Without intending to minimize the importance of the study of mummies, especially those performed by our American colleagues, one must be aware that, for the foreseeable future consideration of skeletal material will be the mai source of information in Paleopathology. Therefore, it seems interesting to define the present state of our discipline, at the very time when it is coming back into favour, and is stated as an exact science. Three aspects of Paleopathology will be discussed: its possibilities, its limits and its prospects.  相似文献

This study investigates cross sectional growth patterns in the human skeleton using a recent skeletal sample of known age and sex. Measurements were selected to reflect different functional regions of the cranium, mandible and post cranial skeleton, and growth is evaluated using a single phase Gompertz curve. Different parts of the skeleton vary in the proportion of adult size attained at birth and in their subsequent rate of attainment of adult size. The paper introduces a method for the objective and quantitative comparison of the growth of different samples, and is used in this instance to analyze sexual differences in the growth of the post cranial skeleton. The development of sexual dimorphism is evaluated in terms of differences in the rate and duration of male and female growth. Adult sexual dimorphism is generally lower in early growing variables than in later-growing variables. There is considerable diversity in the ontogenetic basis of sexual dimorphism in the human skeleton demonstrating that the development of sexual dimorphism within a species should not be regarded as a uniform phenomenon. 1. Philosophically, the term ‘river health’ is useful because it is readily interpreted by the general public and evokes societal concern about human impacts on rivers. The common goal of achieving healthy rivers unites ecologists and the general public because the value of the ecologists’ contributions is clear (and, hence, funded). The difficulty arises in the choice of relevant symptoms because there is a wide variety that can be measured with varying accuracy at a broad range of spatial scales. These indicators may respond to impacts at different time scales, and no single indicator is a ‘silver bullet’ that reveals river health unequivocally. 2. In practice, choice of indicator often shows personal bias, technical considerations, and constraints of knowledge. Selection of appropriate spatial and temporal scales for these measures is crucial. Although most measurements are spot samples (e.g. concentration, abundance, species richness), assessment of river health based on changes in ecological processes such as post-disturbance recovery rate or nutrient spiralling lengths may be more suitable in some cases. 3. Problems include validation of the indicator, its response time at a range of scales, and the reliability of its measurement. Assessment of river health should be accurate, timely (warning of deterioration instead of waiting until the patient is terminal), rapid (so that the response is swift), and inexpensive. The connectedness of running waters with their floodplains and catchments must be explicitly recognized. Hydrological and geomorphological modifications of rivers usually affect their health by severing or impairing the linkages, and the ‘cure’ may lie in addressing these causes. Often, we need landscape-level data for management because this is the scale where cumulative effects of impacts are evident. 4. The prognosis is uncertain. We need to explore further the use of integrative measures of river health, and focus on establishing a link between the measure and impaired ecological integrity. Ecosystem-level variables (e.g. estimates of production or respiration) show promise and recent technological advances make these more accessible. Data analytical approaches (e.g. multimetric vs. predictive models) need further debate but must not overlook the importance of high quality and relevant input data. Appropriate choice of indicators, rigorous sampling and analysis, and careful data interpretation must be matched with effective communication to policy-makers and the public. When this occurs, the concept of ‘river health’ becomes more than just a rhetorical tool.‘  相似文献

The typing of C. albicans by MLEE (multilocus enzyme electrophoresis) is dependent on the interpretation of enzyme electrophoretic patterns, and the study of the epidemiological relationships of these yeasts can be conducted by cluster analysis. Therefore, the aims of the present study were to first determine the discriminatory power of genetic interpretation (deduction of the allelic composition of diploid organisms) and numerical interpretation (mere determination of the presence and absence of bands) of MLEE patterns, and then to determine the concordance (Pearson product-moment correlation coefficient) and similarity (Jaccard similarity coefficient) of the groups of strains generated by three cluster analysis models, and the discriminatory power of such models as well [model A: genetic interpretation, genetic distance matrix of Nei (d(ij)) and UPGMA dendrogram; model B: genetic interpretation, Dice similarity matrix (S(D1)) and UPGMA dendrogram; model C: numerical interpretation, Dice similarity matrix (S(D2)) and UPGMA dendrogram]. MLEE was found to be a powerful and reliable tool for the typing of C. albicans due to its high discriminatory power (>0.9). Discriminatory power indicated that numerical interpretation is a method capable of discriminating a greater number of strains (47 versus 43 subtypes), but also pointed to model B as a method capable of providing a greater number of groups, suggesting its use for the typing of C. albicans by MLEE and cluster analysis. Very good agreement was only observed between the elements of the matrices S(D1) and S(D2), but a large majority of the groups generated in the three UPGMA dendrograms showed similarity S(J) between 4.8% and 75%, suggesting disparities in the conclusions obtained by the cluster assays.  相似文献

It is of central interest in biology to understand how gene activity networks are coordinated and integrated in the cell. Within the field of genomics, microarray technologies have become a powerful technique for monitoring simultaneously the expression patterns of thousands of genes under different sets of conditions. A main task now is to propose analytical methods that can suggest which groups of genes are activated by similar conditions. We review several techniques based on self-organizing map and clustering algorithms but implemented through a network of units controlled by biologically inspired functions (see Table 1). The computer tool, named NBIA, permits a categorization that generates a set of gene groups with coordinated expression patterns.  相似文献

Blood glucose is the primary cellular substrate and in vivo must be tightly maintained. The liver plays a key role in glucose homeostasis increasing or decreasing glucose output and uptake during fasting and feeding. Glucokinase (GCK) is central to this process. Its activity is modulated in a coordinated manner via a complex set of mechanisms: in the postprandial period, the simultaneous rise in glucose and insulin increases GCK activity by enhanced gene expression, changes in cellular location, and interaction with regulatory proteins. Conversely, in the fasting state, the combined decrease in glucose and insulin concentrations and increase in glucagon concentrations, halt GCK activity. Herein we summarize the current knowledge regarding the regulation of hepatic GCK activity.  相似文献

Aluminium (Al) is associated with many clinical disorders in renal patients. Al accumulation in brain has also been related to the neurodegenerative processes in Alzheimer’s disease. In order to better understand Al transport in the human body, it is necessary to identify and quantify chemical species in which Al is present in body fluids and tissues. Among a variety of biological samples, Al speciation was the most frequently investigated in human serum. Improvements were made in the development of analytical techniques for the determination of the amount and composition of high molecular mass Al (HMM-Al) and low molecular mass Al (LMM-Al) species in human serum. However, due to the complex chemistry of Al in serum, its low total concentration and the high risk of contamination, speciation of Al in biological samples is still a difficult task for analytical chemists. In this work, problems related to speciation of Al in human serum are critically discussed. An overview of the progress that was made by the use of different analytical procedures, in order to propose analytical protocols for reliable speciation of Al in serum at low ng mL−1 concentration range, is presented.  相似文献

This research evaluates the interobserver error when the macroscopic methods recommended by American and European anthropologists to estimate age at death of a skeleton, were applied to a sample of the Terry Collection (Smithsonian Institution, Washington D.C.). Although no statistical differences among observers were found for any of these methods, small dissimilarities suggest that techniques using a narrower scale of categories produce greater agreement among researchers. The present study is within a wider research project designed to evaluate the accuracy of these methods, when applied to an identified (age known) sample of 963 skeleton from the Terry Collection.  相似文献

In the larval fat body of Drosophila gibberosa, polytene chromosome structure and activity exhibit cytological differences from chromosomes of midgut and salivary glands. These differences include long-persisting puffs, transient puffs and long-persisting band modulations. Some early ecdysteroid-induced puffs are present in all three organs but few late puffs are present in the fat body. Comparative studies reveal, therefore, that late larval-early pupal puffing is enhanced in salivary glands relative to gut, fat body and Malpighian tubules. After the fat body breaks up in the prepupa, the rate of programmed cell death and the corresponding slow decline of chromosomal activity also differ from cell to cell and from other organs.by M.L. Wolbachia is a genus of bacterial endosymbionts that impacts the breeding systems of their hosts. Wolbachia can confuse the patterns of mitochondrial variation, including DNA barcodes, because it influences the pathways through which mitochondria are inherited. We examined the extent to which these endosymbionts are detected in routine DNA barcoding, assessed their impact upon the insect sequence divergence and identification accuracy, and considered the variation present in Wolbachia COI. Using both standard PCR assays (Wolbachia surface coding protein--wsp), and bacterial COI fragments we found evidence of Wolbachia in insect total genomic extracts created for DNA barcoding library construction. When >2 million insect COI trace files were examined on the Barcode of Life Datasystem (BOLD) Wolbachia COI was present in 0.16% of the cases. It is possible to generate Wolbachia COI using standard insect primers; however, that amplicon was never confused with the COI of the host. Wolbachia alleles recovered were predominantly Supergroup A and were broadly distributed geographically and phylogenetically. We conclude that the presence of the Wolbachia DNA in total genomic extracts made from insects is unlikely to compromise the accuracy of the DNA barcode library; in fact, the ability to query this DNA library (the database and the extracts) for endosymbionts is one of the ancillary benefits of such a large scale endeavor--which we provide several examples. It is our conclusion that regular assays for Wolbachia presence and type can, and should, be adopted by large scale insect barcoding initiatives. While COI is one of the five multi-locus sequence typing (MLST) genes used for categorizing Wolbachia, there is limited overlap with the eukaryotic DNA barcode region.  相似文献

Methods for the determination of aflatoxins in paprika, peanut butter, pistachio paste, fig paste and baby food were developed. The methods employ an immunoaffinity cleanup step and reversed-phase liquid chromatography. All steps of the analysis were tested for their suitability for all matrices with focus on method robustness, simplicity, toxicology, environment, and user friendliness. Extraction procedures, chromatographic separation and post column derivatisation techniques were elaborated for this purpose. The methods were statistically validated in collaborative trials at currently established legal limits for aflatoxins and are in the process for adoption as official methods by CEN and AOAC.  相似文献
