迅速提高,苯丙烷类代谢途径的关键酶苯丙氨酸解氨酶的活性在1 h后急速提高,目的产物紫杉醇在诱导后72 h达到峰值,比对照组提高了6倍,且细胞凋亡的出现与紫杉醇合成的峰值具有时间上的一致性.  相似文献

寡聚糖是由2~10个单糖组成的一类聚合物,它属于碳水化合物,其结构特征、理化性质和生理功能却与单糖和多糖有着重大差别.经研究表明:寡聚糖具有低热值、甜味、稳定、安全无毒、粘度低、吸湿性大、不被胃肠道消化等理化特征以及具有改善肠道微生物区系、降低血清胆固醇和中性脂肪含量、改善血糖含量等生理功能.最近,国外研究人员又发现寡果糖还具有提高动物的兔疫性能、降低死亡率、促进生长、提高饲料利用效率和改善畜产品质量等功能.  相似文献

海洋寡聚糖对仔猪生产性能及血液理化指标的影响   总被引:2,自引:0,他引:2  
选择0-35日龄200头哺乳仔猪和35-70日龄160头断乳仔猪各分为对照组和寡聚糖组,每组10个重复,各重复仔猪数相等。研究探讨了海洋寡聚糖作为猪饲料添加剂对生产性能及血液理化指标的影响。结果表明:寡聚糖组与对照组相比,0-35日龄哺乳仔猪和35-70日龄保育期仔猪的死亡率、次品率均显著降低(P〈0.05),而平均体重分别提高18%和14.2%,显著高于对照组(P〈0.05);寡聚糖组白细胞、粒细胞、红细胞、血红蛋白、压积有减少的倾向,而淋巴细胞和血小板含量增加,但差异都不显著。与对照组相比寡聚糖组谷草转氨酶、谷丙转氨酶、碱性磷酸酶及乳酸脱氢酶的活力略有降低,谷草/谷丙比和乎谷氨酰转肽酶的活性呈上升趋势,但差异均不显著。寡聚糖组的肌酐含量比对照组显著增加外(P〈0.05),其余尿酸、葡萄糖、甘油三脂、高密度脂蛋白胆固醇、球蛋白、载脂蛋白A1的含量均下降,而尿素、胆固醇、低密度脂蛋白胆固醇、白蛋白、总蛋白含量比对照组略有增加的趋势,但差异均不显著。  相似文献

A method for determining low-molecular carbohydrates based on the use of HPLC with refractometric detection was modified. In contrast to previous methods, the determination of the qualitative and quantitative composition of sugars in plant extracts was performed without their preliminary separation into monosaccharides, disaccharides, and oligosaccharides. The exclusion of an additional separation stage made it possible to reduce twofold the time of analysis of a single sample, to save expensive materials necessary for carbohydrate HPLC, and to increase the useful life of the analytical column.  相似文献

Biotic elicitors produced by plant pathogens or herbivore pests rapidly activate a range of plant chemical defenses when translocated to plant tissue. The fatty acid conjugate volicitin has proven to be a robust elicitor model for studying herbivore-induced plant defense responses. Here we review the role of insect-derived volicitin (N-[17-hydroxylinolenoyl]-L-glutamine) as an authentic elicitor of defense responses, specifically as an activator of signal volatiles that attract natural enemies of herbivore pests. Comparisons are drawn between volicitin as an elicitor of plant defenses and two other classes of signaling molecules, C6 green-leaf volatiles and C4 bacterial volatiles that appear to prime plant defenses thereby enhancing the capacity to mobilize cellular defense responses when a plant is faced with herbivore or pathogen attack.  相似文献

Metabolites of a phytopathogenic fungus Botrytis cinerea Pers. were analyzed for the presence of biotic elicitors. Three groups of elicitors competent in inducing defense responses inAllium cepa cells were identified and partly purified. The recognition of the elicitor signal in onion cells was shown to elevate the concentration of reactive oxygen species (ROS), namely, superoxide anion-radical (O2^{\overset{-}.}) and hydrogen peroxide (22). The intensity of ROS release depended on chemical identity of elicitor and its concentration. The most active ROS production in onion cells was induced by a protein fraction isolated from the medium for fungus culturing. The carbohydrate elicitors extracted from the fungus cytoplasm and cell walls of mycelia were much less effective. The dynamics of ROS generation comprised two stages. The first stage represented fast and low-amplitude changes that peaked in 15 min after the elicitor treatment. The second stage was more durable and extensive; it occurred in 1.5–6 h after the treatment.  相似文献

Host genotype specificity in interactions between biotrophic fungal pathogens and plants in most cases complies with the gene-for-gene model. Success or failure of infection is determined by absence or presence of complementary genes, avirulence and resistance genes, in the pathogen and the host plant, respectively. Resistance, expressed by the induction of a hypersensitive response followed by other defence responses in the host, is envisaged to be based on recognition of the pathogen, mediated through direct interaction between products of avirulence genes of the pathogen (the so-called race-specific elicitors) and receptors in the host plant, the putative products of resistance genes. The interaction between the biothrophic fungusCladosporium fulvum and its only host tomato is a model system to study fungus-plant gene-for-gene relationships. Here we report on isolation, characterization and biological function of putative pathogenicity factors ECP1 and ECP2 and the race-specific elicitors AVR4 and AVR9 ofC. fulvum and cloning and regulation of their encoding genes. Disruption ofecp1 andecp2 genes has no clear effect on pathogenicity ofC. fulvum. Disruption of theavr9 gene, which codes for the race-specific 28 amino acid AVR9 elicitor, in wild type avirulent races, leads to virulence on tomato genotypes carrying the complementary resistance geneCf9. The avirulence geneavr4 encodes a 105 amino acid race-specific elicitor. A single basepair change in the avirulence geneavr4 leads to virulence on tomato genotypes carrying theCf4 resistance gene.  相似文献
