五十年代成功地解决了核乳胶和核素标记化合物的制备,为放射自显影术在生物学研究中的应用创造了必要的条件。1955年Hughes用氚标记胸腺嘧啶核苷(以下简称_3H-TdR)成功(Cronkite et al., 1959),它是参与胞核DNA合成代谢的特异性大分子核苷类标记化合物,为细胞动力学研究提供了新的较好的试剂。但计数标记的核分裂象或标记的细胞数工作量较大。1956年Friedkin等成功地用~14C-TdR先后掺入到鸡胚羢毛-尿囊膜、鸡骨髓、胸腺、睾丸、肺、小肠粘膜、兔胸腺等组织DNA中。以后~14C-TdR广泛用于细胞动力学的研究。它比用~3H-TdR简便、省时,对环境污染易于监测(Cronkite 相似文献

前列腺素(PGS)作为体内一种活性物质,几乎存在于哺乳动物体内的各重要组织及体液中。其含量虽微,但代谢迅速,活力很高。对机体的各系统组织具有多种生理生化效应,因而国外的研究甚为活跃。国内自1979年建立PGS的放射免疫测定方法(陈四传,1982)后,应用家兔等动物开展对PGS的实验研究日益增多。为实验设计需要,我们用放射免疫法测定了正常家兔血浆及大脑皮质、肾髓质内PGA_2、PGE_1、及PGE_(2a)含量,总结成本文。 相似文献

本文报道用Singer氏法测得40例正常儿童(2—12岁)的胎儿血红蛋白值为1.27±1.05%;36例正常成人为0.89±0.83%。根据测定结果初步说明2岁以上各年龄组儿童胎儿血红蛋白无明显差异。40名正常儿童胎儿血红蛋白的测定结果与36名成人测定结果比较也无显著差异(P>0.05),表明2岁以上儿童胎儿血红蛋白值已趋稳定。 相似文献

1997年Asahara等在人外周血中发现内皮祖细胞(endothelial progenitorcell.EPC),随后有文献报道,从骨髓中分离出EPC.异体骨髓移植研究显示成体外周血EPC来源于骨髓。许多作者报告EPC参与生理性和病理性血管新生.具有潜在的临床应用前景。目前关于EPC的不少基本生物学问题尚不完全清楚,其生理和病理意义也有争议。 相似文献

为探索组织化学方法检测标志基因在正常骨髓细胞及白血病细胞的表达,本研究利用脂质体介导将NeoR基因和LacZ基因共转移正常人及白血病人骨髓细胞。然后,采用X-gal染色的方法,我们观察了LacZ基因在转导细胞的表达。结果显示:LacZ基因在正常人及白血病人骨髓细胞表达的阳性率分别为13.33%±2.68%和15.39±3.69%。经G418筛选后,转导阳性细胞率达到46.06%±3.47%和48.22%±4.47%。提示:经脂质体介导LacZ基因在正常骨髓细胞及白血病细胞可获得高效的转移率。同时表明,利用组织化学方法可有效地检测标志基因的表达。 相似文献

多发性骨髓瘤的骨髓细胞置于Dexter长期培养体系中,浆细胞可以长期生长。非贴壁层细胞形成CFU-GM的能力达6周以上,但随着培养时间的延长,CFU-GM的形成活性明显下降。浆细胞产生兔疫球蛋白单克隆特异性在Dexter长期培养体系中仍维持不变,但Ki-67阳性的浆细胞随着培养时间的延长而减少,以上结果说明浆细胞虽然可以在Dexter培养体系中生长并保持单克隆特异性,但此种培养体系不是浆细胞最佳的生长条件。本研究还表明Dexter长期培养不宜作为骨髓净化的手段用于多发性骨髓瘤的自体骨髓移植。 相似文献

本文以101/E1和C3H/E1的杂种第一代小鼠为材料,一次性腹腔注射秋水仙碱(COM)后,于不同时间取材,观察分析了小鼠骨髓细胞有丝分裂指数(MI)和C-有丝分裂的变化。结果表明:COM处理后2小时,MI和C-有丝分裂均已达到最高;并随处理时间延长而降低;至18小时,MI已降到(1mg/kg)或显著低于(3mg/kg)对照水平,而C-有丝分裂仍显著高于对照组。并对COM影响MI的可能机制以及C-有丝分裂效应与非整倍体诱导活性之间的关系进行了讨论。 相似文献

体外化学诱导人骨髓间充质干细胞分化为心肌样细胞 总被引:1,自引:0,他引:1
为了探讨人骨髓间充质干细胞(MSCs)的体外培养及化学诱导向心肌细胞分化的过程及条件,我们用1.073g/mL密度梯度离心法分离健康人骨髓单个核细胞,经骨髓间充质干细胞培养基传代培养后用流式细胞仪检测细胞表面抗原,在完全培养基中分别加入3、5、10μmol/L的5氮胞苷(每组n=5)进行化学诱导分化,阴性对照组采用完全培养基培养,诱导后21天细胞爬片免疫荧光法鉴定,透射电镜观察细胞超微结构。结果显示人MSCs为形态均一的梭形细胞,生长旺盛时呈旋涡样分布,流式细胞仪检测细胞表面CD44阳性,CD34、CD45阴性;5、10μmol/L的5氮胞苷进行化学诱导后细胞形态变长,诱导后14天时20%-30%细胞融合形成多核肌管样结构,3μmol/L组MSCs未出现肌管结构,诱导后21天5、10μmol/L组MSCs中desmin、心肌早期转录因子GATA4、心肌特异性cTnI及闰盘蛋白connexin43的表达阳性,10μmol/L组cTnI阳性染色细胞数目(65.3±4.7%)高于5μmol/L诱导组(48.2±5.4%)(p<0.05);3μmol/L组及阴性对照组无心肌特异性蛋白的表达。细胞诱导后28天透射电镜下可见肌丝形成。本实验说明,人MSCs在体外经化学诱导可分化为心肌样细胞,而且5-氮胞苷对于心肌相关蛋白的表达呈浓度依赖性正相关。 相似文献

The influence of the bone marrow on the repopulation of the thymus in X-irradiated mice has been investigated.
It was observed that the thymus and a certain population of bone marrow lymphocytic cells were repopulated in parallel in a cyclic fashion. This occurred either after a single exposure of mice to 400 R or after serial weekly X-ray treatments with 170 R. Lethally irradiated recipients which were grafted with bone marrow cells obtained 12-24 days after four weekly irradiations of donor mice with 170 R also exhibited a cyclic repopulation of both the thymus and the bone marrow lymphocytic population. In contrast, mice which were transplanted with bone marrow cells from unirradiated donors, containing an equal number of stem cells (CFU), exhibited a continuous rather than a cyclic recovery of both cell populations. the bone marrow stem cells of mice recovering from X-irradiation were found to have a decreased proliferative activity, since they produced significantly smaller spleen colonies in lethally irradiated recipients than marrow cells from unirradiated mice.
The results were interpreted as indicating that the bone marrow lymphocytic cells may act as thymic precursor cells and that thymic lymphopoiesis is dependent on the presence of such cells. Evidently, the production of lymphocytic cells will decrease when the stimulus for granulocyte production increases due to the limited proliferative activity of the surviving bone marrow stem cells after irradiation. This may result in a cyclic variation of the production of bone marrow lymphocytic cells and it follows that thymic lymphopoiesis will run parallel. 相似文献

本文对几种化学诱变剂诱发小鼠体内脾脏、骨髓和精原细胞的SCE进行了比较研究,同时分析了几类常见化合物在小鼠脾脏细胞中诱发SCE的活力。结果显示诱变剂在脾脏细胞中诱发SCE比骨髓和精原细胞敏感。几类化合物都能显著地诱发小鼠脾脏SCE的增加,与对照相比差异显著(P<0.05)或极显著(P<0.01),说明利用小鼠脾脏细胞检测环境诱变物是相当灵敏的。 相似文献

本文报告狗骨髓长期培养的初步研究结果。采用马血清与狗血清混合加入培养体系中,可建立起比较稳定的贴壁细胞层,可以维持19周以上。但是,这种贴壁层不能有效地维持造血干细胞的增殖与分化,上清液中血细胞与GM-CFU_C的数目,在第二次接种骨髓后2—3周即迅速下降。狗骨髓培养的条件液中存在着抑制GM-CFU_C生长的物质,但它对小鼠骨髓GM-CFU_C和CFU-S无影响。如以贴壁细胞作为底层,用双层琼脂平皿培养法与正常狗骨髓共同培养,发现贴壁细胞不仅没有抑制反而有加强CSF刺激GM-CFU_C生长的作用。因此对狗骨髓长期培养体系不能长期支持造血干细胞生长和繁殖的可能原因尚须作进一步探索。 相似文献
